The aim of this study was to report the occurrence of Austrodiplostomum compactum metacercariae in the eyes of 98 specimens of loricariid fish (Hypostomus ancistroides, H. hermanni, H. iheringii, H. margaritifer, H. regani, H. strigaticeps, Hypostomus sp. and Megalancistrus parananus) from the Chavantes reservoir (23°07′36″S and 49°37′35″W) located in the rio Paranapanema, upper Paraná river basin, municipality of Ipaussu, São Paulo State, Brazil. Fish were collected from October 2007 to February 2009 using nylon monofilament gill nets and transported to the field laboratory where they were euthanized and the eyes were taken and examined under a stereomicroscope. Hypostomus ancistroides and M. parananus were not infected by this diplostomid. Hypostomus hermanni and H. margaritifer were represented by only one specimen but both had a high intensity of A. compactum metacercarie (27 and 35, respectively). Hypostomus strigaticeps (n = 45) and H. iheringii (n = 28) were the most representative specimens and the prevalence, mean intensity of infection and mean abundance were 24.4%, 10.3 and 2.7, and 64.2%, 13.1 and 8.4, respectively. No correlation was observed between the intensity of infection and the standard length (r = − 0.223; P = 0.827) and weight (r = 0.03; P = 0.779) of studied fish. Similarly, linear regression among these variables showed a poor correlation and indicated that the infection by A. compactum metacercariae occurs similarly in small and large fish specimens. A seasonal pattern of infection was not observed. Hypostomus hermanni, H. iheringii, H. margaritifer and H. strigaticeps were new hosts recorded for A. compactum metacercariae. A review of morphometric data of A. compactum metacercariae is presented.

The digenean trematode Diphtherostomum brusinae (Stossich, 1888) Stossich, 1903 presents a complex life cycle that may involve more than one intermediate host. The present study represents the first description of the metacercariae from D. brusinae infecting the labial palps of a new intermediate host, Mytilus galloprovincialis, in the Aveiro estuary, Portugal. The morphology of this parasitic stage was studied by light (LM) and scanning electron microscopy (SEM) and some differences were reported (body and sucker sizes, and spine distribution and shape). In this work, the 18S partial region of the ribosomal DNA was sequenced from D. brusinae metacercariae isolated from M. galloprovincialis collected in different localities of the Aveiro estuary. In addition, sequences from the same region of the 18S rDNA were obtained from D. brusinae cercariae and metacercariae, hosted by Nassarius reticulatus and Cerastoderma edule, respectively. No intraspecific polymorphism was detected in the 18S partial region, since there was 100% homology among all the sequences analysed. The same comparison was made for the ITS1, and we observed intraspecific polymorphism in this region. To our knowledge, this is the first report of D. brusinae metacercariae infecting the mussel M. galloprovincialis with support from morphological and molecular data.

In this study filarial recombinant protein or DNA vaccine constructs encoding BmALT-2 and BmVAH as single or as cocktail antigens were evaluated. Male jirds were immunized intramuscularly with DNA vaccine constructs or were immunized intraperitoneally with protein vaccine. The single and bicistronic DNA constructs induced substantial interferon-γ responses in spleen cells; antigen-specific responses were higher following immunization with the bicistronic cocktail construct and evoked a significant protective response of 57% in jirds challenged with Brugia malayi that was similar in the antibody-dependent cellular cytotoxicity (ADCC) assay and micropore chamber experiment. The cocktail protein vaccines induced a mixture of IgG2a (Th1) and IgG1 (Th2) responses with 80% protective response when challenged with B. malayi infective larvae. However, the single protein vaccine rALT-2 induced Th2 (IgG1/IgG3) with a 70% protective response and rVAH induced Th1 (IgG2a) with a lower proliferative response with 60% protection following challenge with B. malayi infective larvae. These results suggest that filarial cocktail protein vaccines are able to elicit substantial immune and protective responses when compared with single antigen vaccination in suitably vaccinated jirds.

Polystoma vernoni (Polystomatidae) is a new species in the urinary bladder of Ptychadena oxyrhynchus in KwaZulu-Natal province, South Africa. Ptychadena is a true African species and the 49 currently known species in Africa harbour 11 polystome parasites. Ptychadena oxyrhynchus has an extensive distribution through Africa and harbours Polystoma prudhoei in West Africa. This new parasite differs significantly from P. prudhoei. The new species is distinguished by its long body relative to its haptor length, its unique shape and its ratio of marginal hooklets. In a sample of 13 frogs, 53.8% were infected with up to 18 parasites per host (mean intensity 4.86).

