The absorption spectra of haemoglobins from A. glabratus and P. corneus and from B. (P.) globosus and B. (B.) tropicus angolensis were established. There was no difference between the spectra of snails which transmit Schistosoma mansoni or S. haematobium and the spectra from related insusceptible species.

The snail haemoglobins produced absorption spectra which differed from mammalian pigments only in the position of a single peak.

Snail blood occasionally produced additional small peaks in the ultra-violet region, which may have been due to blood proteins other than the respiratory pigment.

Despite many recent advances in the study of nematicides and in the breeding of resistant strains of plants, control of cyst forming nematodes is still best accomplished by crop rotation; to be fully effective, it is also desirable that a field in which it is proposed to grow a susceptible crop should be sampled and an estimate made of the population of nematodes present: such an estimate is susceptible to different sources of error and although it is possible to decrease the magnitude of these errors a compromise has to be reached between the conflicting claims of precision on the one hand and the expenditure of man-power on the other; it is desirable that this compromise be based on factual data rather than guesswork; good estimates of the order of magnitude of the different types of error are therefore desirable.

The possible role of the cockroach Periplaneta americana in the transmission of Toxocara canis eggs and larvae via faeces and tissue migration was studied. Cockroaches fed with 3 × 105 and 5 × 105 embryonated eggs were found to harbour viable eggs and larvae from days 1 to 5 post-infection (DPI). At necropsy on 5 DPI, eggs and larvae were also recovered from the rectal contents but not from the tissues of cockroaches. In addition patent infections were established in pups fed on infected faeces of cockroaches, with eggs first appearing in the faeces of pups at 38 DPI. Adult worms of T. canis were also recovered at necropsy. Therefore the importance of cockroaches as good mechanical disseminators of ascarid eggs, especially T. canis, is discussed.

The species of Onchocerca described in this paper was received some months ago from Mr. J. L. Stewart, M.C., M.R.C.V.S., from Tamale, Gold Coast Colony. The cysts containing the parasites are found most commonly in the subcuticular connective tissue of the inter-costal muscles; but they are also found elsewhere, usually in a superficial position. Mr. Stewart in a letter accompanying the material, draws attention to their superficial resemblance to Cysticercus bovis.

During the winter of 1921–22 I visited the slaughterhouse at Aberystwyth at least once each week for the purpose of studying the helminthic parasites of sheep and pigs in the neighbourhood. An average of 50 sheep and 8 pigs were examined weekly from November 26th, 1921, to April 28th, 1922. Metastrongylus elongatus and M. brevivaginatus were present in the bronchioles of 50 per cent. of the pigs examined, generally both species being present in the same animal, and I took the opportunity of working out the anatomy of these two forms in detail, and indicating the differences between them; but before being able to publish my results Gedoelst covered the ground in his “Le Genre Metastrongylus Molin, 1861,” published in 1923.

Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) regulates a wide range of biological processes, including pathogen evasion. In the present research, the GAPDH gene of Haemonchus contortus (HcGAPDH) was cloned and characterized. Specific primers for the rapid amplification of cDNA ends (RACE) were designed based on the expressed sequence tag (EST, AW670737) to amplify the 3′ and 5′ ends of HcGAPDH. The full length of cDNA from this gene was obtained by overlapping the sequences of 3′ and 5′ extremities and amplification by reverse transcription polymerase chain reaction (RT-PCR). The biochemical activities of the recombinant protein HcGAPDH, which was expressed in prokaryotic cells and purified by affinity chromatography, were analysed by assays of enzymatic activity, thermal stability and pH. The results showed that the cloned full-length cDNA comprised 1303 bp and encoded a peptide with 341 amino acid residues which showed sequence similarity to several known GAPDHs. The biochemical assay showed that the protein encoded by the HcGAPDH exhibited enzymatic activity with NAD+ as a cofactor. HcGAPDH was stable between pH 5 and 9 and maintained activity at high temperatures of up to 75°C. The natural GAPDH of Haemonchus contortus detected by immunoblot assay was approximately 38 kDa in size, and the recombinant HcGAPDH was recognized strongly by serum from naturally infected goats.

