Clay particle aggregation affects a number of environmental processes, such as contaminant sorption/desorption, particle movement/deposition, and sediment structure and stability, yet factors that control clay aggregation are not well understood. This study was designed to investigate how microbial reduction of Fe(III) in clay structure, a common process in soils and sediments, affects clay-particle aggregation. Microbial Fe(III) reduction experiments were conducted with Shewanella putrefaciens CN32 in bicarbonate buffer with structural Fe (III) in nontronite as the sole electron acceptor, lactate as the sole electron donor, and AQDS as an electron shuttle. Four size fractions of nontronite (D5–D95 of 0.12–0.22 µm, 0.41–0.69 µm, 0.73–0.96 µm and 1.42–1.78 µm) were used to evaluate size-dependent aggregation kinetics. The extent of Fe(III) bioreduction and the amount of exopolysaccharide (EPS), a major biopolymer secreted by CN32 cells during Fe(III) bioreduction, were measured with chemical methods. Nontronite particle aggregation was determined by photon correlation spectroscopy and scanning electron microscopy. The maximum extent of Fe(III) bioreduction reached 36% and 24% for the smallest and the largest size fractions, respectively. Within the same time duration, the effective diameter, measured at 95% percentile (D95), increased by a factor of 43.7 and 7.7 for these two fractions, respectively. Because there was production of EPS by CN32 cells during Fe(III) reduction, it was difficult to assess the relative role of Fe(III) bioreduction and EPS bridging in particle aggregation. Thus, additional experiments were performed. Reduction of Fe(III) by dithionite was designed to examine the effect of Fe(III) reduction, and pure EPS isolated from CN32 cells was used to examine the effect of EPS. The data showed that both Fe(III) reduction and EPS were important in promoting clay mineral aggregation. In natural environments, the relative importance of these two factors may be dependent on local conditions. These results have important implications for understanding factors in controlling clay particle aggregation in natural environments.

The data on efficacy and tolerance from four (phase II and III) trials of quetiapine (Seroquel® or ICI 204,636) in a total of 1,461 patients with acute exacerbations of chronic or sub-chronic schizophrenia are briefly reviewed. Together, the results of these trials have demonstrated antipsychotic efficacy in comparison with placebo and equivalent efficacy to haloperidol. In spite of its short plasma elimination half-life, quetiapine is equally effective when given in twice-daily and three-time daily doses. The optimum dose is between 300 mg and 400 mg per day, and the main and most consistent adverse effect is sedation. It has a low extrapyramidal side effect liability and does not produce sustained elevations in plasma prolactin levels.

Marinobacter sp. W1-16 from Antarctic surface seawater was analysed for the production of extracellular polymeric substances (EPSs). Enhancement of the EPS biosynthesis was carried out by evaluating the influences of the carbon source (type and concentration), temperature, pH and salinity. EPS yields varied strongly depending on sugar substrate and temperature, while pH and salinity did not strongly affect levels of EPS production. Marinobacter sp. W1-16 produced the highest quantity of EPSs when growing at 15°C and pH 8, in the presence of 2% glucose and 3% NaCl. The EPS chemical characterization revealed a molecular weight of about 260 kDa. Colorimetric assays determined a higher quantity of carbohydrate than of proteins and uronic acids, as well as the presence of sulphate, in the extracted EPSs. The monosaccharidic composition resulted in Glc:Man:Gal:GalN:GalA:GlcA in relative molar proportions of 1:0.9:0.2:0.1:0.1:0.01. Some biotechnological potentialities (i.e. emulsifying and cryoprotective actions, and heavy metal binding properties) of the EPSs were proved, suggesting possible industrial and bioremediation applications.

Background: This is a report of a 66-year-old male with Parkinson's disease (PD), depression and anxiety who developed tardive dyskinesia (TD) while on fluoxetine.

Methods: The patient underwent psychiatric, neurological and neuroimaging examination.

Results: The patient's neuroimaging examination was normal, his psychiatric assessment revealed depression and anxiety, and his neurological evaluation diagnosed only mild PD. The patient's TD resolved when fluoxetine was discontinued and recurred upon re-exposure.

