The results from analyses of botanical remains (pollen, wood, charcoal, seeds) from several archaeological features excavated in Kluizen (northern Belgium) are presented. The region was largely uninhabited until the Iron Age and Roman period when a rural settlement was established, resulting in small-scale woodland clearance. The site was subsequently abandoned from c. AD 270 till the High Middle Ages. The results of the archaeological and archaeobotanical analyses provide information on changes in land use and resulting dynamics of woodland cover and composition between c.600 BC and AD 1200, with a spatial and temporal resolution unrivalled in northern Belgium. Especially the long period of woodland regeneration following abandonment of the site around AD 270, covering the Late Roman and Early Medieval period, could be reconstructed in detail. Abandoned fields were first covered with pioneer woodland (Salix, Corylus and Betula), then Quercus-dominated secondary forest and finally a late-successional forest with Fagus sylvatica, Carpinus betulus and Ilex aquifolium, an evolution that took over 300 years. The results also indicate that the observed increase of Fagus during the Early Middle Ages, which was never an important element in the woodland vegetation in northern Belgium before, was related to climatic changes rather than anthropogenic factors.

Substances that could play a role in maintaining seed viability (phenolic compounds, α-tocopherol, sterols, ascorbic acid, glutathione and soluble proteins) were estimated in beech (Fagus sylvatica L.) seed lots that had been stored for 2, 5, 7 and 10 years at − 10°C. Germination capacity was strongly and positively correlated with amounts of total phenolic compounds, ultraviolet (UV)-absorbing phenols and soluble proteins. Moderately strong relationships appeared between germination capacity and α-tocopherol and ascorbic acid contents. Total sterol and glutathione contents were not correlated with germination capacity. A strong, negative correlation was found between germination capacity and reactive oxygen species (ROS), such as superoxide radical () and hydrogen peroxide (H2O2), as well as with lipid hydroxyperoxides (LHPOs). The putative role of these compounds in the maintenance of beech seed viability during long-term storage is discussed.

The seed storage behaviour of Fagus sylvatica and F. crenata was investigated. A large fraction of seeds of both species survived desiccation to about 3% moisture content (MC) (in equilibrium with 10% relative humidity at 20°C). Nevertheless, viability was reduced significantly and progressively by desiccation from 14% to 3% MC. In addition, during subsequent hermetic storage at constant temperatures of 20 to –20°C in F. sylvaticaand 10 to –20°C in F. crenata seeds, viability was lost more rapidly with reduction in MC below about 7.6–11.5% (40–71%relative humidity at 20°C). Thus, Fagus sylvatica and F. crenataexhibited intermediate seed storage behaviour. Survival at –20°C with 7.8–11.5% (F. sylvatica) and 7.6% MC (F. crenata) was comparatively good, with 64–84% of seeds remaining able to germinate normally after 2 years of hermetic storage, although this was neither appreciably better nor worse than at 0–10°C. Optimum seed storage environments, within the range investigated, were provided by combining temperatures of –10 to –20°C with 7.8–11.5% (F. sylvatica) or 7.6–9.5%(F. crenata) MC.

Dormancy and desiccation tolerance were studied in Fagus sylvatica seeds subsequent to 0–26 weeks of either moist prechilling (5°C) or warm (15°C) pretreatment at a restricted water content of 34% (fresh weight basis), i.e. c. 5% below full imbibition. After the pretreatments, seeds were either fully imbibed at 5 or 15°C, or dried for 7 d to around 13% moisture content and then fully imbibed at 5°C. Most of the dormancy was released after 5 weeks of prechilling, with nearly 70% of the seeds germinating at 15°C, and dormancy was fully released after 11 weeks of prechilling. Most of the seed desiccation tolerance, measured as the final germination percentage for dried seeds, was lost between 11 and 13 weeks of prechilling. A further decline in desiccation tolerance was recorded from 13 to 19 weeks, with a moderate recovery after 26 weeks. Germination was compared with the expression of a previously described abscisic-acid-responsive GRPF1 mRNA for a glycine-rich protein in F. sylvatica. There was a clear decline in accumulation of the transcript in seeds during prechilling, but not during the warm pretreatment. The amount of GRPF1 expression correlated significantly (P < 0.001) with dormancy status.

