Chrysoperla species include well-known predators of aphids and other soft-bodied arthropods. As such, they are considered important biological control agents of herbivorous pests in agroecosystems where many of green lacewings species occur. Despite the high number of species of the genus Chrysoperla, only a few have been assessed for the predation efficiency of their larvae against pests infesting plants, and even fewer are currently marketed for use in biocontrol practice. Difficulties in species identification within the Chrysoperla carnea complex species in particular has been related to varying success of commercial C. carnea s.l. releases in the field. In this study, we assessed the ability of two Chrysoperla species, Chrysoperla agilis a member of the carnea cryptic species group, and Chrysoperla mutata of the pudica group to consume aphid and mealybug individuals and suppress their populations in sweet pepper plants. We found that third-instar larvae of both species were able to consume a high number of aphids (approximately 120 nymphs per larva) and mealybugs (approximately 105 nymphs per larva) within 24 h. Furthermore, the release of second-instar larvae of both C. agilis and C. mutata was shown to be remarkably efficient in suppressing the pest populations in long-term greenhouse experiments. Aphid populations were suppressed by approximately 98% and mealybugs by 78% as compared to control plants. Our results highlight the predation efficiency and the biocontrol potential of two widespread Chrysoperla species for their use in pest control.

The relative costs and benefits of thermal acclimation for manipulating field performance of pest insects depend upon a number of factors including which traits are affected and how persistent any trait changes are in different environments. By assessing plastic trait responses of Ceratitis capitata (Mediterranean fruit fly) across three distinct operational environments (laboratory, semi-field, and field), we examined the influence of different thermal acclimation regimes (cool, intermediate [or handling control], and warm) on thermal tolerance traits (chill-coma recovery, heat-knockdown time, critical thermal minimum and critical thermal maximum) and flight performance (mark-release-recapture). Under laboratory conditions, thermal acclimation altered thermal limits in a relatively predictable manner and there was a generally positive effect across all traits assessed, although some traits responded more strongly. By contrast, dispersal-related performance yielded strongly contrasting results depending on the specific operational environment assessed. In semi-field conditions, warm- or cold-acclimated flies were recaptured more often than the control group at cooler ambient conditions suggesting an overall stimulatory influence of thermal variability on low-temperature dispersal. Under field conditions, a different pattern was identified: colder flies were recaptured more in warmer field conditions relative to other treatment groups. This study highlights the trait- and context-specific nature of how thermal acclimation influences traits of thermal performance and tolerance. Consequently, laboratory and semi-field assessments of dispersal may not provide results that extend into the field setting despite the apparent continuum of environmental complexity among them (laboratory < semi-field < field).

Laboratory tests were carried out in order to examine the population growth of Prostephanus truncatus (Horn) (Coleoptera: Bostrychidae) and Sitophilus oryzae (L.) (Coleoptera: Curculionidae) on maize. These two species were placed either simultaneously or one species was allowed to colonize the kernels 7 days earlier than the other, at two temperatures, 26 and 30 °C for 65 days. Apart from progeny production, grain quality parameters, such as insect-damaged kernels (IDK) and undamaged kernels (NDK), the weight of frass and kernel weight were measured. Our data confirms that temperature plays a key role in the competition of these two species; P. truncatus seems to perform better at the higher temperature (30 °C), regardless of the presence of an additional species. Moreover, the results of the present study demonstrates that P. truncatus outcompetes S. oryzae. Sitophilus oryzae produced fewer progeny than P. truncatus in all combinations. Given the outcome of a competition, we hypothesize that most of the kernel damage was due to feeding by P. truncatus. Based on these data, we surmise that P. truncatus has a competitive advantage as an invasive species in new areas with stored maize, even in the presence of S. oryzae.

