Seed storage proteins (SSPs) are synthesized during development and the expression of their genes is under tight tissue-specific and temporal transcriptional regulation. In this review we summarize the current knowledge concerning the regulatory steps controlling SSP synthesis in Arabidopsis and cereals, which involves the interaction of cis regulatory elements with corresponding trans-acting factors. In some cases, the regulation of SSP genes requires the concerted action of multiple transcription factors (TFs). There is an evolutionary conservation between the prolamins (the main group of SSPs in many cereal grains) and a major group of dicot seed albumins; this relates to both the regulatory elements and the TFs that are functionally exchangeable between the monocot and dicot species.

The transition from the dormant to the non-dormant state of a viable and mature seed can take place at low hydration by exposure to air-dry storage conditions (dry afterripening; AR). The events occurring during this loss of dormancy are of considerable physiological, ecological and agricultural interest. AR may be attributable to increased sensitivity to germination-stimulating factors and a widening of the temperature window for germination. Genetic, –omics and physiological studies on this mode of dormancy breaking provide support for a key role of the balance between gibberellin (GA) and abscisic acid (ABA) metabolism and sensitivity. Recent evidence also supports a possible role for ethylene (ET) in this complex signalling network that is necessary for AR implementation. However, hormone-independent signals, such as reactive oxygen species (ROS), nitrate () or nicotinamide adenine dinucleotide (NAD+), also appear to be involved. The way in which hormone- and non-hormone-signalling pathways affects each other (cross-talk) is still under study. This review provides updated information on the programmes that overcome seed dormancy. Thus, we have reviewed: (1) the –omic status in dry seeds; (2) the relationship between temperature and nitrate signalling and AR; (3) alterations in ABA/GA synthesis and signalling; (4) the action of hormone molecules other than ABA and GA (i.e. ET, salicylic and jasmonic acids); and (5) participation of reactive oxygen species (ROS), NAD+ and protein carbonylation. Taken together, the acquisition and implementation of dry AR involve a complex signalling network that is difficult to disentangle.

Dehydrins are late embryogenesis abundant (LEA) proteins that accumulate during seed maturation and in response to abiotic stresses in vegetative tissues. They are thought to protect cellular components from dehydration stress. However, whether they play a role in survival in the dry state is not clear. In this study, an RNA interference (RNAi)-construct against the seed-expressed dehydrin of Arabidopsis thaliana, LEA14 (At2g21490), was introduced to wild-type plants, which led to a strong reduction in transcript abundance of the target gene as well as that of two other seed-expressed dehydrin homologues, XERO1 (At3g50980) and RAB18 (responsive to abscisic acid 18, At5g66400) in the transformants. Mature, dry seeds from the RNAi plants germinated to at least 95% after rehydration, indicating that seed desiccation tolerance was not affected, while they exhibited a twofold reduction in longevity. When stored at 75% relative humidity and 35°C, the seeds of two independent RNAi lines lost 50% of their viability in 10 d and 5 d, respectively, while it took 17 d for wild-type seeds to lose 50% viability. In addition, when seeds were imbibed in the presence of 100 mM NaCl, the seeds of RNAi plants exhibited reduced germination compared to wild-type seeds, suggesting that at least one of the three seed-specific dehydrins plays a role both against deterioration during storage at low moisture content and when imbibed tissues are submitted to salt stress at high moisture.

