The prevalence of intramammary infection in 4 commercial goat herds was studied in conjunction with electronic somatic cell count and volume analysis, determined using a Coulter Counter and volume analyser.

Neither streptococci nor mycoplasma were isolated from any half and the prevalence of intramammary infection with Staphylococcus aureus ranged from 0 to 3% between herds. For coagulase-negative staphylococci the range for infected halves was 36–71%. There was no significant difference between the mean total microscopic somatic cell count for halves infected with coagulase-negative staphylococci and those free from infection. A similar trend was observed for electronic somatic cell counts although the mean electronic cell count was greater than the mean total microscopic count on the 2 occasions that they were compared. The correlation coefficients between the 2 cell counting methods were 0·86 and 0·94. Between herds there were significant differences in mean electronic somatic cell count, with herd means ranging from 438×103 to 1684×103 cells/ml. In 2 of the 4 herds studied, milk samples from halves infected with coagulase-negative staphylococci had a significantly higher prevalence of cell volume distributions with a modal cell volume between 65 µ3 and 100 µ3. This was attributed to a higher proportion of polymorphonuclear neutrophils.

Use of electronic somatic cell count and cell volume analysis were considered of little value in predicting infection caused by coagulase-negative staphylococci as there was a high proportion of false negative and false positive predictions.

The effect of ageing fresh milk for 24 h at 4°C on the composition of 4 fractions of milk fat globule membrane (FGM) – microsomal pellet membrane (MPM), deoxycholate soluble membrane (DOCM), high density membrane (HDM) and low density membrane (LDM) – prepared from washed cream treated with sodium deoxycholate (DOC), was studied for milk of individual cows. Total FGM and its fractions were solubilized by treatment with sodium dodecyl sulphate (SDS), EDTA and β-mercaptoethanol, and the dissociated FGM proteins were separated by polyacrylamide gel electrophoresis in the presence of SDS.

Ageing resulted in a greater loss of phospholipid during cream preparation than was found with fresh milk, and also in a reduction in the total amount of FGM isolated. Loss of FGM material was confined to MPM, DOCM and LDM and was accompanied by compositional changes in DOCM and LDM, quantitatively the most significant fractions. Ageing also produced changes in the gel electrophoresis patterns of DOCM, where the band of greatest mobility (mol. wt about 16000) was partially lost, and of LDM, where there was an increase observed in the major protein band.

The implication of the results on the proposed models of FGM structure is discussed.

The ability of Mozzarella cheese to stretch and form long fibres was evaluated by two simple instrumental tests. The first test, which was basically a tensile test, resembled the way the consumer assesses the stretchability of cheese as a pizza topping. The extensibility measurements of the test correlated well with the sensory stringiness scores. The second, fundamental test was a compressive elongation of the melted cheese. The elongational viscosity of the melted cheese was calculated; this could be used as an index of the ability of the cheese to form strings when it is stretched.

Aminopeptidase P was purified 65·3-fold from the cytoplasm of Lactococcus lactis subsp. cremoris AM2 with a 5·8% yield. The purified enzyme was found to consist of one polypeptide chain with a relative molecular mass of 41600. Metal chelating agents were found to be inhibitory and Mn2+ and Co2+ stimulated activity 7-fold and 6-fold respectively. The purified enzyme removed the N-terminal amino acid from peptides only where proline (and in one case alanine) was present in the penultimate position. No hydrolysis was observed either with dipeptides even when proline was present in the C-terminal position or when either N-terminal proline or pyroglutamate was present preceding a proline residue in the penultimate position of longer peptides. On the basis of this substrate specificity either aminopeptidase P or post-proline dipeptidyl aminopeptidase are necessary along with a broad specificity aminopeptidase to effect complete hydrolysis of casein-derived peptides containing a single internally placed proline residue. However, both aminopeptidase P and post-proline dipeptidyl aminopeptidase would be required together with a broad specificity aminopeptidase in order to completely hydrolyse casein-derived peptides that contain two internally placed consecutive proline residues. As bitter casein-derived peptides are likely to contain either single prolines or pairs of prolines, aminopeptidase P appears to be an important enzyme for debittering.