Stenoponia tripectinata tripectinata (Tiraboschi, 1902) is the most prevalent subspecies, within the genus Stenoponia, in the Mediterranean area. This rodent flea is widely distributed throughout southwestern Europe and the North of Africa including Mediterranean islands and the Canary Islands. Nevertheless, from a taxonomical and systematic point, this flea group has been neglected over the years. Therefore, the aim of this study was to carry out a comparative morphometric, phylogenetic, and molecular study of two populations of S. t. tripectinata isolated from rodents collected from different islands from the Canary Archipelago and from Corsica to clarify the taxonomic status of these two isolated populations and to assess the morphological and molecular differentiation between them. For this purpose, we have analyzed several morphological traits and sequenced five molecular markers (EF1-α, ITS2, cox1, cox2, and cytb). We observed slight differences in the overall body size between females of both populations, and two well-defined geographical genetic lineages. This suggests the existence of two cryptic subspecies within S. t. tripectinata corresponding to two different island groups. Furthermore, we bring to light the necessity to provide new and updated morphological, molecular, and phylogenetic data to clarify the taxonomic status of S. tripectinata.

A model for the Aedes aegypti lifecycle is developed that takes into account temperature-dependent maturation and death rates for several life stages, wet and dry egg oviposition with flooding, as well as three classes of larval habitat with different temperature profiles: outdoor (subject to external temperature fluctuations, human-inhabited), indoor (temperature moderated, human-inhabited, interior), and enclosed (temperature moderated, human free, exterior). An equilibrium analysis shows that the temperature range of outdoor viable equilibrium populations aligns closely with reported risk levels. Temperature patterns for El Paso, Texas; New York, New York; New Orleans, Louisiana; Orlando, Florida; and Miami, Florida, are considered. In four of these locations (all but New York), enclosed habitats can support mosquito populations even if all outdoor and indoor habitats are removed. In two locations (El Paso and New York) the model shows that in spite of the disappearance of adult mosquitoes during colder temperatures, populations reach seasonal steady state due to the survival of eggs. The results have implications for both vector and disease control.

Energy homeostasis is essential for organisms to maintain fluctuation in energy accumulation, mobilization. Lipids as the main energy reserve in insects, their metabolism is under the control of many physiological program. This study aimed to determine whether the adipokinetic hormone receptor (AKHR) was involved in the lipid mobilization in the Spodoptera litura. A full-length cDNA encoding AKHR was isolated from S. litura. The SlAKHR protein has a conserved seven-transmembrane domain which is the character of a putative G protein receptor. Expression profile investigation revealed that SlAKHR mRNA was highly expressed in immatural stage and abundant in fat body in newly emerged female adults. Knockdown of SlAKHR expression was achieved through RNAi by injecting double-stranded RNA (dsRNA) into the 6th instar larvae. The content of triacylgycerol (TAG) in the fat body increased significantly after the SlAKHR gene was knockdown. And decrease of TAG releasing to hemolymph with increase of free fatty acid (FFA) in hemolymph were observed when the SlAKHR gene was knowned-down. In addition, lipid droplets increased in fat body was also found. These results suggested that SlAKHR is critical for insects to regulate lipids metabolism.

Several studies have shown the negative effects of mature plant resistance (MPR) on aphids in sugar beet, which is correlated to the formation of black deposits in their stomach. However, the underlying mechanism of MPR still needs to be elucidated, by understanding the toxicity effects of MPR on aphids and the role of the plant phenological stage and the environment. Here, we report that MPR in sugar beet does not only affect Myzus persicae mortality rate and the formation of a black deposit in the aphid stomach, but also aphid fecundity and behaviour. In addition, experiments in climate-controlled and field settings showed quantitative variation in MPR to M. persicae between six genotypes of sugar beet. Our results indicate that environmental effects, such as temperature, play a major role in MPR and underscore the importance of proper climate-controlled experiments for investigating MPR. In climate-controlled experiments, 83.3% of aphids on old leaves developed a black deposit, in contrast to only 16.8% of aphids on young leaves. This shows that not only plant age, but also leaf age plays a major role in the intensity of MPR. Further research will be needed to identify the underlying mechanism, before MPR can be used as a viable and sustainable solution to aphid pests in sugar beet.

