Four restriction enzymes (AluI, HhaI, MspI and RsaI), either individually or in combination, were used in terminal restriction fragment length polymorphism (T-RFLP) analysis to: (i) characterize the chicken intestinal bacterial community; and (ii) tentatively identify intestinal bacterial groups related with increased performance parameters in broiler chickens. Balanced commercial diets free of any feed antibiotics were offered to broilers assigned randomly to one of the five dietary treatments: control (C) (commercial diet with no additive), inulin (I), fructose caramel, propyl propane thiosulphonate (PTS-O)-45 and PTS-O-90. Except for the inulin-supplemented diet, multivariate statistical analysis of T-RFLP profiles based on individual enzymes or their combinations showed that the caecal bacterial community composition was significantly different among diets, particularly between the control and the supplemented diets. Individual RsaI and the combination AluI + RsaI proved to be the most useful to discriminate between dietary treatments. Clostridiaceae 1, Lachnospiraceae, Ruminococcaceae and Micrococcaceae were tentatively identified as those families most likely to be implicated in defining the caecal microbiota composition of growing broiler chickens, and also as those most closely related with differences in productive parameters.

Overfeeding minerals to dairy herds will raise diet cost, increase their excretion into the environment and for minerals such as copper (Cu) can lead to poisoning and cow death. In contrast, underfeeding may compromise cow performance, health and fertility. Despite this, the level of mineral intake on commercial dairy units is poorly documented. To determine the mineral intake on commercial dairy herds in central and northern England over the winter of 2011/12 and compare these to recommended levels, samples of compound feed, forage mix, supplementary sources (including lick blocks, rumen boluses, free access minerals and drenches) and drinking water were collected from 50 herds over the winter feeding period and analysed for 10 macro and trace minerals. For cows in early lactation the mean dietary concentration of phosphorus (P) was 4·5 g/kg dry matter (DM) (s.d. 0·70), 0·1 g/kg DM below UK requirements, and for calcium (Ca) was 10·2 g/kg DM (s.d. 2·94), 5·9 g/kg DM above requirements. Trace mineral concentrations were also in excess of requirements in early lactation, with a mean dietary Cu concentration of 28 mg/kg DM (s.d. 9·85), approximately 18 mg/kg DM above UK requirements, with 32 of the 50 herds feeding above the UK industry recommended maximum of 20 mg/kg DM and 6 above the EU limit of 40 mg/kg DM. Dietary mineral concentrations were generally lower in late lactation but still higher than requirements. The forage mix (including supplementary feeds and minerals) contributed the greatest amount of minerals, with percentile ranges (10th–90th) of 2·1–4·4 g/kg diet DM for P, 1·4–3·2 g/kg diet DM for magnesium (Mg) and 5·3–25·0 mg/kg diet DM for Cu. Compounds fed in the milking parlour supplied (10th–90th percentile) 0·0–1·4 g P g/kg diet DM, 0·0–1·2 g Mg/kg diet DM and 0·0–11·6 mg Cu/kg diet DM. For the upper 90th percentile of dairy herds, water supplied proportionally 0·08 of Ca requirements recommended in early lactation in the UK, whilst supplementary mineral sources supplied up to 0·64 of Cu and 0·43 of zinc (Zn) requirements. High dietary concentrations of Cu were not justified by the presence of the dietary antagonist molybdenum (Mo), with no relationship between the two minerals in early or late lactation diets. In conclusion, most dairy herds were feeding excess amounts of minerals over the winter feeding period when compared to UK or other national recommended guidelines, with the implications of a higher diet cost and negative impact on the environment and animal health.