7-Ketocholesterol and 7-ketositosterol were chosen as reliable markers of the oxidation of cholesterol and phytosterols in infant milk formulas and infant milk cereals. A reversed-phase HPLC method was developed to measure them simultaneously in infant formulas. This method was then tested on a wide range of infant milk formulas and milk cereals on sale in Italy whose lipid composition is representative of the most common commercial formulas. The analytical results revealed no significant differences in the extent of oxidation of cholesterol and sitosterol. As the level of 7-ketocholesterol often followed the cholesterol level, a cholesterol content similar to that of human milk produced amounts of cholesterol oxides with possible negative effects on infant health. In contrast, the low cholesterol content of milk cereals never produced amounts of cholesterol oxides high enough to cause concern. The contents of phytosterols and hence their oxides were always low.

A method for detection of proteolysis in milk was evaluated. Amino acids and peptides soluble in trichloroacetic acid were estimated by the Lowry–Folin procedure with expression of results in terms of colour yield equivalent to that of tyrosine (the ‘tyrosine value’ (TV)). Some variables and alternatives in the method of estimation were investigated.

TVs for milk freshly drawn from individual cows were extremely varied (0·31–0·92 mg/ml). TVs for samples of refrigerated milk from daily supply farms were in the range 0·40–0·58 mg/ml; no relationships between TV and bacterial counts were evident with these samples. With bulk refrigerated raw milk supplied to Brisbane for liquid consumption from country depots and local farms equipped with bulk vats, significant positive relationships were found between TV and total bacterial count (TBC) for supplies from some sources, but in no instance was a significant relationship found between TV and psychrotroph or proteolytic psychrotroph count. Significant positive relationships were found between TV of bulk milk supplies from some sources and atmospheric temperature, and between TBC of the supplies and atmospheric temperature. The significance of these various relationships for bulk milks is discussed.

Natural variation in TV imposed limitations on reliability of the method to provide an index of proteolysis; in general the results indicated that its application was restricted to bulk milk cold-stored for at least 3 days.

α-Adrenergic mimetics inhibit milk removal whereas β-adrenergic mimetics have a favourable effect upon milk removal in dairy cows. This work aimed to relate these effects to the milk ejection reflex mechanism and/or milk flow from the cisternal cavities only. The α-adrenergic agonist phenylephrine and the β-adrenergic agonist isoproterenol were injected prior to and after milk ejection. Applied after milk ejection, the phenylephrine treatment reduced intramammary pressure (IMP) and milk yield markedly. Phenylephrine application before milk ejection prevented IMP from rising during teat stimulation or after oxytocin injection. Milking produced small amounts of milk at minimal milk flow rates. The very different responses to phenylephrine application prior to and after milk ejection indicate separate adrenergic inhibitory mechanisms for milk ejection and milk flow. The application of the β-adrenergic agonist isoproterenol did not alter the IMP. Nevertheless, this compound improved milk removal by increasing milk flow. The observation of bimodal milk flow at milkings without preceding teat stimulation indicates a delayed milk ejection. Isoproterenol injection without teat stimulation enhanced milk flow rates, but the bimodality remained unchanged. We conclude that isoproterenol enhances milk flow without affectiiig milk ejection.

A method for measuring the increasing rigidity moduli and internal viscosities of milk during rennet setting is described. Curves are given showing the degree of reproducibility on replicate samples of dried and liquid milks, and of the effects of changes in calcium levels in the case of the former.

A tentative interpretation of the shape of the main part of the setting curve is proposed. The upper part of the curve generally becomes erratic with the onset of syneresis, but some dried milks do not show this behaviour.

Some curious phenomena concerning the production of syneretic liquid (whey) are described and methods for obviating the disturbing effects of syneresis are discussed.

1. A method is described for following the intensity of reduction (or oxidation-reduction potential) at different levels in the interior of ripening Cheddar cheese.

2. Variations in this potential throughout the course of ripening are described and mean values assessed.

3. Cheese are shown to be not homogeneous throughout their mass; outer zones of more highly oxidised condition exist near the surface of the cheese and round cracks and borings. The depth of these zones increases with the age of the cheese.