Insects are mass-reared for release for biocontrol including the sterile insect technique. Insects are usually reared at temperatures that maximize the number of animals produced, are chilled for handling and transport, and released into the field, where temperatures may be considerably different to those experienced previously. Insect thermal biology is phenotypically plastic (i.e. flexible), which means that there may exist opportunities to increase the performance of these programmes by modifying the temperature regimes during rearing, handling, and release. Here we synthesize the literature on thermal plasticity in relation to the opportunities to reduce temperature-related damage and increase the performance of released insects. We summarize how and why temperature affects insect biology, and the types of plasticity shown by insects. We specifically identify aspects of the production chain that might lead to mismatches between the thermal acclimation of the insect and the temperatures it is exposed to, and identify ways to harness physiological plasticity to reduce that potential mismatch. We address some of the practical (especially engineering) challenges to implementing some of the best-supported thermal regimes to maximize performance (e.g. fluctuating thermal regimes), and acknowledge that a focus only on thermal performance may lead to unwanted trade-offs with other traits that contribute to the success of the programme. Together, it appears that thermal physiological plasticity is well-enough understood to allow its implementation in release programmes.

The highest natural mortality rate of larval Lepidoptera in field populations occurs in the first instar, but it is highly variable. The pattern and degree of survival is not easily predicted but depends on their ability to establish on host plants. Lepidopteran larval dispersal behaviour, known as ‘drop-off’, happens when the host is unsuitable for larvae to settle and begin feeding. Understanding drop-off behaviour of Helicoverpa armigera (Hübner) with and without physiological resistance to Bt toxins on Bt and non-Bt cotton plants is an important component for resistance management strategies for this insect. We examined the drop-off behaviour of H. armigera to determine: (1) whether they move the same way or differently in response to Bt and non-Bt, and (2) could H. armigera larvae detect Bt toxin levels in cotton plants or did they move independently of toxin levels? In this study, we assessed the drop-off behaviour of Bt-resistant and Bt-susceptible H. armigera neonates on artificial diets and cotton plants with and without Bt toxin during the first 12 h after hatching. Bt-resistant and Bt-susceptible H. armigera neonates behaved differently on Bt and non-Bt substrates. The percentages of Bt-resistant larvae that dropped off Bt and non-Bt cotton plants were not significantly different. In contrast, significantly more Bt-susceptible larvae dropped off Bt cotton than non-Bt cotton plants over time. Although Bt-susceptible larvae could not detect Bt toxin, they showed preference on non-Bt toxin substrates and were more likely to drop off substrates with Bt toxin.

Aedes-borne diseases are on the rampant rise despite continued application of chemical insecticide-based interventions. The appearance of high degree of insecticide resistance in Aedes species and noxious effects on environment and non-targets have raised further concerns. Among new chemical interventions, neonicotinoids are considered a safe and effective approach. The present study investigated the control potency of acetamiprid and development of resistance in Aedes aegypti larvae; and the involvement of CYP450 monooxygenases in inducing resistance. The early fourth instars of Ae. aegypti parent susceptible strain (PS) were selected with acetamiprid for 15 generations (ACSF strain) increasing the resistance to 19.74-fold in ACSF-10 and 36.71-fold in ACSF-15. The ACSF-10 larvae were assayed with acetamiprid combined with piperonyl butoxide (PBO) in three different ratios (1:1, 1:5 and 1:10) and selected for next five generations with 1:10 combination. Selection with synergized acetamiprid (APSF strains) reversed as well as reduced the rate of resistance development resulting in only 1.35-fold resistance in APSF-15. The APSF strains showed %monooxygenase dependency ranging from 86.71 to 96.72%. The estimation of the monooxygenases levels in parent and selected larvae showed increased monooxygenase level in the ACSF strains by 2.42–2.87-fold. The APSF-15 strains exhibited 57.95% lower enzyme production than ACSF-15 strain. The reduction and reversion of resistance by using PBO and the elevated levels of monooxygenases in ACSF and reduction in APSF strains recommend the involvement of CYP450-mediated mechanism in the development of acetamiprid resistance in Ae. aegypti. These studies could help in devising resistance management strategies in order to preserve the efficiency of pre-existing insecticides.

