A statistically derived disease reaction index based on parasitological, clinical and haematological measurements observed in 309 5 to 8-month-old Boran cattle following laboratory challenge with Theileria parva is described. Principal component analysis was applied to 13 measures including first appearance of schizonts, first appearance of piroplasms and first occurrence of pyrexia, together with the duration and severity of these symptoms, and white blood cell count. The first principal component, which was based on approximately equal contributions of the 13 variables, provided the definition for the disease reaction index, defined on a scale of 0–10. As well as providing a more objective measure of the severity of the reaction, the continuous nature of the index score enables more powerful statistical analysis of the data compared with that which has been previously possible through clinically derived categories of non-, mild, moderate and severe reactions.

Thymidylate synthase, dihydrofolate reductase and dUTPase specific activities were found to remain at a high and constant level in crude extracts from adult worms of Trichinella spiralis, as well as from muscle larvae of both Trichinella spiralis (isolated 1–24 months after infection) and Trichinella pseudospiralis (isolated 5·5–13 months after infection). The results obtained with Trichinella pseudospiralis muscle larvae isolated with the use of pepsin did not differ from those obtained when pepsin was not used. No thymidine kinase activity could be detected in muscle larvae of either species and thymidine phosphorylase could be found only in T. pseudospiralis larvae isolated without the use of pepsin. Muscle larvae of both species contained orotidylate phosphoribosyl transferase activity, pointing to a possibility of 5-fluorouracil activation. Uridine phosphorylase, another enzyme involved in 5-fluorouracil anabolism, was also present in T. pseudospiralis muscle larvae. Results of comparative studies on inhibition of purified T. spiralis and rat thymidylate synthases by substrate (4-thio-5-fluoro-dUMP, 2-thio-5-fluoro-dCMP and N4-hydroxy-dCMP) and cofactor (ZD 9331) analogues indicated only dUMP analogues to show feeble selectivity towards the parasite enzyme. A hypothesis is discussed, assuming high expression of thymidylate synthase in muscle larvae to be connected with their cells being arrested in the cell cycle.

Male and female schistosomes are generally assumed to form stable monogamous pairs for the whole span of their long existence in the mammalian host. Recent evidence from mixed infections has shown that Schistosoma mansoni males can displace S. intercalatum males from their homologous partners, but no information exists about the existence of similar phenomena within a single schistosome species. Here, we determine whether male S. mansoni can displace males of the same species from pre-formed pairs in vivo. The availability of clear-cut genetic markers of drug resistance in schistosomes was exploited to show that hycanthone sensitive S. mansoni males can displace homospecific hycanthone resistant males from pre-formed pairs and vice versa. The frequency of changes is dependent on the magnitude of the excess single males competing with paired worms. The possible mechanics and the biological significance of mate changing are discussed.

Trypanosomiasis is a major veterinary problem over much of sub-Saharan Africa and is frequently associated with undernutrition. There is growing evidence that nutrition can have a profound effect on the pathophysiological features of animal trypanosomiasis. These features include anaemia, pyrexia, body weight changes, reduced feed intake and diminished productivity including reduced draught work output, milk yield and reproductive capacity. Anaemia is a principal characteristic of trypanosomiasis and the rate at which it develops is influenced by both protein and energy intakes. Pyrexia is associated with increased energy demands for maintenance which is ultimately manifested by reductions in voluntary activity levels and productivity. Weight changes in trypanosomiasis are markedly influenced by the levels of protein intake. High intakes allow infected animals to grow at the same rate as uninfected controls providing energy intake is adequate whilst low energy levels can exacerbate the adverse effects of trypanosomiasis on body weight. Reductions in feed intake are less apparent in animals which are provided with high protein diets and where intake is limited by the disease animals will often exhibit preferential selection of higher quality browse. Further studies are required to evaluate the minimum levels of protein and energy supplementation required to ameliorate the adverse effect of trypanosomiasis, the nature and quality of protein supplement to achieve these benefits and the influence these have on digestive physiology.