The surface topography of 15-day-old adult Echinoparyphium recurvatum (Von Linstow, 1873) sensu stricto, from an isolate of the parasite utilizing Lymnaea peregra as first intermediate host in southern England, is described and illustrated using scanning electron microscopy, and is compared to those of other Echinoparyphium species from Europe, and with those of E. recurvatum of East Asian origin. The general morphology of tegumental features was found to be very similar to that of worms of the same age observed in a previous study on a Korean isolate of E. recurvatum. Comparison of collar and body spination of E. recurvatum with other 45-collar-spined members of the genus in Europe revealed some significant differences in morphology. Collar spines of E. recurvatum were found to be shorter and more pointed than those of Echinoparyphium mordwilkoi. The body spines of E. recurvatum are rounded and scale-like, extending just beyond the ventral sucker, contrasting with the pointed, thorn-shaped body spines of E. mordwilkoi, extending posteriorly to the level of the second testis. Body spine shape and distribution in E. recurvatum were found to be more similar to those of Echinoparyphium pseudorecurvatum. The value of SEM studies in elucidating the relationship between members of the genus Echinoparyphium in Britain/Europe and those in Africa, Asia and North America is suggested.

A survey was conducted of gastrointestinal nematode infections and trypanosomosis in Nigerian West African Dwarf (WAD) goats from the savanna region of the country. Animals were screened at two markets, Gboko and Akpagher, from the beginning of April until the end of September, coinciding with the end of the dry season and the first 5 months of the wet season. Of 1054 goats that were examined, 80.5% carried gastrointestinal (GI) nematodes belonging to the genera Haemonchus (61.0%), Oesophagostomum (21.0%) and Trichostrongylus (17.9%). Faecal egg counts (FEC) increased very slowly but significantly from April to maximum levels in September, and varied marginally between the two market sources. The majority of goats (68.8 and 70.1% at the two markets) had low FEC not exceeding 50 eggs/g (epg). FEC did not differ significantly between the sexes or between age classes. Packed cell volume (PCV) also declined significantly with month of the study, but was affected by host sex (a significant month × sex interaction) being generally higher in male animals throughout the period. There was a highly significant negative correlation between log10(FEC+1) and PCV, when all other factors had been taken into account. Body condition scores (BCS) also declined with month of the study, but there was a marked difference between the two sexes, with male animals generally showing a greater stability of BCS across the months compared with females. Trypanosome infections were found in only 4% of the goats and only during the rainy season. Most infections (92.86%) were caused by Trypanosoma brucei alone although T. vivax and T. congolense were occasionally detected. Overall, the majority of goats sampled each month maintained generally good body condition (BCS 3.0–5.0), normal or slightly reduced PCV, even when concurrently infected with trypanosomes and GI nematodes. However, four concurrently infected goats showed signs of overt anaemia during periods of peak infection, during the late rainy season, with marked reductions in PCV ( < 15%). Two of the infected goats were also in poor body condition with BCS of < 2.0. There was no evidence of additive or synergistic pathogenic effects of the two parasites. These results are discussed in the context of the unexpectedly strong resistance and resilience of the savanna WAD ecotype to its native strains of GI nematode and trypanosome parasites.

The gills of 41 Cichla piquiti and 39 C. kelberi from Itaipu and Lajes reservoirs, respectively, Brazil, were examined to describe the ectoparasite assemblages of these two non-native peacock-bass populations. All ectoparasite species of the two studied hosts (C. piquiti and C. kelberi) were dominant, but Ascocotyle sp. (metacercariae) was the prevalent (58.53%) and most abundant helminth species in C. piquiti hosts, while Sciadicleithrum ergensi was the dominant species in C. kelberi hosts. Gill ectoparasites of C. piquiti and C. kelberi showed a typical pattern of overdispersion or aggregation, which is commonly reported for many other freshwater fishes. Ectoparasite prevalence and abundance did not vary between host sexes of the two Cichla populations. The prevalence and abundance of Ascocotyle sp. were positively correlated with C. piquiti standard length (SL), but only the abundance of S. ergensi showed a positive correlation with C. kelberi SL. Although environmental differences between reservoirs might also have influenced the results, we anticipated that the presence of a close congener in Itaipu reservoir and the lack of other Cichla species in Lajes reservoir were the key factors to explain the contrasts between C. piquiti and C. kelberi gill ectoparasites. Overall, our results suggest that the trend of parasite species loss through the invasion process may have contributed to the establishment of non-native C. piquiti and C. kelberi populations in Brazilian reservoirs.

