If physiological mechanisms similar to cat Y and X cells explain faster detection of low spatial frequencies by humans, then cats should show the same effect. We have tested this prediction by determining the visual reaction time of cats over a range of spatial frequencies and contrasts by training them to respond quickly when a vertical sine-wave grating was presented. At 50% contrast, the cat's visual reaction time increased monotonically from 0.25–2.0 cpd (cycle/deg). At every spatial frequency tested, the cat's reaction time increased monotonically as contrast decreased. By determining contrast threshold (70% detection) at each spatial frequency, it was possible to determine reaction times for different spatial frequencies at equal physical contrasts and equal “threshold equivalent” contrasts. Some of the cat's faster detection of low spatial frequencies was due to sensitivity differences and some was not. To determine if faster detection of low spatial frequencies was based upon Y cells, we took advantage of the fact that Y cells show a strong peripheral effect while X cells do not. Low and high spatial frequencies were detected in the presence of a flickering (7 Hz) or steady (70 Hz) surround. Surround frequency had no effect upon reaction times to 2.0 cpd but the flickering surround increased reaction times to 0.25 cpd. These results indicate that, in cats, rapid detection of low spatial frequencies is by Y cells and slower detection of high spatial frequencies is by X cells.

In this study, we investigated the potency of dopamine for being an intrinsic signal for cyclic events in the fish retina. Dopaminergic activity was measured during different light/dark cycles, during continuous darkness, and during short-term light and dark adaptation within 1 h. During a 12-h light/12-h dark cycle, the total content of endogenous dopamine was high during the dark phase and low during the light phase. The potassium-induced release of endogenous dopamine followed a parallel time course. The concentration of the dopamine breakdown product 3,4-dihydroxyphenylacetic acid (DOPAC), which reflects the endogenous dopaminergic activity, was high during the light phase and low during the dark phase. Similar alterations occurred in accelerated 6-h light/6-h dark cycles, again indicating a strong coupling of dopaminergic activity with light. The cyclic alterations in the total endogenous dopamine content persisted during continuous darkness after an entrainment of the fish to a 12-h light/12-h dark cycle. Although the magnitude of the change was weaker, changes in dopamine content, potassium-induced dopamine release, and DOPAC were also measured during 1 h of light or dark adaptation. During a 1-h period of dark adaptation, the total content of dopamine and the potassium-induced release of endogenous dopamine increased, while DOPAC values decreased. These values changed in the opposite direction during 1 h of light adaptation.

Our findings strongly suggest that dopamine is the intrinsic signal for light during both the light and dark phases and during short-term adaptation. Light seems to be the major trigger for dopaminergic activity within the fish retina. An endogenous clock might participate in the control of the dopaminergic activity. This has to be, however, confirmed by further investigations.

The morphological development of the tectofugal pathway in the zebra finch has recently been described in a series of studies from our laboratory. No data are currently available on the development of visual responsiveness in this pathway. We therefore investigated the development of visually evoked potentials (VEPs) in the ectostriatum, the telencephalic target area of the tectofugal pathway. Contralateral VEPs could already be recorded in 20-day-old birds, whereas ipsilateral VEPs could first be recorded in 40-day-old birds. The latencies of contralateral VEPs decrease to adult values between 20 and 40 days of age, probably due to an increase in the myelination of afferent fibers. The amplitudes of the contralateral VEPs increase continuously from day 20 to day 60; however, between 60 and 80 days of age the responses diminish substantially (−60%). Thus, contralateral VEPs in 80-day-old birds are not significantly different from those in 20-day-old birds. Thereafter the responses recover and reach their final amplitude values at about 150 days of age. The relationship of these results to morphological studies and possible mechanisms which may cause the double-peaked development of visual-evoked potentials in the ectostriatum are discussed.

The receptive-field profile of the cone in cat retina was computed. The computation was based on (1) the known anatomical circuit connecting cones via narrow-field bipolar cells to the on-beta ganglion cell; (2) the known physiological receptive-field profile of the on-beta (X) cell at the corresponding eccentricity; and (3) a model in which the beta receptive field arises by linear superposition of cone receptive fields. The computed cone receptive field has a center/surround organization with a center almost as broad as that of the beta cell center. The cone surround is comparably broad to that of the beta cell but somewhat lower in peak amplitude. The problems to which the center/surround receptive field are the solution, namely, signal compression and noise reduction, apparently must be solved before the first synapse of the visual pathway.