A cDNA library derived from mRNA of tachyzoites of Neospora caninum (NC-Liverpool strain) was screened with antisera from a cow naturally infected with N. caninum. The DNA sequence of 1 recombinant isolated predicted a significant protein sequence homology of the gene product to the 28 kDa (GRA2) antigen of Toxoplasma gondii. Studies on the N. caninum gene coding for this antigen demonstrated the presence of a single intron flanked by 2 exons; the gene was also highly expressed in culture-derived tachyzoites. The antigen was expressed in Escherichia coli; when injected into mice it stimulated the production of antibodies which detected a 29 kDa antigen of N. caninum. Secondary structure predictions made for the N. caninum protein showed support for several amphipathic helices separated by loops and turns. The available evidence indicates maintenance of protein secondary structure, and not DNA or amino acid sequence, has occurred during the evolution of GRA2 proteins in N. caninum and T. gondii.

Gyrodactylus salaris infects numerous salmonid species, ranging from the fully susceptible (Norwegian strains of Salmo salar), through species which, though initially susceptible, eventually eliminate their infections (Salvelinus alpinus and S. fontinalis) to entirely resistant (Salmo trutta) species. Here we describe experiments in which Salvelinus alpinus, S. fontinalis and Salmo trutta, implanted with hydrocortisone acetate to simulate stress-induced immunosuppression, were challenged with G. salaris. With previously uninfected Salvelinus fontinalis, G. salaris infections on fish treated with hydrocortisone acetate grew larger, and for longer, than on sham-treated controls. A similar result was obtained with S. trutta. Patterns of infection on Arctic charr, Salvelinus alpinus, were more complex, because individual fish varied from susceptible to highly resistant. Fish were therefore initially infected with G. salaris, and the most highly resistant group of individuals identified and disinfected. After 6 months recovery from this primary infection, hydrocortisone acetate was administered to half the fish, and all were challenged with G. salaris. Parasite populations on the hydrocortisone-treated individuals were consistently larger than those on the sham-treated controls, exceeding 30 parasites per fish after 5 weeks, in comparison with less than 10 parasites per fish on controls. These results indicate that hydrocortisone administration can lead to enhanced gyrodactylid populations on a range of salmonids. This suggests that the response to G. salaris is mediated by the immune system, and that the spectrum of responses observed in different species are, at least in part, due to the same mechanism. At a practical level, stress-induced immunosuppression during handling and transport of cultured salmonids may prove an important factor in the dissemination of G. salaris between watersheds.

Experiments were conducted to investigate the potential for transmission of Orientia tsutsugamushi, the aetiological agent for scrub typhus, when naturally infected mite larvae were co-feeding with uninfected larvae. Larvae from colonies of Leptotrombidium deliense and L. imphalum infected with O. tsutsugamushi were used. Transmission of O. tsutsugamushi to previously uninfected L. deliense and Blankaartia acuscutellaris co-fed with infected L. deliense was shown to occur. The overall minimum rate of acquisition was 1·6% (4/258) for L. deliense and 2·5% (3/119) for B. acuscutellaris. When individual infected L. deliense were co-fed with B. acuscutellaris acquisition of O. tsutsugamushi was not detected. However, when 4 and 8 infected larvae were co-fed with B. acuscutellaris acquisition of O. tsutsugamushi was detected. Transmission of O. tsutsugamushi was not observed when uninfected L. deliense were co-fed with infected L. imphalum. This novel transmission route may explain the occurrence of rickettsiae in genera other than Leptotrombidium spp, which are considered to be the main vectors of O. tsutsugamushi.

Bidirectional signalling between Trypanosoma brucei brucei and CD8+ T cells involves reciprocal action of the parasite-derived trypanosome derived lymphocyte triggering factor (TLTF) and lymphocyte-derived IFN-γ. Herein we further characterize this relationship and report quantitation of TLTF secretion into culture supernatants using a newly developed ELISA. Secretion is induced by IFN-γ in a dose–response manner, with 100 U/ml giving maximal yields of bioactive TLTF as assessed by ELISA and ELISPOT. This is a constitutive and active secretion. Furthermore, IFN-γ-induced secretion was dependent on tyrosine protein kinase activity. Specific blockers of this signalling system resulted in lower yields of TLTF in the culture supernatants.

In this paper we address the question of detecting immunity to helminth infections from patterns of infection in endemic communities. We use stochastic simulations to investigate whether it would be possible to detect patterns predicted by theoretical models, using typical field data. Thus, our technique is to simulate a theoretical model, to generate the data that would be obtained in field surveys and then to analyse these data using methods usually employed for field data. The general behaviour of the model, and in particular the levels of variability of egg counts predicted, show that the model is capturing most of the variability present in field data. However, analysis of the data in detail suggests that detection of immunity patterns in real data may be very difficult even if the underlying patterns are present. Analysis of a real data set does show patterns consistent with acquired immunity and the implications of this are discussed.

