1. Experiments were conducted to study the effects of dietary vicine (2, 6-diamino-4, 5 dihydroxy pyrimidine-5 (β-D-glucopyranoside)) and supplemental vitamin E on the performance of laying hens and growing chicks.

2. Chicks fed on diets that contained vicine had similar growth rates but slightly higher levels of spontaneous haemolysis of erythrocytes than birds fed on a control diet.

3. Vicine when fed to laying hens had a very dramatic effect. It depressed food consumption, egg weight, fertility and hatchability of eggs, packed cell volume and erythrocyte haemoglobin levels and led to increased liver weights, liver glutathione levels, liver and plasma lipid levels, plasma lipid peroxide levels and erythrocyte haemolysis in vitro. Liver protein and plasma vitamin E:lipid levels were not altered. Vitamin E supplementation slightly increased egg weights, markedly improved fertility and hatchability of eggs and lowered liver weights and lipid levels but did not affect the other factors examined.

4. It is concluded that vicine which was isolated from faba beans (Vicia faba L.) has a marked influence on the metabolism of the laying hen and only a slight effect on the growing chick. Vicine or its metabolites or both cause peroxidation of cellular components which result in abnormal lipid transport or synthesis or both, increased fragility of erythrocytes, and reduced fertility. These effects are overcome to varying extents by supplemental vitamin E.

1. Male rats were undernourished either during the geslational and suckling periods or for a period of time immediately following weaning. Some rats were killed at the end of the period of undernutrition; others were nutritionally rehabilitated for lengthy periods of time before examination. Two muscles, the extensor digitorum longus (EDL) and soleus (SOL) were studied from each rat. Histochemically-stained transverse sections of these muscles were used to determine total number of fibres, the fibre cross-sectional areas and the relative frequency of the various fibre types.

2. All rats killed immediately following undernutrition showed significant deficit sin body-weight, muscle weight and fibre cross-sectional area compared to age-matched controls.

3. Animals undernourished during gestation and suckling and then fed normally for 5 months showed persistent and significant deficits in body-weight, muscle weight and total fibre number. There were also significant deficits in mean fibre cross-sectional area of each fibre type except for red fibres in the EDL. No difference in the volume proportion of connective tissue was found.

4. Rats undernourished after weaning and then fed ad lib. for approximately 7 months had normal body-and muscle weights. Their muscles showed no significant differences in total fibre number, relative frequency of the various fibre types, fibre size or volume proportion of connective lissue.

5. These results indicate that, although the effects on rat skeletal muscle of a period of undernutrition after weaning can be rectified, undernutrition before weaning causes lasting deficits.

1. Rats were fed to appetite on a stock laboratory diet or on diets consisting of the stock diet and in addition 50 or 200 g triolein/kg, 50 g palmitic acid/kg or 50 g/kg of a concentrate mixture of methyl branched-chain fatty acids (Me-BCFA) which had been prepared from sheep adipose triacylglycerols.

2. No differences could be detected in the Δ9-desaturase activity or fatty acid synthetase activity of liver preparations from rats which had been fed on either the stock diet, the 50 g palmitic acid/kg or the 50 and 200 g triolein/kg diet; the palmitic acid diet was therefore taken as the control diet in subsequent experiments.

3. Rats consuming the 50 g Me-BCFA/kg diet exhibited a marked reduction in the capacity of their liver microsomes for Δ9-desaturation when compared with animals receiving the control diet. The Δ6-desaturase activity also showed an inhibitory trend with the Me-BCFA diet.

4. Microsomal ω-oxidation of fatty acids, mitochondrial succinate oxidation and the activity of cytosolic fatty acid synthetase (FAS) were unaffected by the ingestion of the Me-BCFA mixture compared with the diet which included palmitic acid.

5. There were no differences in the plasma concentrations of thyroxin, insulin and glucagon between animals fed on the diets containing palmitic acid or the Me-BCFA.

6. For a given concentration of fatty acids the Me-BCFA had a greater inhibitory effect when added to incubations of liver microsomes from rats fed on the standard diet than did the addition of palmitic acid.