This study has shown that the retinae of Prototherian (egg-laying) mammals possess two neuronal types that are present in non-mammalian retinae, but absent or morphologically different in the retinae of Eutherian (placental) mammals. First, endogenous serotonin-like immunoreactivity has been localized in a population of presumptive amacrine cells in the platypus retina, the first such report in a mammalian retina. Second, the protein kinase C-immunoreactive (PKC-IR) bipolar cells in the echidna retina appear similar to the PKC-IR bipolars in the chicken retina, in that their dendrites give rise to a Landolt's club and their axons are multistratified. By contrast, the PKC-IR rod bipolar cells in the rabbit and in the brushtail possum, a Metatherian (marsupial) mammal, have no Landolt's clubs and their axons form terminal lobes in the innermost stratum of the inner plexiform layer.

Immunohistochemical and tracing techniques were used in combination to reveal the source of a neuropeptide Y-like immunoreactive (NPY-LI) plexus in the nucleus of the basal optic root (nBOR) of the pigeon accessory optic system. Injections of rhodamine-labeled latex microspheres into nBOR produced retrograde labeling of a population of neurons interposed between the principal optic nucleus of the dorsolateral thalamus (equivalent to the mammalian dorsal lateral geniculate nucleus) and the ventral lateral geniculate nucleus. The retrogradely labeled neurons were distributed mainly in the immediate vicinity of the lateral, dorsal, and ventral aspects of the nucleus rotundus. Immunohistochemical methods revealed many NPY-containing somata within the same intergeniculate thalamic area. Double-labeling immunohistochemical and retrograde tracing experiments evidenced that many NPY-LI neurons in the intergeniculate area contained rhodamine microspheres that had been previously injected into the ipsilateral nBOR. The projection of that general thalamic area to the nBOR was then confirmed by means of anterograde transport of Phaseolus vulgaris leucoagglutinin. In these experiments, the intergeniculate region was demonstrated to project to all divisions of the nBOR and to every other retino-recipient structure, including the suprachiasmatic nucleus. Finally, electrolytic lesions of the intergeniculate area produced a dramatic reduction in the number of NPY-LI axons and terminals within the ipsilateral nBOR and also within other retino-recipient structures. These data indicate the existence of a thalamic NPY-LI projection to the pigeon nBOR of the accessory optic system. This chemically specific projection originates from the intergeniculate area, which was shown in this study to project to all other retino-recipient structures. Thus, NPY may have a role in the functional organization of the accessory optic system and also of the avian visual system as a whole.

We studied quantitatively the receptive-field properties of 74 units recorded from the representation of the central visual fields in the cat's lateral suprasylvian (LS) visual cortex. In agreement with previous workers, we found that LS receptive fields tended to be large and to lack discernible spatial structure. They resembled the complex receptive fields of areas 17 and 18 in their general organization. We examined the responses of these neurons to moving optimally oriented sinusoidal gratings that varied in spatial and temporal frequency of drift. Most LS neurons were selective for the spatial frequency of sinusoidal gratings; 7% responded to all spatial frequencies below a cutoff value. In agreement with previous reports, the optimal spatial frequencies for LS neurons covered a wider range than is seen in either area 17 or 18 alone (0.05–1 cycle/deg), but are certainly included in the range covered by both these afferent areas. Individual neurons in LS responded to a range of spatial frequencies broader than is typical for neurons in areas 17 and 18. The effect of varying the drift rate of otherwise optimal gratings was similar in LS to that reported for areas 17 and 18. Most neurons were optimally responsive to drift rates between 0.5 and 4 Hz, and resolved frequencies as high as 10–30 Hz. A few neurons had optima higher than 6 Hz and resolved frequencies in excess of 30 Hz. We conclude that the receptive fields of LS neurons reflect rather closely the properties of their afferents from areas 17 and 18. Apart from the increased incidence of directional selectivity in LS and the increase in receptive-field size seen there, we find no evidence for a significant reorganization of visual signals.

An eletrical potential recorded from the cornea, the a-wave of the ERG, is evaluated as a measure of human photoreceptor activity by comparing its behavior to a model derived from in vitro recordings from rod photoreceptors. The leading edge of the ERG exhibits both the linear and nonlinear behavior perdicted by this model. The capability for recording the electrical activity of humans photoreceptors in in vivo opens new avenues for assessing normal and abnormal receptor activity in humans. Furthermore, the quantitative model of the receptor response can be used to isolate the inner retinal contribution, Granit's PII, to the gross ERG. Based on this analysis, the practice of using the trough-to-peak amplitude of the b-wave as a proxy for the amplitude of the inner nuclear layer activity is evaluated.