The Millennium Ecosystem Assessment estimates that between 60,000 and 100,000 plant species are threatened with extinction–equivalent to around one-quarter of the total number of known plant species. Why should we care? There are a number of reasons. The first is that these plants may be useful to us in unknown ways. Secondly, ecology has taught us that resilience is found in diversity. Thirdly, we should be saving plant species from extinction because we can–there is no technological reason why any plant species should become extinct. Where we can't protect and manage plant diversity in situ, we should be employing ex situ conservation techniques, ranging from seed banks to habitat restoration. The Millennium Ecosystem Assessment describes such interventions as ‘techno-gardening’. This is not an abstract concept–it is already a reality in the majority of man-managed landscapes. In this context the perception of ex situ conservation as simply a back-up strategy for in situ conservation is mistaken. We are all involved in ex situ conservation to some degree, from cultivating our back gardens, to farming, to management of protected areas. Ex situ conservation should be seen as a complementary approach to in situ conservation and on the same spectrum. Kew's Millennium Seed Bank Partnership, comprising more than 120 plant science institutions in 50 countries, epitomizes this philosophy in action. We work actively on every seed collection we bank, finding out how useful it is and how we can grow it to enable human innovation, adaptation and resilience. Challenges remain at the policy level; for example, the need to factor-in the value of natural capital to development decision making, and better defining a role for public-sector science. At the technical level, also, there is much to do. Perhaps the greatest technical challenges relate to the restoration and management of complex, self-sustaining habitats or species assemblages. If we are to techno-garden effectively, in order to maintain ecosystem services and sustain biodiversity, then a multidisciplinary approach will be required. Many plant science institutions have recognized this and are becoming engaged increasingly in restoration activities and in situ management. Ultimately, humanity's ability to innovate and adapt is dependent on our having access to the full range of plant species and the alleles they contain.

Starch is globally important as a source of food and, in addition, has a wide range of industrial applications. Much of this agriculturally produced starch is synthesized in developing seeds, where its biological function is to provide energy for seedling establishment. Storage starch in developing seeds is synthesized in heterotrophic plastids called amyloplasts and is distinct from the transient synthesis of starch in chloroplasts. This article reviews our current understanding of storage starch biosynthesis occurring in these organelles and discusses recent advances in research in this field. The review discusses starch structure and granule initiation, emerging ideas on the evolution of the pathway, the enzymes of starch synthesis, and the post-translational modification and regulation of key enzymes of amylopectin biosynthesis.

Lettuce (Lactuca sativa L.) and onion (Allium cepa L.) seeds have relatively short longevity during storage and their germination is sensitive to environmental stress. Seed priming (controlled hydration followed by drying) can improve seed germination under stressful conditions, inducing faster and more uniform germination over broader temperature ranges, but it can also reduce seed longevity in storage. Controlled deterioration (CD) tests are often employed to study longevity by ageing seeds rapidly at elevated temperature and moisture content, and primed seeds are particularly sensitive to CD conditions. As reactive oxygen (O2) species are thought to be involved in seed deterioration, we tested whether storage under reduced O2 atmospheres (0 and 2% O2) would extend the longevity of primed and non-primed seeds under low relative humidity (RH) (33% RH+37°C) and CD (75% RH+50°C) storage conditions. The longevity of both non-primed and primed lettuce seeds in low RH storage was extended by anaerobic environments, but the effect of O2 was much less under CD conditions. In onion, only primed seeds exhibited a beneficial effect of low O2 atmospheres under both types of ageing conditions. In both species, storage under anaerobic conditions was beneficial for extending the longevity of primed seeds, but was not able to ameliorate fully the negative effect of priming on storage life.

There is little information about seed dormancy release and germination behaviour in Vitis amurensis, which is one of the most important wild Vitis species. This work aimed to investigate the dormancy release and germination characteristics and their variation in V. amurensis seed. Seed dormancy was released by: (1) stratification at 5, 10, 15, 20, 25 and 15/5°C, respectively; (2) stratification at 25°C followed by stratification at 5°C; (3) 5°C stratification before or after seed drying; and (4) stratification at a series of relative humidities (RHs) at 5 and 25°C. The freshly harvested and dormancy-released seeds were germinated at three fluctuating and four constant temperatures. In V. amurensis: (1) dormancy release could occur at temperature ≤ 25°C, but was maximal at 5°C for all varieties; (2) dormancy release and induction occurred simultaneously during stratification; (3) the rates of dormancy release and induction were dependent on temperature; (4) seed moisture content affected dormancy release with a temperature-dependent pattern; (5) seeds germinated better at fluctuating temperature than at constant temperature. However, the optimum condition for stratification and germination, the rates of dormancy release and induction, and the response of seeds to warm stratification, to drying and to germination temperature varied significantly among the V. amurensis varieties. Our data imply that seed dormancy and germination in V. amurensis not only have intra-species similarity, but also vary within species, which may be due to differences in environmental conditions at the original growth and cultivated sites.

