For decades, it has remained unclear how the Asian swim bladder nematode Anguillicola crassus was able to supplant the previously stable population of its relative from New Zealand Anguillicola novaezelandiae in the Lake Bracciano, Italy. Previously, researchers have hypothesized that A. crassus possesses an ecological advantage due to a more efficient life cycle in combination with a pattern of unidirectional hybridization between A. novaezelandiae females and A. crassus males. The present study focuses on the viability of hybrid offspring and their allelic pattern, particularly in developed adult stages of the hybrid F1 generation. While the percentages of hybrid individuals from A. novaezelandiae mothers and A. crassus fathers increased from egg to adult stages, it was more distinct in egg stages of A. crassus females and A. novaezelandiae males, but did not occur in adult F1 individuals at all. Therefore, we corroborate the hypothesis of unidirectional hybridization by differentiating between egg and adult stages, and suggest this as another explanatory factor for the extinction of A. novaezelandiae in Lake Bracciano in Italy and the predominance of A. crassus.

In teleosts, vitellogenin (Vtg) is a phospholipoglycoprotein synthesized by the liver, released into the blood circulation and incorporated into the oocytes via endocytosis mediated by the Vtg receptor (VTGR) to form the yolk granules. The VTGR is crucial for oocyte growth in egg-laying animals but is also present in non-oviparous vertebrates, such as human. The VTGR belongs to the low-density lipoprotein receptor superfamily (LDLR) and is also named very-low-density lipoprotein receptor (VLDLR). In this study, we identified and phylogenetically positioned the VTGR of a basal teleost, the European eel, Anguilla anguilla. We developed quantitative real-time PCR (qRT-PCR) and investigated the tissue distribution of vtgr transcripts. We compared by qRT-PCR the ovarian expression levels of vtgr in juvenile yellow eels and pre-pubertal silver eels. We also analyzed the regulation of ovarian vtgr expression throughout vitellogenesis in experimentally matured eels. The Vtg plasma level was measured by homologous ELISA experimental maturation. Our in silico search and phylogenetical analysis revealed a single vtgr in the European eel, orthologous to other vertebrate vtgr. The qRT-PCR studies revealed that vtgr is mainly expressed in the ovary and also detected in various other tissues such as brain, pituitary, gill, fat, heart, and testis, suggesting some extra-ovarian functions of VTGR. We showed that vtgr is expressed in ovaries of juvenile yellow eels with no higher expression in pre-pubertal silver eels nor in experimentally matured eels. This suggests that vtgr transcription already occurs during early pre-vitellogenesis of immature eels and is not further activated in vitellogenic oocytes. European eel Vtg plasma level increased throughout experimental maturation in agreement with previous studies. Taken together, these results suggest that vtgr transcript levels may not be a limiting step for the uptake of Vtg by the oocyte in the European eel.

Anguillicola crassus is the most invasive species of its genus and it is a successful colonizer of different eel species worldwide. It is so far the only species of the genus Anguillicola whose life cycle has been studied completely. To analyse whether differences in life cycle may explain differences in invasiveness, we infected European eels with Anguillicola novaezelandiae under laboratory conditions. Anguillicola novaezelandiae shows a synchronized development in the European eel. Eggs with second-stage larvae appeared 120 days after infection. No density-dependent effect in parasite development could be found for A. novaezelandiae. The present study shows that the life cycle of A. novaezelandiae differs on final host level compared with A. crassus in ways which result in a less successful invasion of new host species.

The effect of Anguillicola crassus (Nematoda) on gas composition in the swimbladder of eels (Anguilla anguilla) was studied using mass spectrometry in feral eels from two German rivers and in experimentally infected eels. In both naturally and experimentally infected eels significant correlations were observed between the proportion of oxygen in the swimbladder and level of infection with Anguillicola crassus. In swimbladders of naturally infected feral eels the contribution of oxygen to swimbladder gas was reduced by 36–62·9% and in experimentally infected eels it was reduced by 11·4–57% compared to uninfected controls. The proportion of CO2 appeared to be lower in infected swimbladders compared to uninfected ones. However, this change was not significant. The findings are discussed in relation to an altered structure of the swimbladder wall due to the parasite.

