Many mechanisms, functions and structures of life have been unraveled. However, the fundamental driving force that propelled chemical evolution and led to life has remained obscure. The second law of thermodynamics, written as an equation of motion, reveals that elemental abiotic matter evolves from the equilibrium via chemical reactions that couple to external energy towards complex biotic non-equilibrium systems. Each time a new mechanism of energy transduction emerges, e.g., by random variation in syntheses, evolution prompts by punctuation and settles to a stasis when the accessed free energy has been consumed. The evolutionary course towards an increasingly larger energy transduction system accumulates a diversity of energy transduction mechanisms, i.e. species. The rate of entropy increase is identified as the fitness criterion among the diverse mechanisms, which places the theory of evolution by natural selection on the fundamental thermodynamic principle with no demarcation line between inanimate and animate.

Reaction-diffusion equations based on a polymerization model are solved to simulate the spreading of hypothetic left and right-handed life forms on the Earth's surface. The equations exhibit front-like behavior as is familiar from the theory of the spreading of epidemics. It is shown that the relevant time scale for achieving global homochirality is not, however, the time scale of front propagation, but the much longer global diffusion time. The process can be sped up by turbulence and large scale flows. It is speculated that, if the deep layers of the early ocean were sufficiently quiescent, there may have been the possibility of competing early life forms with opposite handedness.

It has long been assumed that the planet Jupiter acts as a giant shield, significantly lowering the impact rate of minor bodies upon the Earth, and thus enabling the development and evolution of life in a collisional environment which is not overly hostile. In other words, it is thought that, thanks to Jupiter, mass extinctions have been sufficiently infrequent that the biosphere has been able to diversify and prosper. However, in the past, little work has been carried out to examine the validity of this idea. In the second of a series of papers, we examine the degree to which the impact risk resulting from objects on Centaur-like orbits is affected by the presence of a giant planet, in an attempt to fully understand the impact regime under which life on Earth has developed. The Centaurs are a population of ice-rich bodies which move on dynamically unstable orbits in the outer Solar system. The largest Centaurs known are several hundred kilometres in diameter, and it is certain that a great number of kilometre or sub-kilometre sized Centaurs still await discovery. These objects move on orbits which bring them closer to the Sun than Neptune, although they remain beyond the orbit of Jupiter at all times, and have their origins in the vast reservoir of debris known as the Edgeworth–Kuiper belt that extends beyond Neptune. Over time, the giant planets perturb the Centaurs, sending a significant fraction into the inner Solar System where they become visible as short-period comets. In this work, we obtain results which show that the presence of a giant planet can act to significantly change the impact rate of short-period comets on the Earth, and that such planets often actually increase the impact flux greatly over that which would be expected were a giant planet not present.

One of the most fundamental questions in exoplanetology is to determine whether a given planet is habitable. We estimate the relative likelihood of a planet's propensity towards habitability by considering key physical characteristics such as the role of temperature on ecological and evolutionary processes, and atmospheric losses via hydrodynamic escape and stellar wind erosion. From our analysis, we demonstrate that Earth-sized exoplanets in the habitable zone around M-dwarfs seemingly display much lower prospects of being habitable relative to Earth, owing to the higher incident ultraviolet fluxes and closer distances to the host star. We illustrate our results by specifically computing the likelihood (of supporting life) for the recently discovered exoplanets, Proxima b and TRAPPIST-1e, which we find to be several orders of magnitude smaller than that of Earth.

Two thermodynamic principles offer considerable insight into the climatic and geological settings for life on other planets, namely (1) that natural systems tend to actually achieve the ideal (‘Carnot’) limit of conversion of heat into work and (2) if a fluid system such as an atmosphere has sufficient degrees of freedom, it will choose the degree of heat transport that maximizes the generation of work (equivalently, that which offers maximum entropy production). The first principle agrees well with results on terrestrial cumulus convection, and the mechanical energy released by tectonic activity. The second principle agrees with the observed zonal climates of Earth, Mars and Titan, and shows promise for planetary interiors too; I discuss applications in the investigation of paleoclimates and habitability. I compare the work performed by planetary atmospheres and interiors on the terrestrial planets and thereby predict a weakly eroded landscape on Titan. The association of life with the production of entropy is also noted, and the possibility of evaluating planetary entropy production by telescopic observation is discussed.