Although seed-size preference by granivores is relative to the sizes of seeds offered by researchers, preferences are often inappropriately discussed and interpreted as if they are for absolute seed sizes. Granivores often prefer relatively small seeds in forests and relatively large seeds in herbaceous communities, although the absolute size of preferred seeds in each is often similar. These results are consistent with predictions of optimal foraging theory. Researchers are encouraged to take heed of optimal foraging theory and both relative and absolute seed size when interpreting seed-size preferences.

Heretofore, no detailed account was available on seed dormancy and germination of a member of the Colchicaceae (Liliales). Thus, the primary aim of this study was to do an in-depth investigation of the temperature requirements for dormancy break and germination in seeds of Merendera montana (Colchicaceae) at the embryo and whole-seed levels under near-natural temperatures in a non-heated frame shade-house and under controlled conditions in the laboratory. Mean embryo length in fresh seeds was c. 0.57 mm and embryos had to grow to at least 1.30 mm before radicle emergence. Embryos grew to full size and seeds completed germination (radicles emerged) when they were stratified at 28/14°C for 60 d followed by a cool temperature for 60 d and then incubated at a cool temperature for 30 d. The optimum cool stratification temperature for dormancy-break was 10°C. Thus, after the moist pretreatment at 28/14°C+10°C, radicle emergence was>93% at all incubation temperatures (5, 15/4 and 20/7°C). In its natural habitat, M. montana seeds are dispersed in June, the embryo elongates to full size in autumn and radicles emerge from early November to early February. Although the shoot does not emerge until March and April, it is not physiologically dormant. The shoot emerged from 80% of the radicle-emerged seeds in 13 d at 20/7°C without a previous cold pretreatment. Seeds of M. montana have non-deep complex morphophysiological dormancy, C1b1aB-C1a. This is the first study on seeds with complex MPD to show a delay in shoot emergence following root emergence despite the shoot being physiologically non-dormant.

Fritillaria tubiformis subsp. moggridgei (Liliaceae) is a rare, endemic species that inhabits open mountains and alpine grasslands of the Ligurian Alps. At the time of seed dispersal, the underdeveloped embryos were 27% the length of the seed. Here we report the results of laboratory experiments carried out to determine the temperature preferences for embryo growth and radicle emergence. Embryo growth commenced immediately after sowing at 4°C. Once the embryo had grown the length of the seed, the radicle emerged. The time required for embryo growth and radicle emergence was longer when seeds were placed through a seasonal sequence of temperatures, commencing with late summer (10/20°C), compared with seeds immediately placed at a temperature to simulate winter conditions (4°C). Prematurely transferring seeds from winter to spring temperatures (5/10°C) also slowed the progress of germination. Radicle emergence did not occur at 10 or − 5°C and less than 20% germination occurred in seeds placed at constant 0°C. Addition of gibberellic acid (GA3) did not promote embryo growth of seeds placed at 20°C. Overall, the temperature preferences for embryo growth and subsequent radicle emergence are such that, in situ, seed germination may occur during the winter under snow cover or at the end of winter to coincide with snow melt and warming temperatures.

