Lactoperoxidase (LPO) is a glycosylated antimicrobial protein present in milk with a molecular mass of 78 kDa. LPO is included in many biological processes and is well-known to have biocidal actions, acting as an active antibiotic and antiviral agent. The wide spectrum biocidal activity of LPO is mediated via a definite inhibitory system named lactoperoxidase system which plays a potent role in the innate immune response. With the current advancement in nanotechnology, nanoformulations can be developed for stabilizing and potentiating the activity of LPO for several applications. In the research described in this Research Communication, fresh LPO purified from bovine mammary gland secretions was used for nanoparticle synthesis using a simple thermal process at different pH and temperatures. The round-shaped nanoparticles (average size 229 nm) were successfully synthesized at pH 7.0 and a temperature of 75°C. These nanoparticles were tested against four different bacterial species namely S. flexineri, P. aeruginosa, S. aureus, and E. coli. The prepared nanoparticles exhibited strong inhibition of the growth against all four bacterial species as stated by their MIC and ZOI values. These results may help in increasing the efficiency of lactoperoxidase system and will assist in identifying novel avenues to enhance the stability and antimicrobial function of LPO in drug discovery and industrial processes.

The main objective of this research communication was to explore the extent of milkability changes caused by the incidence of clinical mastitis. Our second objective was to investigate if the milkability of cows shortly before mastitis incidence significantly differed in comparison with healthy cows. Milk yield, milking time, average milk flow, partial milk flows and the occurrence of bimodal milk flows were monitored during the first 120 d in milk for all cows that calved on the farm during the experimental period (n = 127). A veterinarian diagnosed 27 cows with clinical mastitis, while the remaining healthy cows served as a control group. The period surrounding the mastitis was monitored in a timeline from 2 weeks before to 4 weeks after the diagnosis. We did not observe any significant differences in milkability between the healthy cows and the cows in the pre-mastitis period, which suggests that monitoring these might not be useful for early mastitis detection. Milk yield and milk flow were significantly decreased in the week of mastitis diagnosis compared to the pre-mastitis period. Milking time was not affected in the week of diagnosis, but was significantly increased in the following week, because milk yield returned to pre-mastitis values, but average milk flow was still significantly decreased. Milk yield was normalized to pre-mastitis values as the first of monitored parameters. Milk flow returned to pre-mastitis values in the fourth week after mastitis diagnosis. Milking time was the only parameter that was significantly affected up to a month after diagnosis. Our results showed that milkability changes caused by mastitis are significant and should not be ignored. The ability to adjust milking settings for cows diagnosed with mastitis could become a useful tool for improving mastitis treatment.

This research reflection examines the physiological links between redox balance, skeletal health and gastrointestinal functionality in dairy cows. With the increase in demand of animal products caused by the growth in human population, the dairy industry needs to develop and implement innovative strategies which are profitable, sustainable and cow friendly. Redox balance, skeletal heath and gastrointestinal functionality are three key physiological systems that are often seen as independent entities. In this research reflection we intend to stress that the antioxidant system, bone health and the microbiome are intimately intertwined. Antioxidants are crucial for the maintenance of redox homeostasis and optimal immune function. Optimal gastrointestinal functionality is important to maintain animal performance, health and welfare. In particular, the intestinal microbiome is increasingly seen as a driver of health and disease. Vitamin D metabolism is pivotal not only for optimal skeletal health, but in light of all the extra-skeletal effect of vitamin D, it is the foundation for optimal productive life. It makes sense to ask the question ‘how are redox balance and the microbiome involved in the modulation of bone health and immune function?’ In other words, are bugs and bones connected in dairy cows! The existing data available in the literature suggests that this might be the case. The characterization of the interactions between redox balance, skeletal health and the microbiome, will allow the development of a multisystem biological approach to refine nutritional interventions to improve dairy cattle health, welfare and productive longevity.

Teat dipping is widely used in dairy cattle, especially to protect against contagious mastitis. Here we determine the effect of the device called ‘Electrical Teat Dipping’ (ETD), which was developed by combining teat dipping application and electrical field stimulation technique on teats. For this purpose, the front teats of 100 Holstein breed milking cows were evaluated in two groups, with ETD being applied once to the left front teat of these cows, and conventional teat dipping (CTD) being applied once to the right front teat, both after milking. Ultrasonographic measurements of the teats were made before milking and after teat dipping. We found that the width of the teat canal (1.88 ± 0.07 mm) in the teat using ETD was narrower after the application compared to those with CTD (2.28 ± 0.05 mm). Based on our findings, we conclude that the effects of ETD on the teat are very positive and can potentially be used as a new approach in the preventative control of mastitis in cows.

