The antibacterial, antifungal and antioxidant effects of halloysite nanoclay, Cloisite 10A (C10A) and Cloisite 15A (C15A) organonanoclays were examined in this study. The antimicrobial action was assessed using the agar-well method and the disc diffusion method. The free radical-scavenging effects of the clays were determined using the 2,2-diphenyl-1-picrylhydrazyl method. Halloysite showed antimicrobial activity against Pseudomonas aeruginosa, Enterococcus faecalis and Staphylococcus aureus. C10A was effective against both Gram-positive bacteria (S. aureus, Listeria monocytogenes, Bacillus subtilis and E. faecalis) and Gram-negative bacteria (Escherichia coli, Klebsiella pneumoniae and P. aeruginosa). Additionally, only C10A was found to have an antimicrobial effect on Candida glabrata of 18 mm amongst the tested clays. C15A showed an antimicrobial effect on S. aureus and K. pneumoniae. It was determined that the antifungal properties of organoclays were higher than those of halloysite. The most effective clay type was determined to be C10A. The positively charged inner surface of the halloysite nanoclay can provide a large area to which negatively charged free radicals can attach. The modified C15A used in this study has two long-chain alkyl groups attached, whereas the modified C10A has a single long-chain alkyl group and a benzyl group attached. It is proposed that the differences in these antimicrobial effects are due to the structures of the molecules. According to these results, organoclays as green source materials could be used as additives and coatings in food processing, biomedical devices, filters and paints due to their antimicrobial and antioxidant properties.

Polymer–filler interactions play a major role in determining the antibacterial activity of organoclay in nanocomposites. The objective of the current study was to determine the effect of polymer type on the antibacterial properties of an organically modified clay – cloisite 10A (C10A) – using poly-ε-caprolactone (PCL) and poly-L-lactic acid (PLA) polymeric systems. Nanocomposite characterization using atomic force microscopy (AFM) showed an increase in roughness upon addition of the clay mineral, and X-ray diffraction (XRD) showed intercalation of the selected polymers into the interlayer spaces of the clay. Transmission electron microscopy (TEM) analysis supported the XRD findings. C10A in PCL thin films enhanced the bactericidal activity against Staphylococcus aureus when compared to the C10A in PLA. The observed change could be the result of pronounced levels of interaction between the filler and polymer matrix in the C10A-PLA nanocomposite when compared to C10A-PCL. The higher interaction levels could hinder the diffusion of bactericidal agents from the nanocomposite membranes. The present study provided insight into the nature of interaction between nanocomposite components and its impact on bioactivity, which can have applications in terms of generating engineered antibacterial materials.

The worldwide emergence of infectious diseases, together with the increasing incidence of antibiotic-resistant bacteria, elevate the need to properly detect, prevent, and effectively treat these infections. The overuse and misuse of common antibiotics in recent decades stimulates the need to identify new inhibitory agents. Therefore, natural products like clays, that display antibacterial properties, are of particular interest.

The absorptive properties of clay minerals are well documented for healing skin and gastrointestinal ailments. However, the antibacterial properties of clays have received less scientific attention. French green clays have recently been shown to heal Buruli ulcer, a necrotic or ‘flesh-eating’ infection caused by Mycobacterium ulcerans. Assessing the antibacterial properties of these clays could provide an inexpensive treatment for Buruli ulcer and other skin infections.

Antimicrobial testing of the two clays on a broad-spectrum of bacterial pathogens showed that one clay promotes bacterial growth (possibly provoking a response from the natural immune system), while another kills bacteria or significantly inhibits bacterial growth. This paper compares the mineralogy and chemical composition of the two French green clays used in the treatment of Buruli ulcer.

Mineralogically, the two clays are dominated by 1Md illite and Fe-smectite. Comparing the chemistry of the clay minerals and exchangeable ions, we conclude that the chemistry of the clay, and the surface properties that affect pH and oxidation state, control the chemistry of the water used to moisten the clay poultices and contribute the critical antibacterial agent(s) that ultimately debilitate the bacteria.

