The quality of a seed (germination and vigour) is established during its development and maturation, but can be improved by post-harvest processing and pre-sowing treatments. During commercial seed production, maturity is usually estimated visually, relying on experience of the growers, but seed researchers are working to find molecular markers that can be applied easily to help in establishing optimal harvest time. One marker is cell cycle activity expressed as DNA replication in the seeds, analysed by flow cytometry. This fast and accurate method for the estimation of DNA content in plant nuclei allows detection of nuclei at different replication stages in different seed tissues and thus makes it possible to follow changes in the physiological state of a seed. DNA replication, as a late event during germination, can also be used to mark completion of germination and transition to early seedling growth. This information can be useful in the evaluation of seed quality and for following the advancement of priming. Flow cytometric analysis of ploidy can be also used as a basis for control of purity of some polyploid species seed lots.

Seeds of Medicago truncatula, a genomic model species for legumes, exhibit physiological and physical dormancy. Here, the factors influencing the germination behaviour of freshly harvested and stored seeds were investigated using several genotypes. Hardseededness is promoted when mature seeds are equilibrated at relative humidities (RH) below 75%. The release of physical dormancy during imbibition was dependent on the initial water content/RH that the seeds were dried to: the drier the seeds, the longer the imbibition time needed to break physical dormancy. The kinetics of physical dormancy release was slower than that of physiological dormancy, making it possible to uncouple both phenomena. Freshly harvested embryos without seed coverings germinated at the same speed as afterripened seeds. The depth of dormancy varied between different M. truncatula genotypes, from more to less dormant: DZA315.16>A17 (Jemalong)>R108>DZA45.5. This difference was eliminated by removing the endosperm. Collectively, these observations indicate that the endosperm is likely the main factor in the reduced germination of freshly harvested seeds. White light decreased germination speed of dormant seeds whereas it had no effect on non-dormant seeds. Recently harvested seeds were most dormant at temperatures above 17°C, whereas afterripened seeds germinated over a wider range of temperature. Fluridone could efficiently break dormancy, reinforcing the role of abscisic acid (ABA) synthesis. However, dormancy was not affected by gibberellic acid (100 μM GA3) or nitrate. The particular dormancy features unravelled here for M. truncatula, combined with the available genomic resources, make it a new, useful model for genetic and molecular studies which can complement those developed for Arabidopsis.

Since experimentation with plants began in space, a wide range of information has been gained regarding how this unique environment affects the biology of seeds. Seed biology experiments in this milieu have addressed aspects of seed storage, seed germination and metabolism, seedling orientation and seed production by flowering plants. Construction of hardware that provides a suitable growth environment in microgravity has been especially challenging because of the consequences posed by microgravity for fluid and gas distribution around the plant. Fluid shifting causes seed hydration kinetics to occur at a faster rate in microgravity than in 1 g; however, it also induces hypoxic metabolism during the seed germination process. In the absence of a detectable gravitational force, seedling roots grow according to their embryonic orientation and then initiate random walk movements. Light and oxygen gradients are the primary stimuli that orient root growth in this environment. For seed development to occur in spaceflight, well-ventilated growth chambers are necessary to support the carbohydrate supply needs of the developing embryos, and to provide the necessary humidity gradient for anthers to successfully dehisce and release pollen. The dry weight of seeds formed in space is lower than that in ground controls, and seed storage reserves are altered. Seed storage phenomena in spaceflight depend on whether or not oxygen and moisture are present – if not, viability exceeds that of seeds stored under comparable conditions on the ground. Because of the key role to be played by seeds in future advanced life support scenarios in space, more research is needed on the implications of this unique environment for seed biology.

In the range of water contents studied (0.1–0.4 g H2O g dw−1), Coffea arabica seeds were less sensitive to desiccation than C. costatifructa, C. racemosa and C. sessiliflora seeds. At 0.20 g H2O g dw−1, 53% of C. arabica seeds germinated after direct immersion in LN (rapid cooling, 200°C min−1), but none of them developed into normal seedlings. By contrast, in C. costatifructa, C. racemosa and C. sessiliflora, when seeds were dehydrated to the optimal water content (0.19, 0.28 and 0.31 g H2O g dw−1, respectively), the percentages of seeds which developed into normal seedlings after LN exposure were 26, 78 and 31% of the desiccation control, respectively. Normal seedlings could be recovered from cryopreserved C. arabica seeds only if they were desiccated to 0.20 g H2O g dw−1 and precooled slowly to −50°C prior to immersion in LN. Precooling seeds at 2°C min−1 allowed 25% of seeds to develop into normal seedlings. The thawing rate had no effect on the survival of cryopreserved C. arabica seeds. In all cryopreservation experiments, the total germination did not reflect the percentage of seeds which developed into normal seedlings. Examination of excised embryos indicated a partial explanation of this difference since only the shoot apex was destroyed in abnormal embryos, whereas the hypocotyl and radicle were normal.

