Bromochloromethane (BCM), a halogenated methane analogue, was evaluated for its anti-methanogenic activity in both batch and continuous fermentation systems. For batch fermentation, a roughage-based substrate was incubated with mixed rumen microflora for 24 h at two concentrations of BCM (5 and 10 μm). A 89–94 % reduction in methane was obtained in terms of volume and truly degraded substrate (TDS; P < 0·05). The partitioning factor (an index of efficiency of microbial protein production; expressed as mg TDS/ml net gas produced) increased from 3·55 to 3·73 (P < 0·05), while the acetate:propionate proportion decreased from 2·80 to 2·22 (P < 0·05). A complete inhibition of methanogens was associated with a 48 % decrease in Ruminococcus flavefaciens, a 68 % increase in Fibrobacter succinogenes and a 30 % increase in rumen fungi when quantified using qPCR. The continuous fermentation was carried out using the roughage-based substrate in four fermenters, two fermenters being control and the other two fed with BCM (5 μm) once in a day, for nine consecutive days. A persistent effect of BCM on methane reduction (85–90 %) was obtained throughout the study (P < 0·05) with no effect on gas production, SCFA production, acetate:propionate proportion, true degradability and efficiency of microbial mass synthesis (P>0·05). The complete inhibition of methane resulted in a significant decrease in R. flavefaciens and methanogens (P < 0·05) and an increase in fungal population (P < 0·05), while there was no effect on total bacterial and F. succinogenes populations (P>0·05). The batch fermentation confirms the anti-methanogenic activity of BCM, while the continuous fermentation indicates the persistency of this effect under in vitro conditions.

Zn is an essential trace element required throughout the life cycle. Although suboptimal Zn status is thought to be common in many sub-Saharan countries, there is a paucity of data in the Democratic Republic of Congo. The objective of the study was to determine Zn status in non-pregnant Congolese women. We measured plasma Zn and indicators of nutritional status (albumin, prealbumin, retinol-binding protein) and inflammation (C-reactive protein (CRP), ceruloplasmin, α1-acid glycoprotein (AGP)) in seventy-seven lactating and thirty non-lactating women (mean age 28 and 31 years, respectively). Blood samples were collected in summer 1989 in rural Bas-Congo during a survey on Fe status. Mean lactation period was 8·3 months. Mean parity was higher in lactating (3·6) than in non-lactating (2·2) women (P < 0·05). Mean biochemical indicators of nutritional status, CRP and ceruloplasmin were within normal range and not different between groups. Mean AGP concentrations were above normal (>1·2 g/l) and higher in lactating (1·365 g/l) than in non-lactating (1·178 g/l) women (P < 0·05). Mean Zn concentration (540 μg/l) of the overall study population was below normal (700 μg/l); and the mean was lower in lactating (455 μg/l) than in non-lactating (759 μg/l) women (P < 0·05). Multiple regression analysis suggested that parity (P < 0·05), but not inflammation, was the most important factor associated with low Zn levels. Despite the lack of data on dietary intake, the results suggest that suboptimal Zn status may be common in the studied population.

Dietary supplements of conjugated linoleic acid (CLA) containing trans-10, cis-12 CLA decrease milk fat secretion in the lactating cow and sheep, but their effects on mammary lipogenesis in the goat are less well defined. Eight lactating goats were used in two 4 × 4 Latin-square experiments with 14 d experimental periods to examine the effects of calcium salts of CLA methyl esters (CaCLA) containing trans-10, cis-12 on milk fat synthesis. Experimental treatments consisted of incremental inclusion of 0, 30, 60 or 90 g of CaCLA/d (corresponding to 7·47, 14·9 and 22·4 g/d of trans-10, cis-12 CLA) offered during the first 10 d of each experimental period that replaced maize grain in concentrates (Experiment 1) or calcium salts of palm oil fatty acids (Experiment 2). Relative to the control, inclusion of 30, 60 or 90 g CaCLA/d in the diet reduced milk fat yield by 19·8, 27·9 and 32·3 % and 17·5, 39·0 and 49·3 % in Experiments 1 and 2, respectively. Decreases in milk fat were due to reductions in the secretion of fatty acids synthesised de novo rather than the uptake of fatty acids from the peripheral circulation. Indirect comparisons with the studies in the lactating cow indicated a lower efficacy of CaCLA supplements on mammary lipogenesis in the goat. In conclusion, CaCLA in the diet inhibits milk fat synthesis in the goat, responses that are dependent on the supply of dietary fatty acids, with evidence that the caprine is less sensitive to the anti-lipogenic effects of trans-10, cis-12 CLA compared with the bovine or ovine.

