Two types of artificial neural networks, multilayer perceptron (MLP) and self-organizing feature map (SOM) were used to detect mastitis by automatic milking systems (AMS) using a new mastitis indicator that combined two previously reported indicators based on higher electrical conductivity (EC) and lower quarter yield (QY). Four MLPs with four combinations of inputs were developed to detect infected quarters. One input combination involved principal components (PC) adopted for addressing multi-collinearity in the data. The PC-based MLP model was superior to other non-PC-based models in terms of less complexity and higher predictive accuracy. The overall correct classification rate (CCR), sensitivity and specificity of this model were 90·74%, 86·90% and 91·36%, respectively. The SOM detected the stage of progression of mastitis in a quarter within the mastitis spectrum and revealed that quarters form three clusters: healthy, moderately ill and severely ill. The clusters were validated using k-means clustering, ANOVA and least significant difference. Clusters reflected the characteristics of healthy and subclinical and clinical mastitis, respectively. We conclude that the PC based model based on EC and QY can be used in AMS to detect mastitis with high accuracy and that the SOM model can be used to monitor the health status of the herd for early intervention and possible treatment.

Diet is a relatively simple way to modify milk fat yield and composition in dairy cows as the end-products of digestion are precursors or inhibitors of milk fat synthesis. The individual effects of these end-products are well-known, but it is still not known whether these nutrients have an additive effect or an interaction effect on milk fat secretion. Thus our objective was to investigate the effects of two of these nutrients on milk fat secretion, trans-10, cis-12 conjugated linoleic acid (CLA) and propionic acid (C3) supplied alone or together, under the same experimental conditions. Four Holstein dairy cows were used in a 4×4 Latin square design with 14-d periods. Treatments were control, CLA (duodenal infusion of 1·85 g/d of trans-10, cis-12 CLA), C3 (ruminal infusion of 500 g/d of C3) and CLA+C3 (duodenal infusion of 1·85 g/d of trans-10, cis-12 CLA plus ruminal infusion 500 g/d of C3). Infusions of trans-10, cis-12 CLA reduced milk fat content and yield by 18% whereas C3 infusions had no significant effect on milk fat secretion. Trans-10, cis-12 CLA decreased the yields of all milk fatty acids (FA). This reduction was proportionally greater for FA synthesized de novo than for preformed long-chain FA. Infusions of C3 decreased the yields and percentages of 4:0 and 18:0 and increased the yields and percentages of all odd-chain FA. Interactions between trans-10, cis-12 CLA and C3 infusions on milk fat content, yield and FA composition were never significant. Overall, this study showed that trans-10, cis-12 CLA has different and greater effects on milk fat secretion than C3. Moreover, under our experimental conditions, their effects on milk FA yields, which reflect their effects on mammary lipogenesis, were additive, whatever their individual effect.

Donkey's milk is well tolerated by human infants with cow's milk allergy and is useful in the treatment of human immune-related diseases and in the prevention of atherosclerosis. Thyroid hormones (TH) stimulate lactation and active triiodothyronine (T3) in colostrum and milk could take paracrine action supporting lactogenesis in the mother, and play physiological roles for the suckling offspring (systemic or within the gastrointestinal tract). The aims were to measure TH concentrations in donkey blood and milk, validate ELISA methods, evaluate the effects of sample collection and post-collection handling and the stability of TH in milk and blood serum and plasma samples. In milk and blood samples obtained from lactating jennies total concentrations of TH were assayed using competitive-type ELISA kits. Good validation results were obtained for both TH concentrations in blood serum and plasma and T3 in milk samples extracted with cold (−20°C) ethanol alkalinized (pH 9·0) with NH4OH. In most of the milk extract samples, thyroxine (T4) concentrations resulted below the sensitivity threshold. Intra- and inter-assay coefficients of variations of TH concentrations in different blood and milk samples were below 10%. Parallelism tests gave displacement lines parallel to those of the calibrators for both TH in blood serum and plasma and for T3 in milk extracts. Mean recovery rates were between 95% and 123%, but the concentration values approaching the highest calibrators were overestimated. Therefore, serum and plasma samples for T3 assay must be previously diluted with buffer. Both TH concentrations in blood serum and plasma and T3 in milk did not change during storage for up to 6 months at −20°C. In conclusion, the ELISA methods tested in the present study are suitable for determination of both TH concentrations in donkey blood samples, and for T3 measurement in milk, after extraction with cold alkaline ethanol.