This study reports further observations on encystment and excystment of the paramphistomid trematode Zygocotyle lunata. Of numerous substrates tested in the laboratory for cercarial encystment, i.e. plastic and glass dishes, Styrofoam™, aluminium foil, snail shells, and cheesecloth, all but cheesecloth allowed for 100% encystment within 4 h of cercarial release at 28°C. Numerous physical and chemical factors were tested to kill cysts, and their capacity to do so was determined by the subsequent inability of cysts to excyst within a chemical excystment medium. Vinegar, Italian salad dressing, soy sauce and 10% solutions of sucrose, acetic acid and sodium chloride were the most detrimental. Freezing, boiling and storage of cysts for more than 2 years also killed them. Several marinades such as lemon juice, Worcestershire sauce, and various concentrations of potassium permanganate, did not kill the majority of cysts tested. Since Z. lunata is a potential foodborne trematode for humans and animals, it is important to determine suitable ways to control or kill these cysts.

Angiostrongylus cantonensis infection causes eosinophilic meningitis in humans. Baicalein is a flavonoid originally isolated from the roots of Scutellaria baicalensis Georgi. In this study we evaluated the efficacy of the combination of albendazole and baicalein for treating eosinophilic meningitis in BALB/c mice. Therapeutic efficacy included the survival time, body weight, neurological function, leucocyte and eosinophil counts, eotaxin concentration, matrix metalloproteinase-9 (MMP-9) activity, larval recovery and histopathological examination. The results showed that the combination of albendazole and baicalein was more effective than either drug administered singly. Combination therapy increased the survival time, decreased body weight loss, neurological dysfunction, leucocyte response, eotaxin concentration and MMP-9 activity. Our results suggest that the combination of albendazole and baicalein may exhibit synergistic beneficial effects in the treatment of eosinophilic meningitis induced by A. cantonensis.

Ethanolic and aqueous extracts of selected medicinal plants from Cameroon and Ghana were assessed for their in vitro anthelmintic activity by using the bovine filarial parasite Onchocerca ochengi and the free living nematode Caenorhabditis elegans, a model organism for research on nematode parasites. Worms were incubated in the presence of different concentrations of extracts and inhibitory effects were monitored at different time points. Among the extracts used in this study, ethanolic extracts of Anogeissus leiocarpus, Khaya senegalensis, Euphorbia hirta and aqueous extracts from Annona senegalensis and Parquetina nigrescens affected the growth and survival of C. elegans and O. ochengi significantly. The mortality was concentration dependent with an LC50 ranging between 0.38 and 4.00 mg/ml for C. elegans (after 72 h) and between 0.08 and 0.55 mg/ml for O. ochengi after a 24 h incubation time. Preliminary phytochemical screenings on these extracts revealed the presence of flavonoids, alkaloids, saponins, carbohydrates and tannins in the extracts. Accordingly, application of A. leiocarpus, K. senegalensis, E. hirta and A. senegalensis extracts could provide alternatives in the control of helminthic infections.

Cystic liver disease (CLD), presenting with solitary or multiple cysts in the liver, is a common diagnosis today, primarily due to the frequent application of modern radiological methods. There is a wide range of possible causes. CLD of infective origin is usually caused by an echinococcal species. During the past three decades a number of cystic echinococcosis (CE) control programmes have led to a significant decrease in the incidence of human hydatidosis in some endemic areas. The aim of this study was to determine the seroprevalence of E. granulosus infection in Croatian patients with CLD. A total of 540 serum samples from patients with hepatic cysts detected by imaging methods were screened for the presence of E. granulosus IgG antibodies using semiquantitative enzyme-linked immunosorbent assay. The Western blot technique was used as a confirmatory test for the CE diagnosis. The overall E. granulosus seroprevalence rate in patients with CLD was 3.9%. There was no significant difference in seroprevalence rate between male and female patients (P = 0.541). According to age groups, there was a significant difference in seropositivity among age groups (P = 0.002). The highest seroprevalence rate was detected in the youngest age group (up to 18 years), both in males and females (20% and 13%, respectively). This study indicates that CE still represents a public health problem in Croatia. Preventive measures should be used to control Echinococcus infections, including avoidance of contact with infected dogs, egg-contaminated soil or plants; control and treatment of dogs with antihelmintics; hand washing, improved sanitation and health education.

