Animal evidence indicates that green tea may modulate insulin sensitivity, with epigallocatechin-3-gallate (EGCG) proposed as a likely health-promoting component. The purpose of this study was to investigate the effect of dietary supplementation with EGCG on insulin resistance and associated metabolic risk factors in man. Overweight or obese male subjects, aged 40–65 years, were randomly assigned to take 400 mg capsules of EGCG (n 46) or the placebo lactose (n 42), twice daily for 8 weeks. Oral glucose tolerance testing and measurement of metabolic risk factors (BMI, waist circumference, percentage body fat, blood pressure, total cholesterol, LDL-cholesterol, HDL-cholesterol, TAG) was conducted pre- and post-intervention. Mood was evaluated weekly using the University of Wales Institute of Science and Technology mood adjective checklist. EGCG treatment had no effect on insulin sensitivity, insulin secretion or glucose tolerance but did reduce diastolic blood pressure (mean change: placebo − 0·058 (se 0·75) mmHg; EGCG − 2·68 (se 0·72) mmHg; P = 0·014). No significant change in the other metabolic risk factors was observed. The EGCG group also reported feeling in a more positive mood than the placebo group across the intervention period (mean score for hedonic tone: EGCG, 29·11 (se 0·44); placebo, 27·84 (se 0·46); P = 0·048). In conclusion, regular intake of EGCG had no effect on insulin resistance but did result in a modest reduction in diastolic blood pressure. This antihypertensive effect may contribute to some of the cardiovascular benefits associated with habitual green tea consumption. EGCG treatment also had a positive effect on mood. Further studies are needed to confirm the findings and investigate their mechanistic basis.

1. To obtain a quantitative model for nitrogen pathways in sheep, a study of ammonia and urea metabolism was made by using isotope dilution techniques with [15N]ammonium sulphate and [15N]urea and [14C]urea.

2. Single injection and continuous infusion techniques of isotope dilution were used for measuring ammonia and urea entry rates.

3. Sheep were given 33 g of chaffed lucerne hay every hour; the mean dietary N intake was 23.4 g/d.

4. It was estimated that 59% of the dietary N was digested in the reticulo-rumen; 29% of the digested N was utilized as amino acids by the micro-organisms, and 71% was degraded to ammonia.

5. Of the 14.2 g N/d entering the ruminal ammonia pool, 9.9 g N/d left and did not return to the pool, the difference of 4.3 g N/d represented recycling, largely within the rumen itself (through the pathways: ruminal ammonia → microbial protein → amino acids → ammonia).

6. Urea was synthesized in the body at a rate of 18.4 g N/d from 2.0 g N/d of ammonia absorbed through the rumen wall and 16.4 g N/d apparently arising from deamination of amino acids and ammonia absorbed from the lower digestive tract.

7. In the 24 h after intraruminal injection of [15N]ammonium salt, 40–50% of the N entering the plasma urea pool arose from ruminal ammonia; 26% of the 15N injected was excreted in urinary N.

8. Although 5.1g N/d as urea was degraded apparently in the digestive tract, only 1.2g N/d appeared in ruminal ammonia; it is suggested that the remainder may have been degraded in the lower digestive tract.

9. A large proportion of the urea N entering the digestive tract is apparently degraded and absorbed and the ammonia incorporated in the pools of nitrogenous compounds that turn over only slowly. This may be a mechanism for the continuous supply to the liver of ammonia for these syntheses.

10. There was incorporation of 15N into bacterial fractions isolated from rumen contents after intraruminal and intravenous administration of [15N]ammonium salts and [15N]urea respectively.

11. A model for N pathways in sheep is proposed and, for this diet, many of the pool sizes and turn-over rates have been either deduced or estimated directly.

1. A study has been made of the zinc, copper, iron, manganese, protein (nitrogen x 625) and phytic acid contents of nineteen soya-bean-based textured-vegetable-protein (TVP) meat-extenders and meat-substitutes and of three ‘ready-prepared’ canned meals containing TVP.

2. Phytate analysis was performed using a newly-developed method based on Holt's (1955)procedure. This method enabled the phytate content of milligram quantities of TVP to be estimated, with an SD for six replicates of 3%.