Drug resistance to helminth parasites is one of the most serious problems to threaten the livestock industry. The problem also poses a major threat to public health. Therefore, novel and safe agents should urgently be investigated to control parasitic infections. The current study was conducted to evaluate the possible antiparasitic effects of zinc oxide nanoparticles (ZnO-NPs) on one of the most prevalent gastrointestinal nematodes, Teladorsagia circumcincta. The worms were incubated with various concentrations of ZnO-NPs: 1, 4, 8, 12 and 16 ppm for 24 hours. Mobility and mortality of the parasites were recorded at four-hour intervals. At the endpoint, several biomarkers of oxidative/nitrosative stress, including superoxide dismutase, glutathione peroxidase and catalase, as well as lipid peroxidation, protein carbonylation, total antioxidant status, nitric oxide contents and DNA damage, were measured in the homogenized samples. ZnO-NPs showed significant anthelminthic effects, depending on time and concentration. Furthermore, the nanoparticle induced severe oxidative/nitrosative stress and DNA damage. ZnO-NPs could be considered as a novel and potent anthelminthic agent.

A central and critical step in the molecular detection of soil-transmitted helminths from environmental sources is the extraction of DNA from the eggs. In this study, we investigated the yield of DNA extracted from known quantities (500, 100, 50, 20, 10 and 5) of Ascaris suum eggs, as well as directly from wastewater and sludge samples containing Ascaris spp. eggs, using six commercial DNA extraction kits. The amount of DNA extracted was quantified with NanoDrop, Qubit and Ct values from quantitative polymerase chain reaction (qPCR) assay using CFX96 Touch™ real-time PCR equipment. The PowerLyzer Ultraclean Microbial DNA isolation kit and PowerSoil DNA isolation kit gave the highest yield of DNA based on the NanoDrop, Qubit and Ct values. However, the qPCR results indicate that in some of the kits, PCR inhibitors may have been carried over to the PCR reaction. DNA extraction kits that incorporate a bead-beating step as well as other mechanical eggshell disruption steps were superior in extracting DNA from Ascaris spp. eggs. Additionally, for the accurate quantification of extracted DNA, the use of Ct values from qPCR and Qubit readings gives better results compared to the NanoDrop readings. For efficient downstream applications, the use of DNA extraction kits with superior inhibitor removal technology is essential, in addition to a high yield of DNA.

Biological control is a strategy to decrease parasitic populations, and the action takes place through natural antagonists in the environment. We studied the predatory activity of the fungus Duddingtonia flagrans in infective larvae (L3) of gastrointestinal nematodes after gastrointestinal transit. Ten heifers were divided into two groups: treated (animals received pellets containing fungus) and control (animals received pellets without fungus). Twelve hours after administration, faeces samples were collected for in vitro efficacy tests. The animals then remained for 7 h in the experimental pasture area. At the end of this period, 20 faecal pads (ten treated and ten control) were selected at random. Pasture, faecal pad and soil collections occurred with an interval of 7 days, totalling four assessments. In vitro activity demonstrated that fungi effectively preyed on L3, achieving a reduction percentage of 88%. In the faecal pad of the pasture area, there was a difference (P < 0.05) between collections 3 and 4 for both groups; in the treated group a reduction of 65% was obtained, while in the control group there was an increase of 217% in the number of L3. The recovery of L3 in the soil and in the pasture was similar in both groups. There was no influence (P = 0.87) of the passage time on the fungus predatory activity. Duddingtonia flagrans demonstrated the ability to survive gastrointestinal transit in the animals, reducing the number of L3 in the faeces, indicating that this biological control has great potential in the control of worm infections.

The objective of this study is to determine the prevalence of Fasciola hepatica infection in cattle slaughterhouses, as well as its association with climatic/environmental factors (derived from satellite data), seasonality and climate regions in two states in Mexico. Condemned livers from slaughtered animals were obtained from three abattoirs in the states of Puebla and Veracruz. The overall prevalence of the parasite in cattle between January and December of 2017 was 20.6% (1407 out of 6834); the highest rate of condemnation was observed in Veracruz (26.3%; tropical climate), and the lowest rate was found in Puebla (15.5%; temperate climate). The seasonal prevalence of fluke infection was 18.6%, 14.8% and 28.4% during the wet season, and 17.1%, 12.4% and 22.8% during the dry season in the three abattoir sites, located in the districts of Zacatlán, Teziutlán and Ciudad Alemán, respectively. Liver condemnations due to bovine fasciolosis were prevalent in the Zacatlán, Teziutlán and Ciudad Alemán districts during summer, autumn and summer, respectively. Using generalized estimating equations analysis, we determined six variables – rainfall (wet/dry), land surface temperature day, land surface temperature night, normalized difference vegetation index, seasonality and climate regions (temperate/tropical) – to be significantly associated with the prevalence of condemned livers. Climate region was the variable most strongly associated with F. hepatica infection (odds ratio (OR) 266.59; 95% confidence interval (CI): 241.90–353.34), followed by wet and dry seasons (OR 25.56; 95% CI: 20.56–55.67).

