Dairy goat farming is an important sector of the agricultural industry in Greece, with an annual total milk production exceeding 450 000 l and accounting for over 25% of all goat milk produced in the European Union; this milk is used mainly for cheese production. Despite the importance of goat milk for the agricultural sector in Greece, no systematic countrywide investigations in the bulk-tank milk of goats in Greece have been reported. Objectives were to investigate somatic cell counts (SCC) and total bacterial counts (TBC) in raw bulk-tank milk of goat herds in Greece, study factors influencing SCC and TBC therein and evaluate their possible associations with milk content. Throughout Greece, 119 dairy goat herds were visited for milk sampling for somatic cell counting, microbiological examination and composition measurement. Geometric mean SCC and TBC were 0.838 × 106 cells ml−1 and 581 × 103 cfu ml−1, respectively. Multivariable analyses revealed annual frequency of check-ups of milking system and total milk quantity per goat (among 53 variables) to be significant for increased SCC; no factor emerged (among 58 variables) to be significant for increased TBC. Negative correlation of SCC with total protein was found; mean total protein content in the bulk-tank milk in herds with SCC >0.75 × 106 cells ml−1 was 5.1% lower and in herds with SCC >1.5 × 106 cells ml−1, it was 7.8% lower.

Gall-making Cynipidae manipulate the leaves of host plant to form galls where offspring find shelter and food. The relationship between oak gallwasp and biochemical mechanisms of galls still requires a better understanding. So, in this research, protein and phenolic compound contents, as well as the activity of antioxidative enzymes and pathogenesis-related (PR) proteins were determined. Galls caused by asexual generation of Cynips quercusfolii L., Neuroterus numismalis (Fourc.) and N. quercusbaccarum L., as a model were used. All cynipid species modified the protein levels of gall tissues, but they cannot be treated as protein sinks. Significantly higher levels of phenols were observed in the galled leaves and galls of all cynipid species when compared with the control tissues. Peroxidase and polyphenol oxidase activity was usually low or showed no activity in galled tissues of all species. PR proteins, such as chitinase and β-1,3-glucanase, had a similar activity profile. Their activity significantly increased in the leaves with galls of all cynipid species, especially those infested with C. quercusfolii. Data generated in this study clearly indicate that galling Cynipidae manipulate the biochemical machinery of the galls for their own needs. However, the pattern of the biochemical features of leaves with galls and galled tissues depends on gall-making species.

The effects of twelve soil metabolites of trifluralin [2,6-dinitro-N,N-dipropyl-4-(trifluoromethyl)benzenamine] on soybean [Glycine max (L.) Merr.] growth, yield, and seed oil and protein content were determined. The metabolites included oxidative dealkylated, reduced nitro group, benzimidazole, azoxy, azo, oxidized, and hydroxylated derivatives of trifluralin. When applied at 0.22 kg ai/ha and incorporated 7.5 cm deep, no individual metabolite had a significant adverse effect on growth or yield. Three of the metabolites significantly decreased seed oil content and two of the metabolites significantly increased seed protein content. These effects are not considered important in that the differences, while statistically significant, are very small. Also, the amounts of metabolites applied greatly exceeded those expected to be found after many years of continuous use of trifluralin. Potential accumulation of individual metabolites from long-term use of trifluralin does not appear to adversely affect soybean growth, yield, or quality.

