Fifty-one metaphase figures from male Mesocricetus newtoni bone-marrow cells were analysed with respect to the distribution of chromosomes. The peripheral location of the sex chromosomes (XY) and that of the chromosome pairs 3 and 9, as well as the non-peripheral location of the chromosome pairs, 16, 17, 18 was revealed. The significance of the peripheral location of the X chromosome and a possible explanation of the characteristic distribution of the chromosomes are discussed.

We have studied the association of the ten possible B blood-group genotypes with mortality during incubation in the 1966 generation of two relatively noninbred populations of Light Sussex chickens. These were related substrains 6D and 6F derived in the 1964 generation by equal division of strain 6, which we had previously studied in its 1962 generation. The B blood-group genotypes of zygotes but not of dams were associated with marked mortality differences in 6F, while in 6D almost no differences were found. Comparisons of the findings in 6D and 6F with the results previously published for the ancestor strain 6, after adjustment for overall between-population differences, showed marked changes between 6 and 6F in the mortalities associated with the five most frequent genotypes. In four of these, the mortalities in 6D were about half way between those in 6 and 6F. Taken together, these results provide evidence that the selective pressures on the B locus from differential embryonic mortality vary from generation to generation and even between populations differing apparently only slightly in environment and genetic composition.

In progeny of crosses between females from 6D and 6F with males from 5D and 5F respectively, the latter being two related Rhode Island Red/New Hampshire substrains, embryonic mortality was much lower than in the pure matings within 6D and 6F, and only a few associations with B blood-group genotypes of zygotes were found.

The results are discussed in relation to the general lack of association of polyallelic polymorphisms with aspects of fitness reported by other workers; and the importance is stressed of studying associations of genotypes rather than genes or serological factors.

By crossing a pair of albino strains each with a different adjacent nutritional marker and then crossing the same pair of albino markers with the nutritional markers transposed it was possible to order a series of al mutants with a resolution approaching that available for nutritional markers. A genetic map with approximated distances is provided demonstrating a grouping of mutant sites for a range of discrete carotenoidless phenotypes.

Purine-excreting mutants of yeast have been obtained by selecting for (a) adenine-insensitive pigment accumulation in a strain carrying ad-2 and (b) by isolating prototrophic mutants which crossfeed to an adenine-requiring indicator strain. The first method yielded both auxotrophic and prototrophic mutants. The auxotrophs were assigned to ad-12 which specifies adenylosuccinate synthetase. The prototrophs were affected by a gene, su-pur, which prevented purine excretion, and the majority of them showed anomalous behaviour of mating-type a associated with the mutations causing excretion. Mutants obtained by the second method were allocated to six unlinked genes. Those assigned to pur-1 to pur-5 are recessive and affected by su-pur. pur-6 is a complex locus with both dominant and recessive alleles; it is closely linked or allelic to su-pur and ad-4. Functional tests involving mutants of pur-3 and pur-6 in combination with pur-1 and pur-4 did not show normal intergenic complementation. The suppressor shows gene-specific dominance/recessiveness. Some metabolic lesions which could give rise to purine excretion are discussed.

An analysis of populations of Drosophila melanogaster which had ceased to respond to selection for increased scutellar bristle number has disclosed the existence of an overall negative genetic correlation between replicate lines in the frequency of the two major component bristle types, viz. anteriors and posteriors. Negative phenotypic correlations among component bristle sites have also been detected within populations. A model involving competition among sites for the available resources of a particular limiting substrate is therefore proposed. Genetic changes have been effected in exceptional populations which lead either to an increase in the rate of production of the limiting substrate, or to a lowering of the concentration of the substrate necessary for bristle initiation. The allelic substitutions concerned are recessive, and have been described as decanalizing alleles in view of their effects on both bristle number variability and a measure of developmental regulation at individual anterior sites. Genetic variation has also been demonstrated for the mean allocation of resources to each of the four component bristle types, viz. anteriore interstitials, posteriors and apicals. A brief discussion is given of the implications of the model for breeding practice in domestic species.

A system has been developed for the study of reversion of an amber mutation responsible for a deficiency in DNA synthesis in T4 phage E51. When complexed with bacteria able to suppress the amber mutation the induced mutation rate per phage genome per rad is

When complexed with bacteria unable to suppress the amber mutation (and being thus unable to synthesize phage DNA) the induced mutation rate is at least 14 times lower indicating that DNA synthesis is necessary for the production of the majority of functional reversions at the amber site. The induced mutation rate in suppressor-containing bacteria is independent of multiplicity of infection between 0·2 and 5, suggesting that recombination immediately after irradiation between phage genomes is unlikely to be a requirement for the mutation process.

