Maintenance of the cotton bollworm, Heliothis armigera Hübner (Lepidoptera: Noctuidae) in laboratory culture — I. Rearing on semi-synthetic diet

Abstract

Based on a semi-synthetic diet, a standard rearing method for the cotton bollworm, Hetiothis armigera Hübner, is described. The diet is composed of chickpea (Cicer arietinum) flour, yeast powder, sucrose, ascorbic acid, cholesterol, choiine chloride, wheat germ oil, vitamin mixture and a combination of preservatives. A plastic container for individual rearing of the cannibalistic larvae and a cage for mating of adult moths are described. After inoculation with the neonate larvae, development of the different stages was synchronouson this diet. The larval and pupal development period was 14.3 ± 1.2 and 11.7 ± 0.82 days, respectively. Females reared on this diet produced an average of 2000 eggs. Adult longevity was 22 days. The female moths retained their oviposition rhythm even after 3 years of laboratory rearing. A method for introduction of wild populations into the laboratory culture is discussed. The reliability of the technique was checked by monitoring the insects' performance for at least 25 generations.

Résumé

Dans le présent mémoire, nous présentons la composition d'un régime artificiel ainsi que la mise au point d'un procédé permettant l'élevage au laboratoire de la chenille des épis du maïs (Heliothis armigera). Ce régime se compose de farine de pois chiche (Cicer arietinum), de poudre de levure, de saccharose, d'acide ascorbique, de cholestérol, de chlorure de choiine, d'huile de germe de blé, d'un mélange de vitamines et de 4 antibiotiques. Nouse décrivons une simple boîte à compartlments qui sert à l'élevage individuel des larvescannibales ainsi qu'une cage d'accouplement pour les papillons adultes. En utilisant ce régime, le développement des différentes stades était bien synchronisé. La durée du développernent s'élevait à 14.3 ± 1.2 jours pour les larves et à 11.7 ± 0.82 jours pour les pupes. Des observations d'oviposition ont montré que l'élevage des femelles sur ce régime entraîne la ponte d'un grand nombre d'oeufs. Une longévité de 22 jours a été mise au point pour les insectes adultes; les papillons femelles ont pu maintenir leur rythme de ponte mêrne au bout de plusieurs années d'élevage au laboratoire. On discute une méthode sûre pour introduire des populations sauvages dans la culture de laboratoire. La solidité des techniques décrites ici a été mise à l'épreuve en examinant l'activité des insectes au cours de plusieurs générations.
