Information from a field perspective on temperature thresholds related to physical dormancy (PY) alleviation and seed resistance to high temperatures of fire is crucial to disentangle fire- and non-fire-related germination cues. We investigated seed germination and survival of four leguminous species from a frequently burned open Neotropical savanna in Central Brazil. Three field experiments were conducted according to seed location in/on the soil: (1) fire effects on exposed seeds; (2) fire effects on buried seeds; and (3) effects of temperature fluctuations on exposed seeds in gaps and shaded microsites in vegetation. After field treatments, seeds were tested for germination in the laboratory, together with the control (non-treated seeds). Fire effects on exposed seeds decreased viability in all species. However, germination of buried Mimosa leiocephala seeds was enhanced by fire in an increased fuel load treatment, in which we doubled the amount of above-ground biomass. Germination of two species (M. leiocephala and Harpalyce brasiliana) was enhanced with temperature fluctuation in gaps, but this condition also decreased seed viability. Our main conclusions are: (1) most seeds died when exposed directly to fire; (2) PY could be alleviated during hotter fires when seeds were buried in the soil; and (3) daily temperature fluctuations in gaps also broke PY of seeds on the soil surface, so many seeds could be recruited or die before being incorporated into the soil seed banks. Thus seed dormancy-break and germination of legumes from Cerrado open savannas seem to be driven by both fire and temperature fluctuations.

Solidago shortii is endemic to a small area in northcentral Kentucky (USA), whereas two of its sympatric congeners, S. altissima and S. nemoralis, are geographically widespread. Seeds (achenes) of S. shortii (0.370 mg) are significantly larger (PLSD, P=0.05) than those of S. altissima (0.070 mg) and S. nemoralis (0.068 mg). Germination percentages of freshly-matured seeds of the three Solidago species collected in November 1991, 1992 and 1994 were 0–2% in light at 15/6°C, 1–37% at 20/10°C, 9–56% at 25/15°C and 10–85% at 30/15 and 35/20°C. Stratification increased the percentage and rate of germination and decreased the time to the onset of germination (measured by Timson's index only at 20/10°C in light) in the three species. Following 12 weeks of cold stratification in light, seeds of the three species germinated to 72–100% in the light and to 22–100% in darkness over the range of thermoperiods; those cold-stratified in darkness germinated to 39–100% in light. Freshly-matured seeds of S. altissima and of S. nemoralis germinated to 0–4% in darkness, whereas those cold-stratified for 12 weeks in darkness germinated to 0–28% in darkness. On the other hand, freshly-matured and cold-stratified (in darkness) seeds of S. shortii germinated to 0–13 and 13–73%, respectively, in darkness. Under near-natural temperatures in a glasshouse without temperature control, germination of the three species peaked in March. Thus, the primary difference in dormancy-breaking and germination requirements of the three species is that the endemic germinates to a much higher percentage in darkness than its two congeners. Seeds of S. shortii do not have any special dormancy-breaking or germination requirements that could not be fulfilled outside its present-day geographic range.

The stimulatory role of 3-methyl-2H-furo[2,3-c]pyran-2-one, a smoke-derived butenolide, on germination and post-germination events is well documented. Previous studies have involved germinating seeds in the continuous presence of the compound. However, commercial growers cannot exploit the potential of the butenolide for large-scale production of crops due to limited availability and environmental constraints. The present study was undertaken to assess the potential of the butenolide as a priming agent of tomato (Solanum esculentum Mill.) seeds. Flow cytometry data revealed that butenolide-primed seeds had a higher percentage of nuclei at the 4C stage than water-primed seeds. Emergence of the radicle was much faster in the primed seeds. After 36 h of imbibition, a higher percentage of the butenolide-primed seeds (22%) exhibited radicle emergence compared to the water-primed seeds (12%). While butenolide-primed seeds initially germinated more rapidly than either water-primed or unprimed seeds in a 48-h period, water-imbibed seeds reached a similar germination level as the butenolide-primed seeds by 60 h of incubation. The butenolide-primed seeds produced significantly (P ≤ 0.05) more vigorous seedlings than water-primed seeds or seeds in the continuous presence of either water or butenolide. A gradual decrease in the seedling vigour index was recorded for both water and butenolide-primed seeds with increased seed storage at room temperature. Nevertheless, the vigour index was significantly greater in the butenolide-primed seeds than the water-primed seeds. Vigour indices were significantly (P ≤ 0.05) higher for the butenolide-primed seeds under various stress conditions (salinity, osmoticum or temperature) compared to control or water-primed seeds. Results of the present study suggest that the butenolide is a good seed-priming agent. Additionally, primed seeds retained the promotive effect for a considerable time. This was also the case for tomato seeds under various simulated field stress conditions.

