The relationships between the Japanese beetle (JB) Popillia japonica Newman, 1841 and the grapevine agroecosystem were investigated in Piedmont in 2020 and 2021, to assess the impact of the species and its distribution within vineyards in relation to the proximity of environmental risk factors. Grubs were sampled by soil coring in the inter-rows of vineyards, whereas both adult beetles and defoliation were counted directly on grapevine plants. The presence of spatial autocorrelation was assessed and the influence of environmental variables (distance from woodlands, meadows and the margin of the vineyard, soil parameters, year of sampling, and year of first detection of the JB) was evaluated through generalized linear mixed models. Beetles and defoliation were more clustered at the edges of vineyards, whereas grubs were localized in few hot spots, generally close to meadows. Spatial autocorrelation was weaker for grubs with respect to adults and defoliation. Grub density depended on distance from meadows, and partially on soil features. Adults abundance was influenced by the proximity to meadows, woodlands, and their presence was clustered at the margin of vineyards. The JBs seem to rely on grapevine mainly as a food source rather than a reproductive site, preferring meadows for egg-laying: therefore, pest management in vineyards should be more focused on adult beetles rather than larvae in the vineyard inter-rows.

We evaluated the lethal and sublethal effects of two novel Betaproteobacteria-based insecticides (Burkholderia spp. strain A396 as Venerate® XC; Chromobacterium subtsugae strain PRAA4-1 as Grandevo® WDG) and two neem-based insecticides (1.2% azadirachtin A and B as Azatrol and 3% azadiractin as Molt-X) on the cotton aphid, Aphis gossypii, and its natural enemy, Coccinella septempunctata. Aphids were given both residual and direct treatments, i.e. exposed to residues applied by leaf dipping, or by spraying the insects and foliage, while the predator was treated directly with insecticides. Well-established spirotetramat (Movento® 240 SC) was used as standard due to its effectiveness against a wide range of pests, its unique mode of action, and its systemic properties. All insecticides were effective against aphid mostly in concentration-dependent manner, as do exposure time, but at different magnitudes. Spirotetramat and Azatrol induced the highest toxicity to adult aphids, while spirotetramat and Molt-X were more noxious to aphid nymphs. C. subtsugae and Burkholderia were less effective, inducing only moderate levels of aphid mortality. Azatrol and spirotetramat were more detrimental to the fecundity of aphid compared to other products. Insecticides significantly increased the development time of nymphs surviving exposure to insecticides, except Burkholderia. Azatrol were more destructive to eggs, larvae and adult of Coccinella septempunctata, together with spirotetramat for young larvae and adults, relative to other treatment. The development time of predator larvae remained unaffected by treatment. New Betaproteobacteria- and neem-based insecticides except Azatrol seem to be a promising tool to suppress population of Aphis gossypii and integrate pest management programmes.

The canopy of forests as the ‘last biotic frontier’ has often been neglected in insect biodiversity studies because it is harder to access compared to the understorey, even in relatively well-known temperate ecosystems. We investigated the diversity, abundance, and body size patterns of macromoths (Lepidoptera) in the canopy and understorey in a central European deciduous forest. We collected moths at two sites during 19 trapping nights and three lunar phases between July and September 2021 using a weak ultraviolet light emitting diode (LED) lamp (LepiLED mini). Overall, we captured 4368 individuals (165 species) from 11 families. Based on a number of metrics, richness and diversity was significantly lower in the canopy than in the understorey. Non-metric multidimensional scaling ordinations show that communities largely overlap, but the proportion of species that only occur in the understorey was higher. While Noctuidae and Erebidae species were abundant in both strata, Geometridae species were most frequently observed in the understorey. We identified 16 indicator species for the understorey but only three for the canopy. Forewing length of moths in the canopy was on average 1.7 mm longer than of those in the understorey. Overall, the understorey is far more important for moths than the canopy in a temperate forest. The canopy is dominated by fewer and larger species and probably has a higher proportion of dispersers.