Three species of poeciliids (Gambusia holbrooki, Xiphophorus helleri and X. maculatus) and 15 species of ecologically similar native freshwater fishes (mainly eleotrids, ambassids, melanotaeniids and retropinnids) were examined for parasite richness to investigate parasite flux, qualitative differences, quantitative differences and the structuring factors in parasite communities in the 2 fish types in Queensland, Australia. Theory suggests that poeciliids would harbour depauperate parasite communities. Results supported this hypothesis; poeciliids harboured more species-poor parasite infracommunities and regional faunas than natives (P < 0·0001), despite greater sampling effort for the former. Cluster analysis of presence/absence data for poeciliids and the 6 most-sampled native fishes revealed that parasite communities of the 2 fish groups are qualitatively distinct; the proportion of parasite species with complex life-cycles was lower in poeciliids than in native species, and Myxosporea, Microspora, Coccidia and parasitic Crustacea were all absent from poeciliids. Limited exchange of parasite species has occurred between natives and poeciliids. Logistic ordinal regression analysis revealed that fish origin (exotic or native), environmental disturbance and host sex were all significant determinants of parasite community richness (P < 0·05). Theoretical modelling suggests that poeciliids are at a competitive advantage over native fishes because of their lack of parasites.

Acanthocephalans are polygamous parasites of vertebrates and some species are known to aggregate in sexual congress to mate. Such a reproductive behaviour could lead to male–male competition for access to females and could have consequences for sexual selection. We dissected 87 gobiid fish, Gobius bucchichii, harbouring 891 acanthocephalans, Acanthocephaloides propinquus. The parasites were sexed and their body sizes were measured. Testicular volume was also evaluated in 82 males in order to establish their phenotypic sexual investment in relation to the estimated sex ratio. We found that parasite intensity (i.e. the number of individuals/fish) was not correlated with fish size, but that parasite size was significantly related to host size. Our results showed that there was a significant relationship between the mean female body size and their number within one host. We found that when the percentage of male parasites in a host increased, presumably increasing male–male competition for access to females, males had a larger testicular volume. We discuss these results in terms of energy allocation, sexual and sperm competition. We conclude that competition for space should be less important for males than competition for access to females. Moreover, increasing testis size should confer advantages to males especially for their reproductive success when sperm competition occurs.

The study aims were to quantify the survivorship of Anguillicola crassus in eels in seawater and investigate transmission in estuarine (50% seawater) and marine (100% seawater) simulated conditions. Most A. crassus were able to survive and reproduce in 50% and 100% seawater eels for up to 6 months and therefore could survive during the spawning migration of eels to the Sargasso Sea. Up to 10% of the parasite metapopulation were damaged after long-term exposure to 50% or 100% seawater. Transmission of A. crassus was completed in 50% and 100% seawater in the laboratory, although infection levels in 100% seawater hosts were always lower. Transmission in estuaries is ecologically possible since the copepod Eurytemora affinis is susceptible to infection and is the dominant autochthonous species in most tidal estuaries in the Northern hemisphere. Transmission at sea is unlikely due to lack of suitable intermediate hosts. The data support the suggestion that dissemination of A. crassus has been facilitated by natural movement of eels in coastal waters. It is possible that A. crassus could impair the success of the eel spawning migration to the Sargasso Sea, although there is no direct evidence to support this.

Nitric oxide (NO) produced by cytokine-activated macrophages is reported to be cytotoxic against the helminth Schistosoma mansoni, although this is a controversial issue. Previous work in our laboratory identified a fraction of S. mansoni soluble adult worm antigenic preparation (SWAP), named PIII, able to elicit significant in vitro cell proliferation and at the same time lower in vitro and in vivo granuloma formation when compared either to soluble egg antigen (SEA) or to SWAP. Here we report that, in comparison to other S. mansoni antigenic preparations (SEA and SWAP), supernatants of PBMC cultivated with PIII possess higher concentrations of interleukin-10 (IL-10) and macrophage inflammatory protein (MIP-1α), concomitantly with lower concentrations of monocyte chemoattractant protein (MCP-1) and regulated on activation, normal T expressed and secreted (RANTES). In the particular case of NO inhibition, supernatants of PBMC cultivated with PIII present decreased IL-10 levels. Altogether, these results indicate that IL-10, MIP-1α, MCP-1 and RANTES are distinctively important elements in the PIII modulating role, while NO seems to be pivotal in the regulation of granulomatous responses.

