Of 17 Trichinella isolates from domestic pigs and wild boars (Sus scrofa) in regions where Trichinella nativa is widespread among sylvatic animals, two wild boars from Estonia were found to be naturally infected with this Trichinella species. The other 15 animals were infected with Trichinella spiralis. Trichinella nativa is tolerant to freezing when in the muscles of carnivores. The biological characteristics and temperature tolerance of this species in swine need to be further investigated if pork is certified for consumption following freezing.

Echinostoma caproni tail loss was studied in vitro in the presence of the toxicant copper sulphate (CuSO4) in concentrations ranging from 1 to 10 000 mg l?1 in standardized artificial spring water (pH 7.4, osmolarity 34 mOsm kg?1 H2O, Ca2+ 20 mg l?1) at 23°C. Tail loss was also studied in the absence of toxicants during in vivo encystment of the cercariae in juvenile Biomphalaria glabrata. As the concentration of CuSO4 increased, the percentage of cercarial tail loss increased. By 2 h in 10 000 mg l?1, 1000 mg l?1 and 100 mg l?1 CuSO4, 50%, 23% and 13%, respectively, of the cercariae had lost their tails. In the in vivo studies, by 1 h PI, 59±5% of cercariae had lost their tails and only 4±1% of the cercariae were actively swimming in the multi-well dishes. At 3 h PI, 72±3% of the cercariae began to form cysts within the snails.

The survival characteristics of the cercariae of Hypoderaeum conoideum and Euparyphium albuferensis (Trematoda: Echinostomatidae) at 20°C and 30°C are described, and the age dependency of their infectivity at 20°C is studied to determine their respective transmission efficiencies. Cercarial survival was found to be age-dependent and was higher at 20°C. For both cercariae, the maximum life-span was 26 h at 20°C and 16 h at 30°C, and their respective times to 50% mortality were similar at each temperature. Both cercariae seem to be well adapted to transmission in their natural habitat, though cercarial infectivity of H. conoideum was higher than that of E. albuferensis, this being correlated with their prevalences in nature. The age-dependency of cercarial survival may be related to steadily diminishing endogenous energy levels, though the delay in attaining maximum infectivity suggests that other factors not related to energy considerations are involved in the delimitation of the cercarial infective period. However, this latter observation may constitute an adaptative mechanism allowing cercarial dissemination.

Epidemiological studies of Echinococcus multilocularis infections in definitive hosts require a reliable and economic diagnostic method. In this study, the current copro-DNA examination technique was modified by increasing the faecal amounts tested and adding a step to neutralize the faeces before DNA extraction. Reliability of the modified method was evaluated using rectal faecal samples from red foxes and comparing them with intestinal worms detected using the sedimentation and counting technique (SCT) following necropsy. The modified copro-DNA examination method demonstrated 93.9% sensitivity (138/147) on the SCT. Its detectability increased depending on the worm burden, and the sensitivity was 100% in cases harbouring over 1000 worms. From 111 SCT-negative cases, six (5.4%) were copro-DNA-positive, and all were confirmed as E. multilocularis via sequencing analysis. Five of the remaining 105 SCT-negative cases (4.8%) retained polymerase chain reaction (PCR) inhibitors in the extracted solution, suggesting that approximately 5% of the red fox faeces retained these inhibitors after treatment with the present copro-DNA extraction method. Although further evaluation is needed for faeces deposited in the wild, the present copro-DNA examination technique will help monitor the E. multilocularis prevalence in definitive hosts. When used for detailed evaluations of endemicity (e.g. changes in infection pressure or spread in non-endemic areas), the absence of PCR inhibitors should be confirmed, and multiple trials on faecal subsamples are recommended.

Thada populus n. sp. was found in the rhizosphere of Populus euphratica in the city of Dezful, south-western Iran. The new species is characterized by its 365–453 μm body length, cuticle with transverse annuli 0.9–1.2 μm wide, lateral fields with four lines, lip region low with one or two annuli, 7.0–8.5 μm wide at base, longitudinal or slightly sigmoid amphidial slit, delicate stylet 8.4–9.8 μm long with posteriorly directed knobs, dorsal pharyngeal gland opening at 1.0–1.5 μm posterior to the stylet knobs, non-muscular and valve-less median bulb, pyriform and offset basal bulb, almost oval spermatheca, posterior position of vulva (V = 75–79%) without lateral membrane, short post-vulval uterine sac and conical tail with finely to broadly rounded terminus. Morphological differences of the new species with those of the type species, Thada striata, are discussed. Molecular phylogenetic studies of the new species using partial sequences of small subunit ribosomal DNA revealed that the new species formed a clade with Malenchus spp., Filenchus facultativus, F. fungivorus and Filenchus sp. in Bayesian inference. Morphological and molecular phylogenetic studies were also performed on Tenunemellus indicus, the second recovered species, the latter analysis using partial sequences of large subunit ribosomal DNA. In the inferred Bayesian tree, T. indicus formed a clade with Labrys fuzhouensis, Lelenchus leptosoma from the Netherlands, Malenchus spp. and Filenchus discrepans.