Conclusion: This case shows that fluoxetine as monotherapy can be associated with TD especially in patients with concomitant PD. Clinicians must be aware of this side-effect and monitor for features of TD due to antidepressants that are often used to treat comorbid depression in patients with PD.

This study highlights the role of specific outer bacterial structures, such as the glycocalix, in calcium carbonate crystallization in vitro. We describe the formation of calcite crystals by extracellular polymeric materials, such as exopolysaccharides (EPS) and capsular polysaccharides (CPS) isolated from Bacillus firmus and Nocardia calcarea. Organic matrices were isolated from calcifying bacteria grown on synthetic medium—in the presence or absence of calcium ions—and their effect on calcite precipitation was assessed. Scanning electron microscopy observations and energy dispersive X-ray spectrometry analysis showed that CPS and EPS fractions were involved in calcium carbonate precipitation, not only serving as nucleation sites but also through a direct role in crystal formation. The utilization of different synthetic media, with and without addition of calcium ions, influenced the biofilm production and protein profile of extracellular polymeric materials. Proteins of CPS fractions with a molecular mass between 25 and 70 kDa were overexpressed when calcium ions were present in the medium. This higher level of protein synthesis could be related to the active process of bioprecipitation.

La dernière décennie a vu apparaître de nombreuses solutions technologiques destinées àaméliorer le confort des automobilistes. L’une d’entre elles, la direction assistéeélectrique (DAE), a considérablement évolué : de plus en plus de véhicules en sontéquipés. L’assistance, délivrée par l’intermédiaire d’un moteur électrique et d’unréducteur roue et vis sans fin, est contrôlée par un calculateur. Cette solution offre despossibilités plus intéressantes en terme de consommation d’énergie que les systèmeshydrauliques encore utilisés sur certains véhicules. La compréhension et lacaractérisation du comportement dynamique des DAE sont devenues des préoccupationsmajeures. En effet, afin de répondre à des exigences vibratoires et acoustiques toujoursplus sévères, le comportement sonore et vibratoire de la direction est étudié. Dans cecontexte, le comportement dynamique de l’ensemble de la DAE est simulé. L’accent anotamment été mis sur la modélisation des deux engrenages présents dans le système(roue-vis sans fin et pignon-crémaillère) et leurs systèmes à rattrapage de jeu. Lamodélisation de la ligne poussoir des DAE est présentée, ainsi que sa validation et sonintégration dans un modèle dynamique d’ensemble. Cette étude consiste en une approche à lafois numérique et expérimentale de la problématique étudiée.

Forty seven exopolysaccharide (EPS) producing mesophilic lactic acid bacteria have been isolated from Dahi and raw milk and selected cultures were evaluated for their influence on rheological and sensory properties of fat-free Dahi. Two isolates namely B-6 and KT-24 that showed promising technological attributes were identified as Lc. lactis subsp. lactis strains. B-6 produced 184±2 mg/l EPS in deproteinized whey medium compared with 193±1 mg/l by KT-24. EPS produced by B-6 was a heteropolysaccharide (consisting of glucose and mannose, 1:7·4) with molecular weight of 3·0×104 Da whereas KT-24 EPS was a homopolysaccharide (rhamnose) having molecular weight of 4·5×104 Da. Both EPS producing cultures showed significant changes in rheological and sensory properties of fat-free Dahi. Dahi prepared by these cultures was more viscous, adhesive, sticky, showed lower susceptibility to whey separation, and received higher sensory scores than Dahi prepared with non-EPS producing culture.

Biofilms are assemblages of microorganisms and their associated extracellular products at an interface and typically with an abiotic or biotic surface. The study of the morphology of biofilms is important because they are associated with processes of biofouling, corrosion, catalysis, pollutant transformation, dental caries, drug resistance, and so forth. In the literature, biofilms have been examined by atomic force microscopy (AFM), which has proven to be a potent tool to study different aspects of the biofilm development on solid surfaces. In this work, we used AFM to investigate topographical changes during the development process of Enterococcus faecalis biofilms, which were generated on sterile cellulose nitrate membrane (CNM) filters in brain heart infusion (BHI) broth agar blood plates after 24, 36, 72, 192, and 360 h. AFM height images showed topographical changes due to biofilm development, which were used to characterize several aspects of the bacterial surface, such as the presence of extracellular polymeric substance, and the biofilm development stage. Changes in the development stage of the biofilm were shown to correlate with changes in the surface roughness as quantified through the mean roughness.