7. The observations in vivo and in vitro strongly suggested that the Me-BCFA were having a specific inhibitory effect on the desaturation reaction.

1. Basal levels of serum 25-hydroxy vitamin D (25-OHD) were estimated in ten normal children and twenty-seven children with protein–energy malnutrition.

2. Five normal children and fourteen malnourished children were administered a single massive dose of 15 mg vitamin D orally and the remaining children received oral supplements of 50 μg vitamin D daily for 20 d. Blood samples were obtained after 10 and 20 d of dosing and serum 25-OHD levels were repeated.

3. The basal serum 25-OHD levels were significantly lower (P < 0·05) in malnourished children than in normal children.

4. Administration of a single massive dose of vitamin D as well as daily supplementation of small doses resulted in significant increases in serum 25-OHD levels within 10 d. However, the increase with a massive dose was of a higher magnitude. There was no significant difference between the normal and malnourished children.

5. The binding capacity of serum to 25-OHD was similar in both the groups.

6. These results suggest that vitamin D metabolism is not altered in protein–energy malnutrition.

1. The concentrations of vitreous humour and plasma glucose were closely correlated in both healthy and fatty liver and kidney syndrome-affected chicks at time of death.

2. The values of vitreous humour glucose and lactate decreased rapidly after death, such that they were not reliable indicators of the presence of hypoglycaemia immediately ante mortem.

3. Hepatic glycogen was extremely low in fatty liver and kidney syndrome-affected birds, whereas significant quantities remained in healthy birds up to at least 24 hpost mortem.

1. The contractile properties of soleus and extensor digitorum longus muscles from animals at 12 months of age whose mothers had been undernourished during pregnancy and lactation (PU rats) have been compared with those of muscles from age-matched controls.

2. Body-weight and muscle wet weight of PU rats was significantly reduced. Muscle: body-weight values were, however, no different from controls.

3. No significant alterations in whole muscle speeds of contraction or relaxation could be delected when compared with those of age-matched controls.

4. Twitch and tetanic forces of both SOL and EDL were greater per unit weight of muscle in PU rats. Apart from SOL twitch these differences were significant. The tetanus:twitch values were, however, not different.

1. Wether sheep were used, some of which were prepared with a Thiry-Vella loop of jejunum. The long-term use of these loops was ensured by regular perfusion with fresh nutrient solution.

2. The net calcium absorption rale from a Thiry-Vella loop of jejunum increased with increasing intraluminal Ca concentration and was increased by the addition of 1 α-hydroxy-cholecalciferol (3μg/l) to the loop fluid.

3. When the diet of sheep was changed from one which was normal in Ca to a diet low in Ca there was an increase in the efficiency of net Ca absorption from the jejunal loop. This dietary change was accompanied by an increase in the circulating concentration of 1,25-dihydroxy-cholecalciferol (1.25(OH)2D3).

4. An intravenous infusion of the Ca chelating agent EDTA increased the efficiency of net Ca absorption from the jejunal loop. The use of 47Ca demonstrated that this reflected an increase in the true absorption rate of Ca.

5. Dietary phosphorus deficiency reduced the efficiency of intestinal Ca absorption and was associated with a reduction in the plasma concentration of 1,25(OH)2D3.

1. Iron absorption was studied in weanling rats using balance techniques from semi-synthetic diets containing dried white bread (60·5 g dietary fibre/kg, White & Southgate, unpublished results), brown bread (130·2 g dietary fibre/kg) or wholemeal bread (221·2 g dietary fibre/kg) at a level of 300 g/kg and compared with a control group given a diet containing added FeSO4 at a similar Fe level to that for the bread groups. The dried bread contained 6·2–6·4 g phytate/kg.

2. Absorption of Fe was significantly higher (P < 0·001) in the control group (0·45) than in the white (0·28), brown (0·31) or wholemeal (0·24) groups.

3. A second experiment was carried out on 6-week-old rats in which the dried bread was extrinsically labelled with 59Fe and absorption from a single meal measured by both faecal excretion and incorporation of 59Fe into the blood. Control animals were given 59FeSO4 for comparison.