Nitrogen-fixing cyanobacteria are vital photosynthetic microorganisms that contribute to soil fertility by fixing atmospheric nitrogen and are also important for maintaining ecosystem stability. These microorganisms can be very sensitive to herbicides because they possess many characteristics of higher plants. Six days after the application of monosulfuron at 0.03 to 0.3 nmol L−1 under laboratory conditions, growth of the nitrogen-fixing cyanobacteria Anabaena flos-aquae, Anabaena azollae, and Anabaena azotica was stimulated, but at higher concentrations (30 to 300 nmol L−1) protein synthesis was inhibited. The production of 16 amino acids in A. flos-aquae was reduced from 7 to 69% with increasing monosulfuron concentration. Application of monosulfuron at 3 to 300 nmol L−1 substantially inhibited in vitro acetolactate synthase (ALS) activity as indicated by 50% inhibition index values of 3.3, 65.2, and 101.3 nmol L−1 for A. flos-aquae, A. azollae, and A. azotica, respectively. In contrast, extractable ALS activity was not affected in these algal species with monosulfuron treatments ranging from 0.03 to 300 nmol L1 except in A. flos-aquae at higher concentrations (30 to 300 nmol L−1). The most sensitive species to monosulfuron was A. flos-aquae, followed by A. azollae and A. azotica. Molecular analyses showed that the genomic DNA of A. azollae and A. azotica differed in only one amino acid. Results from photogenetic analyses revealed a high degree of homology between these algae. In contrast, the genomic DNA of A. flos-aquae differed from that of A. azollae and A. azotica in 44 and 45 amino acids, respectively. Our findings support the view that monosulfuron toxicity in these three nitrogen-fixing cyanobacteria is due to interference with protein metabolism via inhibition of branch-chain amino acid biosynthesis, and particularly ALS activity.

Chickpea (Cicer arietinum L.) is an important food legume crop grown mostly in Asia and Africa as well as in other parts of the world, and serves as an important source of protein in the diets of vegetarian people. Lot of variation in grain quality traits has been observed in cultivated chickpea in both desi and Kabuli types. It has often been observed that seeds with smaller size have more protein when compared with those with larger size. The joint dependence of these two traits on the same primary characteristics results in this negative relationship. This negative correlation coupled with breeding efforts aimed at increasing grain yield has hampered the progress of improving these two traits simultaneously. Therefore, a germplasm exhibiting wider variability for these quantitative traits is an important resource for the identification of novel alleles. Herein, we report the wide variation observed for five important quantitative traits including days to flowering, days to maturity, plant height, 100-seed weight and protein content, and also report the significant positive correlation observed between 100-seed weight and protein content in a diverse collection of chickpea germplasm comprising both desi and Kabuli types.

Annual wild soybean characterized by low 100-seed weight (100SW), high protein content (PRC) and low oil content (OIC) may have favourable exotic genes/alleles for broadening the genetic base of the cultivated soybean. To evaluate the wild alleles/segments, a chromosome segment substitution line population comprising 151 lines with N24852 (wild) as the donor and NN1138-2 (cultivated) as the recurrent parent was analysed using single-marker analysis, interval mapping, inclusive composite interval mapping and mixed linear composite interval mapping. On 14 segments of ten chromosomes, 17 quantitative trait loci (QTL) were identified, with two segments each containing two QTL for 100SW and OIC and one segment containing two QTL for PRC and OIC, respectively. All the seven wild alleles/segments for 100SW were associated with negative effects and three were associated with positive effects, but one was associated with a negative effect for PRC, and five were associated with negative effects, but one was associated with a positive effect for OIC. Except Satt216 and Sat_224 for 100SW, the identified QTL/segments have been reported from cultivated soybean mapping populations. The detected wild segments may provide materials for further characterization, cloning and pyramiding of the alleles conferring the seed-quality traits.

Phenotyping of genetic resources remains the bottleneck in the characterization of genetic resources, since the advent of modern next-generation sequencing technologies has made genotyping much more cost- and time-effective. This article reports on the phenotyping of agriculturally important traits in perennial ryegrass (Lolium perenne). In the present study, water-soluble carbohydrate (WSC), crude protein and dry matter contents were recorded for 1320 individuals, pooled into 132 samples from 33 perennial ryegrass ecotypes and cultivars at five different harvest time points across the 2004 growing season. While, in general, the cultivars had higher WSC contents than the ecotypes, individual ecotypes did show potential to be used in breeding programmes, as they had higher values than all other accessions at particular cutting time points. In correlation analyses, positive relationships were observed between dry matter and glucose contents both early and late in the growing season. Principal components analysis allowed the split either between cultivars and ecotypes or between tetraploid cultivars and the rest of the accessions at four of the five cutting time points. In the analysis of variance, cutting time was the most significant factor influencing the variation in traits.

