Efficient life support systems are needed to maintain adequate oxygen, water and food for humans in extraterrestrial conditions. On the near-Earth missions, these are supplied by transport from the Earth, and by physical and chemical cleaning and recycling, but on long-term missions to far-away destinations, such as Mars, on-site production of the consumables may be required. Molecular oxygen and organic biomass can be most efficiently produced biologically, i.e. by photosynthesis. The conditions on Mars are distinctly harsh, and they strictly limit the growth and survival of any photosynthetic organisms to artificially maintained containments. For obtaining most economical growth conditions, minimal parameters need to be determined which still allow efficient growth of photosynthetic organisms. In this work we are testing how reduced air pressures (hypobaria) and increased CO2 concentrations, i.e. features typical for Martian conditions, affect the durability, growth and photosynthesis of laboratory strains of cyanobacteria, a group of prokaryotic organisms capable of plant-like photosynthesis. Our preliminary results show that air pressures down to 0.1 atm or CO2 concentrations up to 20% have no harmful effect on the photosynthetic oxygen production or growth rate of the cyanobacterial model species, Synechocystis sp. PCC 6803.

Astrobiology is a discipline that is expanding its field of investigation not only in the natural sciences, but also in the social sciences. It is for this reason that the ethical aspects are progressively emphasized leading to a point where the whole field requires a specific handling. The appellation ‘astrobioethics’ is now considered as not only relevant, but also a true issue for the future of Astrobiology. Astrobioethics is the subsection within astrobiology that is accountable for studying the moral implications of, for example, bringing humans to Mars, the Planetary Protection Policy, the social responsibility of the astrobiologist to society, etc. It is in this way that the present article outlines a path for astrobioethics, as being a fertile field of study and an opportunity to trade scientific knowledge in a transdisciplinary way.

In a previous paper, we showed that Earth-type habitable planets around 47 UMa are in principle possible if a distinct set of conditions is warranted. These conditions include that the Earth-type planets have successfully formed and are orbitally stable and, in addition, that the 47 UMa star–planet system is relatively young ([lsim ]6 Gyr). We now extend this study by considering Earth-like planets with different land/ocean coverages. This study is again based on the so-called integrated system approach, which describes the photosynthetic biomass production taking into account a variety of climatological, biogeochemical and geodynamical processes. This approach implies a special characterization of the habitable zone defined for a distinct type of planet. We show that the likelihood of finding a habitable Earth-like planet on a stable orbit around 47 UMa critically depends on the percentage of the planetary land/ocean coverage. The likelihood is significantly increased for planets with a very high percentage of ocean surface (‘water worlds’).

The astonishing capability of life to adapt to extreme conditions has provided a new perspective on what ‘habitable’ means. On Earth extremophiles thrive in hostile habitats, such as hot and cold deserts or Antarctic sub-glacial lakes considered as Earth analogues of Mars and icy moons of Jupiter and Saturn. Recently desert cyanobacteria were exposed to ground-based simulations of space and Martian conditions and to real space and Martian conditions simulated in low Earth orbit using facilities attached outside the International Space Station. When exposure to such conditions does not exceed repair capabilities, more data are available regarding the physico-chemical constraints that life can withstand. When the accumulated damage exceeds the survival potential, the persistence of biomarkers contributes to the search for life elsewhere. Knowledge concerning the endurance of desert cyanobacteria under space and Martian conditions contributes to the development of life support systems.

The National Aeronautics and Space Administration (NASA) and the European Space Agency (ESA) are studying how samples might be brought back to Earth from Mars safely. Backward planetary protection is key in this complex endeavour, as it is required to prevent potential adverse effects from returning materials to Earth's biosphere. As the question of whether or not life exists on Mars today or whether it ever did in the past is still unanswered, the effort to return samples from Mars is expected to be categorized as a ‘Restricted Earth Return’ mission, for which NASA policy requires the containment of any unsterilized material returned to Earth. NASA is investigating several solutions to contain Mars samples and sterilize any uncontained Martian particles. This effort has significant implications for both NASA's scientific mission, and the Earth's environment; and so special care and vigilance are needed in planning and execution in order to assure acceptance of safety to Earth's biosphere. To generate a technically acceptable sterilization process across a wide array of scientific and other stakeholders, on 30–31 January 2019, 10–11 June 2019 and 19–20 February 2020, NASA informally convened a Sterilization Working Group (SWG) composed of experts from industry, academia and government to assess methods for sterilization and inactivation, to identify future work needed to verify these methods against biological challenges, and to determine their feasibility for implementation on robotic spacecraft in deep space. The goals of the SWG were:

1. (1) Understand what it means to sterilize and/or inactivate Martian materials and how that understanding can be applied to the Mars Sample Return (MSR) mission.