4. The excretion of 59Fe (% of administered dose) was significantly lower (P < 0·001) in the control group (31) than in the white (48), brown (45) or wholemeal (47) groups. After 10 d the control group had significantly more 59Fe in the blood than the bread groups, but there were no differences between the bread groups.

5. It appears that wheat bran fibre itself has no effect on the retention of Fe from the diet in the rat, when supplied in amounts similar to those found in commercially-available bread.

1. Sheep were fed on different diets of juice-extracted herbage to determine what effect juice-extraction had on reticulo-rumen motility.

2. The frequency of A and B sequences of contraction of the reticulo-rumen were recorded during eating, rumination and inactivity for continuous periods of 24–72 h by using integrated electromyograms obtained from electrodes implanted in the musculature of the reticulum and cranial dorsal rumen.

3. Animals were fed on herbage in which approximately 200 g/kg dry matter had been removed in juice extracted from ryegrass (Lolium perenne), white clover (Trifolium repens), mixed ryegrass–white clover and lucerne (Medicago saliva).

4. Over all the frequency of A sequences of contraction did not differ in animals fed on pressed herbage or the unpressed material from which it was derived, although it was slower during rumination on some of the pressed material. In contrast, the frequency of B sequences was higher on the pressed material. The frequencies of contraction of A and B sequences in animals fed on pressed herbage was related to the activity of the animals in the order eating > rumination > inactivity.

5. Changes in reticulo-rumen motility due to juice extraction were small and the frequencies of A and B sequences of contraction in sheep fed on pressed herbage were in the range encountered in ruminants consuming more conventional foods.

1. A detailed investigation was made of the motor performance of rats which had impaired cerebellar and muscular growth resulting from early-life undernutrition.

2. Developing rats were growth-retarded by underfeeding their mothers during pregnancy and lactation. They were fed ad lib. from 25 d of age.

3. Adult male previously-undernourished (PU) and well-fed control rats were given tests of motor behaviour commencing at 5 months of age. They were required to run backwards to maintain their position on a revolving drum. Aspects of the test situation were varied systematically: visual environment, interval between trials, test duration and drum speed.

4. PU rats had a 29% body-weight deficit compared with controls at 5 months.

5. There was no evidence of impaired motor performance by the PU rats under any of the test conditions.

1. Whole-body retention in vivo and uptake of 59Fe-labelled ascorbate and nitrilotriacetate chelates by intestinal slices in vitro were determined in groups of normal control rabbits and rabbits with experimentally-induced Fe deficiency.

2. Over-all absorption as measured by retention of doses of either chelate was greatly increased in conditions of Fe deficiency.

3. Intestinal Fe uptake in vitro was inhibited up to 77% in the presence of 2,4-dinitrophenol and sodium fluoride. Initial rates showed saturation within the concentration range 18–450 μmol/l, suggesting that uptake was brought about by an active transport process.

4. When studied at chelate concentrations of 450 μmol/l, significant regional differences in uptake rates were observed. Uptake in duodenal slices was increased when compared with slices from jejunum and ileum.

5. Fe uptake from ferric and ferrous chelates was greatly enhanced in Fe deficiency. This was chiefly due to increases in uptake by slices from the duodenum, but uptake into slices of distal intestine was also stimulated.

6. Kinetic analysis of Fe uptake by duodenal slices from animals rendered Fe deficient by diet or repeated bleeding indicated in both groups an increased apparent maximum velocity (Vmax) for influx of Fe without significant changes in apparent affinity for Fe.

7. The experiments provide further insight into the nature and regional distribution of transport of Fe into the intestine and suggest, in the rabbit, that important control of Fe absorption may be exerted by an active process operating at this initial entry step.

1. Glucose turnover (i.e. glucose entry and utilization rates) in fed and 48 h starved goats during pregnancy and lactation was determined using a continuous infusion of [U-14C]- and [3-3H]glucose.

2. Glucose synthesis and utilization increased during pregnancy and lactation in fed but not in starved goats.

3. Recycling of giucosc-C was approximately 10% in fed animals and 15–20% in starved animals and was unaffected by the stage of pregnancy or lactation.

