Sarcoptes scabiei is an important parasitic mite in both man and animals. Little is known about the molecular interactions between this pathogen and its host. This is in part explained by the paucity of mite-derived material, including antigens. To extend the knowledge of the molecular repertoire in S. scabiei, we have performed a gene survey by an expressed sequence tag (EST) analysis. A total of 1020 ESTs were generated from an S. scabiei cDNA library. The average sequence read was 510 bp after editing and the overall sequencing success was 89%. Clustering of the sequences resulted in 76 clusters, comprising 36% of the ESTs. Sequence similarity searches showed that almost half of the S. scabiei ESTs could be assigned a putative identity. Many of these transcripts shared similarity with genes involved in basic metabolism and cellular organization. In the data set we also identified several proteases and other types of potential allergens implicated in various disease mechanisms. A relatively large fraction of the ESTs coded for different proteins carrying protease inhibitor-like domains. The clones with no similarity to previously identified genes constituted 11% of our transcripts. The EST data generated in this study will be a valuable resource in further studies of the biology of S. scabiei and in the identification of genes that can serve as potential targets in the control of the parasite.

Following the introduction of E. granulosus into Australia with domestic animals during European settlement, the parasite quickly became established in the E. granulosus-naive native animals of the continent. The distribution of E. granulosus in wildlife in Australia is restricted by rainfall, but nevertheless the parasite is currently widespread and highly prevalent in many areas including numerous national parks and privately owned farms. The human population of Africa is rapidly increasing resulting in ever more pressure on wild-life populations and habitat. National parks, reserves and conservation areas now provide important tracts of preserved habitat for maintaining populations of wildlife that are also important in the maintenance of E. granulosus. In some parts of Africa, hydatid-infected humans provide a source of E. granulosus infection to wildlife definitive hosts. In many areas felids may also act as important definitive hosts for E. granulosus with the parasite being maintained in a prey/predator relationship between lions and a range of intermediate hosts. Populations of E. granulosus-infected wild-life both in Australia and Africa act as important reservoirs in perpetuating the transmission of E. granulosus to both domestic animals and humans. In Australia, E. granulosus-infected wild-life is infiltrating urban areas and currently represents a potentially important new public health problem.

The present study was designed to assess the role of inulin and sugar beet fibres (SBF) on adult O. dentatum in growing pigs. Four experimental diets were formulated based on barley flour with added insoluble fibre from oat husk (Diet 1), a pure carbohydrate source inulin (Diet 2), soluble fibre from sugar beet fibre (SBF) with a high proportion of soluble fibre components (Diet 3) or inulin plus SBF (Diet 4). Thirty-two 10-week-old pigs were divided randomly into 4 groups each of 8 pigs. After 3 weeks adaptation on Diet 1 all pigs were infected with a single dose of 6000 L3O. dentatum. At week 7 p.i. one group was switched to Diet 2, another group to Diet 3 and another group to Diet 4. The remaining 8 pigs continued on Diet 1 until the end of the experiment and served as controls. At week 13, all pigs were necropsied and their worm burdens determined. The worm recoveries from the pigs on the inulin supplemented diet (Diet 2) were reduced by 97% compared to the controls (Diet 1). Further, the inulin-fed pigs exhibited markedly reduced faecal egg counts. The pigs on inulin plus SBF diet (Diet 4) and on SBF diet (Diet 3) had 86% and 70% adult worm reductions compared with the controls, respectively. The results from this study indicate that highly degradable and rapidly fermentable carbohydrates such as dietary inulin have a profound deworming effect on O. dentatum infection.

