Five Euphorbiaceae species (Jatropha macrocarpa, J. hieronymi, J. excisa, Cnidoscolus tubulosus and Manihot guaranitica) were analysed for seed oil content and fatty acid profiles in order to evaluate their potential as sources of biodiesel. The five species are perennial shrubs adapted to arid and semi-arid environments of north-western Argentina. The seed oil content for all species ranged from 27.9 to 38.7% by dry weight. Fatty acid composition consisted mainly of linolenic, myristic, palmitic, stearic, arachidic, oleic and linoleic acids, with linoleic and oleic acid being the most abundant. Among the five species analysed, J. macrocarpa has the best potential to be used as a biodiesel source. The average total seed oil content (35.8%) and a high value of oleic acid (27.3%) together with the lowest values of linoleic acid (55.8%) make this species a promising crop as a source of biodiesel in arid and semi-arid regions.

Narcissus longispathus (Amaryllidaceae) is a perennial geophyte and a Mediterranean narrowly endemic species. At dispersal time, N. longispathus seeds are dormant and have underdeveloped embryos. This work aimed to determine requirements for dormancy break and germination and to compare dormancy traits with those of the two endemic Iberian congeners. Phenology of embryo growth and germination were studied by regularly exhuming seeds sown in near-natural conditions. Temperature and light requirements for embryo growth, breaking of dormancy and germination were determined by incubating seeds under controlled laboratory conditions. Mean embryo length in fresh seeds was 1.50 mm, and embryos had to grow to 3.80 mm before radicle emergence. Embryos grew to full size and seeds germinated when they were warm stratified for 2 months (optimum 1 month at 20/7°C+1 month at 15/4°C), then cold stratified at 5°C for 2 months, and finally incubated at cool temperatures (15/4°C) for 30 d. However, in seeds only subjected to either warm or cold stratification, the embryos hardly grew and did not germinate. In natural conditions, the embryos elongate in autumn–winter, and in late winter–early spring (March) almost all radicles and seedlings emerged. Velocity of embryo growth and germination percentages increased with seed storage duration. Seeds of N. longispathus have non-deep complex morphophysiological dormancy (MPD). This is the first report of such a level of MPD in Narcissus. Our data suggest that non-deep complex MPD may have been derived from intermediate complex MPD in the section Pseudonarcissi.

Mangroves are highly adapted to extreme environmental conditions that occur at the interface of salt and fresh water. Adaptations to the saline environment during germination are a key to mangrove survival, and thereby, its distribution. The main objective of this research was to study the effect of salinity on seed germination of selected mangrove species and the application of a hydrotime model to explain the relationship between water potential of the medium and rate of seed germination. Germination of seeds was examined at 15, 25 and 35°C in light/dark over a NaCl gradient. Germination time courses were prepared, and germination data were used to investigate whether these species behave according to the principles of the hydrotime model. The model was fitted for the germination of Acanthus ilicifolius seeds at 25°C. Final germination percentage was significantly influenced by species, osmotic potential and their interaction at 25°C. Moreover, temperature had a clear effect on seed germination (Sonneratia caseolaris and Pemphis acidula) which interacted with osmotic potential. Only A. ilicifolius seeds behaved according to the hydrotime principles and thus its threshold water potential was –1.8 MPa. Optimum germination rates for seeds of the other species occurred at osmotic potentials other than 0 MPa. The descending order of salinity tolerance of the tested species was Aegiceras corniculatum > Sonneratia caseolaris > Acanthus ilicifolius > Pemphis acidula > Allophylus cobbe, suggesting that the viviparous species (A. corniculatum) is highly salt tolerant compared with the non-viviparous species. The results revealed that seeds of the study species exhibited facultative halophytic behaviour in which they can germinate over a broad range of saline environments. Use of a hydrotime model for mangroves was limited as germination of their seeds did not meet model criteria.

While investigating germinability in the new-crop meadowfoam (Limnanthes alba Benth.), it was desired to gain maximum information despite minimal seed material and prior knowledge. Extended use of the Richards function in a factorial germination experiment proved very powerful. The functions yielded estimates of four correlated coefficients, requiring multivariate analysis of variance (MANOVA) to provide valid F-tests among germination profiles. These germination functions provided more rigorous discrimination among treatments than the univariate final germination level. The functions for the imbibants KNO3 and GA4+7, in darkness at 10/5°C, were best, being characterized by high mean absolute germination rate, relatively short duration, and high final germination (upper asymptote). The results provided insights into possible dormancy mechanisms.