The aim of this study was to assess the concentration of penicillin G in mammary tissue and secretion of dry heifers following systemic administration of penethamate hydriodide. Six dairy heifers in late gestation received a single intramuscular injection of 10 g penethamate hydriodide and were sacrificed 24, 48 or 144 h after treatment. Penicillin G concentrations were measured in mammary tissue and secretion samples using HPLC. Penicillin G was detected in the udder of two animals euthanized at 24 h (mammary tissue and secretion) and at 48 h post treatment (mammary secretion only) after administration at concentrations still close to or above MIC90 values reported for the pathogens associated with heifer mastitis. Antibiotic concentration shortly after administration will have been substantially higher indicating a potential for systemic treatment with penethamate hydriodide to control prepartum intramammary infections in heifers without the disadvantages of local therapy such as teat contamination or risk of trauma for the administrator.

The oxidative process in milk fat, resulting in spontaneous oxidized off-flavour (SOF), is commonly assumed to depend on contents of pro- and antioxidants in milk and availability of fatty acids acting as their substrate. An important antioxidant in milk is α-tocopherol whereas the most potent prooxidant is the metal ion copper. The separate effects of α-tocopherol, copper, and milk fatty acid profile, and their combined effect on SOF development were examined in milk from 44 multiparous cows fed different roughage types and different amounts of dietary, unsaturated fat. A clear association between concentrations of copper and poly-unsaturated fatty acids in milk and the risk for developing SOF was found. Heritability estimates suggest that occurrence of SOF is partly under genetic control which indicates that milk quality may be compromised if breeding bulls are selected that carry genotypes predisposing for milk prone to develop SOF.

The aim of this study was to investigate the effects of unhydrolysed/intact casein and eight different sodium casein hydrolysates (a–h) on the viability and growth of human cancer cell lines. Both human Jurkat T cells and Caco-2 cells were incubated with increasing concentrations of the test compounds (0·5–10% v/v) for 24 h. Cell viability was assessed using the MTT, lactate dehydrogenase (LDH) release and Trypan Blue assays. Cell growth was monitored using the MTT, Trypan Blue and Bromodeoxyuridine (BrdU) proliferation assays. Casein hydrolysates b, c and f had an inhibitory effect on the viability and growth of both cell lines. The casein hydrolysates did not negatively affect the membrane integrity of both Jurkat and Caco-2 cells. In Jurkat cells hydrolysates a and h had an inhibitory effect on DNA synthesis after 24 h, while in Caco-2 cells DNA synthesis was not affected. In conclusion, we found that the different casein hydrolysates had cell-specific effects which target particular functions within the cell. Overall, casein hydrolysates had no effect on membrane integrity while they had varied effects on mitochondrial activity and DNA synthesis in the different cell lines.

Microscopic conformation, growth behaviour and freezing sensitivity of seven strains of Geotrichum candidum, a ripening starter, were studied and compared according to their macroscopic morphotypes. It has been shown that the thallus forming units (TFU)×ml−1/OD600nm ratio as a function of time is an interesting parameter to follow G. candidum sporulation through the growth behaviour. Microscopic conformation, growth behaviour and freezing sensitivity are clearly strain specific and mostly related to their corresponding morphotypes “yeast”, “mould” or “intermediate”. The two “mould” strains that sporulate weakly (UCMA103, UCMA499) showed a low survival rate to freezing stress whereas the “yeast” strains expressed a significant resistance owing to the arthrospore abundance. Interestingly, one strain (UCMA96) which appeared on solid medium in accord with the “mould” morphotype respond similarly to freezing stress.