This study examined the route of infection by free-swimming larvae of 2 monocotylid monogeneans that inhabit the gills (Neoheterocotyle rhinobatidis) and the nasal tissue (Merizocotyle icopae) of the shovelnose ray, Rhinobatos typus, from Heron Island on the Great Barrier Reef, Australia. Larvae of N. rhinobatidis and M. icopae attached directly to the gills and the nasal tissue of the ray, respectively, and did not first settle on the skin. Initial development of the post-oncomiracidium of N. rhinobatidis was rapid and hamuli formed between 6 and 24 h p.i. at a mean temperature of 26 °C. However, growth then slowed markedly and was variable; only 2 fully mature individuals were found 20 days p.i. at a mean temperature of 24·5 °C. Development of M. icopae was slow and variable throughout; hamuli did not appear until 10 days p.i. and no mature individuals were obtained even 22 days p.i. at a mean temperature of 24·5 °C. No character could be found as an indicator of parasite age for N. rhinobatidis or M. icopae due to the high variability in development in both species.

The northern biotype of Echinococcus granulosus occurs throughout the holarctic zones of tundra and taiga, from eastern Fennoscandia to the Bering Strait in Eurasia and in North America from arctic Alaska approximately to the northern border of the United States. The cycle of the cestode is complex in taiga at lower latitudes, because of the greater diversity of potential hosts. In the Arctic and Subarctic, however, four patterns of predator/prey relationships may be discerned. Two natural cycles involve the wolf and wild reindeer and the wolf and elk (moose), respectively. Where deer of the two species coexist, both are prey of the wolf; the interactions of the wolf and elk are here described on the basis of long-term observations made on Isle Royale (in Lake Superior near the southern limit of taiga), where only the wolf and elk serve as hosts for E. granulosus. A synanthropic cycle involving herding-dogs and domesticated reindeer caused hyperendemicity of cystic echinococcosis in arctic Eurasia, mainly in northeastern Siberia. The 4th pattern, a semi-synanthropic cycle, formerly existed in Alaska, wherein sled-dogs of the indigenous hunters became infected by consuming the lungs of wild reindeer. The sequence of changes in life-style inherent in the process of acculturation affected the occurrence of cystic echinococcosis among nomadic Iñupiat in arctic Alaska. When those people became sedentary, the environs of their early villages soon became severely contaminated by faeces of dogs, and cases of cystic echinococcosis occurred. Compared to cystic echinococcosis caused by E. granulosus adapted to synanthropic hosts (dog and domestic ungulates), the infection produced by the northern biotype is relatively benign. Nearly all diagnosed cases of cystic echinococcosis (>300) in Alaska have occurred in indigenous people; only one fatality has been recorded (in a non-indigenous person). After sled-dogs were replaced by machines, cases have become rare in Alaska. A similar effect has been observed in Fennoscandia, in the Saami and domesticated reindeer. Recent records indicate that the prevalence of cystic echinococcosis is increasing in Russia, suggesting that dogs are used there in herding.

The population biology of the schistosome-vector snail Bulinus truncatus was studied in an irrigation area near Marrakech, Morocco, using demographic approaches, in order to estimate life-history parameters. The survey was conducted using 2 capture-mark-recapture analyses in 2 separate sites from the irrigation area, the first one in 1999 and the second one in 2000. Individuals larger than 5 mm were considered. The capture probability varied through time and space in both analyses. Apparent survival (from 0·7 to 1 per period of 2–3 days) varied with time and space (a series of sinks was considered), as well as a square function of size. These results suggest variation in population intrinsic rate of increase. They also suggest that results from more classical analyses of population demography, aiming, for example at estimating population size, should be interpreted with caution. Together with other results obtained in the same irrigation area, they also lead to some suggestions for population control.

Non-pathogenic trypanosomes of the subgenus Herpetosoma are normally host specific, and laboratory models include Trypanosoma lewisi in rats and Trypanosoma musculi in mice. Two isolates of Trypanosoma grosi, originating from Apodemus agrarius and Apodemus peninsulae, grew well in Mongolian jirds, Meriones unguiculatus, after intraperitoneal inoculation of 2×105 or a minimum 500 bloodstream forms. The course of T. grosi infection in jirds resembled T. musculi infection in mice, rather than T. lewisi infection in rats. At week 2 to 3 p.i. trypanosomes disappeared from the bloodstream, and neither prednisolone treatment nor splenectomy prevented parasite elimination from the bloodstream. However, these treatments induced a marked increase in peak parasite counts. Regardless of prednisolone treatment or splenectomy, all jirds after day 21 p.i. became resistant to the reinfection. Although no trypanosomes were detected in the bloodstream of recovered jirds, dividing parasites persisted in the medullary capillaries of the kidney, like T. musculi infection in mice. We propose the T. grosi infection in jirds as an additional laboratory model for the study of non-pathogenic trypanosomes.

