Giant panda Ailuropoda melanoleuca exhibits are popular attractions for zoos and wildlife parks. However, it remains to be investigated whether such exhibits enhance visitor knowledge about pandas and broader conservation issues. We conducted questionnaire surveys at giant panda exhibits at three city zoos and five wildlife parks in China. Although visitors were generally interested in the giant panda, this was not reflected in their post-exhibit knowledge of giant panda biology. Socio-demographically, men were more knowledgeable of giant panda biology than women. Knowledge correlated positively with respondent level of education. Younger respondents (< 45 years) knew most about giant pandas and expressed an interest in learning more about them using social media. The most informed respondents had visited other giant panda exhibits previously. Respondents were generally satisfied with the giant panda exhibits (mean score 4.44/5). Wildlife parks delivered a better educational outcome than city zoos. We recommend approaches to improve the visitor experience further and to leverage public interest in broader conservation engagement and action in China.

The Endangered giant panda Ailuropoda melanoleuca is one of the most threatened mammals. The species has experienced declines in its population and habitat as a result of human disturbance. We investigated the influence of human disturbance on habitat use by giant pandas in the Daxiangling Mountains, in China's Sichuan Province. We mapped all signs of giant panda and all locations of seven types of human disturbance in the study area. We used correlation analysis, generalized linear models, and Akaike information criteria to analyse the influence of the various types of human disturbances on habitat use by the giant panda. Our results showed that habitat use was positively correlated with elevation and distance from roads, residences, hydropower stations and logging or tree-felling sites, but negatively correlated with distance from bamboo shoot collection sites and trap sites. We found that the road-effect zone spanned a distance of c. 1,200 m and that human residence could affect the intensity of habitat use by giant pandas at distances > 2,500 m. The effect of roads on habitat use was probably influenced by the association of roads with residences, hydropower stations and mines. In the area occupied by giant pandas, we recommend increased regulation to minimize the expansion and impact of roads, residences, hydropower stations and logging activities.

The effect of landscape on populations is of great importance, especially given the number of species inhabiting patchy landscapes. Developments in geographical information systems are facilitating a greater application of spatial analyses to threatened species, such as the Endangered giant panda Ailuropoda melanoleuca, for which habitat patchiness and quality are key limiting factors. Of all factors that influence the suitability of habitat for the giant panda, topography is not subject to change. Here, we report a spatial and statistical analysis of the high quality topographic habitat preferred by the giant panda across its stronghold in the Qinling Mountains, China. High quality topographic habitat, as indicated by a combination of elevation, slope and aspect, covers 92,788 ha, accounting for 15% of the current range of the species. The distribution of the giant panda closely follows patterns of topography and areas with patches of high quality topographic habitat are strongly associated with areas supporting greater numbers of giant pandas. However, comparisons between our model and the existing reserve system reveals a number of inadequacies. Some of the reserves contain little high quality topographic habitat and many patches of high quality topographic habitat are unprotected. Given the importance of topography and the decisive role it plays in shaping habitat, landscapes containing high quality topographic features must be a critical consideration in the design of reserves for the giant panda. The existing system of nature reserves is heavily weighted towards judicial and administrative boundaries, to the detriment of other factors such as topography.

The giant panda Ailuropoda melanoleuca, endemic to China, is perhaps the most powerful and well-known global icon of biodiversity conservation, having been the symbol of WWF since it was formed in 1961. It is categorized as Endangered on the IUCN Red List. Although there have been conservation successes for the giant panda in the last 2 decades, with 59 reserves established for the species, many conservation challenges remain and the species' status on the IUCN Red List has not changed since 1990, when it was first categorized as Endangered. Habitat fragmentation, synchronous bamboo flowering and die-off, and mismanagement of reserves are continuing challenges.

Insulin-like growth factor-1 (IGF-1) is a polypeptide that regulate growth, differentiation and muscle regeneration in several species. This study was designed to analyse the cDNA sequence of IGF-1 in giant panda, its gene expression and tissue distributions in relation to growth, development and reproduction of captive giant pandas. A cDNA encoding IGF-1 was cloned by reverse transcriptase-PCR from a male giant panda cub liver tissue. Sequence alignment analysis showed the cysteine residues and other amino acid residues of the mature IGF-1 peptide of giant panda are highly conserved in mammalian species. DNA encoding the mature IGF-1 peptide of the giant panda was ligated with pET-DsbA expression vector and expressed in E. coli BL21. Western blot confirmed the IGF-1 antigen activity of the recombinant protein. By in situ hybridization and immunohistochemistry analysis it is demonstrated that the IGF-1 mRNA distribution abundance were significantly higher in the liver, muscles and heart than those in the other tissues studied. However, the IGF-1 peptide distribution abundances in all the tissues studied did not show any significant differences. The localization of the IGF-1 peptide was in accordance with that of mRNA for each tissue. These results may suggest that IGF-1 play an autocrine or paracrine rôle in regulation of the giant panda growth and development.

As a solitary species, giant pandas Ailuropoda melanoleuca have a chemical communication system that allows for avoidance with conspecifics throughout most of the year and facilitates breeding during their brief reproductive period. To date, most studies have focused on adults during the breeding season, but much remains to be learned about developmental and seasonal effects on chemical communication. Here, using odour discrimination methodology, we examine these contextual influences on chemosignalling in giant pandas for the first time. During the breeding season both adult and subadult giant pandas overtly discriminated conspecific sex using chemical cues in urine. Male urine was consistently investigated more than female urine by pandas across all age–sex categories. This preference for male urine was observed for both adult and subadult urine donors, indicating that adult levels of reproductive hormones are not necessary for the production of sex-specific urinary odour cues. By contrast, giant pandas did not overtly discriminate sex during the non-breeding season. This finding underscores that negative results for discrimination studies need to be interpreted with caution, as many contextual factors may influence overt expression of this ability. Evidence points to seasonal changes in motivation, rather than in the availability of sex-specific chemical cues in urine, as the causal factor for changing odour discrimination patterns. These and other findings are discussed in the light of the potential role that discrimination of odour plays in the regulation of competitive and reproductive interactions in nature.