In December 2017 the Venetian Region (local Authority), financed the creation of the Operational Group (OG) ‘Serinnovation’, within the framework of the Rural Development Plan supported by the European Community. The OG aims at coordinating and spreading innovation in sericulture through mechanisation of processes and centralisation of some rearing steps, the use of waste as by-products and traceability to promote local productions. The project acts on perceived quality by increasing the added value, through production cost efficiency, and on the recovery of the waste material for further applications (circular economy). The final target was to develop a niche-process to obtain traceable ‘Made-in-Italy’ silk for the luxury market and non-textile applications. A first strategy to increase the efficiency of the process was to build an automatic leaf cutting machine to prepare the feed for the first three instars of the silkworm (Bombyx mori Linnaeus). This new machine – based on a patent – was validated through several tests and compared with the cutting system previously used. The study was completed by a bioassay of production and survival rate associated with the introduction of this innovation. The results showed that labour saving is in the order of 10% compared to a semi-manual process, the leaf quality is not affected, survival of larvae and silk production are not significantly different from the control. This methodology is proposed as a study case for other similar mechanisation processes in entomological production, as the impact of innovations on insect physiology should be carefully considered.

One of the most prominent problems related to biological invasions is the variation of local species composition, which often leads to ex novo interspecific interactions. Here, we explored and analysed the native species composition of gall inducers and their associated parasitoids and inquilines in Spanish areas invaded by Dryocosmus kuriphilus Yasumatsu 1951 (Hymenoptera: Cynipidae), an invasive pest of chestnut trees. After a quantitative description of these species' assemblages, we analysed through bipartite networks the level of the trophic specialisation of parasitoids and inquilines when considering either the host taxonomic identity, the host plant species or the host gall morphological type. We sampled galls of D. kuriphilus and native species of Cynipidae in different Spanish areas, including those where the exotic parasitoid Torymus sinensis Kamijo 1982 (Hymenoptera: Torymidae) had been released for D. kuriphilus biological control. The results indicate that the native parasitoids recruited by D. kuriphilus come almost exclusively from native communities on Quercus galls, except for one species from Rosa. Galls of D. kuriphilus had the second most diverse species composition; despite this species assemblage arose ex novo in less than a decade. The bipartite networks resulted more specialised when considering host plant taxa than when gall types and the host taxa were accounted. In such trophic webs, there were few parasitoid/inquiline specialist and many generalist species, which agrees with the rapid recruitment by D. kuriphilus. Higher parasitoid species richness in D. kuriphilus galls is likely due to their being a largely unexploited available resource for the native natural enemies of cynipid wasps.

Triatomines (Hemiptera: Heteroptera: Reduviidae) are hematophagous insects, well-known for their vectorial role in transmitting Trypanosoma cruzi Chagas (Kinetoplastida: Trypanosomatidae) parasites, the etiological agent of Chagas disease. Trapping these insects would limit human-triatomine interaction and, thus, control the disease. In this context, there is a critical need for effective lures to control triatomines. Through double-choice bioassays, we investigated the preference of Triatoma infestans Klug, T. pallidipennis Stal, and Rhodnius prolixus Stal triatomines for: (a) volatiles from fermented products (various fermentation types and substrates) and (b) commercial insect lures. Furthermore, we identified the chemical composition of these volatiles through headspace collection using Solid Phase Micro Extraction coupled with Gas Chromatograph-Mass Spectrometer (HS-SPME-GC-MS). Volatiles from lactic fermentation and certain fermented fruits, along with commercial lures, attracted triatomines, while other products exhibited possible repellent or dislodging properties. These findings hold promise for the control of triatomines and, ultimately, Chagas disease.

Negative relationships between the parental age and offspring life history traits have been widely observed across diverse animal taxa. However, there is a lack of studies examining the influence of parental age on offspring performance using mites, particularly phytoseiid predators as subjects. This study explored the influence of maternal age on offspring life history traits in Amblyseius herbicolus (Chant) (Acari: Mesostigmata), a phytoseiid predatory mite reproducing through thelytokous parthenogenesis. We hypothesised that increased maternal age negatively impacts offspring traits, including developmental duration, body size, fecundity and lifespan. Amblyseius herbicolus was reared under controlled laboratory conditions, and the life history traits of offspring from mothers of varying ages were analysed using linear mixed-effect models. Our results showed that the increase in maternal age significantly reduced individual egg volume, but did not significantly affect offspring developmental duration, body size, fecundity or lifespan. These findings indicate that while older A. herbicolus females produced smaller eggs, the subsequent performance (i.e. body size, fecundity and lifespan) of offspring remained largely unaffected, suggesting possible compensatory mechanisms in the offspring or alternative maternal provisioning strategies. The results of this study offer useful insights into the reproductive strategies of phytoseiid predators and asexually reproducing species, enhancing our understanding of how maternal age affects offspring fitness. Further studies can examine how offspring of A. herbicolus from mothers of different ages perform under adverse environmental conditions.