Tannic acid-stabilized silver nanoparticles were synthesized in situ on halloysite clay nanotubes. The synthesis strategy included simple steps of tannic acid adsorption on clay nanotubes and further particle formation from silver salt solution. Pristine halloysite nanotubes as well as amino-modified clays were used for silver stabilization in water or ethanol. The materials were tested for antibacterial performance using three different methods. All of the materials produced showed antimicrobial activity. The pristine halloysite-based material with ~5 nm particles produced using ethanol as the solvent and tannic acid as the reducing agent showed the greatest antibacterial activity against Serratia marcescens. The materials were tested in vivo on Caenorhabditis elegans nematodes to ensure their safety, and they showed no negative effects on nematode growth and life expectancy.

Previous scientific studies have listed chemical compounds from sea cucumber species. The ability to produce bioactive metabolites derives from their response to various ecological stresses and avoidance of predation. In view of this, the present work aimed to evaluate the antibacterial activity and quantitatively assess the active components of different organs of the sea cucumber Holothuria foskali collected from the north-east of Tunisia. The antibacterial activity of ethyl acetate extracts was tested against Bacillus subtilis, Bacillus cereus, Escherichia coli and Pseudomonas aeruginosa. It was revealed that Escherichia coli and Bacillus subtilis were inhibited by the sea cucumber extracts. To identify the composition of these extracts, Gas Chromatography-Mass Spectroscopy (GC-MS) and Liquid Chromatography-Mass Spectroscopy (LC-MS) methods were used. The GC-MS revealed the presence of 25 major components, the most concentrated ones were characterized as C26H27N5O6 Morphinan-4,5-epoxy-3,6-diol, 6 amino;C17H31F3O2 3-trifluoroacetoxypentadecane; C31H50O2 Stigmasta-5,22-dien-3-ol,acetate; C11H14O2 1-Phthalanol,1,3,3-trimethyl 1-phenyl; C30H48O2 Ergosta-5,22-dien-3-ol,acetate and C25H40O2 (9E)-8-Phenyl-9-octadecenoic acid methyl ester; while the LC-MS revealed the presence of eight molecules, identified as phenolic acid, phytosterol and fat-soluble vitamins. Some molecules are interesting and deserve to be explored for their potential uses as ingredients in functional foods or as raw materials for new drug forms.

Considerable inhibitory antibacterial actions were exerted by the soybean 11S subunit comparable with nisin on the proliferation of total viable count, Pseudomonas count and Enterobacteriaceae count in bovine milk stored at 4 or 25 °C for 30 d and 48 h, while 7S and lysozyme were much less effective. The maximum magnitudes of bacterial reduction by 11S and nisin were in the range 2–4 log CFU/ml. The proliferation of 3 pathogenic bacteria (Listeria monocytogenes, Salmonella Enteritidis and Escherichia coli O157:H7) artificially inoculated into raw milk stored at 4 or 25 °C were particularly and significantly (P<0·05) reduced by 11S subunit and nisin (0·5% w/v), but only slightly by 7S and moderately by lysozyme. Lactose consumption, acidity development and casein degradation during storage of bovine raw milk were attenuated during storage at 4 or 25 °C and sensorial traits were better maintained by supplementation with 11S (0·5% w/v). 11S subunit may be used a safely food preservative, if permitted.

Bioactive compounds of gonad, respiration tree, cuvierian organ, and body wall of the sea cucumber Holothuria leucospilota collected from the north coast of the Persian Gulf were extracted using ethyl acetate, methanol and water–methanol solvents. Extracts were evaluated for their antibacterial and antifungal activities against Aspergillus niger, Candida albicans, Staphylococcus aureus, Pseudomonas aeruginosa and Escherichia coli. The activity was determined using the disc diffusion test. Cytotoxic activities of the extracts were determined by brine shrimp lethality assay. Results demonstrated that the A. niger was shown to be the most sensitive microorganism followed by C. albicans. The inhibition zone against A. niger and C. albicans had minimum inhibitory concentrations ranging from 3 to 7 μg/ml. The highest antifungal activity was found in G (water–methanol) with an inhibition zone of 50 mm against A. niger at 18 μg/ml extract concentration continuing with CT (methanol) with 46 mm inhibition zone against A. niger at 18 μg/ml extract concentration. The highest cytotoxic effect of methanol extract was found with LC50 values of about 40.31 μg/ml in the gonad organ from H. leucospilota continuing with the respiration tree organ with LC50 values of about 72.49 μg/ml.