The effects of temperature, oxygen availability and salinity on seed germination in a population of Zostera marina L. from the Fleet, a tidal lagoon in southern England, were investigated in the laboratory. The fate of seeds under natural conditions was also monitored throughout the year from core samples and from seed samples buried in the sediment layer in nylon sachets. Supported with temperature, salinity and oxygen level measurements at the study site, field data confirmed that following seed dispersal in September/October, the majority of seeds germinated in the Fleet during the winter and early spring. In the laboratory, seeds germinated more rapidly and to a higher final percentage under anaerobic conditions compared to aerobic conditions. Under anaerobic conditions, rapid, near complete germination was possible at full salinity (30 g l-1) or when the salinity was reduced by 50%(15 g l-1). Under aerobic conditions, seeds germinated more rapidly at lower salinity. The effects of temperature were strongly dependent on the presence or absence of oxygen. Under anaerobic conditions, there was a pronounced optimum constant temperature for germination around 6°C, corresponding closely to the average water temperature recorded during the period mid-October to the end of March. On the other hand, under aerobic conditions, very low levels of germination were recorded at low temperatures. These findings support previously published evidence that seeds of Z. marina are well adapted to germinate in anaerobic sediments and that water temperature is the key environmental variable that determines the timing of germination under natural conditions. Previous misuse of dormancy terms to describe seed populations of Z. marina is discussed.

On the premise that seed ageing may be largely a result of free-radical lipid autoxidation, a study was made of the relationship between lipid stability and longevity in seeds of soybean (Glycine max), lentil (Lens culinaris), mungbean (Vigna radiata), chickpea (Cicer arietinum), broadbean (Vicia faba), pea (Pisum sativum), lettuce (Lactuca sativa), and tomato (Lycopersicon esculentum). Seed lipids were examined for α-, γ- and δ-tocopherols, and levels of unsaturated and saturated fatty acids. Using this information, analysis was attempted using linear, multiplicative or exponential models to correlate aspects of lipid stability with seed longevity values from the published literature. No statistically significant correlations could be found between longevity and total lipid unsaturation, tocopherol levels or two protection formulae obtained from the oil chemistry literature. When values for tomato were excluded, a good correlation (r = 0.89, P = 0.007) was obtained using a multiplicative regression model for levels of linolenic acid per unit of total tocopherols in relation to longevity. Possible factors contributing to a lessening of the relationship between lipid stability and seed longevity are discussed.

Mature and immature axes of Theobroma cacao (cocoa) seeds tolerated desiccation under a rapid-drying regime to critical water contents of 1.0 and 1.7 g g-1 dw, respectively. These critical water contents corresponded to water contents below which activities of free radical-scavenging enzymes (ascorbate peroxidase, peroxidase and superoxide dismutase) decreased rapidly during desiccation. The decline in axis viability below the critical water content was correlated with sharp increases in lipid peroxidation and cellular leakage. Cotyledon tissues were more desiccation-tolerant than axes, with a low critical water content of 0.24 g g–1dw. Desiccation sensitivity in cotyledon tissues was also correlated with the decrease in superoxide dismutase activity and increased lipid peroxidation products. However, in the cotyledons, no ascorbate peroxidase activity was detected at any water content, and peroxidase activity was gradually reduced as desiccation proceeded. Cocoa embryonic axes contained large amounts of sucrose, raffinose and stachyose but only traces of reducing monosaccharides. Desiccation sensitivity of recalcitrant cocoa axes did not appear to be due to the lack of sugar-related protective mechanisms during desiccation, and it was more likely related to the decrease of enzymic protection against desiccation-induced oxidative stresses.