A characteristic mechanism of gene expression regulation during seed germination is the selective translation of mRNAs. Previous findings indicate that the two cap-binding complexes eIF4F (with eIF4E and eIF4G subunits) and eIF(iso)4F [with eIF(iso)4E and eIF(iso)4G subunits] are differentially expressed during maize seed germination. In addition, several studies in vitro have suggested that these factors may participate in selective mRNA translation. The translational activities of eIF4E and eIF(iso)4E were tested in vitro using transcripts from two different sets: dry (0 h) and 24-h-imbibed maize embryonic axes. In vitro translation of these mRNA pools in the presence of the recombinant eIF4E or eIF(iso)4E, and the native cap-binding complexes from dry- or 24-h-imbibed axes, produced different profiles of proteins which were visualized by two-dimensional protein gels and autoradiography. The data indicated that eIF(iso)4E was particularly required for translation of the stored mRNAs from dry seeds, and that eIF4E was unable to fully replace the eIF(iso)4E activity. In addition, the dry seed mRNA pool was translated by the cap-binding complex isolated from dry seeds better than by the complex isolated from 24-h-imbibed seeds, whereas the translational efficiency of the mRNA pool from 24-h-imbibed seeds was similar between the cap-binding complexes from these two stages. Interestingly, eIF(iso)4E was more abundant than eIF4E in dry seeds, while both cap-binding proteins were present at similar levels in 24-h-imbibed seeds. These results suggest that the ratio of eIF(iso)4E to eIF4E in the corresponding eIF4F complex is critical for the mechanisms of translational control during maize germination.

Several of the procedures necessary for cryopreservation of excised zygotic embryonic axes are known to be accompanied by emission of damaging levels of reactive oxygen species (ROS). These have been shown to be associated with shoot apical meristem necrosis, curtailing subsequent axis development to root production only, particularly for tropical/subtropical recalcitrant-seeded species. Here we report on the application of the principles of electrochemistry in the generation of strongly reducing, high pH cathodic water, by electrolysis of a solution containing calcium and magnesium chloride. The cathodic water in which Strychnos gerrardii axes were immersed for 30 min following dehydration, and importantly, after dehydration followed by cryopreservation, was shown to have strongly antioxidative properties in counteracting the damaging effects of ROS bursts and promoting shoot development. In a parallel experiment, axes of Boophane disticha exhibited enhanced total antioxidant activity when exposed to cathodic water both immediately following excision, and after flash-drying. For both species, the efficacious effects of cathodic treatment were manifest after the axes had been in culture for 4 h, suggesting that ROS were not quenched at source, but probably counteracted by enhancement of activity of endogenous antioxidants. Cathodic water therefore affords a non-toxic means of amelioration of oxidative, stress-related damage, which, coupled with the strongly fungicidal activity of the acidic, anionic water fraction, offers significant, and apparently non-injurious, advances towards successful cryopreservation of germplasm – and probably generally improved success of in-vitro-based procedures for plant tissues.

The potential longevity of japonica rice (Oryza sativa L. subsp. japonica) seed is particularly sensitive to high temperature – and thus climate change – during development and maturation. Cultivar Taipei 309 was grown at 28/20°C (12 h/12 h) and then from 19 DAA (days after 50% anthesis), when seeds were just over half filled, at 28/20°C, 30/22°C, 32/24°C or 34/26°C (12 h/12 h). Whereas ability to germinate ex planta had been achieved in almost all seeds by 24 DAA, only half the population were desiccation tolerant. Desiccation tolerance continued to increase over the subsequent 28 d, similarly at all four temperatures. Subsequent longevity, assessed by p50 (period in days to reduce viability to 50% in hermetic storage at 40°C with c. 15% moisture content), increased progressively at 28/20°C until 38 DAA, and remained constant until the final harvest (52 DAA). The three warmer temperature regimes provided similar longevity to 28/20°C at any one harvest, except at 38 DAA where the warmest (34/26°C) was poorer. That temperature regime also provided greater seed-to-seed variability within each survival curve. The results confirm that appreciable improvement in seed quality occurs during seed development and also subsequent maturation in japonica rice, but that increase in temperature from 28/20°C to 34/26°C during late seed filling onwards has comparatively little effect thereon. Comparison with previous investigations suggests that seed quality development may be less sensitive to high temperatures during late development and maturation than during the early seed development that precedes it.