Immunomodulatory actions of vitamin D have been recognised for over a quarter of a century, but it is only in the last few years that the significance of this to normal human physiology has become apparent. Two key factors have underpinned this revised perspective. Firstly, there are increasing data linking vitamin insufficiency with prevalent immune disorders. Improved awareness of low circulating levels of precursor 25-hydroxyvitamin D in populations across the globe has prompted epidemiological investigations of health problems associated with vitamin D insufficiency. Prominent among these are autoimmune diseases such as multiple sclerosis, type 1 diabetes and Crohn's disease, but more recent studies indicate that infections such as tuberculosis may also be linked to low 25-hydroxyvitamin D levels. The second factor expanding the link between vitamin D and the immune system is our improved knowledge of the mechanisms that facilitate this association. It is now clear that cells from the immune system contain all the machinery needed to convert 25-hydroxyvitamin D to active 1,25-dihydroxyvitamin D, and for subsequent responses to 1,25-dihydroxyvitamin D. Mechanisms such as this are important for promoting antimicrobial responses to pathogens in macrophages, and for regulating the maturation of antigen-presenting dendritic cells. The latter may be a key pathway by which vitamin D controls T-lymphocyte (T-cell) function. However, T-cells also exhibit direct responses to 1,25-dihydroxyvitamin D, notably the development of suppressor regulatory T-cells. Collectively these observations suggest that vitamin D is a key factor linking innate and adaptive immunity, and both of these functions may be compromised under conditions of vitamin D insufficiency.

The relative antibacterial activities of excretion/secretion (ES) from two carrion-feeding insects, Calliphora vicina Robineau-Desvoidy and Dermestes maculatus DeGeer, and a detritivore, Tenebrio molitor Linnaeus, were compared to that of Lucilia sericata Meigen, a species with ES of known antibacterial capacity, in order to explore the antimicrobial potential of other carrion and detritivore species. Viable counts were used to assess time-kill of ES against five bacterial species, Staphylococcus aureus, Escherichia coli, Bacillus cereus, Pseudomonas aeruginosa and Proteus mirabilis. Antibacterial activity was recorded in all four insect species although T. molitor and D. maculatus were the most effective in controlling growth of P. mirabilis. The blowflies were more effective in controlling a wider range of both Gram-positive and Gram-negative bacteria. The larval ES from all species was shown to reduce bacterial growth rate although differences in antibacterial spectrum were noted and the degree of potency varied between the four species. These differences may be explained ecologically by the different colonisation times of each insect species on the corpse. Overall, this study demonstrates that research into other carrion-feeding insect species has potential to provide an increased source of antimicrobial chemicals to broaden the range of bacterial species beyond that currently controlled using L. sericata.

The in vitro digestion of caprine whey proteins was investigated by a two-step degradation assay, using human gastric juice (HGJ) at pH 2·5 and human duodenal juice (HDJ) at pH 7·5. Different protein and peptide profiles were observed after the first (HGJ) and second (HDJ) enzymatic degradation. The minor whey proteins serum albumin, lactoferrin and Ig were rapidly degraded by HGJ, while α-lactalbumin (α-LA) and β-lactoglobulin (β-LG) were more resistant and survived both 30 and 45min of the enzymatic treatment. Further digestion with HDJ still showed intact β-LG, and the main part of α-LA also remained unchanged. The protein degradation by HGJ and HDJ was also compared with treatment by commercial enzymes, by using pepsin at pH 2·5, and a mixture of trypsin and chymotrypsin at pH 7·5. The two methods resulted in different caprine protein and peptide profiles. The digests after treatment with HGJ and HDJ were screened for antibacterial effects on some selected microorganisms, Escherichia coli, Bacillus cereus, Lactobacillus rhamnosus GG and Streptococcus mutans. Active growing cells of E. coli were inhibited by the digestion products from caprine whey obtained after treatment with HGJ and HDJ. Cells of B. cereus were inhibited only by whey proteins obtained after reaction with HGJ, while the products after further degradation with HDJ demonstrated no significant effect. Screenings performed on cells of Lb. rhamnosus GG and S. mutans all showed no signs of inhibition.