It is pointed out that two of the three major groups of anthelmintic act by opening membrane ion-channels. It is appropriate, therefore, to use electrophysiological methods to study the properties of the sites of action of these drugs and the changes in the properties of these receptor sites associated with resistance. This paper describes the use of the patch- clamp technique to observe the currents that flow through the levamisole-activated channels as they open and close in levamisole-sensitive and levamisole-resistant isolates. It was found that, on average, the proportion of time the channels are open, is less in the resistant isolate. The patch-clamp technique also showed that the ion-channels are heterogeneous and that one of the subtypes is lost with the appearance of resistance. The use of the current clamp technique is illustrated to record a site of action of ivermectin in the pharyngeal muscle of Ascaris.

Hookworms are gut-dwelling, blood-feeding nematodes that infect hundreds of millions of people, particularly in the tropics. As part of a program aiming to define novel drug targets and vaccine candidates for human parasitic nematodes, genes expressed in adults of the human hookworm Necator americanus were surveyed by the expressed sequence tag approach. In total 161 new hookworm genes were identified. For the majority of these, a function could be assigned by homology. The dataset includes proteases, protease inhibitors, a lipid binding protein, C-type lectins, an anti-bacterial factor, globins and other genes of interest from a drug or vaccine development viewpoint. Three different classes of small, secreted proteins were identified that may be involved in the host–parasite interaction, including potential potassium channel blocking peptides. One third of the genes were novel. These included highly expressed, secreted (glyco)proteins which may be part of the excretory–secretory products of these important pathogens. Of particular interest are a family of 9 genes with similarity to the immunomodulatory protein, neutrophil inhibitory factor, that may play a role in establishing an immunocompromised niche for this successful parasite.

Schistosoma mansoni depends for its survival on glycolysis. Two glycolytic enzymes, glyceraldehyde-3P-dehydrogenase and triose-phosphate dehydrogenase, found in both the adult and schistosomular tegument, have been reported to confer partial protection against cercarial infection. This paper describes the immunogenic properties of phosphofructokinase (PFK), a rate-limiting enzyme of glycolysis, and its localization in the tegument and adjacent tissues. Recombinant schistosome PFK was used as antigen. A polyclonal antibody against purified PFK from Fasciola hepatica was affinity purified using recombinant PFK and used in combination with immunogold labelling to identify PFK by transmission electron microscopy in cryosections. In both adult worms and in schistosomula most immunogold label localized in the cytoplasmic syncytial region with less being found in the tegument. There was no significant PFK localization within or external to the outer membrane. Sera from mice immunized with recombinant S. mansoni PFK with Freund's adjuvant or alum plus rIL-12 demonstrated high titres of anti-PFK IgG, but no protection against cercarial infection. Sera from mice that were acutely or chronically infected or multiply exposed to irradiated cercariae did not recognize recombinant schistosome PFK in either Western blotting or ELISA. Similarly, sera from humans infected with S. mansoni did not recognize PFK. We conclude that in spite of the high immunogenicity of rPFK in mice, it is not a significant immunogen during the course of infection and does not confer protection from schistosomiasis. One main difference between PFK and the other 2 glycolytic enzymes seems to be the inaccessibility of PFK to the outside surface of the tegument.

When two parasite species are manipulators and have different definitive hosts, there is a potential for conflict between them. Selection may then exist for either avoiding hosts infected with conflicting parasites, or for hijacking, i.e. competitive processes to gain control of the intermediate host. The evidence for both phenomena depends largely on the study of the relative competitive abilities of parasites within their common intermediate host. We studied the effects of simultaneous infection by a fish acanthocephalan parasite, Pomphorhynchus laevis, and a bird acanthocephalan parasite, Polymorphus minutus, on the behaviour of their common intermediate host, the amphipod Gammarus pulex. We compared the reaction to light and vertical distribution of individuals infected with both parasites to those of individuals harbouring a single parasite species and uninfected ones under controlled conditions. Compared to uninfected gammarids that were photophobic and tended to remain at the bottom of the water column, P. laevis-infected gammarids were attracted to light, whereas P. minutus-infected individuals showed a modified vertical distribution and were swimming closer to the water surface. The effects of both P. laevis and P. minutus appeared to be dependent only on their presence, not on their intensity. Depending on the behavioural trait under study, however, the outcome of the antagonism between P. laevis and P. minutus differed. The vertical distribution of gammarids harbouring both parasites was half-way between those of P. laevis- and P. minutus-infected individuals, whereas P. laevis was able to induce altered reaction to light even in the presence of P. minutus. We discuss our results in relation to the occurrence of active avoidance or hijacking between conflicting manipulative parasites and provide some recommendations for future research.