The native tobacco, Nicotiana attenuata, synchronizes its germination with the immediate post-fire environment with a combination of germination stimulants found in wood smoke and inhibitors from the unburned litter of the dominant vegetation. The inhibitors override the stimulants and prevent seeds from germinating maladaptively in unburned habitats adjacent to burns. To understand the physiological basis of this environmental control of germination, we tested several previously isolated signals, phytohormones and their respective biosynthesis inhibitors. The germination inhibitors methyl jasmonate (MeJA, a constituent of sagebrush litter), bornane-2,5-dione (BD, a constituent of juniper litter extract, JLE) and JLE did not alter abscisic acid (ABA) content of imbibed seeds. Treatment with the ABA biosynthesis inhibitor, fluridone, inhibited the dormancy-inducing effects of BD, JLE and MeJA, but surprisingly did not affect endogenous ABA levels in treated seeds. However, ABA leached from litter of the species, which dominate the plant community before fires, plays an important role in germination control. We conclude that N. attenuata seeds, which can lie dormant in the soil for 150 years between fires, time their germination with the post-fire environment by responding to smoke, ABA and four terpenes (BD, 1,8-cineole, β-thujaplicin and camphor) leaching from the litter of the dominant vegetation.

Cycling of physically dormant (PY) seeds between states insensitive and sensitive to dormancy-breaking factors in the environment has recently been demonstrated in Fabaceae and Convolvulaceae, and it may be a common phenomenon in seeds with water-impermeable seed coats. In contrast to seeds of many species with physiological dormancy (PD), those with PY cannot cycle between dormancy and non-dormancy (ND). In this paper, we evaluate the role of sensitivity cycling in controlling the timing of germination of seeds with PY in nature, and show that sensitivity cycling in seeds with PY serves the same ecological role as dormancy cycling in seeds with PD. Thus, sensitivity cycling in seeds with PY ensures that germination in nature occurs only at (a) time(s) of the year when environmental conditions for growth are, and are likely to remain, suitable long enough for the plant to complete its life cycle or to form a perennating structure. Further, we describe the experimental procedures necessary to determine whether sensitivity cycling is occurring, and discuss briefly the possible relevance of sensitivity cycling to dormancy classification.

Understanding how plant traits interact with climate to determine plant niches is decisive for predicting climate change impacts. While lifespan and seed size modify the importance of germination timing, germination traits such as base temperature and base water potential directly translate climatic conditions into germination timing, impacting performance in later life stages. Yet we do not know how base temperature, base water potential, seed mass, lifespan and climate are related. We tested the relationships between base temperature and base water potential for germination, seed size and lifespan while controlling for bioclimatic regions. We also quantified the phylogenetic signal in germination traits and seed size using Pagel's λ. We used a worldwide data set of germination responses to temperature and moisture, seed size and lifespan of 240 seed plants from 49 families. Both germination temperature and moisture are negatively related to seed size. Annual plants show a negative relation between seed size and base water potential, whereas perennials display a negative relation between base temperature and seed mass. Pagel's λ highlighted the slow evolution of base temperature for germination, comparable to seed mass while base water potential was revealed to be labile. In the future, base water potential and seed mass can be used when moisture niches of plants are to be predicted. Lifespan, seed size and base temperature should be taken into account when analysing thermal limits of species distributions.

