Glasshouse experiments were conducted to evaluate the effects of preemergence applications of metribuzin, pendimethalin, alachlor, and imazaquin on the competitive relationship between soybean and sicklepod, tall morningglory, or common cocklebur. An equal-ratio series design was employed where the proportion of species in a mixture remained constant and total plant density was varied. The ratio of soybean:sicklepod fresh weights was altered by alachlor and imazaquin while the ratio of soybean:tall morningglory fresh weights was altered only by imazaquin. The ratio of soybean:common cocklebur fresh weights was altered by metribuzin and imazaquin. Increasing herbicide rates resulted in higher soybean:weed fresh weight ratios and higher herbicide response coefficients.

Field experiments were conducted in 1986 and 1987 to evaluate the effect of herbicide incorporation to various depths on common cocklebur control with imazaquin, a tank mix of imazaquin and metribuzin, and a preformulated mixture of chlorimuron and metribuzin in soybeans. Herbicide rates included were two-thirds and the full registered rate. All treatments, except chlorimuron plus metribuzin (50 plus 320 g ai/ha, respectively), controlled more common cocklebur when incorporated to depths of 2.5 to 7.6 cm than when left unincorporated. Also, imazaquin at 90 g ai/ha and the tank mix of imazaquin plus metribuzin at 90 plus 280 g ai/ha, when incorporated, controlled more common cocklebur than higher rates of the same combination when not incorporated. Common cocklebur control was similar with 2.5-, 5.0-, and 7.6-cm soil incorporation depths.

Smutgrass is an invasive, well-rooted perennial that has long been recognized as an aggressive weed throughout Florida and in the subtropical regions of the United States. Small smutgrass and giant smutgrass are the two predominant smutgrass varieties found in Florida. The native soil pH of Florida flatwoods is 4.5 to 5.0 which is lower than the level of soil pH recommended for optimum bahiagrass growth. Therefore, replacement series experiments were conducted in a greenhouse in 2010 and 2011 to compare the competitive ability of bahiagrass with each of the two varieties of smutgrass at three levels of soil pH (4.5, 5.5, and 6.5), two densities; 4 (low) and 8 (high) plants pot−1, and at five planting ratios of 100 : 0, 75 : 25, 50 : 50, 25 : 75, and 0 : 100. Relative competitive ability and aggressivity of giant smutgrass was greater than bahiagrass across all pH levels and densities, whereas relative competitive ability and aggressivity of bahiagrass was greater than small smutgrass in all pH levels and densities, except at pH 6.5. At pH 5.5, biomass accumulation of giant smutgrass was at least 73% higher than bahiagrass, whereas small smutgrass biomass was at least 46% lower than bahiagrass at equal planting ratios of both low and high densities. Differential responses were observed on bahiagrass competitive ability with small and giant smutgrass. Amending soil pH is not a likely option to increase the growth and competitive ability of bahiagrass over giant smutgrass. However, for small smutgrass, it is likely to increase the aggressivity of bahiagrass in bahiagrass–small smutgrass mixture, unless the soil pH is raised above 5.5.

The implementation of a successful glyphosate resistance management strategy requires a simple and cost-effective method for detecting resistance in key weeds. To date, however, glyphosate resistance is still routinely confirmed via laborious and time consuming whole-plant pot assays using seeds collected at the end of the growing season. Here, we describe a simple, early-season bioassay for detecting evolved glyphosate resistance in grass and broadleaf weeds. It involves transplanting suspected glyphosate resistant seedlings alongside known sensitive and resistant standards into agar containing informative rates of herbicide and recording percentage survival 14 d after plating. The method was validated using sensitive and resistant populations of Lolium, Eleusine, Conyza, and Amaranthus species encompassing the main glyphosate resistance mechanisms, namely, impaired translocation, EPSPS gene duplication, and mutations. The whole plant pot and agar-based seedling tests generated comparable resistance indices in dose-response assays and percentage survival at discriminating glyphosate rates. The method was applied successfully to detect resistance in a rigid ryegrass population collected from a French vineyard well before glyphosate was applied in the field for the current season. Additionally, the test was shown to be highly transferable to several other grass and broadleaf weeds that have evolved resistance to glyphosate. One major attribute of the method is that it is capable of detecting resistance regardless of the mechanism involved. In addition to being very simple, quick and, cost-effective, it allows determination of glyphosate resistance in weeds prior to field application. It thus offers the opportunity for an informed choice of herbicides for effective weed control.