Insulin-like growth factor 2 (IGF2) is considered to be a regulator of post-natal growth and differentiation of the mammary gland. In the present work, associations of two single nucleotide polymorphisms in the bovine IGF2 gene with milk production traits were studied in dairy Holstein-Friesian cows: the already described g.8656C>T transition in exon 2 (RFLP-BsrI) and the newly found g.24507G>T transversion in exon 10 (RFLP-HaeIII), found by sequencing 273-bp exon 10 of the IGF2 gene in six individuals. Associations were analysed individually and in combination with the multi-trait repeatability test-day animal model. The CT/GT haplotype appeared to be associated with most of the milk traits studied (differences were significant at P⩽0·001). The most frequent CT/GG haplotype seemed inferior to others in fat and protein content and daily yield of fat and protein but superior (together with the TT/GG genotype) when the daily milk yield is considered.

The stability of camel α-lactalbumin (α-la) against heat denaturation was measured, using circular dichroism (CD) and fluorescence spectroscopy, as well as differential scanning calorimetry (DSC). The experiments were performed in the presence of saturating concentrations of calcium as well as in the presence of EDTA, yielding to the apo form of α-la. The change in heat capacity (ΔCp) suggests a greater contribution of hydrophobic interactions to the stability of holo camel α-la than in its bovine counterpart. Overall the results obtained in this study suggest a greater stability of camel α-la than the bovine protein in both holo and apo states. Also CD experiments showed similar secondary structure for camel and bovine α-la and secondary structure of camel α-la was better preserved than that of bovine α-la during heat denaturation. The differences in thermal stability between the proteins from two species can be primarily ascribed to the difference in the quantity of hydrophobic interactions involved in their folding.

The combination of ultrasound and heat (thermoultrasound) is an emerging food preservation technique that retains higher quantities of bioactive components compared with current thermal pasteurization practice, but has not yet been assessed for pasteurizing human milk. Artificially contaminated human milk samples were treated with ultrasound (20 kHz, 150 watts) with and without heating. The retention of four human milk proteins was quantified by biochemical assay and laser scattering particle sizing was used to determine the extent of homogenization. While ultrasonic treatment was effective at inactivating Escherichia coli (D4 °C=5·94 min), Staphylococcus epidermidis exhibited resistance (D4 °C=16·01 min). Thermoultrasonic treatment was considerably more effective (Esch. coli D45 °C=1·74 min, D50 °C=0·89 min; Staph. epidermidis D45 °C=2·08 min, D50 °C=0·94 minutes) with a predicted retention (2·8 min treatment, 50°C) of secretory IgA lysozyme, lactoferrin and bile salt stimulated lipase of 91, 80, 77, and 45%, respectively. Homogenization of the milk samples occurred after 5 min and 2 min of ultrasonic and thermoultrasonic treatment, respectively. Thermoultrasonic treatment is an effective method for pasteurizing donor human milk and retaining a greater proportion of bioactive components compared with current practices. However, further studies are required to assess the practicality of applying this technique routinely to donor human milk.

This paper reports the results of a study that aimed to assess whether liver functionality defined by liver activity index (LAI) is associated with inflammatory and immune parameters in blood and milk. LAI is an index including the average blood levels of albumin, lipoproteins and retinol-binding protein measured three times in the first month of lactation (at 5, 15 and 30 days in milk). The aim was to assess the relationship of this index with blood and udder immune and inflammatory status as a means of identifying as early as possible cows at risk of disease. The research was carried out using 10 multiparous Italian-Friesian dairy cows of average genetic merit. Cows were retrospectively ranked in three groups according the LAI level. Blood samplings were performed at different intervals before and after calving; quarter milk samples were taken only after calving with the same schedule as blood samples. Leucocytes, oxidative burst, blood lysozyme and N-acetyl-β-d-glucosaminidase (NAGase) curves showed large overlapping among the three LAI group curves during the follow-up period. Four blood (complement, sialic acid, haptoglobin and reactive oxygen metabolites) and three milk (somatic cell count, lysozyme and NAGase) parameters showed larger and more consistent differences among LAI groups. Complement showed higher values and sialic acid showed lower values in high LAI group when compared with the other two LAI groups. Two other markers of inflammatory status (haptoglobin and reactive oxygen metabolites) showed the lowest values in high LAI cows. A consistent and significant reduction of milk NAGase and milk lysozyme in high LAI group was observed. The results suggest that cows with the highest liver functionality index have also the highest levels of some immune markers and the lowest levels for inflammatory markers at blood (already before calving) and mammary levels. Finally, cows with low LAI index, being more susceptible to metabolic and infectious diseases, should be carefully monitored to identify as early as possible the development of a disease.

