The success of Phasmarhabditis hermaphrodita (Schneider) Andrássy (Rhabditida: Rhabditidae) as a biological control agent of molluscs has led to a worldwide interest in phasmarhabditids. However, scant information is available on the lifecycle development of species within the genus. In the current study, the development of P. hermaphrodita, Phasmarhabditis papillosa, Phasmarhabditis bohemica and Phasmarhabditis kenyaensis were studied using ex vivo cultures, in order to improve our understanding of their biology. Infective juveniles (IJs) of each species were added to 1 g of defrosted homogenized slug cadavers of Deroceras invadens and the development monitored after inoculated IJ recovery, over a period of eight–ten days. The results demonstrated that P. bohemica had the shortest development cycle and that it was able to produce first-generation IJs after eight days, while P. hermaphrodita, P. papillosa and P. kenyaensis took ten days to form a new cohort of IJs. However, from the perspective of mass rearing, P. hermaphrodita has an advantage over the other species in that it is capable of forming self-fertilizing hermaphrodites, whereas both males and females are required for the reproduction of P. papillosa, P. bohemica and P. kenyaensis. The results of the study contribute to the knowledge of the biology of the genus and will help to establish the in vitro liquid cultures of different species of the genus.

Of the total 47 species in the subgenus Acanthosentis, 43 have been reported from the freshwater fishes of Asia. Amin et al. (2017) provided a key to the 23 species of the genus Acanthogyrus reported from the Indian subcontinent. The present study reports two new species: Acanthogyrus bispinosa n. sp. and A. garciai n. sp. from Cirrhinus mrigala Hamilton and Labeo calbasu Hamilton, respectively, and two previously described species: A. golvani Gupta and Jain, 1980 and A. hereterospinus Khan and Bilqees, 1990 from L. rohita Hamilton and L. catla Hamilton, respectively. A. bispinosa n. sp. comprises 3 circles of 6 proboscis hooks each. Trunk spines in A. bispinosa n. sp are divided into two groups: anterior and posterior separated by unarmed region, which has not been previously reported in the subgenus. Anterior spines are present in 7–8 and 7–10 circles in females and males, respectively, whereas posterior spines are in 23–28 and 31–38 circles in males and females, respectively. A. garciai n. sp. comprises 3 circles of 6–8 hooks each and a single set of trunk spines is present in A. garciai n. sp., comprising 35–42 and 25–45 circles in males and females, respectively. All four species were also characterised based on the 18S, 28S, and ITS1-5.8S-ITS2 rRNA molecular markers. The Bayesian inference tree generated based on these markers showed distinct identities of all the species, with a significant molecular divergence, ranging from 3.2 to 53.6%.

Two new species of diplectanid monogeneans, Diplectanum grouperi and Pseudorhabdosynochus coioidesis from the marine finfish Epinephelus coioides and E. aerolatus(Serranidae), cultured in floating cages in Malaysia, Indonesia and Hong Kong, were described and compared with P. lantauensis Beverley-Burton and Suriano, 1981 from the same hosts. There is no other close species sharing the features of Diplectanum grouperi. The possession of a copulatory organ with an open cup-like proximal part and a narrow tubular distal part is a main distinguished feature of D. grouperi. Pseudorhabdosynochus coioides is most closely related to P. epinepheli Yamaguti, 1938, but can be distinguished from it by the shape and the size of the haptor, hamuli, copulatory organ, the lack of a muscular bulbus ejaculatorius, a greater number of rows in the squamodiscs and the location of the testis. Pseudorhabdosynochus lantauensis from the present hosts differs from that described from the original hosts, Epinephelus brunneus and E. fario in the shape of the dorsal bar and vagina.

