The present paper is aimed to celebrate Franz J. Kallmann's memory, by illustrating the twin-family method, which he developed. His phenomenal powers of organization are testified to by his extensive samples. Their representative nature has the built-in check of the ratio of MZ: DZ twins. He brought a refinement of his own to the existing methods of calculating expectancy from nett figures, that permits comparison of studies of varying age distributions relative to manifestation period. His investigative design allows not only for the standard global concordance comparisons between DZ and MZ twins as well as for those reared together and apart, but exploits the implications of the hitherto neglected comparison of the categories of sibs and DZ twins also. Further, his combined twin-family method comprehends in its extended range the powerful analytical tool of comparative figures for the series step-sibs, half-sibs, sibs, DZ twins and MZ twins (Kallmann and Sander, 1947; Kallmann, 1953). Kallmann (1954a, 1954b) has moreover given minute attention to controversies concerning legitimate inferences from twin studies, and has drawn attention to a point made by Darlington (1953), which is often overlooked by the best of geneticists. Discordance between MZ twins is not a measure merely of postnatal or even of prenatal environmental effects: it may also have a genetic component through the action of genes sensitive to cytoplasmic asymmetry.

The biometrical genetical method of analysis of continuously varying human traits is described and shown to unify a number of common approaches to the analysis of twin data. The method, which is based on the analysis of variance, goes beyond alternative approaches by providing tests of basic assumptions and by investigating the possibility of assortative mating and dominant gene action.

Twins form an indispensable group for the application of the biometrical genetical method but the value of including other groups, often more easily collected, is demonstrated.

The techniques are illustrated through the reanalysis of data on the behavioural measures, neuroticism and intelligence quotient (I.Q.). For both measures fundamental assumptions are met. The results suggest the absence of dominant gene action for neuroticism, indicating an evolutionary history of stabilising selection, and directional dominance for high I.Q., indicating an evolutionary history of strong directional selection. Both these findings seem reasonable in view of psychological knowledge concerning the two measures.

The theoretical relationship between the distribution of a given trait in a population of twin pairs and several genetic parameters has been examined. In particular, a series of mathematical models has been worked out, that, when applied to a twin population, nonselected for the occurrence of a given trait and nondiagnosed as to zygosity, leads to an estimate of:

1) The MZ: DZ ratio in the population;

2) The frequency of the genotype responsible for a given trait;

3) The probability of manifestation of the trait;

4) The value of epistatic factors.

A further mathematical model affords the estimate of linkage in the hypothesis of simultaneous recording of more than one trait.

In ogni paese si annette una rilevante importanza alla disponibilità di gemelli MZ, nelle specie normalmente unipare, per la ricerca sperimentale di base e dimostrativa.

In zootecnia, la specie più interessante a questo riguardo è quella bovina. Le ricerche attuate nei laboratori dei vari paesi riguardano: (1) l'utilizzazione e conversione di diversi alimenti e sistemi di razionamento; (2) l'accrescimento diario in p.v.; (3) il rendimento quanti-qualitativo di prodotto (latte, carne, grasso); (4) la precocità ed il comportamento sessuale (produzione spermatica ed ovulazione); (5) il comportamento alle variazioni di determinati fattori ambientali (luminosità, temperatura, ventosità).

With the exception of disorders related to gestation and birth, twins are usually found to be medically representative of the general population; diseases occur among twins with the same frequency as among singletons. As a corollary, in a large series of twins, if members of each pair are assigned randomly to two groups, A and B, both groups will be representative of the population. For example, if a disease occurs in 1% of the population, then we can expect it in 1% of twin-group A and in 1% of twin-group B.

Tab. I diagrams a hypothetical population of 15000 twin pairs in which 1% of all persons have the disease in question. Twins A include, in the left column, 150 sick persons. Each of these has a partner, twin B, who may be either sick or well, and so the total number of pairs in the left column is 150. The figures are the same when the table is read down or across; that is, the table is symmetrical with respect to twins A and twins B because assignment to A and B was random.

We present the results of a noncentral χ2 test applied to 63 like-sexed twin pairs.

We have separated the pairs into two groups: 30 serologically identical and 33 serologically nonidentical pairs for nine sera of the blood groups ABO, MN, and Rh.

