Nine different clones of six species of Salix (Salix cordata Muhlenb. non Michaux, S. fragilis L., S. caprea L., S. cinerea L., S. burjatica Nazarov. and S. viminalis L.) and one hybrid (S.×calodendron Wimm.) were exposed to heavy metals in solution culture in an attempt to increase innate metal resistance. Resistance was estimated using comparative root measurements, and metal uptake was also studied. The first experiment entailed pretreatments with background nutrient solution, or 0·25 and 0·50 mg Cu l−1 amendments, and re-exposure to each of the same concentrations. In a second experiment clones were exposed to sub-toxic concentrations of single metals (0·15 mg Cu l−1, 0·15 mg Cd l−1 or 2·5 mg Zn l−1) and dual-combination treatments (0·075 mg Cu l−1+0·075 mg Cd l−1, 0·075 mg Cu l−1+1·25 mg Zn l−1 or 0·075 mg Cd l−1+1·25 mg Zn l−1)1 with concentrations gradually raised 10-fold over 128 d. Plants tested in the first experiment, following pre-exposure to Cu, were no more resistant to subsequent exposure to this metal. In the second experiment, gradual cumulative doses resulted in reduced phytotoxicity and increased resistance, most notably to Cd. There appeared to be an inverse relationship between metal uptake and resistance. Copper uptake was restricted to the roots, whereas Cd and Zn were more evenly distributed throughout the plant. Exposure to dual combinations of metals resulted in several interaction effects on uptake: increased root-bound Cu in all combinations, and the increase in uptake of both Cd and Zn into the root tissues when supplied with Cu. The implications of these results for the use of willows in phytoremediation programmes are discussed.

Plants from metalliferous soils often exhibit combined tolerance to different heavy metals. Such tolerance could rely either on the possession of a combination of different metal-specific tolerance mechanisms (‘multiple tolerance’), or on less specific mechanisms that pleiotropically confer tolerance to different metals (‘co-tolerance’). In this study, we examined the co-segregation of tolerances to Cu, Zn, Ni, Co and Cd in crosses between distinctly tolerant ecotypes of Silene vulgaris (Moench) Garcke. The results demonstrate non-pleiotropic genetic control of tolerance to Cu, Zn and Cd. Tolerance to Ni and Co, on the other hand, seems to represent the pleiotropic by-product of the tolerance allele of one particular locus for zinc tolerance.

Quasi-steady-state measurements of root hydraulic conductance (KR) of Olea oleaster Hoffmgg. et Link potted seedlings were performed using a pressure chamber with the aim of: (a) measuring the impact of different water-stress levels on a KR; (b) measuring the kinetics of KR recovery several days after soil rewetting; (c) relating changes in KR to changes in root anatomy and morphology. Increasing water-stress was applied in terms of ratio of leaf water potential (ΨL) measured at midday to that at zero turgor (ΨTLP), i.e. ΨL/ΨTLP=0·5, 1·0, 1·2, 1·6; KR was measured initially and at 24, 48, 72, 96 h after irrigation.

Values of KR in seedlings stressed to ΨL/ΨTLP=1·2 increased for 48 h after irrigation from 0·23 to 0·97×10−5 kg s−1 m−2 MPa−1 i.e. from 16% to 66% of that measured in unstressed seedlings. A marked shift of the x-axis intercept of the straight line relating flow to pressure (zero flow at non-zero pressure) was recorded initially after irrigation and persisted up to 48 h. Recovery of KR occurred within 24 h after irrigation in seedlings at ΨL/ΨTLP=0·5 and 48 h later in those at ΨL/ΨTLP=1·0.

Severe drought stress (ΨL/ΨTLP=1·6) caused anatomical changes to roots which formed a two-layered exodermis with thicker suberized walls and a three- to four-layered endodermis with completely suberized tangential walls. Recovery of KR in these roots required resumed growth of root tips and emergence of new lateral roots.

Seedlings of the annual grass Vulpia ciliata ssp. ambigua were grown in sterilized sand with a dark septate root fungus, Phialophora graminicola, which had been isolated from a natural population of the grass. Tiller number and shoot, root and total biomass of seedlings grown with P. graminicola were enhanced relative to uninoculated control plants in a growth room and a glasshouse experiment. Root length of seedlings grown with P. graminicola was significantly increased, but no effects of the fungus on root diameter, number of root hairs or specific root length were recorded. Root nitrogen content and shoot, root and total phosphorus contents of seedlings grown with the fungus were enhanced, but shoot nitrogen concentration of these plants was reduced. Shoot biomass and specific root length of inoculated plants were positively associated with the number of P. graminicola colonies re-isolated from roots. These data indicate that P. graminicola acts as a beneficial associate of V. ciliata ssp. ambigua seedlings under controlled conditions.