4. Plasma glucose concentrations were maintained during pregnancy and lactation in fed goats but decreased during 48 h starvation in pregnant goats. Little change was seen in the plasma concentration of lipids and their metabolites during pregnancy and lactation in fed goats, but increases were observed after 48 h starvation.

5. The control of glucose metabolism in ruminants during pregnancy and lactation is discussed.

1. Utilizing the bromodeoxyuridine differential-chromatid labelling technique the in vitro proliferation of lymphocytes from children with kwashiorkor was followed in phytohaemagglutinin-stimulated cultures and compared with controls.

2. Analyses of first, second and third or subsequent division cells as a function of culture time between 40 and 96 h showed that cell-cycle duration was prolonged in kwashiorkor cultures. The extent of this increase was approximately 4.5 h for the first-division cells and 1.5 h for third-division cells.

3. The peak for second-division cells was depressed in kwashiorkor cultures.

4. A decreased number of third or subsequent-division cells was observed in kwashiorkor cultures at all time points studied.

5. These findings suggest that the loss in lymphocyte activity to PHA in malnourished children in general could be due to an increase in cell-cycle duration of responding lymphocytes.

1. The frequency of association between methanogenic bacteria and ovine rumen ciliates was studied in the rumen fluid of a fistulated sheep.

2. A period of fasting and flushing of the rumen content with nitrogen resulted in a relatively high association, whereas the intake of food and flushing with hydrogen caused a detachment of the methanogenic bacteria from the ciliates.

3. The changes in the frequency of association can be correlated with the relative attribution to the H2 production by hydrogenogenic bacteria and rumen ciliates.

1. The effects of two isoenergetic diets differing only in the values for polyunsaturated: saturated fat (P : S values of 0·2 v. 2·0) were studied in twenty adult human volunteers.

2. A period of 14 d on the high P:S diet failed to produce significant changes in fasting triglyceride levels, though there were individual variations. On the other hand, fasting cholesterol levels dropped by 10% (P < 0.005). High-density-lipoprotein-cholesterol concentrations were not influenced by changes in the P:S value.

3. Investigations into the mechanism by which changes in the P:S value might affect plasma triglyceride values revealed no consistent effects on very-low-density-lipoprotein kinetics, insulin secretion, insulin sensitivity or free fatty acid concentrations.

4. The results of this study suggest that the largest increase in dietary P:S values that is likely to be obtained on a long-term basis may have only a small effect on plasma triglyceride and cholesterol concentration.

1. Twelve young adult male volunteers were given a low-fibre white bread diet (9 g neutral-detergent fibre (NDF)/d) and a medium-fibre coarse-bran bread diet (22 g NDF/d), each lasting 20 d. In a third period of 20 d the volunteers were subdivided in groups of four, consuming a high-fibre coarse-bran bread diet (35 g NDF/d). a medium-fibre fine-bran bread diet (22 g NDF/d, bran particle size < 0.35 mm) or a wholemeal bread diet (22 g NDF/d), Retention of calcium, magnesium, iron, zinc and copper were determined during each 20 d period.

2. An increase of the amount of dietary fibre (through bran in bread) from 9 g to 22 g NDF/d resulted in a significantly increased mineral intake, but also faecal excretion increased significantly; mineral retention remained almost constant.

3. Both intake and faecal excretion of all minerals studied, except faecal Ca. increased further (P < 0.05) on the diet providing 35 g NDF/d: only Fe balance decreased significantly. No significant differences with respect to intake, excretion (except urinary Ca) and balance of the minerals could be detected between the coarse-bran bread and fine-bran bread diets providing 22 g NDF/d. Faecal Fe, Cu balance and Mg balance increased significantly during the wholemeal bread period compared to the coarse-bran bread diet providing 22 g NDF.

4. Serum cholesterol increased significantly, i.e. by 0.3 mmol/1, during the coarse-bran bread diet providing 22 g NDF, compared to the white-bread diet.

5. It is concluded that increasing the amount of bran in bread does not appear to affect mineral balance considerably but there seems to be an influence on mineral availability. The increased intake was accompanied by increased faecal excretion.