Commonly measured metabolite and hormone concentrations used to describe the metabolic status of lactating cows undergo diurnal variation resulting in distinct patterns. Studies have shown that feeding events can modulate these diurnal patterns as cows respond to the nutrient intake. What is less clear is the extent to which cow genetics and diet interact to modify the diurnal patterns of specific nutritionally related metabolites and hormones. The objective of this study was to investigate diurnal patterns in circulating metabolite and hormone concentrations in divergent strains of Holstein-Friesian cows (North American, NA; and New Zealand, NZ) offered either fresh pasture (FP) or a total mixed ration (TMR). Plasma concentrations of growth hormone (GH), insulin-like growth factor-1 (IGF-1), glucose, insulin, β-hydroxybutyrate (BHBA), and non-esterified fatty acids (NEFA) were determined at 4-h intervals for a minimum of three consecutive days. All of the above metabolites and hormones exhibited within-day variability. Main effects of genetic strain and diet were observed for plasma IGF-1, and a strain by diet interaction was observed for GH. Time of day×diet interactions were observed for both glucose and insulin. Three-way interactions (time of day×diet×strain) were observed for BHBA and NEFA. These data indicate different levels of diurnal variation, with glucose, insulin, NEFA and BHBA having the largest daily variation. These diurnal patterns need to be considered in future investigations of these metabolites and hormones.

The effect was quantified of coagulase-negative staphylococci (CNS) intramammary infections on quarter- and cow-level somatic cell count (SCC) and on bulk milk somatic cell count (BMSCC) in different BMSCC cohorts in Dutch dairy herds. Two datasets were used for this purpose. In the first dataset, on 49 randomly selected dairy farms a total of 4220 quarter milk samples of 1072 cows were collected of all cows and heifers with a test-day SCC ⩾250 000 and ⩾150 000 cells/ml, respectively, and of 25% of cows and heifers below these thresholds. In the second dataset, on 39 selected dairy farms a total of 8329 quarter milk samples of 2115 cows were collected of all cows with a test-day SCC ⩾250 000 cells/ml following two consecutive SCC <250 000 cells/ml, and of heifers using the same SCC criteria but with a threshold of 150 000 cells/ml. These cows and heifers were defined as new high SCC. In both datasets, CNS was the most frequently isolated pathogen, 11% in the first dataset and 12% in the second dataset. In both datasets, quarters with CNS IMI had a lower SCC than quarters infected with major pathogens, and a higher SCC than culture-negative quarters. The same was found for SCC at cow level. Coagulase-negative staphylococci were more often found in quarters with SCC ⩾200 000 cells/ml in dairy farms with a BMSCC <150 000 cells/ml compared with dairy farms with a higher BMSCC. Prevalence of CNS in cows and heifers with a high SCC was higher in dairy farms with a BMSCC <150 000 cells/ml compared with dairy farms with a medium or high BMSCC: 30, 19 and 18%, respectively. This indicates that CNS IMI as a cause of subclinical mastitis is relatively more important in dairy farms with a low BMSCC and may become a point of attention in udder health management on that type of farm.

