Seeds are complex structures that serve as dispersal units in angiosperms. Seeds consist of three specialized tissues with distinct roles and molecular compositions. Hence, the characterization of the genetic regulators that act within individual seed tissues, and how their activity changes during seed development and germination, has been a primary focus of seed research. However, our knowledge of the spatiotemporal modulation of genetic regulators within seeds, across different seed cell types, has been limited by the resolution of available techniques. In the last few years, the development and application of single-cell technologies in plants have enabled the elucidation of gene networks involved in various developmental processes at the cellular level. Some studies have applied these technologies to seeds, enabling further characterization of seed development and germination at the cellular level. Here, we review the current status of the application of single-cell technologies to seeds and present a workflow for conducting single-cell transcriptomics. Additionally, we discuss the integration of single-cell multi-omics, aiming to demonstrate the potential of single-cell technologies in enhancing our comprehension of the spatiotemporal regulations governing seed development and germination.

Endozoochory, the dispersal of seeds through the animal gut passage, plays a significant role in vegetation dynamics. The success of endozoochorous seed dispersal depends on each stage of the process: ingestion by animals, gut passage, and post-dispersal events after defecation. After the deposition of seeds through feces, the effects of feces on the initial stages of seedling establishment, including seed germination and seedling growth, can significantly impact overall survival. The pattern of fecal effects on plant species depends on the animal species. In this study, we investigated the effects of feces presence on seed germination and early seedling growth using feces of the Korean water deer (Hydropotes inermis argyropus). We conducted a germination experiment on 12 plant species belonging to 10 plant families, which are known to germinate in the feces of Korean water deer. The study compared the seed germination rate and seedling length after germination between seeds sown with and without feces of the Korean water deer. In general, we found that the presence of deer feces per se had no significant effects on seed germination and early growth stages. However, additional research on post-dispersal events such as long-term growth, fecal type, and germination conditions is needed to fully understand the costs and benefits of endozoochory.

Echinochloa crus-galli var. crus-galli (L.) P. Beauv. (EC), Echinochloa crus-galli var. mitis (Pursh) Petermann (ECM), and Echinochloa glabrescens Munro ex Hook. f. (EG) are all serious rice (Oryza sativa L.) weeds that are usually treated as a single species in weed management practices. To determine interspecific and intraspecific differences in seed germination responding to different temperatures among the three Echinochloa weeds, we conducted field surveys and collected 66 EC, 141 ECM, and 120 EG populations from rice fields of East China in 2022; and tested their seed germination under 28/15 C (day/night), 30/20 C, and 35/25 C regimes, simulating temperatures of rice-planting periods for double-cropping early rice, single-cropping rice, and double-cropping late rice, respectively. In EC, ECM, and EG, seed percentage germination (cumulative percentage of germinated seed) and germination index (sum of the ratio of germinated seeds to the corresponding days) increased with increasing temperatures. At 28/15 C, the average percentage germination of EC populations (67.5%) was significantly (P < 0.05) higher compared with ECM (46.4%) and EG (43.7%); GD50 (duration for 50% total germination) for EC populations (5.2 d) was significantly shorter compared with ECM (5.9 d) and EG (5.8 d). At 35/25 C, the percentage germinations of EC (90.7%), ECM (80.5%), and EG (80.3%) were all significantly the highest among the three temperature treatments, respectively, and the GD50 values for EC (2.5 d), ECM (2.6 d), and EG (2.7 d) were all significantly the lowest. At 30/20 C and 35/25 C, the average germination percentages of populations collected from transplanted rice fields were significantly higher than those of populations collected from direct-seeded rice fields. Moreover, among EG populations, the longitudes and latitudes of collection locations were significantly correlated with seed percentage germination and germination indices. According to the interspecific differences and intraspecific variations of Echinochloa species, weed management strategies should also be customized according to the species and population characteristics in seed germination.