Food regulation aims to protect public health through a safe and nutritious food supply produced by a compliant food industry. Food standards of developed countries generally do not regulate protein content or protein quality because the risk of dietary protein inadequacy in their national populations is very low. Protein is nevertheless regulated for reasons of product quality or protein labelling or to minimise assessed health risks associated with consumption of certain animal- and vegetable-protein foods; analogue products that extend or simulate commonly available animal-protein foods; and special purpose foods such as infant formula and foods, supplementary and medical foods, and foods for weight loss. The extent and approach to protein regulation varies greatly among jurisdictions but where it occurs, it is applied through minimum and sometimes maximum limits on protein content or quality measures or both using an inter-related approach. Protein quality measures range from amino acid profiles and digestibility corrected scores to protein rating, a rat bioassay and reference proteins not further described. Regulatory methods for protein quality determination are referenced to the published scientific literature or developed nationally. Internationally, the Codex Alimentarius regulates the protein content and quality of some foods. The Codex approach varies according to the food but is similar to the approaches used in national and regional food regulation. This paper provides a comparison of the regulation of protein in foods using examples from the food regulations of Australia New Zealand, Canada, the European Union, the United States of America and the Codex Alimentarius.

Introduction. Chestnut represents one of the most important crops for the Portuguese economy. The most common consumption mode of these fruits is roasted or boiled. In this context, the major aim of our study was to evaluate the amino acid contents and the mineral composition of raw and cooked chestnuts. Materials and methods. Amino acids were determined by HPLC and minerals were determined by molecular absorption spectrophotometry, by atomic absorption spectrophotometry or by flame emission photometry. The most important Portuguese cultivars were evaluated: Aveleira, Boaventura, Côta, Judia, Lada, Lamela, Longal Padrela, Longal Soutos da Lapa, Negra and Martaínha. Results and discussion. The cooking method significantly affected the total amino acid composition with contents in roasted samples 13% and 12 % higher than in boiled and raw chestnuts, respectively. Roasted chestnuts presented higher alanine, arginine, isoleucine, leucine, phenylalanine, threonine, tyrosine and valine contents than either raw or boiled chestnuts. Moreover, serine presented the highest content in raw or roasted chestnut kernels. Regarding the mineral composition, potassium (K) was the predominant macronutrient in the chestnuts, whereas phosphorus (P), calcium (Ca) and magnesium (Mg) were found in low contents. Cooking significantly affected the mineral composition, except for iron (Fe), copper (Cu), and manganese (Mn) contents. In general, calcium decreased upon cooking. K, Ca, Mg and B decreased with boiling. The present data confirm that cooked chestnuts are a good source of amino acids and minerals, both affected by boiling and roasting, which have been associated with positive health benefits.