Keng stiffgrass is a serious farmland grass weed distributed globally in winter wheat fields and rice–wheat double-cropping areas. The intensive use of acetyl-CoA carboxylase (ACCase)-inhibiting herbicides has led to the evolution of resistance in a growing number of grass weeds. In this study, whole-plant pot bioassay experiments were conducted to establish that a Keng stiffgrass population from eastern China, JYJD-2, has evolved high-level resistance to fenoxaprop-P-ethyl and moderate resistance to quizalofop-P-ethyl and pinoxaden. Using the derived cleaved amplified polymorphic sequence method, a tryptophan-to-cysteine mutation at codon position 1999 (W1999C) was detected in the ACCase gene of the resistant population JYJD-2. Of the 100 JYJD-2 plants tested, we found 47 heterozygous resistant and 53 homozygous sensitive individuals. In vitro ACCase assays revealed that the IC50 value of the ACCase activity of the resistant population JYJD-2 was 6.48-fold higher than that of the susceptible population JYJD-1. To the best of our knowledge, this is the first report of the occurrence of W1999C mutation in the ACCase gene of fenoxaprop-P-ethyl–resistant Keng stiffgrass. This study confirmed the resistance of Keng stiffgrass to the ACCase inhibitor fenoxaprop-P-ethyl, cross-resistance to other ACCase inhibitors, and the resistance being conferred by specific ACCase point mutations at amino acid position 1999.

Waterhemp [Amaranthus tuberculatus (Moq.) J. D. Sauer] is a troublesome weed occurring in cropping systems throughout the U.S. Midwest with an ability to rapidly evolve herbicide resistance that could be associated with competitive disadvantages. Little research has investigated the competitiveness of different A. tuberculatus populations under similar environmental conditions. The objectives of this study were to evaluate: (1) the interspecific competitiveness of three herbicide-resistant A. tuberculatus populations (2,4-D and atrazine resistant [2A-R], glyphosate and protoporphyrinogen oxidase [PPO]-inhibitor resistant [GP-R], and 2,4-D, atrazine, glyphosate, and PPO-inhibitor susceptible [2AGP-S]) with soybean [Glycine max (L.) Merr.]; and (2) the density-dependent response of each A. tuberculatus population within a constant soybean population in a greenhouse environment. Amaranthus tuberculatus competitiveness with soybean was evaluated across five target weed densities of 0, 2, 4, 8, and 16 plants pot−1 (equivalent to 0, 20, 40, 80, and 160 plants m−2) with 3 soybean plants pot−1 (equivalent to 300,000 plants ha−1). At the R1 soybean harvest time, no difference in soybean biomass was observed across A. tuberculatus populations. At A. tuberculatus densities <8 plants pot−1, the 2AGP-S population had the greatest biomass and stem diameter per plant. At the R7 harvest time, the 2AGP-S population caused the greatest loss in soybean biomass and number of pods compared with the other populations at densities of <16 plants pot−1. The 2AGP-S population had greater early-season biomass accumulation and stem diameter compared with the other A. tuberculatus populations, which resulted in greater late-season reduction in soybean biomass and number of pods. This research indicates there may be evidence of interspecific competitive fitness cost associated with the evolution of 2,4-D, atrazine, glyphosate, and PPO-inhibitor resistance in A. tuberculatus. Focus should be placed on effectively using cultural weed management practices to enhance crop competitiveness, especially early in the season, to increase suppression of herbicide-resistant A. tuberculatus.