Cysticercus fasciolaris is the larval stage of the cestode Taenia taeniaeformis, whose definitive hosts are mainly cats. This larval stage uses a wide variety of small rodents, and occasionally birds and humans, as intermediate hosts. In the Yucatan, there are no reports of the presence of this cestode in animal populations. The aim of this study was to evaluate the occurrence of C. fasciolaris in rodent populations from the Cuxtal ecological reserve, Yucatan, Mexico. Trapping of rodents was conducted from October 2009 to April 2010 in 40 households in Molas, in which Sherman traps were placed both inside and outside backyards. Rodents were dissected to inspect the liver for the presence of the worm. To determine risk factors associated with infection, univariate analysis was performed using sex, age, species, trapping site, and season as independent variables. Variables with a P value <  0.2 were analysed using a logistic regression model. In this study, 411 individuals of six rodent species were trapped; Mus musculus was the most abundant (78%), followed by Rattus rattus (13%) and the wild species Peromyscus yucatanicus, Ototylomys phyllotis,Heteromys gaumeri and Reithrodontomys gracilis (9%). Only 7.5% (n = 31) of M. musculus and R. rattus were infected with C. fasciolaris (demonstrated by the presence of liver cysts) with a prevalence of 9.0% and 3.5%, respectively. Both adults and male mice were 4.33 and 3.46 (OR values) times more likely to have C. fasciolaris than juveniles and females respectively. We can conclude that in the Cuxtal Reserve, Yucatan, Mexico, the prevalence of C. fasciolaris is higher in M. musculus, and that adult males had a higher probability of infection. Wild species, mainly P. yucatanicus, were not found to be infected with the cestode, but its presence in the backyards of households could result in a potential risk of acquiring this infection.

Cystic echinococcosis (CE) caused by the larval stage of Echinococcus granulosus is a disease that affects both humans and animals. In humans the disease is treated by surgery with a supplementary option of chemotherapy with a benzimidazole compound. During the present study heat-shock protein 60 (HSP 60) was identified as one of the most frequently expressed biomolecules by E. granulosus after albendazole treatment. Data were correlated with 14-3-3 protein signature, and overexpression of this molecule after albendazole induction was an indicator of cell survival and signal transduction during in vitro maintenance of E. granulosus for up to 72 h. This observation was further correlated with a uniform expression pattern of a housekeeping gene (actin II). Out of three β-tubulin gene isoforms of E. granulosus, β-tubulin gene isoform 2 showed a conserved point mutation indicative of benzimidazole resistance.

Thirty-one faecal samples were collected from red deer in the northern area of Varese, in the Italian region of Lombardy, between August and October 2008. The animals had either been hunted or accidently killed. Examination for internal parasites showed a prevalence of 45.2% for Elaphostrongylus cervi larvae and species identification was confirmed by polymerase chain reaction (PCR). Ninety-seven faecal samples were also collected from two goat flocks grazing in the same area between December 2007 and May 2008. These showed a prevalence of 74.7% for lungworms. Furthermore, the central nervous systems from five goats and one sheep from this area with a history of neurologically related lameness were examined. Histopathology confirmed E. cervi cerebro- spinal nematodiasis in five cases out of six. This study demonstrates E. cervi transmission from wild to domestic ruminants when the animals graze in the same area, and the possible occurrence of clinical disease in infected goats and sheep associated with high prevalence in deer.