Emissions of reactive oxidized nitrogen (NO and NO2), collectively known as NOx, from human activities are c. 21 Tg N annually, or 70% of global total emissions. They occur predominantly in industrialized regions, largely from fossil fuel combustion, but also from increased use of N fertilizers. Soil emissions of NO not only make an important contribution to global totals, but also play a part in regulating the dry deposition of NO and NO2 (NOx) to plant canopies. Soil microbial production of NO leads to a soil ‘compensation point’ for NO deposition or emission, which depends on soil temperature, N and water status. In warm conditions, the net emission of NOx from plant canopies contributes to the photochemical formation of ozone. Moreover, the effect of NOx emissions from soil is to reduce net rates of NO2 deposition to terrestrial surfaces over large areas.

Increasing anthropogenic emissions of NOx have led to an approximate doubling in surface O3 concentrations since the last century. NOx acts as a catalyst for the production of O3 from volatile organic compounds (VOCs). Paradoxically, emission controls on motor vehicles might lead to increases in O3 concentrations in urban areas.

Removal of NO and NO2 by dry deposition is regulated to some extent by soil production of NO; the major sink for NO2 is stomatal uptake. Long-term flux measurements over moorland in Scotland show very small deposition rates for NO2 at night and before mid-day of 1–4 ng NO2-N m−2 s−1, and similar emission rates during afternoon. The bi-directional flux gives 24-h average deposition velocities of only 1–2 mm s−1, and implies a long life-time for NOx due to removal by dry deposition.

Rates of removal of O3 at the ground are also influenced by stomatal uptake, but significant non-stomatal uptake occurs at night and in winter. Measurements above moorland showed 40% of total annual flux was stomatal, with 60% non-stomatal, giving nocturnal and winter deposition velocities of 2–3 mm s−1 and daytime summer values of 10 mm s−1. The stomatal uptake is responsible for adverse effects on vegetation. The critical level for O3 exposure (AOT40) is used to derive a threshold O3 stomatal flux for wheat of 0·5 μg m−2 s−1. Use of modelled stomatal fluxes rather than exposure might give more reliable estimates of yield loss; preliminary calculations suggest that the relative grain yield reduction (%) can be estimated as 38 times the stomatal ozone flux (g m−2) above the threshold, summed over the growing season.

Low phosphorus availability is often a primary constraint to plant productivity in native soils. Here we test the hypothesis that root carbon costs are a primary limitation to plant growth in low P soils by assessing the effect of P availability and mycorrhizal infection on whole plant C budgets in common bean (Phaseolus vulgaris L.). Plants were grown in solid-phase-buffered silica sand providing a constant supply of low (1 μm) or moderate (10 μm) P. Carbon budgets were determined weekly during the vegetative growth phase. Mycorrhizal infection in low-P plants increased the root specific P absorption rate, but a concurrent increase in root respiration consumed the increased net C gain resulting from greater P uptake. The energy content of mycorrhizal and non-mycorrhizal roots was similar. We propose that the increase in root respiration in mycorrhizal roots was mainly due to increased maintenance and growth respiration of the fungal tissue. Plants grown with low P availability expended a significantly larger fraction of their total daily C budget on below-ground respiration at days 21, 28 and 35 after planting (29–40%) compared with plants grown with moderate P supply (18–25%). Relatively greater below-ground respiration in low P plants was mainly a result of their increased root[ratio ]shoot ratio, although specific assimilation rate was reduced significantly at days 21 and 28 after planting. Specific root respiration was reduced over time by low P availability, by up to 40%. This reduction in specific root respiration was due to a reduction in ion uptake respiration and growth respiration, whereas maintenance respiration was increased in low-P plants. Our results support the hypothesis that root C costs are a primary limitation to plant growth in low-P soils.