We have reviewed seed dormancy and germination in the Rubiaceae, the fourth-largest angiosperm family (in terms of species richness), in relation to ecology, life form, biogeography and phylogeny (subfamily/tribe). Life forms include trees, shrubs, vines and herbs, and tropical rainforest trees have the greatest number of tribes and species. The family has five kinds of embryos: investing, linear-full, linear-underdeveloped, spatulate and spatulate-underdeveloped, and seeds are non-dormant (ND) or have morphological (MD), morphophysiological (MPD) or physiological (PD) dormancy. Except for the occurrence of the investing embryo only in dry fruits of Dialypetalanthoideae, each kind of embryo is found in dry and fleshy fruits of Dialypetalanthodies and of Rubioideae. In tropical and temperate regions, there are species with ND seeds and others whose seeds have MD, MPD or PD. A complete seed dormancy profile (i.e. some species with ND seeds and others whose seeds have MD, MPD or PD) was found for tropical rainforest trees and shrubs and semi-evergreen rainforest shrubs. Dormancy-break occurs during cold or warm stratification or dry-afterripening, depending on the species. Some tropical species have long periods of dormancy-break/germination extending for 4–5 to 30–40 weeks. Soil seed banks are found in 5 and 15 tribes of Rubiaceae in tropical and temperate regions, respectively. With increased distance from the Equator, diversity of life forms and seed dormancy decreases, resulting in only herbs with PD at high latitudes. We conclude that the low species richness of Rubiaceae in temperate regions is not related to low diversity of seed dormancy/germination.

Nypa fruticans Wurmb is both a relic plant and a true mangrove. In China, wild populations are distributed only on Hainan Island and face significant challenges in regeneration from seedlings. This study explored the underlying causes of recruitment limitation by examining seed morphological traits from three distinct populations (Haikou, Wenchang and Wanning) and analysing seed germination and seedling growth characteristics under varying conditions. The key findings are as follows: fruiting and seed-setting rates for N. fruticans were notably low, standing at only 21 and 40%, respectively. The Wanning population exhibited significantly higher rates compared to the other two populations. Under natural conditions, the germination and seedling emergence rates were also modest, at 36.58 and 22.99%, respectively. The germination and emergence rates of the Wanning population were significantly greater than those of the Haikou and Wenchang populations. Meanwhile, seeds from a single population did not differ in germination rates among three in situ N. fruticans habitats, but seedling emergence rates differed significantly. Optimal conditions for seed germination involved a light intensity of 60%, a salinity of 5‰ and a flooding time of 8 h/day. In natural settings, these three environmental factors fall short of the ideal conditions. The study underscores that light, salinity and flooding are primary factors contributing to the limitations in N. fruticans seedling recruitment. In addition to advocating increased investment in scientific research and technology to address seed source issues, we recommend heightened efforts in habitat restoration, in situ conservation and the optimization of relocation and field return strategies to bolster N. fruticans populations.

Seed germination is a pivotal period of plant growth and development. This process can be divided into four major stages, swelling absorption, seed coat dehiscence, radicle emergence and radicle elongation. Cupressus gigantea, a tree native to Tibet, China, is characterized by its resistance to stresses such as cold, and drought, and has a high economic and ecological value. Nevertheless, given its unique geographic location, its seeds are difficult to germinate. Therefore, it is crucial to explore the mechanisms involved in seed germination in this species to improve the germination efficiency of its seeds, thereby protecting this high-quality resource. Here, our findings indicate that seed germination was enhanced when exposed to a 6-h/8-h light/dark photoperiod, coupled with a temperature of 20°C. Furthermore, the application of exogenous GA3 (1 mg/ml, about 2.9 mM) stimulated the germination of C. gigantea seeds. Subsequently, proteomics was used to detect changes in protein expression during the four stages of seed germination. We identified 34 differentially expressed proteins (DEPs), including 13 at the radicle pre-emergence stage, and 17 at the radicle elongation stage. These DEPs were classified into eight functional groups, cytoskeletal proteins, energy metabolism, membrane transport, stress response, molecular chaperones, amino acid metabolism, antioxidant system and ABA signalling pathway. Most of them were found to be closely associated with amino acid metabolism. Combined, these findings indicate that, along with temperature and light, exogenous GA3 can increase the germination efficiency of C. gigantea seeds. Our study also offers insights into the changes in protein expression patterns in C. gigantea seeds during germination.

Alpine plant species are particularly vulnerable to climate change. Temperature fluctuations are projected to be most severe at high elevations. Even small shifts in temperature have major consequences on phenology, reproduction, and community composition. Early life stages are arguably the most important processes in the fitness of an individual plant and the dynamics and persistence of plant populations. These initial developmental stages are expected to be more vulnerable to changes in climate than adult life stages. To understand how early life stages of alpine plant species will respond to warming temperatures, seeds and seedlings of two species were exposed to three different temperature regimes. Temperatures were based on current and projected conditions under low and high emission scenarios. Two rare alpine species performed better under warmer temperatures at both the germination and seedling stages. The results show that early life stages of alpine plants may not be at high risk from warming temperatures; however, there are many other shifting climatic factors to consider, resulting from climate change beyond temperature alone.