Deformed wing virus (DWV), which is an important honey bee virus transmitted by Varroa destructor (V. destructor), causes colony losses in honey bee colonies. This study aimed to investigate the prevalence and genetic diversity of DWV in honey bees in Türkiye and to determine the role of V. destructor in the transmission of the genetic variants of DWV. Honey bee samples were collected from 62 apiaries, by simple random sampling, during March 2022 and April 2023. The presence of V. destructor in collected bee samples was examined using a stereo microscope. Real-time RT-PCR was used for the detection of DWV-A and DWV-B (Varroa destructor virus-1 (VDV-1)) viruses. Genetic characterisation of the positive samples was conducted by sequencing polyprotein genomic region. Considering the V. destructor infestation rate of 3% as relevant, out of the 62 apiaries examined, 17 (27.4%) were positive. However, DWV-A and VDV-1 specific RNA was not detected in V. destructor samples. VDV-1 specific RNA was detected in 6.5% (4/62) of the apiaries, whereas DWV-A was not detected in the sampled apiaries. Phylogenetic analysis showed that isolates detected in this study were located in a separate cluster from previously characterised DWV-A and VDV-1 isolates. According to RDP4 and GARD analyses, DWV-VDV-1 recombination breakpoints were detected in field isolates. To the best our knowledge, this is the first report of the presence of VDV-1-DWV recombinants in Türkiye. Further studies are needed to determine the impact of VDV-1-DWV recombinants and their virological and antigenic properties.

Paranosema locustae is an environmentally friendly parasitic predator with promising applications in locust control. In this study, transcriptome sequencing was conducted on gonadal tissues of Locusta migratoria males and females infected and uninfected with P. locustae at different developmental stages. A total of 18,635 differentially expressed genes (DEGs) were identified in female ovary tissue transcriptomes, with the highest number of DEGs observed at 1 day post-eclosion (7141). In male testis tissue transcriptomes, a total of 32,954 DEGs were identified, with the highest number observed at 9 days post-eclosion (11,245). Venn analysis revealed 25 common DEGs among female groups and 205 common DEGs among male groups. Gene ontology and Kyoto Encyclopaedia of Genes and Genome analyses indicated that DEGs were mainly enriched in basic metabolism such as amino acid metabolism, carbohydrate metabolism, lipid metabolism, and immune response processes. Protein–protein interaction analysis results indicated that L. migratoria regulates the expression of immune- and reproductive-related genes to meet the body's demands in different developmental stages after P. locustae infection. Immune- and reproductive-related genes in L. migratoria gonadal tissue were screened based on database annotation information and relevant literature. Genes such as Tsf, Hex1, Apolp-III, Serpin, Defense, Hsp70, Hsp90, JHBP, JHE, JHEH1, JHAMT, and VgR play important roles in the balance between immune response and reproduction in gonadal tissues. For transcriptome validation, Tsf, Hex1, and ApoLp-III were selected and verified by quantitative real-time polymerase chain reaction (qRT-PCR). Correlation analysis revealed that the qRT-PCR expression patterns were consistent with the RNA-Seq results. These findings contribute to further understanding the interaction mechanisms between locusts and P. locustae.

Aphis gossypii is one of the most economically important agricultural pests that cause serious crop losses worldwide, and the indiscriminate chemical application causes resistance development in A. gossypii, a major obstacle to successful control. In this study, we selected the up-regulated expression gene AgJHAMT, which was enriched into juvenile hormone pathway though transcriptome sequencing analysis of the cotton aphids that fed on transgenic cotton lines expressing dsAgCYP6CY3 (the TG cotton). The AgJHAMT gene was overexpressed in cotton aphids which fed on the TG cotton, and its expression profile during the nymphs was clarified. Then, silencing AgJHAMT could advance the developmental period of cotton aphids by 0.5 days compared with control groups. The T and t values of cotton aphids in the dsJHAMT treatment group (6.88 ± 0.15, 1.65 ± 0.06) were significantly shorter than that of the sprayed H2O control group (7.6 ± 0.14, 1.97 ± 0.09) (P < 0.05), respectively. The fast growth caused by AgJHAMT silencing was rescued by applying the JH analogue, methoprene. Overall, these findings clarified the function of AgJHAMT in the developmental period of A. gossypii. This study contributes to further clarify the molecular mechanisms of delaying the growth and development of cotton aphids by the transgenic cotton lines expressing dsAgCYP6CY3.