We examined the roles that seasonal shifts in precipitation and temperature played in the ectomycorrhizal (ECM) colonization of pinyon pine (Pinus edulis Engelm.) at two contrasting sites in northern Arizona. Pinyons growing in ash and cinder soils experienced much greater water and nutrient stress than pinyons growing nearby in sandy-loam soils. Over a one year period, we obtained monthly measurements of ECM colonization, root zone soil moisture and temperature, and air temperature and precipitation. Four major patterns emerged. Firstly, although climate as measured by ambient temperature and precipitation did not vary between the two sites, soil temperature was significantly higher and soil moisture significantly lower at the cinder site than at the sandy-loam site. Secondly, ECM colonization was significantly higher at the cinder site for 5 of 12 months. Thirdly, although nearly 70% of the variation in ECM colonization of pinyons growing in cinder soil was predicted by a combination of soil moisture and soil temperature, these same variables had little predictive power for pinyons growing in sandy-loam soils. Air temperature and precipitation were also significantly correlated with ECM colonization at the cinder site but not the sandy-loam site. Fourthly, a watering experiment showed that ECM colonization significantly increased with supplemental water at the cinder site, but not at the sandy-loam site. Thus, in two sites that did not differ in plant community or climate, ectomycorrhizas in cinder soils were far more sensitive to changes in moisture and temperature than ectomycorrhizas in sandy-loam soils.

The metabolism of chlorophylls and related tetrapyrroles directly influences, and is influenced by, the proteins and cell structures with which they are associated. During net accumulation, de-greening and at the steady state, chlorophyll and its derivatives are important elements in the post-translational regulation of the expression of genes for chloroplast proteins. At the same time, they represent potential photodynamic hazards against which green cells need to have protective mechanisms. This review deals with genetic, chemical and environmental perturbations of chlorophyll biosynthesis that impact on protein stability, membrane organization and susceptibility to photodamage. NADPH-protochlorophyllide oxidoreductase is considered in detail as a pigment-protein regulating, and regulated by, chlorophyll metabolism. The question of the extent and significance of chlorophyll turnover at the steady state is addressed, with particular emphasis on the dynamics of the photosystem II reaction centre. The pathway of chlorophyll catabolism is described, along with its interrelationship with protein mobilization in chloroplast senescence. Finally, the structural basis of pigment–protein interaction and stability is examined, and the discussion ends by expressing some general thoughts about the control of protein lifetimes in the living cell.

Uptake, translocation and metabolism of 14C-labelled formaldehyde in the leaves of Epipremnum aureum (Golden Potho) and Ficus benjamina (Weeping Fig) were investigated. Plants were exposed in light and dark to 14C-formaldehyde (500 μg m−3) in gas exposure chambers. The amount of 14C-incorporation into the soluble (water-extractable) and insoluble fractions of leaves, stem sections and roots was determined. The soluble 14C-activity was fractionated by ion exchange chromatography followed by thin-layer chromatography/autoradiography. Approximately 60–70% of the applied 14C-formaldehyde was recovered from the plants. In the light about five times more 14C-formaldehyde was assimilated than in the dark. The amount of 14C-label derived from 14C-formaldehyde, which was incorporated into acid-stable metabolites, was enhanced to an even larger extent in the light. The 14C-activity pattern closely resembled the general labelling spectrum of photosynthates, obtained after a 14CO2 exposure. A substantial amount of labelled material, mostly sucrose, was translocated into the stems and roots. Our results suggest that in the light 14C enters the Calvin cycle after an enzymatic two-step oxidation process of 14C-formaldehyde to 14CO2. The activities of the respective enzymes, formaldehyde dehydrogenase and formate dehydrogenase, were determined. Among 27 ‘leafy’ indoor decorative plants, a screening experiment revealed no outstanding species with regard to its capacity for metabolism of formaldehyde, and rate of uptake through stomata was too low to justify claims that plants contribute usefully to indoor air purification.

