Obtaining corn hybrid seeds (Zea mays L.) with high vigour depends on the parental lines and the direction of the cross, and this relates to seed desiccation tolerance and composition. This research studied reciprocal crosses between pairs of proprietary, elite parent lines (L1 and L5; L2 and L4) producing hybrid seeds with different qualities attempting to correlate vigour with seed composition, focusing on storage proteins, starch and soluble sugar amounts. Four corn hybrid seed lots produced from reciprocal crosses were compared (HS 15 with HS 51, and HS 24 with HS 42) by assessing germination, vigour, and seedling emergence in the field. Seed composition was assessed in mature, dehydrated seeds. Proteins were extracted, quantified, and analysed by electrophoresis and densitometry. Starch amounts were assessed using a kit and soluble sugars were determined using high performance liquid chromatography with pulsed electrochemical detection. The L1 and L2 lineages, used as female parents, provided seeds with lower vigour; however, the quantification of major protein bands, and sucrose, raffinose and stachyose were similar between seed lot pairs. While both total seed protein and starch varied between reciprocal hybrids for one of the two sets of crosses, the amounts of neither correlated with seed vigour. Interestingly, hybrids with low seed vigour (HS 15, HS 24) accumulated greater amounts of fructose relative to their reciprocal; correlation analysis confirmed these results. We demonstrate different effects on seed vigour dependent on the maternal parent in reciprocal crosses producing hybrid corn seeds. We also show that vigour is negatively correlated with seed reducing sugar contents.

Swartzia langsdorffii seeds have recalcitrant characteristics. Nonetheless, dispersal begins in the month with the lowest precipitation in the studied region, which could lead to seed death by desiccation. Therefore, the objectives of this study were: (1) to characterize the physiological behaviour of S. langsdorffii seeds related to their desiccation sensitivity/tolerance; and (2) to assess the morphophysiological characteristics that enable the seeds to remain viable after dispersal. Fruits and seeds were subjected to biometric evaluation and the anatomical and ultrastructural features of the seeds were determined. Field assessments were performed to determine the capacity of the seeds to maintain viability and to verify the relation between seed viability, diaspore water content and environmental variables. Seeds of this species were found to be recalcitrant and showed pores distributed throughout the seed coat, and contained a large number of stomata in the hypocotyl–radicle axis epidermis. Moreover, phenolic compounds were found throughout the radicle region. Seeds remained viable in the soil for up to 7 months after dispersal without a significant decrease in water content, despite the low precipitation and soil water content. Radicle protrusion began 5 months after dispersal and coincided with partial fruit decomposition at the beginning of the rainy season. Thus, the possible microclimate created by the pericarp, with the moisture content of the aril and the soil, the presence of the structures in the axis, such as the pores and stomata, the chemical composition and the morphology of S. langsdorffii seeds could favour maintenance of their viability until the beginning of the rainy season.

Germination responses to 45 combinations of diurnal mean temperature and amplitude were examined in freshly collected seeds of two wetland perennials: Typha latifolia L. and Phragmites australis (Cav.) Trin. ex Steudel. Nearly all seeds (>95%) germinated in favourable temperature regimes. Mean temperature (range 10–30°C) and amplitude (range 0–20°C) affected final germination of both species. P. australis required a high amplitude (> 10°C) for germination over the entire range of mean temperatures. Final germination of T. latifolia was more sensitive to mean temperature than P. australis. The germinated proportion of T. latifolia had a maximum around 20°C, above which it decreased, and amplitudes were more stimulating at low than at high levels of mean temperature. The germination rate was rapid and increased with mean temperature for both species. More than 50% germination was achieved within 1–3 d at favourable temperatures. It is proposed that the thermal requirements provide the non-dormant seeds with a season-sensing mechanism which postpones germination of seeds dispersed during autumn, winter or early spring, until the soil surface is heated by the sun in the spring and sufficiently large diurnal fluctuations of temperature occur. Furthermore, the amplitude requirement implies a strong avoidance mechanism for germination of P. australis in sites with small temperature fluctuations (e.g. below water tables), whereas seeds of T. latifolia appear to be less exacting in the requirements when the soil or water becomes warmer.