Poor quality of nutrition during fetal development is associated with adverse health outcomes in adult life. Epidemiological studies suggest that markers of fetal undernutrition are predictive of risk of the metabolic syndrome and CHD. Here we show that feeding a low-protein diet during pregnancy programmed the development of atherosclerosis in ApoE*3-Leiden mice. ApoE*3-Leiden mice carry a mutation of human ApoE*3 rendering them prone to atherosclerosis when fed a diet rich in cholesterol. It was noted that fetal exposure to protein restriction led to a greater degree of dyslipidaemia in mice when fed an atherogenic diet, with low-protein-exposed ApoE*3 mice having elevated total plasma cholesterol (34 % higher; P < 0·001) and TAG (39 % higher; P < 0·001) relative to offspring exposed to a control diet in utero. The low-protein group developed more severe atherosclerotic lesions within the aortic arch (2·61-fold greater lesion area; P < 0·001). Analysis of a targeted gene array suggested a potential role for members of the LDL receptor superfamily, along with similar programmed suppression of the mRNA expression of hepatic sterol regulatory element-binding protein-1c. This indicates that disordered lipid metabolism may play a role in the fetal programming of atherosclerosis in this model. Whereas earlier studies have shown early programming of cardiovascular risk factors, these results demonstrate for the first time that the interaction of prenatal undernutrition with a postnatal atherogenic diet increases the extent of atherosclerotic disease.

Enteral tube feeding remains an indispensible strategy to treat disease-related malnutrition. In the present study we evaluated in clinical practice whether prescribed feeding volumes correspond with administered quantities and we highlight possible causes for discrepancies. During a 4-month observation period data from all patients fully depending on tube feeding (1·5–2·5 litres/d) were collected in a Dutch 900-bed academic hospital. The range for administered feeds to be adequate was set at 100 ± 10 % of the prescribed dose. Fifty-five patients (mean age 57 (sd 30) years) were included. Tube feeding was given continuously via pump (n 37) or drip (n 3), in portions (n 14) or by combined modes (n 1). Administered tube feeding amounts were significantly lower than prescribed in 40 % of all patients (P ≤ 0·001). The mean ratio of administered v. prescribed energy was 87 (sd 21) % (all modes), 85 (sd 24) % (pump), 94 (sd 12) % (portions) and 88·3 (sd 18·1) % (drip), respectively. The mean energy deficit amounted to 1089 kJ/d (range − 7955 to +795). Only on intensive care unit wards did feeding administration meet the set goal. Feeding interruptions because of diagnostic or therapeutic procedures were the main reason for decreased intakes. Our findings show that many patients relying on tube feeding do not meet their nutritional goals during hospital stay. This problem can be addressed by adapting feeding schedules and the use of formulations with a higher energy density.

Mate (Ilex paraguariensis) is rich in polyphenolic compounds, which are thought to contribute to the health benefits of tea. Mate tea was administered orally to mice at a dose of 0·5, 1·0 or 2·0 g/kg for 60 d, and changes both in serum lipid concentration and fatty acid composition of liver and kidney were examined. The effects of mate tea on serum and tissue lipid peroxidation were assessed by the evaluation of thiobarbituric acid-reactive substances (TBARS). In tea-consuming mice, both MUFA (18 : 1n-9) and PUFA (18 : 2n-6 and 20 : 4n-6) were increased (P < 0·05) in the liver lipid (approximately 90 and 60 %, respectively), whereas only MUFA (approximately 20 %) were increased in the kidney lipid. The most altered PUFA class was n-6 PUFA, which increased by approximately 60–75 % (P < 0·05). This difference in the fatty acid profile in the liver is reflected in the increased PUFA:SFA ratio. Consistent with these results, mice fed with mate tea had much lower TBARS in the liver. No differences (P>0·05) were found in the levels of serum cholesterol, HDL-cholesterol and TAG under the conditions of the present study. These results suggest that treatment with mate tea was able to protect unsaturated fatty acids from oxidation and may have selective protective effects within the body, especially on the liver.