The study reported in this Research Communication was conducted to investigate the molecular characterisation of buffalo SCAP gene, expression analysis, and the association between single nucleotide polymorphisms and milk production traits in 384 buffaloes. Sequence analysis revealed the SCAP gene had an open reading frame of 3837 bp encoding 1279 amino acids. A ubiquitous expression profile of SCAP gene was detected in various tissues with extreme predominance in the mammary gland during early lactation. Moreover, eleven SNPs in buffalo SCAP gene were identified, six of them (g.1717600A>G, g.1757922C>T, g.1758953G>A, g.1759142C>T, g.1760740G>A, and g.1766036T>C) were found to be significantly associated with 305-day milk yield. Thus, buffalo SCAP could sever as a candidate gene affecting milk production traits in buffalo and the identified SNPs might potentially be genetic markers.

CXCL8 (also known as IL-8) is a member of the CXC subfamily of chemokines that binds two of the seven transmembrane G-protein-coupled receptors (GPCRs), CXCR1 and CXCR2, to mediate and regulate leucocyte accumulation and activation at sites of inflammation. They are known to play a critical role in both disease susceptibility and infection outcome. The aim of this study was to investigate the entire sequences of CXCL8 and CXCR2 genes in thirty-one Simmental sires to evaluate the effects of genomic variants on the indexes of the bulls for milk, fat and protein yields, and for somatic cell score (SCS). Five new single nucleotide polymorphisms (SNPs) were found in CXCR2 gene. The analysis of association indicated that one SNP in CXCL8 and two in CXCR2 influenced the considered traits. To evaluate the existence of functional haplotypic effects, combinations among the three genomic variants (SNP 1 in CXCL8, SNP 6 and SNP 7 in CXCR2) were investigated. Four different haplotypic alleles were identified in the experimental population, one of which at a high frequency (61%). Bulls with Hap 4 (G-C-G at SNP 1, SNP 6, and SNP 7 respectively) had more favourable indexes for SCS (P < 0.05). These results suggest that the SNPs in CXCL8 and CXCR2 may be potential genetic markers to improve udder health in the Simmental breed.

Sensors are increasingly being used to monitor animal behaviour. Data handling methods have, however, lagged behind the continuous data stream to some extent, often being limited to summarizing data into daily averages at group level. This research reflection presents our opinion of the neglected application of 24-h pattern analysis. Recent studies of dairy cow behaviour have demonstrated that additional ways of analysing data improve our understanding of animal behaviour and add value to data that were already retrieved. The terminology for the described 24-h patterns differs between these studies, making them difficult to compare. Thus, diurnal, circadian, daily, periodicity and 24-h pattern are all terms used to describe dairy cow activities over a 24-h period. Several studies have shown that the 24-h behavioural pattern at herd level is relatively consistent over time, and that with well-established management routines, a specific herd signature will be evident. However, within a herd, individual cows may have individual 24-h patterns with more or less variability. Recent studies suggest that deviations from herd and/or individual 24-h patterns can be used to describe cow robustness, as well as to predict disease. We strongly believe that individual and herd 24-h patterns provide a great deal of information about behaviour and that these patterns offer opportunity for more precise and timely health management and welfare monitoring.

One of the major challenges in lameness management is prompt detection, especially before visible gait disturbance. This scoping review describes the potential biomarkers for lameness in dairy cows reported in the literature, their relevance in lameness diagnosis, identifying cows at risk of developing claw lesions and monitoring recovery after treatment. Using specific keywords, a comprehensive literature search was performed in three databases: PubMed, Google Scholar and ScienceDirect to retrieve relevant articles published between 2010 and 2022. A total of 31 articles fulfilling the inclusion criteria were analysed. The categories of potential markers for lameness reported in the literature included acute phase proteins (APPs), nociceptive neuropeptides, stress hormones, proteomes, inflammatory cytokines and metabolites in serum, urine and milk. Cortisol, APPs (serum amyloid A and haptoglobin) and serum, urinary and milk metabolites were the most studied biomarkers for lameness in dairy cows. While APPs, nociceptive neuropeptides and blood cortisol analyses assisted in elucidating the pain and stress experienced by lame cows during diagnosis and after treatment, evidence-based data are lacking to support their use in identifying susceptible animals. Meanwhile, metabolomic techniques revealed promising results in assessing metabolic alterations occurring before, during and after lameness onset. Several metabolites in serum, urinary and milk were reported that could be used to identify susceptible cows even before the onset of clinical signs. Nevertheless, further research is required employing metabolomic techniques to advance our knowledge of claw horn lesions and the discovery of novel biomarkers for identifying susceptible cows. The applicability of these biomarkers is challenging, particularly in the field, as they often require invasive procedures.