Genetic variation in feed efficiency may have a significant impact on sheep production in integrated crop livestock farming systems in dry areas, where the shortage and poor quality of feed is widespread. Thus, the present study was carried out to investigate the effects of sheep genotype and feed source on liveweight gain, feed conversion efficiency and dry matter (DM) intake in feedlot lambs finished on diets based on low-cost forages or a high-cost concentrated feed as a means of assessing the efficiency of this feeding system. Early weaned lambs of the purebred fat-tailed Akkaraman breed were compared with synthetic Anatolian Merino (0·80 German Mutton Merino×0·20 Native Akkaraman) breed. The lambs were kept in individual pens for 8 weeks and fed four diets: daily harvested forages of triticale (T), Hungarian vetch (HV), a triticale-Hungarian vetch mixture (T+HV), and a concentrate-based feed (CF). Lamb liveweight gain (LWG) was monitored during the early (18 April–16 May) and late (17 May–13 June) spring periods. Diet×period and diet×breed interactions were detected in LWG of the lambs. Lambs from both genotypes on the concentrate-based diet had higher liveweight gains, DM intakes and better feed conversion ratios compared with lambs finished on the forage-based diets. The LWG of lambs offered triticale forage decreased from 177 g/head/day in the early spring to 95 g/head/day in the late spring period, as plant maturity increased. Liveweight gains did not change for the other forage rations during the same period. The LWG of Akkaraman lambs were similar for both the early (189 g/head/day) and the late (183 g/head/day) spring periods, whereas Anatolian Merino lambs gained 41 g/head/day less LW and had 3·8 higher feed conversion rate for the late spring period compared with the early spring period. The present study showed that fat-tailed Akkaraman lambs were better able to utilize forages with low nutritive value compared to Anatolian Merino lambs, and may be better suited to semi-arid areas, where crop and livestock are highly integrated in the farming system.

Four Holstein steers (297 ± 56 kg of body weight (BW)) fitted with duodenal cannula and rumen catheter, were housed in metabolism cages and used in a 4 × 4 Latin Square trial to evaluate the effect of both protein source and Acacia mearnsii tannin extract on digestibility, ruminal fermentation, rumen microbial protein synthesis, N utilization and on duodenal flow of individual amino acids. The diet was offered at restricted amount of 25 g of dry matter (DM)/kg BW and consisted of maize silage plus concentrate, in a proportion of 0·7:0·3 (DM basis) respectively. Concentrate was formulated with either soybean meal or canola meal as protein source, with or without 50 g/kg of A. mearnsii tannin extract (i.e. 15 g/kg of total dietary DM). There was no effect of protein source on most variables. The apparent and true organic matter (OM) digestibilities, as well as neutral detergent fibre (NDF) digestibility were negatively affected by tannin extract inclusion without, however, affecting digestible OM intake. The amount of nitrogen (N) excreted in faeces increased whereas the urinary N excretion decreased in tannin extract treatments. No interaction time × treatment was detected for any rumen variable and no treatment effect was observed for rumen fluid pH and reducing sugars concentration. Rumen fluid concentration of ammonia N was lower for the canola meal plus tannin extract treatment. Rumen concentration of α-amino compounds was not affected by tannin extract but was higher when canola meal was the protein source. The duodenal flow of OM, total N, α-amino N and non-ammonia non-microbial N increased with tannin extract inclusion, whereas the duodenal flow of microbial N was not affected by treatment. For both protein sources, the amount of most individual amino acids flowing to the duodenum increased due to tannin extract addition. In conclusion, the dietary inclusion of 15 g/kg DM of tannin extract from A. mearnsii improved the amino acid supply independently of whether the protein source was canola meal or soybean meal, without affecting the amino acid profile, to steers fed maize silage plus concentrate, with a minor but significant impact on OM digestibility.