Among closely related citrus genotypes growing in high phosphorus (P) orchard soils, there is a tendency for less mycorrhizal-dependent (M-dependent) species to have lower rates of root colonization than more M-dependent species. We hypothesized that the less M-dependent the citrus species the more limited is their carbohydrate (CHO) allocation to the M fungus. In a glasshouse study at low and high P supply, lower total incidence of Glomus intraradices FL 208, intensity of vesicles formation, and accumulation of 16[ratio ]1W5cis fungal fatty acid as measures of root colonization were related to lower mycorrhizal dependency (MD; M plant d. wt/non-mycorrhizal (NM) plant d. wt expressed at low P supply) in five citrus genotypes. At high P supply, when host carbon (C) production was not affected by P nutrition, less M-dependent genotypes had consistently lower starch concentrations in their root and shoot tissues than did more M-dependent genotypes, irrespective of M inoculation. At low P supply, M plants were more heavily colonized by G. intraradices and had lower starch levels than the M plants supplied with additional P. At high P, M plants of the more dependent genotypes allocated more C to starch pools relative to the NM plant than the least dependent genotype. Concentration of sucrose in tissues did not vary consistently with the dependency of citrus genotypes or with M inoculation except in high P NM plants, when less M-dependent genotypes had lower levels of sucrose in their roots than the more dependent species. At high P supply, sucrose concentrations were lower in colonized roots than in NM roots. Across citrus genotypes, sucrose in M fibrous roots decreased relative to that in NM roots with increase in root colonization suggesting that more sucrose was allocated for growth and maintenance of G. intraradices in roots of M-dependent species. The concentration of reducing sugars in root tissues varied in relation to MD of citrus genotypes in the same way as that of starch and sucrose, but was less responsive to M colonization. Responses of total non-structural CHO in tissues indicated that more heavily colonized plants at low P expended more C to acquire P than did M plants of similar biomass and P status grown at high P supply. Total CHO pools increased with MD of citrus genotypes, providing evidence than C allocation patterns in the host affect M colonization.

Cloned cuttings of Betula pendula Roth were grown in field fumigation chambers at Birmensdorf throughout one growing season in filtered air with either <3 (control) or 90/40 nl l−1 O3 (day/night; ozone generated from pure oxygen). Each ozone regime was split into high and low soil nutrient regimes by watering plants with either a 0·05% or a 0·005% solution of a fertilizer which contained macronutrients and micronutrients.

Fertilization had a strong effect on plant growth, enzyme activities and the expression of ozone-induced effects at the biochemical level. The activities of PEPC and Rubisco were enhanced about threefold in the plants with high fertilization (HF). Significant effects of ozone were in most cases found only in the older leaves of the plants with low fertilization (LF). There, sucrose, glucose and fructose levels were enhanced. In both fertilization treatments, the number of starch granules along the minor veins was increased. These ozone effects point to a decreased or inhibited phloem loading. The increased PEPC activity and the enhanced malate levels in the ozone-exposed plants might be the result of a redirection of carbon flow from sucrose synthesis and translocation towards anapleurotic processes, which can feed detoxification and repair of ozone injury as indicated by enhanced respiration. These findings agree well with the observed effects of ozone in lowering the root[ratio ]shoot biomass ratio. Although there was a marked reduction in the O3/LF plants, O3/HF plants showed no significant response. Inositol was decreased under ozone exposure in both fertilizer treatments, contrasting with the pattern for carbohydrates.

These results demonstrate the role of fertilization as an important modifier of ozone-induced effects at the plant biochemical level. Well fertilized plants appear to cope better with the impact of ozone on metabolism.

The relationships between leaf structure, nitrogen concentration and CO2 assimilation rate (A) were studied for 14 grass species grown in the laboratory under non-limiting nutrient conditions. Structural features included leaf thickness and density, and the proportion of leaf volume occupied by different types of tissue (mesophyll, epidermis, vessels and sclerenchyma). Relationships were assessed for data expressed per unit leaf area and fresh mass. The latter was found to be closely related to leaf volume, which allowed us to use A per unit leaf fresh mass (Afm) as a surrogate of A per unit leaf volume. Assimilation rate per unit leaf area (Aa) was positively correlated with leaf thickness and with the amount of mesophyll per unit leaf area; the relationship with leaf nitrogen content per unit area was only marginally significant. Afm was negatively correlated with leaf thickness and positively with fresh mass-based leaf organic nitrogen concentration. A multiple regression involving these two variables explained 81% of the variance in Afm. The value of Afm was also significantly related to the proportion of mesophyll in the leaf volume, but surprisingly the correlation was negative. This was because thin leaves with high Afm and nitrogen concentration had proportionally more mechanically supportive tissues than thick ones; as a consequence, they also had a lower proportion of mesophyll. These data suggest that, in addition to leaf nitrogen, leaf thickness has a strong impact on CO2 assimilation rate for the grass species studied.

