The anti-haemagglutinin antibody response in adult human volunteers to inactivated whole virus or tween ether split influenza A/Victoria/75 (H3N2) and A/Scotland/74 (H3N2) virus vaccines was investigated using antibody adsorption and single-radial-haemolysis (SRH) techniques. The concentrations of haemagglutinin (HA), nucleoprotein (NP) and matrix (M) antigens measured by single radial diffusion (SRD) and rocket immunoelectrophoresis were similar for both the whole virus and split vaccines. Whole virus and split vaccines induced cross-reactive (CR) antibody in 87% of vaccinees. Strain specific (SS) antibody to A/Hong Kong/1/68 or the homologous virus was induced less frequently than CR antibody. Higher anti-haemagglutinin antibody titres were detected in persons receiving the split virus vaccines than in those receiving the whole virus vaccines. No antibody to the type-specific matrix protein was detectable, but 33% of volunteers developed an antibody rise to type-specific nucleoprotein antigen.

The specificity of the anti-haemagglutinin antibody response in human adults to natural infection with A/Port Chalmers/73 (H3N2) virus was similar to that induced by inactivated vaccines in that a high proportion of subjects developed CR anti-haemagglutinin antibody, which reacted with A/Hong Kong/68 virus and the homologous A/Port Chalmers/73 virus, and SS antibody for A/Hong Kong/68 virus but SS antibody for A/Port Chalmers/73 virus was infrequently stimulated by natural infection.

A collection of 346 strains of Streptococcus milleri from a variety of human sources was examined culturally and biochemically, and for the presence of Lancefield group antigens. Most of the strains were non-haemolytic and ungroupable, but 25% were β haemolytic and 19% were α haemolytic; 28% possessed a group antigen (A, 5%; C, 6%, F, 14%, G, 3%). These antigens were present in 69% of β-haemolytic but in only 13% of α-haemolytic or non-haemolytic strains; β haemolysis occurred in 82% of group-F strains, 43% of other groupable strains and 11% of ungroupable strains.

The following reactions were given by > 80% of S. milleri strains: hydrolysis of arginine and aesculin, a positive Voges–Proskauer reaction, and acidification of trehalose, lactose, salicin and sucrose. A minority of strains showed enhancement of growth by CO2, bile tolerance, NaCl tolerance, and ability to acidify other sugars, notably mannitol, raffinose and melibiose. Departures from the modal pattern of biochemical reactions showed a weak correlation with the type of haemolysis and the presence or absence of a group antigen but were not sufficiently systematic for clear-cut subdivisions to be recognized within the species.

S. milleri therefore appeared to comprise a ‘central’ group of non-haemolytic strains that rarely formed a Lancefield-group antigen, in which the aesculin reaction was nearly always positive, lactose was usually acidified, and a considerable minority showed enhancement of growth by CO2 and bile tolerance. Deviations from this pattern were of two main types. (1) ‘Loss’ of one or more of these reactions, which tended to be associated with β-haemolysis and presence of a group antigen. In these respects, α-haemolytic strains tended to occupy an intermediate position. (2) ‘Gain’ of the ability to acidify additional sugars, notably raffinose and melibiose or mannitol; this occurred mainly among otherwise typical non-haemolytic strains that were rarely groupable.

Only 12% of isolations from the bloodstream of patients suffering from systemic infections were β haemolytic and only 18% possessed a group antigen, but a considerably greater proportion of those from visceral abscesses were β haemolytic (28%). Among isolations from superficial lesions in some body sites there were considerably greater proportions of β-haemolytic and groupable strains; thus, nearly one-half of those isolated from the abdomen other than the female genital tract were β-haemolytic and over one-half were groupable. On the other hand, strains from the teeth and gums were nearly always non-haemolytic and ungroupable, and most vaginal isolations were of non-haemolytic strains with a wide sugar-fermentation pattern.

Swine Vesicular Disease virus (SVDV) did not survive drying at high relative humidities (r.h.) but there was little virus loss at low r.h.

Purified virus dried in films was inactivated by formaldehyde fumigation only at high r.h. Inactivation was also influenced by the suspending medium from which the virus was dried. Purified virus resuspended in distilled water and then dried, was rapidly killed, but that in tissue culture fluid survived.