The present study evaluated the size and germination characteristics of seeds of seven species of Syngonanthus (distributed among four sections) that occur in different microhabitats in campos rupestres vegetation in Minas Gerais State, to determine if (1) the germination responses of the various species characterize a single germination pattern for the genus; and (2) there is a correlation between the germination characteristics and the geographic distribution as well as the microhabitat of the different species. The experiments were undertaken with recently collected seeds exposed to a 12-h photoperiod and continuous darkness, at constant temperatures of 10–40°C (at intervals of 5°C) in germination chambers. The results indicated the existence of a pattern of reduced seed size and light requirements for germination within the genus. The geographic distribution of different species of the genus cannot be explained by the responses of seed germination to different temperatures, but suggests some relationship to their edaphic microhabitats. Therefore, germination characteristics of the populations studied may have been selected to colonize specific environments at different soil water conditions.

Legumes are crops that develop in cropping systems with relatively low inputs and are suitable to a more sustainable agriculture. Successful crop establishment, which is crucial for reliable plant production, depends on seed quality, environmental factors and genotypes. We studied pea and bean germination and seedling growth at various temperatures (5–40°C) and water potentials ( − 0.2 to − 1.5 MPa) using winter and spring pea and two common bean seeds produced in different conditions. The germination base temperature was − 1.1°C for pea seeds, and seeds of the winter genotype germinated more rapidly than those of the spring genotype. The base temperature for bean seed germination was 5.1–9.6°C, depending on the seed lot. The germination base water potential was about − 2 MPa for both species. The base temperatures for shoot elongation were higher (3–6°C) than those for germination. A review of the literature on other legumes confirmed that the differences in the responses of the legume seeds and seedlings to different temperatures were associated with their geographic origin. These results help understanding of pea and bean crop establishment, provide crop model parameter values and contribute to the search for genetic variability.

This study demonstrates that Anthemis chrysantha, a ‘Critically Endangered’ annual plant, produces two morphs of achenes: white and dark achenes, which differ in size, mass, anatomy and germination behaviour. Fresh white achenes germinated at all temperatures assayed from 10 to 25°C in both continuous darkness and 12-h photoperiod, ranging between 24% at 25°C in darkness and 89% at 12/20°C in light, whereas fresh dark achenes did not germinate under any temperature or light conditions. To identify differences in dormancy type between the two morphs, germination of dry-stored achenes, and achenes stratified at 5 or 25°C for 2 months were tested in both darkness and light at 5, 15 and 12/20°C for dry-stored and warm-stratified (25°C) achenes; and at 15, 25 and 12/20°C for cold-stratified (5°C) achenes. Of the white achenes, 90% germinated during the cold stratification period. In general, dry storage and warm stratification did not increase germination compared to fresh achenes. However, dark achenes did not germinate under any conditions. Dark achene dormancy was only broken by mechanical scarification or by excising the embryo (germination reached 71%). An anatomical study showed that the mesocarp of dark achenes had no intercellular spaces and was much thicker and stronger than that of white achenes, making the entry of water difficult, and also preventing germination by mechanical restriction. This study demonstrated that dormancy in the dark achenes is likely caused by the thickness of their pericarp, physically impeding germination and hampering imbibition of water.

Although mature seeds of the monogeneric conifer family Cephalotaxaceae sensu stricto have underdeveloped embryos, no definitive studies have been done to classify dormancy in this family. Our primary purpose was to determine the kind of dormancy in seeds of Cephalotaxus wilsoniana and to put the results into a broad phylogenetic context for gymnosperms. The species is of horticultural and medicinal value, and information is needed on how to propagate it efficiently from seeds. Embryo growth and germination were monitored for seeds at warm, cold and warm plus cold temperatures, and germination was monitored for seeds subjected to: (1) cold →  warm →  cold →  warm; and (2) warm →  cold →  warm →  cold →  warm temperature sequences. The effects of gibberellic acids GA3 and GA4 were tested on radicle emergence in ungerminated seeds and on shoot emergence in root-emerged seeds. Germination was promoted by ≥ 36 weeks of warm stratification followed by ≥ 8 weeks of cold stratification, but only if seeds were returned to high temperatures. The underdeveloped embryo must increase in length by >120% before the radicle emerges. Neither GA3 nor GA4 was effective in promoting radicle emergence; however, both plant growth regulators increased rate (but not percentage) of shoot emergence in root-emerged seeds. We conclude that seeds of C. wilsoniana have the deep simple level of morphophysiological dormancy (MPD), C1b-C3-B1b; thus, warm stratification followed by cold stratification and then warm-temperature incubation are required for germination. In gymnosperms, MPD is known in cycads, Ginkgo and now in three families of conifers.