Dry deposition of NH3 and NOx (NO and NO2) can affect plant metabolism at the cellular and whole-plant level. Gaseous pollutants enter the plant mainly through the stomata, and once in the apoplast NH3 dissolves to form NH4+, whereas NO2 dissolves to form NO3− and NO2−. The latter compound can also be formed after exposure to NO. There is evidence that NH3-N and NOx-N can be reversibly stored in the apoplast. Temporary storage might affect processes such as absorption rate, assimilation and re-emission. Once formed, NO3− and NO2− can be reduced, and NH4+ can be assimilated via the normal enzymatic pathways, nitrate reductase (NR), nitrite reductase and the glutamine synthetase/glutamate synthase (GS/GOGAT) cycle. Fumigation with low concentrations of atmospheric NH3 increases in vitro glutamine synthetase activity, but whether this involves both or only one of the GS isoforms is still an open question. There seems to be no correlation between fumigation with low concentrations of NH3 and in vitro GDH activity. The contribution of atmospheric NH3 and NO2 deposition to the N budget of the whole plant has been calculated for various atmospheric pollutant concentrations and relative growth rates (RGRs). It is concluded that at current ambient atmospheric N concentrations the direct impact of gaseous N uptake by foliage on plant growth is generally small.

Availability of nitrogen (N) and phosphorus (P) might significantly affect N2 fixation in legumes. The interaction of N and P was studied in common bean (Phaseolus vulgaris), considering their effects on nodulation and N2 fixation, nitrate reductase activity, and the composition of N compounds in xylem sap. The effect of N on the uptake of P by plants was estimated by analysing rhizospheric pH and P concentration in xylem sap and in plant shoots. Inoculated bean plants were grown in pots containing perlite/vermiculite in two experiments with different amounts of P and N. In a third experiment, bean plants were grown on two soil types or on river sand supplied with different concentrations of N. At harvest, shoot growth, number of nodules and mass, and nitrogenase activity were determined. Xylem sap was collected for the determination of ureides, amino acids, nitrate and phosphate concentration. At low nitrate concentration (1 mM), increasing amounts of P promoted both nodule formation and N2 fixation, measured as ureide content in the xylem sap. However, at high nitrate concentration (10 mM), nodulation and N2 fixation did not improve with increased P supply. Glutamine and aspartate were the main organic N compounds transported in the xylem sap of plants grown in low nitrate, whereas asparagine was the dominant N compound in xylem sap from plants grown in high nitrate. Nitrate reductase activity in roots was higher than in shoots of plants grown with low P and high N. In both soils and in the sand experiment, increased application of N decreased nodule mass and number, nitrogenase activity and xylem ureides but increased the concentration of asparagine in xylem sap. Increasing P nutrition improved symbiotic N2 fixation in bean only at low N concentrations. It did not alleviate the inhibitory effect of high nitrate concentration on N2 fixation. A decrease in plant P uptake was observed, as indicated by a lower concentration of P in the xylem sap and shoots, correlating with the amount of N supplied. Simultaneously with the specific inhibition of N2 fixation, high nitrate concentrations might decrease P availability, thus inhibiting even further the symbiotic association because of the high P requirement for nodulation and N2 fixation.

The seasonal patterns of xylem embolism and xylem transport properties in Quercus pubescens Willd. and Quercus ilex L. trees growing in a natural mixed coppice stand in conditions of severe water stress were investigated. Xylem embolism was evaluated in both dehydrating branches and in apical twigs during a whole year. Measurements of xylem water potential were conducted from predawn to sunset on selected sunny days. On the same days, diurnal courses of leaf conductance were monitored. Measurements of half-hourly sap flow were made by the heat-pulse technique throughout the summer. At the onset of summer, a sharp decrease in water potential was observed in both species. Full recovery of water potentials was observed for both species after the first major rainfall event in September. Both experienced serious embolism throughout the year, ranging between minima of c. 60% (expressed as percentage loss of hydraulic conductivity) after the rains in autumn and after bud burst in spring, and maxima of c. 80% during summer and after freezing-thawing events during the winter season. A significant negative linear relationship was found between water potential and xylem embolism in branches dehydrating in air for Q. pubescens and Q. ilex. Q. pubescens had greater efficiency in hydraulic transport (higher specific conductivity and leaf specific conductivity) by the xylem than Q. ilex. In June, leaf conductance was high early in the morning and decreased gradually during the day. Midday depression of leaf conductance, as a result of high evaporative demand combined with water deficit, was observed in both species. In August, leaf conductance of both species was greatly reduced, as water potential dropped to extremely low values, and the stomata were almost completely closed during the afternoon. No hysteresis resulting from plant capacitance was observed in the relationship between shoot water potential and sap flow. Q. pubescens exhibited very high values of whole-tree hydraulic resistance between July and September, whereas Q. ilex generally showed lower values. The effect of soil moisture depletion on the relationship between sap flow and shoot water potential appears as a lowering of water potential at zero flow. A significant decrease of whole-tree hydraulic resistance in both species was observed with the onset of the autumn, preceding the partial recovery of twig hydraulic conductivity. The results demonstrate that both Q. pubescens and Q. ilex, although highly tolerant of severe water stress and tissue dehydration, operate at the limits of safety which are surpassed under severe droughts, and prolonged climatic stress might predispose these Quercus species to decline.