Hormone-sensitive lipase (LIPE) plays a fundamental role in the regulation of energy balance by releasing free fatty acids from adipose triacylglycerol stores. These fatty acids can be subsequently transferred to other body compartments to be oxidized or employed in other biochemical reactions. This enzymic function is particularly important in lactating animals because the synthesis of milk components involves the mobilization of lipid depots to satisfy the large energy demands of the mammary gland. In the current study, we partially sequenced the goat LIPE gene in several individuals. In doing so, we identified two synonymous polymorphisms at exons 2 (c.327C>A>T, triallelic polymorphism) and 3 (c.558C>T). Moreover, we found a mis-sense polymorphism at exon 6 (c.1162G>T) that involves an alanine to serine substitution at position 388. Analysis with Polyphen and Panther softwares revealed that this amino acid replacement is expected to be neutral. Performance of an association analysis with a variety of milk traits revealed that goat LIPE genotype has highly suggestive effects on milk yield (P=0·0032) as well as on C18:3 n-6g (P=0·0051), trans-10 cis-12 CLA (P=0·007) and C12:0 (P=0·0084) milk contents. These associations are concordant with the preference of LIPE to selectively mobilize medium-chain and unsaturated fatty acids.

This study explains how active coating can serve to prolong the shelf life of Fior di latte cheese. The active coating was prepared by dissolving, in two sodium alginic acid solutions (5 and 8% w/v), different concentrations of lysozyme (0·25, 0·50 and 1·00 mg ml−1)+50 mm of Ethylene-Diamine Tetraacetic Acid (EDTA). Samples of Fior di latte cheese packaged in brine and active brine (lysozyme+EDTA, at the above concentrations) were also used as controls. The quality decay of the Fior di latte cheese stored at 10°C was assessed by monitoring the viable cell concentration of the main spoilage microorganism, as well as its sensory quality (i.e., external appearance, consistency, colour and flavour). The concentration of rod-or coccus-shaped Lactic Acid Bacteria (LAB) was also monitored to assess the effect of the proposed packaging strategies on the flora type of Fior di latte cheese. The results show that an increase in the shelf life equal to 104% was recorded for the coated samples, compared with controls packaged in brine without active compounds. This shelf life increase is slightly lower than that recorded with samples packaged in the active brine (151%), as a result of a more pronounced microbial proliferation; however, the coating could be a better packaging solution for the reduced weight of tray.

The primary objective of this study is to validate a new fast method for determination of uric acid in milk. The method is based on an enzymatic-fluorometric technique that requires minimal pre-treatment of milk samples. The present determination of uric acid is based on the enzymatic oxidation of uric acid to 5-hydroxyisourate via uricase where the liberated hydrogen peroxide reacts with 10-acetyl-3,7-dihydroxyphenoxazine via peroxidase and the fluorescent product, resorufin, is measured fluorometrically. Fresh composite milk samples (n=1,072) were collected from both Jersey (n=38) and Danish Holstein (n=106) cows from one local herd. The average inter- and intra-assay variations were 7·1% and 3·0%, respectively. Percent recovery averaged 103·4, 107·0 and 107·5% for samples spiked with 20, 40 or 60 μm of standard, respectively, with a correlation (r=0·98; P<0·001) observed between the observed and expected uric acid concentrations. A positive correlation (r=0·96; P<0·001) was observed between uric acid concentrations using the present method and a reference assay. Storage at 4°C for 24 h resulted in lower (P<0·01) uric acid concentrations in milk when compared with no storage or samples stored at −18°C for 24 h. Addition of either allopurinol (a xanthine oxidase inhibitor) or dimethylsulfoxide (a solvent for allopurinol) did not affect milk uric acid concentrations (P=0·96) and may indicate that heat treatment before storage and analysis was sufficient to degrade xanthine oxidase activity in milk. No relationship was observed between milk uric acid and milk yield and milk components. Authors recommend a single heat treatment (82°C for 10 min) followed by either an immediate analysis of fresh milk samples or storage at −18°C until further analysis.