A large fraction of tree species forming persistent soil seed-banks and with dormant seeds are expected to be found in strongly seasonal ecosystems such as Neotropical savannas, where seedling recruitment could be highly variable. In the savannas of Central Brazil, we studied seed characteristics (type of dormancy, longevity and moisture content) of 14 representative woody species differing in seed dispersal season. We also studied the dynamics of soil seed-banks and similarity patterns in woody species composition among seed rain, soil seed-bank, seedling bank and standing vegetation along shallow topographic gradients that differ in canopy cover. Woody species composition of the soil seed-bank largely differed from the standing vegetation, the seed rain and the seedling bank species composition, suggesting low recruitment of woody species from the soil seed-bank. Seeds of the 14 woody species remained viable for less than 16 months in laboratory dry-storage conditions. Of those, most seeds dispersed in the dry season were dormant and exhibited low moisture content, while most seeds dispersed in the wet season were non-dormant and exhibited high moisture content. Longevity of these seeds dispersed in the dry and the wet seasons did not differ significantly. This study shows that both timing of seed dispersal and dormancy appear to control timing of seed germination and seedling recruitment of most Neotropical savanna woody species, which did not form persistent soil seed-banks. This study contributes to the understanding of tree/grass coexistence and tree density variations along topographic gradients in tropical savannas.

The soil seed-bank is an important component of vegetation dynamics. Its presence affects both ecosystem resistance and resilience. A persistent seed-bank is especially important in disturbed habitats and harsh environments. In the hilly-gullied Loess Plateau region, serious soil erosion causes decreases in soil water capacity and constrains vegetation recolonization. A stable and long-term persistent soil seed-bank is necessary for natural vegetation recolonization. We used an integrated measure of the depth distribution of seeds in the soil and the seasonal dynamics of soil seed-banks to analyse the persistence of seeds in soil and to investigate the correlation of seed longevity with seed size/shape and the species' life history. The results showed a significant tendency for small seeds and seeds of annuals/biennials to persist longer in soil than large seeds and seeds of perennials. However, seed shape was not related to persistence. The main dominant speciesArtemisia scoparia, Lespedeza davurica, Heteropappus altaicus, Stipa bungeana, Artemisia gmelinii, and Bothriochloa ischaemun in the different successional stages in this region can form a persistent and stable soil seed-bank. The pioneer species A. scoparia is especially significant because it can form a large, long-term, persistent seed-bank. These species can play a role in the recolonization of the eroded abandoned slope lands by vegetation.

Whether and how seed mass and shape are related to seed persistence in soil is a controversial topic in plant ecology. There is little information on this relationship in sandy habitats. We tested whether or not this pattern is retained among 141 species within a range of growth forms, from a wide range of families, in the Horqin sandy land of northern China, which belongs to the inland sand ecosystem. We collected information on seed mass, shape and persistence of these species in this region from published articles. Seed mass and shape were found to be significantly and negatively correlated to persistence in the soil in the Chinese flora, in the same way as in most other floras examined, but not in the same way as the Israeli coastal sand dune flora. The value of seed mass and shape as good predictors of persistence in the soil could be applied to the Horqin sandy land. Although many factors affect the relationship between seed mass, shape and persistence, ease of burial appears to be the main potential interpretation in the Horqin sandy land.
