The establishment of reliable serological methods for cysticercosis in pigs is important for the surveillance, control and prevention of taeniosis/cysticercosis in humans as well as in pigs to prevent economic loss. Both ELISA and immunoblot using glycoproteins (GPs) purified by a single step of preparative iso-electric focusing, which are highly useful for human cysticercosis, have been applied for a serological study in pigs naturally infected with Taenia solium. All sera from pigs showed similar responses to those in human cysticercosis. Therefore, it is expected that both ELISA and immunoblots using GPs would be useful in differentiating infected pigs from uninfected ones.

This study reports further observations on encystment and excystment of the paramphistomid trematode Zygocotyle lunata. Of numerous substrates tested in the laboratory for cercarial encystment, i.e. plastic and glass dishes, Styrofoam™, aluminium foil, snail shells, and cheesecloth, all but cheesecloth allowed for 100% encystment within 4 h of cercarial release at 28°C. Numerous physical and chemical factors were tested to kill cysts, and their capacity to do so was determined by the subsequent inability of cysts to excyst within a chemical excystment medium. Vinegar, Italian salad dressing, soy sauce and 10% solutions of sucrose, acetic acid and sodium chloride were the most detrimental. Freezing, boiling and storage of cysts for more than 2 years also killed them. Several marinades such as lemon juice, Worcestershire sauce, and various concentrations of potassium permanganate, did not kill the majority of cysts tested. Since Z. lunata is a potential foodborne trematode for humans and animals, it is important to determine suitable ways to control or kill these cysts.

Aedes aegypti (black-eyed Liverpool strain) were exposed to a sublethal dose (LD25) of Bacillus sphaericus and were fed to Mastomys coucha infected with Brugia malayi. The development of the filarial parasite was found to be arrested mostly at the second larval stage. The infection (P< 0.05), infectivity rates (P< 0.001) and L3 load (P< 0.001) were found to be reduced significantly in the treated group.

Echinostoma caproni tail loss was studied in vitro in the presence of the toxicant copper sulphate (CuSO4) in concentrations ranging from 1 to 10 000 mg l?1 in standardized artificial spring water (pH 7.4, osmolarity 34 mOsm kg?1 H2O, Ca2+ 20 mg l?1) at 23°C. Tail loss was also studied in the absence of toxicants during in vivo encystment of the cercariae in juvenile Biomphalaria glabrata. As the concentration of CuSO4 increased, the percentage of cercarial tail loss increased. By 2 h in 10 000 mg l?1, 1000 mg l?1 and 100 mg l?1 CuSO4, 50%, 23% and 13%, respectively, of the cercariae had lost their tails. In the in vivo studies, by 1 h PI, 59±5% of cercariae had lost their tails and only 4±1% of the cercariae were actively swimming in the multi-well dishes. At 3 h PI, 72±3% of the cercariae began to form cysts within the snails.

It has been, for some time, a recognised fact that intense eelworm infections are, in some instances, accompanied by only slight damage to the plants, while in other areas where the infection is much less intense, the plants show every symptom of “potato sickness” and the crop is a complete failure.

The lungworm, Aelurostrongylus abstrusus, is a common parasite of the domestic cat in Great Britain and produces pulmonary lesions in that species which are occasionally severe. The disease may be reproduced experimentally either by the oral administration of third stage larvae which have been extracted from infected molluscs or by feeding the latter, themselves. For experimental purposes, the former procedure is preferred but the main disadvantage is that an infecting dose must be given on several occasions and over a period of days if sickness, with consequent loss of larvae, is to be avoided. Parenteral administration would, therefore, be advantageous. It is known that pulmonary infections have been produced in calves by the subcutaneous administration of larvae of Dictyocaulus viviparus (Wade and Swanson, 1958), in pigs by the intravenous and intratracheal injection of Metastrongylus spp. larvae (Kelley & Krous, 1961) and in sheep by the intravenous inoculation of Dictyocaulus filaria larvae (Michel and Sinclair, 1963). As neither intravenous nor intratracheal administration is particularly desirable in cats, it was resolved to attempt the reproduction of lungworm disease in that species by the subcutaneous injection of third stage larvae of Aelurostrongylus abstrusus.

A defined medium is described as a basis for in vitro culture work with larval Fasciola hepatica. This medium, termed BCM, can be quickly made up by using a system of stock solutions. BCM contains inorganic salts, glucose, amino acids, vitamins and antibiotics, but no lipid or proteins. Rediae can be dissected from infected snails for culture, but many appear to be contaminated with bacteria. Large rediae cannot survive in BCM but free immature cercariae can complete their final maturation in vitro. This final maturation, from the 30th to the 35th day after miracidial penetration of donor snails, includes tail growth and appearance of body pigmentation. Cercariae matured in vitro encyst successfully when transferred from BCM to water. Small rediae survive in BCM for 5 days, but show no growth or development measured as dry weight and total nitrogen.