Grain production of two wheat cultivars (Triticum aestivum L. cv Recital and Scipion), known for their different germination behaviour, was studied at two different temperatures. The study of dormancy onset during grain development showed that in both cultivars, dormancy developed on the mother plant. In Recital grain, after a period of high germinability, dormancy developed for a transient period. However, full germination was obtained on medium supplemented with fluridone, showing that dormancy was associated with abscisic acid (ABA) biosynthesis inside the grain. Dormancy progressively disappeared during natural drying, at a slower rate for grains developing at 15°C than for grains developing at 25°C. However, at both temperatures, the release from dormancy was complete at maturity. In Scipion grain, dormancy was almost total throughout grain development irrespective of the temperature. However, grains could germinate in the presence of fluridone: changes in sensitivity to fluridone were observed during grain development, showing an increase in dormancy during the first half of development, followed by a progressive decrease during the second half. This decrease occurred later for grains developing at 15°C than for those developing at 25°C. In Scipion grain, unlike the cultivar Recital, release from dormancy was not completed before the end of development on the mother plant. An additional dry storage period was necessary which was shorter for grains developed at 25°C than for grains produced at 15°C. A comparison of embryo ABA levels after a 24-h culture in the presence or absence of fluridone, allowed the ABA synthesis to be estimated. It appears that the depth of dormancy was related to the estimated ABA synthesis capacity of the embryos.

Hydration memory is a phenomenon in which a seed can tolerate discontinuous hydration periods, displaying enhanced germination after one or multiple hydration–dehydration (HD) cycles; it was described physiologically in a few cactus species around 15 years ago. Although no additional work was done on this subject, it has great biotechnological potential since its analysis would permit predictions about whether a seed can withstand discontinuous hydration; in the long run, the knowledge about its regulation might lead to induction of this resistance, so we aimed to provide an initial approach to the molecular mechanisms that underlie hydration memory. This phenomenon was reproduced successfully in our lab with Ferocactus peninsulae seeds. Using two-dimensional (2D) electrophoresis, we compared expression patterns of proteins involved in seed maturation of seeds and seedlings subjected to an HD cycle treatment. We found differential expression of several proteins possibly involved in primary metabolism, ubiquitination pathway and reserve protein availability regulation in seeds and seedlings subjected to an HD cycle. We also found differential stability of total RNA. These results strongly suggest that the differential expression of proteins is at least partially related to the hydration memory process.

Poor wheat seed quality in temperate regions is often ascribed to wet production environments. We investigated the possible effect of simulated rain during seed development and maturation on seed longevity in wheat (Triticum aestivum L.) cv. Tybalt grown in the field (2008, 2009) or a polythene tunnel house (2010). To mimic rain, the seed crops were wetted from above with the equivalent of 30 mm (2008, 2009) or 25 mm rainfall (2010) at different stages of seed development and maturation (17–58 DAA, days after 50% anthesis), samples were harvested serially, and subsequent air-dry seed longevity estimated. No pre-harvest sprouting occurred. Seed longevity (p 50, 50% survival period in experimental hermetic storage at 40°C with c. 15% moisture content) in field-grown controls increased during seed development and maturation, attaining maxima at 37 (2008) or 44 DAA (2009); it declined thereafter. Immediate effects of simulated rain at 17–58 DAA in field studies (2008, 2009) on subsequent seed longevity were negative but small, e.g. a 1–4 d delay in seed quality improvement for treatments early in development, but with no damage detected at final harvests. In rainfall-protected conditions (2010), simulated rain close to harvest maturity (55–56 DAA) reduced longevity immediately and substantially, with greater damage from two sequential days of wetting than one; again, later harvests provided evidence of recovery in subsequent longevity. In the absence of pre-harvest sprouting, the potentially deleterious effects of rainfall to wheat seed crops on subsequent seed longevity may be reversible in full or in part.