Laboratory reared Culex quinquefasciatus Say, 1823 originating from the vicinity of Belem, in northern Brazil, were allowed to engorge on caimans (Caiman c. crocodilus) infected with Hepatozoon caimani (Carini, 1909) and boas (Boa constrictor) infected with H. cf. terzii (Sambon and Seligmann, 1907) both from Para State. Engorged mosquitoes killed on successive days post-feeding (p.f.) were studied by transmission electron microscopy. Images of oocysts from 13 days p.f., caiman-fed mosquitoes were also examined by scanning electron microscopy. The Hepatozoon spp. from the respective hosts differed in their ability to develop in C. quinquefasciatus. Most female mosquitoes fed on caimans, became fully engorged, and survived beyond 22 days p.f., whereas those engorged on boa became debilitated and did not survive beyond 9 days p.f. In boa-fed mosquitoes oocysts were found on the 6th day p.f. The few mosquitoes surviving to the 9th day p.f. contained either undivided oocysts or those that had already commenced sporogenic division. By 8–10 days p.f. caiman-fed mosquitoes contained uninucleate oocysts. Sporogonic divisions were observed from day 12 p.f. onwards. Although sporogenic development conformed in general with the previously reported accounts, the study allowed a more detailed examination of the plasmalemmal endocytotic system, and the consolidation of the crystalline body in specialized ‘factories’ of cystalline material. Sporozoite differentiation occasionally started on the 18th day p.f., but otherwise was observed only on day 22 p.f.

The intestinal macroparasite communities of freshwater eels (Anguilla anguilla) captured in the south of England from Windsor (River Thames) during August 2001, and Leckford (River Test) during late June/July 2000, are reported for the first time. Parasite component communities were among the most species rich and diverse recorded from European eels. A total of 13 intestinal macroparasite species were encountered during the study, 8 from each eel host population with 3 being common to both. Acanthocephalans, nematodes and cestodes were recovered from each host population. Eels from Windsor additionally harboured Nicolla gallica (Digenea), which was also the most prevalent and abundant macroparasite species in these hosts. Each component community followed a log normal rank abundance distribution and demonstrated reduced species dominance and increased species equitability compared with previous studies. As such, the study component communities were suitable for testing the hypothesis of low infracommunity diversity in European eel hosts. Specifically, this hypothesis predicts that the intestinal macroparasite infracommunities of European eels are species-poor, displaying low density and diversity with high dominance, irrespective of component community diversity, and that this may be more pronounced in UK host populations. This hypothesis was not upheld; study findings demonstrate that higher infracommunity diversity in eel hosts is possible, and suggest that infracommunity diversity in individual eel hosts may be a simple, stochastic reflection of component community diversity.