The functional morphology and the replication pattern of the male X-chromosome in an autosome-X insertion stock (T(1;3) 05) of Drosophila melanogaster have been examined. In larval salivary glands carrying this insertion neither the enlargement and pale staining of the single male X, nor the characteristic early completion of replication cycle, as revealed by 3H-TdR autoradiography is in any way changed. The normal properties of the inserted autosomal segment are also unaltered. The results appear to support a ‘piecemeal’ type of dosage compensation mechanism in Drosophila operating through the male.

Over 400 temperature-sensitive mutants of Ustilago maydis have been tested for DNA synthesis at the restrictive temperature of 32°C by measuring 14C adenine incorporation into DNA and RNA. Five mutants were defective in DNA synthesis but none was completely blocked. One mutant, tsd-1, which is unlinked to the others, forms long uninucleate filaments at 32°C which die exponentially after 4h temperature treatment. The phenotype is comparable to that of thymine-starved bacteria, but it is possible that rather than being specifically defective in DNA synthesis the mutant is blocked in nuclear division.

Effects of the incompatibility factors in Schizophyllum commune Fries on the process of hyphal fusion are described. A role for the A incompatibility factor in hyphal fusion is indicated. Matings between strains with different mating types have higher fusion frequencies than matings between strains with the same mating types. Evidence is presented that the differences in fusion frequencies are not due to genetical factors other than mating types. When two strains of different mating types are grown in the same culture plate, but separated by a cellophane membrane, the strains are altered in some unexplained manner in such a way that even matings between strains of the same mating type have a higher fusion frequency than occurs in matings between compatible strains not so treated. Matings leading to the formation of common-B and dikaryotic mycelia have comparable fusion frequencies while those leading to the formation of common-A mycelia have a far lower frequency of fusions. It has been demonstrated that high fusion frequency is associated with heterozygosity at the A locus. It is suggested that a repression–derepression mechanism involving a cell wall degrading enzyme or enzymes may be involved in the regulation of hyphal fusion.

A synthetic lethal system involving homozygous combination of the non-allelic eye mutants eye-gone and eyeless is described. The major lethal crisis is towards the end of the pupal instar although a number of double homozygotes die at pupation. The pupal lethal crisis can be overcome in a number of individuals by the experimental removal of the operculum from differentiated pupae. However, only a few of the resulting enclosed double homozygotes proved fully viable, the remainder dying almost immediately after emergence. Dissection of the surviving adults revealed that the brain is located in the anterior thorax.

The genetic and phenotypic regressions and correlations between ovulation rate and body weight were examined in a random bred strain (Q) of laboratory mice during the course of three experiments. These experiments were (1) a sib analysis; (2) selection for natural and induced primiparous ovulation rate; and (3) replicated selection for 6-week weight. The following results were obtained:

(a) The genetic correlations between body weight and natural and induced ovulation rate were positive, and approximately equal to 0·4 and 0·6 respectively.

(b) The genetic regressions of natural and of induced ovulation rate on body weight were approximately 0·4 and 0·9 eggs per gram respectively.

(c) The genetic regressions of body weight on natural and on induced ovulation rate were approximately 0·5 and 0·25 g per egg respectively.

(d) The phenotypic correlation between natural ovulation rate and body weight was approximately 0·4 and the corresponding regression of ovulation rate on body weight approximately 0·4 eggs per gram.

(e) The phenotypic correlation between induced ovulation rate and body weight declined from 0·4 at 6 weeks of age to zero at the time of scoring, the corresponding regressions of ovulation rate on body weight declining from 0·1 eggs per gram to zero.

It was concluded that natural ovulation rate itself, and both its components (FSH activity and ovarian sensitivity) are positively genetically correlated with body weight. Furthermore, the observation that large mice shed at least as many eggs as small ones in response to the same dose of PMS showed that the response was more closely related to the absolute dose than to the resultant concentration.

A new spontaneous mutant bubble (bu) with a marked effect on early morphogenesis in the multicellular green alga, Ulva mutabilis is described. It behaves as a recessive chromosomal mutant. The bu+ gene product formed in heterozygous sporophytes exerts a predetermining effect on morphogenesis of haploid gametophytes with the bu genotype. It is suggested that the product of the bu+ gene affects the orientation of the mitotic spindles during early development.