The legume pod-borer, Maruca vitrata Fabricius (Lepidoptera: Crambidae) (LPB), is an important insect pest of pigeon pea. Chemical pesticides are generally employed to manage this pest, but because of the soil residue issues and other environmental hazards associated with their use, biopesticides are also in demand. Another benign alternative is to use entomopathogenic nematodes (EPNs) to manage this vital pest. In the present study, the infectivity of ten native EPNs was evaluated against LPB by assessing their penetration and production in the LPB. The effectiveness of the promising EPNs against second-, third- and fourth-instar LPB larvae was also studied. Heterorhabditis sp. (Indian Agricultural Research Institute-Entomopathogenic Nematodes Rashid Pervez (IARI-EPN RP) 06) and Oscheius sp. (IARI-EPN RP 08) were found to be most pathogenic to LPB, resulting in about 100% mortality within 72 h, followed by Steinernema sp. (IARI-EPN RP 03 and 09). Oscheius sp. (IARI-EPN RP 04) was found to be the least pathogenic to LPB larva with 67% mortality. Maximum penetration was exhibited by Heterorhabditis sp. (IARI-EPN RP 06) followed by Oscheius sp. (IARI-EPN RP 08), whereas the lowest rate of penetration was exhibited by Oscheius sp. (IARI-EPN RP 01). The highest rate of production was observed with Oscheius sp. (IARI-EPN RP 08), followed by Oscheius sp. (IARI-EPN RP 04 and 10). Among the tested instars of the LPB larvae, second-instar larvae were more susceptible to EPNs, followed by third- and fourth-instar larvae. The results indicate that Heterorhabditis sp. (IARI-EPN RP 06) and Oscheius sp. (IARI-EPN RP 08) have a good potential to the manage LPB.

Cement glands are one of the most conspicuous and distinctive elements of taxonomic interest in male Acanthocephala. Cement glands vary in shape, number and arrangement in different classes of the taxon. The glands and their products have a fundamental role in the reproductive process. Light and electron microscopy were used to investigate the ultrastructure of the cement apparatus, which includes both cement glands and the cement reservoir, in mature males of Centrorhynchus globocaudatus (Zeder, 1800). Centrorhynchus globocaudatus is an enteric parasite of birds of prey, including Falco tinnunculus (Linnaeus, 1758) and Buteo buteo (Linnaeus, 1758) from the province of Ferrara (northern Italy). The four elongated cement glands of C. globocaudatus are situated posterior to the testes. Sections through the cement glands show each gland is surrounded by a fibrous envelope with an approximate thickness of 0.6 μm. Beneath this envelope is an outer cytoplasmic layer thickness ranging from 22 to 26 μm, which contains a number of nuclei with diameters variable from 20 to 22 μm. The cytoplasmic layer is filled with prominent free ribosomes and many mitochondria with lamellar cristae. Secretory granules, measuring from 1 to 1.3 μm in diameter, are formed within the cytoplasmic layer. The cytoplasmic layer surrounds the luminal area for storage of the cement material in each gland. Cement gland ducts arise from the gland and extend towards a common cement reservoir in close contact with the seminal vesicle and Saefftigen's pouch. Microtubules, large secretory granules and rest of undefined organelles were also observed within the cement reservoir.

The objective of this study was to evaluate the predatory activity of the nematophagous fungus Duddingtonia flagrans on infective larvae of gastrointestinal nematodes from dairy heifers in different conditions (shaded and sunny) of a silvopastoral system (SPS) on an agroecological farm. Ten Jersey heifers were divided into two groups: treated (received pellets containing fungus); and control (received pellets without fungus). Twelve hours after fungus administration, faeces samples were collected for in vitro efficacy tests. The animals then remained for 8 h in the experimental pasture area. At the end of this period, 20 faecal pads (10 treated and 10 control) were selected. Pasture, faecal pad and soil collections occurred at intervals of seven days (d), totalling four assessments over 28 d. To evaluate the influence of the conditions shaded and sunny, we registered the condition of the location of each faecal pad per hour. After 12 h of gastrointestinal transit in dairy heifers, a reduction of 65% was obtained through the in vitro test. The treated group presented a lower number of infective larvae (L3) in the faecal pad and upper pasture. Differences in numbers of L3 were observed between the conditions (sunny and shaded) in the faecal pad of the control group; while in the treated group there were no differences between the conditions. The predatory activity of the fungus was efficient over time in the shaded and sunny conditions of an SPS, decreasing the parasite contamination during the pasture recovery time in a subtropical climate.