Seed germination is vital for persistence in species that rely on seeds for post-disturbance regeneration. It is a high-risk phase and vulnerable to environmental parameters. Here, I assessed temperature sensitivity for germination in Banksia L.f. (Proteaceae) from south-western Australia, screening all 38 endemic obligate seeder species. A bi-directional temperature gradient plate with 49 temperature combinations (constant and fluctuating) between 5 and 40°C was used to profile germination temperature requirements and identify upper and lower temperature thresholds for germination. Using these data the impact of increasing temperatures on germination in these species was modelled under high and low greenhouse gas scenarios for 2050 and 2070. The results suggest that many Banksia species from the region have wide physiological tolerance for high germination temperatures, although a number of common, but geographically restricted species, such as B. praemorsa, B. oreophila and B. quercifolia, have more narrow temperature windows for germination than at least one of the rarer species (B. verticillata). Only B. dryandroides is expected to decline in germination in the future; however, the optimal germination timing for many species is predicted to occur later under climate warming. In conjunction with declining rainfall, this germination delay will place seedlings closer to the summer dry in this seasonal Mediterranean-climate ecosystem and thus they will be more vulnerable to desiccation. The framework developed here can be used to identify vulnerable species for monitoring of early population decline.

Changes in soluble sugars in cauliflower seeds were followed during 50 h of imbibition in relation to desiccation tolerance. Sucrose and stachyose contents decreased, and glucose and fructose accumulated. This occurred in radicles first and subsequently in hypocotyls and cotyledons. Loss of desiccation tolerance in the various seed parts coincided with an increase in glucose and fructose and the complete loss of stachyose, but sucrose content, the major sugar, was still high. Drying imbibed seeds over silica gel did not evoke resynthesis of stachyose, but did increase sucrose and decrease glucose and fructose contents. Seeds primed in solutions of 30% polyethylene glycol for 10 days showed a loss of stachyose, while sucrose remained high and glucose and fructose contents were still very low. Redrying of primed seeds did not change the sugar contents. The primed seeds were still tolerant of desiccation. We conclude that stachyose is not a prerequisite for desiccation tolerance, but that sucrose may be. We suggest that glucose and fructose may be involved in desiccation damage.

Cotton (Gossypium spp.) is an important source of edible oil and protein meals. Complex genetics and strong environmental effects hinder progress in seed quality trait breeding in this species. The use of molecular markers can improve an understanding of the genetic factors conditioning seed quality traits, and is expected to assist in selection of superior genotypes. This study was conducted to identify quantitative trail loci (QTL) associated with seed physical and nutrient traits in cotton. To achieve this objective, a population of 140 BC1S1 lines developed from a cross between ‘TM-1’ and ‘Hai7124’ was evaluated in 2003 and 2004. A linkage map consisting of 918 markers from this population was used to identify QTL using QTLNetwork-2.0 software. Eleven single QTL were identified for kernel percentage, kernel oil percentage, kernel protein percentage and seven amino acids (Asp, Ser, Gly, Ile, Leu, Phe and Arg). Phenotypic variation explained by each individual QTL ranged from 10.89 to 46.28%. Two epistatic QTL for Cys and Leu were detected, explaining 9.55 and 4.43% of the phenotypic variation. These QTL detected for seed quality traits in cotton are expected to be useful for further breeding programmes targeting development of cotton with improved nutrient quality.