Understanding the timing of weed emergence is crucial to effective management. Management practices implemented too early may fail to completely control late-emerging seedlings, whereas management practices implemented too late will suffer from low efficacy. Weed emergence times reflect biological factors, such as seed dormancy and germination requirements, as well as environmental conditions. We conducted a systematic review of studies that developed models to predict weed emergence temporal patterns. We screened 1,854 studies, 98 of which were included in the final dataset. Most of the studies included were conducted in North America (51%) or Europe (30%). A wide variety of weed species (102) and families (21) were included, and many studies modeled several weeds. Grass weeds (Poaceae) were modeled most frequently (83 instances). Most weeds (40%) had base temperature ${T_{\rm{b}}}$ values between 0 and 5 C, and 38% had base water potential ${{\rm{\psi }}_{\rm{b}}}$ ranging from −1.0 to −0.5 MPa. Most studies used empirical parametric models, such as Weibull (40%) or Gompertz (30%) models. Nonparametric and mechanistic models were also represented. Models varied in their biological and environmental data requirements. In general, empirical parametric models based on hydrothermal time (i.e., time above base temperature and water potential thresholds) represented a good balance between ease of use and prediction accuracy. Soft computing approaches such as artificial neural networks demonstrated substantial potential in situations with complex emergence patterns and limited data availability, although they (soft computing approaches) can be susceptible to overfitting. Our study also demonstrated variability in model performance and limited generalizability across species and regions. This finding underscores the need for context-specific and well-validated weed emergence models to inform management, especially in the context of climate change.

Desiccation tolerance (DT) of seeds, one of the plant's environmental adaptation mechanisms, allows them to survive as seeds in a quiescent state under extremely water-deficient conditions during the plant's life cycle, followed by seed germination and seedling establishment under favourable water conditions. The seed-DT is lost after radicle emergence; however, there is a developmental period called the ‘DT window’ during which the germinating seeds can re-induce DT following a cue from their ambient low water potential (i.e. mild osmotic stress). The DT re-inducibility within the DT window has been used as a model biosystem for understanding molecular mechanisms that activate/supress DT in a number of plant species. However, the characteristics of the DT window for species particularly important to the agroindustry are still largely fragmented. Here, physiological analyses were performed, aiming to elucidate the properties of the DT window in tomato, a model species for Solanaceae, holding a key strategic position for the seed industry and commercial use around the world. We revealed that (i) the DT window of tomato seeds is closed when the developing radicle reaches about 4 mm after germination, (ii) the most effective ambient water potential to re-induce DT into seeds is about −1.5 MPa and (iii) there is organ specificity of DT re-induction with hypocotyls, showing a longer DT window than cotyledons and roots in post-germination seeds.

Small isolated plant populations are one of the consequences of fragmentation of natural habitats by humans. We asked what effect does the creation of smaller populations from larger ones has on the plant fitness-related trait seed germination. Using information on 119 species (142 species entries) in 50 families, we found that seeds in only 35.2% of the species entries from larger populations germinated to higher percentages than those from smaller populations. In the other entries, seeds from large and small populations germinated equally well (57.7% of total entries) or seeds from small populations germinated better (7.0% of total entries) than those from large populations. These results indicate that population size is not a reliable predictor of seed germinability. Furthermore, there was little relationship between seed germination and either seed mass, genetic diversity or degree of population isolation, or between population size and genetic diversity.

The intraspecific variations of phenotypic traits in the early life stages, such as seed germination and seedling establishment, are important components affecting species adaptation and differentiation. As one of the most common ways in which seeds are maintained, dry storage usually affects these traits, by either increasing or decreasing variation among populations. However, little is known about how the interaction between dry storage and population variation of a species affects the performance and adaptability of early life-cycle traits. In this study, we conducted experiments with seeds from ten populations of Elymus nutans along an altitudinal gradient on the eastern Qinghai-Tibet Plateau. Fresh seeds and seeds stored dry for 6 months were used to quantify the temperature thresholds for germination, determine seedling emergence and survival in two common gardens and examine the correlation between these traits and environmental conditions of population provenance. Dry storage increased germination percentage, germination speed and seedling emergence, and decreased intraspecific variation in germination traits of seeds (reduced by 33.36, 52.05 and 20.45% for Tb, θT(50) and σθT, respectively). Dry storage had little effect on the intraspecific variation of seedling emergence and survival. In addition, the temperature threshold for germination cannot be used to predict seedling emergence and survival in either common garden, regardless of whether seeds were stored or not. These results indicated that it is feasible to use dry-stored seeds to evaluate and select suitable provenances in ecological restoration, and using dry-stored seeds in ecological restoration projects is expected to achieve better vegetation restoration results than fresh seeds. On the other hand, the status of seeds (fresh vs. dry stored) should be considered in an evaluation of the adaptive value of plant functional traits, especially in the early life stages, otherwise, inconsistent conclusions may be drawn.