In Mediterranean fire-prone ecosystems, annual species specific to post-fire habitats should have a soil seed bank and should be able to germinate after a fire. Therefore, various fire-related cues can be expected to stimulate germination in post-fire annuals. Germination patterns of the rare annual Chaenorhinum rubrifolium (Plantaginaceae) were examined in response to mechanical scarification, heat shock, aqueous smoke, nitrogenous compounds, gibberellic acid, karrikinolide (KAR1), and mandelonitrile (a cyanohydrin analogue, MAN) under dark and photoperiod conditions in the laboratory. Combinations of these treatments were also included in the experiment. Strong physiological dormancy in the seeds of C. rubrifolium was partially broken by several fire-related germination cues, including smoke and nitrate, under light conditions. KAR1 and MAN also stimulated germination, and the highest improvement in germination was achieved in the KAR1 treatment in the presence of light. Heat shock + smoke and KAR1 + MAN combinations had positive synergetic and additive effects on germination under light conditions, respectively. The light played a crucial role in the promotion of germination. The results suggest that multiple fire-related cues operate to stimulate germination in C. rubrifolium, an annual species from the Mediterranean Basin. However, the species may have a broader germination niche than a fire-restricted one.

Conserving alpine ecosystems and the plant communities they contain using ex situ conservation requires an understanding of seed longevity. Knowledge of seed longevity may determine the effectiveness of ex situ seed banking for alpine plant conservation, and may provide insight into plant recruitment in situ. We sought to determine the influence of elevation and climatic variables, as well as plant and seed traits, on the seed longevity of 57 species inhabiting a unique biome, (sub-)alpine regions of mainland Australia. Seed longevity was estimated using controlled accelerated ageing tests to determine the time taken for seed viability to fall by 50%. We found that, across the study species, like alpine seeds elsewhere in the world, Australian alpine seeds are relatively short-lived and overall shorter-lived than Australian plants in general. Seed mass and elevation explained most of the variation in seed longevity among the Australian alpine species considered. Species with larger seed mass, and collections made at higher elevations, were found to have relatively short-lived seeds. Phylogeny, however, explained very little of the variation in longevity. Our results suggest that viability testing for Australian alpine seeds in ex situ seed banks should be conducted with shorter intervals than for the non-alpine flora. This study highlights how seed longevity in the Australian Alps is not dictated primarily by evolutionary lineage but rather by a complex combination of environmental variables and intrinsic seed characteristics. Potential implications for conservation ex situ and in situ in the context of climate change are discussed.

Post-harvest drying prolongs seed survival in air-dry storage; previous research has shown a benefit of drying moist rice seeds at temperatures greater than recommended for genebanks (5–20°C). The aim of this study was to determine whether there is a temperature limit for safely drying rice seeds, and to explore whether the benefit to longevity is caused by high-temperature stress or continued seed development. Seeds of two rice varieties were harvested at different stages of development and dried initially either over silica gel, or intermittently (8 h day–1) or continuously (24 h day–1) over MgCl2 at temperatures between 15 and 60°C for up to 3 days. Seeds dried more rapidly the warmer the temperature. Subsequent seed longevity in hermetic storage (45°C and 10.9% moisture content) was substantially improved by increase in drying temperature up to 45°C in both cultivars, and also with further increase from 45 to 60°C in cv. ‘Macassane’. The benefit of high-temperature drying to subsequent longevity tended to diminish the later the stage of development at seed harvest. Intermittent or continuous drying at high temperatures provided broadly similar improvements to longevity, but with the greatest improvements detected in a few treatment combinations with continuous drying. Heated-air drying of rice seeds harvested before maturity improved their subsequent storage longevity by more than that which occurred during subsequent development in planta, which may have resulted from the triggering of protection mechanisms in response to high-temperature stress.