Drosophila suzukii is a pest of global concern due to its great impact on several crops. The entomopathogenic nematode Steinernema carpocapsae was highly virulent to the larvae of the fly although some immune mechanisms were triggered along the infection course. Thus, to understand the gene activation profile we performed a comparative transcriptome of D. suzukii larvae infected with S. carpocapsae and Xenorhabdus nematophila to map the differentially expressed genes involved in the defence response. The analysis exposed the induction of genes involved in the humoral response such as the antimicrobial peptides and pattern-recognition receptors while there was a suppression of the cellular defence. Besides, genes involved in melanisation, and clot formation were downregulated hindering the encapsulation response and wound healing. After the infection, larvae were in a stress condition with an enrichment of metabolic and transport functionalities. Concerning the stress response, we observed variations of the heat-shock proteins, detoxification, and peroxidase enzymes. These findings set a genetical comprehensive knowledge of the host-pathogen relation of D. suzukii challenged with S. carpocapsae which could support further comparative studies with entomopathogenic nematodes.

Many parasitoids alter their reproductive behaviour in response to the quality of encountered hosts. They make adaptive decisions concerning whether to parasitise a potential host, the number of eggs laid on an accepted host, and the allocation of sex to their offspring. Here we present evidence that Goniozus jacintae Farrugia (Hymenoptera: Bethylidae), a gregarious ectoparasitoid of larval tortricids, adjusts its reproductive response to the size and developmental stage of larvae of the light brown apple moth (LBAM), Epiphyas postvittana (Walker) (Lepidoptera: Tortricidae). Goniozus jacintae parasitises instars 3–6 of LBAM, but most readily parasitises the later, larger, instars. Brood sizes were bigger on larger hosts and brood sex ratios were female biased (proportion of males = 0.23) with extremely low variance (never >1 male in a brood at emergence), perhaps the most precise of all studied bethylids. Host size did not influence brood development time, which averaged 19.64 days, or the body size of male offspring. However, the size of females was positively correlated with host size and negatively correlated with brood size. The sizes of individual males and females were positively related to the average amount of host resource available to individuals within each brood, suggesting that adult body size is affected by scramble competition among feeding larvae. Average brood sizes were: 3rd instar host, 1.3 (SE ± 0.075); 4th instar, 2.8 (SE ± 0.18); 5th instar, 4.7 (SE ± 0.23); 6th instar, 5.4 (SE ± 0.28). The largest brood size observed was 8 individuals (7 females, 1 male) on the 6th instar of LBAM. These results suggest that later instars would give the highest yield to optimise mass-rearing of G. jacintae if used for augmentative biological pest control.

The social-sexual environment is well known for its influence on the survival of organisms by modulating their reproductive output. However, whether it affects survival indirectly through a variety of cues without physical contact and its influence relative to direct interaction remain largely unknown. In this study, we investigated both the indirect and direct influences of the social-sexual environment on the survival and reproduction of the mite Tyrophagus curvipenis (Acari: Acaridae). The results demonstrated no apparent influence of conspecific cues on the survival of mites, but the survival and reproduction of mated female mites significantly changed, with the females mated with males having a significantly shortened lifespan and increased lifetime fecundity. For males, no significant difference was observed across treatments in their survival and lifespan. These findings indicate that direct interaction with the opposite sex has a much more profound influence on mites than indirect interaction and highlight the urgent need to expand research on how conspecific cues modulate the performance of organisms with more species to clarify their impacts across taxa.