Acid-precipitated rabbit ‘whole casein’ was digested by trypsin, chymotrypsin, pepsin, and clostripain to screen for possible peptides with antibacterial properties. The peptide fragments were separated by reversed-phase chromatography. The collected fractions were pooled and their antibacterial properties tested against Escherichia coli, Bacillus subtilis and Staphylococcus lentus. Three antibacterial peptide fragments derived from tryptic digestion of rabbit casein were isolated and identified. Their sequences were found as follows: HVEQLLR (residues 50–56 of β-casein), ILPFIQSLFPFAER (residues 64–77 of β-casein), and FHLGHLK (residues 19–25 of αs1-casein). The three peptides were synthesized and found to exert antibacterial effect against Gram positive bacteria only. Proteolytic digestion of rabbit casein by chymotrypsin, pepsin and clostripain yielded several peptide fragments with antibacterial activity. Since antibiotic peptides can be released from casein during the digestion of milk proteins, our results suggest a possible antibacterial function of rabbit caseins. It is conceivable that antibacterial peptides can be generated by endopeptidases of the mammalian gastrointestinal tract possibly providing protection for new-born rabbits against aggression of micro-organisms.

Mussel (Mytilus galloprovincialis) plasma contains cytotoxic activity against both vertebrate (erythrocytes and mouse tumour) and protozoan cells. Procaryotes (Escherichia coli and Vibrio alginolyticus) were not sensitive to the cytotoxicity. The activity was still present in dialyzed samples but was inhibited by heating at 45 °C. Large individual variability which was not correlated with protein concentration and an increasing number of reactive specimens following injection, suggested that naive mussels were in various stages of immune response. Purification by anion exchange chromatography followed by gel filtration revealed a 320 kDa cytotoxic polymeric protein that acts through a polymerization process after binding onto target cell membranes as revealed by ultrastructural observation.

European and Pacific oysters (Ostrea edulis and Crassostrea gigas) expressed antibacterial activity against both Gram negative and Gram positive bacteria which was most probably due to small proteins. When tested against the marine pathogenic Vibrio alginolyticus, hemocyte lysates of both species were more active than cell-free plasma. Antibacterial activity showed significant individual variability that was dramatically reduced by stimulation through mechanical stress or injection. The number of spontaneously active Pacific oysters increased from 50 to 100% following a single injection of bacteria.

These results strongly support the view that bivalve molluscs possess sensitive immuno-defense mechanisms that will greatly aid the development of aquaculture systems by employing refined techniques of transgenesis.

Glycerol and ichthammol (G & I) has been used for generations by otologists. However, there is a paucity of information on both its mode of action and its anti-bacterial properties. The aim of this paper was to ascertain firstly, what the most common organisms found in discharging ears were and secondly, what antibacterial activity G & I had against these organisms. All ear swabs from 1992–1994 in our unit were reviewed to ascertain the prevalence of the commonly isolated organisms. Fresh isolates of these organisms were collected and plated onto agar with wells of glycerol, ichthammol and a combination of both as used in clinical practice. The diameters of the zones of inhibition observed after incubation were measured in millimetres. Common isolates were: Pseudomonas aeruginosa, Staphylococcus aureus, Proteus mirabalis, Streptococcus pyogenes in descending order of frequency. Pure glycerol showed no significant zones of inhibition against any of the organisms tested. The average zones of inhibition for G & I and ichthammol alone were for Staphylococcus aureus 15 mm and 18 mm and for Streptococcus pyogenes: 16 mm and 23 mm. Ichthammol alone was significantly more effective than G & I (p<0.001). There was no significant activity against Proteus mirabalis and Pseudomonas aeruginosa. The therapeutic benefit of G & I is due in part to the inherent anti-bacterial activity of ichthammol against the Gram positive organisms as well as its anti-inflammatory action and the dehydrating effect of the glycerol.

Three 9-acridone alkaloids, melicopicine, tecleanthine and 6-methoxy tecleanthine were isolated from the bark of Teclea trichocarpa. Melicopicine and tecleanthine exhibited mild antifeedant activity against the African armyworm, Spodoptera exempta. All the three alkaloids showed antimicrobial activity against the fungus, Cladosporium cucumerinum and the bacterium, Bacillus subtilis.