The endosperm of dormant wild oat (Avena fatua L.) grains contains a number of mRNAs, including low levels of transcripts encoding avenin (AV10), puroindoline-like (AV1) and serine proteinase inhibitor-like (Z1) proteins. During dry afterripening, a period of warm dry storage that relieves dormancy, the abundance of these intact transcripts gradually declined to undetectable levels in pooled grain samples. In contrast, the three mRNAs did not decline in grains stored for the same period at −20°C, and levels of mRNA decay products derived from these transcripts did not change during afterripening. Disappearance of the three intact mRNAs was not a general phenomenon, since abundance of more than 5000 other seed mRNAs did not change during this period. Additional experiments were conducted to test the idea that the presence of these transcripts was positively correlated with seed dormancy status. First, the rate of intact Z1 mRNA decline was similar to the rate of dormancy relief during 9 months of afterripening. Second, cultivated oat (A. sativa) grains (which are not dormant at maturity) contained much lower levels of these mRNAs than did highly dormant wild oat grains. Third, levels of full-length AV1 and Z1 mRNAs remained constant in dormant grains during 6 d of imbibition. However, Northern analyses of individual grains from a partially afterripened seed lot showed that amounts of intact AV1 or Z1 mRNAs were highly variable in germinating and dormant grains and were not correlated with dormancy status. Even though loss of these transcripts occurred under the same environmental conditions that promote afterripening, a causal relationship between the two processes seems unlikely. The apparently specific mechanism(s) by which these mRNAs are degraded in endosperm tissue may represent a novel post-transcriptional process for regulating levels of certain transcripts in dry grains.

Vitamin E is known to scavenge lipid peroxy radicals and has a purported role in preventing seed deterioration during storage. In our previous studies using 20 rice varieties from different variety groups, the specific ratio of vitamin E homologues rather than total vitamin E content was associated with seed longevity. To validate this result, we extended the experiment to a rice panel composed of 185 Aus (semi-wild rice) varieties. Seed longevity values were determined through storage experiments at 45°C and 10.9% seed moisture content (MC). Eight types of vitamin E homologues (α-, β-, γ- and δ-tocopherol/tocotrienol) were quantified by ultra-performance liquid chromatography. The theoretical initial viability in NED, Ki, was positively correlated with γ- and δ-tocopherols and negatively correlated with α-tocotrienol. The time for viability to fall to 50% during storage at elevated temperature and relative humidity, p50, was positively correlated with δ-tocopherol. The harvest MC was negatively correlated with all seed longevity traits. Taking this factor into account in a genome-wide association (GWA) analysis, we were able to correct false positives. A consistent major peak on chromosome 4 associated with −σ−1 was detected with a mixed linear analysis. Based on rice genome annotation and gene network ontology databases, we suggest that RNA modification, oxidation–reduction, protein–protein interactions and abscisic acid signal transduction play roles in seed longevity extension of Aus rice. Although major GWA regions were not overlapped across traits, three genetic markers, on chromosomes 1, 3 and 4, were associated with both δ-tocopherol and Ki and two markers on chromosome 1 and 8 were associated with both δ-tocopherol and p50.

The seasonality of the Mediterranean climate has important implications for plant physiology and some specific conditions must be met before germination can occur. Most plants sprout during the spring season although a few germinate during the autumnal rainy period. To understand the patterns of germination under autumn conditions we selected five Mediterranean autumn-flowering geophytes that usually co-exist in fire-prone habitats: Leucojum autumnale, Narcissus cavanillesii, Narcissus serotinus, Scilla autumnalis and Urginea maritima. Experiments were performed under conditions simulating those prevailing in the habitat during the autumnal Mediterranean season when seeds of these species usually germinate. In all species, germination occurred quickly ( < 7 d), was usually higher at the lowest temperatures tested (15°C, 20°C; >89%) and no signs of delayed germination were detected. Nevertheless, species usually had different patterns of seed germination, a strategy that might avoid interspecific competition when co-occurring in the same habitats. Germination was lower and slower in light than in darkness, suggesting that under natural conditions germination of seeds directly exposed to sunlight can be severely restricted. Several fire-dependent treatments (heat and ash) revealed a lack of fire-stimulated germination, and treatments even had a negative effect. In addition, only a small fraction of seeds remained viable after 1 year of dry lab storage and their germinability was very low, suggesting the absence of a permanent soil seed bank in the life cycle of these geophyes.