The probabilities of monozygosity of the serologically identical pairs, calculated by the method of Maynard-Smith and Penrose, range from 0.83 to 0.92 — rather small values. With the help of Belgian frequencies of DZ and MZ twins, we could prove that there are about 50% chances of having less than two DZ pairs amongst the 30 serologically identicals.

Consequently, our first group with 30 pairs is mostly MZ (less than 7 DZ pairs in 96% of the cases); while our second group, with 33 pairs, is completely DZ.

We did not try to extract out of the 30 serologically identicals, such pairs that might “seem” DZ. For instance, we have kept in our first group two twins — one with blue and one with brown eyes — because the genetics of eye colour is not yet enough well-known to be safely used in twin-diagnosis; and also in memory of Madame de Sévigné, who had one brown and one blue eye. Imperfect as the composition of our sample is, we have avoided at least one mistake: adding, to surely DZ pairs, twins who are “seemingly” DZ, but not proven to be so.

I nostri studi ed esami radiografici si riferiscono alle ossa delle mani e dei piedi, del cranio, della gabbia toracica e della colonna vertebrale, e agli organi interni del torace.

I nostri esami hanno dimostrato che la forma delle ossa delle mani e dei piedi nei gemelli MZ presenta sempre netta analogia, mentre nei gemelli DZ presenta delle differenze. L'analogia nella forma delle ossa si riferisce pure ai centri di ossificazione delle epifisi, alla presenza di epifisi accessorie e di pseudoepifisi.

Per quanto riguarda le dimensioni, è stato constatato che più sovente esse si distinguono, specialmente nei gemelli più adulti, a causa del differente sviluppo dei gemelli sotto l'azione dei fattori esterni. Gli esami radiometrici dello scheletro delle mani e dei piedi hanno dimostrato che le differenze omolaterali nella misura delle ossa omonime sono minori nei MZ rispetto ai DZ, e che il confronto eterolaterale fra le dimensioni delle ossa omonime della mano sinistra e della mano destra dimostra la presenza di differenze a carico della mano destra, essendo questa funzionalmente più operosa della sinistra. Gli esami radiometrici permettono di accertare in modo più esatto la concordanza e la discordanza delle dimensioni delle ossa dei due gemelli.

Recent years have witnessed a growing interest in dermatoglyphics: an increasing number of associations between dermatoglyphic alterations and different diseases (especially chromosomal aberrations) are actually being reported, which is of particular concern to human and medical geneticists, as well as to clinicians and pediatricians.

Having been shown to undergo practically complete genetic conditioning, fingerprints are also being applied, by different authors, to the diagnosis of zygosity in twins.

On the other hand, although, on account of available data, a large impact of heredity may be assumed in their determination, dermatoglyphics as a whole still fail to find a clear genetic definition.

Furthermore, with respect to clinical aspects and applications, a clear quantitative definition of dermatoglyphic alterations, and therefore of dermatoglyphic normality and its variability, is still needed.

Last, but not least, a wider and generalized application of dermatoglyphics to the diagnosis of zygosity in twins would require a clear, morphological and genetic, quantitative definition of each trait, as well as the introduction of simple and standardized statistical methods for calculating the probabilities of monozygosity and dizygosity.

Les résultats statistiques sont exacts dans la mesure où nous disposons de séries gémellaires MZ et DZ non sélectionnées. Il est nécessaire que cette condition soit réalisée, si l'on veut déterminer la probabilité de manifestation d'une tare.

D'autre part, pour les maladies dont l'âge d'apparition est très variable (diabète sucré, schizophrénie, folie circulaire), un certain nombre de jumeaux sont considérés comme normaux, alors qu'ils n'ont peut-être pas encore atteint l'âge de manifestation. Pour ces cas, nous devons utiliser une méthode correctrice, tenant compte de l'âge. Certaines séries gémellaires de nos statistiques n'ont pas subi cette correction, et de ce fait les valeurs peuvent être plus ou moins faussées.

