Dongxiang wild rice (Oryza rufipogon Griff.) (DXWR) is the northernmost distributed wild rice found in the world. Similar to other populations of O. rufipogon, DXWR contains a large number of agronomically valuable genes, which makes it a natural gene pool for rice breeding. Molecular markers, especially simple repeat sequence (SSR) markers, play important roles in plant breeding. Although a large number of SSR markers have been developed, most of them are derived from the genome coding sequences, rarely from non-coding sequences. Meanwhile, long non-coding RNAs (lncRNAs), which are derived from the transcription of non-coding sequences, play vital roles in plant growth, development and stress responses. In our previous study, we obtained 1655 lncRNA transcripts from DXWR using strand-specific RNA sequencing. In this study, 1878 SSR loci were detected from the lncRNA sequences of DXWR, and 1258 lncRNA-derived-SSR markers were developed on the genome-wide scale. To verify the validity and applicability of these markers, 72 pairs of primers were randomly selected to test 44 rice accessions. The results showed that 42 (58.33%) pairs of primers have abundant polymorphism among these rice materials; the polymorphism information content values ranged from 0.04 to 0.87 with an average of 0.50; the genetic diversity index of SSR loci varied from 0.04 to 0.88 with an average of 0.56; and the number of alleles per marker ranged from 2 to 11 with an average of 4.36. Thus, we concluded that these lncRNA-derived-SSR markers are a very useful source for future basic and applied research.

Okra (Abelmoschus esculentus L. Moench) is considered as a treasure house of nutrients and it is one of the major vegetables widely spread all over tropical, subtropical and warm temperate regions of the world. Yellow vein mosaic virus (YVMV) and enation leaf curl virus are the most destructive diseases of okra as they affect both crop growth and yield. Due to the frequent breakdown of resistance and lack of a stable source of resistance in the cultivated species, interspecific hybridization is considered as a reliable approach for durable resistance. Cultivated species from The United States Department of Agriculture and wild accessions from The National Bureau of Plant Genetic Resources were screened at YVMV hotspot (Guntur, Andhra Pradesh) to identify the potential donors for disease resistance. Accessions IC141032 and IC141012 were found to be free from both viruses and categorized as resistant lines. Interspecific hybridization between A. tetraphyllus and A. esculentus revealed a high crossability index of around 80% when A. esculentus was utilized as a female parent. The bottleneck of hybrid sterility was partially overcome by the colchicine treatment of interspecific F1 hybrids. Good seed set was observed when raw colchiploids were backcrossed to the recurrent parent.

Due to the high yield losses caused by late blight in potato cultivation, the development of resistant pre-breeding material is of great importance for cultivar breeding. The gene pool of the Julius Kühn Institute (JKI) includes a large collection of resistant clones whose resistance has not yet been analysed in detail with markers for relevant resistance genes. A panel of 52 pre-breeding potato clones developed at the JKI via interspecific crosses and highly resistant to late blight were tested for the presence of seven resistance genes (Rpi-blb1/Rpi-sto1, Rpi-blb2, Rpi-blb3/R2/Rpi-abpt, R1, R3a, R3b, Rpi-phu1) and one QTL allele (QTL_phu-stn) from Solanum species S. bulbocastanum, S. demissum, S. phureja and S. stoloniferum, respectively. Molecular marker assays based on sequence-specific primers revealed that 36 of the 52 pre-breeding clones carried either 1, 2, 3 or 4 resistance genes introgressed from these wild Solanum species. Results indicate that these resistance genes were retained over generations of breeding. Although highly resistant to late blight, 16 pre-breeding clones did not carry any of these resistance genes. Resistance in the gene pool may, thus, be based not only on individual resistance genes but also on QTL effects. Results help to better understand both inheritance and expression of late blight resistance of this unique gene pool and may be used for breeding programmes.

Big bud caused by several different phytoplasmas is an emerging threat to tomato production worldwide. The development of resistant varieties would be an effective approach to manage this problem, but it requires an appropriate screening technique. Recently, we have described a simple and efficient chip graft inoculation assay (CGIA) for the first time to screen tomato germplasm against Tomato leaf curl New Delhi virus. The present study was conducted to first validate the CGIA for phytoplasma transmission, then to assess the resistance of 74 genotypes belonging to different Solanum species against 16SrII-D phytoplasma. CGIA success rate and phytoplasma transmission was 100% since all the grafts survived and phytoplasma was detected in these plants using nested polymerase chain reaction. No genotype was found resistant as all the grafted plants showed typical disease symptoms. In addition to phytoplasma transmission, CGIA can be used for better understanding the plant–phytoplasma interactions and biology of phytoplasmas in tomato.

Seeds kept in long-term storage are essential for maintaining genetic resources of crops and other plant materials in seed banks and national germplasm systems. But seeds undergo chemical changes over time as part of the ageing process that result in reduced germination rates and seedling normalcy. For example, oilseed crops such as peanuts are particularly vulnerable to oxidation and rancidification due to their high oil content. To test the effect of storage time on seed oil in peanuts, we grew different accessions and harvested fresh seeds to compare the oil composition of new seeds with seeds aged in storage for varying lengths of time. Out of the nine fatty acids detected and measured by gas chromatography, five including oleic, gadoleic, behenic, lignoceric and cerotic acids differed significantly between new and old seeds. Additionally, old seeds accumulated up to four oxidation products together averaging about 1% of the total oil composition. The concentration of these oxidation products was positively correlated with the age of the seeds, increasing linearly up to more than 6% of the total in the oldest seeds. The presence and concentration of oxidation products measured using simple chromatography techniques can be used as an initial indication of quality and viability in older seed inventories stored in germplasm repositories.

Emmer wheat (Triticum turgidum ssp. dicoccum) is one of the most promising gene sources for drought tolerance improvement of durum wheat (Triticum turgidum ssp. durum). Achieving desired results requires a conscious choice of crossing parents based on general and specific combining ability (GCA and SCA) and also understanding the genes action involved in controlling the desired traits. In this study a 12 × 12 full diallel cross was performed using four emmer and eight durum wheats. The 132 hybrid progenies along with their parental lines were field evaluated under water-stressed and non-stressed conditions. Based on the Griffing diallel analysis both GCA and SCA effects were highly significant for all measured traits under both water treatments indicating possibility of improvement for drought tolerance. In this respect, the amount of additive effect was higher than the non-additive suggesting the chance for genetic advancement through selection. Based on Hayman's graphical analyses under the two water conditions it was revealed that several grain yield component traits were under the control of partial dominance. In contrary, grain yield and most morphological traits showed either dominance or over-dominance gene action. Grain yield had a significant positive correlation with the number of kernels per spike, kernel diameter, grain weight per spike and harvest index. These traits also had greater share of additive effects, relatively high narrow-sense heritability and high Baker ratio suggesting effective indirect selection for grain yield. Most durum × emmer hybrids had grain yield and drought tolerance indices better than the parents indicating that Iranian emmer wheats have a great genetic potential for drought tolerance improvement of durum wheat.