The European eel (Anguilla anguilla) is drastically declining in all its distribution area and listed in the red list of the IUCN (International Union for Conservation of Nature). A rebuilding plan was adopted at European level in 2007, Regulation R(CE) 100/2007, to restore eel abundance to the level observed during the seventies. Its implementation started on the 1st of January 2009. This species is heavily threatened by numerous activities including fishing, and its management can only be effective through a systemic approach minimizing the whole range of human-induced impacts on the resource and its habitat. In the framework of the European interregional programme INTERREG IIIB-Atlantic Area– the INDICANG project aimed at elaborating abundance indicators of the European eel in the central part of its distribution area (http://www.ifremer/fr/indicang/). A methodological guide was elaborated by this project to define the indicators needed for this resource assessment. In this framework, Ifremer (Institut Français pour l'Exploitation de la Mer) and the UPPA University (Université de Pau et des Pays de l'Adour) have developed a method to estimate the daily and seasonal biomass of glass eels with the view to evaluate the fishery's impact on the estuarine eel recruitment to the Adour catchment. The estimation method uses observations from nightly scientific surveys to estimate glass eels' densities in the water column during various flood tides characterized by different hydrodynamic conditions. Information on these conditions allows the estimation of the glass eels biomass migrating during the night. From these estimates and reported catches made by the fishery during the same night, the exploitation rate applied by the fishery on the flow of glass eels progressing upstream during night flood tides is estimated. The relationship between the exploitation rate, fishery catches and hydrodynamic conditions allowed the estimation of the exploitation rate and nocturnal biomass fluctuations during the fishing season, from November 1st to March 31st of the following year. Finally, from the chronological series of biomass migrating at night, the total biomass migrating every day and the total recruitment into the estuary, during the main glass eel migration period, are estimated. Estimations made between 1998 and 2005 indicate that the overall rate of exploitation of the marine and continental fisheries, on average, is of 15.7%, ranging between 8 and 25% according to fishing seasons.

The studies carried out on the Adour estuary on the migration of glass eels lead to the characterisation of their swimming behavior. Individuals migrate passively with the flood tide current behind the dynamic tide front, and their movements into the water column are linked to the surrounding luminosity and water turbidity. A numerical model was built from observations gathered on glass eel densities during scientific surveys. It uses the outputs of a hydrodynamic model accounting for the variations in the river flow and tide coefficient. It allows the simulation of the displacement of a particle in the longitudinal axis of the estuary. The variation of the vertical movements through the water column takes into account the cloud cover, the moon phases, the alternation of days and nights and the water turbidity. The results allow displaying the migration speed of groups of glass eels entering the Adour estuary. These simulations are validated by the comparison with in situ observations and the outputs of the behavioral model make possible, on the one hand, to simulate the migration speed of glass eels according to hydrological data and, on the other hand, to define the environmental conditions that stop or slow down their displacement into the estuary.

Chub and eels were experimentally infected via intermediate hosts harbouring cystacanths, with Pomphorhynchus laevis alone, or Acanthocephalus anguillae alone, or simultaneously with mixtures of both species in varying proportions, and sampled at 7, 56 or 112 days post-infection. Examination of chub revealed that both species showed low establishment and growth rates, differing markedly from British field data, where chub is apparently one of the most important hosts, and preventing further meaningful experiments. Both species showed higher establishment rates in eels, but A. anguillae both grew faster and matured to a greater extent than P. laevis, reflecting field data. No evidence of any interactions, competitive or otherwise, was found in the mixed infections in eels at low or high infection levels. The results were interpretated with regard to British field data and current theories of the role of interspecific competition in structuring fish helminth communities.

The parasitic nematode Anguillicola crassus was recently introduced into populations of the European eel, Anguilla anguilla. We investigated, under experimental conditions, the regulation of A. crassus infrapopulations. We tested the effects of (1) the resource-limited habitat of the parasite and (2) the coexistence of several developmental stages in its niche (the swim-bladder) on the composition of the infrapopulations. The results revealed that the respective effects of these factors differed substantially during the course of the infection. Third-stage larvae (L3s) establishment would not be constrained by the size of the swim-bladder. Their moult to fourth-stage larvae (L4s) would be accelerated as the number of L3s increased. The moulting time of L4s to adults would be reduced by males and would be constrained by the size of the swim-bladder. However, the moult of L4s to adults and their further development would be synchronized with those of the opposite sex. At the time of mating, the number of males and the body weight of adults would depend on the size of the swim-bladder. Soon after the laying of eggs, the developmental constraint on the late L3s would decrease. When adults die, constraints would cease and late larval stages would moult to become adults.