3. The Fe, Cu and Mn contents (mg/kg) of the meat-extenders or meat-substitutes varied, wzith values of 59.4–144, 14.1–19.7 and 19.5–29.1 respectively. The protein content of these products was approximately 500 g/kg.

4. The phytate content of the meat-extenders and meat-substitutes ranged from 11.0to 20.2 g/kg and the Zn content from 35.0 to 49.4 mg Zn/kg. The calculated molar ratio, phytate:Zn varied from 25 to 42.

5. The trace element, phytate and protein contents of the ‘ready-prepared’ canned meals were 30–50 %: lower than the meat-extenders and meat-substitutes.

6. Cooking the ‘ready-prepared’ meals as specified by the manufacturers was without effect on the trace element or phytate content.

7. When TVP was fed to rats as the only protein source, they had significantly lower growth rates and plasma Zn concentrations than rats given an egg-albumen-based diet of similar Zn content (14.5mg Zn/kg). Supplementation of the TVP diet with Zn (100 mg Zn/kg) significantly increased growth rate and plasma Zn concentration whereas Zn supplementation of the albumen diet was without effect.

8. The possible implications of consumption of TVP products in relation to Zn status of the human population is discussed.

The health benefits currently associated with increased dairy intake may be attributable to the whey component of dairy proteins. The present study evaluated the effects of whey protein supplementation on body composition, lipids, insulin and glucose in comparison to casein and glucose (control) supplementation in overweight/obese individuals for 12 weeks. The subjects were randomised to whey protein, casein or glucose supplementation for 12 weeks according to a parallel design. Fasting blood samples and dual-energy X-ray absorptiometry measurements were taken. Seventy men and women with a mean age of 48·4 (sem 0·86) years and a mean BMI of 31·3 (sem 0·8) kg/m2 completed the study. Subjects supplemented with whey protein had no significant change in body composition or serum glucose at 12 weeks compared with the control or casein group. Fasting TAG levels were significantly lowered in the whey group compared with the control group at 6 weeks (P = 0·025) and 12 weeks (P = 0·035). There was a significant decrease in total cholesterol and LDL cholesterol at week 12 in the whey group compared with the casein (P = 0·026 and 0·045, respectively) and control groups (P < 0·001 and 0·003, respectively). Fasting insulin levels and homeostasis model assessment of insulin resistance scores were also significantly decreased in the whey group compared with the control group (P = 0·049 and P = 0·034, respectively). The present study demonstrated that supplementation with whey proteins improves fasting lipids and insulin levels in overweight and obese individuals.

The iodine intake level in a population is determined in cross-sectional studies. Urinary iodine varies considerably and the reliability of studies of iodine nutrition and the number of samples needed is unsettled. We performed a longitudinal study of sixteen healthy men living in an area of mild to moderate iodine deficiency. Iodine and creatinine concentrations were measured in spot urine samples collected monthly for 13 months. From these data we calculated the number of urine samples needed to determine the iodine excretion level for crude urinary iodine and for 24 h iodine excretion estimated from age- and gender-specific creatinine excretions. We found that mean urinary iodine excretion varied from 30 to 87 μg/l (31 to 91 μg/24 h). Sample iodine varied from 10 to 260 μg/l (20 to 161 μg/24 h). Crude urinary iodine varied more than estimated 24 h iodine excretion (population standard deviation 32 v. 26; individual standard deviation 29 v. 21; Bartlett's test, P < 0·01 for both). The number of spot urine samples needed to estimate the iodine level in a population with 95 % confidence within a precision range of ± 10 % was about 125 (100 when using estimated 24 h iodine excretions), and within a precision range of ± 5 % was about 500 (400). A precision range of ± 20 % in an individual required twelve urine samples or more (seven when using estimated 24 h iodine excretions). In conclusion, estimating population iodine excretion requires 100–500 spot urine samples for each group or subgroup. Less than ten urine samples in an individual may be misleading.