4. The values obtained can be correlated with the known bacteriological data for Cheddar cheese.

5. The significance of the data obtained in relation to cheese faults is discussed.

British Saanen dairy goats (n = 10) were treated with bromocriptine or vehicle from day 147 of pregnancy to day 4 post partum, a treatment duration of 8·8±1·7 d (mean ± SEM). The periparturient prolactin surge was abolished by this treatment, but there were no significant effects on plasma growth hormone or insulin concentrations. Lactogenesis was delayed in the bromocriptine-treated goats, milk yields being significantly depressed (P < 0·01) for the first week of lactation. Yields had recovered to control values by day 10 when prolactin concentrations were still significantly depressed. Mammary gland biopsies were taken on day 4 post partum from five animals in each group. Using this tissue, no significant differences could be shown in mammary morphology or DNA synthesis, but the RNA:DNA ratio was significantly reduced (P < 0·05). After week 1, there were no significant differences between bromocriptine-treated and control goats in milk yield, milk composition, udder volume, time of peak yield or persistence. The goats given short-term bromocriptine treatment at parturition showed prolonged effects on prolactin secretion, their seasonal prolactin rise being severely blunted (P < 0·001). A normal lactation is therefore not prevented in goats by a delay in lactogenesis, suppression of prolactin at parturition or the resulting prolonged depression of circulating prolactin. Goats in established lactation given bromocriptine for 8 d showed, by contrast, a rapid recovery of plasma prolactin concentrations within 5 d post treatment. Milk yield declined significantly (P < 0·03) compared with pretreatment values during and for 1 week after bromocriptine but then began to recover, with no significant change in vehicle-treated goats.

Swabs taken from the rectum and vulva of 14 dairy cows on 7 occasions at approximately weekly intervals revealed a pattern of isolations of Streptococcus uberis which were not random. Two of the 14 cows were responsible for 80 and 40% respectively of the swabs of the rectum and vulva which were found to be positive. It is suggested that these patterns are consistent with these 2 cows being intestinally colonized with Str. uberis. The implications of this work are discussed with reference to the sources of Str. uberis in the dairy herd.

Explants from lactating bovine mammary gland were cultured in vitro in serum-free medium through 1–9 d. Casein synthesis was determined by [32P] incorporation into newly synthesized Ca rennin precipitable fraction. High correlation (γ = 0·98) was found between incorporation of [32P] and [3H]amino acids in explants cultured under different hormonal regimes, thus indicating that post-translational phosphorylation is not a rate-limiting step in casein synthesis.

Hormonal effects on casein synthesis were studied by supplementing the incubating medium with insulin (I), prolactin (PRL), cortisol (F), thyroxine (T4) and triiodothyronine (T3). It was found that both PRL and I were required absolutely for maximal synthesis and almost maximal effect was achieved with 50 ng/ml. The effect of F was less clear, but some increase was achieved at the 200–1000 ng/ml range. T4 and T3 did not affect casein synthesis at a range of 10-11–10-7M while a significant inhibition was observed at 2 × 10-5 M. A time-course study of casein synthesis further substantiated the dominant role of PRL in maintenance or even elevation of the initial rate of casein synthesis in the explants, through the first 4 d of incubation.

This research study aimed to evaluate the use of the milk leukocyte differential (MLD) to: (a) identify quarter milks that are culture-positive; and (b) characterize the milk leukocyte responses to specific groups of pathogens causing subclinical mastitis. The MLD measures the absolute number and relative percentage of inflammatory cells in milk samples. Using the MLD in two dairy herds (170 and 172 lactating cows, respectively), we studied all lactating cows with a most recent monthly Dairy Herd Improvement Association somatic cell count (SCC) >200 × 103 cells/ml. Quarter milk samples from 78 cows meeting study criteria were analysed by MLD and aseptically collected milk samples were subjected to microbiological culture (MC). Based upon automated instrument evaluation of the number and percentage of inflammatory cells in milk, samples were designated as either MLD-positive or – negative for subclinicial mastitis. Positive MC were obtained from 102/156 (65·4%) of MLD-positive milk samples, and 28/135 (20·7%) of MLD-negative milk samples were MC-positive. When MC was considered the gold standard for mastitis diagnosis, the calculated diagnostic Se of the MLD was 65·4% (IC95% = 57·4 to 72·8%) and the Sp was 79·3% (IC95% = 71·4 to 85·7%). Quarter milks positive on MC had higher absolute numbers of neutrophils, lymphocytes and macrophages, with higher neutrophils% and lymphocytes% but lower macrophages%. The Log10 (N/L) ratios were the most useful ratio to differentiate specific subclinical mastitis quarters from healthy quarters. Use of the MLD on cows with monthly composite SCC > 200 × 103 cells/ml for screening at quarter level identified quarters more likely to be culture-positive.