A broad variability for seed desiccation sensitivity, as quantified by the water content and the water activity at which half of the initial viability is lost, has been previously observed within nine African coffee species. In order to investigate if these different degrees of desiccation sensitivity correspond to an adaptive trait, additional data, such as the duration of seed development and seed water content at maturity, were measured for these species, and the relationships between these parameters and some climatic characteristics of their specific native environments were investigated. Since flowering in all coffee species occurs only a few days after the main rainfall marking the end of the dry season, simulations could be made, based on the continuous sequences of rainfall data compiled in databases of nine climatic stations, chosen for their appropriate location in the collecting areas. The simulations revealed a highly significant correlation between the duration of seed development and that of the wet season. Consequently, mature seeds are shed at the beginning of the following dry season. Moreover, the mean number of dry months that seeds have to withstand after shedding was significantly correlated with the parameters used to quantify seed desiccation sensitivity. By contrast, seed moisture content at maturity was not correlated with the level of seed desiccation tolerance. All these results are discussed on the basis of more detailed descriptions of the natural habitats of the coffee species studied.

Seed dormancy was analysed in Geraniaceae, testing 29,103 seeds of 35 species for seed-coat impermeability to water. Seeds of wild and cultivated species from a number of different climatic/ecological zones (northern Germany, the Canary Islands and South Africa) were investigated. Seeds from cultivated plants and natural populations had similar (±2%) percentages of water-impermeable seeds. The percentages of impermeable seeds at maturity were also mostly constant (±2%) for two different years. The species analysed contained from 0 to 100% impermeable seeds at maturity, and could be classified into three quantitative types of physical dormancy: four species with no impermeable seeds (PY0), 10 species with a maximum of 80% impermeable seeds (PY80), and 21 species with more than 80% impermeable seeds (PY100). Species with different life cycles and from very different habitats, e.g. Geranium pratense or Pelargonium capitatum, belonged to the PY100 group. However, all of the succulent shrubs investigated from the genus Pelargonium were PY0 species. Five years of dry storage at 20 ± 5°C decreased physical dormancy in a species-dependent fashion. Erodium cicutarium, for example, had 95% impermeable seeds at maturity; all of them became permeable during the dry storage and began to germinate immediately after placing them on a moist substrate. Physical dormancy of other species, e.g. Pelargonium zonale, Pelargonium vitifolium or Geranium pratense, was not affected by dry storage. Thus, physical dormancy is a diversely differentiated feature in Geraniaceae with regard to both percentages of impermeable seeds at maturity and maintenance of dormancy under particular conditions.

The effect of two priming treatments in PEG solutions on nuclear replication stages of pepper (Capsicum annuum L.) embryos were evaluated using flow cytometry. Priming at −1.1 MPa for 10 d induced almost 40% of nuclei in the embryo root tips to enter the synthetic phase of nuclear division, while no DNA synthesis occurred during 6 d at−1.5 MPa and in less than 10% of the nuclei at 10 d. Both priming treatments effectively increased seed performance. A decrease of 2.7 d in mean germination time (MGT) was observed after priming in PEG solution at −1.1 MPa, while the treatment at−1.5 MPa lowered the MGT by 2.1 d. Primed and untreated seeds were subjected to controlled deterioration treatments by exposure to 45°C after equilibration at 75% RH. Both osmotic treatments considerably lowered seed tolerance to adverse storage conditions as compared with untreated seeds. However, seeds in which DNA replication was induced by priming were more sensitive to controlled deterioration than seeds in which priming did not induce nuclei to enter the synthetic phase. This could be a consequence of a higher DNA content, which is a more vulnerable target for mutation inducing factors. Alternatively the increased sensitivity could be a consequence of their more advanced progress in germinative events, making seeds less resistant to deteriorative factors imposed during storage. On the whole, from the present results it appears that the nucleic acid DNA content of the embryo only plays an additive role in influencing seed storability.