Two fruit fly (Diptera: Tephritidae) species of economic importance: Ceratitis rosa Karsch and Ceratitis quilicii De Meyer, Mwatawala & Virgilio are present in South Africa. The two species were considered as one species prior to 2016, but were subsequently separated. In this study, the distribution and abundance of the two species were quantified in seven provinces in South Africa through trapping with Enriched Ginger Oil as an attractant. Trapping was conducted over three seasons across two years (2020 and 2021): late summer, autumn-winter, and spring-early summer. Host ranges of the two species were investigated by fruit sampling in and outside of trapping sites. Ceratitis quilicii was more widely distributed than C. rosa with the latter being recorded in only three north-eastern provinces. There were geographical limits for both species with no records of them in Northern Cape Province. Catches of C. quilicii were higher in summer with average temperatures varying from 15 to 27°C while for C. rosa, catches remained low and consistent between seasons. Ceratitis quilicii catches decreased at lower rates than those of C. rosa at temperatures below 15°C. The two species were reared from 13 plant species from nine families. Four of these hosts were infested by both C. quilicii and C. rosa in the same province where they occurred. Preferred hosts of the two species belonged to the Myrtaceae family. The characterisation of the distribution, abundance and host ranges of these pests will provide a baseline for pest status determination and implementation of management actions.

Aphidius colemani is an important biological control agent, used in greenhouses and open fields against aphid pests. Despite this economical importance, A. colemani, along with A. transcaspicus and A. platensis, has gone through a complex taxonomical history. The three species have only recently gained status as separate species again, comprising the morphologically defined Aphidius colemani species group. Other than sporadic records probably as a consequence of escape from greenhouses, the A. colemani species group members prefer warmer regions and there are numerous records from South America, Southern Europe and Asia. Based on slide-mounted material collected in the period 1964–2001 in Africa, we describe five new species belonging to this group, and report A. colemani, A. transcaspicus and A. platensis from several African countries. This data opens questions about the origin of the group and presents potential for the diversification of biological control agents against aphid pests.

The eggplant fruit and shoot borer (EFSB) is a devastating pest of eggplants (Solanum aethiopicum L. and Solanum melongena L.) in Ghana, causing significant economic losses. Although initially thought to be the Leucinodes orbonalis Guenee species found in Asia, recent European and Mediterranean Plant Protection Organization reports suggest its absence in Africa. However, eight Leucinodes species have been recently described in Africa, including two new species, Leucinodes africensis sp. n. and Leucinodes laisalis Walker, which were intercepted in eggplant fruits exported from Ghana to the United Kingdom. Despite the reported absence of L. orbonalis in Africa, it remains on the pest list of Ghana as a species known to attack eggplants. To accurately determine the identity of the EFSB complex occurring on eggplant in Southern Ghana, molecular and morphological taxonomic tools were employed, and adult male populations were monitored in on-farm conditions. Our results revealed the presence of two EFSB species, L. africensis and L. laisalis, in the shoot and fruits of eggplants, with L. africensis being the dominant species and widely distributed in Southern Ghana. Notably, L. africensis males were attracted to the pheromone lure of L. orbonalis despite the two species being biologically distinct. This study provides crucial information on correctly identifying the EFSB species attacking eggplants in Southern Ghana and has significant implications for developing management interventions against these pests and their effects on international eggplant trade.

Puparia are commonly found in tsetse fly larviposition sites during studies on larval ecology. This chitinous shell is representative of past or ongoing exploitation of these sites by tsetse flies. The morphological characteristics of the puparium are not sufficiently distinctive to allow identification of the species. This study explores the applicability of biomolecular techniques on empty puparia for tsetse fly species identification. Five techniques were compared for DNA extraction from tsetse fly puparia, 1/Chelex® 100 Resin, 2/CTAB, 3/Livak's protocol, 4/DEB + proteinase K and 5/QIAamp® DNA Mini kit, using two homogenisation methods (manual and automated). Using a combination of two primer pairs, Chelex, CTAB, and DEB + K proved the most efficient on fresh puparia with 90, 85, and 70% samples identified, respectively. Shifting from fresh to one- to nine-month-old puparia, the Chelex method gave the best result allowing species identification on puparia up to seven months old. The subsequent testing of the Chelex extraction protocol identified 152 (60%) of 252 field-collected puparia samples at species level. The results show that reliable genetic identification of tsetse flies species can be performed from empty puparia, what can prove of great interest for future ecological studies on larviposition sites. The Chelex technique was the most efficient for DNA extraction, though the age-limit of the samples stood at seven months, beyond which DNA degradation probably compromises the genetic analysis.