The present work was undertaken to study the gastrointestinal effects of wheat and oat dietary fibre (DF) using 40–50 kg pigs cannulated in the terminal ileum. The variables studied were: chemical characteristics of the DF, ileal and faecal digestibility of nutrients and bulking properties of polysaccharides and other major constituents. The wheat products studied included refined wheat flour and wheat fractions rich in the following botanical components: aleurone, pericarp/testa and bran. The oat products used were rolled oats and oat bran. The products varied considerably in DF content (g/kg dry matter) and composition; non-starch polysaccharides (NSP) and Klason lignin content ranged from 34 and 1 g/kg respectively in wheat flour, to 465 and 92 g/kg in pericarp/testa. The main NSPs in the wheat were arabinoxylans (AX) (64–69%) and cellulose (15–31%) and in oats mixed linked β(1 → 3; 1 → 4)-D-glucans (β-glucans; 46–63%) and AX (28–32%). The lowest content of soluble NSP was found in the lignified wheat fractions (bran and pericarp/testa) and the highest in oat bran. Eight diets were produced using the wheat and oat products and studied in two series of experiments using wheat flour as the DF-depleted control. The diets in Expt 1 were based on wheat flour and three iso-DF enriched diets prepared by adding DF from the fractions rich in wheat aleurone, pericarp/testa or bran. In Expt 2, oat bran was added to wheat flour to achieve the same DF intake level as in Expt 1. This series also included diets based on rolled oats and rolled oats plus oat bran. Starch was almost completely digested in the small intestine (0.97–1.00). However, there was a tendency to a slightly lower digestibility of oat starch compared with wheat starch. The recovery of wheat NSP in ileal digesta was 82–104 % compared with 64–66% for oats. The low recovery of NSP in oat diets was primarily due to the low recovery of β-glucans (25–36%). In the large intestine NSP and starch residues were extensively degraded. For the DF-depleted control diets or diets based on oats, 8–17% NSP survived breakdown while in the diets enriched with aleurone, pericarp/testa or bran fractions, NSP recovery was 33, 50 and 38 % respectively. Fermentative breakdown of carbohydrates in the large intestine was estimated to contribute between 10 and 24 % of the energy for maintenance. Energy derived from the inflow of organic acids from the ileum contributed an additional 1–4% of maintenance energy. In wheat endosperm, AX were broken down to a greater extent than cellulose, while the breakdown of AX in pericarp/testa was similar to that of cellulose. This difference in NSP breakdown can be explained by structural differences in the two types of cell walls. The breakdown of oat AX was lower than that of wheat flour. Wheat DF increased faecal bulk primarily by virtue of its physical presence and its water-holding capacity, while the oat DF stimulated faecal output through an increase in microbial biomass (Bach Knudsen et al. 1991). The result was a higher excretion of protein and fat. The higher fat excretion with the oat diets was probably due to a higher bile acid excretion caused by the more extensive fermentation of carbohydrates and the lower lumen pH.

Dietary pattern analysis, which reflects the complexity of dietary intake, has received considerable attention by nutritional epidemiology. For a long time, two general approaches have been used to define these summary variables in observational studies. The exploratory approach is based only on the data of the study, whereas the hypothesis-oriented approach constructs pattern variables based on scientific evidence available before the study. Recently, a new statistical method, reduced rank regression, was applied to nutritional epidemiology that is exploratory by nature, but can use scientific evidence by focusing on disease-related dietary components or biomarkers. Several studies, both observational and clinical, suggest that dietary patterns may predict the risk of CHD and stroke. In the present review, we describe the results of these studies and the available evidence regarding the relationships between dietary patterns and risk of CVD and we discuss limitations and strengths of the statistical methods used toextract dietary patterns.

Dietary phytochemicals, of which polyphenols form a considerable part, may affect the risk of obesity-associated chronic diseases such as type 2 diabetes. This article presents an overview on how phytochemicals, especially polyphenols in fruits, vegetables, berries, beverages and herbal medicines, may modify imbalanced lipid and glucose homeostasis thereby reducing the risk of the metabolic syndrome and type 2 diabetes complications.