Structural studies in seeds with physical dormancy (PY) are important to better understand its causes and release when subjected to treatments for dormancy breaking. The aims of this study were to (1) characterize the PY break; (2) examine the role of different seed structures in water uptake; and (3) identify the water gap in Senna multijuga seeds. Imbibition patterns of dormant and non-dormant (subjected to dormancy breaking treatments) seeds and the morphological changes during dormancy breaking and germination were evaluated. To identify the water gap, the micropyle and lens were blocked separately, and the water absorption by seed parts was determined. Structural characteristics of the seed coat were also examined. Immersion in water at 80°C was efficient in breaking seed dormancy and imbibition occurred first at the hilar region, through the lens. Water was not absorbed through the micropyle or the extra-hilar region. S. multijuga seeds have a testa with a linearly aligned micropyle, hilum and lens. The seed coat consisted of a cuticle, macrosclereids, one (hilar region) or two (extra-hilar region) layer(s) of osteosclereids and parenchyma cell layers. The lens has typical parenchyma cells underneath it and two fragile regions comprised of shorter macrosclereids. Heat treatment stimulated the lens region, resulting in the opening of fragile regions at the lens, allowing water to enter the seeds. It is concluded that short-term exposure to a hot water treatment is sufficient for the formation of a water gap in S. multijuga seeds, and only the lens acts in the imbibition process.

The influence of the storage environment on seed viability and antioxidant potential was examined for four species native to Western Australia: Acacia bivenosa DC., Anigozanthos manglesii D. Don, Banksia ashbyi E.G. Baker, and Mesomelaena tetragona (R. Br.) Benth. Seeds were stored at four water contents (at c. 5%, 11–15%, 20–23% and 50% relative humidity) at each of five temperatures (–196, –18, 5, 23 and 50°C), and seed germination and seedling vigour monitored over an 18-month period. Deterioration was apparent in all species (except A. bivenosa) stored at 50°C, with 11% RH maximizing longevity for B. ashbyi and M. tetragona seeds, and 5% or 11% RH preventing deterioration for A. manglesii seeds. Seed viability generally remained high for all species stored at 23°C or less. Notably, however, germination and seedling vigour of A. manglesii and M. tetragona seeds gradually declined when stored at –18°C, suggesting that storage at this temperature was detrimental. The antioxidant activity of lipid extracts of seeds after 18 months storage at 5, 23 and 50°C was also examined to determine whether the seed viability decline was associated with a loss of antioxidants. Antioxidant activity varied between storage treatments and was not related to seed viability.

The ability of orthodox seeds to survive long-term dry storage is a key prerequisite for ex situ seed conservation in genebanks. However, only a few credible observations of seed survival for ≥ 200 years have been reported. In this paper, seed survival is shown for three species under suboptimal storage conditions for a documented time of ≥ 203 years and carbon dated at 218–270 years. Two species that germinated are in the Fabaceae (Liparia sp. and Acacia sp.). A water-impermeable seed coat, and hence an inability to equilibrate with ambient relative humidity, may contribute to long-term survival of these species. The third species that germinated (Leucospermum sp.) does not have a water-impermeable seed coat, and long-term survival may be linked to an oxygen-impermeable barrier in the seed. These results for species from the Cape Floristic Region, South Africa, suggest adaptation for extreme longevity in seeds of species of seasonally dry, Mediterranean environments.

X-radiography is a simple, rapid and non-destructive method for analysing the morphology of embryos in dry, mature seeds of Arabidopsis thaliana. In wild type seeds, the cotyledons, hypocotyl and radicle tip can be readily distinguished. In seeds of the mutant types knolle, keule, and the double mutant keulelgnom, aberrations in embryo morphology can be visualized. X-radiography may therefore be useful in the isolation of embryo mutants from Arabidopsis seed samples.