In conclusion, the MLD can provide an analysis of mammary quarter status more detailed than provided by SCC alone; however, the MLD response to subclinical mastitis was not found useful to specifically identify the causative pathogen.

We determined the effects of feeding canola oil or infusing it into the abomasum on rumen fermentation, nutrient digestibility, duodenal flows of fatty acids, and milk composition in Holstein cows. Five ruminally and duodenally cannulated Holstein cows in late lactation were used in a 3×5 incomplete Latin square design. Treatments were 1) Control: basal diet (CON), 2) Control+supplementation of canola oil at 1 kg/d in the feed (FED), and 3) Control+abomasal infusion of canola oil at 1 kg/d (INF). Compared with CON, feed intake, ruminal fermentation characteristics, ruminal and total tract digestibilities of nutrients were not significantly affected by FED treatment but duodenal flows and milk concentrations of fatty acids (FA) such as trans-11 18[ratio ]1 and cis-9 trans-11 18[ratio ]2 (conjugated linoleic acid, CLA) were increased. In contrast to the effects of FED, INF reduced feed intake, total VFA production, intestinal flows of nutrients, FA digestibility and yields of milk and milk fat. Both FED and INF significantly reduced the proportions of saturated and medium-chain FA, and increased cis 18[ratio ]1 in milk. Concentrations of 18[ratio ]2n-6 and 18[ratio ]3n-3 in milk were increased nearly 2-fold with INF relative to CON. Dietary or postruminal supplementation of canola oil to late-lactation cows reduced saturated FA and increased unsaturated C18 in milk but nutrient digestion was adversely affected with abomasal infusion of canola oil.

The effect of the stage of lactation, the number of foals and age of the mare on changes in the fat content and fatty acid composition of colostrum and milk of primitive Konik horses was investigated. Colostrum and milk samples from 12 lactating mares were collected at the beginning of lactation, on the days 1 and 2 after foaling and then, starting from the first month of lactation, at 4-week intervals up to the sixth month of lactation. Significant differences were observed in fat content as well as the composition of some analysed fatty acids between colostrum and milk of mares of the Konik breed. The number of foalings and the age of mares did not have a statistically significant effect on the fat content in milk and had only a slight effect on the fatty acid composition. Milk produced by mares of the Konik breed is characterized by a considerable content of polyene fatty acids with 18 carbon atoms, a low ratio of n-6 fatty acids to n-3 fatty acids as well as low, highly advantageous values of atherogenic and thrombogenic indices.

The hyperinsulinaemic euglycaemic insulin clamp technique was used to study the effect of insulin on the arterio-venous concentration differences of glucose and amino acids across the mammary gland in dairy goats. Insulin was given in conjunction with K to prevent insulin hypokalaemia. Appropriate amino acid infusion was used to blunt insulin-induced hypoaminoacidaemia or to create hyperaminoacidaemia and maintain this state under insulin treatment. Hyper-aminoacidaemia alone only stimulated mammary leucine uptake but did not significantly modify the net metabolism of other amino acids and glucose. Insulin infusion at physiological level in conjunction with glucose, KCl-NaCl and amino acids failed to alter mammary uptake of glucose and essential amino acids; occasional increase in arginine extraction and decrease in tyrosine extraction were exceptions. Thus these new experimental conditions did not reveal any galactopoietic effect of insulin.

For six successive milkings, six dairy cows were relocated immediately before milking to an unfamiliar operating theatre, a procedure previously shown to inhibit oxytocin release and milk ejection. Two control milkings were performed in familiar surroundings. After milk flow had ceased, two i.v. injections of 1 i.u. oxytocin were given to remove the remaining milk. Milk flow was recorded continuously and blood samples were taken every minute during milking and 10 min after milking. During the first milking in unfamiliar surroundings, no oxytocin was released. Thereby, only 13% of the total milk yield, the cisternal milk, was available and the alveolar milk fraction could only be removed after injection of oxytocin. During subsequent relocations oxytocin release steadily increased toward the control level, although the timing of oxytocin release remained delayed as compared with controls. However, the milk fraction available before oxytocin injection increased with increasing number of removals, following an asymptotic approach to control levels. The concentrations of β-endorphin, cortisol (and perhaps also of prolactin) gradually declined with the number of times the animal was moved to unfamiliar surroundings, i.e. hormone concentrations gradually adjusted to control level. During milking, concentrations of prolactin and cortisol increased, while β-endorphin concentrations decreased (except for the first relocation). We conclude that milking-related oxytocin release and therefore milk ejection adapted gradually to repeated relocations to unfamiliar surroundings. This adaptation was inversely related to β-endorphin concentrations, so it is possible that oxytocin release was suppressed by high circulating β-endorphin concentrations.