Echinococcosis is a zoonotic infection caused by adult or larval (metacestode) stages of cestodes belonging to the genus Echinococcus. The purpose of this study was to evaluate the antigenic ability of hydatid cyst fluid antigen for the diagnosis of hydatidosis in cattle using enzyme-linked immunosorbent assay (ELISA) and indirect haemagglutination test (IHA). The source of the antigens for the serological tests was fertile crude cyst fluids collected from naturally infected sheep at the Addis Ababa abattoir. A total of 502 sera were collected from 329 uninfected cattle and 173 hydatid-infected cattle which were confirmed by post-mortem examination. Most cysts were sterile and multiple organ infection predominated. Of 173 infected cattle, 166 (96.0%; confidence interval (CI) 91.8–98.4) were positive using ELISA while 7 (4.0%) were negative. Of 329 sera from uninfected cattle, 274 (83.3%; CI 78.8–87.2) were found to be negative and the remaining 55 (16.7%) were positive by ELISA. Of 173 infected cattle, 151 (87.3%; CI 81.4–91.9) were positive and 22 (12.7%) were negative by IHA. Of 329 negative sera tested using IHA, 266 (80.9%; CI 76.2–85.0) were negative and the remaining 63 (19.1%) were positive. The false positive and negative values of ELISA were 4.0 and 16.7%, respectively, and the corresponding values of IHA were 12.7 and 19.1%. The sensitivity and diagnostic efficiency of IHA were 87.2 and 83.6%, respectively. Crude hydatid cyst fluid antigen seems to have reasonable antigenic properties and hence could be employed for epidemiological surveillance of cattle hydatidosis.

We evaluated the relationship between infection by proteocephalid cestodes and the sex and weight classes of tucunaré (Cichla piquiti) captured between August 1999 and June 2001 in the Volta Grande Reservoir, Minas Gerais, Brazil. A total of 96 fish, 75.9 ± 9.3% males and 88.9 ± 6.4% females, were parasitized by Proteocephalus macrophallus and P. microscopicus, with total mean intensities of 76.6 ± 23.9 and 145.2 ± 36.7, respectively, during this period. In the majority of the months analysed, males showed 71.4–100% prevalence of parasitism and females 80–100%. Although there was no significant difference, females showed a higher mean intensity of infection (145.2 ± 36.7) than males (76.6 ± 23.9). Fish weighing 300–800 g showed a higher mean abundance of parasites (P < 0.05) compared with the biggest specimens weighing 801–2750 g. Analysing both males and females together, the greatest mean intensities of infection were found in October and December (P < 0.05) independent of the year, which coincides with the months of highest rainfall. These results show that fish living in reservoirs may be more susceptible to intermediate hosts than those that live in rivers.

A wild population of the lymnaeid snail Pseudosuccinea columella infected by larval stages of Fasciola hepatica was discovered in the Pinar del Río Province, Cuba. One of 100 snails was infected in a rice culture field. This is the first time this species has been found acting as intermediate host of F. hepatica under natural conditions, not only for Cuba but also for the Caribbean area.

This study evaluated the existence of different genotypes of Haemonchus contortus prevailing among goats in West Bengal, India. These parasites were isolated from the abomasum of goat intestine and the molecular characterization was performed by comparing variation of nucleotide sequences of the internal transcribed spacer 1 (ITS-1) gene region. Single-strand conformation polymorphism (SSCP) analysis of ITS-1 amplified product showed the presence of three distinct conformations both in male and female parasites. The sequence analysis of conformations showed two single nucleotide polymorphisms (SNP) in male parasites at nucleotide positions 106 and 107 and one SNP was detected in female parasites at nucleotide position 157. These nucleotide variations in different isolates did not alter the interior loop structure of the predicted secondary RNA, therefore we believe these variations may not be responsible for any evolutionary changes among conformations.

Five out of ten ocellated skinks (Chalcides ocellatus) examined in Dubai between 2007 and 2010 were infected with cestodes of the genus Oochoristica. Out of the 36 collected tapeworms seven specimens were used to describe a new species. Oochoristica chalcidesi n. sp. belongs to the group of species with 25 to 35 testes arranged in two clusters. The lobes of the ovary are subdivided into 4–5 lobules in a similar way as O. ubelakeri described from Agama atra in Namibia. Both species differ in the presence of a neck, a lower number of mature segments in O. chalcidesi n. sp. and a different position of the cirrus pouch in relation to the ovary, as well as in the distribution of uterine capsulae in gravid segments.

A total of 687 adult nematomorphs of four species of Gordiida: Chordodes brasiliensis (393 specimens), Noteochorododes cymatium (47 specimens), N. talensis (162 specimens) and Pseudochordodes dugesi (85 specimens) were collected during a period of 1 year from El Simbolar stream, Argentina. Free-living worms were abundant during autumn and spring, but their number decreased during winter and summer. Males were shorter and significantly more abundant than females. The presence of N. cymatium, N. talensis and P. dugesi was correlated with water temperature and these species were most abundant in winter and spring. The presence of C. brasiliensis was correlated with flow rate and pH; this species was more abundant in autumn and winter. These four species are sympatric.