While the desire for systemic repellents is high, ineffective repellents put one at risk of insect-vectored pathogens. Vitamin B1, or thiamine, has been touted as a systemic insect repellent since 1943, and denounced as an ineffective placebo for just as long. This paper presents a scoping review of 104 relevant case reports, research studies, and review articles to trace the evolution of this idea and identify an evidence-based, scientific consensus. Reports of thiamine's systemic repellency are primarily anecdotal and based on uncontrolled trials and/or used bite symptoms as a proxy for reduced biting. Controlled experiments on insect landing and feeding found no evidence of repellency. Of the 49 relevant review papers, 16 insect bite prevention guidelines, and 4 government documents, none after the 1990s claimed thiamine is a repellent. The findings of this review are that thiamine cannot repel arthropods in any dosage or route of administration. Due to limited available evidence, the possibility that thiamine reduces the subjective symptoms of insect bites cannot currently be ruled out. Unfortunately, many medical professionals and travelers today still believe thiamine may be effective despite the evidence stating otherwise. Continued promotion of debunked repellents on the commercial market poses a serious risk in countries with the endemic, mosquito-vectored disease.

Whiteflies are one of the major pests of tomato under greenhouses, and their control partly relies on biocontrol strategies. Among those biocontrol agents, parasitoids or predators are widely used. However, the introduction of a biocontrol agent in a new area is not trivial. For that reason, we investigated the use of a tropical native mirid, Nesidiocoris volucer (Hemiptera: Miridae), for the biological control of whiteflies among other insect pests on tomato crops under greenhouses in the subtropical island of La Réunion, France. Nesidiocoris volucer life history traits and plant injury were examined. Nymphs developed and survived between 15 and 30°C and required on average 49.41 days at 15°C and on average 10.50 days at 30°C to develop (nymph survival >94%). At 25°C, each female produced on average 65 eggs. Nesidiocoris volucer was able to feed on several prey species, but performed better on whiteflies than on spider mites or thrips. No N. volucer feeding injury was observed on tomato. Nesidiocoris volucer has also been found in tropical countries of Africa, and we believe that the data presented on this natural enemy could be of great importance for the biocontrol of whiteflies in tropical areas.

In this study, we present the first characterization of herbivore-induced plant volatiles (HIPVs) released from infested olive leaves. The gall midge Dasineura oleae is a specific pest of Olea europaea and endemic of the Mediterranean Basin, an area in which severe outbreaks currently occurred. Little is known about the damage caused by the pest and the relationship with its host. Since gall formation and larval feeding activity may lead to the release of specific plant volatile compounds, we investigated the volatile profiles emitted from infested plants compared with healthy plants under both laboratory and field conditions. Additionally, the volatiles emitted from mechanically damaged plants were considered. A blend of 12 volatiles was emitted from olive trees infested by D. oleae. Of these, β-copaene, β-ocimene, cosmene, unknown 1 and unknown 3 were found to be exclusively emitted in infested plants. The emission of germacrene-D, (E,E)-α-farnesene, and (Z,E)-α-farnesene, α-copaene, (E)-4,8-dimethylnona-1,3,7-triene, (E)-β-guaiene and heptadecane significantly increased in infested trees. Linalool, β-copaen-4-α-ol, β-bourbonene, β-cubebene, β-elemene, β-copaene and δ-amorphene were found only in the field trial and showed differences depending on the level of infestation and the plant stage. (Z)-3-Hexenol, (E)-4-oxohen-2-enal, and 2-(2-butoxyethoxy)-ethanol, were exclusively emitted from the leaves after mechanical damage. In a field trial in Italy, we also demonstrated spring synchronization between adults of D. oleae and O. europaea trees. Analyses of morphoanatomical malformations of gall leaves showed that tissue alterations occur at the spongy parenchyma causing an increase of the leaf blade thickness. We speculate that tissue alterations may lead to HIPV release, in turn potentially attracting D. oleae natural enemies.