The effects of cold atmospheric plasma (CAP) of dielectric barrier discharges on the wettability, imbibition and germination of Leucaena leucocephala were investigated. It was established that CAP treatment markedly hydrophilized the seed coat, especially at longer treatment times. From the profile of the imbibition curve and visual observation, it was possible to verify that there are two resistance barriers to water penetration: integument surface and region of the macrosclereid cell wall (light line). Although the plasma interacts only in the integument, increasing the density of hydrophilic sites increases the capacity of water absorption, producing enough driving force to overcome the second resistance barrier. The existence of these two barriers changes the three-phase pattern generally observed during seed germination. Despite an increase in imbibition, the plasma treatment conditions used in this work, were not enough to overcome completely the dormancy barrier.

Seeds were collected at weekly intervals from mid-maturation to the fully ripened stage. As seed development progressed, desiccation tolerance increased. Desiccation tolerance of C. lansium seeds was greatest at 67 days after anthesis (DAA), when they tolerated air drying for 9 days; 74 DAA was considered as physiological maturity, and their full viability was only maintained for up to 3 days of drying; overripened seeds (88 DAA) had the lowest desiccation tolerance. In L. chinensis, the desiccation sensitivity of seeds at 98 DAA (fully mature) was higher than that at 84 and 91 DAA (less mature); among the excised embryonic axes at different developmental stages, the less mature ones were less sensitive to desiccation than the fully mature ones; excised embryonic axes of the same stage were more tolerant of desiccation than whole seeds.

Seed germination of Grand Rapids lettuce (Lactuca sativa L.) was inhibited at 38°C (thermoinhibition). Thermoinhibition was totally relieved by a combination of 100% oxygen and 10 mg I−1 kinetin, to a lesser extent by a combination of oxygen and ethylene but not at all by oxygen plus GA3. The combination of oxygen with kinetin or ethylene was more effective in preventing thermoinhibition than a combination of all three hormones and CO2 in air. The beneficial effect of oxygen plus kinetin was not related to either increased production of CO2 or ethylene by the seeds. In air, germination was partially inhibited by 2-aminoethoxyvinyl glycine (AVG) or 2,5-norbornadiene (NBD) and totally inhibited by AVG plus NBD. Oxygen plus kinetin almost completely alleviated their inhibitory effects. It was concluded that oxygen plus kinetin caused seeds to bypass an ethylene requirement for germination or increased the sensitivity of seeds to ethylene. In air at 38°C, seeds exhibited a high level of ethanolic fermentation, while in oxygen plus kinetin they respired aerobically. At 38°C in air, contents of ATP and total adenylates were reduced, while in seeds treated with oxygen plus kinetin these contents were similar or higher than in 25°C controls. A similar response was also observed for adenylate energy charge. It was concluded that part of the mechanism of thermoinhibition could be the failure of ATP content to reach a sufficient level to satisfy the requirement for germination at 38°C. It appeared possible that the prevention of thermoinhibition by oxygen plus kinetin could be related to increased ATP content in seeds caused by this treatment.