A bacteriological study was carried out on the first Leisure Centre swimming pools in the United Kingdom to be disinfected with ozone/chlorine. Results suggested that a free chlorine concentration of approximately 0·8 mg/l was necessary to maintain the pools in a bacteriologically satisfactory condition. This amount of free chlorine was similar to that required when the pool was disinfected with chlorine alone. However, the associated amount of combined chlorine was much lower when disinfection was by ozone/chlorine and this gave more acceptable bathing conditions. Implications for the management of pools disinfected by this method are discussed.

Antibiotic resistant strains of Escherichia coil were isolated from the domestic sewage of Palmerston North. The ability of these strains to transfer the drug resistance markers to two different recipient E. coli strains was assessed. The ‘O’ and ‘H’ serotypes of all strains were determined and attempts at correlation with the drug resistance studies were made. Some typically ‘human’ serotypes were found as well as a number of rarely described ones. No correlation between serotype and resistance pattern was observed.

Re-evaluation of recently published figures comparing the tuberculin Tine Test+ and the Mantoux showed that, contrary to original opinion, the Tine Test+ did perform satisfactorily, the reason for the apparent ineffectiveness lying in the original interpretation of the results.

Thirty-two different types of anaerobic bacteria isolated from chickens have been tested for anti-salmonella activity in vitro. Under the conditions of the test only Bacteroides hypermegas and a Bifidobacterium sp. were shown to inhibit the salmonellas and this was attributed to the production of volatile fatty acids (VFA's) coupled with a low pH. When these organisms were tested in newly hatched chicks no inhibition of s. typhimurium occurred. Possible explanations for this observation are considered.

The pH value and concentration of VFA's in the caecal material were determined in chicks from 0–84 days. In vitro tests with s. typhimurium indicated that, whilst the organism would be able to multiply at the pH and concentration of VFA's found during the first few days after hatching, the rapid increase inVFA concentration during the first 21 days would make this increasingly difficult. The significance of the developing caecal flora in relation to VFA production and pH is discussed.

Because certain feed additives are known to influence the carriage of salmonellas, the sensitivity of various caecal anaerobes to these compounds was determined in vitro, generally at 1, 10 and 100 μg/ml. The additives tested included flavomycin, furazolidone, nitrovin, tetracycline, tylosin, sulphaquinoxaline, virginiamycin and zinc bacitracin. All the organisms tested were inhibited by 100 μg/ml furazolidone; none were inhibited by 500 μg/ml sulphaquinoxaline.

Changes occurring in the VFA concentration, pH value and microflora of the caeca of chicks fed for 49 days or longer on a normal starter diet or the same diet containing 10 or 100 mg/kg nitrovin have been compared. When the chicks were fed on the diet containing 100 mg/kg nitrovin, the Gram-negative non-sporing anaerobes were eliminated as a significant part of the caecal flora. However, the VFA concentration combined with a low pH in chicks from 2 weeks onwards was still sufficient to inhibit salmonella multiplication. Other possibly interrelated factors which might lead to an increased salmonella carrier rate in the nitrovintreated chickens are discussed.

During the H1N1 outbreak of 1977/8, the virus was isolated in embryonated eggs from 59 out of 76 patients (78%) with the serologically confirmed infection. A similar isolation frequency has been achieved during a period of six H3N2 outbreaks since 1972/3. The H1N1 strains were isolated less frequently from late specimens (collected 4–6 days from the onset of illness) and more often only in the second passage compared with the H3N2 viruses. The new H1N1 strains resembled those prevalent in the 1950s with respect to their ability to agglutinate erythrocytes of certain laboratory animals and to be eluted from these, and thus differed from the H3N2 viruses.

The behaviour of f2 coliphage during activated-sludge treatment was influenced by the temperature, flow-through-time, concentration of mixed liquor suspended solids and the virus load.