1. Restriction of food to ewes bearing twin lambs showed a trend to decrease the level of ω6 fatty acids in muscle phospholipids of the foetus though this was not significant statistically (P < 0·05).

2. There was a significant increase in the fatty acid ratio, 20:3ω9:20:4ω6 in the twin lambs as compared with single lambs.

3. There were no symptoms of polyunsaturated fatty acid deficiency.

1. The abilities of cattle, sheep, goats, equines and rabbits to digest mature timothy (Phleum pratense) hay were compared. Apparent digestibilities were partitioned into true digestibility, metabolic faecal output (MFO) and fibre digestibility. The aim of the study was to determine the relative effects of fermentation site (among groups) and of body-weight (within groups) on the efficiency of digestion.

2. The ruminants were superior to equines, which were in turn superior to rabbits, in digesting fibre components of the hay. A large individual variation in digestibility was noted only for the equines. Increasing body-weight was associated with higher digestibility in ruminants, but no such trends were seen in the non-ruminants.

3. The MFO expressed as a proportion of dry matter intake gave similar values for all groups (0·085–0·118). As a proportion of available microbial substrate originating from the feed, the values were found to be 0·167 for the ruminant, 0·425 for the equines and 2·13 for the rabbits. The value for the rabbits shows that their lower tract microflora must obtain energy from non-fibre components of the feed. No appreciable digestion of the generated microbes by the host was suggested by the values obtained for the equines.

1. The effects of feeding a palatable and varied ‘cafeteria’ diet on energy balance were studied in young (5·5 week) and adult (5·5 month) lean male Zucker (+/?) rats.

2. Estimates of metabolizable energy (ME) intake derived from food composition tables were almost identical to values obtained from bomb calorimetry of foods, urine and faeces, and ME intake was elevated by approximately 73% in all ‘cafeteria’ animals compared to stock-fed controls.

3. ‘Cafeteria’ feeding had no effect on the body-weight of young rats but induced excess weight gains in the older animals and resulted in increased deposition of fat and energy in both groups. Energy expenditure, calculated from ME intake and body-energy gain, was elevated by 77 and 57% in young and adult cafeteria rats respectively. The energy cost of fat deposition could account for only a small proportion of this increased expenditure.

4. The present results confirm previous findings in another strain of rat and show that the increased energy expenditure (i.e. diet-induced thermogenesis, DIT) which occurs in response to hyperphagia is not restricted to young animals but is also seen in older rats. Measurements of resting oxygen consumption after injections of noradrenaline or a β-adrenergic antagonist (propranolol), and changes in brown adipose tissue mass are consistent with the suggestion that the DIT of ‘cafeteria’-fed rats results from sympathetic activation of brown fat.

1. A study was undertaken to investigate the distribution of ascorbic acid between various cellular components of blood, in normal individuals, and its relation to the plasma concentration. Forty-one unsupplemented individuals and sixteen supplemented (2 g/d for 5 d) individuals were studied.

2. Granulocytes, mononuclear leucocytes, platelets and erythrocytes were separated by differential sedimentation and centrifugation. Ascorbic acid contents were measured by the dinitrophenylhydrazine method.

3. Ascorbic acid content per cel was higher in mononuclear leucocytes and granulocytes than in platelets and erythrocytes. Intracellular ascorbic acid concentrations, calculated from published values for cell volumes, when compared with the plasma concentration showed a marked ability to concentrate ascorbic acid in mononuclear leucocytes (80 times), platelets (40 times) and granulocytes (25 times).

4. Erythrocytes showed little ability to concentrate ascorbic acid over the normal range of plasma concentration but because of their relative numbers they and the plasma fraction accounted for most of the blood-borne ascorbic acid (> 70%).

5. The ascorbic acid content of granulocytes, platelets and erylhrocytes showed a significant positive correlation with the plasma concentration and supplementation with ascorbic acid significantly increased the content of these cell types. Mononuclear leucocytes in contrast did not show any such relationship.

6. The ability of the mononuclear leucocytes to maintain the highest levels of ascorbic acid in the cell types studied, despite variation in plasma availability, warrants further study, particularly in view of the importance of these cells in immunocompetence.