The observation that the total abundance of adult nematodes in the abomasum of Svalbard reindeer increases between October and April suggests adaptation to cope with the Arctic winter. Here we investigate the extent to which selection has led to similar life-history strategies in the 3 most numerous trichostrongyle species. The life-histories are found to differ markedly. We use flexible statistical models for the abundance and dispersion of parasites in the host population. One of the taxa, Marshallagia marshalli, was most abundant and had its highest egg output in the winter. In contrast, the abundance of the most common taxa, Ostertagia gruehneri, m. gruehneri was stable or declined from autumn to late winter, and the closely related taxa, O. gruehneri, m. arcticus, showed a similar over winter drop. The faecal egg output of these 2 taxa was highest in summer, as found in temperate trichostrongyle species. Despite the apparent contamination of summer pastures with O. gruehneri, calves showed negligible burdens until their second summer and the abundance of infection reached an asymptote within their third year. In contrast, the abundance of M. marshalli in calves showed a rapid increase over the first summer and by late winter was similar to peak levels found in adults (8000 worms). This increase could not be accounted for by the developing abomasum larvae population and is therefore evidence for transmission over the winter for this taxa. While M. marshalli showed little between-year variation, O. gruehneri showed 2-fold fluctuation in the abundance of infection. O. gruehneri may therefore play a role in the fluctuating population dynamics of the host. Since there was no apparent decline in abundance with host age in any of the 3 taxa there was no evidence of reindeer mounting an immune response.

Neuropeptide F (NPF) is an abundantly expressed neuropeptide in platyhelminth nervous systems, and exhibits a moderate, myogenic effect on muscle preparations of parasitic flatworms. NPF displays structural similarities to peptides from molluscs and vertebrate members of the neuropeptide Y (NPY)-superfamily of peptides. NPY is one of the most abundant and highly conserved neuropeptides within vertebrates and similarities between the gene organization of NPY, pancreatic polypeptide (PP) and peptide tyrosine tyrosine (PYY), suggest a common evolutionary origin of this peptide family. Dual localization analyses coupled with confocal scanning laser microscopy revealed a close spatial relationship between NPF-containing nerves and muscle fibres in M. expansa. Molecular cloning techniques identified the M. expansa NPF (mxNPF) precursor and characterized the isolated transcript which encodes an open reading frame of 57 amino acids. The transcript possesses a 17 amino acid signal peptide and the mature NPF sequence (39 amino acids) followed by a carboxyterminal glycyl extension. Sequence analysis of genomic DNA identified a product which possessed a 54 base pair intron with consensus sequences for 5′ and 3′ splice sites. The M. expansa npf gene possesses a phase 2 intron within the penultimate arginyl residue, a characteristic feature of NPY superfamily peptide-genes. The intron–exon organization of the npf gene, coupled with the abundant expression of NPF within the nervous systems of flatworms, suggests an early evolutionary origin for this peptide transmitter family within the nervous systems of basal bilaterian metazoans.

Changes in gene expression in Biomphalaria glabrata following infection with Schistosoma mansoni have been investigated using a modified differential display approach. RNA was extracted from ovotestis, mantle tissue and anterior nephridium of control and exposed snails at 2 time-points (4 h and 24 h) post-exposure and analysed by RNA fingerprinting. A number of transcripts were identified; some novel and some homologous to mRNAs in GenEMBL that were previously unknown in B. glabrata. Down regulation of one 241 bp mRNA expressed sequence fragment – with an open reading frame showing 48% identity to a cytochrome p450 over 80 residues – has been confirmed using semi-quantitative RT-PCR. Preliminary classification of B. glabrata cyp450 sequence shows it to fall into CLAN 2 of the cytochrome p450 superfamily. Differential display has been successful in identifying changes in gene expression in Biomphalaria glabrata upon infection with Schistosoma mansoni and promises to be a useful technique for the investigation of the interaction between host and parasite.