The original host of the swimbladder nematode Anguillicola crassus, the Japanese eel (Anguilla japonica) and the recently colonized European eel (Anguilla anguilla) were immunized with 40 irradiated (500 Gy) 3rd-stage larvae (L3) of this parasite and challenged with an infection of 40 normal L3. The immunization induced a significant reduction of the number of adult worms developing from the challenge infection in A. japonica, but not in A. anguilla. The induced resistance (calculated using the relation of the number of adult worms in immunized eels and in non-immunized control eels) in A. japonica was 87·3%±30·4%. Following a single infection, the percentage of adult worms found in A. japonica was lower as compared to A. anguilla, and the few adult worms were much smaller, revealing a lower susceptibility of A. japonica to A. crassus in comparison to A. anguilla. Both eel species developed an antibody response against A. crassus, but the level of antibody responses was not positively correlated with the protection against infection, suggesting that the antibody response is not a key element in resistance of eels against A. crassus. This study suggests that the original host of A. crassus is able to mount efficient protective immune responses against its parasite, whereas the newly acquired host seems to lack this ability.

To investigate whether the stress response of European eels infected with Anguillicola crassus is influenced by environmental pollutants, experimentally infected eels were exposed to Cd and/or to 3,3′, 4,4′, 5-pentachlorobiphenyl (PCB 126). Serum cortisol and glucose concentrations of these eels were monitored over a period of 103 days and were compared with data from infected, unexposed eels as well as with data from uninfected eels. Additionally, the levels of cortisol were correlated with concentrations of Anguillicola-specific antibodies. All eels showed an initial increase of the cortisol levels until day 63. This general elevation of plasma cortisol is most likely due to handling stress, as all eels were repeatedly netted and afterwards inoculated with a feeding tube. At the end of the exposure period eels which were infected and those which were infected and simultaneously exposed to Cd and PCB showed significantly higher levels than the controls. The general course of serum glucose levels in eels resembled that of cortisol. Accordingly, Spearman correlation analysis revealed that an increase in serum cortisol concentrations is correlated with rising levels of glucose. With respect to immune-endocrine interactions a significant negative correlation between cortisol and anti-A. crassus antibodies was found. Our data show that A. crassus is the most potent stressor for European eels among the treatments tested within this study. This is important in terms of ecotoxicological studies as the main effects are caused by parasites rather than chemicals. Accordingly, effects of parasites on the physiological homeostasis of organisms must be considered in ecotoxicology. From the parasitological point of view our results suggest that probably as part of an unbalanced host-parasite interaction A. crassus evokes a strong cortisol response in A. anguilla, thereby suppressing the immune response which in turn enables the parasite to establish. The parasite-induced stress response in the newly adopted European eel might be one of the factors which contributes to the extremely effective colonizing strategy of A. crassus.

After their oceanic migration, juvenile European eels Anguilla anguilla enter estuaries as glass eels, develop into pigmented elvers and migrate into fresh water. Fisheries often transfer such eels abruptly between salinities, principally glass eels and elvers from estuarine to fresh water. It is usually assumed that survival rates are high, but this required systematic investigation. Survival was found to be 100% over 21 days of glass eels and semi-pigmented elvers transferred abruptly from estuary conditions into fresh water, 50% sea water and full sea water. Fully pigmented elvers, however, showed significantly reduced survival when transferred into sea water. Salinity preference experiments with juvenile eels have historically been inconclusive. Here, in a choice chamber design, a clear developmental shift in salinity preference was found, with glass eels preferring 100% sea water, semi-pigmented elvers showing no clear preference and fully pigmented elvers preferring fresh water. We conclude that eel fisheries enhancement by abrupt transfer of juveniles among salinities is largely vindicated. In addition, developmental shifts in salinity preference have been clarified and this aids in the interpretation of eel migration patterns.

Data from a long-term study of the intestinal helminth parasite community of eels, Anguilla anguilla, stocked into the shallow eutrophic Neusiedler See, Austria, were collected over an 8 year period (1994–2001). In total, 720 eels from 2 sampling sites were examined. The parasite community showed characteristics similar to those in the natural eel populations in rivers of the UK and mainland Europe: it was species poor, with only 5 species (Acanthocephalus lucii, Acanthocephalus anguillae, Raphidascaris acus, Proteocephalus macrocephalus, Bothriocephalus claviceps) comprising the component community and a maximum infracommunity richness of 4 species. Over the period, the intestinal parasite community of the sampling site in Illmitz, which was originally dominated by A. lucii, changed. As levels of A. anguillae increased to a point at which it dominated the community, diversity increased whilst dominance of a single species decreased. By contrast the community in the southern sampling site remained rather constant with a continuous high infection level of A. anguillae and low abundance of A. lucii. Both acanthocephalan species exhibited higher infection levels in larger eels and in different seasons of the year and the infection parameters were significantly different between the years of study. The significant differences in the infection levels of the 2 acanthocephalan species at the 2 sampling sites were surprising as both acanthocephalan species use the same intermediate host, Asellus aquaticus, and the sampling sites were in close proximity and were similar in terms of water quality, host size and invertebrate abundance. Differences in the fish communities of the 2 sampling sites and eel movements rather than interspecific competition are discussed as possible explanations for the differences in the parasite communities of the 2 sampling sites.

