Studies on the in vitro exsheathment of Brugia pahangi—2. The in vitro exsheathment of B. pahangi microfilariae incubated with mosquito tissues and cells

Abstract

Brugia pahangi microfilariae obtained from peritoneal washings of jirds (Meriones unguiculatus) were incubated with midgut tissue of Culex quinquefasciatus and Aedes aegypti (susceptible stock), in the presence or absence of blood and also in thoracic tissue and salivary glands in various media. Exsheathment took place in the presence of midgut tissue with or without blood and in thoracic tissue, but not in the presence of salivary glands.

Percentage exsheathment was variable and not reproducible for each type of tissue. Exsheathment also occurred in the presence of mosquito cells in suspension and in monolayers. In the latter, percentage exsheathment in some cases was higher than that observed in vivo in the midgut of C. quinquefasciatus, which is refractory to B. pahangi microfilariae.

Résumé

Les microfilaires du Brugia pahangi obtenus á partir des produits de lavage (periotenal) des mériones (Meriones unguiculatus) étaient soumis á l'accouvage avec des tissus des mi-intestins de la souche du Culex quinquefasciatus et d'une éspece susceptible d'Aedes aegypti, en présence ou en l'absence du sang, dans des tissus thoraciques et dans des glandes salivaires soumis a divers environnements.

Le dégagement eut lieu en présence des tissues des mi-intestins, en présence on en l'absence de sang, dans des tissus thoraciques, mais pas en presence des glandes salivaires.

Le niveau du dégagement était variable et non reproductible par chaque catégorie de tissu. Le dégagement eut également lieu en présence des cellules de moustique en suspension ou en couches simples. Dan le dernier cas, le niveau de dégagment était, dans certains das, plus élevé que celui observé avec des mi-intestins de la souche du C. quinquefasciatus in vivo, qui est réfractair aux microfilaires du B. pahangi.