Two experiments were carried out to evaluate different dosing and sampling regimes for estimating the dry matter intake (DMI) of vegetation by grazing beef cattle. Experiment 1 compared the DMI of a perennial ryegrass pasture by 48 grazing beef cattle dosed with C32n-alkane and faecally sampled at different time intervals. The cattle, of dairy and suckler origin, were balanced by origin and allocated to one of three alkane-dosing treatments (n=16): T1, dosed once daily (10.30 h) with two 500 mg boluses of C32 alkanes; T2, dosed twice daily (09.00 and 16.00 h) with a 500 mg bolus of C32n-alkane; and T3, dosed twice daily (07.00 and 19.00 h) with a 500 mg bolus of C32n-alkane. Faecal samples were collected concurrently with dosing in each treatment. Dosing interval had no effect on estimated DMI, when calculated using the n-alkane content of faeces sampled concurrently with dosing, suggesting that once-daily dosing with concurrent faecal sampling is adequate to estimate DMI by beef cattle. The objective of Expt 2 was to compare the accuracy of herbage DMI estimated using a once-daily C32 alkane dosing regime and n-alkane concentrations in faeces sampled at 09.00, 13.00, 17.00 and 21.00 h, against DMI measured directly. Twelve Holstein-Friesian bulls (mean body weight 265 kg) were housed individually in digestibility stalls and offered harvested perennial ryegrass or harvested semi-natural upland vegetation, both ad libitum. The DMI estimated by once-daily dosing with n-alkane and concurrent per rectum faecal sampling did not differ significantly from the directly measured DMI for either herbage type. It is concluded that a once-daily alkane dosing and concurrent faecal sampling protocol is adequate to reliably estimate the DMI of both upland and lowland pasture vegetation by beef cattle.

The objective of the present study was to investigate the effect of dietary neutral detergent fibre (NDF) : crude protein (CP) ratio on duodenal microbial crude protein (MCP) flow and nitrogen (N) losses. The study was completed in a 5 × 5 Latin square design with five lactating Holstein dairy cows and 5 high-concentrate total mixed rations (TMR) with different forage combinations, typical for Northern China. The rations with a fixed forage-to-concentrate ratio (39 : 61) resulted in different dietary NDF : CP ratios: TMR1 3·03 : 1 (428·2 g NDF/kg : 141·4 g CP/kg); TMR2 2·74 : 1 (392·7 g NDF/kg : 143·2 g CP/kg); TMR3 2·55 : 1 (368·3 g NDF/kg : 144·4 g CP/kg); TMR4 1·84 : 1 (304·8 g NDF/kg : 165·8 g CP/kg); TMR5 1·60 : 1 (285·0 g NDF/kg : 178·0 g CP/kg). Rumen content, milk, blood, urine and faeces were sampled on the last 3 days of five 18-day periods. Purine derivatives in the urine samples were determined to estimate rumen MCP flow into the small intestine. Milk yield and milk protein yield increased linearly with decreasing dietary NDF : CP ratio although slight differences in dry matter intake were observed due to feed intake restriction. Diurnal ammonia N in the rumen and duodenal MCP flow increased linearly, but blood urea N, urinary N and faecal N linearly decreased with decreasing dietary NDF : CP ratio. The enhanced N utilization in the maize-silage-based TMRs (TMR4–5) in comparison with maize-stover-based TMRs (TMR1–3) increased milk yield and the synthesis of milk protein instead of milk fat in the lactating cows, probably due to high transfer of ammonia N into rumen MCP with a considerable increase of dietary non-fibre carbohydrate content and the decrease of NDF : CP ratio. The present results indicate that not only increasing dietary non-structural carbohydrate content but also adjusting the ratio of structural carbohydrate to CP ratio are important diet formulation strategies for mitigating N losses in lactating cows.

The effect on performance of adding calcium (Ca)-butyrate and Oleobiotec (a flavouring agent) additives to dairy calf starter diet was investigated. Thirty-two newborn Holstein calves (16 males and 16 females) were used in a completely randomized design with four treatments as: (1) diet with no additives, (2) diet containing Ca-butyrate, (3) diet containing Oleobiotec and (4) diet containing Ca-butyrate plus Oleobiotec. Milk of calves was supplemented with additives in the first 20 days of the study and then additives were top-dressed into the starter. Calcium-butyrate had no effect on starter and alfalfa intake during pre-weaning, post-weaning and over the whole trial period but it significantly increased the average daily gain and feed efficiency of calves during post-weaning and the whole trial period. Adding Oleobiotec did not change calf performance significantly. Furthermore, no synergistic effect of experimental additives on calf performance was observed.