Striate cortical cells were classified according to whether or not their preferred orientation was close to one of the “primary” orientations (horizontal, vertical or radial, i.e. directed to the area centralis) and according to their ordinal position on the afferent pathway from the dorsal lateral geniculate nucleus (dLGN). Among the neurones that could be driven monosynaptically from the dLGN, there was a high representation of those with a preference for the primary orientations. This was particularly evident in the case of C (complex) cells. There was no such preponderance of primary orientations among the polysynaptically activated cells. It is proposed that the asymmetry of distribution seen among the first-order cells reflects the asymmetry seen subcortically in neurones that show orientation biases. It may be that the cortex elaborates a more uniform representation of orientations only at the higher ordinal levels.

The projection of the thalamic intralaminar nuclei (ILN) upon the visual cortex in the squirrel monkey was studied using anterograde, autoradiographic techniques. In area 17, the ILN afferents terminate in the inner and outer portions of lamina V, whereas in areas 18 and 19 the fibers terminate more diffusely along the laminae V–VI boundary. Widespread labeling of layer I is seen throughout the occipital cortex.

We have examined the distribution of immunoreactivity for the monoclonal antibody Cat-301 in visual cortex of the macaque monkey. Remarkably, those portions of striate cortex (V1) and extrastriate cortex that are most immunoreactive for Cat-301 are anatomically interconnected and are dominated by inputs arising from the magnocellular layers of the LGN (which are themselves highly immunoreactive). In particular, we found that a band of Cat-301 labeled neurons known to exist in layer 4 of V1 is centered on the boundary between layers 4Cα and 4B and thus includes portions of both the primary target of the magnocellular LGN and its subsequent relay through layer 4B. We also demonstrated consistently strong Cat-301 immunoreactivity in all three extrastriate targets of layer 4B: areas V3, MT, and the cytochrome-oxidase (CO) enriched thick stripes of V2. In V2, there was a close correlation between Cat-301 labeling and clusters of cells projecting to MT but not to V4. This was true even in regions where the CO pattern was equivocal or irregular, indicating that Cat-301 is a more reliable marker than CO for the thick-stripe subregions of V2. Finally, we found strong Cat-301 immunoreactivity in at least parts of areas V3A, the MST complex, and the posterior parietal complex, but not in area V4 or inferotemporal cortex. The molecular specificity revealed by this single marker thus correlates with functionally specific subdivisions at each hierarchical level over nearly the entire known extent of the visual pathway in macaques. This supports the notion that these subdivisions form an anatomically, physiologically, and now molecularly distinct pathway known as the M-stream.

Although gradient schemes for detecting the motion of images and measuring their velocities are commonly used in computer vision, and although there is increasing evidence to support the existence of such schemes in biological vision, little attention has been directed to suggesting how such computations might be realized by neural hardware. This paper proposes two simple models, consisting of physiologically realistic networks of neurons, that approximate the gradient scheme. Computer simulations demonstrate that the models measure the speed of an object or pattern independently of its structural properties.

Ganglion cells in the albino rat retina were retrogradely labeled with the fluorescent dye, diamidino-yellow, from the superior colliculus. Preembedding and postembedding immunocytochemical techniques were employed in conjunction with computer-assisted image processing to visualize SP- and GABA-immunoreactivity. Examination of flatmount and sectioned retinas revealed that approximately 3% of the ganglion cells projecting to the contralateral superior colliculus exhibit SP-immunoreactivity. Moreover, these cells were found to comprise a subpopulation of the GABA-immunoreactive cells projecting to the rat tectum.

The role of GABA in the outer plexiform layer of the turtle retina has been examined by intracellular recordings from L- and C-type horizontal cells in the isolated retina preparation.

GABA (1–5 mM) slightly depolarized the L-type horizontal cells, reduced the amplitude of their photoresponses, and slowed down the rate of hyperpolarization during the ON component of the photoresponse. These effects could not be replicated by either muscimol or baclofen. When synaptic transmission from the photoreceptors had been blocked by either kynurenic acid or cobalt ions, GABA depolarized L-type horizontal cells and augmented the remaining photoresponses. Neither muscimol nor baclofen exerted any effect on L-type horizontal cells under these conditions. Nipecotic acid, a competitive inhibitor of the GABA-uptake system, induced effects on turtle L-type horizontal cells which were similar to those exerted by GABA. Thus, the complex GABA effect on turtle L-type horizontal cells seems to represent the summation of at least two actions; an indirect one mediated by the red cones via GABAa-type receptors and a direct one which probably reflects the activation of an electrogenic GABA-uptake system.