With rapid progress being made in deciphering plant genomic sequences, determining the function of these genes is one of the main challenges that plant molecular biologists face today. Herbicidal inhibitors have been very useful for understanding gene function in at least two examples, represented by herbicidal inhibitors of hydroxyphenylpyruvate dioxygenase (HPPD) and deoxyxylulosephosphate reductoisomerase (DXR). In the first, an albino mutant of Arabidopsis isolated during the study of carotenoid biosynthesis was found to have an intact carotenoid biosynthetic pathway. A number of “bleaching herbicides” in development at about the same time (e.g., sulcotrione) produced similar symptoms by strongly inhibiting HPPD, a key enzyme in plastoquinone biosynthesis. Examination of the Arabidopsis mutant revealed that the HPPD gene had been inactivated in the albino plants. Inhibition of the HPPD pathway also led to reduced levels of tocopherol (vitamin E), an end product of the pathway. Further studies and manipulation of the pathway produced plants with significantly higher levels of vitamin E. This result is a clear demonstration of how an herbicidal inhibitor was able to lead to the identification of a gene that was responsible for a particular phenotype. As a second example, identification of fosmidomycin as a specific inhibitor of DXR in the recently elucidated nonmevalonate pathway of isopentenyl pyrophosphate (IPP) biosynthesis was instrumental in furthering the understanding of an important route to synthesis of many important terpenoid products.

Seedling emergence traits of susceptible (S) and resistant (R) blackgrass subpopulations isolated from a single non–target-site resistant (NTSR) population were studied in controlled conditions. The seedling emergence of the R subpopulation was lower and slower than that of the S subpopulation, especially at low temperature and deep burial. The burial depth inhibiting final emergence by 50% for the R subpopulation was significantly lower than that of the S subpopulation at low temperature. The present study revealed that under suboptimal conditions the NTSR loci conferring herbicide resistance were correlated with a fitness cost in relation to seedling emergence traits. The results suggest that deep soil cultivation and delayed sowing of autumn-sown crops can hamper germination of the R more than of the S subpopulation and thus potentially reduce the prevalence of the R subpopulation in the blackgrass population.

Annual sedge (Cyperus compressus L.) populations with resistance to halosulfuron were identified in turfgrass at two new locations in Georgia. Research was conducted to evaluate (1) resistance levels to two acetolactate synthase (ALS) inhibitors, (2) ALS enzyme susceptibility, (3) genetic differences associated with resistance, and (4) differential levels of ALS gene expression in these biotypes. In dose–response experiments, the biotypes were >160 times resistant to halosulfuron but only 12 times more resistant to imazaquin compared with a susceptible biotype. In vitro enzyme assays indicated that resistant (R) biotypes required 6.1-fold greater concentrations of imazaquin to reduce ALS activity 50% compared with the susceptible (S) biotype. Both R biotypes had a similar Pro-197-Ser amino acid substitution in the ALS gene that confers resistance to sulfonylureas. Compared with the S biotype, R biotypes had 4.4 times higher ALS gene expression than the S biotype. No differences in gene copy number were found between any biotypes for the ALS gene. Overall, ALS-resistant C. compressus selected by halosulfuron use in turfgrass may be exhibiting partial susceptibility to imazaquin but complete resistance to sulfonylureas. Differential levels of susceptibility to ALS inhibitors for these biotypes are associated with the Pro-197-Ser substitution and enhanced expression of the ALS gene.

Herbicide-resistant Echinochloa species are among the most problematic weeds in agricultural crops globally. Recurring herbicide selection pressure in the absence of diverse management practices has resulted in greater than 20% of sampled Echinochloa populations from rice (Oryza sativa L.) fields demonstrating multiple resistance to herbicides in Arkansas, USA. We assessed the resistance profile and potential mechanisms of resistance in a multiple herbicide–resistant junglerice [Echinochloa colona (L.) Link] (ECO-R) population. Whole-plant and laboratory bioassays were conducted to identify the potential mechanisms of non–target site resistance in this population. ECO-R was highly resistant to propanil (>37,800 g ha−1) and quinclorac (>17,920 g ha−1) and had elevated tolerance to cyhalofop (R/S = 1.9) and glufosinate (R/S = 1.2) compared to the susceptible standard. The addition of glufosinate (590 g ha−1) to cyhalofop (314 g ha−1), propanil (4,500 g ha−1), or quinclorac (560 g ha−1) controlled ECO-R 100%. However, cyhalofop applied with propanil (48% control) or quinclorac (15% control) was antagonistic. The application of the known metabolic enzyme inhibitors malathion, carbaryl, and piperonyl butoxide increased control of ECO-R with propanil (>75%) but not with other herbicides. Neither absorption nor translocation of [14C]cyhalofop or propanil was different between ECO-R and ECO-S. [14C]Quinclorac absorption was also similar between ECO-R and ECO-S; however, translocation of quinclorac into tissues above the treated leaf of ECO-R was >20% higher than that in ECO-S. The abundance of metabolites was higher (∼10%) in the treated leaves of ECO-R than in ECO-S beginning 48 h after treatment. The activity of β-cyanoalanine synthase, which detoxifies hydrogen cyanide, was not different between ECO-R and ECO-S following quinclorac treatment. Resistance to propanil was due to herbicide detoxification by metabolic enzymes. Resistance to quinclorac was due to a detoxification mechanism yet to be understood. The reduction in sensitivity to cyhalofop and glufosinate might be a secondary effect of the mechanisms conferring high resistance to propanil and quinclorac.