Florida red scale, Chrysomphalus aonidum (L.) is a species of armored scales, which attacks citrus crops. Biological control, particularly ladybeetles, are critical for its management in citrus production systems. We evaluated predation of C. aonidum by adult and larvae (3rd–4th instar) of the metallic blue ladybeetle, Curinus coeruleus. C. coeruleus were tested against C. aonidum in three separate treatments on Valencia orange leaves in Petri-dish arenas (1) individuals with armor intact, (2) individuals with armor removed, and (3) mix of individuals with armor intact or removed, to determine if armor inflicts distraction to predator consumption of scale and impact. Within 24 h of exposure to C. aonidum, adult beetle consumption rate averaged 64–68% in the two treatments containing all or half of the individuals with armor, compared to 100% in the treatment with armor removed. The consumption rate in the former two treatments increased to 83–89% within 72 h. Larval consumption of C. aonidum with armor intact was 25% at 24 h and 41% at 72 h, compared to 92–100% in the treatment with armor removed. The adults resulting from the larvae developed on C. aonidum with the armor intact or between the diets of armor intact or removed consumed more scales without armor than with armor. Significant consumption of C. aonidum by adults and larvae of C. coeruleus indicates that it is an efficient predator of this pest species. These novel findings suggest that this predator could be useful for suppressing C. aonidum populations in citrus production systems, particularly in habitats where both species are established such as Florida.

Eristalinus aeneus (Scopoli, 1763) is a suitable candidate for artificial rearing due to its pollination efficiency and subcosmopolitan distribution. However, the high mortality found at the larval stage of this species needs to be overcome. In this research, two different larval media were used to study the life cycle of E. aeneus: brewery spent grain (BSG) from a local craft-beer factory and soaked oat grains (SOG). The age-stage, two-sex life table method was used to analyze the results, which were compared using the paired bootstrap test. The greatest mortality was found at the larval stage with both media. Individuals fed on SOG presented a shorter preadult developmental time (22.05 days) than those reared with BSG (26.97 days). This fact had a direct impact on the total preoviposition period, it being shorter with SOG (34.36 days) than BSG (38.29 days), although the second provided a larger total number of eggs (19,242 eggs) and a faster adult maturation (10.67 days). The population parameters indicated that both populations will display a positive growth under the studied conditions, being the mean generation time (T) significantly shorter when using SOG (38.71 days) than BSG (45.95 days). Despite the preadult results pointing to SOG being a more efficient medium, the promising fecundity values provided by BSG, as well as it's lower cost and ecological benefits, suggest that this second medium could be improved and used as an alternative to SOG in the near future.

Insect crop pests are a major threat to food security in sub-Saharan Africa. Configuration of semi-natural habitat within agricultural landscapes has the potential to enhance biological pest control, helping to maintain yields whilst minimising the negative effects of pesticide use. Fall armyworm (Spodoptera frugiperda, J. E. Smith) is an increasingly important pest of maize in sub-Saharan Africa, with reports of yield loss between 12 and 45%. We investigated the patterns of fall armyworm leaf damage in maize crops in Ghana, and used pitfall traps and dummy caterpillars to assess the spatial distribution of potential fall armyworm predators. Crop damage from fall armyworm at our study sites increased significantly with distance from the field edge, by up to 4% per m. We found evidence that Araneae activity, richness and diversity correspondingly decreased with distance from semi-natural habitat, although Hymenoptera richness and diversity increased. Our preliminary findings suggest that modifying field configuration to increase the proximity of maize to semi-natural habitat may reduce fall armyworm damage and increase natural enemy activity within crops. Further research is required to determine the level of fall armyworm suppression achievable through natural enemies, and how effectively this could safeguard yields.

Agriculture is one of the major farming activities, representing 32% of the gross domestic product of Madagascar and 74.3% of the population is involved in this activity. Fruit flies of the Tephritidae family are considered as the most destructive pests for agriculture in the country, nevertheless, few data exist on host plants and distribution of those pests. In the present study, we address those questions by conducting a large survey between November 2016 and July 2018 across the six agroecological regions of Madagascar. Fruit and vegetable were sampled from 198 plant species (wild and cultivated) and represented 37,965 fruits from all regions of Madagascar. The infestation index ranged from 0.06 to 538.46 pupae/kg, the infestation percentage was up to 54.84% in some samples, 63 plant species were considered as host of Tephritidae. Twelve fruit fly species were identified, seven of which were previously described as endemic, five species could be considered as widespread (altitudinal gradients between 1 and 1634 m asl) and major pests in Madagascar: Ceratitis malgassa (23 plant species from 12 families), Neoceratitis cyanescens (16 plant species from one family), Bactrocera dorsalis (18 plant species from 12 families), Dacus demmerezi (ten plant species from one family), Dacus vertebratus (six species from one family). Those results are of importance for implementation of control measures.