A Polymerase Chain Reaction (PCR)- based assay developed for the specific identification of Thelazia gulosa, Thelazia rhodesi and Thelazia skrjabini (Nematoda, Spirurida), which cause bovine ocular thelaziosis, was evaluated for its usefulness in detecting the intermediate hosts and in estimating the infection prevalence of vectors in field conditions throughout 5 years (from 1997 to 2001). A total of 5190 flies were captured and identified as Musca larvipara, Musca osiris, Musca autumnalis, Musca tempestiva or Musca domestica. Genomic DNA was extracted from pools constituted by heads, thoraces, abdomens and wings of 10 flies of each species, and 2076 samples were subjected to a PCR assay to specifically detect the ribosomal ITS-1 sequence of bovine Thelazia. Amplicons were sequenced and subjected to digestion with CpoI restriction enzyme. M. autumnalis, M. larvipara, M. osiris and M. domestica species were shown to be PCR positive. T. gulosa was specifically detected by PCR in M. autumnalis, M. larvipara, M. osiris and M. domestica, whereas T. rhodesi is in M. autumnalis and M. larvipara. Of 27 positive samples, 23 were positive for T. gulosa and 4 for T. rhodesi, with a mean prevalence of 2·86% in the whole fly population collected. The highest mean prevalence values of infection were detected in M. autumnalis (4·46%) and M. larvipara (3·21%), and the former species was confirmed to be the vector of T. gulosa and T. rhodesi. This study is the first report of M. osiris as a vector of T. gulosa and M. larvipara as a vector of T. gulosa and T. rhodesi under natural conditions. The occurrence of Thelazia in fly populations in the Apulia region of Italy (in the 5 grazing seasons considered) indicates that cattle thelaziosis is enzootic in southern Italy. This molecular assay should be a useful epidemiological tool for assessing the role of different species of flies as intermediate hosts of thelaziae.

Human alveolar echinococcosis (AE), caused by the metacestode of the fox tapeworm Echinococcus multilocularis, is the most pathogenic zoonosis in temperate and arctic regions of the northern hemisphere. Prospective collection of human cases in some areas and mass screenings using ultrasound imaging and confirmation with serological techniques have markedly improved our knowledge of the epidemiology of the disease in humans during the past two decades. Transmission occurs when eggs of the tapeworm, excreted by the final hosts (usually foxes but also dogs, wolves and cats), are ingested accidentally by humans or during normal feeding by a variety of rodents and small lagomorphs. However, the species of host animals differ according to regional changes in mammalian fauna. This review mostly focuses on epidemiology of alveolar echinococcosis in those parts of the world where new and more accurate epidemiological data are now available, i.e. China and Europe, as well as on new epidemiological trends that can be suspected from recent case reports and/or from recent changes in animal epidemiology of E. multilocularis infection. The People's Republic of China (PRC) is a newly recognized focus on AE in Asia. Human AE cases were firstly recognized in Xinjiang Uygur Autonomous Region and Qinghai Provinces at the end of 1950s and infected animals were first reported from Ningxia in central China and north-east of Inner Mongolia in the 1980s. E. multilocularis (and human cases of AE) appears to occur in three areas: (1) Northeastern China (northeast focus): including Inner Mongolia Autonomous region and Heliongjiang Province (2) Central China (central focus): including Gansu Province, Ningxia Hui Autonomous Region, Sichuan Province, Qinghai Province and Tibet Autonomous Region and (3) Northwestern China: including Xinjiang Uygur Autonomous Region, bordered with Mongolia, Russia, Kazakhstan and Kyrgyzstan. The highest prevalence of the disease, up to 15 per cent of the population in some villages, is reached in China. In Europe, data from the European Echinococcosis Registry (EurEchinoReg: 1982–2000) show 53 autochthonous cases of AE in Austria, 3 in Belgium, 235 in France, 126 in Germany, 1 in Greece, and 112 in Switzerland, and 15 ‘imported’ cases, especially from central Asia; 14 cases were collected in Poland, a country not previously considered endemic for AE. Improved diagnostic technology, as well as a real increase in the infection rate and an extension to new areas, can explain that more than 500 cases have been reported for these 2 decades while less than 900 cases were published for the previous 7 decades. New epidemiological trends are related to an unprecedented increase in the fox population in Europe, to the unexpected development of urban foxes in Japan and in Europe, and to changes in the environmental situation in many countries worldwide due to climatic or anthropic factors which might influence the host–predator relationship in the animal reservoir and/or the behavioural characteristics of the populations in the endemic areas.