Plasmodia of P. polycephalum homozygous and heterozygous for a mutation (act) which confers resistance to actidione on the haploid amoebal stage have been tested for resistance to actidione in liquid semidefined medium. Growth of homozygous resistant (act/act) plasmodia determined by protein or pigment assay was inhibited less by actidione than growth of homozygous-sensitive or heterozygous plasmodia. The mutant (act) is therefore recessive but, contrary to a previous report, confers resistance on homozygous plasmodia. The same mutation affects the morphology of homozygous (act/act) plasmodia on agar medium, causing slow growth, fragmentation and excessive sliminess.

Translocation T(III–VIII) in Aspergillus nidulans has been analysed by the detection of meiotic linkage between markers previously located separately on linkage groups III and VIII. The breakage points have been mapped by the detection of linkage between the crinkled type and genetic markers in the region of the break. A segment from linkage group III, approximately 43 units long and including the markers moC96, sC12, sA1 and cnxH3, has been translocated into linkage group VIII. The breakage point is between su6proA and moC96 and the attachment point is close to cha in linkage group VIII. It seems probable that the segment has been inserted into linkage group VIII.

The frequency and nature of the changes in ‘state’ of the mouse X-chromosome controlling element (inactivation centre) have been investigated on an inbred background. The results indicate with near-certainty that meiotic crossing over is the responsible mechanism and that the frequency of recombination between the T(1; X)Ct breakpoint and the locus of the controlling element is approximately 3%. Maize-type ‘changes in state’ may occur under other experimental conditions. The data do not distinguish on which side of the autosomal insertion the element lies but when combined with observations of other investigators suggest that the location must be on the Mo-Ta side and very close to Ta.

The F factor and the K resistance determinant form an R factor (KF) in E. coli K12. In this R factor the two plasmids are independent of each other and occupy different attachment sites in the host cell.

Treatment of an F+(K) strain with acriflavine and acridine orange yielded the following classes: F+K−; F−K+; and F−K−. The K determinant was thus curable independently of F, which supports the hypothesis of independence of the two plasmids. In Hfr(K) strains, K remains curable with acridines while F is immune. Thus, K remains independent of F in both the F+ and Hfr states.

The use of higher concentrations of acridine orange than those needed for cure of K in the F+(K) strain resulted in no elimination of K, although the F factor was absent from all surviving lines tested. It is suggested that this is caused by equal inhibition of replication of both the chromosome and K, and greater inhibition of that of F.

The variety Chinese Spring of Triticum aestivum is susceptible to cold treatment applied at the juvenile plant stage, while the variety Cappelle-Desprez shows resistance to such treatment. By cytological and backcross procedures single homologous pairs of chromosomes from Cappelle-Desprez were substituted for their homologues in Chinese Spring. Assay experiments carried out on each of the 21 possible substitution lines indicated that three chromosomes, 4D, 5D and 7A of Cappelle-Desprez were involved in the determination of cold resistance. The resistance expressed by the three substitution lines carrying these chromosomes was less than the reaction of Cappelle-Desprez to cold treatment. Also the magnitude of this resistance supported a hypothesis that the action of the three chromosomes was additive on the scale of measurement used. The possible relationships of the genes for cold resistance to the established genes controlling other developmental characters, also carried by these three chromosomes, are discussed.

Females heterozygous for the T(1; X)CtX-autosome translocation tend to have lower levels of c-variegation when their rearranged X is inherited from the father rather than from the mother. The difference is not due to a maternal effect. It is postulated that a paternal or parental-source effect, such as that found to modify position effect variegation in Drosophila, is operating but the possibility that a bias in the inactivation of the maternal and paternal X chromosomes is responsible cannot be ruled out.

A new type of apparatus for replica plating is described, which reduces operator error and fatigue. Several replica plates can be made immediately, thus eliminating the need for a master plate in many cases.

Some features of gene conversion in fungi and their bearing on the hybrid DNA models are discussed. Available experimental data from tetrad analysis seem to give a more complex picture of polarity in intra-genic recombination and of the relations between conversion and post-meiotic segregation, and between conversion and crossing-over, than predicted by the models.

A new hypothesis of the mechanism of gene conversion with special attention given to the aspect of asymmetry in this phenomenon is proposed as an alternative to the mechanism suggested by the DNA hybrid models.