A clear understanding of the mechanisms involved in releasing seed dormancy is important for effective plant conservation and regeneration of desert species. The dormancy types and dormancy release mechanisms of 19 Fabaceae species from four cold deserts in Xinjiang province, China, were studied. An imbibition experiment was performed to determine the presence or absence of physical dormancy. Other treatments included mechanical scarification, sulphuric acid scarification and simulation of summer temperatures typically encountered in the field. High summer temperature treatments included a maximum soil temperature of 80°C, 65°C and 40°C, conducted under dry and wet conditions. The results showed that all the species had orthodox seeds. Caragana korshinskii, Caragana intermedia, Caragana microphylla and Onobrychis taneitica seeds were not dormant. Glycyrrhiza uralensis, Ammodendron bifolium, Vicia costata and Eremosparton songoricum seeds had combinational dormancy (PY+PD) whereas the other 11 species had physical dormancy. The mechanical scarification and sulphuric acid scarification treatments were significant to break dormancy (>80%). Melilotus suaveolens and Oxytropis sp. seeds were temperature-insensitive. Sophora alopecuroides, S. salsula, E. songoricum, A. bifolium and Cassia tora seeds were wet-heat sensitive. Glycyrrhiza glabra seeds were dry-heat sensitive, while those of Halimodendron halodendron and Astragalus lehmannianus were sensitive to both dry and wet heat. Although high temperatures, typical of summer, promoted the breaking of physical dormancy, the proportion of seeds of which dormancy was broken differed among species. These differences were indicative of the conditions in which the species are found naturally. Species may have different adaptations that promote favourable regeneration. Given suitable temperature and moisture conditions similar to those in their natural environment, most species were capable of regeneration.

Torilis japonica (Apiaceae) has a widespread distribution, extending from western Europe to eastern Asia. In Europe, it usually behaves as a spring-germinating biennial species. Ripe seeds of T. japonica have an underdeveloped embryo and can persist in the soil for several years. The aim of this research was to reveal the mechanisms regulating the seasonal emergence pattern of seedlings. Experiments in a natural environment were performed to study phenology of seedling emergence and embryo growth. Seasonal changes in the dormancy status of T. japonica seeds were examined by regularly exhuming buried seeds and incubating them in controlled conditions. The action of temperature and light in regulating dormancy, embryo growth and germination was studied in the laboratory. Results showed that seeds of T. japonica have non-deep, simple morphophysiological dormancy (MPD), whereby physiological dormancy is broken by moist chilling (5°C). Once MPD was broken, embryo growth and subsequent germination started in spring, when appropriate temperature and light conditions were present. Seeds buried at a depth where light could not reach them showed cyclic changes in their dormancy status; embryo growth in these seeds could not be initiated because of the lack of a light stimulus. As far as we know, this is the first extensive study of seasonal dormancy cycles in a spring-germinating species of the Apiaceae. T. japonica occurs in a temperate climate with cool winters and warm, moist summers. In this climate, spring is often the most favourable season for seedling establishment.

Various sources of germination heterogeneity of carrot (Daucus carota L.) seeds were investigated: germination conditions (temperature, oxygen tension), size of seeds, position of the umbels on the mother plants, and pollination conditions of male-sterile plants in hybrid seed production. All seeds tested germinated over a large range of temperatures (5–35°C). However, low temperatures (5–10°C) and temperatures above 30°C reduced germination. Seeds were also sensitive to oxygen deprivation, but their sensitivity to hypoxia depended on the cultivar. The germination responses of seeds to temperature and oxygen depended on their size, particularly at sub- and supra-optimal temperatures. The largest seeds (1.8–2.1 mm) of commercial lots usually germinated better at 5°C and were more sensitive to oxygen deprivation than the smallest ones (1.2–1.8 mm). Experiments performed with open pollinated plants showed that seeds produced by umbels of first and second orders germinated faster and at higher percentages than those collected on third-order umbels. Evidence for the involvement of pollination conditions in the germination quality of carrot seeds was given by pollination by honey bees of male-sterile plants cultivated at various distances from the pollen donors. The longer the distance of the mother plants from the pollinating plants, the lower the seed yield, the heavier the mean seed weight, the more difficult the germination of seeds, and the higher their sensitivity to oxygen deprivation.