The most sensitive way to detect behavioural changes was to examine the regression coefficients for the rate of uptake or loss of virus by the mixed liquor solids. This type of analysis revealed, for instance, high values when the solids concentration was high and even greater values occured when high inocula were used. At high temperature the rate of loss of virus titre after inoculation had stopped was greater than the rate of uptake of virus during inoculation although in all other conditions uptake occurred at a greater rate than the loss of virus. The coefficients were relatively low when the flow rate was increased, when the temperature was low or when the inoculum was small.

The distribution of virus between the solids and liquid fractions of the mixed liquor varied somewhat for all conditions but was notably different when (a) the plant was incubated at 5 °C when there was much less virus in the solids fraction than usual, and (b) when the inoculum was low and a much higher proportion of virus was found in the solids.

The efficiency with which virus was removed across the plant was the least-sensitive determinant of viral behaviour and the value was about the same for most treatment conditions. However, low or high inocula did result in some increased or decreased removal of virus, respectively.

In two controlled clinical trials of a polyvalent pseudomonas vaccine, vaccinated burned patients showed higher antibody titres to the 16 antigens in the vaccine and higher titres of a passively transferable protective antibody than was found in unvaccinated burned patients.

A test is described for assessing the sanitizing effect of napkin treatment products on naturally urine-wetted and faecally-contaminated napkins. This test defines in-use conditions which closely resemble typical domestic situations. One napkin treatment product (‘Napisan’), tested at two different concentrations and with challenges of different numbers of babies' napkins, performed satisfactorily under the conditions used.

Porcine enterovirus type 2 or porcine adenovirus type 3 were seeded into samples of pig slurry, and a bovine enterovirus was seeded into cattle slurry, and samples of the slurry were aerated in the laboratory for 21 days. The viruses were inactivated more rapidly in the aerated slurry than in control slurry which was not aerated. The difference in inactivation rate was greatest for the porcine adenovirus and least for the bovine enterovirus. Inactivation of the porcine enterovirus in aerated distilled water and in aerated, autoclaved pig slurry proceeded at a similar rate as in the same materials which were not aerated. Ten samples of aerated slurry were collected from an aeration tank which received weekly additions of raw pig slurry which was sampled at the same times. Each sample yielded a porcine enterovirus after concentration with the polyelectrolyte PE-60, but in three comparative titrations the viral infectivity titre in concentrates of the raw slurry was at least 1000 times greater than in the aerated slurry. Porcine enterovirus type 2 and porcine adenovirus type 3, which were seeded into pig slurry, and a bovine enterovirus seeded into cattle slurry, were inactivated by treatment of the slurry with calcium hydroxide at pH 11·5. The inactivation rate was highest for the bovine enterovirus and lowest for the porcine adenovirus.

Three detergent preparations (bar soap, ‘Hibiscrub’ base and ‘LIC 76’), two antiseptic preparations (0·5% chlorhexidine in 95% ethyl alcohol and an alcohol jelly, ‘Alcogel’), and one antiseptic-detergent solution (4% chlorhexidine gluconate in a detergent base, ‘Hibiscrub’) were compared for their effectiveness, on a single use, in reducing the yield of bacteria from the hands of volunteers. The antiseptic and antiseptic-detergent preparations were more effective than the detergents, with a mean reduction in yield of skin bacteria of 96·0% after use of alcoholic chlorhexidine and of 81·2% after use of Hibiscrub. One of the detergents, LIC 76, appeared more effective than the others, causing a mean reduction in the yield of skin bacteria of 41·5%, compared with reductions of 4·6% by the Hibiscrub detergent base and an increase of 3·2% with bar soap; unlike the other detergents, LIC 76 was found to have appreciable bacteristatic and bactericidal properties.

Antibiotic production by dermatophyte fungi has been demonstrated in vivo in the lesions of patients with dermatomycoses. Patients infected with antibiotic-producing strains more frequently carried cocci resistant to penicillin and other antibiotics than did patients infected with non-producer strains. The total bacterial load was less in lesions caused by producer fungi. In vitro studies demonstrated the selection of penicillin-resistant S. aureus from mixed populations of resistant and sensitive cells.