Antioxidants protect seeds from oxidative damage during storage, supporting the maintenance of seed viability and the ability to germinate post-storage. No data on antioxidants during long-term storage are available for the seeds of the halophyte Suaeda maritima. Therefore, changes in lipid-soluble antioxidants in the tocopherol family (α-, γ-, δ-tocopherol), were investigated in seeds stored for up to 16 years at 4°C at a seed moisture content of 10–13%, as well as changes in the water-soluble antioxidant glutathione (GSH) and electrolyte leakage. Seed oil content was also measured and determined to be 22%. During the first 3 years of storage, seed viability remained high and the concentration of total tocopherol was stable. Thereafter, both seed viability and α-tocopherol concentration rapidly decreased and electrolyte leakage increased, while γ-tocopherol and δ-tocopherol concentrations did not correlate with seed viability. Although the concentrations of neither GSH nor glutathione disulphide (GSSG) alone were correlated with seed viability, the glutathione half-cell reduction potential (EGSSG/2GSH) was strongly correlated with viability throughout storage and increased before the onset of viability loss. Hence, in this species EGSSG/2GSH appeared to be an ‘early warning’ system preceding viability loss while α-tocopherol concentration changed concomitantly with viability.

Specific chitinases accumulate in seeds of many species as part of their normal developmental programme. Some chitinases can also be induced in developing and germinating seeds in response to microbial attack. All known seed isoforms belong to classes I, II, IV and VII, which are encoded by Chia genes, as well as to the more divergent class III encoded by Chib genes. The study of seed-specific chitinases has contributed significantly to current knowledge of this ubiquitous protein family, including antifungal properties, structure, specificity and catalytic mechanism. Indeed, the first plant chitinase for which a three-dimensional structure was solved had been isolated from barley seeds. Moreover, the finding that a chitinase could rescue a somatic embryo mutant was the first evidence of a non-defensive function. Several lines of evidence have substantiated the biotechnological potential of chitinases to counter plant fungal disease. The recent identification of several seed and fruit chitinases as major panallergens should be taken into account when selecting the genes to be introduced into food crops.

Scaling of water absorption and water loss by seeds on various soil surfaces was simulated using seed analogues constructed with paper pulp. Three sizes of analogue seeds (large, medium and small) were laid on three types of soil surface (coarse, medium and fine texture). To estimate the amount of water absorbed by a seed during a fixed time interval, the difference in seed weight from the start of the experiment was used. The scaling of water absorption necessary for germination was also studied using actual seeds of 14 species representing a range of seed sizes. Scaling coefficients between the amount of absorbed water by an analogue seed (net water gain) and seed mass were usually lower than 1: small seeds absorbed water more rapidly than large ones. The water loss of analogue seeds was also correlated with seed mass with a scaling coefficient lower than 1, but the amount of water loss itself was far smaller than the absorption. On the other hand, the germination of actual seeds revealed that the amount of water necessary to start germination was proportional to seed mass. Thus, smaller seeds have an advantage over larger seeds in more rapidly attaining the water content necessary for germination. Moreover, small seeds can penetrate through small cracks in the soil surface and thus enjoy a double advantage in a microsite that promotes water absorption and minimizes desiccation.

Senna multijuga and Plathymenia reticulata are tropical tree species native to the Brazilian Atlantic Forest and the Brazilian Cerrado, respectively. Seed-coat dormancy variation was evaluated within and among natural populations of these two species. Scarified and non-scarified seeds from different plants within populations were germinated at 28°C, and the percentage of germinated seeds was estimated for both species. Mean germination percentages of non-scarified seeds tended to be higher for P. reticulata populations (40 and 62%) than for S. multijuga populations (9 and 35%). After scarification, germination percentages increased significantly in both species, with all populations showing mean values above 84%. The level of seed dormancy, evaluated through the experiment with non-scarified seeds, differed significantly within and among populations of both species (P < 0.05). The values of the coefficient of genotypic determination were high for populations of both species (b = 0.85). Although this coefficient is an overestimation, since it includes non-genetic maternal effects, its high values suggest that a considerable part of the phenotypic variation in seed dormancy in S. multijuga and P. reticulata is of genetic origin. Variation in seed dormancy can be an important factor for increasing genetic diversity in populations of these species, making them able to respond to environmental changes.