Invasive nonindigenous species pose a serious threat to native biodiversity and ecosystem functioning. Understanding how species’ performance varies under conditions in the current and invaded range can help to predict the dynamics of the invading species in its new environment. Plants with the ability to alter growth in response to variation in light conditions may be favored in landscapes that experience frequent disturbance, as these species may be able to exploit a wide range of niches. Seedbank persistence may also play a critical role in successful plant invasion, as extended seed viability may increase the chance of outlasting unfavorable conditions, maintain population genetic diversity, and allow reinvasions. This study investigated seed longevity and the effect of light intensity on germination of wavyleaf basketgrass [Oplismenus undulatifolius (Ard.) Roem. & Schult.], a newly established invasive species in U.S. mid-Atlantic forest understories. Oplismenus undulatifolius seeds were collected across 5 yr from the original site of introduction in Maryland, USA, and stored in standard lab conditions, then subjected to germination trials under four light conditions in a controlled growth chamber. Seeds remained viable for at least 9 yr, and light intensity did not significantly impact seed germination. Our study demonstrates the importance of evaluating environmental and temporal effects on germination traits, because the scope of surveillance in the field may need to be expanded based on new information about environmental tolerance. Long-term monitoring may also be necessary to effectively control invasive plant populations capable of forming a persistent seedbank.

Verbascum blattaria L., commonly known as moth mullein, naturalized in the USA that produces white or yellow flowers could be considered as a potential ornamental plant. However, genetic characterization using molecular markers and leaf morphology, colourimetric analysis and flowering of V. blattaria influenced by low temperature treatments was not investigated to evaluate as a potential horticultural and landscape plant use. The basal leaves developed during the rosette-growth stage were oblanceolate with an obtuse leaf apex and incisions at the margin. Leaves produced on the stem during the reproductive development were ovate or lanceolate with an obtuse or acute leaf apex. Regardless of the colour of the petiole and leaf blade during the rosette-growth stage, there were no differences in the sequences of nuclear ribosomal ITS and chloroplast interspacer. All plants produced creamy white flowers with a purplish base corolla. All leaves formed during the vegetative and reproductive development were glabrous. Numerous stalked glandular trichomes were observed in the sepal, pedicel and bracts. Seeds started to germinate in 10 days at 25°C and reached the plateau in 30 days after sowing. The earliest flowering occurred in 131 days when the plants received 20 days of low temperature treatment (CD) (20 CD), producing 76 flowers, the highest number compared to the number of flowers produced by plants that received 0, 40 and 60 CD. Plants that received 20 CD exhibited early flowering, probably because of the early transition from vegetative growth to reproductive development, as judged by the short stem to the first flower.

Southern European peninsulas have repeatedly played notable roles as refuges in the natural history of flora during periods of glaciation. Euonymus latifolius (Celastraceae) is a relict species from the last Würmian glaciation in the Iberian Peninsula. It still lives with isolated populations in favourable, cool mountainous microhabitats, being an extremely rare, critically endangered species. These Iberian populations are often high-priority targets for conservation due to their long-term persistence and unique evolutionary trajectory. Previously, it has not been feasible to promote significant programmes for reintroduction and/or population reinforcement of this singular plant species due to the great difficulties of conventional propagation. In this study, we analysed the effects of temperature, light and gibberellic acid (GA3) on the germination responses of E. latifolius to develop an effective protocol for seed germination as a main outcome. The results are coherent with the climatic temperature conditions recorded broadly in the Iberian Peninsula in the past and in the current refuge locations for the taxon. The germination responses of E. latifolius are compatible with those of seeds with intermediate physiological dormancy. In particular, the seeds required a 10-week warm period (20/7°C + 15/4°C) followed by 20 weeks of cold period (5 + 1.5°C) to break dormancy and achieve germination values over 90%. GA3 also promoted germination (80%). Therefore, we developed the first effective protocol for promoting E. latifolius seed germination and, thus, sexual propagation, to facilitate urgent ex situ actions in the current climate change context.