Salmonella dublin is primarily adapted to bovines and is a relatively rare cause of human illness. An outbreak is described in which it was estimated that at least 700 persons were infected from milk which had not been subjected to heat treatment. Although the organism was isolated from retail samples of milk, investigations at the dairy farm were inconclusive and a number of questions are posed. Attention is drawn to the value of inter-disciplinary cooperation in the investigation of the outbreak.

Coliforms isolated from infections over a period of 22 months were examined and 16° were shown to be klebsiellas. The biochemical reactions and serotypes of these klebsiellas were determined and the relationship between species, serotype and type of infection investigated. Although no obvious outbreaks of infection occurred during this period a number of clusters of isolations of the same serotype of klebsiella were found.

Using an enrichment method for the isolation of klebsiellas from faeces, sero and bacteriocin typing, and the examination of ten colonies, it was demonstrated that half of the patients carried a klebsiella of the same type in the bowel as caused the infection.

For 29 months the Veterinary Investigation Centres, covering the whole of Great Britain, forwarded trimethoprim-resistant gram negative enteric bacteria to the Wellcome Research Laboratories. These were examined for degree of resistance, presence and type of R factors. Trimethoprim resistance was found in 0·6 % of the total number of strains examined by the Veterinary Investigation Centres. Trimethoprim R factors were demonstrated in one quarter of the resistant strains, and R factors were found in two strains of Salmonella typhimurium. It was concluded that while the incidence of trimethoprim resistance revealed by the survey gave no cause for alarm, the detection of resistant strains, and particularly R factors, indicated that the drug should continue to be used only for specific therapeutic purposes.

In a warfarin-resistant population of wild mice reared in the laboratory, a dominant gene for adiposity, Ad, was found to segregate. The onset of obesity is at 4–6 months, and adipose mice suffer from hyperinsulinaemia; the sexes differ in penetrance, males having greater penetrance then females. Linkage backcrosses show the gene to be situated on chromosome 7 with about 25% recombination with the closely linked warfarin-resistance genes War, and frizzy, fr. The finding of adipose in two other wild populations also carrying War is discussed as an ecological and physiological problem.

The flora of both faeces and small bowel lumen was studied in children with gastroenteritis from London, England, and Kingston, Jamaica.

Clinical and laboratory differences between these two groups are described.

All bacterial groups in the faeces were greatly altered during gastroenteritis and this particularly affected anaerobic organisms. These changes generally reverted rapidly to normal after the illness.

The small bowel flora was also altered during gastroenteritis; there was a tendency for a wider range of organisms including anaerobes to be isolated from the children in Jamaica than from those in England.

The indirect immunofluorescence technique has been used to titrate the specific immunoglobulins in 200 sera from 64 patients with varicella, and 195 sera from 67 patients with herpes zoster. IgG and IgM antibodies were detected in all patients with varicella, and IgA in 59 (92%). All three classes of antibody appeared 2–5 days after the onset of the rash, increased virtually simultaneously and reached maximum titres during the second and third weeks. IgG then declined slowly, but never became undetectable and was still present in five subjects who were retested after 2–4 years; it was present in 88 out of 100 healthy young adults and probably persists indefinitely after varicella. IgA and 1gM antibodies declined more rapidly and were not detected in specimens taken more than a year after the illness. IgA, however, may possibly persist in some cases since low titres were found in 8 out of 88 young adults who possessed IgG antibody and had presumably had varicella in the past. IgA responses were significantly weaker in children under the age of 6 years than in older children and adults.

Six out of 67 patients with zoster were tested at various times before the onset of the rash: IgG antibody was detected in all. IgG was present in all sera taken after the onset of the rash, increased rapidly after 2–5 days, reached maximum titres during the second and third weeks and then declined slowly. IgA antibody was detected in 66 patients (99%) and IgM in 52 (78%); both types of antibody followed transient courses, as in varicella.

Maximum titres of IgG and complement-fixing antibodies were greater after zoster than after varicella, but the differences were not significant. IgA and IgM titres in young adults with zoster were significantly lower than in older patients, and also lower than in young adults with varicella.

Increases in varicella-zoster antibody in patients with herpes simplex virus infections consisted mainly of IgG, sometimes IgA, but never IgM.