As a potential ultimate cause of microhabitat restriction for ectoparasites of fish, it has been postulated that selection has favoured narrow microhabitats to facilitate mating. The evidence for this hypothesis is evaluated with males and females of the parasitic copepod Lernanthropus cynoscicola, by using methods commonly used in community ecology, and considering the influence of host ontogeny on site selection. No evidence supporting the mating hypothesis was found after comparison of niche measurements between both sexes. Furthermore, aggregation among individuals of the same sex was stronger than among males and females, and the co-occurrence of both sexes did not depart from that expected by chance; also, negative correlations between the intensity of infection of both sexes were observed. Restriction of the microhabitats cannot therefore be due to facilitation of mating. Moreover, both sexes displayed a sequential displacement over the gill arches and differential preferences by specific gill sections in response to an increasing host size, probably as a consequence of a host-size related cline of optimal conditions where fitness is improved. Thus, reproductive benefits other than increased chances of mating may have played a role in the evolution of restricted niches in parasites, and selection may have favoured a narrowing of the niche around sites where fitness is maximized.

Concentrations of lead, determined by electrothermal atomic absorption spectrometry, were compared between the cestodes Hymenolepis diminuta and Taenia taeniaeformis and its host rat (Rattus norvegicus). Rats were sampled at 2 sites, which differed in respect to lead pollution as quantified from road dust, adjacent to the city of Cairo, Egypt. Comparing lead levels among host tissues and the parasites the significantly highest accumulation was found in H. diminuta, followed by rat kidney and larvae of T. taeniaeformis. Calculation of bioconcentration factors showed that H. diminuta contained 36-, 29-, 6- and 6-fold higher lead levels than intestinal wall, liver, kidney and larvae of T. taeniaeformis, at the more polluted site. At the less contaminated site lead bioconcentration factors for H. diminuta were found to be 87, 87 and 11 referred to intestine, liver and kidney of the host. Due to a high variability of the lead concentrations in H. diminuta it was not possible to indicate differences in metal pollution between both sampling sites. This variability may be influenced by different age structures of cestode infrapopulations. It is likely that younger worms contain lower metal levels than older worms due to a shorter exposure period. Thus, it is necessary to standardize the sampling of worms which should be used for indication purposes. Due to a lack of adequate sentinel species in terrestrial habitats more studies are required to validate and standardize the use of helminths as accumulation bioindicators in order to obtain mean values with low standard deviations. The host–parasite system rat–H. diminuta appears to be a useful and promising bioindication system at least for lead in urban ecosystems as rats as well as the tapeworm are globally distributed and easily accessible.

A β-1,3-glucan-binding lectin from the penetration glands of Diplostomum pseudospathaceum cercariae was isolated by affinity chromatography using yeast glucan and curdlan as affinity matrices. Further purification to homogeneity was performed by cation-exchange chromatography. The protein migrated as a double band around 24 kDa in gels after SDS–PAGE. The protein is of strongly basic nature – its pI shown by native IEF was around 10. The mass of the protein determined by MALDI-TOF mass spectrometry was 23·9 kDa. N-terminal sequence as well as some internal sequences showed significant alignments with several cysteine protease sequences found in databases. The protein bound a biotinylated synthetic analogue of the irreversible inhibitor of cysteine proteases, E-64 and, moreover, its proteolytic activity was demonstrated in substrate gels. The enzymatic activity could be inhibited by the cysteine protease inhibitor E-64; therefore, the investigated protein was considered to be a bifunctional molecule possessing both lectin and enzyme activities. Glycanohydrolytic activity was not proved. The detected characters of this molecule lead to a hypothesis on its role in penetration of Diplostomum cercariae into fish hosts – that of binding to the carbohydrates of fish mucus and concurrent cleaving of protein components of the mucus and skin.

Manipulative endoparasites can alter the behaviour and the physiology of their intermediate hosts in ways that increase the probability of successful transmission to the final host. This requires that the parasite is able to circumvent its host's immune defence. Successful immune evasion may depend on host–parasite coevolutionary history and the appearance of new hosts invading the local host population may promote local parasite maladaptation. To test this hypothesis, we examined the effect of 2 acanthocephalan parasites, Pomphorhynchus laevis and Polymorphus minutus, on the immunity of their local and new invasive gammarid intermediate hosts, respectively Gammarus pulex and Gammarus roeseli. We found that infection by each parasite was correlated with a decrease, at different degrees, of the standing level of immune defence in local hosts – measured as the phenoloxidase (PO) enzyme activity – whereas invasive hosts infected by P. laevis had their PO-enzyme activity enhanced. These results suggest that these acanthocephalans evade their local host immune response through immunosuppression but cannot evade the immune response of their new invasive host. The potential role of this maladaptation on the success of invasive species is discussed.