The swim-bladder nematode Anguillicola crassus originates from the Far East where it is a parasite of the Japanese eel (Anguilla japonica). After A. crassus was introduced to Europe, it became a predominant parasite of the European eel (Anguilla anguilla). A study performed with experimentally infected eels (98 days, 23 °C) revealed significant differences in the susceptibility of the two eel species to this parasite. The recovery rate of 30 administered infective A. crassus larvae (L3) from A. japonica was less than half of that from A. anguilla (33·2% and 13·8%, respectively). Almost 60% of the worms recovered from A. japonica were found as dead, encapsulated and necrotic larvae in the swimbladder wall. In contrast, no dead larvae were found in A. anguilla. Additionally, the development of the worms was shown to be significantly slower in A. japonica compared with A. anguilla. The lower survival rate of the worms, together with their slower development, resulted in a significantly lower adult worm burden (11 and 428 mg wet weight, respectively) and in a decreased reproductive success in A. japonica compared with A. anguilla. These results demonstrate that the original host, A. japonica, possesses more effective defence mechanisms against A. crassus than does the non-adapted host, A. anguilla.

A nested (non-random) species composition was observed among the intestinal macroparasite communities of European eels, Anguilla anguilla. Nestedness was observed in 8 out of 10 component community samples from the rivers Thames and Test between April 2000 and October 2001. Parasite component communities consisted of mainly generalist and specialist, but also accidental species, and included acanthocephalans, cestodes, nematodes and digeneans. Nestedness was observed when component population size differed markedly between all or most parasite species, but not when the majority of species present was similarly abundant. Nestedness could not be explained in terms of host weight, log normal distribution of parasite species, or mean intensity of infection. It is proposed that nestedness occurred as a result of a sufficiently graded abundance between parasite species, which was established early in the year by colonization processes.

The individual and combined effects of cadmium (Cd) and 3,3′,4,4′,5-pentachlorobiphenyl (PCB 126) on the antibody response of fish against metazoan parasites were tested. Eels experimentally infected with the swim bladder nematode Anguillicola crassus were exposed to sublethal concentrations of Cd and PCB 126. Cd was added to the water resulting in an effective concentration of 21·7±12·8 μg/l (mean±S.D.). PCB 126 was applied orally at a dose of approximately 100 ng PCB 126 per g body weight. At the end of the experiment, 76 days post-infection (p.i.), eels were found to be infected with 2–3 worms. Immunoblot analyses revealed that the body wall of adult worms was the most suitable crude antigen, and was subsequently used for an ELISA to evaluate the immune response of A. anguilla under various conditions. A significant increase of Anguillicola-specific antibodies in the peripheral blood was first detected 61 days p.i., indicating that it was not the invasive larvae but the adult worms which elicit the antibody response. The presence of Cd in the concentrations applied did not appear to modulate the production of antibodies. In contrast, the exposure to PCB 126 resulted in a complete suppression of the antibody response. A similar effect was also found for the combined exposure of the infected eels to Cd and PCB 126. A suppressed immune response, as demonstrated here, may be the reason why hosts exposed to environmental pollution became often much more easily infected than unexposed conspecifics.

The intestinal macroparasite communities of freshwater eels (Anguilla anguilla) captured in the south of England from Windsor (River Thames) during August 2001, and Leckford (River Test) during late June/July 2000, are reported for the first time. Parasite component communities were among the most species rich and diverse recorded from European eels. A total of 13 intestinal macroparasite species were encountered during the study, 8 from each eel host population with 3 being common to both. Acanthocephalans, nematodes and cestodes were recovered from each host population. Eels from Windsor additionally harboured Nicolla gallica (Digenea), which was also the most prevalent and abundant macroparasite species in these hosts. Each component community followed a log normal rank abundance distribution and demonstrated reduced species dominance and increased species equitability compared with previous studies. As such, the study component communities were suitable for testing the hypothesis of low infracommunity diversity in European eel hosts. Specifically, this hypothesis predicts that the intestinal macroparasite infracommunities of European eels are species-poor, displaying low density and diversity with high dominance, irrespective of component community diversity, and that this may be more pronounced in UK host populations. This hypothesis was not upheld; study findings demonstrate that higher infracommunity diversity in eel hosts is possible, and suggest that infracommunity diversity in individual eel hosts may be a simple, stochastic reflection of component community diversity.