We report a survey of four viruses (beet western yellows luteovirus (BWYV), cauliflower mosaic caulimovirus (CaMV), turnip mosaic potyvirus (TuMV), turnip yellow mosaic tymovirus (TYMV)) in five natural populations of Brassica oleracea in Dorset (UK). All four viruses were common; 43% of plants were infected with BWYV, 60% with CaMV, 43% with TuMV and 18% with TYMV. For each virus there were significant differences in the proportion of infected plants among populations, which were not completely explained by differences in the age of plants. Multiple virus infections were prevalent, with 54% of plants having two or more virus types. There were statistically significant associations between pairs of viruses. The CaMV was positively associated with the other three viruses, and BWYV was also positively associated with TuMV. There was no detectable association between BWYV and TYMV, whereas TuMV and TYMV were negatively associated. We suggest these associations result from BWYV, CaMV and TuMV having aphid vectors in common, as aphids are attracted to plants that already have a virus infection. Infected plants were distributed randomly or were very weakly aggregated within populations. The implications of widespread multiple virus infections in natural plant populations are discussed with respect to the release of transgenic plants expressing virus-derived genes.

In vesicular–arbuscular mycorrhizal symbioses, specialized fungal structures (the arbuscules) are formed which are in intimate contact with plant root cortical cells. It is assumed that these arbuscules are the major sites of solute transfer between the plant and fungus, but there have been no studies that definitively show the extent or types of transfer processes that occur in this structure. Phosphate is one of the major nutrients that is acquired by mycorrhizal fungi and transferred to plants. In this study a single Lycopersicon esculentum cDNA was cloned and shown to be identical to LePT1, a previously cloned inorganic-phosphate transporter. Expression studies revealed that LePT1 transcript levels remained constant in mycorrhizal plants, but increased in phosphate-starved, non-mycorrhizal plants. Localization of the LePT1 transcript by in situ hybridization showed that this gene is highly expressed in arbuscule-containing cortical cells in mycorrhizal plants. In non-mycorrhizal plants LePT1 expression was localized to the stele and cortex. The expression studies suggest that this transporter is involved in phosphate nutrition of L. esculentum and its localization in cells that contain arbuscules indicate that it may be the mechanism used by the plant to take up phosphate that is effluxed across the fungal plasma membrane of the arbuscule. Based on our findings and those of others, an integrated model of inorganic phosphate uptake and transfer in mycorrhizal and non-mycorrhizal plants is presented.

The effect of temperature on germination rate was assessed for seeds of 31 wild plant species and four cultivated species growing in the UK. The temperature at which seed first germinated ranged from 2 °C (Brassica rapa) to 11 °C (several species). As the temperature progressively increased, the percentage of seed that germinated rapidly increased to near 100%, whereas the duration for germination progressively decreased up to the thermal optimum. Above the thermal optimum, the duration for germination initially remained constant, and then rapidly increased until the thermal maximum was exceeded when no seed germinated. To assess the effect of temperature on germination rates, the reciprocal of the duration (1/duration) for 50% germination was calculated and regressed against temperature. For most species a linear regression accounted for >90% of the variation in germination rates for temperatures up to the thermal optimum. From the regressions, the base temperatures (Tb, below which no germination is expected) and the thermal constant (S, expressed in degree days) for 50% germination were calculated. The extremes were the alpine species Dryas octopetala, which had a high Tb (c. 12.0 °C) and low S (26 degree days), and Saxifraga tridactylites which had a low estimated Tb (c. −1.8 °C) and a high S (136 degree days). The estimated value of S increased with increasing percentage germination, and although S varied considerably between species with similar values of Tb, a weak inverse relationship between Tb and S was indicated. A thermal time approach was shown to be useful when analysing and comparing thermal requirements for germination of the seed of wild plant species. It showed, for example, that percentage germination often decreased as temperatures approached Tb, and that for some seed germination ceased at temperatures well above Tb.