Adults and plerocercoids of a new species of Heteronybelinia (Trypanorhyncha, Tentaculariidae) were recovered from the skate Sympterygia bonapartii (Rajiformes: Rajidae) and two species of teleosts, Raneya brasiliensis (Ophidiiformes: Ophidiidae) and Nemadactylus bergi (Perciformes: Cheilodactylidae), respectively. Heteronybelinia mattisi n. sp. differs from its congeners in its possession of a tentacular armature consisting of hooks with slight differences in size and shape on opposite surfaces of the tentacle (uncinate with rounded bases on the bothrial surface and uncinate with elongate bases on the antibothrial surface), without a characteristic basal armature, hooks increasing in size toward the tip of the tentacles, and pars bothrialis slightly overlapping with bulbs. The description of Heteronybelinia is emended regarding the distribution of testes to include species without postovarian testes (H. palliata and H. mattisi), and without testes anterior to cirrus sac on the poral side (H. robusta and H. mattisi). Host data are summarized for all 15 valid species of Heteronybelinia. Host specificity of adults and plerocercoids of Heteronybelinia is variable among species, the adults being more host specific than the plerocercoids. With the exception of H. mattisi, the definitive hosts of all species of Heteronybelinia for which adults are known include carcharhiniform sharks. Heteronybelinia mattisi seems to have oioxenous specificity for its definitive batoid host, S. bonapartii, and to be more specific for the intermediate hosts than most of its congeners.

Male and female Schistosoma japonicum worms have dissimilar appearances in their final host. In this study, a morphometric and morphological assessment of whole worms derived from unisexual and mixed infections in mice was conducted using confocal laser scanning microscopy. Worms from mixed infections showed significant morphological changes between 15 and 25 days post-infection (PI). On the fifteenth day PI, 33% of males had formed the conspicuous gynecophoric canal, but only 8% of them had testicular lobes containing a few germinative cells; 13% of females had incipient ovaries with a few immature ovarian cells inside. On the twentieth day PI, the testicular lobes contained more germinative cells in all male worms, while female worms presented vitelline glands. On the twenty-fifth day PI, more germinative cells were observed in the male testicular lobes, and differentiated cells were present in the female ovaries. All worms had fully developed reproductive organs from 30 days PI onwards. Morphometric analysis showed significant differences between mixed and unisexual infections at 35 days PI. Ovaries of worms from unisexual infections contained cells in one stage of maturation and vitelline glands had undifferentiated cells. Our study of S. japonicum provides a detailed comparison of different morphological traits from worms of mixed and unisexual infections throughout development.

Aulonocephalus pennula is a heteroxenous nematode that commonly infects a declining game bird, the northern bobwhite quail (Colinus virginianus). There is a lack of information on the life cycle of A. pennula and the potential effects of infection on bobwhites. In order to better understand the life cycle of this parasite, various species from the order Orthoptera were collected from a field site in Mitchell County, Texas. Using polymerase chain reaction (PCR), nine potential intermediate hosts were identified from the 35 orthopteran species collected. Later, ten live specimens were collected to identify larvae within the potential intermediate hosts. Larvae were present in three of these and were sent for sequencing. Similarly, the presence of larvae was confirmed from extra tissues of samples identified as positive with PCR. This was the first study to document potential intermediate hosts, but future studies are needed to confirm that these species are capable of transmitting infection to bobwhite. However, this study demonstrates that PCR has increased sensitivity and may be a valuable tool when determining intermediate hosts.