The aim of the present work was (a) to investigate trypsinolysis of denatured purified T phaseolin (Phaseolus vulgaris) subunits by MS and (b) to test the effect of raw T phaseolin inclusion level in diets fed chronically to rats on digestion in the small intestine. The diets contained casein as the sole protein source, or casein substituted with 33, 67 and 100 % of purified T phaseolin. Rats were fed for 10 d and then euthanised. Digesta and tissues from the first and second halves of the small intestine were prepared for electrophoresis, immunoblotting and densitometry. α-Phaseolin subunit for the T phaseolin was more resistant to trypsinolysis than β-phaseolin subunit. Nearly intact phaseolin subunits (molecular weight, MW 44–54 kDa) and partially digested phaseolin fragments (MW 17–19 and 20–24 kDa) were identified in small intestinal digesta. The concentration of intact phaseolin and of most undigested phaseolin fragments in digesta increased in the second half of the small intestine with increasing phaseolin intake (P < 0·05–0·01). The concentration of phaseolin fragments of a MW of 21–22·5 and 23–24·5 kDa in the mucosa increased linearly (P = 0·016–0·084) when the level of the T phaseolin was increased in the diet. In conclusion, the present work provides evidence that denatured T phaseolin subunits display different trypsinolysis patterns in vitro. Moreover, a high intake of raw T phaseolin impacts digestion in the small intestine of rats.

Trace amines, including tyramine and β-phenylethylamine (β-PEA), are constituents of many foods including chocolate, cheeses and wines and are generated by so-called ‘friendly’ bacteria such as Lactobacillus, Lactococcus and Enterococcus species, which are found in probiotics. We therefore examined whether these dietary amines could exert pharmacological effects on the gut and its vasculature. In the present study we examined the effects of tyramine and β-PEA on the contractile activity of guinea-pig and rat ileum and upon the isolated mesenteric vasculature and other blood vessels. Traditionally, these amines are regarded as sympathomimetic amines, exerting effects through the release of noradrenaline from sympathetic nerve endings, which should relax the gut. A secondary aim was therefore to confirm this mechanism of action. However, contractile effects were observed in the gut and these were independent of noradrenaline, acetylcholine, histamine and serotonin receptors. They were therefore probably due to the recently described trace amine-associated receptors. These amines relaxed the mesenteric vasculature. In contrast, the aorta and coronary arteries were constricted, a response that was also independent of a sympathomimetic action. From these results, we propose that after ingestion, trace amines could stimulate the gut and improve intestinal blood flow. Restriction of blood flow elsewhere diverts blood to the gut to aid digestion. Thus, trace amines in the diet may promote the digestive process through stimulation of the gut and improved gastrointestinal circulation.

The purpose of this study was to determine whether n-3 PUFA result in an effect on endothelial function that is in addition to that of acute exercise. For 4 weeks, male Sprague–Dawley rats were subjected to a diet based on n-3 PUFA or a standard diet. In each diet group, ten rats were submitted to an acute treadmill exercise while the remaining ten acted as sedentary controls. The running speed was progressively increased until the animals were exhausted. Endothelial function was then assessed by measuring isometric tension in rings of the thoracic aorta. In vessels precontracted with 0·1 μm-phenylephrine, responses to acetylcholine (ACh) were significantly improved following acute exercise in all diet groups. When PUFA supplementation was compared to the standard diet no significant difference was found in response to ACh, either at rest or after an acute exercise. Pretreatment of rings with Nω-nitro-l-arginine methyl esther (50 μm) inhibited the ACh-mediated vasorelaxation in all groups. Response to 10 μm-nifedipine, an L-type Ca2+ channel antagonist, was similarly enhanced after acute exercise in both standard and PUFA diets. Furthermore, response to 0·01 μm-nifedipine was significantly higher after acute exercise only in the PUFA diet. In conclusion, in our ‘healthy’ rat model with ‘normal’ baseline endothelial function, acute exercise improves response to ACh while PUFA supplementation alone or in combination with acute exercise has no effect on endothelium-dependent vasorelaxation. However, PUFA may potentiate the acute exercise effect on smooth muscle cell relaxation via L-type Ca2+ channel modifications.