2. (2) Assess methods for sterilization and inactivation, and identify future work needed to verify these methods.

3. (3) Provide an effective plan for communicating with other agencies and the public.

This paper provides a summary of the discussions and conclusions of the SWG over these three workshops. It reflects a consensus position based on qualitative discussion of how agencies might approach the problem of sterilization of Mars material. The SWG reached a consensus that sterilization options can be considered on the basis of biology as we know it, and that sterilization modalities that are effective on terrestrial materials and organisms should be part of the MSR planetary protection strategy. Conclusions pointed to several industry standards for sterilization to include heat, chemical, UV radiation and low-heat plasma. Technical trade-offs for each sterilization modality were discussed while simultaneously considering the engineering challenges and limitations for spaceflight. Future work includes more in-depth discussions on technical trade-offs of sterilization modalities, identifying and testing Earth analogue challenge organisms and proteinaceous molecules against chosen modalities, and executing collaborative agreements between NASA and external working group partners to help close data gaps, and to establish strong, scientifically grounded sterilization and inactivation standards for MSR.

The Search for Extraterrestrial Intelligence (SETI) has thus far failed to provide a convincing detection of intelligent life. In the wake of this null signal, many ‘contact-pessimistic’ hypotheses have been formulated, the most famous of which is the Rare Earth Hypothesis. It postulates that although terrestrial planets may be common, the exact environmental conditions that Earth enjoys are rare, perhaps unique. As a result, simple microbial life may be common, but complex metazoans (and, hence, intelligence) will be rare. In this paper we use Monte Carlo realization techniques to investigate the Rare Earth Hypothesis, in particular the environmental criteria considered imperative to the existence of intelligence on Earth. By comparing with a less restrictive, more optimistic hypothesis, the data indicate that if the Rare Earth hypothesis is correct, intelligent civilization will indeed be relatively rare. Studying the separations of pairs of civilizations shows that most intelligent civilization pairs (ICPs) are unconnected: that is, they will not be able to exchange signals at lightspeed in the limited time that both are extant. However, the few ICPs that are connected are strongly connected, being able to participate in numerous exchanges of signals. This may provide encouragement for SETI researchers: although the Rare Earth Hypothesis is in general a contact-pessimistic hypothesis, it may be a ‘soft’ or ‘exclusive’ hypothesis, i.e. it may contain facets that are latently contact-optimistic.

We discuss whether sulphur traces on Jupiter's moon Europa could be of biogenic origin. The compounds detected by the Galileo mission have been conjectured to be endogenic, most likely of cryovolcanic origin, due to their non-uniform distribution in patches. The Galileo space probe first detected the sulphur compounds, as well as revealing that this moon almost certainly has a volcanically heated and potentially habitable ocean hiding beneath a surface layer of ice. In planning future exploration of Europa there are options for sorting out the source of the surficial sulphur. For instance, one possibility is searching for the sulphur source in the context of the study of the Europa Microprobe In Situ Explorer (EMPIE), which has been framed within the Jovian Minisat Explorer Technology Reference Study (ESA). It is conceivable that sulphur may have come from the nearby moon Io, where sulphur and other volcanic elements are abundant. Secondly, volcanic eruptions in Europa's seafloor may have brought sulphur to the surface. Can waste products rising from bacterial colonies beneath the icy surface be a third alternative significant factor in the sulphur patches on the Europan surface? Provided that microorganisms on Europa have the same biochemical pathways as those on Earth, over geologic time it is possible that autochthonous microbes can add substantially to the sulphur deposits on the surface of Europa. We discuss possible interpretations of the non-water-ice elements (especially the sulphur compound mercaptan) in the context of the studies for future missions. To achieve reliable biosignatures it seems essential to go back to Europa. Our work highlights the type of biogenic signatures that can be searched for when probing Europa's icy and patchy surface.