The type species of the genus Epacrolaimus, Epacrolaimus declinatoaculeatus, is studied from the re-examination of type material of Aporcelaimus vorax, its junior synonym, and the observation of several Iberian populations and a few Iranian specimens. Morphologically, it displays a recognizable morphological pattern characterized by, among other features, the incurved nature of its odontostyle aperture, presence of perioral liplets or lobes, lip region 24–31 μm wide, odontostyle 21–25 μm long and comparatively anterior location of S2N pharyngeal gland nuclei. Nevertheless, variations in some morphological traits (vagina shape and tail shape) and in several morphometrics (body length, uterus length, vulva position, tail length and spicule length) are also noted. Sequences of D2–D3 domains of the 28S rDNA, 18S rDNA and COI mtDNA were obtained from several Iberian populations. Their analyses, in particular those from D2–D3 sequences, revealed the existence of a highly supported clade ((Epacrolaimus + Sectonema) + Metaporcelaimus), with a closer relationship between Epacrolaimus and Palaearctic populations of Sectonema, whereas the remaining aporcelaimid genera occupied placements in other clades. These results are discussed, with especial emphasis on the intricate separation of Epacrolaimus and Sectonema, which display significantly different protruding stomatal structure in spite of their close evolutionary relationship as derived from molecular trees.

Slugs are important agricultural pests causing yearly yield losses. However, parasitizing helminths potentially could affect the size of the slug population. Here, a survey of terrestrial slug-parasitic helminths (nematodes and trematodes) was conducted for the first time in Sweden. In total, 268 terrestrial slugs were collected from 27 agricultural field edges in three seasons over 2020 and 2021 and dissected for presence of helminth parasites. Slugs belonging to the genus Arion were molecularly identified by mitochondrial DNA cytochrome c oxidase subunit I (COI) while parasites were identified using ribosomal RNA (18S). Overall, 13% of the collected slugs had helminth parasites and the likelihood of a slug being parasitized was highest in autumn. Slugs identified as Arion vulgaris were more likely to be parasitized than native slug species. The prevalence of nematodes and trematodes were similar; the dominant species found were Alloionema appendiculatum and Brachylaima thompsoni, respectively. This is the first record of the presence of these two species in Sweden.

The ability of two isolates of the nematode-trapping fungus Duddingtonia flagrans to reduce the numbers of gastrointestinal nematode larvae on herbage was tested in three plot studies. Artificially prepared cow pats containing Ostertagia ostertagi eggs, with and without fungal spores, were deposited on pasture plots two or three times during the grazing season in 1995, 1996 and 1997. The herbage around each pat was sampled fortnightly over a period of 2 months and the number of infective larvae was recorded. At the end of the sampling period, the remainder of the faecal pats was collected to determine the wet weight, dry weight, and content of organic matter. The infective larvae remaining in the pats were extracted. Faecal cultures showed that both fungal isolates significantly reduced the number of infective larvae. Significantly fewer larvae were recovered from herbage surrounding fungus-treated pats compared with control pats in all three experiments, reflecting the ability of the fungus to destroy free-living larval stages in the faecal pat environment. After 8 weeks on pasture there were no differences between control and fungus-treated pats with respect to wet weight, dry weight, and organic matter content. This indicates that the degradation of faeces was not negatively affected by the presence of the fungus.

Systematic review and meta-analysis were conducted to provide comprehensive information on the prevalence of amphistome infections in domestic ruminants in sub-Saharan Africa. A systematic search of peer-reviewed articles published between 2002 and 2023 was conducted. Prevalence estimates and meta-analysis were based on 76 peer-reviewed articles which met the inclusion criteria. Of the 55,122 domestic ruminants screened, 12,858 were infected, and the overall pooled prevalence was 22% (95% confidence interval [CI], 10-37). The highest prevalence was recorded in southern Africa 25% (95% CI, 0-62), and central Africa 16% (95% CI, 0-61) the lowest. Cattle were the most frequently sampled hosts (76.56%, n = 42,202) and sheep (8.78%, n = 4838) the lowest, and cattle recorded the highest pooled prevalence of 28% (95% CI, 12-47), and goats the lowest at 5% (95% CI, 0-14). Prevalence rate was the high in males 32% (95% CI, 21-44), adult ruminants 37% (95% CI, 15-62) and animals with poor body condition 47% (95% CI, 34-60), and during the wet season 36% (95% CI, 0-94). The highest pooled prevalence was recorded at postmortem 23% (95% CI, 8-43) compared to coprology 20% (95% CI, 6-39) studies. The meta-regression model demonstrated that the body condition score, host, and period, and the interactions of different factors significantly influenced the prevalence. The lowest prevalence rate was noted for the period between 2013 and 2023. This is the first systematic review and meta-analysis in sub-Saharan Africa that provides a comprehensive review of the prevalence of amphistome infections in domestic ruminants in the past 20 years.