In order to assess nutritional adequacy, valid estimates of nutrient intake are required. One of the main errors in dietary assessment is misreporting. The objective was to review the extent, nature and determinants of misreporting in dietary assessment, how this affects reported intakes of micronutrients and how this is identified and measured, and to identify the best ways of dealing with misreporting when interpreting results. A systematic literature search was conducted for studies of misreporting of dietary intake in adults by 24 hour recalls or by estimated or weighed food records, published up to March 2008. Thirty-seven relevant studies were identified. Possible causes of misreporting were identified. Methods most used to identify misreporting were the Goldberg cut-off (46 % studies) and the doubly labelled water technique (24 % studies). The magnitude of misreporting of energy intake was similar in all three dietary assessment methods. The percentage of under-reporters was about 30 % and energy intake was underestimated by approximately 15 %. Seven papers presented usable data for micronutrient intake. Absolute intakes of Fe, Ca and vitamin C (the three micronutrients addressed in all papers) were on average 30 % lower in low-energy reporters (LER) than that in non-LER and, although results were not consistent, there was a tendency for micronutrient density to be higher in LER. Excluding underreporters or using energy adjustment methods for micronutrient intakes is discussed. Residual method of energy adjustment seems to be a good tool for practice to decrease an influence of misreporting when interpreting results of studies based on food records and 24 hour recalls.

Insulin resistance (IR) and non-alcoholic fatty liver disease (NAFLD) are found in 35 and 30 % of US adults, respectively. Trans-10, cis-12-conjugated linoleic acid (CLA) has been found to cause both these disorders in several animal models. We hypothesised that IR and NAFLD caused by CLA result from n-3 fatty acid deficiency. Pathogen-free C57BL/6N female mice (aged 8 weeks; n 10) were fed either a control diet or diets containing trans-10, cis-12-CLA (0·5 %) or CLA+flaxseed oil (FSO) (0·5 %+0·5 %) for 8 weeks. Weights of livers, concentration of circulating insulin, values of homeostatic model 1 (HOMA1) for IR and HOMA1 for β cell function were higher by 160, 636, 985 and 968 % in the CLA group compared with those in the control group. FSO decreased fasting glucose by 20 % and liver weights by 37 % compared with those in the CLA group; it maintained circulating insulin, HOMA1-IR and HOMA1 for β cell function at levels found in the control group. CLA supplementation decreased n-6 and n-3 wt% concentrations of liver lipids by 57 and 73 % and increased the n-6:n-3 ratio by 58 % compared with corresponding values in the control group. FSO increased n-6 and n-3 PUFA in liver lipids by 33 and 342 % and decreased the n-6:n-3 ratio by 70 % compared with corresponding values in the CLA group. The present results suggest that some adverse effects of CLA may be due to n-3 PUFA deficiency and that these can be corrected by a concomitant increase in the intake of α-linolenic acid, 18 : 3n-3.

We evaluated the effect of undegradable intake protein (UIP) on urea kinetics and microbial incorporation of urea-N in ruminally and duodenally fistulated steers (n 4; 319 kg) provided ad libitum access to grass hay in a 4 × 4 Latin square. Casein was continuously infused abomasally in amounts of 0, 62, 124 and 186 mg N/kg body weight per d to simulate provision of UIP. Periods were 13 d long with 7 d for adaptation and 6 d for collection. Jugular infusion of [15N15N]urea followed by determination of urinary enrichment of [15N15N]urea and [14N15N]urea was used to measure urea kinetics. Forage and N intake increased (quadratic, P < 0·02) with increasing UIP. Urea synthesis was 27·1, 49·9, 82·2 and 85·8 g urea-N/d for 0, 62, 124 and 186 diets, respectively (linear, P < 0·01). The proportion of urea synthesis that entered the gastrointestinal tract was 0·96 for steers receiving no UIP and decreased linearly (P = 0·05) to a low of 0·89 for steers receiving 186. The amount of urea entering the gastrointestinal tract was least for 0 (26·3) and increased (linear, P < 0·01) to 48·7, 77·2 and 76·6 g urea-N/d for 62, 124 and 186 diets, respectively. Microbial incorporation of recycled urea-N increased quadratically (P = 0·04) from 13·9 for 0 to 47·7 g N/d for 124. The proportion of microbial N derived from recycled urea increased (quadratic, P = 0·05) from 0·31 to 0·58 between 0 and 124 and dropped to 0·44 for 186 mg N/kg body weight per d. UIP increased intake of hay and provided a N source for ruminal microbes via urea recycling.