Introduction. Physalis has been known for a long time in Egypt. Amongunexploited tropical fruits, Physalis is a very promising fruit. Recently, the economicimportance of Physalis has risen, due to its high acceptance for local consumption,achieving great success in the African, Latin American and European markets. One of thechallenges of recent years has been to participate in the development of Physalis as acommercial crop of economic utility. In order to understand the nutraceutical andmedicinal characteristics of Physalis fruits cultivated in Egypt, the biochemicalcomposition of the raw Physalis pubescens juice was determined.Materials and methods. Whole fresh fruits of Physalis pubescensfrom Egypt were preserved at –20 °C for the duration of the experiment. Thejuice was extracted from Physalis fruits by using a fruit pulper then filtered oncheesecloth to separate seeds and skins. Contents of oil, fatty acids, proteins, aminoacids, sugars and minerals of the juice were analyzed, and were compared and discussed inrelation to the biochemical composition of other fruits and vegetable oils.Results. Yield of the juice was high (64%) and it is a rich source ofminerals such as potassium (11.32 g·100 g–1 dm), phosphorus(5.55 g·100 g–1 dm), zinc (0.02 g·100 g–1 dm) and boron(0.01 g·100 g–1 dm), polyphenols (76.6 mg·100 mL–1) andcarotenoids (70 µg·mL–1). It contained good amounts of vitamin C(38.77 mg·100 mL–1) and it could be a good potential source of essentialamino acids such as isoleucine, valine and tryptophan [(4.2, 3.9 and3.9) g·100 g–1 protein], whose amounts were higher than those recommended bythe FAO / WHO / UNU. Additionally, it contains a low amount of oil(1.1 g·100 g–1 dm). Conclusion. Physalis juice should attractgreat interest because of its composition. Its potential nutraceutical quality couldparticipate in the development of Physalis as a commercial crop. Certain molecules such asantimicrobial molecules and bioactive withanolides, which have the famous ethnomedical andmedical effects of Physalis juice, still have to be studied.

Eggs of Melongena melongena develop inside round, flat egg capsules which contain a gelatinous intracapsular fluid. To determine if this gel represents a nutritional source for the developing embryos, we measured the amount of proteins of the embryos throughout their development from the egg to the hatching stage as well as the protein content of the intracapsular liquid at the same stages of development. Egg capsules of M. melongena were collected at Golfete de Cuare, Venezuela between 1–2 m depth. Uncleaved eggs measured 352–480 μm and contained 8–15 μg of protein/egg. This amount of protein was not significantly different at the trochophore, veliger and pediveliger stages, however, it decreased significantly at the hatching stage to 6 μg/hatchling. About 95–98% of the eggs develop to the hatching stage, the remaining 2–5% remain intact in the egg capsule. Hatching takes place as a pediveliger measuring around 720 μm in shell length. The protein concentration of the intracapsular liquid was 0.18 μg/μl at the egg stage and it reached 0.13 μg/μl at the prehatching stage; however, the total amount of protein in the intracapsular fluid was not significantly different throughout the development from one stage to another. Results indicate that embryos of M. melongena use neither the intracapsular liquid as an extraembryonic food source, nor nurse eggs.

The protein content of fresh homogenates and their corresponding TCA precipitated fractions of 10 different species of helminths was estimated by the methods of Lowry et al. and Spector using the Folin phenol reagent and Coomassie brilliant blue G-250 respectively. The former method gives exaggerated values as compared to the latter method. The parasite phenols, phenolic proteins and catecholamines could be responsible for interference in the Lowry's procedure. The TCA non-precipitable moieties also give colour only with the Folin phenol assay. The pronounced intra-specific differences in the total protein content of helminths reflect their metabolic variations and adaptations. Habitat does not appear to influence the protein content of parasites, however, the effect of host variation was evident in the pouched amphistome G. crumenifer. It is concluded that the dye binding method gives more consistent results and it can be conveniently applied to crude tissue homogenates of helminths.

The variation for protein content has been assessed in a series of pea (Pisum sativum L.) lines differing in mean seed size. The range of seed size within each genotype was manipulated by growing the plants in different environments and by altering the structure of the plant. The response of the lines in terms of seed weight to the environments and treatments was inconsistent, but 90% of the total variation was accounted for by differences between genotypes. In contrast, for percentage protein, the genetic component was much weaker with nearly half the variation being accounted for by within (residual) variation. The correlations between seed weight and percentage protein were highly genotype specific. For the largerseeded genotype, percentage protein increased with increasing seed size (r= +0.9, P < 0.01), while for the small-seeded genotype there was mid-range negative value (r = −0.5, P <0.01). It is suggested that any assessment of percentage protein in pea genotypes must take account of seed size.