An abscisic acid (ABA)-induced cDNA fragment encoding a putative protein kinase (PK) was obtained by differential screening of a cDNA library from Fagus sylvatica seeds. The full-length clone, named FsPK1, was produced by 5′ rapid amplification of cDNA ends (RACE) extension. This clone contained the 11 catalytic domains present in all protein kinases, but displayed unusual characteristics found only in a few plant PKs. FsPK1 exhibits features of both serine/threonine and tyrosine protein kinases within the catalytic domain, a putative nuclear localization signal within the regulatory domain and the consensus sequence involved in binding of 14-3-3 proteins. The catalytic domain, expressed in Escherichia coli as a fusion protein, showed Ca2+-dependent in vitro kinase activity and dual serine/threonine and tyrosine specificity. Transcription of the FsPK1 gene was reduced by seed stratification at 4°C, and clearly increased when seeds were treated with 0.1 mM ABA, correlating with the inhibition of germination. Interestingly, FsPK1 transcripts were enhanced when ABA (0.1 mM) and calcium (1 mM) were added together, while the addition of EGTA (calcium chelator) and 3,4,5,-trimethoxibenzoic acid 8-(diethylamino) octyl ester (TMB-8, a calcium antagonist) decreased its expression. Furthermore, FsPK1 transcript expression was tissue specific and accumulated only in ABA-treated seeds, but not in any ABA-treated vegetative tissues examined. These results suggest that the expression of the corresponding protein could be related to the inhibition of germination mediated by ABA in a calcium-dependent pathway.

European beech (Fagus sylvatica L.) is an important forest tree species which was always considered being nonproblematic with regard to its susceptibility to pathogens and insects. However during the past decade, in Europe and the north-eastern USA an increasing number of trees and stands were showing symptoms typical for Phytophthora diseases: increased crown transparency, abnormally small and often yellowish foliage, a dieback of the crown, tongue-shaped necroses of the inner bark and the cambium with tarry or rusty spots on the surface of the bark either extending up to 7m from the stem base (collar rot) or occurring isolated higher up the stem (aerial bleeding cankers), fine root destructions and necrotic lesions on suberized roots. Large-scale investigations on both continents using specific isolation methods and ELISA kits for detection, and morphological and physiological parameters as well as ITS-DNA sequence analysis for identification demonstrated that several Phytophthora species were regularly involved as inciting agents of the decline. The most important species were Phytophthora citricola, P. cambivora and P. cactorum in Europe, and P. inflata in the USA. Their pathogenicity to beech was shown by stem inoculation and soil infestation tests performed by various groups. A small-scale nursery survey in Germany revealed that beech fields are regularly infested with various Phytophthora species. Options for disease management and control are discussed, and an emphasis is put on the prevention of disease spread via infested nursery stock.

Growth responses of two provenances of European beech (Fagus sylvatica) were studied. The seedlings were grown in closed-top chambers at four temperature regimes (−2.9 °C below ambient, ambient, +2.3 °C and +4.8 °C above ambient) in combination with two CO2 partial pressures (40 Pa and 74 Pa). Growth was followed by making destructive harvests c. every 25 d from germination in early June to senescence in late September. Allocation patterns were significantly affected by the temperature regimes. However, changes in dry matter allocation and morphology associated with the different treatments at a given time were mostly a result of differences in tree size. Temperature regimes only had a significant effect on the relative growth rate, RGR, at the beginning of the experiment. In contrast to temperature, high [CO2] increased RGR throughout the experiment when compared with plants of equal size. As the trees increased in size net assimilation rate, NAR, decreased but the effect of [CO2] on both NAR and RGR had a tendency to increase. Increases in NAR caused by elevated [CO2] were partly counteracted by reductions in the leaf area ratio, LAR. Reductions in LAR were caused by concomitant reductions in specific leaf area, SLA, whereas the level of [CO2] did not significantly affect leaf weight ratio, LWR, nor other dry weight ratios. The interactions between temperature and [CO2] are highly dependent on whether they are expressed as instantaneous values for plants at a common age or instantaneous values at a common size (and thereby extracting the effects of ontogenetic drift). When comparing instantaneous values at common sizes, the positive effect of [CO2] on RGR increased with plant size in every temperature regime. This also occurred in every temperature regime when comparing plants of equal age but the response to [CO2] was less. The effect of [CO2] on RGR was dependent on growth temperature. The positive effects of elevated [CO2] on RGR were less than the positive effect on photosynthesis. The two provenances did not differ significantly in the response of RGR to [CO2] which is in agreement with measurements of photosynthesis.