Humankind's innate curiosity makes us wonder whether life is or was present on other planetary bodies such as Mars. The EuroGeoMars 2009 campaign was organized at the Mars Desert Research Station (MDRS) to perform multidisciplinary astrobiology research. MDRS in southeast Utah is situated in a cold arid desert with mineralogy and erosion processes comparable to those on Mars. Insight into the microbial community composition of this terrestrial Mars analogue provides essential information for the search for life on Mars: including sampling and life detection methodology optimization and what kind of organisms to expect. Soil samples were collected from different locations. Culture-independent molecular analyses directed at ribosomal RNA genes revealed the presence of all three domains of life (Archaea, Bacteria and Eukarya), but these were not detected in all samples. Spiking experiments revealed that this appears to relate to low DNA recovery, due to adsorption or degradation. Bacteria were most frequently detected and showed high alpha- and beta-diversity. Members of the Actinobacteria, Proteobacteria, Bacteroidetes and Gemmatimonadetes phyla were found in the majority of samples. Archaea alpha- and beta-diversity was very low. For Eukarya, a diverse range of organisms was identified, such as fungi, green algae and several phyla of Protozoa. Phylogenetic analysis revealed an extraordinary variety of putative extremophiles, mainly Bacteria but also Archaea and Eukarya. These comprised radioresistant, endolithic, chasmolithic, xerophilic, hypolithic, thermophilic, thermoacidophilic, psychrophilic, halophilic, haloalkaliphilic and alkaliphilic micro-organisms. Overall, our data revealed large difference in occurrence and diversity over short distances, indicating the need for high-sampling frequency at similar sites. DNA extraction methods need to be optimized to improve extraction efficiencies.

Nitrogen is an essential element for life. Specifically, fixed nitrogen (i.e. NH3, NH4+, NOx or N that is chemically bound to either inorganic or organic molecules and can be released by hydrolysis to form NH3 or NH4+) is useful to living organisms. Nitrogen on present-day Mars has been analysed only in the atmosphere. The inventory is a small fraction of the amount of nitrogen presumed to have been received by the planet during its accretion. Where is the missing nitrogen? Answering this question is crucial for understanding the probability of the origin and evolution of life on Mars, and for its future astrobiological exploration. The two main processes that could have removed nitrogen from the atmosphere include: (1) non-thermal escape of N atoms to space and (2) burial within the regolith as nitrates and ammonium salts. Nitrate would probably be stable in the highly oxidized surface soil of Mars and could have served as an NO3− sink. Such accumulations are observed in certain desert environments on Earth. Some NH4+ nitrogen may also be fixed and stabilized in the soil by inclusion as a structural cation in the crystal lattices of certain phyllosilicates replacing K+. Analysis of the Martian soil for traces of NO3− and NH4+ during future missions will provide important information regarding the nitrogen abundance on Mars. We hypothesize that Mars soil, as typical of extremely dry desert soils on Earth, is likely to contain at least some of the missing nitrogen as nitrate salts and some fixed ammonium bound to aluminosilicate minerals.