The goal of the present study was to determine the best quarter milk flow for teat cup detachment depending on lactational stage and milking interval to optimize the milking process in automatic milking systems (AMS). Milking characteristics and post-milking teat condition were recorded in an AMS with all actions from teat cleaning to post-milking teat dipping occurring in the same teat cup and liner (GEA DairyRobot R9500). In 24 dairy cows, 12 in early (<80 DIM) and 12 in late lactation (>180 DIM), 294 milkings were recorded during 12 consecutive days. Teat cup detachment was set at a quarter milk flow of 50, 87.5 or 125 g/min. Voluntary milking intervals varied similarly in cows in early (6.1–14.8 h) and late lactation (6.3–15.7 h). Total milk yield, milk production per h and average milk flow were higher in early than in late lactation cows. Total milk yield per milking did not differ between detachment levels. The mean milking time was reduced by up to 1.5 min at the highest compared to the lowest detachment level mainly in early lactation cows (P < 0.05). However, no significant effect of the detachment level on milking time was observed at milking intervals >10 h. Average milk flow was higher at milking intervals >10 h than ≤10 h (P < 0.05). In the early lactation cows (only) the average milk flow increased with higher detachment levels (P < 0.05). Teat condition did not differ among detachment settings or milking intervals. In conclusion, teat cup detachment up to 125 g/min reduces milking time in both early and late lactation without a loss of milk yield or affecting the teat condition. Combined with a higher average milk flow through avoiding too short milking intervals, the total milking time and hence stall occupancy can be optimized by early teat cup detachment.

The main objective of this experiment was to evaluate the effects of two milking intervals (8 and 16 h) on milk constituents (fat, protein, lactose, dry matter, and log10 SCC) of nineteen Istrian × Awassi × East-Friesian crossbred ewes in different milk fractions (0–25, 25–50, 50–75 and 75–100%) during the course of milking and in machine stripping (MS) milk. Furthermore, we sought to determine the effect of the two milking intervals on milking characteristics (average milk flow rate, peak milk flow rate, machine-on time, total milk yield, and milk production rate) and whether each milk constituent within each milking interval is best described by a linear, quadratic, or cubic function. Average milk flow rate and milk yield per milking decreased in the 8 h milking interval compared to the 16 h milking interval (P < 0.05). Peak milk flow rate, machine-on time, and milk production rate were not different between the two milking intervals. Overall, milk fat content, dry matter content, and log10 SCC increased in the 8 h milking interval compared to the 16 h milking interval (P < 0.05). Milk protein content did not change through the main milk fractions at either milking interval. Milk lactose content did not change through the milk fractions at the 8 h milking interval, whereas it decreased in the 75–100% and stripping milk fractions at the 16 h milking interval (P < 0.05). The 0–25% and stripping milk fractions contained the highest log10 SCC compared to all other milk fractions (P < 0.05). Changes of milk fat and dry matter content throughout milking were best described by quadratic functions, whereas milk protein content, milk lactose content, and log10 SCC were best described by different functions depending on the milking interval. These results demonstrate that milking interval influenced all milk constituents in various milk fractions during the course of ewe milking. Moreover, milking characteristics such as average milk flow and total milk yield, and the appropriate mathematical function to characterize milk constituents throughout a milking, were affected by milking interval.

We investigated the effect of different types of mother-young contacts (fenceline, restricted and no contact) on the health, growth performance, behaviour and physiological state of Murrah buffalo calves. A total of 24 calves were allocated to three groups of 8: NCM (no calf–mother contact), RCM (restricted calf–mother contact) and FCM (fenceline calf–mother contact). At three months of age, the FCM calves had a higher average body weight (58.6 ± 1.5 kg) than the RCM (52.8 ± 1.3 kg) and NCM (53.6 ± 1.3 kg) calves (P < 0.05). The average daily gain (ADG) at three months of age was greater in FCM (0.6 ± 0.1 kg/d) than RCM (0.5 ± 0.1 kg/d) and NCM (0.5 ± 0.1 kg/d) calves (P < 0.05). The mean immunoglobulin G (IgG) concentrations were significantly greater on d 7, 28, 42 and d 56 of sampling (P < 0.05 for all comparisons) in FCM than RCM and NCM calves. The mean cortisol levels were highest in the NCM calves followed by RCM and the lowest values were observed in FCM calves on d 0, d 7, d 28, d 42 and d 56, respectively (P < 0.05, for all recorded days). The diarrhoea score and eggs per gram of faeces (EPG) were significantly greater in the NCM group compared to the RCM and FCM groups. The time spent in cross sucking, licking inanimate objects and self-licking was greatest (P < 0.05) in NCM calves, followed by RCM calves, and was almost non-existent in the FCM group on all the recorded weeks. Time spent in backward ear position was greater (P < 0.05) in FCM than RCM and NCM calves. The average time spent in forward ear position was greatest (P < 0.05) in the RCM calves followed by NCM with the lowest time observed in FCM calves. The mean duration of time spent by calves in the asymmetrical and axial ear position differed significantly (P < 0.05) among NCM, RCM and FCM calves, P < 0.05. The full and fenceline buffalo calf–mother contact system showed higher growth rates, lower levels of stress responses, oral stereotypies and eggs per gram of faeces. In addition, ear postures may be used as a dependable, and reliable measure of positive, low arousal emotional states.

The destruction of vitamin C in milk under the action of light was studied in different containers. Brown glass bottles showed little destruction, green glass bottles considerably more, and wax impregnated cartons still more. Nevertheless, in the latter, twice as much vitamin C survived as in clear glass bottles.