Physiological responses of two provenances of European beech (Fagus sylvatica) were studied in seedlings grown at two [CO2] in combination with four temperature treatments. For the local Danish provenance, the average effect of elevated [CO2] during growth was to increase light-saturated net photosynthesis (An) and instantaneous water-use efficiency or transpiration efficiency (ITE). These increases were strongly related to the temperature treatment. Stomatal conductance (gs) was reduced in seedlings in high [CO2], but there was no statistically significant effect of temperature treatment. Stomatal conductance was 13–26% lower at elevated [CO2] and ITE was 89–156% higher, depending on growth temperature. The effects of [CO2] on An were considerably larger than those shown for many other woody species, but similar to those in other studies on European beech. The absolute value of An for a Romanian provenance of beech was 5–18% lower than in the Danish provenance at low [CO2] and 14–26% lower at high [CO2]. There was no statistically significant interaction between the provenances and [CO2], or between provenance and temperature. A model of the response of An to [CO2] at different temperatures gave predictions close to the measured results, except at the lowest temperature treatment where the model over-predicted the effect of elevated [CO2]. This and measurements of An made at a common, low [CO2] indicated a down-regulation of photosynthesis in the lowest temperature treatment at high [CO2]. Root plus soil respiration on a whole-tree basis (Rtr) was increased by elevated [CO2] at all but the lowest temperature, but no effect was seen of [CO2] on root respiration per unit root d. wt. Mean Rtr on any given date was significantly correlated with An, except at the lowest temperature treatment. It is hypothesized that low temperature limited the ability of the roots to use photosynthates resulting in a feedback inhibition of An when elevated [CO2] was combined with low temperature.

Seedlings of 12 provenances of European beech (Fagus sylvatica) were exposed to ambient, non-filtered air (NF) or NF+50 nl l−1 ozone (NF50) for 8 h d−1 in open-top chambers (OTCs), from 1 June to 4 October 1995. In 1996 exposure was continued from 31 May to 1 October at four levels: charcoal-filtered air (CF), NF, NF50 and NF+100 nl l−1 ozone (NF100). Provenances were grown for both seasons in outside reference plots. All treatments were replicated. Ozone did not affect gas exchange in the provenances until late in the second season. NF100 reduced photosynthesis by 18% in August 1996 compared to CF. In September, photosynthesis was reduced by 22% in NF50 and by 29% in NF100. After two seasons, ozone reduced the root:shoot ratio by 24% when comparing CF and NF100; this was caused by reductions in the root biomass. Ozone did not affect height growth or stem diameter, and there were no ozone×provenance interactions for any growth parameter. There was, however, a significant ozone×provenance interaction for photosynthesis, showing northwest European provenances to be more sensitive to ozone than southeast European provenances when comparing dose–response estimates. This is interpreted in terms of genetic adaptation of the photosynthetic apparatus to regional growing conditions. Seedlings in the chambers grew 45% taller, and had 28% more shoot biomass and 29% smaller root biomass, resulting in a 44% reduction of root:shoot ratios compared to seedlings outside. Increased temperature and decreased PAR inside the chambers relative to the outside were probably the main causes for the differences. The magnitude of the chamber effects in OTCs raises doubts about conclusions drawn from ozone exposures in such chambers. This and previous ozone experiments with OTCs may have reached inaccurate conclusions concerning the size of ozone responses due to chamber effects.

Young spruce (Picea abies [L.] Karst; 2-yr) and beech trees (Fagus sylvatica L.; current-year and 1-yr-old seedlings) were grown under field conditions at a rural site near the city of Basel (Schönenbuch; 400 m above sea level) and 2-yr-old spruce trees at an elevated montainous site (Wengernalp; 1890 m above sea level). The plants were exposed in open-top-chambers (OTC) either to charcoal-filtered air or ambient air with ozone being the predominant air pollutant at both sites. The exposures lasted up to 1 yr. At the end of the growth period the plants were harvested and the pool sizes of the major non-structural carbohydrates were determined. Exposure to ambient air compared with filtered air controls caused an increase of glucose, fructose, pinitol, starch and, in some cases, sucrose in needles whereas carbohydrate contents in roots and twigs of spruce were reduced, in particular at Wengernalp. In beech saplings at Schönenbuch, a significant rise of sucrose and, in some cases, of raffinose, together with a significant reduction of glucose and fructose was observed in buds, leaves, twigs, and roots in ambient air compared with filtered air. In phloem exudates of current year beech seedlings, a 54–160% higher content of carbohydrates was found in ambient air than in filtered air. In roots of the beech saplings, the amount of soluble carbohydrates increased and starch contents mostly decreased. In general, starch storage in roots and stems was more abundant in beech trees. Spruce, especially at the mountainous site, accumulated high amounts of soluble carbohydrates in needles, in particular glucose.