The effects in vivo of 5, 10, and 20 mg/kg of luxabendazole (LBZ) on the tegument of Fasciola hepatica have been examined 48 h, 7 days and 14 days post-treatment of experimentally-infected rats. As early as 48 h post-treatment, the drug is shown to provoke significant damage to the tegument. The pathological phenomena characterizing LBZ damage are blebbing of the apical plasmalemma, formation of microvillus-like projections over the free surface, swelling of the basal infolds and stimulation of autophagy. The spines are often fractured; the tegument in the vicinity of spines seems more strongly altered than that in other foci. The basal layer is often changed, from increase of electron density to lack of integrity with the apical cytoplasm. The progress of the ultrastructural damage with time is not expressed. However, cytochemical data show that at longer post-treatment intervals the surface-coat structure becomes irregular and patches of ruthenium red positive material of variable thickness are focally accumulated. Only a slight dose-effect is noted 48 h after LBZ application when the alterations provoked by 5 mg/kg are less evident than those by 10 and 20 mg/kg.

The parasite was found in the bile ducts of three sperm whales, which were caught in the 1957–58 whaling season.

As specimens were found independently in bile ducts of three whales this position in the host appears to be the natural habitat. All the worms were in pieces and incomplete. The pieces, in each case, totalled approximately 18 metres, 0·9 metres and 21 metres respectively. The holotype (length 18 metres) was probably complete except for a few terminal proglottids because it consisted of two very long pieces and one short piece which could be fitted together. The type material has been deposited in the British Museum (Natural History) Registered No. 1961·12·12·1 (Holotype) and 1961·12·12·1 (Para types).

Infective larvae (third-stage larvae) of both Angiostrongylus cantonensis and A. costaricensis from their snail intermediate host were subjected to either X-ray or gamma-ray irradiation. The viability of the irradiated larvae was assayed by oral inoculation of the larvae into rodents (A. cantoflensis in mice and rats, A. costaricensis in mice only). From the results of worm recovery, the minimal dose of irradiation that inhibited the infectivity of the third-stage larvae of A. cantonensis and A. costaricensis was 2 and 4 kGy, respectively.

In an earlier paper Goodey (1940) gave a detailed description of the morphology of three species of Anguillulina viz.: A. multicincta (Cobb) Goodey, A. erythrinae (Zimmermann) Goodey and A. robusta (de Man) Goodey. All three species are associated with plant roots: A. multicincta is a parasite of banana roots, A. erythrinae is parasitic on the roots of certain common grasses and cereals such as Agrostis stolonifera, Lolium perenne and oats (the writer has also found it on hop roots), A. robusta though associated with grass roots has not, to the writer's knowledge, been found within plant tissues. In the case of all three species phasmids were easily recognized on the female tails and were described and figured but they were not seen on the male tails and it was assumed that they were probably absent.

In two previous papers (Dissanaike et al., 1957a and 1957b) we described the technique of tagging Culex fatigans and Armigeres subalbatus (=obturbans) with P32 to obtain high beta activities. Filarial larvae were made to develop to the infective stage in these activated mosquitoes and were found to have beta activities high enough for detection individually.

The complex life cycle of Trichinella spiralis includes the migration of newborn larvae through the bloodstream to their encystment in muscle. The parasite establishes an intimate contact with the erythrocytes of the host both during the migration of the newborn larvae and when encysting, as this parasite causes intense vascularization in the muscle cell. The goal of this work was to study the effects of various concentrations of T. spiralis muscle larvae (ML) on erythrocyte membranes. The treatment was performed by incubating human erythrocytes with equal volume of different concentrations of ML for 30 minutes, with controlled agitation (37°C). The control erythrocytes (with no contact with the larvae) were incubated in the same way with an equal volume of physiological solution. To evaluate the alterations to the erythrocytes by the action of the larvae and in the respective controls, an Erythrocyte Rheometer and a Digital Image Analysis technique were used. The results indicated that when the larval concentration was higher, the aggregation and erythrocyte membrane alterations were also higher. Also, the erythrocyte deformability index and the erythrocyte elasticity increased. The values of isolated cell coefficient varied from 0.51 in the treatment with 100 larvae/ml to 0.91 in the incubation with 1000 larvae/ml. This experiment shows that T. spiralis muscle larvae affect significantly the red blood cell aggregation and the erythrocyte viscoelastic properties.