The Mojave Desert has been long considered a suitable terrestrial analogue to Mars in many geological and astrobiological aspects. The Silver Lake region in the Mojave Desert hosts several different rock types (talc, marble, quartz, white carbonate and red-coated carbonate) colonized by hypoliths within a few kilometres. This provides an opportunity to investigate the effect of rock type on hypolithic colonization in a given environment. Transmission measurements from 300 to 800 nm showed that the transmission of blue and UVA varied between rock types. The wavelength at which the transmission fell to 1% of the transmission at 600 nm was 475 nm for white carbonate and quartz, 425 nm for red-coated carbonate and talc and 380 nm for marble. The comparative analysis of the cyanobacterial component of hypoliths under different rocks, as revealed by sequencing 16S rRNA gene clone libraries, showed no significant variation with rock type; hypoliths were dominated by phylotypes of the genus Chroococcidiopsis, although less abundant phylotypes of the genus Loriellopsis, Leptolyngbya and Scytonema occurred. The comparison of the confocal laser scanning microscopy-λ (CLSM-λ) scan analysis of the spectral emission of the photosynthetic pigments of Chroococcidiopsis in different rocks with the spectrum of isolated Chroococcidiopsis sp. 029, revealed a 10 nm red shift in the emission fingerprinting for quartz and carbonate and a 5 nm red shift for talc samples. This result reflects the versatility of Chroococcidiopsis in inhabiting dry niches with different light availability for photosynthesis.

The study of life remote in space has strong parallels with the study of life remote in time. Both are dependent on decoding those historic phenomena called ‘fossils’, here taken to include biogenic traces of activity and waste products. There is the shared problem of data restoration from incomplete data sets; the importance of contextual analysis of potentially viable habitats; the centrality of cell theory; the need to reject the null hypothesis of an abiogenic origin for candidate cells via morphospace analysis; the need to demonstrate biology-like behaviour (e.g., association with biofilm-like structures; tendency to form clusters and ‘mats’; and a preference for certain substrates), and of metabolism-like behaviour (e.g., within the candidate cell wall; within surrounding ‘waste products’; evidence for syntrophy and metabolic cycles; and evidence for metabolic tiers). We combine these ideas into a robust protocol for demonstrating ancient or extra-terrestrial life, drawing examples from Earth's early geological record, in particular from the earliest known freshwater communities of the 1.0 Ga Torridonian of Scotland, from the 1.9 Ga Gunflint Chert of Canada, from the 3.4 Ga Strelley Pool sandstone of Australia, and from the 3.46 Ga Apex Chert also of Australia.

For over 40 years the formalism known as the Drake equation has helped guide speculation about the likelihood of intelligent extraterrestrial life contacting us. Since the equation was formulated there have been significant advances in astronomy and astrophysics, sufficient to merit a review of the significance of the Drake equation. The equation itself is as a series of terms which, when combined, allow an informed discussion of the likelihood of contact with an alien intelligence. However, whilst it has a mathematical form (i.e. a series of terms multiplied together to give an overall probability) it is best understood not as an equation in the strictly mathematical sense. Some of the terms have a physically quantifiable, numerically based meaning (e.g. obtainable from astronomy) and some are more social in content in that they describe the behaviour and evolution of societies and thus are more social science in nature and not truly estimable without observation of a set of societies. Initially, almost all the terms had to be estimated based on informed guesswork or belief. However, in the intervening period since the early 1960s, many of the a priori scientific terms which were themselves initially so uncertain as to require estimation by guess work or belief are now, or will soon be, directly measurable from current or planned astronomical projects. This leaves the non-scientific terms as a distinct class of their own, still subject to analysis only by discussion. Thus observational astronomy has nearly caught up with parts of the Drake equation and will soon quantify the purely physical science parts of the equation. The social parts (concerning intelligent societies, etc.) are still a priori unknowable. In addition, the growth of the subject called astrobiology (i.e. the study of life in the Universe) has developed so fast that communicating with intelligent life is now increasingly seen as just one small part of a much larger discipline. The knowledge as to whether there is life per se (apart from on Earth) in our galactic neighbourhood may be obtainable in the near future directly from observation. Such knowledge will have a profound impact on mankind and will be obtained without the form of communication envisaged by the Drake equation.