The aim of the present study was to evaluate the cariogenicity of cows', human and infant formula milks, supplemented or not with fluoride, in rats. Sixty female Wistar rats were desalivated and infected with Streptococcus sobrinus 6715.Animals were divided into six groups: group 1, sterilised deionised distilled water (SDW; negative control); group 2, 5 % sucrose added to SDW (positive control); group 3, human milk; group 4, cows' milk; group 5, Ninho® formula reconstituted with SDW; group 6, Ninho® formula reconstituted with 10 parts per million F and SDW. At day 21 the animals were killed and their jaws removed to quantify total cultivable microbiota, Strep. sobrinus and dental caries. The concentration of carbohydrate and fluoride in the milks was analysed. The Kruskal–Wallis test (α = 5 %) was used to analyse the data. The caries score by the milk formula was as high as that provoked by sucrose. Regarding smooth-surface caries, human milk was statistically more cariogenic than cows' milk, which did not differ from the SDW and the Ninho® with fluoride (P>0·05). Groups 2–6 showed higher Strep. sobrinus counts when compared with the negative control group (P < 0·05) but no statistically significant difference was found among them (P>0·05). HPLC analysis showed that infant formula had 9·3 % sucrose and 3·6 % reducing sugars. The infant formula should be considered cariogenic due to the sugars found in it, but fluoride supplementation reduced its cariogenic effect. The human milk was more cariogenic than the cows' milk but not as much as the formula milk.

Very low-carbohydrate diets are often used to promote weight loss, but their effects on bowel health and function are largely unknown. We compared the effects of a very low-carbohydrate, high-fat (LC) diet with a high-carbohydrate, high-fibre, low-fat (HC) diet on indices of bowel health and function. In a parallel study design, ninety-one overweight and obese participants (age 50·6 (sd 7·5) years; BMI 33·7 (sd 4·2) kg/m2) were randomly assigned to either an energy-restricted (about 6–7 MJ, 30 % deficit) planned isoenergetic LC or HC diet for 8 weeks. At baseline and week 8, 24 h urine and faecal collections were obtained and a bowel function questionnaire was completed. Compared with the HC group, there were significant reductions in the LC group for faecal output (21 (sd 145) v. − 61 (sd 147) g), defecation frequency, faecal excretion and concentrations of butyrate ( − 0·5 (sd 10·4) v. − 3·9 (sd 9·7) mmol/l) and total SCFA (1·4 (sd 40·5) v. − 15·8 (sd 43·6) mmol/l) and counts of bifidobacteria (P < 0·05 time × diet interaction, for all). Urinary phenols and p-cresol excretion decreased (P ≤ 0·003 for time) with no difference between diets (P ≥ 0·25). Faecal form, pH, ammonia concentration and numbers of coliforms and Escherichia coli did not change with either diet. No differences between the diets were evident for incidences of adverse gastrointestinal symptoms, which suggests that both diets were well tolerated. Under energy-restricted conditions, a short-term LC diet lowered stool weight and had detrimental effects on the concentration and excretion of faecal SCFA compared with an HC diet. This suggests that the long-term consumption of an LC diet may increase the risk of development of gastrointestinal disorders.