A new genus, Mystocestus, is proposed to accommodate a new species, Mystocestus anindoi n. g., n. sp. from Mystus vittatus (Bloch) (type host) in West Bengal and Mystus cavasius (Hamilton) (Siluriformes: Bagridae) in Maharashtra, India. The new genus is most similar to Lucknowia Gupta, 1961 in the shape of the body, which is elongate, slightly tapering towards the anterior end, and scolex, which is digitiform, but differs in the shape of the ovary, which is H-shaped (vs. inverted A-shaped in Lucknowia), the absence of a seminal receptacle (present in Lucknowia) and exclusively cortical vitelline follicles (vs. some follicles in the medulla in the latter genus). Molecular data support the erection of the new genus and place it close to Bovienia Fuhrmann, 1931, species of which can be easily distinguished by exclusively lateral vitelline follicles (lateral and median in the new genus), the presence of a seminal receptacle (absent in Mystocestus) and scolex shape (digitiform, with blunt or slightly concave anterior edge in the new genus vs. small, unspecialized or spatulate in Bovienia). The convoluted taxonomy of tapeworms placed in Mystoides Mathur, 1992 is critically reviewed to clean up taxonomic chaos in Indo-Malayan caryophyllideans. Mystoides was erected in an unpublished PhD thesis and thus its generic name becomes unavailable and also, its type species is conspecific with Lucknowia fossilisi Gupta, 1961 from the stinging catfish, Heteropneustes fossilis (Bloch). Other species of this genus are also conspecific with L. fossilisi. In addition, specific names of most of these taxa are unavailable because they were described in unpublished theses or conference abstracts. Based on recent revisions of Indo-Malayan caryophyllideans, the following nine genera with 15 species are considered valid (numbers of species of individual genera are provided in parentheses): Bovienia (3), Djombangia (1), Lucknowia (2), Lytocestus (2), Mystocestus (1), Pseudocaryophyllaeus (2) (all family Lytocestidae); Adenoscolex (1), Lobulovarium (2), Paracaryophyllaeus (1) (all family Caryophyllaeidae).

This is the first study reporting parasites from the freshwater cyprinid Oxynoemacheilus angorae (Steindachner 1897) caught in Nilüfer Stream, Bursa, in the Northwest Anatolian Region of Turkey. Allocreadium bursensis n. sp. was described from the intesine of O.angorae based on morphological and genetic characteristics. Allocreadium bursensis n. sp. was differentiated from other Allocreadium spp. in having a combination of external (ventral and oral suckers ratio; body length and width and its ratio to forebody) and internal (cirrus pouch position; uterus extension in hindbody; egg size; disposition of anterior border of vitellarium; esophagus length) features. Phylogenetic hypotheses based on maximum parsimony, maximum likelihood, Bayesian inferrence, and neighbor joining analyses of sequence data strongly supported the hypothesis that A. bursensis is nested within the clade of Allocreadium species hosted by cypriniform fish, and it is more closely related to the Far Eastern species A. pseudoisoporum (Primorsky region, Russia) than to the African A. apokryfi. According to genetic p-distances, the taxonomic status of trematodes collected in Turkey was established as independent relative to nine of the valid Allocreadium spp.: 1.8–5.8% in 28S gene and 18.8–22.6% in cox1 gene. The present study increases the number of Allocreadium species and their definitive hosts recorded in Turkey and raises the number of Palearctic representatives of Allocreadium spp. to 26.

The complex life cycle of Trichinella spiralis includes the migration of newborn larvae through the bloodstream to their encystment in muscle. The parasite establishes an intimate contact with the erythrocytes of the host both during the migration of the newborn larvae and when encysting, as this parasite causes intense vascularization in the muscle cell. The goal of this work was to study the effects of various concentrations of T. spiralis muscle larvae (ML) on erythrocyte membranes. The treatment was performed by incubating human erythrocytes with equal volume of different concentrations of ML for 30 minutes, with controlled agitation (37°C). The control erythrocytes (with no contact with the larvae) were incubated in the same way with an equal volume of physiological solution. To evaluate the alterations to the erythrocytes by the action of the larvae and in the respective controls, an Erythrocyte Rheometer and a Digital Image Analysis technique were used. The results indicated that when the larval concentration was higher, the aggregation and erythrocyte membrane alterations were also higher. Also, the erythrocyte deformability index and the erythrocyte elasticity increased. The values of isolated cell coefficient varied from 0.51 in the treatment with 100 larvae/ml to 0.91 in the incubation with 1000 larvae/ml. This experiment shows that T. spiralis muscle larvae affect significantly the red blood cell aggregation and the erythrocyte viscoelastic properties.