Seeds of Dodonaea viscosa (Sapindaceae) have a water-impermeable seed coat, i.e. physical dormancy (PY). Although mechanical scarification, dry heat, sulphuric acid and hot water treatment make seeds permeable under laboratory conditions, the mechanisms by which dormancy is alleviated in natural environments have not yet been understood completely. The present investigation aims to understand the pattern of dormancy alleviation in D. viscosa seeds using an artificial burial approach for 2 years. Freshly collected seeds held in hydrated soil at 10/20°C, 15/20°C, 15/30°C, 20/35°C and 25°C for 32 weeks germinated to less than 15%, irrespective of storage temperature. Dry storage of seeds at 15, 20, 25 and 30°C for 1 year did not break dormancy. Hot water treatment at 80 and 90°C for 30 s broke dormancy in 90% of the seeds. On the other hand, burying seeds at a depth of 3–5 cm in the natural environment for 2 years increased germination from 7 to 71%. In particular, seeds exhumed after summer in both years showed a significant increase in germination percentage (P< 0.05). However, seeds buried after summer did not germinate to a higher percentage when exhumed prior to summer. We suggest that a high summer temperature, rising above 60°C in the top soil layer of the tropics, is a likely factor breaking dormancy. Most seeds germinated during burial, which indicates that light is not a cue for germination. We conclude that germination of D. viscosa following summer is an adaptive mechanism to tolerate summer droughts, which are common in the dry tropics.

Dormancy in rice (Oryza sativa L.) seed is imposed by certain physical and chemical factors associated with its covering structures, i.e.hull and pericarp. The nature of these germination blocks, their mode ofaction, and processes regulating the release of dormancy are not fully understood. Of nine rice cultivars studied, Ching-shi 15, Stejaree 45, PTB10, and Mahsuri are weakly dormant, and Bansphul, Benaful, Kataktara, Dular, and N22 are dormant. Release of seed dormancy in rice by various treatments, oxidative processes and enzymic changes associated with dormancy, and parallelism between natural and artificially imposed dormancy patterns were examined. The influence of the hull in imposing dormancy was stronger and more prolonged than that of the pericarp. Application of GA3 was effective in inducing germination only in weakly dormant cultivars. Dormancy was completely released in all cultivars by subjecting the seeds to moist heat treatment, by removing the hull and pericarp, and by applying GA3 after dehulling. Dormant cultivars had higher O2 uptake rate and peroxidase activity and lower amylase and dehydrogenase activities than the weakly dormant ones. Hull removal substantially decreased peroxidase activity but enhanced amylase and dehydrogenase activities. Nonanoic acid (C90), a short-chain saturated fatty acid (SCSFA), when exogenously applied to non-dormant seeds imposed dormancy. Dry heat treatment or presoaking in 0.01 m KNO3 or 0.1 m H2O2 was very effective in releasing SCSFA-imposed dormancy. Amylase activity was greatly reduced by treatments with nonanoic acid (C90) or ABA. Considering earlier reports and results of the present study, it is proposed that seed dormancy in rice is regulated both by the presence of SCSFAs and ABA in the hull and the pericarp. The relative significance of these substances in cultivars of tropical and temperate origins and its implications in terms of ecogeographic adaptability are discussed.

The soil seed-bank is an important component of vegetation dynamics. Its presence affects both ecosystem resistance and resilience. A persistent seed-bank is especially important in disturbed habitats and harsh environments. In the hilly-gullied Loess Plateau region, serious soil erosion causes decreases in soil water capacity and constrains vegetation recolonization. A stable and long-term persistent soil seed-bank is necessary for natural vegetation recolonization. We used an integrated measure of the depth distribution of seeds in the soil and the seasonal dynamics of soil seed-banks to analyse the persistence of seeds in soil and to investigate the correlation of seed longevity with seed size/shape and the species' life history. The results showed a significant tendency for small seeds and seeds of annuals/biennials to persist longer in soil than large seeds and seeds of perennials. However, seed shape was not related to persistence. The main dominant speciesArtemisia scoparia, Lespedeza davurica, Heteropappus altaicus, Stipa bungeana, Artemisia gmelinii, and Bothriochloa ischaemun in the different successional stages in this region can form a persistent and stable soil seed-bank. The pioneer species A. scoparia is especially significant because it can form a large, long-term, persistent seed-bank. These species can play a role in the recolonization of the eroded abandoned slope lands by vegetation.