Recently, forest seed banks were proven to not only reflect former (decades-old) but also ancient (centuries-old) land use. Yet, as land-use intensity determines the magnitude of seed-bank changes in recent forests, this study aims to identify whether an ancient land-use gradient would also be reflected in the seed bank. On a forested 1600-year-old archaeological site, five different land-use intensities were mapped and sampled. Apart from seed density, species richness and composition, functional seed-bank types, defined by nine seed-bank-related plant traits, were related to the land-use intensity gradient. The land-use gradient from gardens to undisturbed sites was still clearly reflected in the soil seed bank. Six emergent functional seed-bank types, characterized by specific plant traits, changed significantly in abundance, parallel to the land-use gradient. In particular, dispersal agent (and related traits) proved an important explanatory trait of present (functional) seed-bank patterns. Poor dispersers (large and heavy seeds) were not found in the intensively used areas, contrary to animal-dispersed species. Wind-dispersers may have been inhibited in the extension of their distribution by recruitment bottlenecks (low seed production) and/or competitive exclusion. Additionally, the agricultural land-use probably introduced ruderal species into the seed bank of the most intensively used areas, yielding a simultaneous increase in vegetation–seed-bank dissimilarity with land-use intensity, eliminating present vegetation as a driver behind the differences over the seed-bank gradient. We conclude by arguing how coppice-with-standards management possibly maintained the seed-bank gradient.

The distribution of ATP in tomato seeds was visualized by monitoring the luminescence of frozen sections on top of a gel containing all the components of the luciferase reaction, but excluding ATP. ATP was imaged in germinating tomato seeds at intervals of 3, 6, 17, 24 and 48 h and in seeds with primary or secondary dormancy. ATP was present mainly in the embryo and concentrated in the radicle tip towards the completion of germination. In contrast to germinating seeds, ATP was distributed more evenly in dormant seeds. For germination, the ratio of ATP concentration in the radicle tip versus cotyledons was decisive, rather than the absolute concentration.

Seed dormancy and germination involve the concerted operation of molecular and biochemical programmes. It has become feasible to study these processes in great detail, using the current methods for transcriptome, proteome and metabolome analysis. Yet, the large amounts of data generated by these methods are often dazzling and demand efficient tools for data visualization. We have used the freely available PageMan/MapMan package (http://MapMan.gabipd.org) to visualize transcriptome and metabolome changes in Arabidopsis thaliana seeds during dormancy and germination. Using this package we developed two seed-specific MapMan pathways, which efficiently capture the most important molecular processes in seeds. The results demonstrated the usefulness of the PageMan/MapMan package for seed research.

Miconia chartacea is a widely distributed tree in Brazil, occurring at altitudes ranging from 300 m to 1900 m in the Caatinga, Cerrado and Atlantic Forest biomes. In this work we attempted to classify M. chartacea seeds regarding their behaviour during storage and their germination syndrome and to determine, from a storage test in Cerrado soil and laboratory conditions in situ and ex situ, the longevity of seeds, as well as the capacity of the species to form a soil seed bank. The results suggested that M. chartacea seeds form a transient soil seed bank in the Cerrado and can be classified as orthodox in terms of storage behaviour, although the seeds are dispersed with a relatively high water content. The life span of seeds was favoured in soil-stored seeds in comparison with dry storage at 25°C, whereas storage at low temperatures prevented a decrease of the seed's germinability with storage time (330 d). M. chartacea seeds are dispersed during the dry season and germinate during the next rainy season, which can be classified as an intermediate–dry germination syndrome. Seeds of this species are dispersed in the Cerrado when temperatures and soil moisture are relatively low, which favours the formation of a soil seed bank, considering that the seeds tolerate desiccation and their longevity is favoured by low temperatures. A transient seed bank type is favoured by the loss of viability in storage at warm temperatures linked to the rainy season, and the predictable seasonal variations in climate in the region, with germination being restricted to the beginning of the rainy season.

This paper is a résumé of research on imbibitional chilling injury of seeds. The following summarizes the experimental results obtained and some views and concepts derived from the studies conducted at Beijing Botanical Garden during the last decade. A lot of evidence has supported and confirmed the hypothesis for imbibitional chilling injury in seeds, i.e. the primary target of harmful effects during the chilling rehydration in seeds is the membrane systems, and the subsequent degree of lasting damage depends highly on the repair of the membrane systems themselves. The two processes of repair, physical and physiological–biochemical processes, can complement each other to a certain extent.