Introduction. Pour introduire Ricinodendron heudelotii (Euphorbiacée) dans les agrosystèmes, des plantules ayant un système racinaire pivotant peuvent être produites par embryogenèse somatique in vitro. Nous avons étudié l’effet de quelques phytohormones exogènes sur certains paramètres biochimiques caractéristiques de l’embryogenèse somatique de l’espèce. Matériel et méthodes. Des cals et des embryons somatiques ont été induits par culture de fragments de feuilles sur un milieu de base enrichi en (1, 2 ou 3) mg 2,4-D·L–1 ou de combinaisons [2,4-D / kinétine] ou [2,4-D / BAP] impliquant différentes concentrations des hormones. Les extractions et analyses des sucres totaux, phénols, protéines totales et activité des peroxydases ont été faites après (30, 50 et 70) j de développement des cals. Résultats. Après 30 j de culture, les meilleurs pourcentages d’explants formant des cals ont varié de 72,1 % à 100 % selon les phyto-hormones utilisées. Après 50 j de culture, ces cals ont formé des amas globulaires de cellules en division (nodules embryonnaires). Après 70 j de culture, 17,4 % des cals ont différencié les embryons somatiques multiformes en présence de 3 mg 2,4-D·L–1; 23,1 %, en présence de [1 mg 2,4-D·L–1 + 1 mg kinétine·L–1] et 27,6 %, [1 mg 2,4-D·L–1 + 2 mg BAP·L–1]. La teneur en sucres a été la plus élevée à 30 j (formation des cals) et 50 j (formation des nodules embryonnaires sur cals), alors que les teneurs en phénols, en protéines et l’activité des peroxydases ont été plus élevées à 70 j (différenciation des embryons somatiques sur cals). Discussion et conclusion. À l’instar de ce qui se passe chez d’autres espèces végétales, chez R. heudelotii, les phytohormones exogènes interviendraient dans le contrôle du métabolisme des paramètres biochimiques impliqués dans la formation des embryons somatiques et de leur développement.

In the course of the cell cycles of synchronous cultures of the chlorococcal alga Scenedesmus quadricauda, the following were monitored: total protein and RNA accumulation as a measure of growth processes, the timing of the commitment points at which the cells trigger the sequence of reproductive processes (DNA replication, nuclear and cellular division) and the course of the reproductive processes. The synchronous cultures were grown either under various lighting regimes, or in the temporary presence of specific inhibitors of either proteosynthesis (cycloheximide) or DNA replication (5-fluorodeoxyuridine). By adjusting the length of the light period, the cell cycle could be manipulated. Cell cycle patterns could be altered to give different numbers of sequences of reproductive processes. The extent of their mutual overlap could be influenced and the number of daughter cells produced could be altered. Schematic illustrations of various cell cycle patterns and comparisons with those of higher plants and other algal species are presented.

An experiment was carried out to study the effect of changes in either the quality or the quantity of dietary protein intake on the free essential amino acid profiles in plasma, muscle or liver of growing chickens. Following a randomized paired-feeding design based on metabolic body weight (kg M0·75), White Rock male broilers were allocated to one of three isoenergetic (14·5 kJ metabolizable energy per g dry matter (DM)) semisynthetic diets containing different levels of protein (60, 120, 180 or 240 g/kg DM). All diets were based on soya-bean meal, as the sole source of protein, either unsupplemented (diets S) or supplemented with 20 g/kg L-lysine (diets SL) or 2 g/kg DL-methionine (diets SM). Samples of blood, biceps muscle and liver were taken and amino acid analysis was determined by high performance liquid chromatography. Plasma was only adequate to detect the effect of the supplementation with methionine to balance the dietary amino acid (AA) profile. Plasma concentrations of free methionine significantly increased as a result of the supplementation with methionine (P < 0·001) and remained unaffected by the amount of protein ingested. When lysine was added to diets S to induce an AA imbalance, a significant increase in muscle concentrations of free valine, isoleucine and phenylalanine (P <0·05) and a significant decrease in liver concentrations of free arginine and phenylalanine were observed (P<0·05). The supplementation with DL-methionine gave rise to a significant fall in the concentrations of free histidine, glycine and threonine (P < 0·05) and increased those of isoleucine and methionine in the skeletal muscle (P < 0·05), while in liver it significantly lowered the concentrations of free arginine, valine, leucine, phenylalanine and methionine (P < 0·001 to P < 0·05) and raised that of free glycine (P < 0·001). The ratios lysine: phenylalanine, lysine: valine and methionine: glycine in muscle together with that of methionine: glycine in liver seem to be appropriate indexes of the adequacy of the dietary AA profile to requirements.