Instrumental neutron activation analysis (NAA) was used to quantify the amounts of K, Mg, Cl, Ca and Mn leaked into water from imbibing dry seeds, dry fruits and isolated seed parts. A number of intact monocot and dicot dry seeds or fruits were studied as were some isolated parts such as testa, endosperm and embryo. All samples studied leaked detectable amounts of the five elements measured but the amounts leaked varied greatly. In most cases K was leaked in greater amounts than the other elements while Mn was leaked least. The leakage of Ca was highest from isolated seed coats and from mericarps of three umbelliferous species. Comparisons of the amount of element leaked with the total amount of that element present in the starting material showed that leakage, especially of K, represents a substantial loss of mineral nutrient reserves. The degree of leakage of elements did not appear to be closely related to the types of organic storage reserves present.

Cell cycle activities are initiated following imbibition of non-dormant seeds. However, it is not known whether cell cycle related events other than DNA replication also remain suppressed in imbibed dormant seeds. The objective of this study was to demonstrate that the transitions between the non-dormant and dormant (both primary and secondary) states are reflected in cell cycle events, such as DNA replication and the changing patterns of the microtubular cytoskeleton involved in the processes of growth and development. The present studies were conducted on seeds from tomato (Lycopersicon esculentum cv. Moneymaker) that possessed primary dormancy or were manipulated to attain secondary dormancy. In addition, a non-dormant abscisic acid (ABA)-deficient mutant, sitw, was used. DNA replication, as measured by flow cytometry, and β-tubulin accumulation, analysed by immunoblotting, were compared with immunocytological studies of active DNA synthesis and microtubular cytoskeleton formation. It is shown that the depth of dormancy, which distinguishes primary and secondary dormancy, may depend on the progression of the cell cycle prior to the induction of dormancy.

While red rice (Oryza sativa L.) can remain dormant and viable for many years when fully imbibed, the environmental factors that stimulate germination or induce secondary dormancy in the field have not been characterized. In this study, the interactions between the extent of dry-afterripening and germination temperature have been evaluated as possible factors. Red rice dispersal units (florets) were afterripened for 0–10 weeks at 30°C and incubated in water at 1, 5, 15, 20, 25, 30 and 35°C for 2 weeks; then, all the ungerminated florets were transferred to 30°C for two additional weeks. Germination at the end of each of the two sequential treatments was compared to define both the effect of differing temperatures on germination (first treatment), and the effect of these temperatures on subsequent germination at the optimum temperature (30°C, second treatment). In afterripening red rice, the opening of the temperature window for germination begins at 25–35°C. Fully dormant florets acquired the ability to germinate ≥ 90% at 30°C after 4 weeks of dry-afterripening. However, imbibing florets for 2 weeks at 15°C followed by 2 weeks at 30°C, yielded suboptimal germination and induced a degree of secondary dormancy, dependent upon the extent of previous dry-afterripening. Cold stratification (1°C) had a consistent promotive effect on the subsequent germination, particularly when preceded by 1–2 weeks of dry-afterripening at 30°C. To monitor the effects of germination temperature, median afterripening time was utilized as a relative dormancy index, and changes in this index have been interpreted as an overlapping of germination and the temperature-induced changes in the dormancy status. Field weather data suggested that low-temperature stratification may be a germination trigger in the field, even in southern Louisiana, and this merits further investigation in studies of soil-buried seeds.

The lipid concentration (LC) of the embryo–endosperm fraction was determined for the seeds of 60 species in 22 families from an Australian tropical lowland rainforest. It was negatively related to the mass of the embryo–endosperm across all species, but the relationship was not significant at P < 0.05. LC was significantly and positively correlated with N concentration when all species were considered, but not within the families represented by the most species (Lauraceae, 13; Sapindaceae, 9; Proteaceae, 6). LC was significantly and markedly higher in more heavily defended seeds (medians: 27 and 6.9%); there is evidence for the view that seeds rich in either lipid or nitrogen are markedly defended. Lipid concentration was also markedly higher, on average, in seeds taken by the scatter-hoarding Uromys caudimaculatus (white-tailed rat) than in seeds of comparable size not taken by Uromys, but not known to be protected by any toxin or irritant (medians: 35 and 3.7%, respectively). However, LC was not notably high in one major group of seeds taken by Uromys – the largest-seeded species of Beilschmiedia and Endiandra (Lauraceae). The comparisons mostly involved different phyletic lines, and were not consistently supported by contrasts within phyletic lines.