Weeds have acquired evolutionary adaptations to the diverse crop and weed management strategies used in cropping systems. Therefore, changes in crop production practices such as conventional to organic systems, tillage-based to no-till systems, and diversity in crop rotations can result in differences in weed community composition that have management implications. A study was carried out to understand the weed community dynamics in a long-term alternative cropping systems study at Scott, SK, Canada. Long-term (18-yr) weed community composition data in wheat (Triticum aestivum L.) in ORG (organic), RED (reduced-input, no-till), and HIGH (high-input, conventional tillage) systems with three levels of crop rotation diversity, LOW (low diversity), DAG (diversified annual grains), and DAP (diversified annuals and perennials), were used to study the effect of different cropping systems and the effect of environment (random temporal effects) on residual weed community composition using the principal response curve (PRC) technique. The interaction between cropping systems and year-to-year random environmental changes was found to be the predominant factor causing fluctuations in weed community composition. Furthermore, the single most predominant factor influencing the weed composition was year-to-year random changes. Organic systems clearly differed from the two conventional systems in most years and had more diverse weed communities compared with the two conventional systems. The two conventional systems exhibited similar weed composition in most years. In this study, the use of the PRC method allowed capture of the real temporal dynamics reflected in the cropping systems by time interaction. This study further concludes that moving from a tillage-based, high-input conventional system to a no-till, reduced-input system did not cause significant changes in the weed community composition throughout the time period, but diversity in organic systems was high, probably due to increased occurrence of some difficult to control species.

A plastic envelope device is described which allows separate exposure of seeds, roots, and coleoptiles of emerging grass seedlings to soil-incorporated herbicides.

After 10 months, diphenamid residues were found throughout the 0–9 inch soil depth. In silt loam soils under climatic conditions prevailing in and around the bluegrass region of Kentucky, diphenamid residues in soils may be present at phytotoxic levels in the plow layer 10–11 months after application at rates required for weed control in tobacco.

C14-ring-labeled 2-chloro-4-diethylamino-6-isopropylamino-s-triazine (ipazine) was soil-applied to young cotton plants or placed in solution cultures. C14-labeled constituents accumulated in the lysigenous glands of roots, stems, and leaves. Retranslocation of C14-labeled components into new growth after removal of C14-ipazine from solution cultures did not occur. No evolution of of C14O2 by C14-ipazine-treated plants was detected, but evolution of C14O2 occurred from C14-ipazine-treated soil. Some unaltered C14-ipazine was present in leaves and stems sampled 3 months after treatment. No C14-ipazine could be detected in seed formed on treated plants. Free hydroxyipazine did not constitute a major degradation product. However, hydroxyipazine and other basic substances appeared to be released upon hydrolysis of a chloroform-soluble, non-distillable component that accumulated in roots and leaves. An ethanol-insoluble, non-dialyzable component also accumulated in leaves and roots.

Water lettuce (Pistia stratiotes L.) was susceptible to applications of 6,7-dihydrodipyrido(1,2-a:2’,1′-c)pyrazidiinium salt (diquat). The most satisfactory rates of application were 1.0 to 1.5 lb/A of diquat. The addition of a surfactant did not increase the effectiveness of diquat. Several other chemicals gave unsatisfactory control of water lettuce.