Epidemiological studies of Echinococcus multilocularis infections in definitive hosts require a reliable and economic diagnostic method. In this study, the current copro-DNA examination technique was modified by increasing the faecal amounts tested and adding a step to neutralize the faeces before DNA extraction. Reliability of the modified method was evaluated using rectal faecal samples from red foxes and comparing them with intestinal worms detected using the sedimentation and counting technique (SCT) following necropsy. The modified copro-DNA examination method demonstrated 93.9% sensitivity (138/147) on the SCT. Its detectability increased depending on the worm burden, and the sensitivity was 100% in cases harbouring over 1000 worms. From 111 SCT-negative cases, six (5.4%) were copro-DNA-positive, and all were confirmed as E. multilocularis via sequencing analysis. Five of the remaining 105 SCT-negative cases (4.8%) retained polymerase chain reaction (PCR) inhibitors in the extracted solution, suggesting that approximately 5% of the red fox faeces retained these inhibitors after treatment with the present copro-DNA extraction method. Although further evaluation is needed for faeces deposited in the wild, the present copro-DNA examination technique will help monitor the E. multilocularis prevalence in definitive hosts. When used for detailed evaluations of endemicity (e.g. changes in infection pressure or spread in non-endemic areas), the absence of PCR inhibitors should be confirmed, and multiple trials on faecal subsamples are recommended.

Polycystic echinococcosis (PE) is a zoonosis endemic in the Neotropical region of the Americas. It is caused by the larval stage of the cestode Echinococcus vogeli, which develops as harmful cysts that slowly grow in the liver, lungs and other organs of humans and other host species. Human PE diagnosis is usually based on clinical and epidemiological aspects and imaging techniques, often requiring confirmation by immunological assays. The currently available serological tests for detecting antibodies against Echinococcus spp. are mostly based on complex, variable and poorly characterized mixtures of native parasite antigens, which impairs specificity and/or sensitivity. In this scenario, the evaluation of well-characterized alternative antigens is urgently needed for the improvement of PE diagnosis. Here, two subunits (AgB8/1 and AgB8/2) of the major secretory antigen from Echinococcus granulosus (antigen B (AgB)), of diagnostic value for cystic echinococcosis, were validated for PE diagnosis. These antigens, produced as pure recombinant proteins (rAgB8/1 and rAgB8/2) in Escherichia coli, allowed detecting specific immunoglobulin G antibodies in sera from PE patients in an enzyme-linked immunosorbent assay, with sensitivities of 83.72% and 81.40%, respectively, and specificities of 83.12% and 80.09%, respectively. The use of recombinant proteins overcomes difficulties to obtain parasite material and reduced non-specific reactions and costs. Our results demonstrated reproducibility and accuracy high enough to be considered valid according to the acceptance criteria for Food and Drug Administration assay validation. This qualifies rAgB8/1 and rAgB8/2 as potential substitutes for the currently used parasite crude or partially purified antigens.

Via molecular and morphological analyses, we describe adult specimens of a new species of Versteria (Cestoda: Taeniidae) infecting mink and river otter (Carnivora: Mustelidae) in Western Canada, as well as larval forms from muskrat and mink. These sequences closely matched those reported from adult specimens from Colorado and Oregon, as well as larval infections in humans and a captive orangutan. We describe here a new species from British Columbia and Alberta (Canada), Versteria rafei n. sp., based upon morphological diagnostic characteristics and genetic distance and phylogeny. Versteria rafei n. sp. differs from the three other described species of the genus in the smaller scolex and cirrus sac. It also differs from V. mustelae (Eurasia) and V. cuja (South America) by having an armed cirrus, which is covered in hair-like bristles, and in the shape of its hooks, with a long thorn-like blade, and short or long handle (vs. a short sharply curved blade and no difference in handle size in previously described species). The poorly known V. brachyacantha (Central Africa) also has an armed cirrus and similarly shaped hooks. However, it differs from the new species in the number and size of hooks. Phylogenetic analysis of the cox1 and nad1 mitochondrial regions showed that our specimens clustered with isolates from undescribed adults and larval infections in North America, and separate from V. cuja, confirming them to be a distinct species from the American Clade.

Here we describe a new species of the genus Saccocoelioides found parasitizing Astyanax dissimilis Garavello & Sampaio, Psalidodon bifasciatus (Garavello and Sampaio) and Bryconamericus ikaa Casciotta, Almirón & Azpelicueta from the Iguazu National Park, Misiones province, Argentina. Saccocoelioides miguelmontesi n. sp. was studied based on morphological and molecular (28S rDNA and COI mtDNA sequences) data. The COI mtDNA tree indicated that the specimens collected from the three fish hosts are conspecific, with an intragroup p-distance of 0%. The new species shows an intermediate morphological configuration between the diminutive and robust forms described for Saccocoelioides by Curran (2018). Although, in the 28S rDNA tree, it is placed in a well-supported clade with the two robust species analysed (S. elongatus and S. magnus; p-distance of 1 and 2%, respectively), it differs from the robust group by the range of body size, mature egg size, oral and ventral sucker size, sucker ratio, oral sucker to pharynx ratio, and post-cecal or post-testis/body length percentage. Our results led us to redefine the robust group as having eggs shorter or equal in length to the pharynx. Saccocoelioides miguelmontesi n. sp. the 10th species reported from Argentina and the 7th species within the robust group.