We investigated parasitic zoonoses caused by protozoans and helminths in urban and peri-urban rat populations (Rattus norvegicus and R. rattus) in Spanish cities. Rats were trapped and then dissected to remove adult helminths, and the contents of the large intestine were retrieved for the study of parasitic forms. The Midi Parasep® solvent free (SF) technique was used to concentrate the parasites in the intestinal contents. Some of the rats studied (n = 8) were infected by the rat lungworm, Angiostongylus cantonensis, whose first stage larvae (L1) are shed in rat faeces. After the concentration technique, L1 larvae were found in the sediment of 6 of the 8 positive rats. The two negative sediment samples were due to the presence of either only adult females or, in addition to males, only young females in the lungs of the rats. In view of our results, Midi Parasep® SF turned out to be a simple, rapid, inexpensive, and sensitive method to detect nematode larvae, such as the L1 larvae of A. cantonensis (or A. costaricensis), in natural and experimentally infected rats.

The ultrastructure of the dorsal oesophageal gland ampulla and its relationship with the oesophagus, oesophageal ultrastructure, and control mechanisms in oesophageal activity were studied. Terminal ducts of the sub-ventral glands open through the oesophageal crown at the base of the buccal cavity. The terminal duct of the dorsal oesophageal gland running through the dorsal gutter opens to the exterior at the rim ‘groove’ of the buccal capsule. The posterior oesophageal region is clavate and the cuticle of the lumen folds to form outlet valves, ‘valvulae’. An inconspicuous oesophago-intestinal valve (three lobes) connects oesophagus and intestine and is visualized in the open and shut position. In the female reproductive tract, with the exception of the uterus, the cells lie on a thick, irregular (convoluted) basal lamina. The apical plasma membrane of the uterus, and seminal receptacle, extend into the lumen by microvilli-like projections with which spermatozoa make intimate contact. The lumen of the uterus is filled with oocytes, fertilized and unfertilized. Testicular cells have two parts linked by a rachis. Spermatocytes are elongated with a large nucleus, distinct nuclear membrane, and many granules. The apical membrane of the rachis forms long microvilli-like projections with balloon-like tips. The amoeboid spermatozoa contain membrane specializations, a nucleus devoid of a membrane, and are enclosed by a pseudopodial-like extension.

Plasma etching and ashing for demonstrating the three-dimensional ultrastructure of the internal organs of helminths is described. Adult worms of the cestode Caryophyllaeides fennica were dehydrated through an ethanol series, critical point dried (Polaron E3000) and sputter coated with 60% gold-palladium (Polaron E5100) and glued to a standard scanning electron microscope (SEM) stub positioned as required for ashing. After initial SEM viewing of worm surfaces for orientation, stubs were placed individually in the reactor chamber of a PT7150 plasma etching and ashing machine. Worms were exposed to a radio frequency (RF) potential in a low pressure (0.2 mbar) oxygen atmosphere at room temperature. The oxidation process was controlled by varying the times of exposure to the RF potential between 2 to 30 min, depending on the depth of surface tissue to be removed to expose target organs or tissues. After each exposure the oxidized layer was blown from the surface with compressed air, the specimen sputter-coated, and viewed by SEM. The procedure was repeated as necessary, to progressively expose successive layers. Fine details of organs, cells within, and cell contents were revealed. Ashing has the advantage of providing three dimensional images of the arrangement of organs that are impossible to visualize by any other procedure, for example facilitating testes counts in cestodes. Both freshly-fixed and long-term stored helminths can be ashed. Ashing times to obtain the desired results were determined by trial so that some duplicate material was needed.