Several robotic exploration missions will travel to Mars during this decade to investigate habitability and the possible presence of life. Field research at Mars analogue sites such as desert environments can provide important constraints for instrument calibration, landing site strategies and expected life detection targets. We have characterized the mineralogy, organic chemistry and microbiology of ten selected sample sites from the Utah desert in close vicinity to the Mars Desert Research Station (MDRS) during the EuroGeoMars 2009 campaign (organized by International Lunar Exploration Working Group (ILEWG), NASA Ames and ESA ESTEC). Compared with extremely arid deserts (such as the Atacama), organic and biological materials can be identified in a larger number of samples and subsequently be used to perform correlation studies. Among the important findings of this field research campaign are the diversity in the mineralogical composition of soil samples even when collected in close proximity, the low abundances of detectable polycyclic aromatic hydrocarbons (PAHs) and amino acids and the presence of biota of all three domains of life with significant heterogeneity. An extraordinary variety of putative extremophiles, mainly Bacteria and also Archaea and Eukarya was observed. The dominant factor in measurable bacterial abundance seems to be soil porosity and lower small (clay-sized) particle content. However, correlations between many measured parameters are difficult to establish. Field research conducted during the EuroGeoMars 2009 campaign shows that the geological history and depositional environment of the region, as well as the mineralogy influence the ability to detect compounds such as amino acids and DNA. Clays are known to strongly absorb and bind organic molecules often preventing extraction by even sophisticated laboratory methods. Our results indicate the need for further development and optimization of extraction procedures that release biological compounds from host matrices to enable the effective detection of biomarkers during future sampling campaigns on Earth and Mars.

Cryptoendolithic microbial communities living within Antarctic rocks are an example of survival in an extremely cold and dry environment. The extinction of these micro-organisms formerly colonizing sandstone in the Mount Fleming area (Ross Desert), was probably provoked by the hostile environment. This is considered to be a good terrestrial analogue of the first stage of the disappearance of possible life on early Mars. To date, only macroscopically observed indirect biomarkers of the past activity of cryptoendoliths in Antarctic rocks have been described. The present paper confirms, for the first time, the existence of cryptoendolith microbial fossils within these sandstone rocks. The novel in situ application of scanning electron microscopy with backscattered electron imaging and simultaneous use of X-ray energy dispersive spectroscopy allowed the clear detection of microfossils left behind by Antarctic endoliths. Careful interpretation of the morphological features of cells, such as preserved cell walls in algae, fungi and bacteria, cytoplasm elements such as chloroplast membranes in algae and organic matter traces, mineral associations, and the spatial context of these structures all point to their identification as cryptoendolith microfossils. This type of investigation will prompt the development of research strategies aimed at locating and identifying the signs that Martian microbiota, probably only bacteria if they existed, may have been left for us to see.

New observations of Titan by the Cassini spacecraft suggest the presence of cryovolcanism on the surface. Cryovolcanism has important astrobiological implications, as it provides a means of exposing Titan's organics to liquid water, transforming hydrocarbons and nitriles into more evolved and oxidized prebiotic species. One possible cryovolcano – the 180 km structure Ganesa Macula – resembles the pancake domes seen on Venus by the Magellan spacecraft. To assess the potential of Ganesa Macula for prebiotic chemistry, we estimate its height using radarclinometry and other methods, and calculate the freezing timescale assuming an initially completely liquid dome. Given height constraints of ~200 m to 4 km, we find that liquid water or water–ammonia environments could be sustained in Ganesa Macula for timescales of the order of 102–105 years. These timescales open a window for prebiotic chemistry far wider than can be explored in terrestrial laboratory experiments.

The recently approved ExoMars rover is the first element of the ESA Aurora programme and is scheduled to deliver the Pasteur exobiology payload to Mars by 2015. The 0.7 kg Panoramic Camera will provide multi-spectral stereo images with 65 ° field-of-view (1.1 mrad/pixel) and high-resolution (85 μrad/pixel) monoscopic ‘zoom’ images with 5 ° field-of-view. The stereo wide-angle cameras (WAC) are based on the Beagle 2 Stereo Camera System heritage (Griffiths et al. (2005). Planet. Space Sci. 53, 1466–1488). The Panoramic Camera instrument is designed to fulfil the digital terrain mapping requirements of the mission as well as to provide multi-spectral geological imaging, colour and stereo panoramic images, water vapour abundance and dust optical depth measurements. It can also be used for high-resolution imaging of inaccessible locations on crater walls and to observe retrieved subsurface samples before ingestion into the rest of the Pasteur payload.