The use of diets with increasing proportions of concentrate to fibre can ensure appropriate energy levels and result in greater efficiency in Nellore feedlot steers. It was hypothesized that higher proportions of concentrate in the diet of these Nellore steers may affect ruminal fermentation and microbiota as a consequence of ruminal pH reduction. The present study was conducted to evaluate the effect of diets with four different roughage (hay Tifton 85) : concentrate ratios on intake, digestibility, rumen fermentation and rumen microbiota of Nellore feedlot steers. Higher proportions of concentrate in the diet did not affect intake and digestibility of dry and organic matter. The concentration of N-NH3, total rumen volatile fatty acid, acetic (C2), butyric (C4), isobutyric, valeric and isovaleric acids, and microbial nitrogen did not differ among diets. However, increasing proportions of concentrate in the diet resulted in a linear reduction in average rumen pH and increased propionic acid (C3) concentration, resulting in lower relative C2 : C3. Bacterial population of Fibrobacter succinogenes, Ruminococus flavefaciens and Ruminococcus albus decreased in the rumen. However, bacteria that are consumers of lactic acid (Selenomonas ruminantium and Megasphaera elsdenii) and producers of lactic acid (Lactobacillus sp. and Streptococcus bovis) increased when animals were fed with high-concentrate diets. The total number of protozoa was similar for the different roughage : concentrate ratios. Protozoan counts were only influenced by diet for the genus Dasytricha. The findings point to diets with increasing concentrate to Tifton 85 hay ratios as inhibiting the growth of some cellulolytic bacteria and reducing fibre digestibility, and indicate Tifton 85 hay as a possible modulated rumen fermentation in the Nellore steer feedlot.

Focusing on cattle (Bos taurus, Bos indicus), the present paper discusses the concept of the breed in the context of the dynamic history of livestock domestication. A breed categorization is proposed on the basis of how the breeds came into existence. The online Appendix (available at http://journals.cambridge.org/AGS) presents a survey of the cattle breeds of the world consisting of: (1) a list of breeds per country and then subdivided according to the proposed categories; (2) a list of breed names, including synonyms and translations, ordered according to a comprehensive breed classification; and (3) an alphabetical list of these names. The commonly accepted perceptions of breeds and how these are influenced by the nomenclature are described. Considering the history of breeds, it is argued that conservation of genetic diversity does not necessarily imply conservation of breeds. However, breeds are instrumental for the conservation of genetic diversity as independent genetic management units. These considerations may very well be extrapolated to other domestic species.

Moulds belonging to Penicillium section roqueforti are common contaminants of feedstuffs and produce several mycotoxins that can cause health hazards when ingested by farm animals. Among these, PR toxin (PR), mycophenolic acid (MY) and roquefortine C (RC) have been frequently detected in forages, particularly silages. The aims of the current trials were to study the effects of the presence of pure mycotoxins on in vitro rumen fermentation parameters and to assess their stability in the rumen environment. Two successive in vitro gas production experiments were carried out: a central composite design with four replications of central point (CCD) and a completely randomized design with a fully factorial arrangement of treatments (FFD). In CCD, the effects of PR, MY and RC concentrations in diluted rumen fluid (i.e. 0·01, 0·30, 1·01, 1·71 and 2·00 μg of each mycotoxin/ml) were tested. Gas volume produced after 48 h of incubation (Vf) decreased linearly as concentrations of RC and MY in diluted rumen fluid increased, with marginal effects similar for two mycotoxins, being respectively −14·6 and −13·4 ml/g organic matter (OM) for each 1·0 μg/ml of increment in mycotoxin concentration. Similarly, total volatile fatty acid (VFA) production decreased quadratically as concentrations of RC and MY increased, with marginal effects about two times higher for MY than RC, being −4·22 and −2·62 mmol/l for each 1·0 μg/ml of increment in mycotoxin concentration. With respect to maximum Vf (i.e. 410·6 ml/g OM) and VFA (98·06 mmol/l) values estimated by the model, decreases of 13·6 and 15·2% were obtained when incubating the highest RC and MY concentrations, respectively. The PR did not interfere with rumen fermentation pattern and it was not recovered after 48 h of incubation, whereas the stabilities of MY and RC in rumen fluid were similar and on average equal to about 50%. On the basis of CCD results, a second experiment (FFD) was carried out in which only effects of MY and RC concentrations (i.e. 0, 0·67, 1·33 and 2·00 μg of each mycotoxin/ml of diluted rumen fluid) were tested. Data from FFD showed Vf decreased linearly when concentrations of MY and RC increased, with marginal effect two-folds higher for MY than for RC (−11·1 ml/g OM and −6·7 ml/g OM, respectively). Similar marginal effects of MY and RC in decreasing VFA production were recorded: −2·38 and −2·86 mmol/l for each 1·0 μg/ml of increment in mycotoxin concentration, respectively. At the highest RC and MY tested concentrations, Vf and VFA decreased by 8·7 and 10·7%, respectively, over maximum estimated values. In FFD, the average amounts of MY and RC recovered in rumen fluid after 48 h of incubation were 79·0 and 40·6%, respectively. In conclusion, the MY and RC from standards interfered with rumen microorganisms at relatively low levels and were partially stable in the rumen environment after 48 h of incubation. These findings suggested that MY and RC could interfere with digestive processes and might represent a potential risk for ruminants fed diets containing feeds contaminated by mycotoxins produced by P. roqueforti.