Cement glands are one of the most conspicuous and distinctive elements of taxonomic interest in male Acanthocephala. Cement glands vary in shape, number and arrangement in different classes of the taxon. The glands and their products have a fundamental role in the reproductive process. Light and electron microscopy were used to investigate the ultrastructure of the cement apparatus, which includes both cement glands and the cement reservoir, in mature males of Centrorhynchus globocaudatus (Zeder, 1800). Centrorhynchus globocaudatus is an enteric parasite of birds of prey, including Falco tinnunculus (Linnaeus, 1758) and Buteo buteo (Linnaeus, 1758) from the province of Ferrara (northern Italy). The four elongated cement glands of C. globocaudatus are situated posterior to the testes. Sections through the cement glands show each gland is surrounded by a fibrous envelope with an approximate thickness of 0.6 μm. Beneath this envelope is an outer cytoplasmic layer thickness ranging from 22 to 26 μm, which contains a number of nuclei with diameters variable from 20 to 22 μm. The cytoplasmic layer is filled with prominent free ribosomes and many mitochondria with lamellar cristae. Secretory granules, measuring from 1 to 1.3 μm in diameter, are formed within the cytoplasmic layer. The cytoplasmic layer surrounds the luminal area for storage of the cement material in each gland. Cement gland ducts arise from the gland and extend towards a common cement reservoir in close contact with the seminal vesicle and Saefftigen's pouch. Microtubules, large secretory granules and rest of undefined organelles were also observed within the cement reservoir.

Thada populus n. sp. was found in the rhizosphere of Populus euphratica in the city of Dezful, south-western Iran. The new species is characterized by its 365–453 μm body length, cuticle with transverse annuli 0.9–1.2 μm wide, lateral fields with four lines, lip region low with one or two annuli, 7.0–8.5 μm wide at base, longitudinal or slightly sigmoid amphidial slit, delicate stylet 8.4–9.8 μm long with posteriorly directed knobs, dorsal pharyngeal gland opening at 1.0–1.5 μm posterior to the stylet knobs, non-muscular and valve-less median bulb, pyriform and offset basal bulb, almost oval spermatheca, posterior position of vulva (V = 75–79%) without lateral membrane, short post-vulval uterine sac and conical tail with finely to broadly rounded terminus. Morphological differences of the new species with those of the type species, Thada striata, are discussed. Molecular phylogenetic studies of the new species using partial sequences of small subunit ribosomal DNA revealed that the new species formed a clade with Malenchus spp., Filenchus facultativus, F. fungivorus and Filenchus sp. in Bayesian inference. Morphological and molecular phylogenetic studies were also performed on Tenunemellus indicus, the second recovered species, the latter analysis using partial sequences of large subunit ribosomal DNA. In the inferred Bayesian tree, T. indicus formed a clade with Labrys fuzhouensis, Lelenchus leptosoma from the Netherlands, Malenchus spp. and Filenchus discrepans.

The legume pod-borer, Maruca vitrata Fabricius (Lepidoptera: Crambidae) (LPB), is an important insect pest of pigeon pea. Chemical pesticides are generally employed to manage this pest, but because of the soil residue issues and other environmental hazards associated with their use, biopesticides are also in demand. Another benign alternative is to use entomopathogenic nematodes (EPNs) to manage this vital pest. In the present study, the infectivity of ten native EPNs was evaluated against LPB by assessing their penetration and production in the LPB. The effectiveness of the promising EPNs against second-, third- and fourth-instar LPB larvae was also studied. Heterorhabditis sp. (Indian Agricultural Research Institute-Entomopathogenic Nematodes Rashid Pervez (IARI-EPN RP) 06) and Oscheius sp. (IARI-EPN RP 08) were found to be most pathogenic to LPB, resulting in about 100% mortality within 72 h, followed by Steinernema sp. (IARI-EPN RP 03 and 09). Oscheius sp. (IARI-EPN RP 04) was found to be the least pathogenic to LPB larva with 67% mortality. Maximum penetration was exhibited by Heterorhabditis sp. (IARI-EPN RP 06) followed by Oscheius sp. (IARI-EPN RP 08), whereas the lowest rate of penetration was exhibited by Oscheius sp. (IARI-EPN RP 01). The highest rate of production was observed with Oscheius sp. (IARI-EPN RP 08), followed by Oscheius sp. (IARI-EPN RP 04 and 10). Among the tested instars of the LPB larvae, second-instar larvae were more susceptible to EPNs, followed by third- and fourth-instar larvae. The results indicate that Heterorhabditis sp. (IARI-EPN RP 06) and Oscheius sp. (IARI-EPN RP 08) have a good potential to the manage LPB.