Cu appears to have many important functional roles in the body that apparently relate, among others, to the maintenance of immune function, bone health and haemostasis. Some have suggested a role for long-term marginal Cu deficiency in the aetiology of a number of degenerative diseases. Accurate diagnosis of marginal Cu deficiency, however, has remained elusive despite an increased understanding of the biochemistry of Cu and its physiological roles in the body. Traditional markers of Cu status, such as serum Cu and caeruloplasmin protein concentrations are insensitive to subtle changes in Cu status. Cu-containing enzymes, such as Cu–Zn-superoxide dismutase, cytochrome c oxidase and diamine oxidase, may be more reliable but evidence to date is not conclusive. Development of markers sensitive to marginal Cu status is essential before conclusions can be drawn concerning the risks of long-term intake of suboptimal dietary Cu. As Cu appears to be essential for maintenance of immune function, activities of specific immunological markers, altered in Cu deficiency, offer alternatives. This review evaluates a selection of immunological markers that could be considered potentially sensitive markers of marginal Cu status. The indices of immune function reviewed are neutrophil function, interleukin 2 production, blastogenic response to mitogens and lymphocyte subset phenotyping.

The present study assessed the effects of mild dehydration on cognitive performance and mood of young males. A total of twenty-six men (age 20·0 (sd 0·3) years) participated in three randomised, single-blind, repeated-measures trials: exercise-induced dehydration plus a diuretic (DD; 40 mg furosemide); exercise-induced dehydration plus placebo containing no diuretic (DN); exercise while maintaining euhydration plus placebo (EU; control condition). Each trial included three 40 min treadmill walks at 5·6 km/h, 5 % grade in a 27·7°C environment. A comprehensive computerised six-task cognitive test battery, the profile of mood states questionnaire and the symptom questionnaire (headache, concentration and task difficulty) were administered during each trial. Paired t tests compared the DD and DN trials resulting in >1 % body mass loss (mean 1·59 (sd 0·42) %) with the volunteer's EU trial (0·01 (sd 0·03) %). Dehydration degraded specific aspects of cognitive performance: errors increased on visual vigilance (P = 0·048) and visual working memory response latency slowed (P = 0·021). Fatigue and tension/anxiety increased due to dehydration at rest (P = 0·040 and 0·029) and fatigue during exercise (P = 0·026). Plasma osmolality increased due to dehydration (P < 0·001) but resting gastrointestinal temperature was not altered (P = 0·238). In conclusion, mild dehydration without hyperthermia in men induced adverse changes in vigilance and working memory, and increased tension/anxiety and fatigue.

In the present study, the hypoglycaemic potentials of ginger (Zingiber officinale) were studied in rats. An aqueous extract of raw ginger was administered daily (500 mg/kg, intraperitoneally) for a period of 7 weeks to streptozotocin (STZ)-induced diabetic rats. Fasting blood serum was analysed for blood glucose, cholesterol and triacylglycerol levels. The STZ-injected rats exhibited hyperglycaemia accompanied with weight loss, indicating their diabetic condition. At a dose of 500 mg/kg, raw ginger was significantly effective in lowering serum glucose, cholesterol and triacylglycerol levels in the ginger-treated diabetic rats compared with the control diabetic rats. The ginger treatment also resulted in a significant reduction in urine protein levels. In addition, the ginger-treated diabetic rats sustained their initial weights during the treatment period. Moreover, ginger decreased both water intake and urine output in the STZ-induced diabetic rats. The present results indicate that raw ginger possesses hypoglycaemic, hypocholesterolaemic and hypolipidaemic potential. Additionally, raw ginger is effective in reversing the diabetic proteinuria observed in the diabetic rats. Thus, ginger may be of great value in managing the effects of diabetic complications in human subjects.

High intakes of fruit and vegetables may reduce the risk of cancer at several sites. Evidence has been derived mainly from case–control studies. We reviewed the relationship between consumption of vegetables and fruit and the risk of several common cancers in a network of Italian and Swiss case–control studies including over 10 000 cases of fourteen different cancers and about 17 000 controls. Data were suggestive of a protective role of vegetable intake on the risk of several common epithelial cancers. OR for the highest compared with the lowest levels of consumption ranged from 0·2 (larynx, oral cavity and pharynx) to 0·9 (prostate). Inverse associations were found for both raw and cooked vegetables, although for upper digestive tract cancers the former were somewhat stronger. Similar inverse associations were found for cruciferous vegetables. Frequent consumption of allium vegetables was also associated with reduced risk of several cancers. Fruit was a favourable correlate of the risk of several cancers, particularly of the upper digestive tract, with associations generally weaker than those reported for vegetables. A reduced risk of cancers of the digestive tract and larynx was found for high consumption of citrus fruit. Suggestive protections against several forms of cancer, mainly digestive tract cancers, were found for high consumption of apples and tomatoes. High intakes of fibres, flavonoids and proanthocyanidins were inversely related to various forms of cancer. In conclusion, data from our series of case–control studies suggested a favourable role of high intakes of fruit and vegetables in the risk of many common cancers, particularly of the digestive tract. This adds evidence to the indication that aspects of the Mediterranean diet may have a favourable impact not only on CVD, but also on several common (epithelial) cancers, particularly of the digestive tract.