Il est en outre souvent difficile de différencier l'hérédité polygénique de l'hérédité monofactorielle à pénétrance très incomplète du gène responsable. Penrose (1953) a démontré qu'il est illusoire de vouloir, sur la base des données familiales, trancher en faveur de l'un ou l'autre de ces modes d'hérédité, lorsque la fréquence du trait ou de l'anomalie chez les parents et leurs enfants est moins de dix fois supérieure à la fréquence observée dans l'ensemble de la population.

Soixante-douze paires de jumeaux de même sexe, âgés de 6 à 22 ans, ont été examinées à Bruxelles par l'équipe du Prof. Twiesselmann. Ils ont été séparés en deux groupes, 35 paires de concordants et 37 paires de discordants pour au moins un des neuf sérums des groupes ABO, MN et Rh. Aucun élément subjectif n'est donc intervenu dans cette ségrégation basée sur des facteurs dont le mode de transmission est connu. Nous n'avons fait aucun essai de diagnostic de zygotie basé sur la plus ou moins grande similitude de caractères morphologiques, de façon à rester dans un domaine absolument objectif. Le groupe des discordants est évidemment composé uniquement de jumeaux DZ. Celui des concordants comprend tous les MZ plus vraisemblablement quelques paires de DZ.

Pour l'étude des dermatoglyphes, nous avons retenu trois caractères: le nombre total de triradius digitaux (pattern intensity index), l'indice des lignes principales (main line index) et le nombre de crêtes entre les triradius interdigitaux a et b (a-b ridge-count).

Nous avons d'abord étudié les corrélations entre jumeaux d'une même paire (Fig. 1). Dans le groupe des discordants sérologiques, les intervalles de confiance des coefficients de corrélation englobent la valeur théorique de 0.5. Chez les concordants, ils approchent de l'unité et les coefficients de corrélation vont de 0.83 à 0.85. Aucun de ces intervalles de confiance ne se chevauche, malgré leur étalement dû au petit nombre de sujets.

È noto come l'individualità delle risposte alla degustazione della paraetossifenil-tiocarbamide (PTC) poggi su base ereditaria, risultando nulla l'influenza dei fattori ambientali. Sono meno chiari, invece, i meccanismi che comportano, sia pure eccezionalmente, una discordanza in gemelli MZ.

Nell'ambito di più ampie ricerche, si è voluto quindi rilevare:

1) La frequenza, in una data popolazione di gemelli, del fenomeno della discordanza;

2) Se la discordanza non rappresenti piuttosto un'elevata differenza della soglia della sensibilità;

3) Se, nei casi di discordanza, venga rilevata l'interferenza di fattori neurologici o psichici.

Le ricerche hanno interessato 110 coppie di gemelli MZ: 80 coppie di soggetti “normali” e 30 coppie in cui uno o entrambi i soggetti erano sofferenti per gravi disturbi organici o psichici.

I soggetti sono stati esaminati almeno nove volte, nel corso di tre o più anni, con somministrazioni per carta e per goccia di soluzioni di sostanza in concentrazione scalare da 0.02% a 0.90%. Parallelamente, è stato tenuto conto di tutte le varianti ambientali (fattori fisici e psichici) di possibile rilievo, agenti su uno o su entrambi i soggetti.

Nella popolazione di gemelli «normali» si è osservato il 100% di concordanza per il carattere gustatore-non gustatore e, tra i gustatori, il 100% di concordanza nella determinazione dei diversi sapori (amaro, acido, dolce, salato, gusto vario). Ogni gustatore mostra, alle diverse prove, un'oscillazione della propria soglia di sensibilità, spesso in relazione con varianti biologiche apprezzabili (p. es., un episodio febbrile acuto, una grave sofferenza epatica di origine tossica o infettiva); ma nel 100% dei casi lo spostamento in più o in meno dei valori di soglia tende a tornare ai valori di base non appena vengono a mancare le particolari condizioni ambientali che lo hanno sostenuto. Inoltre, i limiti estremi di questi spostamenti sono parsi strettamente individuali con una concordanza intracoppia del 100% dei casi — rilievo che acquista maggiore significato se raffrontato al 20% di concordanza riscontrato nei gemelli DZ.