Seed remains of common millet (Panicum miliaceum L.) were excavated from sites of ad 4th-century Darhan (Mongolia), and ad 15th-century Budapest (Hungary). Because the 15th-century medieval grains looked so intact, a germination test was carried out under aseptic conditions, which resulted in swelling of the grains but no cell proliferation or germination. Ancient DNA (aDNA) was extracted from the aseptic grains; analysed for amplified fragment length polymorphisms (AFLP), simple sequence repeats (SSR) and mitochondrial DNA (mtDNA); and compared with the modern millet cultivar ‘Topaz’. AFLP analysis revealed that extensive DNA degradation had occurred in the 4th-century ancient millet, resulting in only 2 (1.2%) AFLP fragments (98.8% degradation) amplified by MseCAA–EcoAGT, compared to the 15th-century medieval millet, with 158 (40%) fragments (60% degradation), and modern millet cultivar ‘Topaz’ with 264 fragments (100%). EcoAGT–MseCAA was found to be the most effective selective-primer combination for the analysis of medieval and modern millet. Eight AFLP fragments were sequenced after re-amplification and cloning. Microsatellite (SSR) analysis at the nuclear gln4, sh1, rps28 and rps15 loci revealed one SNP (single nucleotide polymorphism) at the 29th position (A→G) of rps28 locus, compared to modern millet. An mtDNA fragment (MboI), amplified at the 18S–5S ribosomal DNA (rDNA) locus in the medieval millet, showed no molecular changes compared to modern millet. The results underline the significance of aDNA extraction and analysis of excavated seeds for comparative analysis and molecular reconstruction of ancient and extinct plant genotypes.

In vitro techniques are essential for Striga research and the development of appropriate control methods. In the laboratory, pre-screening of non-host or false-host plants of Striga for trap cropping or the screening of hosts for resistance involves visual evaluation of Striga seed germination that may be stimulated by plant parts or root exudates. This technique is presently laborious because the small Striga radicles are colourless. A number of solutions were evaluated to visualize the radicles of Striga hermonthica to obtain a reliable, simple and fast staining method yielding good contrast for visual observation, with readily available, inexpensive and minimally toxic dyes and staining solutions. The inks Brilliant Blue (Pelikan), Blue (Geha) and Brilliant Green (Pelikan), in either vinegar or lactic acid, produced radicles with excellent contrast, whereas radicles stained with Brilliant Red (Pelikan), Black (Sheaffer), Brilliant Black (Pelikan) gave good contrast. Striga radicles stained with Aniline Blue in vinegar or lactic acid, or Coomassie Brilliant Blue R250 in lactic acid, showed excellent contrast. Radicles stained with Ink Black (Sheaffer), Cotton Brown, or Rubin S in either vinegar or lactic acid also showed good contrast. With water as the diluent, only Lactophenol Blue showed excellent contrast. For health, safety and environmental concerns, availability and staining time, Blue ink in household vinegar (5% acetic acid) appears to be an excellent dye for Striga radicles and could be used in routine Striga germination assays.

Low soil water potential limits or prevents germination and emergence of rainfed winter wheat (Triticum aestivum L.). This phenomenon is particularly pronounced in the winter wheat–summer fallow region of the US Inland Pacific Northwest, where wheat is routinely sown deep to reach moisture with 12–15 cm of soil covering the seed. Wide differences in seedling emergence among winter wheat cultivars have been reported, but few previous experiments have examined germination differences among cultivars as a function of water potential. The objective of our laboratory study was to quantify seed germination of five commonly sown winter wheat cultivars (Moro, Xerpha, Eltan, Buchanan and Finley) at seven water potentials, ranging from 0 to − 1.5 MPa. Germination was measured as a function of time for a period of 30 d. At higher water potentials (0 to − 0.5 MPa), all cultivars had germination of more than 90%. At the lowest water potentials ( − 1.0 to − 1.25 MPa), however, Moro consistently exceeded the other cultivars for speed and extent of germination, with total germination of 74% at − 1.0 MPa and 43% at − 1.25 MPa. Since its release in 1966, Moro has been sown by farmers when seed-zone water conditions are marginal. Scientists have long known that coleoptile length is an important factor controlling winter wheat seedling emergence from deep sowing depths. In addition to having a long coleoptile, our data suggest that Moro's known excellent emergence ability from deep sowing depths in dry soils can also be attributed to the ability to germinate at lower water potentials than other cultivars.