A variety of fluorescent probes have been used to study the acidic compartments in cercariae and schistosomula of Schistosoma mansoni. Freshly transformed schistosomula treated with the LysoTracker Red dye specific for lysosomes showed large acid-containing compartments (0·5–10 μm in size). The uptake of the dye is an energy-dependent process that depends on the metabolic activity of schistosomula. The compartments were quantified individually with respect to area, quantity of fluorescence and the total number/schistosomulum. Under normal conditions these compartments were not found in untreated cercariae, but appeared in cercariae slightly damaged by poly-L-lysine. The formation of these compartments seemed to be related to the development of cercariae into schistosomula as the number of compartments and uptake of fluorescence increased with time after transformation. Also, the method of transformation as well as the in vitro incubation of the parasite affected the percentage area of compartments/schistosomulum. Acid phosphatase enzyme activity was assessed using an endogenous phosphatase probe. Living and fixed schistosomula displayed the presence of enzyme activity in compartments of the same size and distribution as the acid-rich compartments. This was confirmed by histochemical staining showing deposition of enzyme-generated lead at the sites of phosphatase activity. We suggest that the development of acidic compartments is important during the transformation process or as a consequence of damage.

A laboratory population of a Danish Gyrodactylus salaris variant founded by 1 single specimen was established and infection studies performed. Rainbow trout as well as Atlantic salmon of 3 different stocks were infected both in cohabitation systems and as single-parasite infections on isolated hosts. Both infection systems revealed that this particular morphotype exhibits low virulence towards Atlantic salmon. Thus, in isolated hosts, the parasites could either not establish or only reproduce to a limited degree on salmon. Rainbow trout, in contrast, proved to be rather susceptible to infection with this G. salaris variant and abundances on this host species always attained significantly higher values in cohabitation systems compared to salmon. Detailed morphological examination confirmed the very high resemblance to G. salaris (sensu stricto), as the range of variation in sclerite characters of the morphotype is almost fully covered by the total range of variation reported for reference G. salaris. Morphological similarities to the closely related congeneric species G. bohemicus were noted. Molecular studies, however, showed that the morphotype most likely represents a G. salaris variant, as it differed only slightly from G. salaris sensu Malmberg, 1957, which is also known to inhabit Danish watercourses. The genomic target region investigated does not allow us to rule out the possibility that it represents a variant form of G. thymalli. Sequences of the ribosomal RNA internal transcribed spacer (ITS) revealed that single individuals contained 2 different ITS sequences, one identical to reference sequence of G. salaris while the other differed at 3 positions. This can be interpreted as an example of a hybrid or, more likely, as intra-individual variation of ITS within single individuals. As one of the nucleotide changes in the variant ITS affects an Hae III restriction site, the current G. salaris variant can be distinguished from G. salaris sensu Malmberg by RFLP diagnosis.