I problemi eziopatogenetici riguardanti la deficienza marginale (cioè, il ritardo mentale di lieve grado: Q.I. 70-90) sono stati studiati attraverso l'osservazione di 40 coppie gemellari (20 MZ e 20 DZ) di uguale sesso, di 7-18 anni, vissuti sempre, o solo temporaneamente, in ambienti identici o differenti, presentanti entrambi un ritardo mentale di lieve grado ad eziopatogenesi sconosciuta.

Sono state raccolte 80 schede riportanti in ogni dettaglio le inchieste anamnestiche, socio-familiari e scolastiche, gli esami clinici obiettivi e di laboratorio, generici e neuro-psichiatrici.

1. Q. I. Concordanza o lieve discordanza nel 100% dei MZ, mentre nei DZ non viene rilevata in alcun caso netta concordanza: discordanze lievi nel 15% e marcate nell'85% dei casi. Che i soggetti abbiano vissuto sempre, o solo temporaneamente, in ambienti identici o differenti per condizioni di vita e di educazione, non risulta di rilevante significato.

2. Fenomenologia del disturbo. Il deficit è nel 100% dei casi di tipo settoriale. Le sue espressioni fenomenologiche nei MZ risultano perfettamente concordanti nel 65% dei casi, lievemente discordanti nel 20% e piuttosto discordanti nel 15%; mentre nei DZ portatori entrambi dell'anomalia, non viene rilevato alcun caso di netta concordanza: discordanze lievi nel 30%, marcate nel 70% dei casi. Che i soggetti abbiano vissuto sempre, o solo temporaneamente, in ambienti identici o del tutto differenti, non risulta di rilevante significato.

Il problema del disadattamento sociale, nelle sue diverse forme e gravità, è stato considerato attraverso l'osservazione di 60 coppie di gemelli: 30 MZ e 30 DZ, ♂♂ e ♀♀, dai 15 ai 55 anni, vissuti rispettivamente in ambienti identici o differenti per tutta la vita o solo nell'età infantile-giovanile, oppure nell'età adulta. Sono state escluse, dall'osservazione le coppie di soggetti affetti da sofferenze organiche, neurologiche e psichiche, tali da giustificare anomalie del comportamento e quindi disadattamento sociale. I casi, trovati nel corso di 25 anni, sono stati studiati attraverso inchieste socio-familiari ed esami obiettivi clinici e di laboratorio, generici e specialistici, particolarmente quelli propri della clinica neuro-psichiatrica.

Le inchieste sono state condotte per tutte le età ed ogni ambiente, per raccogliere informazioni attendibili ed obiettive che investissero l'intera vita dei soggetti, contribuendo alla comprensione psicodinamica delle anomalie comportamentali ed in particolare del disadattamento sociale. Gli esami obiettivi e collaterali psichiatrici (colloqui psicodiagnostici e test psicoproiettivi), più volte ripetuti a distanza anche di anni, hanno permesso di precisare i singoli quadri clinici ed hanno fornito gli elementi per la loro interpretazione.

In tutti i gemelli MZ vissuti in ambienti identici (10 coppie), sono riscontrabili rilevanti concordanze. In quelli vissuti in ambienti sostanzialmente differenti (4 coppie), non è stato rilevato alcun caso di perfetta concordanza: concordanze parziali in 2 coppie e marcate discordanze nelle altre 2.

L'hermaphrodisme vrai est une forme relativement rare d'état intersexué.

L'étude chromosomique, dans les cas de ce genre, montre habituellement soit des caryotypes normaux XX ou XY, soit des mosaïques chromosomiques, par exemple XX/XO. Ces mosaïques sont explicables d'une façon simple par les mécanismes généralement invoqués: non-disjonction gamétique, ou bien perte et non-disjonction chromatidienne, lors des premières mitoses du zygote.

Nous avons eu l'occasion d'observer un sujet de 20 ans atteint d'un hermaphrodisme vrai. Elevé comme un garçon, son aspect était ambigu: les poils du visage étaient rares, la pilosité pubienne de type féminin, la forme du bassin féminine, elle aussi; il existait une gynécomastie, la verge était de très petite dimension et les bourses étaient vides.