The efficiency of utilization of protein for retention was analysed in pre-ruminant kid goats of the Granadina breed. Sixty male kids were used. Six were slaughtered at birth and the remaining 54 were offered different protein and fat intakes using nine different milk replacers. The protein concentrations were 200, 240 and 280g/kg dry matter (DM) and those of fat were 200, 240 and 280 g/kg DM. Animals were maintained on experiment until they were 60 days old. All were slaughtered on day 61. Nitrogen (N) balance trials were performed during the last 8 days of the 1st and 2nd months. Body composition of the animals slaughtered at birth and at 61 days were determined. Rates of energy retained as protein and as fat were determined (kJ/kg M0·75 per day) and the corresponding rates of metabolizable energy intake as protein and as fat (kJ/kg M0·75 per day) estimated.

Once the relationships between the rates ofN retained and those of digestible N ingested had been established, it was evident that by increasing the protein content of the diet the efficiency of protein retention was decreased. In contrast, increasing the fat content of the milk replacer increased the efficiency of protein retention. The latter effect was noted for the milk replacers containing the high and medium levels of protein but not for those that contained the lowest level of protein, indicating that the level of protein was then the limiting factor. Having recorded this protein-sparing effect of the fat, the results obtained from the slaughter trials were used to develop generalized equations expressing the rates of energy retention in the form of protein or fat as a function of the rates of metabolizable energy intake achieved as both protein and fat. From the analysis of these equations conclusions are drawn about the variable contribution to protein retention in these animals of energy ingested as fat. This contribution depended on the energy intake achieved both in the form of protein and in the form of fat.

If birds can learn to associate different sensory properties of foods with different metabolic feelings caused by different protein contents, then they should be able to choose a balance of foods which makes them feel most comfortable. Broiler chickens given a choice between two foods, one containing a higher concentration of protein than required and the other a lower concentration, eat amounts of the two which will give a diet that is close to optimum for growth in terms of overall protein content. When both foods have excessive or insufficient protein the birds eat predominantly from that closer to their requirements. As broilers grow they choose a progressively lower protein diet in line with their reducing protein:energy requirements. Strains with higher potential rates of lean deposition voluntarily consume a higher ratio of high-protein to low-protein food compared with low-growth strains, while strains selected for high fat deposition eat less protein per unit of energy than lean birds. Males select higher protein diets than females. Although increases in environmental temperature would be predicted to reduce energy requirements and thus increase the protein:energy ratio in selected diets, heat-stressed birds do not increase the protein content of their diet, probably because of the high heat increment of protein metabolism. Short periods of access to one food alone are followed by a preference for the opposite food when access to both is restored. When given appropriate training, laying hens can balance their diet for protein but not as well as broilers. There is evidence that growing chicks can differentiate high-lysine from low-lysine foods but do not entirely balance their diet for lysine. Methionine-deficient broilers eat more of a methionine supplemented food than normal birds, while laying hens eat some methionine-fortified food when given a choice against a low-methionine food, but not enough to support normal egg production. It is concluded that poultry can show a great degree of ‘nutritional wisdom’ as far as protein is concerned but that they need to be allowed to differentiate between foods with different nutrient profiles by sensory means, and a lack of training might account for some of the poor results obtained.