Ortleppascaris sinensis (Nematoda: Ascaridida) is a dominant intestinal nematode of the captive Chinese alligator. However, the epidemiology, molecular ecology and population genetics of this parasite remain largely unexplored. In this study, the complete mitochondrial (mt) genome sequence of O. sinensis was first determined using a polymerase chain reaction (PCR)-based primer-walking strategy, and this is also the first sequencing of the complete mitochondrial genome of a member of the genus Ortleppascaris. The circular mitochondrial genome (13,828 bp) of O. sinensis contained 12 protein-coding, 22 transfer RNA and 2 ribosomal RNA genes, but lacked the ATP synthetase subunit 8 gene. Finally, phylogenetic analysis of mtDNAs indicated that the genus Ortleppascaris should be attributed to the family Heterocheilidae. It is necessary to sequence more mtNDAs of Ortleppascaris nematodes in the future to test and confirm our conclusion. The complete mitochondrial genome sequence of O. sinensis reported here should contribute to molecular diagnosis, epidemiological investigations and ecological studies of O. sinensis and other related Ascaridida nematodes.

The two-dimensional patterns (isoelectrofocusing-IEF/polyacrylamide gel electrophoresis in the presence of sodium dodecyl sulphate-SDS) of S3 fractions of muscle larvae of four Trichinella isolates were compared. The comparative study concerned six groups of polypeptides. It was observed that the Garkavi isolate of Trichinella pseudospiralis was clearly different from the other isolates, and it showed the simplest IEF/SDS polypeptide pattern. The C-76 isolate of T. nelsoni had only four of the six groups, distinguishing it from the GM-1 isolate of T. spiralis and the Boev isolate of T. nativa that showed all the indicated groups.

In the majority of cases it is almost impossible to identify the species of helminths in our domestic stock by an examination of the eggs found in their droppings. In the case of sheep and goats, this is particularly so since these hosts harbour several worms with eggs which are, apparently, identical in shape, size and stage of development.

The lungworm genus Crenosoma, founded by Molin in 1861, was redescribed in 1928 by Skrjabin and Petrow. To the two existing species, C. striatum (Zeder, 1800) from the hedgehog (Erinaceus europæus L.) and C. vulpis (Dujardin, 1845) from the fox (Vulpes vulpes L.), these writers added a third species, C. taiga, from the lungs of Kolonocus sibiricus (Pall.) in Russia. A generic diagnosis based on a study of these three species was supplied, and the wolf (Canis lupus L.) was recorded as a new host for Crenosoma vulpis.

The infectivity of Apatemon (Australapatemon) minor cercariae to 7 species of freshwater leech was examined under laboratory conditions. The leech species exposed to infection were; Erpobdella octoculata, Helobdella stagnalis, Glossiphonia heteroclita, Glossiphonia complanata, Hemiclepsis marginata, Piscicola geometra, and Theromyzon tessulatum. Five species were capable of acting as second intermediate hosts. Despite this broad specificity only Erpobdella octoculata and Helobdella stagnalis could be considered to show a high degree of compatibility with the parasite. In these 2 leech species more than 38% of the cercariae successfully penetrated and established metacercarial cyst infections. The leeches Piscicola geometra and Theromyzon tessulatum did not become infected. Close parallels were found between the species order of host utilization determined experimentally and that revealed by a range of field surveys in Britain, Eastern Europe and Russia. This suggests that the order of second intermediate host utilization in the natural environment has, at least in part, a physiological basis. The degree to which different leeches are utilized as hosts may relate to the opportunity which they afford for transmission of A. minor to wildfowl definitive hosts.

The potential of using 35S-labelled larvae to determine the number of second-stage Toxocara canis larvae present in the tissues of infected animals was assessed. Infective larvae were labelled by in vitro culture with medium containing 35S-methionine. The amount of radiolabel attached to larvae decayed exponentially with time and had an in vitro mean half life of 3·54±0·65 days. The ‘lost’ radiolabel was incorporated into proteins which formed part of the worm's excretory/secretory products. The levels of radioactivity present in different organs of BALB/c mice, infected with 35S-labelled T. canis larvae, varied over the course of infection. Initially most of the radioactivity was present in liver, but over the course of infection 35S liver levels gradually decreased and brain levels increased. By day 14 post-infection the majority of the isotope was present in the brain (p<0·01). Assessment of antibody levels on day 14 post-infection showed that infection with 35S labelled T. canis larvae induced the production of parasite-specific IgM, IgG and IgG1 antibodies.