We assessed inter-seasonal dynamics of seed banks, dormancy and seed germination in three endemic Chihuahuan Desert succulent species, under simulated soil warming conditions. Hexagonal open top-chambers (OTCs) were used to increase soil temperature. Seeds of Echinocactus platyacanthus (Cactaceae), Yucca filifera and Agave striata (Asparagaceae) were collected and buried within and outside OTCs. During the course of one year, at the end of each season, seed batches were exhumed to test viability and germination. Soil temperature in OTCs was higher than in control plots. Yucca filifera seeds always had high germination independently of warming treatment and season. Agave striata seeds from OTCs had higher germination than those from control plots. Agave striata exhibited low germination in fresh seeds, but high germination in spring. Seeds from this species lost viability throughout the experimental timeframe, and had no viable seeds remaining in the soil. Echinocactus platyacanthus showed high germination in fresh seeds and displayed dormancy cycling, leading to high germination in spring, low germination in summer and autumn, and high germination in winter. Germination of this species was also higher in seeds from OTCs than those from control plots. Echinocactus platyacanthus formed soil seed banks and its cycle of inter-seasonal dormancy/germination could be an efficient physiological mechanism in a climate change scenario. Under global warming projections, our results suggest that future temperatures may still fall within the three studied species’ thermal germination range. However, higher germination for A. striata and E. platyacanthus at warmer temperatures may reduce the number of seeds retained in the seed bank, and this could be interpreted as limiting their ability to spread risk over time. This is the first experimental study projecting an increase in soil temperature to assess population traits of succulent plants under a climate change scenario for American deserts.

The dormancy pattern of buried seeds of Datura ferox was studied by testing germination responses to red light (R), far-red light (FR) or darkness after exhumation at regular time intervals. The seeds were deeply dormant at the time of burial in winter. After three months (spring), exhumed seeds began to respond to an R pulse. Summer temperatures did not induce secondary dormancy. During winter of the second year, seed germination after an R pulse increased further; however, germination after an FR pulse or in darkness also increased and reached values close to R. Changes in seed germination after an R or an FR pulse and in darkness were studied in a factorial design experiment, combining different depths of burial (0.5, 5 and 10 cm), presence of vegetation (with or without vegetation canopies), and date of exhumation (4 or 7 months after burial). The seeds placed at 0.5 cm depth did not germinate in any light treatment. A very high sensitivity to light, indicated by the promotion of germination by an FR pulse compared to darkness, was exhibited only in seeds buried at 5 cm in a soil without vegetation. A typical R/FR reversible response was observed in seeds exhumed at 5 and 10 cm in almost all situations of burial; except in a soil without vegetation, at 10 cm depth, in late summer when even dark controls showed high germination. It is concluded that buried seeds of D. ferox are released from dormancy in two steps, the first step during the spring of the first year and the second step during the subsequent winter. This process is not obviously controlled by summer temperatures. The acquisition of a very high sensitivity to light depends on vegetation cover and depth of burial, and thus it is predicted to be affected by agricultural practices.

We review the literature on effects of inbreeding depression (ID) on seed germination for 743 case studies of 233 species in 64 families. For 216 case studies, we also review the relationship between mass and germination in inbred vs. outbred seeds. Inbred seeds germinated equally well as outbred seeds in 51.1% of 743 case studies, but better than outbred seeds in only 8.1%. In c. 50.5% of 216 cases, mass of inbred seeds was equal to (38.0%) or larger than (12.5%) that of outbred seeds. The magnitude of ID spans most of the − 1 to +1 range for relative performance for germination of inbred vs. outbred seeds; in contrast to what might be expected, seed germinability often is not negatively correlated with the coefficient of inbreeding (F) or positively corrected with population genetic diversity; neither heterosis nor outbreeding depression for germination is common in crosses between populations; and ID in most endemics is low and does not differ from that of widespread congeners. Our results on the effects of ID on seed mass and germination do not agree with the limited number of comparisons Darwin (1876) made on the effects of selfing vs. outcrossing on these two life-history traits. Recommendations are made on how to improve dormancy breaking and germination procedures in order to make the results of studies on ID more relevant to the natural world.

We studied dormancy and germination requirements of seeds of Acacia aroma, A. caven and A. furcatispina from a semi-arid region of central Argentina. Imbibition experiments were performed to determine the rate of water uptake in seeds. To determine optimal temperature for germination, seeds were incubated at three temperature regimes (15/5, 25/15 and 35/20°C) with a 12/12 h daily photoperiod or in total darkness. Additionally, differences in dormancy and germination in seed colour morphs of A. aroma were studied. Seeds of A. aroma and A. caven had impermeable coats, while those of A. furcatispina did not. Seeds of the three species showed the same pattern of germination. Germination percentages were significantly lower at 15/5°C than at 25/15 or 35/20°C. The germination temperature pattern found for these species is probably related to the summer seasonal nature of rainfall in the study area. In A. aroma, seeds of the two colour morphs showed a similar pattern of dormancy and germination.