In this study, the symbiont cells of several endolithic lichens colonizing granite in continental Antarctica and the relationships they have with the abiotic environment were analyzed in situ, in order to characterize the microecosystems integrating these lichens, from a microecological perspective. Mycobiont and photobiont cells, the majority classified as living by fluorescent vitality testing, were observed distributed through the fissures of the granite. The fact that extracellular polymeric substances were commonly observed close to these cells and the features of these compounds, suggest a certain protective role for these substances against the harsh environmental conditions. Different chemical, physical and biological relationships take place within the endolithic biofilms where the lichens are found, possibly affecting the survival and distribution of these organisms. The alteration of bedrock minerals and synthesis of biominerals in the proximity of these lichens give rise to different chemical microenvironments and suggest their participation in mineral nutrient cycling.

How are microphytobenthic biofilms adapted to the high incident irradiances and temperatures, low inorganic nutrient concentrations and high desiccation stresses on intertidal flats present in tropical environments? This study investigated biofilms subject to different environmental conditions in a range of tropical sites in Suva lagoon, Fiji. PAM fluorescence was used to measure photophysiological responses to the light climate. Biofilm colloidal carbohydrate, extracellular polymeric substances (EPS) and low molecular weight (MW) carbohydrate concentrations and diel carbohydrate production patterns were measured. Average biomass (Chl a) ranged from 15 to 36 mg m−2, and was highest in seagrass bed sediments, but biomass was not correlated with water column or sediment porewater nutrient concentrations. Biofilm photophysiology differed significantly along a combined gradient of light and nutrient availability, with Fv/Fm, relative ETRmax and Ek of biofilms highest in mangrove and intertidal main island sites and lowest in subtidal coral reef flats. Subtidal biofilms showed photoinhibition at irradiances >1000 μmol m−2 s−1. Significant correlations between Chl a and colloidal carbohydrate concentrations were present (except on intertidal sandflats), and tropical biofilms had higher ratios of colloidal carbohydrate and EPS to Chl a than temperate estuarine biofilms, probably due to a combination of high irradiance and low nutrient availability leading to the production of excess photoassimilates. The percentage of EPS present in the colloidal fraction was highest in coral sand biofilms (42%), which had the lowest nutrient concentrations, compared with other sites (25–32%). Intertidal biofilms predominantly consisted of large motile taxa and showed strong rhythms of vertical migration. During tidal emersion, high sediment temperatures (41 °C), irradiance (>2300 μmol m−2 s−1) and salinity (49‰) stimulated downward migration. In silty sediments, migration resulted in a reduction in photosynthetic activity during the midday period but, in sands with high light penetration (to a depth of >1700 μm), high production rates of EPS (18.2 μg carbo. μg Chl a−1 h−1) and low MW carbohydrate exudates (40.2 μg carbo. μg Chl a−1 h−1) occurred. Vertical migration, high Ek and high rates of photoassimilate dumping are all adaptations to living in the tropical intertidal zone. Seagrass and reef flat biofilms consisted of a diverse non-migratory flora of motile and non-motile taxa that were not subject to such extreme temperature and irradiance conditions. Low values of photosynthetic parameters and high colloidal and EPS content indicated that these biofilms were nutrient-limited.