Pulsatilla chinensis (Bunge) Regel has been widely used in the pharmaceutical industry. With the deepening of clinical application, the research on its plant resources has attracted much attention. However, the underlying molecular mechanisms of distinct germination during Pulsatilla seed development are still mostly unknown. Therefore, in this study, four germination stages of P. chinensis seeds, with obvious differences in seed appearance traits, were used as materials. Transcriptome sequencing technology was used to analyse the molecular mechanisms of seed germination. A total of 27,601 differentially expressed genes (DEGs) (six different groups) were determined. KEGG enrichment analysis revealed that the up-regulated DEGs were enriched in phenylpropanoid biosynthesis, photosynthesis, photosynthesis–antenna proteins, plant hormone signal transduction, flavonoid biosynthesis and other pathways. A total of 87 DEGs was enriched in phytohormone signal transduction pathways, including auxin (25), abscisic acid (13), gibberellin (6), ethylene (9) and cytokinin (7). Furthermore, a protein–protein interaction network was constructed using these DEGs. Some DEGs were validated by qRT-PCR analysis. This comprehensive analysis provided basic information on the key genes of plant hormone signal transduction pathways involved in the seed germination process of P. chinensis (Bunge) Regel.

Dormancy-breaking requirements and level of morphophysiological dormancy (MPD) were determined for seeds of Ilex formosana and I. uraiensis from the subtropical region, and seeds of I. rotunda from both the subtropical and tropical regions of Taiwan. We hypothesized that some Ilex species would have deep simple MPD broken by warm stratification. Germination of seeds and embryo growth was monitored at 30/20, 25/15, 20/10, 15/5°C and at 25°C. Seeds were cold-stratified and then incubated at 25/15°C, and seeds treated with GA3 and GA4 were incubated at 25/15°C. Fresh seeds reached 50% germination after 11–45 weeks of warm stratification. Pre-treatment with GA increased germination percentages of I. formosana and I. rotunda (tropical) but not I. uraiensis and I. rotunda (subtropical), while cold stratification did not promote germination of either species but increased the germination rate of I. rotunda (tropical). Embryo length in seeds of all species increased ≥710% prior to root emergence, and growth occurred during warm stratification. The positive response to GA and relatively short time for beginning of germination and to reach 50% germination indicate non-deep simple MPD in seeds of I. formosana and I. rotunda (tropical). The negative response to GA and long time for beginning of germination and to reach 50% germination indicate deep simple MPD in seeds of I. uraiensis and I. rotunda (subtropical). Thus, in both the subtropical and tropical regions of Taiwan, the seeds of Ilex species have non-deep simple and deep simple MPD that are broken by warm stratification. Furthermore, GA treatment increases the germination rate and percentage of Ilex seeds with non-deep simple MPD, and cold stratification promotes the seed germination rate of Ilex species with non-deep simple MPD in tropical region.

Reproductive strategies for specific populations are closely related to environmental factors. Consequently, they are fundamental for conservation plans and the management of threatened habitats like salt marshes. From this viewpoint, germination strategy under different temperatures and salt conditions, voltammetric parameters and molecular analysis were performed and compared in six Halocnemum populations (four of H. cruciatum and two of H. strobilaceum) growing on Mediterranean (Balearic, Tyrrenic and Adriatic) and Red Sea coasts to establish the relation to environmental variables. Significant interpopulation differences were found in all the evaluated parameters. The Mediterranean populations showed a variable opportunistic germination strategy that was directly related to the drought period length at the studied sites. Consequently, potential environmental predictors of seed response were identified. The most noteworthy were bioclimate, soil texture, continentality index, winter temperatures and summer precipitations. Additionally, voltammetric parameters were evidenced as indicators of maternal plant stress levels and, thus, as potential determinants of future seed responses. The phylogenetic analyses showed a split into two species that did not correspond to germination response. The phylogeographic analyses showed interpopulation differences in haplotype composition for H. cruciatum, but not for H. strobilaceum. In conclusion, the tight connection between seed responses and the ecological parameters of natural populations as an adaptation for successful seedling emergence was proved regardless of its phylogenetic relations.