The discovery of high concentrations of water-ice just below the Martian surface polar areas made by Mars Odyssey has strengthened the debate about the search for life on Mars. Generally it is believed that life on Earth emerged in liquid water from the processing of organic molecules. Thus, the possible origin of life on early Mars should have been related to the evolution of the planetary water inventory, consequently it is important to know the amount of water-ice stored below the planetary surface. The search and mapping of the present subsurface water and ice reservoirs will be carried out experimentally by Mars Express with its Mars Advanced Radar for Subsurface and Ionospheric Sounding (MARSIS) ground-penetrating radar in the near future. We estimate the present and past water-ice reservoirs, which are and were in exchange with the atmosphere by using the observed D/H ratio in the atmospheric water vapour, measured D/H ratios in Martian SNC meteorites and D/H isotope ratios based on a study by Lunine et al. (2003) regarding asteroid and cometary water delivery to early Mars. Using the results of this study with initial D/H ratios of about 1.2–1.6 times the terrestrial sea water (TSW) ratio and the assumption that these ratios were not fractionated by XUV-driven hydrodynamic escape due to a more active young Sun prior to 3.5 Ga, one finds a present water-ice reservoir, which can exchange with the Martian atmosphere, equivalent to a global ocean layer with a thickness of about 3.3–15 m. By assuming that hydrodynamic escape fractionated the D/H ratio to a value that is stored in the old Martian SNC meteorites with a measured average enrichment of about 2.3 times the TSW ratio we estimate a present water-ice reservoir equivalent to a global layer with a thickness of about 11–27 m. From the obtained range of the estimated present water-ice deposit, we estimate a water-ice reservoir exchangeable with the atmosphere on Mars 3.5 Ga equivalent to a global ocean with a thickness of between 17 and 61 m. All the estimated reservoirs depend on the escape of water from Mars since 3.5 Ga equivalent to a global ocean with a thickness of about 14 m (minimum) to 34 m (maximum). The main uncertainties in the estimate of the minimal and maximal water-ice reservoir is related to the present uncertainties in the efficiency of atmospheric escape rates triggered by plasma instabilities and momentum transfer effects between the solar wind and the ionosphere. However, these uncertainties will be reduced in the near future, since both loss processes will be studied in detail by the Automatic Space Plasma Experiment with a Rotating Analyzer (ASPERA-3) on-board Mars Express. The obtained results combined with the discovery of the present water-ice subsurface reservoirs by the MARSIS radar and isotope studies as presented in this work, will also give us an idea of how enriched the atmosphere was in D compared with H after the heavy bombardment corresponding to a better understanding of the efficiency of the hydrodynamic escape process due to the young Sun.

We present the first model that couples high-resolution simulations of the formation of local group galaxies with calculations of the galactic habitable zone (GHZ), a region of space which has sufficient metallicity to form terrestrial planets without being subject to hazardous radiation. These simulations allow us to make substantial progress in mapping out the asymmetric three-dimensional GHZ and its time evolution for the Milky Way (MW) and Triangulum (M33) galaxies, as opposed to works that generally assume an azimuthally symmetric GHZ. Applying typical habitability metrics to MW and M33, we find that while a large number of habitable planets exist as close as a few kiloparsecs from the galactic centre, the probability of individual planetary systems being habitable rises as one approaches the edge of the stellar disc. Tidal streams and satellite galaxies also appear to be fertile grounds for habitable planet formation. In short, we find that both galaxies arrive at similar GHZs by different evolutionary paths, as measured by the first and third quartiles of surviving biospheres. For the MW, this interquartile range begins as a narrow band at large radii, expanding to encompass much of the Galaxy at intermediate times before settling at a range of 2–13 kpc. In the case of M33, the opposite behaviour occurs – the initial and final interquartile ranges are quite similar, showing gradual evolution. This suggests that Galaxy assembly history strongly influences the time evolution of the GHZ, which will affect the relative time lag between biospheres in different galactic locations. We end by noting the caveats involved in such studies and demonstrate that high-resolution cosmological simulations will play a vital role in understanding habitability on galactic scales, provided that these simulations accurately resolve chemical evolution.

