Impulsive and compulsive problem behaviours are associated with a variety of mental disorders. Latent phenotyping indicates the expression of impulsive and compulsive problem behaviours is predominantly governed by a transdiagnostic ‘disinhibition’ phenotype. In a cohort of 117 individuals, recruited as part of the Neuroscience in Psychiatry Network (NSPN), we examined how brain functional connectome and network properties relate to disinhibition. Reduced functional connectivity within a subnetwork of frontal (especially right inferior frontal gyrus), occipital and parietal regions was linked to disinhibition. Findings provide insights into neurobiological pathways underlying the emergence of impulsive and compulsive disorders.

Given the failure of psychiatry to develop clinically useful biomarkers for psychiatric disorders, and the concomitant failure to develop significant advances in diagnosis and treatment, the National Institute of Mental Health (NIMH) in 2010 launched the Research Domain Criteria (RDoC), a framework for research based on the assumption that mental disorders are disorders of identifiable brain neural circuits, with neural circuitry at the center of units of analysis ranging from genes, molecules, and cells to behavior, self-reports, and paradigms. These were to be integrated with five validated dimensional psychological constructs such as negative and positive valence systems. Four years later, the NIMH stated that the ultimate goal of RDoC is precision medicine for psychiatry, with the assumption that precision medications will normalize dysfunctional neural circuits. How this could be accomplished is not obvious, given that neural circuits are widely distributed, have unclear boundaries, and exhibit a significant degree of neuroplasticity, with multiple circuits present in any given disorder. Moreover, the early focus on neural circuitry has been criticized for its reductionism and neglect of the more recent RDoC emphasis on the integration and equivalence of biological and psychological phenomena. Yet this seems inconsistent with the priorities of the NIMH director, an advocate of the central role of neural circuitry and projects such as the Brain Initiative and the Human Connectome Project. Will such projects, at a cost of at least $10 billion, lead to precision medications for mental disorders, or further diminish funding for clinical care and research?

Personality and individual differences originate from the brain. Despite major advances in the affective and cognitive neurosciences, however, it is still not well understood how personality and single personality traits are represented within the brain. Most research on brain-personality correlates has focused either on morphological aspects of the brain such as increases or decreases in local gray matter volume, or has investigated how personality traits can account for individual differences in activation differences in various tasks. Here, we propose that personality neuroscience can be advanced by adding a network perspective on brain structure and function, an endeavor that we label personality network neuroscience.

With the rise of resting-state functional magnetic resonance imaging (MRI), the establishment of connectomics as a theoretical framework for structural and functional connectivity modeling, and recent advancements in the application of mathematical graph theory to brain connectivity data, several new tools and techniques are readily available to be applied in personality neuroscience. The present contribution introduces these concepts, reviews recent progress in their application to the study of individual differences, and explores their potential to advance our understanding of the neural implementation of personality.

Trait theorists have long argued that personality traits are biophysical entities that are not mere abstractions of and metaphors for human behavior. Traits are thought to actually exist in the brain, presumably in the form of conceptual nervous systems. A conceptual nervous system refers to the attempt to describe parts of the central nervous system in functional terms with relevance to psychology and behavior. We contend that personality network neuroscience can characterize these conceptual nervous systems on a functional and anatomical level and has the potential do link dispositional neural correlates to actual behavior.

A key objective of the emerging field of personality neuroscience is to link the great variety of the enduring dispositions of human behaviour with reliable markers of brain function. This can be achieved by analysing big data-sets with methods that model whole-brain connectivity patterns. To meet these expectations, we exploited a large repository of personality and neuroimaging measures made publicly available via the Human Connectome Project. Using connectomic analyses based on graph theory, we computed global and local indices of functional connectivity (e.g., nodal strength, efficiency, clustering, betweenness centrality) and related these metrics to the five-factor model (FFM) personality traits (i.e., neuroticism, extraversion, openness, agreeableness, and conscientiousness). The maximal information coefficient was used to assess for linear and nonlinear statistical dependencies across the graph “nodes”, which were defined as distinct large-scale brain circuits identified via independent component analysis. Multivariate regression models and “train/test” approaches were used to examine the associations between FFM traits and connectomic indices as well as to assess the generalizability of the main findings, while accounting for age and sex variability. Conscientiousness was the sole FFM trait linked to measures of higher functional connectivity in the fronto-parietal and default mode networks. This offers a mechanistic explanation of the behavioural observation that conscientious people are reliable and efficient in goal-setting or planning. Our study provides new inputs to understanding the neurological basis of personality and contributes to the development of more realistic models of the brain dynamics that mediate personality differences.

Abnormal brain connectivity or network dysfunction has been suggested as a paradigm to understand several psychiatric disorders. We here review the use of novel meta-analytic approaches in neuroscience that go beyond a summary description of existing results by applying network analysis methods to previously published studies and/or publicly accessible databases. We define this strategy of combining connectivity with other brain characteristics as ‘meta-connectomics’. For example, we show how network analysis of task-based neuroimaging studies has been used to infer functional co-activation from primary data on regional activations. This approach has been able to relate cognition to functional network topology, demonstrating that the brain is composed of cognitively specialized functional subnetworks or modules, linked by a rich club of cognitively generalized regions that mediate many inter-modular connections. Another major application of meta-connectomics has been efforts to link meta-analytic maps of disorder-related abnormalities or MRI ‘lesions’ to the complex topology of the normative connectome. This work has highlighted the general importance of network hubs as hotspots for concentration of cortical grey-matter deficits in schizophrenia, Alzheimer's disease and other disorders. Finally, we show how by incorporating cellular and transcriptional data on individual nodes with network models of the connectome, studies have begun to elucidate the microscopic mechanisms underpinning the macroscopic organization of whole-brain networks. We argue that meta-connectomics is an exciting field, providing robust and integrative insights into brain organization that will likely play an important future role in consolidating network models of psychiatric disorders.

The human connectome has been widely studied over the past decade. A principal finding is that it can be decomposed into communities of densely interconnected brain regions. Past studies have often used single-scale modularity measures in order to infer the connectome's community structure, possibly overlooking interesting structure at other organizational scales. In this report, we used the partition stability framework, which defines communities in terms of a Markov process (random walk), to infer the connectome's multi-scale community structure. Comparing the community structure to observed resting-state functional connectivity revealed communities across a broad range of scales that were closely related to functional connectivity. This result suggests a mapping between communities in structural networks, models of influence-spreading and diffusion, and brain function. It further suggests that the spread of influence among brain regions may not be limited to a single characteristic scale.