Infections by larval stages of tapeworms of the genus Echinococcus (echinococcosis or hydatid disease) are zoonotic infections of major public health importance throughout much of the world. Humans become infected through accidental ingestion of eggs passed in faeces of canid definitive hosts. Tibetan populations of China have some of the highest documented levels of infections by both Echinococcus granulosus and E. multilocularis, the causes of cystic and alveolar echinococcosis, respectively. In this study we measured the prevalence of cystic (CE) and alveolar (AE) echinococcosis disease in Tibetan communities in Qinghai, Province, China, and identified putative risk factors for both infections in these communities. 3703 volunteers in three predominately Tibetan counties of Qinghai were surveyed between June 1997 and June 1998. Parasitic lesions were diagnosed by imaging of characteristic space-occupying lesions in abdominal organs (ultrasound) or the lungs (radiographs). Specific serodiagnostic assays (Dot-ELISA and Em2-ELISA) were performed on sera of positively imaged subjects to further distinguish the disease agent. All participants completed a questionnaire documenting age, sex, education level, occupation, lifestyle (nomadic or settled), slaughter practices, drinking water source, hygienic practice and association with dogs. Data were analyzed using SAS version 8. 6·6% of the volunteers had image-confirmed infection with E. granulosus (CE) and 0·8% had E. multilocularis (AE) infection. The significant univariate factors for echinococcal infection (both CE and AE) included livestock ownership, Tibetan ethnicity, female gender, low income, herding occupation, limited education, water source, age greater than 25 years old, poor hygienic practices, offal disposal practices and dog care. Multivariate analysis revealed that livestock ownership was a significant risk factor for both forms of the disease, as well as age greater than 25 years, female gender, herding occupation, and being nomadic (vs semi-nomadic or settled). No additional significant risk factors were identified among the 344 nomadic participants. Being female and being older than 25 years of age were significant factors among the 1906 semi-nomadic participants. Among the 1445 settled participants, allowing dogs to sleep indoors was statistically significant. Issues such as inadequate assessment of animal ownership, selection bias, disease misclassification, and loss of information may have led to reduction in strength of some risk factor associations and need to be addressed in future epidemiologic analysis of echinococcosis in this population.

The population structure and recruitment of Argulus coregoni was monitored at a Finnish fish farm during the open water periods of 1999 and 2001 by weekly sampling of attached argulids. In 2001 the numbers of rainbow trout examined increased in the autumn when the A. coregoni population was declining. When the water temperature exceeded 10 °C, at the end of May, A. coregoni egg hatching commenced. A mean number of 98 (S.D.±5·4) juvenile A. coregoni was recorded on each fish, before the start of female egg laying in July 1999. The abundance of lice was lower in 2001. The main recruitment of A. coregoni juveniles occurred in early summer, but the hatching of eggs continued until September. However, no pulses of hatching were recorded later in the summer and the numbers of lice on fish substantially decreased after mid-July in both years. On average, only 0·6 (S.D.±0·72) parasites/fish were found between August and late October 2001 and none in November. We suggest that there is 1 main A. coregoni generation annually in Central Finland. Environmental conditions, especially temperature, affects the population cycle of A. coregoni: we found a more synchronous and intense population cycle during the summer 1999, when the early summer was warmer than in 2001. The overall sex ratio (female[ratio ]male) of A. coregoni was nearly 1[ratio ]1 in June 1999, but was male biased (1[ratio ]1·4) in June 2001. It was also shown that from July onwards, many females detached from the fish host in order to lay their eggs on the bottom of the pond. Large males were often bigger than the largest females between mid-July and early September 2001, when as many as 3·7 (S.D.±1·48) times more males than females were present in the lice samples.

Two full-length beta-tubulin cDNAs, representing isotypes 1 and 2, were cloned from the cattle nematode Cooperia oncophora. The predicted protein sequences span 448 amino acids, and show a high degree of identity to beta-tubulins from other nematodes. While C. oncophora isotype 1 sequence had the highest identity to Haemonchus contortus isotype 1 and Teladorsagia circumcincta sequences (95% identity), the C. oncophora isotype 2 sequence was most similar to H. contortus isotype 2 and Trichostrongylus colubriformis (92% identity). Alignment of the two C. oncophora sequences with other trichostrongylid beta-tubulins deposited in GenBank showed a clear distinction between isotype 1 and 2 beta-tubulin classes. The two classes differed at 19 amino acid positions, most notably at the carboxy terminus. These isotype-defining residues were conserved among different trichostrongylid species within a class. Analysis of fragments of both genes revealed a high degree of genetic variability in coding and non-coding regions. However, all nucleotide differences detected in the coding region were silent, as they did not result in any amino acid substitution. Analysis of 2 groups of worms for the codon 200 polymorphism associated with benzimidazole resistance revealed a proportion of worms in 1 of the groups bearing a tyrosine at this position.