The most commercialized Bt maize plants in Europe were transformed with genes which express a truncated form of the insecticidal delta-endotoxin (Cry1Ab) from the soil bacterium Bacillus thuringiensis (Bt) specifically against Lepidoptera. Studies on the effect of transgenic maize on non-target arthropods have mainly converged on beneficial insects. However, considering the worldwide extensive cultivation of Bt maize, an increased availability of information on their possible impact on non-target pests is also required. In this study, the impact of Bt-maize on the non-target corn leaf aphid, Rhopalosiphum maidis, was examined by comparing biological traits and demographic parameters of two generations of aphids reared on transgenic maize with those on untransformed near-isogenic plants. Furthermore, free and bound phenolics content on transgenic and near-isogenic plants were measured. Here we show an increased performance of the second generation of R. maidis on Bt-maize that could be attributable to indirect effects, such as the reduction of defense against pests due to unintended changes in plant characteristics caused by the insertion of the transgene. Indeed, the comparison of Bt-maize with its corresponding near-isogenic line strongly suggests that the transformation could have induced adverse effects on the biosynthesis and accumulation of free phenolic compounds. In conclusion, even though there is adequate evidence that aphids performed better on Bt-maize than on non-Bt plants, aphid economic damage has not been reported in commercial Bt corn fields in comparison to non-Bt corn fields. Nevertheless, Bt-maize plants can be more easily exploited by R. maidis, possibly due to a lower level of secondary metabolites present in their leaves. The recognition of this mechanism increases our knowledge concerning how insect-resistant genetically modified plants impact on non-target arthropods communities, including tritrophic web interactions, and can help support a sustainable use of genetically modified crops.

The olive fruit fly, Bactrocera oleae, the most serious pest of olives, requires the endosymbiotic bacteria Candidatus Erwinia dacicola in order to complete its development in unripe green olives. Hence a better understanding of the symbiosis of Ca. E. dacicola and its insect host may lead to new strategies for reduction of B. oleae and thus minimize its economic impact on olive production. Studies of this symbiosis are hampered as the bacterium cannot be grown in vitro and the established B. oleae laboratory populations, raised on artificial diets, are devoid of this bacterium. Here, we sought to develop a method to transfer the bacteria from wild samples to laboratory populations. We tested several strategies. Cohabitation of flies from the field with the laboratory line did not result in a stable transfer of bacteria. We provided the bacteria directly to the egg and also in the food of the larvae but neither approach was successful. However, a robust method for transfer of Ca. E. dacicola from wild larvae or adults to uninfected flies by transplantation to females was established. Single female lines were set up and the bacteria were successfully transmitted for at least three generations. These results open up the possibilities to study the interaction between the symbiont and the host under controlled conditions, in view of both understanding the molecular underpinnings of an exciting, unique in nature symbiotic relationship, as well as developing novel, innovative control approaches.

The European (Melolontha melolontha L.) and Forest (M. hippocastani F.) cockchafer are widespread pests throughout Central Europe. Both species exhibit a 3–5-year life cycle and occur in temporally shifted populations, which have been monitored and documented for more than 100 years. Visual identification of adults and larvae belonging to these morphologically similar species requires expertise and, particularly in the case of larvae, is challenging and equivocal. The goal of the study was the development of an efficient and fast molecular genetic tool for the identification and discrimination of M. melolontha and M. hippocastani. We established a collection of both species from Switzerland, Austria and Northern Italy in 2016, 2017 and 2018. An approximately 1550 bp long fragment of the cytochrome c oxidase subunit 1 (CO1) mitochondrial gene was amplified and sequenced in 13 M. melolontha and 13 M. hippocastani beetles. Alignment of the new sequences with reference sequences (NCBI GenBank and BOLDSYSTEMS databases) and subsequent phylogenetic analysis revealed consistent clustering of the two species. After the identification of M. melolontha and M. hippocastani species-specific single nucleotide polymorphisms (SNPs) in the CO1 alignment, we developed an effective SNP tool based on the ABI PRISM® SNaPshot™ Multiplex Kit for the rapid and accurate species discrimination of adults and larvae.