Peach-potato aphids, Myzus persicae Sulzer (Hemiptera:Aphididae), and cabbage aphids, Brevicoryne brassicae Linnaeus (Hemiptera:Aphididae), are herbivorous insects of significant agricultural importance. Aphids can harbour a range of non-essential (facultative) endosymbiotic bacteria that confer multiple costs and benefits to the host aphid. A key endosymbiont-derived phenotype is protection against parasitoid wasps, and this protective phenotype has been associated with several defensive enodsymbionts. In recent years greater emphasis has been placed on developing alternative pest management strategies, including the increased use of natural enemies such as parasitoids wasps. For the success of aphid control strategies to be estimated the presence of defensive endosymbionts that can potentially disrupt the success of biocontrol agents needs to be determined in natural aphid populations. Here, we sampled aphids and mummies (parasitised aphids) from an important rapeseed production region in Germany and used multiplex PCR assays to characterise the endosymbiont communities. We found that aphids rarely harboured facultative endosymbionts, with 3.6% of M. persicae and 0% of B. brassicae populations forming facultative endosymbiont associations. This is comparable with endosymbiont prevalence described for M. persicae populations surveyed in Australia, Europe, Chile, and USA where endosymbiont infection frequencies range form 0–2%, but is in contrast with observations from China where M. persicae populations have more abundant and diverse endosymbiotic communities (endosymbionts present in over 50% of aphid populations).

Mosquito-borne disease is a significant public health issue and within Australia Ross River virus (RRV) is the most reported. This study combines a mechanistic model of mosquito development for two mosquito vectors; Aedes vigilax and Aedes camptorhynchus, with climate projections from three climate models for two Representative Concentration Pathways (RCPs), to examine the possible effects of climate change and sea-level rise on a temperate tidal saltmarsh habitat in Perth, Western Australia. The projections were run under no accretion and accretion scenarios using a known mosquito habitat as a case study. This improves our understanding of the possible implications of sea-level rise, accretion and climate change for mosquito control programmes for similar habitats across temperate tidal areas found in Southwest Western Australia. The output of the model indicate that the proportion of the year mosquitoes are active increases. Population abundances of the two Aedes species increase markedly. The main drivers of changes in mosquito population abundances are increases in the frequency of inundation of the tidal wetland and size of the area inundated, increased minimum water temperature, and decreased daily temperature fluctuations as water depth increases due to sea level changes, particularly under the model with no accretion. The effects on mosquito populations are more marked for RCP 8.5 when compared to RCP 4.5 but were consistent among the three climate change models. The results indicate that Ae. vigilax is likely to be the most abundant species in 2030 and 2050, but that by 2070 Aedes camptorhynchus may become the more abundant species. This increase would put considerable pressure on existing mosquito control programmes and increase the risk of mosquito-borne disease and nuisance biting to the local community, and planning to mitigate these potential impacts should commence now.

From the 1990s, the Southeast Asia native giant resin bee Megachile sculpturalis (Smith, 1853) was introduced first to North America, and then to many countries in Europe. Despite increasing studies on its invasive potential and geographical expansion, information on nesting behaviour of this species is still extremely scarce. To increase knowledge on the nesting biology of M. sculpturalis, we studied multiple aspects of nesting and pollen provisioning in three consecutive years in artificial nests in Bologna, Italy. We observed 166 bees visiting nests, and followed individual nesting behaviour and success of 41 adult females. We measured cavity diameter in 552 nests and characterised the structure in 100 of them. More than 95% of nest diameters ranged between 0.6 and 1.2 cm, overlapping with several sympatric species of cavity-nesting hymenopterans in the study area. Most nests had a first chamber from the entrance of variable length without brood, followed by an average of about two brood cells with a mean length of 2.85 ± 0.13 cm each. The pollen stored in brood cells was almost monofloral, belonging to the ornamental plant Styphnolobium japonicum (L.) Schott. We estimated that a single female should visit ≈180 flowers to collect enough pollen for a single brood cell. These results fill knowledge gaps on the nesting biology and nest structure of the exotic M. sculpturalis, and they are discussed in relation to possible competition with native bees for nesting sites and foraging resources.
