Allostery describes the ability of biological macromolecules to transmit signals spatially through the molecule from an allosteric site – a site that is distinct from orthosteric binding sites of primary, endogenous ligands – to the functional or active site. This review starts with a historical overview and a description of the classical example of allostery – hemoglobin – and other well-known examples (aspartate transcarbamoylase, Lac repressor, kinases, G-protein-coupled receptors, adenosine triphosphate synthase, and chaperonin). We then discuss fringe examples of allostery, including intrinsically disordered proteins and inter-enzyme allostery, and the influence of dynamics, entropy, and conformational ensembles and landscapes on allosteric mechanisms, to capture the essence of the field. Thereafter, we give an overview over central methods for investigating molecular mechanisms, covering experimental techniques as well as simulations and artificial intelligence (AI)-based methods. We conclude with a review of allostery-based drug discovery, with its challenges and opportunities: with the recent advent of AI-based methods, allosteric compounds are set to revolutionize drug discovery and medical treatments.

Forcibly displaced populations are among the most vulnerable groups in disasters. They experience poorer health conditions compared with nondisplaced individuals. However, a clear picture is lacking regarding the overall health problems encountered by disaster-induced mid- to long-term displaced people. This study investigated these disorders prevalence and identified their correlates among long-settled displaced populations worldwide. The current scoping review follows the PRISMA-ScR guidelines; a systematic search was conducted on PubMed, Web of Science, and CINAHL and included original peer-reviewed studies, commentary, reviews, and grey literature published in English between January 1990 to June 2022. In the thematic and content analysis, the authors applied the narrative review approach to identify themes and sub-themes. Forty-eight documents were identified as fully relevant to this study. The largest number of published papers were from Asia, followed by the Middle East, the United States, and Europe. IDPs in developed countries were the most researched populations. Human-made disasters were addressed by 89% of the included studies. The four main thematic categories included were “physical health,” “mental health,” “inadequate facilities,” and “lack of healthy behaviour.” The worsening of noncommunicable diseases had the highest prevalence, followed by communicable diseases. Due to their condition, forcibly displaced migrants face a triple burden of communicable diseases and noncommunicable diseases such as mental health issues. Health-related research and policy need to consider the links among disasters, health problems, and forced migration as a determinant of health in the new era of climate change-driven displacements.

This article explores the responsibility to protect (R2P) as an organizing concept for preventing, addressing and finding durable solutions to internal displacement in Africa. While the most innovative norms for protecting the forcibly displaced have been conceptualized in Africa, they have not durably addressed displacement, due to limitations in implementation. R2P has similarly faced criticisms emanating from a lack of clarity and distrust. Restated norms underlying frameworks for internally displaced persons (IDPs) and R2P complement each other, and can be operationalized simultaneously through a more credible regional approach, to encourage effective protection of IDPs in Africa. The article explores pillar one, pillar two and the non-coercive elements of pillar three of R2P, and its underlying moral principles, using Kenya as a case study of the process of seeking to secure state responsibility for the protection of displaced civilians victimized by mass atrocities.

The number of internally forcibly displaced persons is growing every year across the globe and exceeds the number of refugees. To date, Ukraine has the highest number of internally displaced persons (IDPs) in Europe, with about 1.4 million people forced to flee from the conflict in eastern Ukraine. Employing Massey’s concept of ‘power geometry’, the modalities of borders, and taking an intersectional approach, this article theorizes how IDPs are situated politically within a protracted conflict. Such an approach offers the chance to see how the reaction to the war brings authorities to see displaced people as a static category and reproduces a war-lexicon in policies, which fractures the space of everyday life. Drawing upon qualitative research on IDPs, the civil society, international organizations, and public officials in Ukraine, the article concludes that intersections of gender and older age with displacement, and the lack of state recognition of these differing groups of IDPs, together with the lack of the economic resources for social policy, produces multiple forms of social exclusion.

The displacement of civilians during a protracted war is a difficult issue that deserves our attention, and Iraq is unfortunately an emblematic example of this phenomenon. Based on the literature produced by humanitarian organizations and academia, this article aims at analyzing what triggers displacement in protracted conflict, highlighting the role of international humanitarian law (IHL) violations. It discusses how Iraq has been struggling with acts of violence, hostilities and IHL violations that have generated displacement and human suffering.

This article provides an overview of the development of the International Federation of Red Cross and Red Crescent Societies’ (IFRC) approach to migration and displacement. The focus of the IFRC and its member National Red Cross and Red Crescent Societies (National Societies) in this regard has traditionally been on refugees and other so-called “displaced persons” – that is, people who have been compelled to flee their place or country of origin and for this reason are deemed to be particularly vulnerable. However, this focus has been extended recently, in the course of the past decade, to cover all people who find themselves in a vulnerable situation in the context of migration. The IFRC Migration Policy, which was adopted in 2009, has offered much-needed guidance to National Societies in dealing with all migrants, including irregular migrants. However, it is argued that there is a need today – taking into consideration the increasing number of displaced people worldwide and the numerous contexts in which National Societies are dealing with refugees, internally displaced persons or cross-border disaster-displaced persons – to better understand the programmatic aspects that are specific to displacement compared with migration. This is a necessary condition in view of the development of more adequate and effective responses to the vulnerabilities and needs of migrants and displaced persons.

This article examines the value of the African Union Convention for the Protection and Assistance of Internally Displaced Persons in Africa (Kampala Convention) in the general quest for the regional and global protection of internally displaced persons (IDPs). It contends that the absence of a globally binding legal instrument for the protection of IDPs underlines the importance of the Kampala Convention and the possible contribution it can make to global and regional efforts to create a binding legal framework for the protection of IDPs. While recognizing some challenges that may impact the full implementation of the Convention, the article concludes by noting its various positive elements that are invaluable in overall efforts to create a comprehensive global legal framework to enhance protection of IDPs.

Addressing the case of a community from rural Urabá, Colombia, this article focuses on the temporality of population displacement and asks: when does the ‘clock’ of displacement start? Drawing upon an in-depth ethnographic fieldwork it challenges the state driven narrative that displacement can be understood from the moment one leaves their residence and advances the argument that displacement is more than just physical relocation. By engaging with the broader social, political and economic context in which displacement occurred and bringing local voices to the fore, this article demonstrates how the experience of violence engendered a sense of displacement before residents actually left.