The prediction that (due to the limited amount of hydrogen available as fuel in the Sun) the future duration of our favourable terrestrial environment will be short (compared with the present age of the Earth) has been interpreted as evidence for a hard-step scenario. This means that some of the essential steps (such as the development of eukaryotes) in the evolution process leading to the ultimate emergence of intelligent life would have been hard, in the sense of being against the odds in the available time, so that they are unlikely to have been achieved in most of the earth-like planets that may one day be discovered in nearby extrasolar systems. It was originally estimated that only one or two of the essential evolutionary steps had to have been hard in this sense, but it has become apparent that this figure may need upward revision, because recent studies of climatic instability suggest that the possible future duration of our biologically favourable environment may be shorter than had been supposed, being only about 1 Gyr rather than 5 Gyr. On the basis of the statistical requirement of roughly equal spacing between hard steps, it is argued that the best fit with the fossil record is now obtainable by postulating the number of hard steps to be five, if our evolution was exclusively terrestrial, or six, if, as now seems very plausible, the first step occurred on Mars.

The search for extraterrestrial intelligence (SETI) has been heavily influenced by solutions to the Drake Equation, which returns an integer value for the number of communicating civilizations resident in the Milky Way, and by the Fermi Paradox, glibly stated as: ‘If they are there, where are they?’. Both rely on using average values of key parameters, such as the mean signal lifetime of a communicating civilization. A more accurate answer must take into account the distribution of stellar, planetary and biological attributes in the galaxy, as well as the stochastic nature of evolution itself. This paper outlines a method of Monte Carlo realization that does this, and hence allows an estimation of the distribution of key parameters in SETI, as well as allowing a quantification of their errors (and the level of ignorance therein). Furthermore, it provides a means for competing theories of life and intelligence to be compared quantitatively.

The Martian regolith is known to contain a maximum of 0.5% (w/v) perchlorate (ClO4−) that is toxic for most living organisms. With such high concentrations of perchlorates on Mars, is there any possibility of life? Here, in order to search and identify potential organisms on Earth, which could survive the perchlorate levels on Mars, we have isolated four perchlorate resistant, halophilic/halotolerant bacterial species from Big Soda Lake (BSL) in Nevada, USA. The 16S ribosomal RNA sequences revealed that these halophiles belong to the genera Bacillus, Alkalibacillus and Halomonas. Growth curves were obtained using a saline medium with different concentrations of magnesium, sodium and/or calcium perchlorate salt to simulate the Martian eutectic brine water. All four species, BSL1-4, grew in high saline media in the presence of perchlorates. This is the first growth experiment using multiple perchlorate salts. BSL3 relative to Halomonas salifodinae showed high maximum growth (Optical Density) comparing with other isolates in the presence of 1% perchlorate salts. Also, BSL1 relative to Bacillus licheniformis survived in the presence of 5% Na-perchlorate, but growth was slower in the absence of Na-perchlorate. The results revealed that these new model microbes are capable of tolerating the hypothesized hypersaline and perchlorate-rich Martian subsurface water environment. Perchlorate-resistant halophile would serve as a new model to understand the biochemistry that may occur on Mars.

In the redshift range 100≲(1+z)≲137, the cosmic microwave background (CMB) had a temperature of 273–373 K (0–100°C), allowing early rocky planets (if any existed) to have liquid water chemistry on their surface and be habitable, irrespective of their distance from a star. In the standard ΛCDM cosmology, the first star-forming halos within our Hubble volume started collapsing at these redshifts, allowing the chemistry of life to possibly begin when the Universe was merely 10–17 million years old. The possibility of life starting when the average matter density was a million times bigger than it is today is not in agreement with the anthropic explanation for the low value of the cosmological constant.