The mallard Anas platyrhynchos is the most abundant water bird species in Austria, but there is no record of its helminth community. Therefore, this work aimed to close that gap by recording and analysing the parasite community of a large number of birds from Austria for the first time. A total of 60 specimens shot by hunters in autumn were examined for intestinal parasites. The following taxa were recovered (prevalence given in parentheses): Cestoda: Diorchis sp. (31.7%) and Fimbriarioides intermedia (1.7%); Acanthocephala: Filicollis anatis (5%), Polymorphus minutus (30%) and one cystacanth unidentified (1.7%); Trematoda: Apatemon gracilis (3.3%), Echinostoma grandis (6.7%), Echinostoma revolutum (6.7%) and Notocotylus attenuatus (23.3%); Nematoda: Porrocaecum crassum (1.7%) and one not identified (1.7%). The frequency distribution of parasites showed a typical pattern in which 39 birds (65%) were either not parasitized or were harbouring up to five worms, whereas more intense infestations occurred in a lesser number of hosts. Compared to other studies from central and eastern Europe, an extremely depauperate helminth community, particularly of the cestodes and nematodes, was found. Polymorphus minutus was observed as having highly variable morphology and, therefore, molecular genetic characterization by DNA barcoding was carried out. Species identification was confirmed by comparing data with the reference cytochrome c oxidase subunit 1 gene sequence from P. minutus available in GenBank.

Eighteen of 56 (32.1%) wild Rana limnocharis from central and south Taiwan were found to contain plerocercoids of Spirometra erinaceieuropaei. This is the first report of S. erinaceieuropaei infections in frogs in Taiwan, with the plerocercoids being recovered from the thigh and back muscles or under the skin. Other species of frogs examined, including nine wild R. latouchii, one wild Buergeria robustus and 110 cultured R. rugulosa were free of infection. The plerocercoids were orally inoculated into four cats; three of which were each given a single plerocercoid and one a dose of three plerocercoids. Daily faecal examination showed that two cats started shedding eggs of S. erinaceieuropaei on day 8 postinfection (PI) and the other two on day 10 PI. The highest eggs per gram and eggs per day for a single worm was found to be 428,000 and 14,416,000 respectively. Only the cat inoculated with three plerocercoids shed proglottids in its faeces during the 2 month observation period.

Kribi is a seaside town that welcomes thousands of tourists each year. However, the poor sanitation condition of its beaches along the Atlantic coast is not without risk for visitors. In this study, we used the formol-ether concentration technique to identify and quantify larvae or eggs of intestinal helminths in waters of three regularly visited Kribi beaches (Mpalla, Ngoyè, and Mboamanga). Results revealed that all identified larvae and eggs were cestodes (Hymenolepis nana) and nematodes (Strongyloides sp., Ascaris sp., Ancylostoma duodenale and Trichuris trichiura). All the helminth eggs and larvae showed high abundance at low tide during rainy seasons. Ancylostoma duodenale eggs, totally absent at Mpalla, were densely present at low tide at Ngoyè (301 ± 15 eggs/L). Trichuris trichiura eggs showed the lowest abundance (0 to 62 eggs/L) at all sites. Abiotic variables indicated that waters at the various beaches were basic (pH: 8.75–9.77), generally warmer (32.44°C at Mpalla in the Short Rainy Season), more oxygenated at low tide, and moderately mineralized at high tide. Positive and significant correlations were observed at Ngoyè at low tide between Strongyloides sp. larvae and dissolved oxygen (P ˂ 0.05); and between Ancylostoma duodenale eggs and temperature (P ˂ 0.05). The overall results indicated that the beaches studied are subjected to fecal pollution. This pollution is more accentuated during low tides than during high tides. Depending on tidal movements, swimmers risk exposure to helminth eggs and larvae known to be responsible for gastroenteritis.

Ring-necked pheasants were exposed to artificially limited 10-day infections with heterakids either from 0-10 or 10–20 days old. The number and length of worms developing from a subsequent challenge dose were compared with those in nonimmunized control birds and with those in birds that had been exposed to a continuous immunizing infection for a total of 49 days: 28 days before challenge and 21 days during the challenge infection.