Germination over 3 years was followed in 25 soil samples of 31 × 31 × 5 cm each from primary and secondary stands in Białowieża forest, Poland. The number of seedlings emerging and the number of species declined over the 3 years. In year 1 the mean densities of emerging seedlings for the primary and secondary stands were similar (2142 m−2 and 2488 m−2 respectively); in year 3 the difference was greater (248 vs 498 m−2) but not statistically significant. The trend reflected the greater longevity of the seed bank species characteristic of the secondary forest. Two functional groups of herb and shrub species were distinguished in the seed bank based on (a) frequency as adults in the present vegetation, and (b) relative abundance in the seed banks of primary and secondary forest. Group A species need large-scale gaps in the tree canopy; most have very small seeds (<0.15 mg mean dry mass) and are very persistent in the soil. Group B species benefit from single-treefall gaps in the tree canopy, but appear to be suited primarily to gaps in the canopy of the herb layer and in the root layer made mainly by animals; their seeds are not as small (mostly >0.15 mg) and not as persistent in the soil. Group B species made up 86% of the seedlings emerging over 3 years in soil from the primary stand, and 56% of those from the secondary stand. In the laboratory all five Group B species tested showed some inhibition of germination in darkness, but only one showed unequivocal inhibition by far-red-enrichment of low irradiance light.

The effects of alternating temperatures on seed dormancy changes, germination and seedling emergence were investigated in ‘Arctic’ and ‘Lineta’ orchardgrass (Dactylis glomerata L.). Thermal time models were successfully developed for 0, 5, 10 and 15°C temperature amplitudes, using 28 constant and alternating temperature regimes. These models were then modified by linking seed germination in Petri dishes and seedling emergence in soil. A field experiment was conducted with four seeding dates over 2 years to validate the modified thermal time models. Temperature regimes with a 5–15°C amplitude enhanced seed germination percentages of orchardgrass, indicating that the conditional dormancy was released by these temperature regimes. Base temperatures decreased with increasing temperature amplitude. Seeds germinated more rapidly under alternating temperatures than under constant temperatures. The dual effects of temperature for dormancy breaking and germination were accounted for by thermal time models based on alternating temperature regimes, which accurately predicted the timing and percentage of ‘Arctic’ and ‘Lineta’ orchardgrass seedlings emerging in the field (R2≥0.88).

Proso millet (Panicum miliaceum L.) is both a crop and a weed in many parts of the world. The weedy biotypes exhibit a wide range of seed colours, but the strains with the most persistent seeds are distinguished by darker seeds. This paper compares the seed biology of a range of biotypes from Canada and demonstrates that darker seeds have heavier seed coats, imbibe and germinate more slowly, and suffer less imbibition damage (measured as electrolyte leakage). It is concluded that all these attributes contribute to the increased persistence in the soil of the dark-seeded weedy biotypes. Imbibition damage is widely implicated in poor emergence and low vigour of crop seeds, but has not previously been considered in the context of weed seed persistence.

Controlled aging experiments were carried out in order to evaluate the changes in seed longevity occurring following the attainment of maximum seed dry weight in foxglove (Digitalis purpurea L.). Seeds harvested in three successive years were stored at ~5% moisture content (fresh weight basis) and 50°C. Seed longevity was greater the later the seeds were harvested. When the data were modelled using the predictive viability model of Ellis and Roberts, these increases were attributable to increases in both the theoretical initial viability (in probits) of the seed-lot, Ki, and in the standard deviation of the normal distribution of seed deaths in time, σ. Furthermore, there was a positive relationship between Ki and σ which was independent of the year of harvest (r = 0.9016, for 11 d.f., P < 0.001). These results do not support the predictive model which relies on the assumption that σ will not differ between seed lots of the same species stored under identical conditions. When seeds were stored at a range of moisture contents (between 5 and 10%) at 50°C, increases in σ during seed development were manifest as changes in the negative logarithmic relationship between σ and moisture content; below an upper limit, the regressions for seed lots at different stages of maturity were significantly different (P < 0.05) but could be constrained to either a common intercept or to a common slope (P < 0.05). It is proposed that it was the inherent variation in individual seed lifespans which increased during seed development. These results raise important concerns regarding the use of predetermined ‘species constants’ to predict the longevity of seed lots during long-term storage in seed banks.