The dynamic-chamber technique was used to investigate the correlation between NH3 and NO2 fluxes and different climatic and physiological parameters: air temperature; relative air humidity; photosynthetic photon fluence rate; NH3 and NO2 concentrations; transpiration rate; leaf conductance for water vapour; and photosynthetic activity. The experiments were performed with twigs from the sun crown of mature beech trees (Fagus sylvatica) at a field site (Höglwald, Germany), and with 12-wk-old beech seedlings under controlled conditions. Both sets of experiments showed that NO2 and NH3 fluxes depended linearly on NO2 and NH3 concentration, respectively, in the concentration ranges representative for the field site studied, and on water-vapour conductance as a measure for stomatal aperture. The NO2 compensation point determined in the field studies (the atmospheric NO2 concentration with no net NO2 flux) was 1.8–1.9 nmol mol−1. The NH3 compensation point varied between 3.3 and 3.5 nmol mol−1 in the field experiments, and was 3.0 nmol mol−1 in the experiments under controlled conditions. The climatic factors T and PPFR were found to influence both NO2 and NH3 fluxes indirectly, by changing stomatal conductance. Whilst NO2 flux showed a response to changing relative humidity that could be explained by altered stomatal conductance, increased NH3 flux with increasing relative humidity (>50%) depended on other factors. The exchange of NO2 between above-ground parts of beech trees and the atmosphere could be explained exclusively by uptake or emission of NO2 through the stomata, as indicated by the quotient between measured and predicted NO2 conductance of approx. 1 under all environmental conditions examined. Neither internal mesophyll resistances nor additional sinks could be observed for adult trees or for beech seedlings. By contrast, the patterns of NH3 flux could not be explained by an exclusive exchange of NH3 through the stomata. Deposition into additional sinks on the leaf surface, as indicated by an increase in the quotient between measured and predicted NH3 conductance, gained importance in high air humidity, when the stomata were closed or nearly closed and/or when atmospheric NH3 concentrations were high. Although patterns of NH3 gas exchange did not differ between different months or years at high NH3 concentrations (c. 140 nmol mol−1), it must be assumed that emission or deposition fluxes at low ambient NH3 concentration (0.8 and 4.5 nmol mol−1) might vary significantly with time because of variation in the NH3 compensation point.

The changes in pigment composition, photosynthesis and PSII photochemistry were investigated in cells of Spirulina platensis adapted to salt stress (<0.75 M NaCl). A decrease in the phycocyanine/chlorophyll and no significant change in the carotenoid/chlorophyll ratio were observed in salt-adapted cells. Salt stress inhibited the apparent quantum efficiency of photosynthesis and PSII activity while stimulating PSI activity and dark respiration significantly. Salt stress also resulted in a decrease in overall activity of the electron transport chain, which could not be restored by diphenylcarbazide, an artificial electron donor to the reaction centres of PSII. Measurements of the polyphasic fluorescence rise in fluorescence transients including phases O, J, I and P showed that salt stress had no effect on the fluorescence yield at phase O but decreased the fluorescence yield at phases J, I and P. Analyses of the JIP test developed from the polyphasic rise of fluorescence transients showed that salt stress led to a decrease in both the maximum quantum efficiency of PSII photochemistry and the maximum quantum efficiency of electron transport beyond the primary quinone electron acceptor. However, salt stress induced no significant changes in the probability of transporting an electron beyond QA, the trapping flux per PSII reaction centre, or the electron transport flux per PSII reaction centre. A theoretical analysis of fluorescence parameters indicated a decrease in the rate constant of excitation energy trapping by PSII reaction centres. In addition, salt stress induced an increase in the complementary area above the fluorescence induction curve in the presence of 3-(3,4-dichlorophenyl)-1,1-dimethylurea, suggesting an increase in the proportion of closed PSII reaction centres in salt-adapted cells. Based on these results, it is suggested that modifications in PSII photochemistry in salt-adapted Spirulina cells maintained a high conversion efficiency of excitation energy, such that no significant change was observed in either the trapping flux or the electron transport flux per PSII reaction centre.