Plant community-level studies on seed dormancy traits are important to understand and determine the significance of seed dormancy in different ecosystems. Hence, we studied seed dormancy and other related seed biological traits of 42 selected tropical lowland rainforest tree species from Sri Lanka, aiming to address seed dormancy class(es) for a biodiverse tropical lowland wet zone forest community and the relationship between dormancy classes, forest strata and seed dispersal mechanisms. Seed germination, imbibition, embryo length:seed length ratio, embryo morphology and the effect of gibberrelic acid on seed germination were determined. Sixty-two percent of the species with T50 < 30 days were identified as having fast-germinating seeds and the remaining 38% with T50 > 30 possessed slow-germinating seeds. Seeds of 33 species had fully developed embryos, while nine species had underdeveloped embryos; three had morphological dormancy (MD) and six morphophysiological dormancy (MPD). Treatment with gibberellic acid revealed physiological dormancy (PD) in seeds of six species, and the response to manual scarification confirmed physical dormancy (PY) in seeds of Pericopsis moonina. The majority of tropical lowland rainforests had non-dormant (ND) species (62%), and 14.3, 14.3, 7 and 2.3% of the species had MPD, PD, MD, and PY, respectively. Non-dormancy decreased for taxa from the upper strata to the lower strata of the forest. ND seeds were dispersed during the rainy season. Thus, non-dormancy seems to be the most dominant germination behaviour among the tree species in the lowland rainforest of Sri Lanka with the class of dormancy related to forest strata and dispersal time.

Nitrogen (N) fertilization affects wheat yield and grain protein concentration; however, its mismanagement can increase plant lodging. While the use of plant growth regulators such as trinexapac-ethyl (TE) can mitigate plant lodging, their effects on seed physiological quality are not well known. The aim of this study was to evaluate the effects of N fertilization and TE on wheat yield, lodging and seed quality of spring wheat varieties. It was carried out in the 2018 growing season in the environments of Londrina and Ponta Grossa, Brazil. A randomized complete block design was used with a 2 × 3 × 3 factorial arrangement to evaluate two wheat genotypes (WT 15008 and WT 15025), three top-dressing N rates (0, 40 and 120 kg ha−1), and three TE rates (0, 50 and 100 g ha−1). Agronomic characteristics related to wheat productivity (hectolitre weight, thousand-grain weight, density of fertile spikes, plant height, lodging and grain yield) and seed physiological quality (seed germination and vigour; length and dry matter of normal seedlings) were evaluated. Increasing N rates up to 120 kg ha−1 increased plant lodging up to 26.4 percentage points for WT 15025 in Londrina. TE impaired some traits of seed physiological quality. Spraying 100 g ha−1 TE on the plants reduced seedling length by 9.4% in the seeds of WT 15008 harvested in Ponta Grossa compared to the TE control (0 g ha−1). The dry matter of the seedlings from the seeds harvested in Londrina declined by 7.2% due to the application of 100 g ha−1 TE, compared to the control. However, a lower rate of TE (50 g ha−1) might be enough to minimize plant lodging without impairing the physiological quality of the seeds, depending on the rate of N fertilization. This study is the first step in providing empirical evidence for the detrimental effects of TE in combination with N on wheat seed quality, suggesting that seed producers should exercise caution in managing TE and N fertilization.

Wild oat is a herbicide resistance-prone global weed species that causes significant economic losses in dryland and horticultural agriculture. As a result, there has been a significant research effort to control this species. A major impediment to this research is the seed coat-mediated dormancy of wild oat, which requires a labor-intensive incision or puncturing of the seed coat to initiate seed germination. This study defines the most efficient settings of a mechanical thresher to overcome wild oat seed dormancy and then validates these settings using multiple populations collected from the Western Australian grain belt. We also compare the effects of rapid mechanical scarification and known germination stimulus tactics such as scarification with sulfuric acid (H2SO4), partial endosperm removal, sandpaper scarification of the seed coat, and immersion in sodium nitroprusside (NO donor SNP) solution on wild oat seedling growth rate. Threshing treatment of 1,500 rpm for 5 s provides equivalent germination compared with manually puncturing individual wild oat seeds, with no difference in seedling relative growth rate. The mechanical scarification of seeds using the thresher resulted in greater germination (66%) than H2SO4 scarification (0%), partial endosperm removal (10%), sandpaper seed coat scarification (25%), and exposure to NO donor SNP (34%). This study demonstrates that the physical dormancy of wild oat can be rapidly overcome using a commercially available mechanical thresher.