At present, bulk liquid water on the surface and near-subsurface of Mars does not exist due to the scarcity of condensed- and gas-phase water, pressure and temperature constraints. Given that the nuclei of soil and ice, that is, the soil solid and ice lattice, respectively, are coated with adsorbed and/or thin liquid films of water well below 273 K and the availability of water limits biological activity, we quantify lower and upper limits for the thickness of such adsorbed/water films on the surface of the Martian regolith and for subsurface ice. These limits were calculated based on experimental and theoretical data for pure water ice and water ice containing impurities, where water ice containing impurities exhibit thin liquid film enhancements, ranging from 3 to 90. Close to the cold limit of water stability (i.e. 273 K), thin liquid film thicknesses at the surface of the Martian regolith is 0.06 nm (pure water ice) and ranges from 0.2 to 5 nm (water ice with impurities). An adsorbed water layer of 0.06 nm implies a dessicated surface as the thickness of one monolayer of water is 0.3 nm but represents 0.001–0.02% of the Martian atmospheric water vapour inventory. Taking into account the specific surface area (SSA) of surface-soil (i.e. top 1 mm of regolith and 0.06 nm adsorbed water layer), shows Martian surface-soil may contain interfacial water that represents 6–66% of the upper- and lower-limit atmospheric water vapour inventory and almost four times and 33%, the lower- and upper-limit Martian atmospheric water vapour inventory. Similarly, taking the SSA of Martian soil, the top 1 mm or regolith at 5 nm thin liquid water thickness, yields 1.10×1013 and 6.50×1013 litres of waters, respectively, 55–325 times larger than Mars’ atmospheric water vapour inventory. Film thicknesses of 0.2 and 5 nm represent 2.3×104–1.5×106 litres of water, which is 6.0×10−7–4.0×10−4%, respectively, of a 10 pr μm water vapour column, and 3.0×10−6–4.0×10−4% and 6.0×10−6–8.0×10−4%, respectively, of the Martian atmospheric water vapour inventory. Thin liquid film thicknesses on/in subsurface ice were investigated via two scenarios: (i) under the idealistic case where it is assumed that the diurnal thermal wave is equal to the temperature of ice tens of centimetres below the surface, allowing for such ice to experience temperatures close to 273 K and (ii) under the, likely, realistic scenario where the diurnal thermal wave allows for the maximum subsurface ice temperature of 235 K at 1 m depth between 30°N and 30°S. Scenario 1 yields thin liquid film thicknesses ranging from 11 to 90 nm; these amounts represent 4×106–3.0×107 litres of water. For pure water ice, Scenario 2 reveals that the thickness of thin liquid films contained on/within Martian subsurface is less than 1.2 nm, several molecular layers thick. Conversely, via the effect of impurities at 235 K allows for a thin liquid film thickness on/within subsurface ice of 0.5 nm, corresponding to 6.0×104 litres of water. The existence of thin films on Mars is supported by data from the Mars Exploration Rovers (MERs) Spirit and Opportunity's Alpha Proton X-ray Spectrometer instrumentation, which have detected increased levels of bromine beneath the immediate surface, suggestive of the mobilization of soluble salts by thin films of liquid water towards local cold traps. These findings show that biological activity on the Martian surface and subsurface is not limited by nanometre dimensions of available water.

The FT-Raman spectra are described of green and red snow algae, Chlamydomona, involved in the colonization of exposed surfaces of the McLeod Glacier, Jane Col, Signy Island, situated at the northern edge of the Weddell Sea, Antarctica. The protective biochemicals produced by these extremophilic algae give rise to the so-called watermelon snow of Alpine regions. The red colour of the senescent algae is shown to derive from the accumulation of carotenoids and a deficiency of chlorophyll believed to arise from UV-radiation induced breakdown into phaecophytin. A comparison of the Raman spectra of young (green) and old (red) algae is effected and possible bio-markers for spectral detection on extraterrestrial icy moons and planets are identified.

In this paper, we describe the development of an International Space Station experiment, BioRock. The purpose of this experiment is to investigate biofilm formation and microbe–mineral interactions in space. The latter research has application in areas as diverse as regolith amelioration and extraterrestrial mining. We describe the design of a prototype biomining reactor for use in space experimentation and investigations on in situ Resource Use and we describe the results of pre-flight tests.