Twenty four French Alpine goats (39 ± 2·0 kg) were individually housed in a completely randomized design and fed a basal diet containing 146 g crude protein and 356 g neutral detergent fibre (NDF)/kg in the absence (control – CTRL) or presence (CELL) of 2 ml of cellulase/kg dry matter intake (DMI) for 70 days, which included a 10-day adaptation period. The feed was offered three times daily at 07·00, 13·00 and 19·00 h, but the single daily dose of cellulase was only fed at 07·00 h. Goats were hand-milked daily; milk production recorded and samples taken for compositional analysis. During the last 5 days of the experimental period, goats from each group were individually housed in stainless steel metabolic cages to enable separate and total collection of faeces and urine for nutrient digestibility and ruminal fermentation determinations. Goats fed CELL had greater DMI and greater digestibility of dry matter (DM), organic matter and NDF than CTRL goats. CELL goats had greater ruminal pH, concentration of acetic acid and concentration of propionic acid than CTRL goats. However, the concentration of ruminal butyric was lower in CELL goats compared with CTRL goats. CELL goats had greater milk yield, energy corrected milk, milk energy content, milk energy output and milk density than CTRL goats and the milk content for total solids, fat, protein and lactose were also greater for CELL goats than for the CTRL goats. The milk of CELL goats had greater palmitoleic acid, cis-10-heptadecanoic acid content and mono-saturated acids than the milk of CTRL goats and lower linoleic acid, linolenic acid contents and saturated fatty acids than the milk of CTRL goats. These results suggest that addition of 2 ml cellulase/kg DM of feed in the diet of lactating French Alpine goats elevated their milk production and improved its composition probably due to improved feed utilization.

Lignification-associated phenolic acids are widely distributed in graminaceous plant cell walls. Nylon bags containing maize bran, wheat bran, millet husk and rice husk were incubated in the rumens of five Charolais (♂) × Nanyang (♀) crossbred steers for 6, 12, 24, 36, 48 and 72 h. The in situ ruminal disappearance of ester-linked phenolic acids linearly increased in the brans with increasing incubation time, and the disappearance was greater for ester-linked ferulic acid (FAest) than for ester-linked p-coumaric acid (PCAest). The disappearances of FAest and PCAest were positively correlated with disappearances of neutral detergent fibre (NDF), cellulose and hemicellulose. The effective degradabilities of NDF, cellulose and hemicellulose in the brans were markedly greater than the effective degradabilities of these components in the husks, and were negatively correlated with the contents of Lignin (sa), ether-linked ferulic acid, PCAest and ether-linked p-coumaric acid in both the cereal brans and husks. These findings suggested that breeding forage crops with modified phenolic acid contents could represent an alternative strategy to promote further increases in fibre digestibility of cereal residue feeds for ruminant animals.