Une laparotomie exploratrice a montré la présence de tissu testiculaire et ovarien dans les gonades, ce qu'ont démontré les examens histologiques (Prof. Nezelof). A gauche, la gonade était constituée exclusivement de tissu ovarien. A droite, il s'agissait d'une glande mixte, d'un ovotestis. Les coupes histologiques ont décelé, en effet, à côté de tissu ovarien et de formations annexielles, des tubes séminifères et une formation épididymaire.

L'examen des frottis buccaux a montré l'existence de masses chromatiniennes dans 20% des noyaux cellulaires.

The authors have studied the variations of the mitotic index and the association index of the acrocentric chromosomes (groups D and G) in eight male twin pairs of assorted age (6 and 60 years) and zygosity. For each individual, two subcultures of peripheral blood lymphocytes have been set up by the standard techniques. After colchicine administration and hypotonic shock, two slides per culture have been prepared and stained with lactoacetic orcein, thus providing the experimental material. The study of variability between individuals, between tests and between age groups indicates the importance of the age factor for the assessment of both indexes.

The study of the variability of intrapair differences indicates that the phenomena are both statistically and dynamically genotype-dependent. The weight of heredity, in the sum of all the factors affecting the phenomena, is 0.79 for the mitotic index and 0.51 for the association index.

One of our patients with chronic myelocytic leukemia has a twin brother. The twins, 31 years old, show similar physical appearance. Both have blue eyes, brown hair and are right-handed. Anthropometric measurements and fingerprint patterns are matching. Blood group antigens, serum factors and PTC tasting are identical. Blood group antigens of the parents have also been determined. It has been calculated that the probability that the twins are MZ is higher than 99.8%.

The twins were the result of an uncomplicated pregnancy. There are no other siblings. No family history of consanguinity, hematological disorders or cancer was given. They spent their childhood on a farm. Both have not been exposed to drugs, benzene or radiological procedures, apart from routine X-ray films of the chest.

Our patient, who is still working on his farm, had been referred to a hospital in June 1959, because of a knife wound of the chest with a myocardial laceration and pericardial tamponade. White blood cell count at that time was 22300 with 3% basophils.

In April 1962, the diagnosis of chronic myelocytic leukemia was established. Findings included hepatosplenomegaly, leukocytosis with a marked shift to the left, hyperplasia of granulopoiesis in the bone marrow and diminished leukocyte alkaline phosphatase activity. In the following years the patient was treated with cyclophosphamide, splenic irradiation and busulfan. Busulfan is continued at present at low doses.

In the course of routine work in the caryological laboratory at the Mendel Institute, we have often noticed the repetition in families, and especially in MZ twins, of alterations in chromosome morphology that can hardly be included among chromosomal aberrations proper (translocations, deletions, etc.). Since such variations do not generally reflect any pathology in the individuals involved, the occurrence of a normal morphological variability of chromosomes has been postulated.

In order to test this hypothesis, we have measured several clearly defined and standardized chromosomal parameters, obtaining the classical structure of the twin test, i.e., the comparison between experimental measures in MZ and DZ twin pairs.

The parameter that was selected for the test is the centromeric index of Group A and Group B chromosomes, intended as the ratio of long arm length to short arm length. In order to avoid method and operator influences, a pilot test was carried out, repeating five times the culture from blood of the same individual, and having the slides read by five different operators; thus, the variability due to the method and to the operator was assessed.

Nous avons eu l'occasion d'examiner une paire de jumelles MZ, apparemment saines, provenant d'un père chez lequel s'étaient associées deux affections: maladie ulcéreuse et hypertension artérielle. Dans ce même symposium, nous présentons en détail les résultats des explorations cliniques concernant la réactivité vasculaire et de sécrétion gastrique chez ces deux jumelles, remarquant que, pendant que l'une des jumelles (K.G.) hérite la réactivité de type hypertensif, l'autre hérite une réactivité de type ulcéreux (K.M.).

Cette constatation nous a déterminé à effectuer une analyse caryologique, afin de dépister les éventuelles modifications chromosomiales, ainsi que leur mode de représentation chez les trois sujets: le père et les deux sœurs jumelles.