The Square Kilometre Array (SKA) will operate in frequency ranges often used by military radar and other communications technology. It has been shown that if extraterrestrial intelligences (ETIs) communicate using similar technology, then the SKA should be able to detect such transmissions up to distances of ~100 pc (~300 light years) from Earth. However, Mankind has greatly improved its communications technology over the last century, dramatically reducing signal leakage and making the Earth ‘radio quiet’. If ETIs follow the same pattern as the human race, will we be able to detect their signal leakage before they become radio quiet? We investigate this question using Monte Carlo realization techniques to simulate the growth and evolution of intelligent life in the Galaxy. We show that if civilizations are ‘human’ in nature (i.e. they are only ‘radio loud’ for ~100 years, and can only detect each other with an SKA-like instrument out to 100 pc, within a maximum communication time of 100 years), then the probability for such civilizations accidentally detecting each other is low (~10−7), much lower than if other, dedicated communication techniques are permissible (e.g. optical SETI or neutrino communication).

Macromolecular organic materials in chondrites display significant variations in carbon and nitrogen stable isotopes. In recent years, these variations have been interpreted as a record of aqueous and thermal processing on asteroids shortly after the birth of the Solar System. In this paper we review and summarize the key data and main interpretative approaches related to this study area. Armed with these methods we attempt to reinterpret the whole rock chondrite data set in the literature.

The ongoing search for life on other worlds and the prospects of eventual human exploration of the Moon and Mars indicate the need for new ethical guidelines to direct our actions as we search and how we respond if we discover microbial life on other worlds. Here we review how life on other worlds presents a novel question in environmental ethics. We propose a principle of protecting and expanding the richness and diversity of life as the basis of an ethic for astrobiology research and space exploration. There are immediate implications for the operational policies governing how we conduct the search for life on Mars and how we plan for human exploration throughout the Solar System.

This paper highlights unique sites in Ladakh, India, investigated during our 2016 multidisciplinary pathfinding expedition to the region. We summarize our scientific findings and the site's potential to support science exploration, testing of new technologies and science protocols within the framework of astrobiology research. Ladakh has several accessible, diverse, pristine and extreme environments at very high altitudes (3000–5700 m above sea level). These sites include glacial passes, sand dunes, hot springs and saline lake shorelines with periglacial features. We report geological observations and environmental characteristics (of astrobiological significance) along with the development of regolith-landform maps for cold high passes. The effects of the diurnal water cycle on salt deliquescence were studied using the ExoMars Mission instrument mockup: HabitAbility: Brines, Irradiance and Temperature (HABIT). It recorded the existence of an interaction between the diurnal water cycle in the atmosphere and salts in the soil (which can serve as habitable liquid water reservoirs). Life detection assays were also tested to establish the best protocols for biomass measurements in brines, periglacial ice-mud and permafrost melt water environments in the Tso-Kar region. This campaign helped confirm the relevance of clays and brines as interest targets of research on Mars for biomarker preservation and life detection.

The ‘holy grail’ in planet hunting is the detection of an Earth-analogue: a planet with similar mass as the Earth and an orbit inside the habitable zone. If we can find such an Earth-analogue around one of the stars in the immediate solar neighbourhood, we could potentially even study it in such great detail to address the question of its potential habitability. Several groups have focused their planet detection efforts on the nearest stars. Our team is currently performing an intensive observing campaign on the α Centauri system using the High Efficiency and Resolution Canterbury University Large Échelle Spectrograph (Hercules) at the 1 m McLellan telescope at Mt John University Observatory in New Zealand. The goal of our project is to obtain such a large number of radial velocity (RV) measurements with sufficiently high temporal sampling to become sensitive to signals of Earth-mass planets in the habitable zones of the two stars in this binary system. Over the past few years, we have collected more than 45 000 spectra for both stars combined. These data are currently processed by an advanced version of our RV reduction pipeline, which eliminates the effect of spectral cross-contamination. Here we present simulations of the expected detection sensitivity to low-mass planets in the habitable zone by the Hercules programme for various noise levels. We also discuss our expected sensitivity to the purported Earth-mass planet in a 3.24-day orbit announced by Dumusque et al. (2012).

Episodes of species mass extinction dramatically affected the evolution of life on Earth, but their causes remain a source of debate. Even more controversy surrounds the hypothesis of periodicity in the fossil record, with conflicting views still being published in the scientific literature, often even based on the same state-of-the-art datasets. From an empirical point of view, limitations of the currently available data on extinctions and possible causes remain an important issue. From a theoretical point of view, it is likely that a focus on single extinction causes and strong periodic forcings has strongly contributed to this controversy. Here I show that if there is a periodic extinction signal at all, it is much more likely to result from a combination of a comparatively weak periodic cause and various random factors. Tests of this unified model of mass extinctions on the available data show that the model is formally better than a model with random extinction causes only. However, the contribution of the periodic component is small compared to factors such as impacts or volcanic eruptions.