GABA (1–5 mM) induced a transient depolarization in C-type horizontal cells but eliminated color opponency in only three cells out of seven studied. This observation is inconsistent with the notion that the only neural mechanism responsible for the chromatic properties of C-type horizontal cells in the turtle retina is a GABAergic negative feedback from the L-type horizontal cells onto the green ones.

The effects of synaptic blocking agents on the antagonistic surround of the receptive field of cone photoreceptors were studied intracellular recording in the retina of hte turtle (Pseudemys scripta elegans) Illumination of a cone's receptive-field surround typically evoked a hybriid depolarizing response composed of two componests: (1) the graded synaptic feedback depolarization and (2) the prolonged depolarization a distinctive, intrinsic response of the cone. The locus of action of synaptic blocking agents was analyzed by comparing their effects on the light-evoked response of horizontal cells, the hybrid cone depolarization evoked by surround illumination, and the pure prolonged depolarization evoked by intracellular current injection.

The excitatory amino-acid antagonists, d-O-phosphoserine (DOS) and kynurenic acid (KynA), suppressed the light responses of horizontal cells and eliminated the surround-evoked, hybrid cone depolarization without affecting the prolonged depolarization evoked by current injection. Cobalt at 5–10 mM suppressed horizontal cell responses and thereby eliminated surround-evoked cone depolarizations. Cobalt (5–10 mM) also blocked the current-evoked prolonged depolarization, suggesting that the intrinsic cone mechanisms responsible for the prolonged depolarization are likely to be calcium-dependent.

Various GABA agonists and antagonists were found to have no effect on the surround-evoked depolarizations of cones. In contrast, a very low concentration of cobalt (0.5 mM) selectively suppressed the light-evoked feedback depolarization of cones without affecting horizontal cell responses or the current-evoked prolonged depolarization. Cobalt at 0.5 mM thus blocks the light-evoked action of the cone feedback synapse while sparing feedforward synaptic transmission from cones to horizontal cells. The implications of the present work for the possible neurotransmitters used at these synapses is discussed.

The telencephalic target of the thalamofugal visual pathway in birds, the visual wulst, is part of the hyperstriatum accessorium/dorsale in the bird's brain. In this study, we tried to determine the exact location of the visually responsive area in the zebra finch by recording visually evoked potentials (VEPs) from different sites throughout the hyperstriatum and calculating current source densities (CSDs). In addition, we examined the influence of ipsilateral and contralateral stimuli on stimulus processing within this area, and tried to get insight into the neuronal machinery of the thalamofugal pathway by application of drugs such as tetrodotoxin (TTX) and picrotoxin.

About two-thirds of the hyperstriatum is responsive to contralateral stimuli but only a small portion responds to ipsilateral stimuli. Contralateral visual information arrives in the hyperstriatum dorsale (HD) and is processed further to the hyperstriatum accessorium (HA).

The small influence of ipsilaterally evoked potentials is not due to inhibition by the activity of the contralateral eye, as could be demonstrated previously for the ectostriatum. Instead, our results show that ipsilaterally evoked potentials are inhibited at least in part by a projection from the contralateral visual wulst.

Postsynaptic responses of tectal cells in the bullfrog (Rana catesbeiana) were intracellularly recorded following electrical stimulation of the optic tract and the nucleus isthmi, and fluorescent dye, Lucifer yellow, was injected into some of the impaled cells to show their morphologies. Two main response types were found: The first type was an EPSP followed by an IPSP, and the second type was single IPSP. The first type predominates in cells responding to the optic tract stimulation and the second type prevails in cells responding to the isthmic stimulation. Fifteen cells stained with Lucifer yellow were localized in layer 6 (11 cells), layer 7 (1 cell), and layer 8 (3 cells). They were mainly identified as pear-shaped cells, large ganglionic cells, and stellate cells. Three injections demonstrated “dye-coupling,” which labeled up to six cells following one injection. Comparisons of postsynaptic potentials with cellular morphologies suggested that the nucleus isthmi could directly excite large ganglionic neurons in layer 6. Synaptic mechanisms for strong isthmic inhibition on the tectal neurons remain unknown.