Impaired nutritional status has been frequently reported in surveys estimating its prevalence amongst patients in hospital. While there is no doubt that protein–energy undernutrition has serious implications for health, recovery from illness or surgery and hospital costs, lack of nationally or internationally accepted cut-off points and guidelines for most nutrition-related variables make nutritional assessment difficult and proper comparisons between studies impossible. In reviewing published work in which the prevalence of undernutrition has been assessed, it can be seen that each study defined undernutrition, or nutritional risk, using different methodology. This present review aims to highlight the problems which arise when deciphering these studies, and the resulting difficulty in determining the true prevalence of undernutrition and nutritional risk, amongst both general and specific groups of hospital in-patients. It is widely agreed that routine hospital practices can further adversely affect the nutritional status of sick patients in hospital. How this occurs, and the potential effects of impaired nutritional status on clinical outcome are examined. The methods currently available to assess nutritional status are evaluated in the knowledge that such assessments are difficult in clinical practice. The review concludes by proposing that if we want the medical and nursing professions to consider the nutritional status of hospital patients seriously, definitions of undernutrition and nutritional risk, and cut-off values for the nutritional variables measured must be agreed to allow evidence-based practice. Outcome measures which allow clear comparisons between groups and treatments must be used in studies assessing the effects of nutritional interventions.

Epidemiological evidence suggests that polyphenols may, in part, explain the cardioprotective properties of fruits. This review aims to summarise the evidence for the effects of fruit polyphenols on four risk factors of CVD: platelet function, blood pressure, vascular function and blood lipids. This review includes human dietary intervention studies investigating fruits and their polyphenols. There was some evidence to suggest that fruits containing relatively high concentrations of flavonols, anthocyanins and procyanindins, such as pomegranate, purple grapes and berries, were effective at reducing CVD risk factors, particularly with respect to anti-hypertensive effects, inhibition of platelet aggregation and increasing endothelial-dependent vasodilation than other fruits investigated. Flavanone-rich fruits, such as oranges and grapefruits, were reported to have hypocholesterolaemic effects, with little impact on other risk factors being examined. However, the evidence was limited, inconsistent and often inconclusive. This is in part due to the heterogeneity in the design of studies, the lack of controls, the relatively short intervention periods and low power in several studies. Details of the polyphenol content of the fruits investigated were also omitted in some studies, negating comparison of data. It is recommended that large, well-powered, long-term human dietary intervention studies investigating a wider range of fruits are required to confirm these observations. Investigations into the potential synergistic effects of polyphenols on a combination of CVD risk markers, dose–response relationships and standardisation in methodology would facilitate the comparison of studies and also provide valuable information on the types of fruits which could confer protection against CVD.

Common infectious diseases (CID) of the airways and the gastrointestinal tract are still a considerable cause of morbidity and mortality in elderly. The present study examined the beneficial effect of a dairy product containing the probiotic strain Lactobacillus casei DN-114 001 (fermented product) on the resistance of free-living elderly to CID. The study was multicentric, double blind and controlled, involving 1072 volunteers (median age = 76·0 years) randomised for consumption of either 200 g/d of fermented (n 537) or control (non-fermented) dairy product (n 535) for 3 months, followed by an additional 1 month's follow-up. The results showed that, when considering all CID, the fermented product significantly reduced the average duration per episode of CID (6·5 v. 8 d in control group; P = 0·008) and the cumulative duration of CID (7 v. 8 d in control group; P = 0·009). Reduction in both episode and cumulative durations was also significant for all upper respiratory tract infections (URTI; P < 0·001) and for rhinopharyngitis (P < 0·001). This was accompanied with an increase of L. casei species in stools throughout the fermented product consumption (2–3·8 × 107 equivalents of colony-forming unit/g of stools, P < 0·001). The cumulative number of CID (primary outcome) was not different between groups nor was the CID severity, fever, pathogens' occurrence, medication, immune blood parameters and quality of life. The fermented product was safe and well tolerated. In conclusion, consumption of a fermented dairy product containing the probiotic strain L. casei DN-114 001 in elderly was associated with a decreased duration of CID in comparison with the control group, especially for URTI such as rhinopharyngitis.