Since the introduction of carbon tetrachloride for the treatment of the hookworm infestations of man it has been ascertained that in certain conditions fatal poisoning may follow oral administration of the drug. If carbon tetrachloride is given in cases of alcoholic cirrhosis of the liver, or if alcohol is taken immediately after the drug, severe gastro-intestinal symptoms are to be anticipated with death in two or three days. Davis (1924) showed that in dogs the toxicity of the drug was intimately associated with certain dietetic factors. He found that a high carbohydrate diet enabled the liver to withstand relatively large doses of the drug. High protein diets also protected the liver against injury (although less efficiently than carbohydrate) but an “all-fat” diet was invariably associated with extensive damage to the hepatic parenchyma. Lambert (1923) while carrying out hookworm treatment on a large scale in Fiji found that where a patient was heavily infested with Ascaris lumbricoides as well as with hookworm it was unwise to exhibit carbon tetrachloride without first getting rid of the ascarids.

Laboratory experiments have shown that cysts of the eelworm Heterodera schachtii adhering to seed potato tubers are killed by immersion of the tubers for 6 hours in 5% formalin solution (Franklin 1939). Further experiments were designed to determine what effect such treatment has on the seed tubers and on the resulting crop.

Temnocephala axenos Monticelli, 1898 was described based on specimens from an unidentified host collected in Blumenau, Santa Catarina, Brazil. Information about type locality was imprecise and the host was later identified as Aegla laevis (Latreille, 1818). However, it is known that A. laevis is not present on the eastern side of the Andes. Also, only histological preparations from one specimen studied by Monticelli are currently available in the Museum für Naturkunde Berlin, but it showed none of the taxonomic characters needed for the characterization of the species. Although the updated description of the species based on Uruguayan specimens, neither the author nor the several previous studies about the species showed a search for the type material, a resolution for the misidentification of the type host or the imprecise type locality due to the subsequent geographical division of the municipality cited in the description. The Uruguayan specimens were not even geographically close to the type locality and a neotype was not designed to validate the species’ taxonomic status again. Specimens from Santa Catarina and Paraná States, Brazil, were studied, as well as restudied Argentinean specimens. The new data were compared with the update description of the species. The historical background and the discussion about geographical origins and hosts of the species, as well as a designation of a neotype, allow comparative material of the type locality and type host to exist, eliminating doubts about the identification of T. axenos.

Molecular techniques are an alternative for the diagnosis of strongyloidiasis, produced by Strongyloides stercoralis. However, it is necessary to determine the best amplification target for the populations of this parasite present in a geographical area and standardize a polymerase chain reaction (PCR) protocol for its detection. The objectives of this work were the comparison of different PCR targets for molecular detection of S. stercoralis and the standardization of a PCR protocol for the selected target with the best diagnostic results. DNA extraction was performed from parasite larvae by saline precipitation. Three amplification targets of the genes encoding ribosomal RNA 18S (18S rDNA) and 5.8S (5.8S rDNA) and cytochrome oxidase 1 (COX1) of S. stercoralis were compared, and the PCR reaction conditions for the best target were standardized (concentration of reagents and template DNA, hybridization temperature, and number of cycles). The analytical sensitivity and specificity of the technique were determined. DNA extraction by saline precipitation made it possible to obtain DNA of high purity and integrity. The ideal target was the 5.8S rDNA, since the 18S rDNA yielded non-reproducible results and COX1 never amplified under any condition tested. The optimal conditions for the 5.8S rDNA-PCR were: 1.5 mM MgCl2, 100 μM dNTPs, 0.4 μM primers, and 0.75 U DNA polymerase, using 35 cycles and a hybridization temperature of 60 °C. The analytical sensitivity of the PCR was 1 attogram of DNA, and the specificity was 100%. Consequently, the 5.8S rDNA was shown to be highly sensitive and specific for the detection of S. stercoralis DNA.