Long space flights and planetary settlement will require sources of nutrition and chemicals that must be generated in space. This will include not only amino acids and vitamins but also oxygen, all of which can be generated by means of biosynthesis. Synthetic biology has the potential to generate organisms designed for supplying human nutritional needs in space. Photosynthetic microbes may be ideal for this purpose, as they are more efficient per volume cultivated than green plants at conversion of light to chemical energy, biomass and nutritional molecules. In addition, microbes are easier and faster to genetically engineer, facilitating not only design and terrestrial manufacture of organisms optimized for growth and nutrient production in the artificial conditions of space, but superior ability in space to develop organisms suited to newly discovered environments. The rapid ability to adapt and create new microbes to suit new circumstances when in space offers significant potential for risk reduction. Development of sun-driven microbial production of nutritional chemicals would also have terrestrial benefits in commerce and sustainability. A synthetic biology approach to chemical production would not be based on fossil fuels as such fuels do not exist on other planets. This approach would highlight a synergistic relationship between outer space and ‘spaceship earth’, illustrating NASA's role in stimulating technology development with terrestrial application. Two specific approaches deserve consideration: production by traditional photosynthetic microbes, or by the newly appreciated capacity of some bacteria to absorb electric current (e.g. solar panels) to drive metabolism. Palatability and sensory stimulation are a key part of food consumption and could be engineered into microbes. As a first step, NASA should test a bioreactor in which genetically engineered, nutrient-producing photosynthetic bacteria are grown and harvested in space.

We studied the effects of soft X-rays radiation on free and clay (montmorillonite, kaolinite) adsorbed DNA. The DNA samples were exposed to X-rays of 1.49, 4.51 and 8.04 keV for exposure times ranging from 2 min up to 16 h. The biological transformation technique was used to estimate the damage of the DNA molecules. Free and clay adsorbed DNA are differently affected by X-rays. The former is damaged by X-rays and the level of damage depends on the energy dose rather than the hardness of the radiation. The clay adsorbed DNA is not damaged by X-rays for energy doses up to 5.8×104 erg. Clays materials could have protected the building blocks of life on the primordial Earth when the solar X-ray emission was much stronger than today.

Subglacially erupted Neogene basaltic hyaloclastites in lava-fed deltas in Antarctica were found to contain putative endolithic microborings preserved in fresh glass along hydrous alteration boundaries. The location and existence over the past 6 Ma of these lava deltas has exposed them to successive interglacials and subsequent percolation of the hyaloclastite with marine water. A statistical study of the hyaloclastites has found that endolithic microborings are distinctly more abundant within samples that show evidence for marine alteration, compared with those that have remained in a strictly freshwater (glacial) environment. Additionally, correlation between elevation and the abundance of microborings shows endolithic activity to be more prolific within lower elevation samples, where the hyaloclastites were influenced by marine fluids. Our study strongly suggests that endolithic microborings form more readily in marine-influenced, rather than freshwater environments. Indeed, marine fluids may be a necessary precondition for the microbial activity responsible. Thus, we suggest that the chemistry and origin of alteration fluids are controlling factors on the formation of endolithic microborings in basaltic glass. The study also contributes to the understanding of how endolithic microborings could be used as a biosignature on Mars, where basaltic lavas and aqueous alteration are known to have existed in the past.

Template-directed polymer assembly is a likely feature of prebiotic chemistry, but the product blocks further synthesis, preventing amplification and Darwinian selection. Nucleic acids are unusual because charge repulsion between opposing phosphates permits salt-dependent association and dissociation. It was postulated (Lathe, R. (2004). Fast tidal cycling and the origin of life. Icarus168, 18–22) that tides at ocean shores provide the driving force for amplification: evaporative concentration promoted association/assembly on drying, while charge repulsion on tidal dilution drove dissociation. This permits exponential amplification by a process termed here the tidal chain reaction (TCR). The process is not strictly contingent upon tidal ebb and flow: circadian dews and rainfalls can produce identical cycling. Ionic strength-dependent association and dissociation of nucleic acids and possible prebiotic precursors are reviewed. Polymer scavenging, chain assembly by the recruitment of pre-formed fragments, is proposed as the primary mechanism of reiterative chain assembly. Parameters determining prebiotic polymer structure and amplification by TCR are discussed, with the suggestion that Darwinian selection may have operated on families of related polymers rather than on individual molecules.