The MAC-T cell line has been used extensively to investigate bovine mammary epithelial cell function. A lactogenic phenotype is generally induced in this cell line by a combination of dexamethasone, insulin and prolactin and has typically been assessed by milk protein production. Few studies have focused on identifying other factors that may affect milk protein synthesis in the MAC-T cell line, and none have considered the lipid class distribution of MAC-T cells as a component of the lactogenic phenotype. Growth hormone (GH) has been shown to increase milk protein synthesis both in vivo and in mammary cell models, and has been shown to alter the lipogenic profile of mammary explant models. We tested the hypothesis that MAC-T cells would respond directly to GH and that the response would include alterations to the lipid class distribution as well as to milk protein gene expression, leading to a more appropriate model for mammary cell function than treatment with dexamethasone, insulin and prolactin alone. Differentiated cells expressed GH receptor mRNA, and addition of GH to the differentiation medium significantly induced production of α-s1 casein and α-lactalbumin mRNA. GH also significantly affected the proportion of triacylglycerol and sphingomyelin. These results indicate that GH is an important factor in inducing a lactogenic phenotype in the MAC-T cell line, and support the possibility of a direct effect of GH on milk synthesis in vivo.

This study aimed to reduce the time taken to detect low numbers of Cronobacter sakazakii inoculated onto environmental swabs (100, 10 or 1 cfu per swab) using a simple plating procedure for application in a dairy testing laboratory. Three types of environmental swabs (Biolab FlexiSwab™, gauze swabs and the Whatman SwabCheck Polywipe™ sponge) were inoculated with either Cron. sakazakii in single culture or Cron. sakazakii together with Citrobacter freundii. A 5-h enrichment procedure of swabs in Cronobacter enrichment broth at 37°C prior to plating was then compared with no enrichment or 24-h enrichment. The 5-h enrichment procedure was as efficient at detecting Cron. sakazakii on environmental swabs at low cell concentrations (100 cfu per swab), and in pure culture or in competition with other coliforms (Citrobacter), as pre-enrichment for 24 h. This protocol was also successful in detecting 10 cfu per swab 80% of the time. The results also indicated that the type of swab selected for use in environmental safety programmes is influential on the outcome, with the FlexiSwab™ and gauze swabs being the most efficient swabbing materials evaluated in this study.

Emulsions were prepared with 20% soy oil and different concentrations of lactoferrin, and tested at pH values from 3 to 7·5. The stability of the emulsions decreased as the pH got closer to the isoelectric point of the protein. A concentration of 1% lactoferrin was determined to be sufficient to provide full coverage of the emulsion droplets. Lactoferrin-stabilized emulsions were then prepared in water at pH 6·6 and their behaviour when added to reconstituted milk was studied. It was observed that lactoferrin emulsions were stable when reconstituted in milk, but they showed aggregation when diluted in milk serum alone. The destabilization was caused by shielding of the charges on the surface of the oil droplets. Stabilization in milk occurred due to interactions at the interface with other soluble proteins. In fact, when β-lactoglobulin or sodium caseinate were added to the serum, stability of the emulsion droplets was restored, indicating that these proteins were able to adsorb at the interface and aid in the stabilization. Since ζ-potential measurements did not show significant overall charge on the emulsion droplets, this suggests that the stabilization forces are not only electrostatic in nature, but that there are other mechanisms at play.

Casein genes have been proved to have an influence on milk properties, and are in addition appropriate for phylogeny studies. A large number of casein polymorphisms exist in goats, making their analysis quite complex. The four casein loci were analyzed by molecular techniques for genetic polymorphism detection in the two dairy goat breeds Bunte Deutsche Edelziege (BDE; n=96), Weiße Deutsche Edelziege (WDE; n=91), and the meat goat breed Buren (n=75). Of the 35 analyzed alleles, 18 were found in BDE, and 17 in Buren goats and WDE. In addition, a new allele was identified at the CSN1S1 locus in the BDE, showing a frequency of 0·05. This variant, named CSN1S1*A′, is characterized by a t→c transversion in intron 9. Linkage disequilibrium was found at the casein haplotype in all three breeds. A total of 30 haplotypes showed frequencies higher than 0·01. In the Buren breed only one haplotype showed a frequency higher than 0·1. The ancestral haplotype B-A-A-B (in the order: CSN1S1-CSN2-CSN1S2-CSN3) occurred in all three breeds, showing a very high frequency (>0·8) in the Buren.