This investigation analysed the applicability of principal component analysis (PCA), a latent variable method, for the early detection of mastitis and lameness. Data used were recorded on the Karkendamm dairy research farm between August 2008 and December 2010. For mastitis and lameness detection, data of 338 and 315 cows in their first 200 d in milk were analysed, respectively. Mastitis as well as lameness were specified according to veterinary treatments. Diseases were defined as disease blocks. The different definitions used (two for mastitis, three for lameness) varied solely in the sequence length of the blocks. Only the days before the treatment were included in the blocks. Milk electrical conductivity, milk yield and feeding patterns (feed intake, number of feeding visits and time at the trough) were used for recognition of mastitis. Pedometer activity and feeding patterns were utilised for lameness detection. To develop and verify the PCA model, the mastitis and the lameness datasets were divided into training and test datasets. PCA extracted uncorrelated principle components (PC) by linear transformations of the raw data so that the first few PCs captured most of the variations in the original dataset. For process monitoring and disease detection, these resulting PCs were applied to the Hotelling's T2 chart and to the residual control chart. The results show that block sensitivity of mastitis detection ranged from 77·4 to 83·3%, whilst specificity was around 76·7%. The error rates were around 98·9%. For lameness detection, the block sensitivity ranged from 73·8 to 87·8% while the obtained specificities were between 54·8 and 61·9%. The error rates varied from 87·8 to 89·2%. In conclusion, PCA seems to be not yet transferable into practical usage. Results could probably be improved if different traits and more informative sensor data are included in the analysis.

An automatic purge and trap apparatus, coupled to a GC-MS was used to study the seasonal variability of the volatile fraction of raw milk Manchego cheese. Both season and dairy significantly affected abundance of most volatile compounds. Most aldehydes, methyl ketones, n-alcohols, and secondary alcohols reached significantly higher concentrations in spring cheeses. Branched chain alcohols showed significantly higher concentrations in autumn and winter cheeses, while significantly higher amounts of diketones were found in summer cheeses. Most ethyl esters reached higher concentrations in spring and winter cheeses and lower in autumn cheeses. Lower concentrations of α-pinene were found in spring cheeses, and higher amounts of limonene were observed in winter cheeses. Heptane and octane were significantly more abundant in summer cheeses. No significant seasonal differences were found either for quality or intensity scores.

Two groups of eight multiparous cows with different calving periods (November or February) were managed in the same way during lactation. During four 4 week experimental periods distributed over 12 months, the cows were fed on a diet composed of hay and concentrate (70[ratio ]30) in a restricted and controlled amount which varied according to their lactation stage, so as to cover the animals' requirements correctly. The animals' average lactation stage varied according to period from 26 to 298 d. Milk from each group was processed on two occasions during each period to make Saint-Nectaire-type cheese; the cheesemaking conditions were the same throughout. Lactation stage had an important effect on milk fat, protein and calcium contents but not on the casein[ratio ]protein ratio or phosphorus content. The milk pH and the urea content were higher in late lactation. The calcium concentration of milk was higher in late lactation but the soluble fraction was higher in early lactation. Despite higher protein contents, the maximal firmness of the coagulum of late-lactation milk was not different from that of early or mid-lactation milks. pH was higher in cheeses from late-lactation milks compared with those from early and mid-lactation milks. Cheeses from early lactation milks were more yellow than the others and had a lower dry matter fat content. In sensory analysis the odour of cheeses from early and late-lactation milks was less pleasant than that of those from mid-lactation milks. Cheeses from late-lactation milks were more melting and less firm than those from early or mid-lactation milks. Their taste was more intense and more persistent. At tasting, they were less appreciated than the others. These differences were linked to increased proteolysis in the cheeses made with late-lactation milks.