Direct sowing is the simplest method of plant establishment for restoration and remediation purposes, but relatively few plants can establish under high salinity conditions. In this study, the ability of different seashore plants and grass cultivars to germinate in different dilutions of seawater (0–400 mM NaCl) was tested. Highest germination was found in distilled water or seawater dilutions up to 100 mM NaCl. When seawater concentrations were increased from 100 to 200 mM NaCl, a strong decline in germination percentage and rate was observed in less salt-tolerant species, such as Matricaria maritima and Achillea millefolium. The more salt-tolerant species, Plantago maritima, Juncus gerardii, Artemisia vulgaris, Agrostis spp. and Rumex spp., had a threshold salinity, where germination was significantly decreased in seawater dilutions between 200 and 400 mM NaCl. Even among the salt-tolerant species, only two, Agrostis stolonifera and Artemisia vulgaris, germinated at 400 mM. Variation in salinity response was observed among populations of Artemisia vulgaris and among cultivars of Festuca spp. Increasing salinity to 200 mM NaCl delayed germination in most species. Ungerminated seeds of most salinity-tolerant species were still viable after 21 d at the highest salinity (400 mM), and showed a rapid and high germination when transferred to distilled water. These species would be able to survive high salinity and germinate when the salinity of the sediments decreases through dilution or leaching of salts. The experiment revealed species and cultivars that will be of interest in further testing for restoration and remediation in saline habitats.

Oilseed crops are major sources of oils for human nutrition, and an increasing proportion is also being utilized for industrial purposes. Recent advances in our understanding of the basic biochemistry of seed oil biosynthesis, coupled with identification of genes for oilseed modification, have set the stage for the genetic engineering of oilseed crops that produce ‘designer’ plant seed oils tailored for specific applications. In this review we provide an overview of seed oil biosynthesis and highlight the enzymatic steps that have already been targeted for genetic manipulation, with the end goal of producing seed oils containing desired amounts of fatty acid components. Furthermore, we describe the identification of genes from various wild plant species that are capable of producing structurally diverse fatty acids, and how these advances open the door to the production of entirely novel oils in conventional oilseed crops. Transgenic oilseeds producing high amounts of these novel fatty acids represent renewable sources of raw materials that may compete with, and eventually replace, some petrochemicals that are derived from non-renewable crude oil.

The seed storage behaviour of Fagus sylvatica and F. crenata was investigated. A large fraction of seeds of both species survived desiccation to about 3% moisture content (MC) (in equilibrium with 10% relative humidity at 20°C). Nevertheless, viability was reduced significantly and progressively by desiccation from 14% to 3% MC. In addition, during subsequent hermetic storage at constant temperatures of 20 to –20°C in F. sylvaticaand 10 to –20°C in F. crenata seeds, viability was lost more rapidly with reduction in MC below about 7.6–11.5% (40–71%relative humidity at 20°C). Thus, Fagus sylvatica and F. crenataexhibited intermediate seed storage behaviour. Survival at –20°C with 7.8–11.5% (F. sylvatica) and 7.6% MC (F. crenata) was comparatively good, with 64–84% of seeds remaining able to germinate normally after 2 years of hermetic storage, although this was neither appreciably better nor worse than at 0–10°C. Optimum seed storage environments, within the range investigated, were provided by combining temperatures of –10 to –20°C with 7.8–11.5% (F. sylvatica) or 7.6–9.5%(F. crenata) MC.

Early plant establishment through seed germination and seedling emergence is a crucial process that determines seedling number, emergence time distribution and the early growth of seedlings, all of which are affected by soil climate and soil structure. In the current context of climate change, in which increasing the diversity of cultivated species is considered desirable, and new tillage practices are considerably modifying top-soil surface characteristics, we need to improve our ability to model the effects of the environment on plant establishment. Using a trait-based and model-based framework, we aimed to identify general relationships between seed and seedling traits (e.g. seed mass and lipid content, seedling diameter, base temperature) and germination and emergence model parameters (e.g. time to mid-germination, shoot elongation rate) measured for 18 genotypes belonging to 14 species. Relationships were also investigated among model parameters or traits. Germination rates were faster for species with a high base temperature and for species with seed reserves located principally in the embryo (rather than the endosperm or perisperm). During heterotrophic growth, maximal shoot length and elongation rate increased with seed dry mass. The sensitivity of seedlings to soil obstacles was negatively related to shoot diameter. Thus apart from the known effects of seed mass on seedling establishment, we found that seed reserve location, seedling shoot diameter and shape affected germination rate and emergence success. Such generic rules linking plant traits to germination and emergence parameters enhance our understanding of the determinants of environmental effects on plant establishment success.