The role of 48-kDa protein in Visual transduction remains unresolved. Two hypotheses for its role in quenching the light activation of cyclic GMP cascade suggest that the protein binds to either phosphodiesterase or phosphorylated rhodopsin. Since the protein is also reported to bind ATP, we anticipated that the protein may have ATP hydrolyzing activity, and in analogy with the GTP-binding protein of the rod outer segments, such activity may be greatly enhanced by the elements of transduction cyclic GMP cascade, permitting the protein to function cyclically as GTP-binding protein does. We found that purified 48-kDa protein hydrolyzes ATP but at a slow rate of 0.04–0.05 per min. The Km for ATP is about 45–65 μM. The activity is inhibited noncompetitively by ADP with a Ki of about 50 μM. The ATPase activity of 48-kDa protein is not affected by rhodopsin, bleached rhodopsin, phosphorylated rhodopsin, unactivated cyclic GMP phosphodiesterase, or phosphodiesterase (PDE) activated by GMP PNP-bound G-protein. These data show that although 48-kDa protein has ATPase activity, lack of regulation of this activity by the elements of visual transduction makes it unlikely for this activity to have a role in quenching the light activation of cyclic GMP cascade.

Cortical connections were investigated by restricting injections of WGA-HRP to different parts of the middle temporal visual area, MT, in squirrel monkeys, owl monkeys, marmosets, and galagos. Cortex was flattened and sectioned tangentially to facilitate an analysis of the areal patterns of connections. In the experimental cases, brain sections reacted for cytochrome oxidase (CO) or stained for myelin were used to delimit visual areas of occipital and temporal cortex and visuomotor areas of the frontal lobe. Major findings are as follows: (1) The architectonic analysis suggests that in addition to the commonly recognized visual fields, area 17 (V-I), area 18 (V-II), and MT, all three New World monkeys and prosimian galagos have visual areas DL, DI, DM, MST, and FST. (2) Measurements of the size of these areas indicate that about a third of the neocortex in these primates is occupied by the eight visual areas, but they occupy a somewhat larger proportion of neocortex in the diurnal marmosets and squirrel monkeys than the nocturnal owl monkeys and galagos. The diurnal primates also have proportionally more neocortex devoted to areas 17, 18, and DL and less to MT. These differences are compatible with the view that diurnal primates are more specialized for detailed object and color vision. (3) In all four primates, restricted locations in MT receive major inputs from short meandering rows of neurons in area 17 and several bands of neurons in area 18. (4) Major feedforward projections of MT are to two visual areas adjoining the rostral half of MT, areas MST and FST. Other ipsilateral connections are with DL, DI, and in some cases DM, parts of inferotemporal (IT) cortex, and posterior parietal cortex. (5) In squirrel monkeys, where injection sites varied from caudal to rostral MT, caudal parts of MT representing central vision connect more densely to DL and IT than other parts. Both DL and IT cortex emphasize central vision. (6) In the frontal lobe, MT has dense connections with the frontal ventral area (FV), but not with the frontal eye field (FEF). (7) Callosal connections of MT are most dense with matched locations in MT of the other hemisphere, rather than with the outer boundary of MT representing the vertical meridian. Targets of sparser callosal connections include FST, MST, and DL.

The results support the conclusions that (1) prosimian primates and New World monkeys have at least ten visual and visuomotor areas in common, (2) the connections of MT are remarkably consistent across four species of primates, (3) the anatomical segregation of visual subsystems in areas 17 and 18 is common to all primates, (4) connections from the part of MT representing central vision with visual areas emphasizing central vision are more dense, and (5) MT and the associated fields MST and FST occupy proportionally more cortex in nocturnal than diurnal primates.

The period of susceptibility for binocular depth vision was studied in kittens by subjecting them to periods of monocular deprivation beginning at different ages. In an initial study, we found that normally reared kittens can learn a depth-discrimination task much more rapidly when tested binocularly than monocularly, even when testing is begun as early at 30 d. In subsequent experiments, kittens were monocularly deprived by eyelid suture, following which their monocular and binocular depth thresholds were measured using the jumping-stand procedure. We obtained the following results: (1) When monocular deprivation is applied before the time of natural eye opening but is discontinued by no later than 30 d, there is very Little effect on binocular depth thresholds. (2) When deprivation is begun at 90 d, binocular depth thresholds are unaffected. (3) When deprivation is begun between these two ages, the magnitude of the deficit varies with the period of deprivation and the age at which it begins. (4) By imposing brief (5 or 10 d) periods of deprivation, beginning at different ages, we were able to demonstrate that the peak of the sensitive period is between the ages of 35 and 45 d, with a fairly rapid decline in susceptibility outside those age limits. (5) Even with as little as 5 d of deprivation, substantial permanent deficits in binocular depth vision can be induced.