Les examens caryologiques furent effectués sur des cellules lymphocytaires, prélevées du sang périphérique et cultivées sur Difco 199, selon la méthode de Moorhead. Les plaques métaphasiques, colorées au Giemsa, furent examinées au microscope à immersion, ainsi que sur des microphotographies. Pour chaque cas à part, l'on a effectué des idéogrammes de 18-25 plaques métaphasiques qui furent ensuite analysées et comparées l'une à l'autre.

L'analyse quantitative des caryogrammes n'a révélé aucune aberration de nombre chez les trois sujets examinés.

Le monozygotisme hétérocaryote est une gémellité pathologique par trouble de la ségrégation chromosomique. Elle fut découverte en 1961 (Turpin et al, 1961), quand l'analyse d'un couple, garçon normal — fille typique de syndrome de Turner, prouva que ces jumeaux, malgré leurs caryotypes différents et leurs dissemblances consécutives, étaient MZ. Depuis, de nouveaux exemples de monozygotisme pathologique ont été publiés, caractéristiques de types nouveaux.

Le couple qui révéla ce monozygotisme anormal, jusqu'alors insoupçonné, était composé d'un garçon et d'une fille âgés de 17 ans et issus d'une grossesse monochoriale, d'après l'aspect macroscopique du placenta.

Le garçon était normal: 56 kg; 165 cm; QI 125.

La fille était anormale: 40 kg; 142 cm; QI 106 (difficultés scolaires).

En dépit de son âge, elle n'avait jamais été réglée. Allant de pair avec cette aménorrhée, l'on notait l'absence des caractères sexuels secondaires sans malformations des organes génitaux externes. Par contre, certaines dystrophies évoquaient un syndrome de Turner: pterygium colli, cubitus valgus. Une laparotomie révéla un développement utéro-tubaire normal, mais l'absence des gonades droite et gauche. Le taux des gonadostimulines dépassait 100 u S/24h. L'étude de cellules jugales ou de culture (fascia lata) en interphase, ne décela aucun corpuscule chromatinien nucléaire.

Deux cousines au 4ème degré de la mère de ce couple avaient eu elles aussi des jumeaux, l'une garçon et fille, l'autre de type ignoré.

È stato più volte segnalato che nelle fratrie di pazienti affette da disgenesia gonadica si verificano parti multipli in numero più elevato del normale. Nelle casistiche riportate (Boyer et al, 1961; Lindsten, 1963; Nance e Uchida, 1964), l'incidenza dei parti gemellari oscilla dal 25% al 63%; considerando globalmente tutti i casi degli autori citati (524 nascite), la frequenza risulta del 32%. Un'elevata incidenza di parti gemellari è segnalata anche fra ascendenti e collaterali delle pazienti.

Il fenomeno viene attribuito a fattori, probabilmente in parte ereditari, che possano provocare un'instabilità nei meccanismi che regolano le divisioni cellulari, tan to nella gametogenesi che nell'embriogenesi (Root et al, 1964). Si deve tuttavia segnalare che nelle fratrie dei pazienti con sindrome di Down, nonostante un errore meiotico nell'ovogenesi e l'età materna più elevata della media, non si osserva un numero di gemelli superiore al normale (∅ster, 1953; Allen e Baroff, 1955).

il quadro clinico e quello cariologico delle disgenesie gonadiche con fenotipo femminile sono alquanto variabili (Giusti et al, 1966). Si possono distinguere: (1) sindrome di Turner tipica, con bassa statura, quadro malformativo completo, compreso il tipico pterigium colli; (2) sindrome di Turner attenuata, con quadro malformativo attenuato, generalmente senza pterigium; (3) casi senza alcuna malformazione, eccettuata la bassa statura; (4) casi con disgenesia gonadica pura che presentano un aspetto somatico completamente regolare, salvo Pinfantilismo. Nella sindrome di Turner tipica il cariogramma rivela quasi sempre il caratteristico ipoploidismo XO; nei casi di disgenesia gonadica pura il cariotipo è molto spesso normale; negli altri casi il quadro è variabile: fra i soggetti con mosaicismo di tipo XO/XX prevalgono quadri di sindrome di Turner attenuata; fra quelli di tipo XO/XY sono abbastanza frequenti le neoplasie gonadiche del tipo del gonadoblastoma.