To understand the ecology of the exotic silver European eel Anguilla anguilla introduced into Japanese waters, the migratory behavior of 106 specimens captured in the coastal waters of Japan between April 1997 and March 2002 was analyzed. Their migratory behavior was apparently correlated with various environmental factors, particularly photoperiod, water temperature, lunar phase, and passage of atmospheric depressions, and was similar to the behavior of the species in European waters. These findings suggest that transplanted European eels retain their ability to respond to environmental cues for seaward migration in similar temperate habitats. The timing of the migration of silver European eels coincided with that of the native Japanese eels A. japonica, suggesting that the silver European eel was synchronized physiologically with the native eel by the same environmental factors.

We have developed an index based on macroscopic criteria to easily assess the health state of the eel swimbladder as a result of infection by the nematode Anguillicola crassus. In the sampling area (brackish lagoons of the French Mediterranean coast), 92% of the host sample (1251 eels covering all size classes) showed pathological signs of infection. The general trend was for increasing damage as eel size increased, thus suggesting accumulation of pathological effects. We also revealed a non-linear relationship between the swimbladder index and the abundance of living worms. In particular, we showed that severely damaged swimbladders harboured very few living nematodes. We argue that the swimbladder degenerative index more closely reflects the parasite pressure than does classic parasite count. We found seasonal variation in the swimbladder index, with maximum damage occurring in July and thereafter a trend for healthy individuals. We discuss the possibility that the seasonal decrease in the swimbladder index could reflect the death of the more severely affected individuals during the warmest months.

European eels (Anguilla anguilla) from 2 sampling sites on the Rhine river (near Karlsruhe and near Worms) were investigated with respect to their parasite communities. Nine different metazoan species were found to live in and on the eels. The highest number of species was recorded from the intestine, which contained up to 6 different helminths. Among these, acanthocephalans were the most prevalent worms with the eel-specific parasite Paratenuisentis ambiguus as the dominant species of the intestinal component communities at both sites. Comparing the intestinal parasites from eels caught near Karlsruhe with those from Worms, the acanthocephalans showed a significantly lower abundance at Worms. A significantly lower mean number of intestinal helminth species as well as a significantly lower Brillouin's Index was found at Worms compared with Karlsruhe. This difference could be related to the abundance of the respective intermediate crustacean hosts. At the sampling site Worms the amphipod Corophium curvispinum was the dominant crustacean. Additionally, only the isopod Jaera istri and the amphipod Dikerogammarus villosus were found. All these crustacean species have only recently colonized the Rhine river system via the Main-Danube canal, built in the early 1990s. They are not known to act as intermediate hosts for any of the acanthocephalans found in the eels. The site near Karlsruhe exhibited a higher crustacean diversity, including Asellus aquaticus and different species of the genus Gammarus, which are all known intermediate hosts for the acanthocephalans found. Therefore, changes of eel parasite diversity can be correlated with the appearance of invading crustacean species (neozoans).

The purpose of this study was to determine under laboratory conditions over a period of 311 days if infection with the nematode Anguillicola crassus induces stress in European eels (Anguilla anguilla), and stimulates the endocrine stress axis as measured by serum cortisol levels. Eels were experimentally infected with 3rd-stage larvae (L3) in different doses to simulate natural conditions with varying infection pressures. Blood samples were drawn from the caudal vein every 2 weeks and serum cortisol concentrations were determined by radioimmunoassay (RIA). The results showed that the application of L3 resulted in a significant increase in the cortisol levels. The period of time at which elevated cortisol values were observed was consistent with the time of larval development and the appearance of adult A. crassus. Thus, there is a stress response to the larval and young adult stages, but no chronic response to older adults. Therefore, it is likely that infection of eels under natural conditions especially with a high number of larvae may be a considerable stressor, in combination with different environmental factors like water temperature, pH, oxygen concentration, pollution and inter-individual relationships.