The effectiveness of catch or trap crops in ridding soil of witchweed was studied by growing the crops 1 through 5 years on heavily infested witchweed land.

Catch or trap crops were about equal in value for witchweed eradication. Soybeans, field peas, and sorghum were grown 3 years before acceptable corn yields were obtained. Two years of millet and 4 of corn were required to obtain acceptable corn yields.

Complete eradication of witchweed was not achieved by any crop during the 5-year period.

Nitrofen (2,4-dichlorophenyl p-nitrophenyl ether) injured meristematic tissues and reduced germination and growth of cabbage (Brassica oleracea var. capitata L.) in the dark. Cabbage was more susceptible to nitrofen when the plants were grown under a low water potential or when they were maintained in the dark after spraying. This toxicity was overcome by exogenous sucrose and light was not required for nitrofen activity. Nitrofen promoted membrane permeability of red beet (Beta vulgaris L.) root sections and this was enhanced by dimethylsulfoxide (DMSO). Sucrose or Carbowax 1500 prevented this increase in permeability. Nitrofen inhibited non-cyclic photophosphorylation and electron transport in isolated spinach (Spinacia oleracea L.) chloroplasts and increased oxygen uptake of cabbage leaf sections. These effects appeared dependent upon permeability. Nitrofen induced stomatal closure, decreased transpiration, and increased leaf temperature. The leaf sustains thermal injury under high temperatures or high light intensities.

Four postemergence herbicides were evaluated for control of horse purslane and barnyardgrass occurring alone and in combination in three separate field experiments at Haryana Agricultural University, Hisar (India). Haloxyfop, fluazifop, and sethoxydim controlled barnyardgrass. Fomesafen controlled horse purslane most effectively when applied at the two-branch stage of horse purslane in mung bean. Combinations of haloxyfop, fluazifop, or sethoxydim with fomesafen applied at the five-branch stage of horse purslane and two-leaf stage of barnyardgrass controlled both weeds. Mung bean treated with fomesafen at 250 g ae/ha plus haloxyfop at 190 or 250 g ae/ha, fluazifop at 750 or 1000 g ae/ha, or sethoxydim at 750 g or 1000 g ae/ha produced dry weights, pods/plant, and grain yield similar to weed-free mung bean.

The root growth rate index of velvetleaf (Abutilon theophrasti Medic.) was significantly greater than that of five other weed species. The root growth rate index of sorghum [Sorghum bicolor (L.) Moench ‘RS-626’] was found to be significantly higher than that of soybeans [Glycine max (L.) Merr. ‘Ford’]. Shoot growth rate indexes of three perennial broadleaf species studied were significantly lower than shoot growth rate indexes of six annual species. Root weights of perennial broadleaf species were not significantly different from root weights of velvetleaf and soybeans. Soybeans were found to have a significantly higher shoot to root ratio than seven weed species but not higher than sorghum. All other species had larger leaf weights and leaf areas than the three broadleaf perennials. While there were significant differences in leaf weights among six species, there were no significant differences in leaf area. Seed weight was not significantly correlated with emergence rate index and root weight but was correlated to all other factors studied. Emergence rate index was correlated with root growth rate index but not with shoot growth rate index.

Jointed goatgrass is most commonly described as a winter annual species. However, it has been observed to produce spikes in spring crops, apparently without being exposed to vernalizing conditions. A controlled environment study was conducted to determine the reproductive response of jointed goatgrass plants grown from seeds of fall- and spring-emerging parent plants to various vernalization durations. Winter wheat was included as a control. Winter wheat spikelet production was dependent on vernalization, and the number of spikes per plant was 10-fold greater if the plants were exposed to 4 C for 10 wk. In contrast, jointed goatgrass spike production without vernalization remained as high as 50% of that produced by plants exposed to 10 wk of vernalization conditions. Jointed goatgrass is thus not as dependent on vernalization for reproduction as the comparative winter wheat standard. Apparently, jointed goatgrass is more a facultative rather than an obligate winter annual. Rotating to a spring-seeded crop should not be expected to completely prevent jointed goatgrass seed production. Fields rotated to spring wheat to eliminate jointed goatgrass seed production should be monitored, and jointed goatgrass should be hand pulled or otherwise controlled to ensure zero seed production.