Marine snow is a ubiquitous feature of the ocean and an important agent in the transport of energy and nutrients through marine ecosystems. Diatom aggregates, which form during blooms and, to a lesser extent, by the resuspension of benthic biofilms, are a primary source of marine snow. Genera commonly found in diatom aggregates are: Nitzschia, Chaetoceros, Rhizosolenia, Leptocylindricus, Skeletonema and Thalassionema. Most fieldwork has been restricted to a limited number of locations in the Northern Hemisphere. To quantify the global impact of diatom aggregation there is a need to conduct fieldwork in a wider range of areas, particularly in the Southern Hemisphere. Aggregates form when particles collide and stick together. Collisions in the water column are affected by turbulence, differential settlement and animal feeding, whereas diatom stickiness is affected by extracellular polymeric substances (EPS). Laboratory experiments have demonstrated that diatoms produce more EPS under nutrient limitation, although little is known about how limitation by different nutrients affects the quantity and composition of EPS and subsequent stickiness. EPS form three pools in the environment: cell coatings, soluble EPS and transparent exopolymeric particles (TEP). There is a need to investigate the dynamics of conversion between the pools of EPS by both abiotic and biological processes and how these conversions affect aggregate concentration and structure. Processes governing disaggregation have been largely overlooked, although they are as important as aggregation in determining the dynamics of aggregate concentrations in the water column. The biogeochemical significance of diatom aggregates as a means of transporting carbon and other nutrients from the euphotic zone to the seabed is well established. However, the internal biogeochemistry of aggregates is not well understood. Aggregates contain anaerobic microsites and further work is required to establish whether aggregates are significant sinks for nitrogen in the water column through anaerobic denitrification. Several hypotheses have been proposed to explain diatom aggregation in the field, but many of these are flawed because the mechanisms and adaptive explanations proposed require natural selection to operate at the level of populations rather than genes or individuals.

The excretion of extracellular polymeric substances (EPS) by an axenic culture of the benthic diatom Cylindrotheca closterium was investigated. Two sequential extraction steps proved to be sufficient to remove the bulk of the EPS present. Soluble EPS was recovered by a simple centrifugation step and represented a fraction that was not or was only loosely associated with diatom cells. For the extraction of bound EPS, different procedures were compared. The best results were obtained using distilled water as extraction solvent (1 h, 30 °C). The sugars that were recovered using this procedure were typically associated with aggregates of diatoms. In addition to the distinct differences in localization of the different types of EPS, their temporal dynamics differed in relation to the light–dark cycle. Soluble EPS were continuously released into the medium at a rate of 1·6 pg cell−1 day−1. In contrast, the production of bound EPS was highly light-dependent. In the dark, this bound EPS rapidly disappeared, probably as the result of its utilization by the diatoms.

Drying of cells leads to damage resulting from crowding of cytoplasmic components, condensation of the nucleoid, increases in the Tm of membrane phase transitions, and imposition of stress upon cell walls. Prolonged desiccation leads to oxidation of proteins, DNA and membrane components through metal-dependent Fenton reactions, while Maillard reactions generate cross-linked products between the carbonyl groups of reducing sugars and the primary amines of nucleic acids and proteins. Although such damage restricts many organisms to aqueous environments, some, including many cyanobacteria, can tolerate the air-dried state for prolonged periods. Cyanobacteria in the Tintenstrich communities of exposed rock faces, Microcoleus and Lyngbya spp. in intertidal mats, chasmoendolithic Chroococcidiopsis spp. in the rocks of hot and cold deserts, and terrestrial epilithic crusts of Tolypothrix and Nostoc are examples that show a marked capacity to withstand the removal of their cellular water. For Nostoc commune, the mechanisms of desiccation tolerance reflect both simple and complex interactions at the structural, physiological and molecular levels.

The production and composition of extracellular polymeric substances (EPS) in axenic batch cultures of the benthic marine epipelic diatoms Navicula salinarum and Cylindrotheca closterium were investigated. EPS was secreted into the medium and the bulk was loosely associated with the cells. Neither N. salinarum nor C. closterium formed a well-defined polysaccharide capsule. EPS of both N. salinarum and C. closterium consisted predominantly of polysaccharide but small quantities of protein were present as well. EPS also contained uronic acids and SO42− groups. Analysis of monosaccharides using gas chromatography showed that for both species glucose and xylose were the main constituents, but several other monosaccharides were present in smaller quantities. Two fractions of EPS were distinguished: a small amount was secreted into the medium and a second fraction was extracted in water at 30°C. For both species the two fractions differed somewhat in composition, indicating that they represented two different types of EPS. The EPS produced by N. salinarum and by C. closterium differed in their composition. The rate of EPS production in batch culture was highest during the transition from exponential growth to stationary growth. Negatively charged groups such as uronic acids and sulphated sugars determine the adhesion capacity of EPS and probably play an important role in the stabilization of intertidal sediments on which these diatoms grow and produce biofilms.