Prebiotics may increase Zn absorption, a mineral known to play a central role in the immune system. Zn-deficient states are characterised by suppressed immune function, while prebiotics may improve both gut and cell-mediated immunity. Our objective was to determine if inulin alters the number and proportion of immune cells in the spleen, mesenteric lymph nodes (MLN) and Peyer's patches (PP), ex vivo cytokine secretion, intestinal permeability and Zn status in healthy as well as Zn-deficient rats. Weanling female rats were fed diets supplemented with 5 % cellulose (CEL) or 5 % inulin (PRE) for 4 weeks. The rats received the CEL or PRE diet ad libitum (ZN) or in restricted amounts (DR), or deficient in Zn (ZD) for another 4 weeks. The PRE-fed rats had a higher number and proportion of dendritic cells in PP, and greater ex vivo secretion of IL-2, IL-10 and interferon-γ from spleen and MLN cells compared with CEL-fed rats. PRE reduced the number and proportion of T cell receptor (TCR)-αβ+CD8+ cells in spleen and CD45RA+ cells in MLN compared with CEL. ZD rats had lower serum IgG2a and T cell numbers in MLN compared with ZN and DR rats. TCRγδ+ cell numbers in PP were higher in ZD-PRE rats compared with ZD-CEL rats. Femur Zn concentrations of DR-PRE rats were higher than those of DR-CEL rats. Intestinal permeability was unchanged. The higher proportion and number of dendritic cells in the PP of inulin-fed rats indicates a need for further research on how prebiotics and their metabolites affect immune function possibly through intestinal dendritic cells.

In humans poor maternal folate status is associated with a decrease in infant birth weight. As low birth weight increases the risk of cardiovascular and metabolic disease in adults, an inadequate supply of folic acid in the mother's diet may increase the susceptibility of the offspring to disease. We have fed laboratory rats diets deficient in folic acid and the related methyl donors methionine and choline to examine the effects on growth, blood pressure and insulin action in the offspring. Poor folate status transiently increased fetal growth but did not produce a long-term change in body weight. There were, however, small changes in the hearts of the female offspring. When folate deficiency was combined with low intakes of methionine and choline, the kidneys of the male offspring were proportionately smaller, probably because of the limited availability of methionine. There was no effect on the blood pressure of either the male or female offspring. The pancreatic insulin content of fetuses from animals fed the folate-deficient diets were higher than those of the controls. Following an oral glucose challenge, there was a weak trend for glucose-stimulated insulin release to be increased in the offspring of dams fed the folate-deficient diet. The changes in insulin concentrations were, however, much smaller than the corresponding changes observed in the offspring of animals fed protein-deficient diets. These results suggest that folate deficiency during gestation causes modest changes to the insulin axis of the fetus.

α-Linolenic acid (ALA; 18 : 3n-3) and stearidonic acid (SDA; 18 : 4n-3) are on the biosynthetic pathway of EPA (20 : 5n-3) and DHA (22 : 6n-3). The n-3 fatty acid in rapeseed oil is ALA while Echium oil contains both ALA and SDA. To determine the comparative efficacy of ALA- and SDA-rich oils in enriching broiler meat with n-3 PUFA, we offered diets supplemented with rapeseed oil (rapeseed group) or Echium oil (Echium group) for 35 d to two groups of chicks (age 21 d). There were no differences in carcass weight (2·20 (sem 0·06) v. 2·23 (sem 0·05) kg), boned, skinless thigh muscle (494 (sem 20·5) v. 507 (sem 16·7) g), boned, skinless breast muscle (553 (sem 13·4) v. 546 (sem 11·6) g) or organ weights (heart, liver and gizzard) between the two groups. The total intramuscular fat (IMF) percentage of thigh (8·0 (sem 0·64) v. 8·1 (sem 0·62) %) and breast muscles (2·3 (sem 0·24) v. 2·0 (sem 0·19) %) were also similar between the groups. In contrast, the concentrations of most of the individual n-3 fatty acids (ALA, SDA, EPA and docosapentaenoic acid) were all higher in the Echium than the rapeseed group (P < 0·05). However, differences in DHA concentrations were significant in breast but not thigh muscle IMF. The total n-3 yields/100 g serve thigh muscle were 265 and 676 mg for the rapeseed and Echium groups, respectively (P < 0·0001). The corresponding values for equivalent breast muscles were 70 and 137 mg, respectively (P < 0·01). We conclude that Echium oil is a better lipid supplement than rapeseed oil in changing the concentration and yield of n-3 fatty acids, except DHA, in broiler meat.