The factors that affect the selection of feeding sites by giant pandas Ailuropoda melanoleuca in the Longxi-Hongkou Nature Reserve were explored in winter from 2000 to 2002, by using strip transects. It was reported in the literature that giant pandas prefer to forage on the edges of bamboo patches because these edges offer them more food than the interiors. The number of feeding sites and the corresponding vegetation indices (including the basal diameter of bamboo stem, height and density of bamboo or bamboo shoot; density and canopy density of trees) were compared close to and far away from the edges of bamboo patches to detect the patterns of feeding habitat selection by giant pandas. Giant pandas preferred to forage at the sites near edges rather than in the interiors of bamboo patches. Such a tendency was independent of bamboo species and individual pandas. By comparing the density, basal diameter of bamboo stem and shoot, tree density and available dry biomass of bamboo, we detected no significant differences between the edges and the interiors of bamboo patches. The possible explanations were analysed and it was deemed that the selection of food habitat near edges by the giant pandas could be attributed to the direct effects of edges, which facilitate movement of the giant pandas in the otherwise dense bamboo patches.

As a solitary species, giant pandas Ailuropoda melanoleuca appear to use chemical signals to maintain a communication network with individuals in the community with whom they have little direct contact. Chemosensory discrimination abilities undoubtedly play a large role in determining the efficiency of this communication system. The hypothesis that pandas could discriminate the age of the signaller was tested by presenting subjects with a simultaneous choice test between odours collected from adult and subadult donors. Twenty-eight pandas were tested with three types of stimuli: male urine; male anogenital gland secretions; female urine. The results indicate that individuals from both age classes readily discriminate between adult and subadult odours emanating from all three odour types, showing a pronounced investigatory preference for adult odours in all cases. Female reproductive condition influenced general responsiveness to male odours, but did not significantly alter adult/subadult discrimination patterns. The prevalence of overt discrimination across odour types and age classes suggests an important and multifunctional role for this ability, and highlights the importance of studying chemical communication at all developmental stages. It is proposed that these results may be explained in part by: (1) assessment of competitive ability, which correlates with the age class of the signaller; (2) identification of potential mates that have attained reproductive maturity.

Giant pandas Ailuropoda melanoleuca are a solitary, seasonally mono-oestrous species, and as such are likely to rely heavily on chemical signals to co-ordinate mating efforts. Perhaps the most crucial chemosensory task facing male pandas is the assessment of female reproductive condition on the basis of chemical cues. To test for this ability, eight male and 10 female giant pandas were presented with urine from oestrous and non-oestrous females. Males but not females displayed a significant investigatory preference for oestrous over non-oestrous female urine, as indicated by sniffing and flehmen. Males also investigated, displayed flehmen, and licked female urine more than did females. These findings indicate that female urine contains chemical cues permitting discrimination of oestrous status, and probably serves a reproductive advertisement function. The prevalence of flehmen and licking by males suggests that males may rely on the vomeronasal organ to assess female reproductive condition. Female odours may play an important role in the reproductive behaviour of male giant pandas and, therefore, may have implications for captive-breeding programmes.

We studied the postnatal body weight gain and development of 11 male and nine female giant panda Ailuropoda melanoleuca cubs born at Beijing Zoo from 1985 to 1998. Growth rates of the cubs appeared to be sexually dimorphic from the fourth month after birth; the male grew slightly faster than the female cubs. Growth rates between artificially fed and naturally fed cubs were significantly different from the fourth month after birth. The growth rate of the artificially fed cubs was slightly higher than that of the cubs fed by their mothers, indicating that the substitute milk satisfied the nutrient needs of the cubs. The body length of the cubs increased rapidly after birth; 8-month-old cubs were three times longer than newborn cubs. Chest circumferences of 8-month-old cubs also increased to twice that of newborn cubs. Tail length relative to body length was reduced from 14.9% at birth to about 8.6% in 8-month-old cubs. Cubs started to grow teeth when they were 3 months old. By the age of 1 year, the cubs had fully grown deciduous teeth. The teeth formula of one 1-year-old cub was 2·1·3·0/2·1·3·0=24. We recorded the changes in fur colour, development of the sense organs and limbs of the giant panda cubs. Finally, we compared the body weight and life-history parameters of giant pandas with those of bears and raccoons and discussed the management regime for the care of captive-born giant panda cubs.

Reproductive activities of six giant panda females (Ailuropoda melanoleuca) living in the Qinling Mountains of China (Shaanxi Province) were recorded from 1989 into early 1995. Data on mating and birth dates, litter size, cub sex and den use were collected for three of the pandas. At this site, pregnant females migrated to lower elevations in advance of other pandas to seek dens for birthing. One dam was videotaped in the den with her cub from birth to the end of the denning phase at 94 days of age. This cub was repeatedly handled during the dam's absences to obtain data on development and weight. During the early part of the denning phase, the cub was maintained on the dam's body virtually continuously. Post-partum fasting for up to 3 weeks was noted. Dams invariably moved cubs to new dens three or four times during the birth season. By the time cubs left the den at about 3–4 months, they were fully furred and capable of surviving winter temperatures without further shelter. Reproductive parameters for the wild population fell within the range of values for captive pandas, based on information in the literature and in the giant panda studbook. The combined data on length of gestation, intervals between births, litter size and weight, infant development, and reproductive life span reveal a range of life-history variables that strongly resemble the Ursidae.