Using germ-free rats inoculated with a human faecal flora (gnotobiotic rats), the effects of three oligosaccharides (β-fructo-oligosaccharides (FOS), β-galacto-oligosaccharides (TOS) and α-gluco-oligosaccharides (GOS)) on intestinal bacterial metabolism were compared. The animals were fed on either a control diet or diets containing 4Og/kg of GOS, FOS or TOS. FOS and TOS were the preferred growth substrates for Bifidobacteria which increased in number by 2 log values in faeces of rats when compared with rats fed on GOS or control diets. Ingestion of TOS specifically induced hydrolysis of the substrate, and did not modify the activity of any other enzymes measured in the caecum. GOS led to a non-specific enzymic induction of β-galactosidase (EC 3.2.1.23), β-glucosidase (EC 3.2.1.21) and α-glucosidase (EC 3.2.1.20) activities whereas β-glucuronidase (EC 3.2.1.31) was lowered. Compared with the control group, FOS and TOS diets led to a significant increase in H2 and CH4 excretion; the GOS diet increased only CH4. Analysis of caecal contents revealed a decrease in pH for all diets compared with controls. Total short-chain fatty acid (SCFA) concentration increased significantly in all groups, but the SCFA profile differed between treatment groups. It was concluded that the three oligosaccharides studied had different effects which may be linked to their chemical structure. Some of these effects may be beneficial to human health

The aim of the present study was to evaluate the importance of social status and lifestyle for dietary habits, since these factors may influence life expectancy. We studied the association of four indicators for healthy dietary habits (fruits and vegetables, fibre, fat and Hegsted score) with sex, age, socio-economic status, education, physical leisure exercise, smoking and personal attention paid to keeping a healthy diet. Data were gathered with a self-administered quantitative food-frequency questionnaire distributed to a representative sample of Norwegian men and women aged 16–79 years in a national dietary survey, of whom 3144 subjects (63%) responded. Age and female sex were positively associated with indicators for healthy dietary habits. By separate evaluation length of education, regular physical leisure exercise and degree of attention paid to keeping a healthy diet were positively associated with all four indicators for healthy dietary habits in both sexes. Socio-economic status, location of residence and smoking habits were associated with from one to three indicators for healthy dietary habits. In a multiple regression model, age, education and location of residence together explained from 1 to 9% of the variation (R2) in the four dietary indicators. Length of education was significantly associated with three of four dietary indicators both among men and women. By including the variable ‘attention paid to keeping a healthy diet’ in the model, R2 increased to between 4 and 15% for the four dietary indicators. Length of education remained correlated to three dietary indicators among women, and one indicator among men, after adjusting for attention to healthy diet, age and location of residence. Residence in cities remained correlated to two indicators among men, but none among women, after adjusting for age, education and attention to healthy diet. In conclusion, education was associated with indicators of a healthy diet. Attention to healthy diet showed the strongest and most consistent association with all four indicators for healthy dietary habits in both sexes. This suggests that personal preferences may be just as important for having a healthy diet as social status determinants.

1. Twelve grain mixtures, one lucerne (Medicago sativa) hay and one maize silage which had been used in mixed diets for which dietary nitrogen undegraded in the rumen (UDN) had been estimated with duodenally-cannulated cows, were studied. Total N in the feeds was fractionated into pool A (N soluble in borate–phosphate buffer), pool B (total N–(pool A + pool C)) and pool C (acid-detergent-insoluble N or residual N after 24 h incubation in protease solution).