When detecting the adventitious presence of transgenic plants (AP), it is important to use an appropriate testing method in the laboratory. Dorfman's group testing method is effective for reducing the number of laboratory analyses, but does not consider the case where AP is diluted below the sensitivity of the analyses, which causes the rate of false negatives to increase. The objective of this study is to propose binomial and negative binomial probabilistic models for determining the required sample size (n), number of pools (g), and size of the pool (k) for detecting individuals possessing AP with a probability ≥ (1 − α) (for a small α) given: (1) pool size (k); (2) estimated proportion of individuals with AP in the population (p); (3) concentration of the trait of interest (AP) in individual seeds (w); and (4) detection limit of the test (c) (AP concentration in a pool below which it cannot be detected). The proposed models consider the different rates of false positives (δ) and false negatives (λ), and the assessment of consumer and producer risks. Results have shown that when using the negative binomial, a required sample size n can be determined that guarantees a high probability that m individuals or g pools containing AP will be found. The pools formed have an optimum size, such that one element with AP will be detected at a low cost. The negative binomial distribution should be used when it is known that the proportion of individuals with AP in the population is p < 0.1; thus, it is guaranteed that m individuals or g pools of individuals with AP will be detected with high probability.

To understand the beneficial effects of cold conditions during the alleviation of dormancy of walnut (Juglans regia L.), lipid mobilization, gluconeogenesis and changes related to ageing were compared in dormant kernels incubated in the cold and under deteriorating warm conditions. Stratifying kernels at 5°C for 30 d enhanced their germination capacity, whereas warm-incubated (27°C) kernels turned rancid after 20 d and had reduced germination. Kernel imbibition was sufficient to bring about lipid mobilization, irrespective of temperature of incubation. Although imbibed kernels displayed high isocitrate lyase activity, starch and soluble sugar accumulation occurred only under cold conditions. Deteriorative 64 kDa fatty acyl-ester hydrolase activity declined in cold-stratified kernels. Cold treatment also led to reduced lipid peroxidation and hydrogen peroxide in kernels. The activity of NADP+-isocitrate dehydrogenase, an NADPH-generating enzyme, declined in warm-incubated kernels. Thus, warm-incubated kernels undergo ageing associated with oxidative stress, but there are beneficial effects of cold stratification in preventing deteriorative ageing-related processes. Imbibition is sufficient to allow lipid mobilization to occur in dormant walnut kernels, although cold stratification accompanied by gluconeogenesis is essential for kernel germination.

Expansins are a multi-gene family of proteins involved in changes in cell wall properties. In seeds where the embryo is completely surrounded by the endosperm, dormancy breakage requires weakening of the micropylar endosperm, increased embryo growth potential or both. Cell wall alterations are fundamental components of these processes, and expansins are thought to participate in them. Here, we explore the possible involvement of expansins in the control of germination by phytochrome in Datura ferox L. seeds. Based on the conserved sequences of known expansin genes, corresponding primers were designed to investigate the expression of expansin mRNAs by reverse transcriptase polymerase chain reaction (RT-PCR). One expansin mRNA was detected in micropylar endosperm of D. ferox, while two mRNAs were present in the embryo prior to radicle emergence. Expansin transcript content is promoted by red (R) light, both in the micropylar endosperm and the embryo; these effects of R are far-red light (FR) reversible, displaying a typical low-fluence response (LFR) in a way fully consistent with the photocontrol of germination. On the other hand, when the promotion of germination by the LFR is antagonized by exposing the seeds to continuous FR through a high-irradiance response (HIR) or by ABA, the inhibitory treatments do not affect expansin mRNA abundance. The results support the participation of expansins in the promotion of germination by LFR of phytochromes, and suggest that inhibition of germination by HIR or ABA does not include a reduction in the transcription of expansin genes.