Human cystic echinococcosis (CE) is a zoonotic disorder triggered by the larval stage of Echinococcus granulosus (E. granulosus) and predominantly occurred in the liver and lungs. The M2 macrophage level is considerably elevated among the liver of patients with hepatic CE and performs an integral function in liver fibrosis. However, the mechanism of CE inducing polarisation of macrophage to an M2 phenotype is unknown. In this study, macrophage was treated with E. granulosus cyst fluid (EgCF) to explore the mechanism of macrophage polarisation. Consequently, the expression of the M2 macrophage and production of anti-inflammatory cytokines increased after 48 h treatment by EgCF. In addition, EgCF promoted polarisation of macrophage to an M2 phenotype by inhibiting the expression of transcriptional factor hypoxia-inducible factor 1-alpha (HIF-1α), which increased the expression of glycolysis-associated genes, including hexokinase 2 (HK2) and pyruvate kinase 2 (PKM2). The HIF-1α agonist ML228 also inhibited the induction of macrophage to an M2 phenotype by EgCF in vitro. Our findings indicate that E. granulosus inhibits glycolysis by suppressing the expression of HIF-1α.

The aim of this study was to develop a simple method to purify Strongyloides eggs from rat faeces using a sucrose gradient centrifugal-flotation technique. This procedure is simple, rapid and possesses a high efficiency in recovering Strongyloides eggs without faecal detritus in less than one hour, thus eliminating the use of complex apparatus and different chemical substances. The possibility of working with pure and live Strongyloides eggs opens up a wide range of future studies on the biology of this parasite. This study constitutes the first report in the scientific literature on purifying Strongyloides eggs using a sucrose density gradient.

In 1988, 1989 and 1990 second year grazing heifers, naturally infected with gastrointestinal nematodes, were separated into two groups, one of which was treated orally with albendazole. In 1988 and 1989 treatment was administered immediately after parturition (February), and in 1990 during the last term of pregnancy (December). Both treated and control animals were grazed on separate plots in a rotational system. Maximum faecal egg counts were observed amund parturition, except in 1990, when treatment was given at the end of gestation. The main genera identified were Cooperia, Trichostrongylus, Ostertagia and Oesophagostomum. The number of Ostertagia larvae in the treated groups increased from 1989 to 1990, while the others decreased. Pasture contamination with third stage larvae (L3) was lower on the piots grazed by treated heifers. Maximum numbers of L3 were found in autumn, at the end of winter, and at the beginning of spring. Mean serum pepsinogen concentrations were significantly higher in the untreated groups. This concurs with the pattern for L3 on pasture. The trial shows that if a single treatment against gastrointestinal nematodes is carried out, and the animals remain on contaminated pastures, the parasitic load tends to level out after 4–5 months under favourable climatic conditions. However, the percentages of nematode genera occurring in the new populations may differ from those in the original infection.

Strongylus (Dicerocola) tremletti sp.nov. is described from the black rhinoceros, Diceros bicornis L. from Kenya.

The affinities of S. (D.) tremletti sp.nov. with the other species of the genus Strongylus Müller, 1780 are discussed and the subgenus Dicerocola subgen.nov. is erected to contain it.

The species of Strongylus in Kenya are noted and original drawings of S. (D.) asini Boulenger, 1920 are included.

Compressed organ autoradiography has been utilized to study the migration of the homogonic strain of Strongyloides ratti from the site of skin penetration to the gut. This transit is characterized by rapid disappearance of parasites from the cutaneous site of infection, followed by vascular dispersal throughout all organs investigated. For the first time, parasite migration in both naive and previously sensitized hosts is compared; the principal stations of migration are the same in the two groups although there is a quantitative difference in accumulation of parasites with time in these stations. Parasite attrition occurs in both groups of animals; however, in naive rats it is not manifest until day 20–25 post challenge, whereas in sensitized rats immune elimination occurs as early as 48 h post challenge.

Infective larvae (third-stage larvae) of both Angiostrongylus cantonensis and A. costaricensis from their snail intermediate host were subjected to either X-ray or gamma-ray irradiation. The viability of the irradiated larvae was assayed by oral inoculation of the larvae into rodents (A. cantoflensis in mice and rats, A. costaricensis in mice only). From the results of worm recovery, the minimal dose of irradiation that inhibited the infectivity of the third-stage larvae of A. cantonensis and A. costaricensis was 2 and 4 kGy, respectively.

The worm burden in grazing ewes was studied. Analysis of the results showed that correcting worm egg counts (e.p.g. fresh faeces) for variations in faecal D.M. content and in total faeces output is not of any great benefit unless precise information is available.

Data are also presented which suggest that under normal circumstances variations in the sub-clinical worm burden of adult grazing ewes are not related to any variations in food intake.