2. N solubilization in protease solution containing 6·6 units/ml (substrate-saturating enzyme concentration) indicated the presence of subfractions in pool B, with different rates of solubilization. Such subfractions were not detectable from in situ, Dacron bag, estimates of N solubilization.

3. UDN was estimated using a dynamic mathematical model and rate-constants obtained from N solubilization in protease solution or in situ.For three grain mixtures tested using the protease technique the model predicted UDN values of 7, 10 and 12% compared with values of 47, 66 and 59% estimated in vivo. The full range of experimental feeds was tested using the in situtechnique and UDN values predicted by the model were used to derive UDN values for twelve mixed diets. The latter values were significantly but not closely correlated with those determined in vivo (r2 0·41, P < 0·05).

4. An attempt was made to simulate rumen proteolysis in vitro by choosing a protease enzyme concentration (0·066 units/ml) providing a proteolytic activity similar to that of whole rumen fluid. The experimental samples of feed were subjected to simulated rumen proteolysis for 18 or 48 h to resemble the mean retention times in the rumen for grain mixtures and roughages respectively. The residual N at the end of incubation was considered as an estimate of UDN. The UDN values estimated from simulated rumen proteolysis and those determined in vivo for twelve mixed diets were in close agreement (r2 0·61, P < 0·01).

5. Simulated rumen proteolysis can serve as a simple, rapid and sensitive method to estimate UDN in a variety of feedstuffs.

1. The physiological response of rainbow trout (Salmo gairdneri) reared on different levels of available carbohydrate in practical trout diets having the same levels of energy and nitrogen for 16–24 weeks was determined.

2. Weight gain was significantly reduced in trout reared on the highest level of available carbohydrate, 210 g cerelose (α-glucose)/kg, and there was a significant linear regression (R2 0.88) of dietary carbohydrate on weight gain.

3. Liver: body-weight values and liver glycogen levels increased in relation to increased dietary carbohydrate.

4. Liver glucose-6-phosphate dehydrogenase (EC 1.1.1.49) activity increased and liver phosphoenolpyruvate carboxykinase (EC 4.1.1.32) activity decreased per kg body-weight of fish with increasing dietary carbohydrate. However, no significant effect was noted on the activity of these liver enzymes above a dietary cerelose level of 140 g/kg.

5. Liver fructose diphosphatase (EC 3.1.3.11) activity increased with increasing dietary carbohydrate has been interpreted as meaning a recycling of triosephosphate to glucose-6-phosphate.

6. Dietary carbohydrate level had no significant effect on the liver pyruvate kinase (EC 2.7.1.40) activity, the rate of glucose utilization or the percentage conversion of [14C]alanine to glucose in the plasma of trout.

7. The results indicate that rainbow trout have a limited ability to adapt to increased dietary carbohydrate and a level in excess of 140 g/kg of the diet is not efficiently utilized.

The dietary inflammatory index (DII) is a new tool to assess the inflammatory potential of the diet. In the present study, we aimed to determine the association between the DII and BMI, waist circumference and waist:height ratio (WHtR). We conducted a cross-sectional study of 7236 participants recruited into the PREvención con DIeta MEDiterránea trial. Information from a validated 137-item FFQ was used to calculate energy, food and nutrient intakes. A fourteen-item dietary screener was used to assess adherence to the Mediterranean diet (MeDiet). Sex-specific multivariable linear regression models were fitted to estimate differences (and 95 % CI) in BMI, waist circumference and WHtR across the quintiles of the DII. All nutrient intakes, healthy foods and adherence to the MeDiet were higher in the quintile with the lowest DII score (more anti-inflammatory values) except for intakes of animal protein, saturated fat and monounsaturated fat. Although an inverse association between the DII and total energy was apparent, the DII was associated with higher average BMI, waist circumference and WHtR after adjusting for known risk factors. The adjusted difference in the WHtR for women and men between the highest and lowest quintiles of the DII was 1·60 % (95 % CI 0·87, 2·33) and 1·04 % (95 % CI 0·35, 1·74), respectively. Pro-inflammatory scores remained associated with obesity after controlling for the effect that adherence to a MeDiet had on inflammation. In conclusion, the present study shows a direct association between the DII and indices of obesity, and supports the hypothesis that diet may have a role in the development of obesity through inflammatory modulation mechanisms.