We tested the hypothesis that 30 min of brisk walking daily would offset the increase in plasma TAG concentrations associated with substituting dietary fat for carbohydrate. Fourteen subjects (six males) aged 57 (sd 5) years underwent three 4 d trials in a counterbalanced order: (i) 4 d on a typical UK diet (40 % energy from fat, 45 % carbohydrate and 15 % protein); (ii) 4 d on an isoenergetic diet but substituting fat for carbohydrate in line with the present recommendations (30 % fat, 55 % carbohydrate and 15 % protein); (iii) 4 d on the same recommended diet with 30 min of brisk walking each day. The food provided for the first 3 d of each trial was isoenergetic with each volunteer's previously determined daily energy intake. On day 4, the subjects consumed breakfast, lunch and an early evening meal, equivalent in total to 90 % of their daily energy intake. Blood samples were collected and substrate utilisation and energy expenditure were determined in the fasted state and for 9 h postprandially. Substrate utilisation differed significantly among trials (P = 0·003); RER was higher on the recommended diet trial than during either of the other two trials (P = 0·012 and 0·021 for the UK diet and recommended diet with walking, respectively). The rise in plasma TAG concentrations over the day was steeper on the recommended diet trial than on the other two trials (trial × time interaction, P = 0·040). The increase in postprandial TAG concentrations associated with substituting dietary fat for carbohydrate was offset by 30 min of brisk walking daily.

Associations between birth weight and CVD in adult life are supported by experiments showing that undernutrition in fetal life programmes blood pressure. In rats, the feeding of a maternal low-protein (MLP) diet during gestation programmes hypertension. The present study aimed to assess the potential for a nutritional insult to impact across several generations. Pregnant female Wistar (F0) rats were fed a control (CON; n 10) or MLP (n 10) diet throughout gestation. At delivery all animals were fed a standard laboratory chow diet. At 10 weeks of age, F1 generation offspring were mated to produce a second generation (F2) without any further dietary change. The same procedure produced an F3 generation. Blood pressure in all generations was determined at 4, 6 and 8 weeks of age and nephron number was determined at 10 weeks of age. F1 generation MLP-exposed offspring exhibited raised (P < 0·001) systolic blood pressure (male 143 (sem 4) mmHg; female 141 (sem 4) mmHg) compared with CON animals (male 132 (sem 3) mmHg; female 134 (sem 4) mmHg). Raised blood pressure and reduced nephron number was also noted in the F2 generation (P < 0·001) and this intergenerational transmission occurred via both the maternal and paternal lines, as all three possible offspring crosses (MLP × CON, CON × MLP and MLP × MLP) were hypertensive (132 (sem 3) mmHg) compared with CON animals (CON × CON; 123 (sem 2) mmHg). No effect was noted in the F3 generation. It is concluded that fetal protein restriction may play a critical role in determining blood pressure and overall disease risk in a subsequent generation.

Fish consumption is associated with a lower incidence of CVD and decreases in risk factors for atherosclerosis. Although fish contains other interesting components than fish oil, few studies focus on total fish composition and the influence food preparation might have on health-beneficial components. In the present cross-over intervention study the effect of a 6-week herring diet compared with a reference diet on CVD risk factors was investigated. Thirty-five healthy, but overweight, men (mean BMI 28·3 kg/m2) were randomised to a 6-week herring diet (150 g baked herring fillets/d, 5 d/week) or a reference diet (150 g baked lean pork and chicken fillets/d, 5 d/week). Diets were switched after a 12-week washout period. Plasma total cholesterol, TAG, HDL, HDL2, HDL3, LDL, C-reactive protein, IL-6, IL-18, intercellular adhesion molecule-1, oxidised LDL, oxygen radical absorbance capacity using perchloric acid (ORACPCA), whole-blood fatty acids, bleeding time and blood pressure were measured at the beginning and end of each dietary period. HDL was significantly higher after the herring diet period compared with after the reference diet period: 1·04 v. 0·99 mmol/l. TAG decreased after both diets, with no significant difference between the two diets. ORACPCA values did not indicate lower concentrations of non-protein plasma antioxidants, and oxidised LDL was not higher after the herring diet than after the reference diet. To conclude, a 6-week herring-rich diet significantly raised HDL compared with a diet of matched lean pork and chicken dishes. No adverse effects on in vivo oxidation or serum antioxidants were found after herring intake.