Sodium sulphite is used in an optional way to remove insoluble proteins from neutral detergent fibre (NDF) residue. To determine whether the recovery of both NDF and insoluble nitrogen (N) in NDF solution (NDIN) are altered by its use, both parameters were measured in a set of 12 feeds, including cereal grains: maize (MG), rye (RG) and wheat (WG); cereal co-products: maize gluten feed (MGF), distilled dried grains from barley (DDGB) and wheat (DDGW) and wheat bran (WB); protein concentrates: rapeseed meal (RSM) and expeller palm kernel (EPK); dehydrated sugar beet pulp (DBP) and oat (OH) and ryegrass (RGH) hays. Associated effects on the in situ effective degradability (ED) of both NDF and NDIN were also studied in DDGW, WB, RSM, EPK, DBP, OH and RGH. Also, ED of acid detergent fibre (ADF) and its N (ADIN) were studied in hays. Errors due to microbial contamination in the rumen on the ED of NDF, ADF, NDIN and ADIN were also established in these last seven samples using 15N infusion methods. Three rumen and duodenum cannulated wethers were used in the study. The sulphite use in NDF solution led to reductions (DDGB, DDGW, RSM and OH) and increases (RG, WG, WB and DBP) of the NDIN proportion, as well as the contribution of crude protein to NDF. These variations were associated with irregular effects on NDF residues and on ED of both NDIN and NDF. As a consequence, sulphite use does not assure the reduction of the insoluble protein contamination and it may even increase it. This methodology may also alter the degradability estimates of NDIN or NDF. Mean ruminal microbial contamination in NDF was 7·0, 10·8, 13·3, 5·4, 12·0, 35·3 and 20·0 g/kg in WB, DDGW, RSM, EPK, DBP, OH and RGH, respectively. The associated contents of microbial N in NDIN were: 59·3, 29·9, 26·2, 19·8, 37·3, 441 and 150 g/kg, respectively. Microbial contamination in ADF and ADIN (g/kg) was 3·6 and 94·5 in OH and 1·7 and 41·2 in RGH. Not correcting this contamination led to consistent undervaluations of ED of NDIN and NDF in all tested feeds, although errors only reached significance for NDIN in hays and DBP. Microbial-corrected ED of NDIN was 0·685, 0·826, 0·481, 0·389, 0·166, 0·718 and 0·425 in WB, DDGW, RSM, EPK, DBP, OH and RGH, respectively, whereas values for ADIN were 0·504 (OH) and 0·469 (RGH).

To investigate the extent to which enteric methane (CH4) emissions from growing lambs are explained by simple body weight and diet characteristics, a 2 × 2 Latin square changeover design experiment was carried out using two sheep breeds and two fresh pasture types. Weaned lambs of two contrasting breed types were used: Welsh Mountain (WM, a small, hardy hill breed) and Welsh Mule × Texel (TexX, prime lamb) (n = 8 per breed). The lambs were zero-grazed on material cut from recently reseeded perennial ryegrass and extensively managed permanent pasture. In each experimental period, individual ad libitum dry matter intake (DMI) was determined indoors following an adaptation period of 2 weeks, and CH4 emissions were measured individually in open-circuit respiration chambers over a period of 3 days. Although total daily CH4 emissions were lower for the WM lambs than for the TexX lambs (13·3 v. 15·7 g/day, respectively) when offered fresh forage, the yield of CH4 per unit DMI was similar for the two breed types (16·4 v. 17·7 g CH4/kg DMI). Total output of CH4 per day was higher when lambs were offered ryegrass compared with permanent pasture (16·1 v. 12·9 g/day, respectively), which was probably driven by differences in DMI (986 v. 732 g/day). Methane emissions per unit DMI (16·4 v. 17·7 g CH4/kg DMI) and proportion of gross energy intake excreted as CH4 (0·052 v. 0·056 MJ/MJ) were both higher on the permanent pasture. No forage × breed type interactions were identified. The results indicate that forage type had a greater impact than breed type on CH4 emissions from growing weaned lambs. It can be concluded that when calculating CH4 emissions for inventory purposes, it is more important to know what forages growing lambs are consuming than to know what breeds they are.
