Efficient seed storage is a shared concern among the growing number of seed banks established for crop improvement or ex situ conservation. Container properties greatly affect seed interactions with the environment and the overall cost and success of seed banking operations. Several material properties contribute to their suitability as seed containers. This paper provides a consolidated list of water vapour permeability properties of thermal plastics commonly used for packaging. Composite packages with layers of film with different properties provide distinct advantages to seed banks. Different seed banks must rank the importance of the various factors depending on their mission and resources. Once the risks, costs and benefits are weighed, an appropriate strategy can be developed that addresses a seed bank's specific needs. Because there are many problems and several solutions, it is likely that strategies will vary among seed banks. This response details variables to consider when selecting seed storage packages, and focuses on water diffusion rates of packages with different compositions. A ‘moisture audit’ will help seed bank operators make informed decisions about packaging.

Physiological dormancy of scarified seeds of Townsville stylo (Stylosanthes humilis HBK) was released by acidic aluminium (Al3+) solution. Antiethylenic substances inhibited germination of low-pH-stimulated dormant seeds, with a correspondingly low ethylene production and low activity of 1-aminocyclopropane-1-carboxylate (ACC) oxidase in seeds. On the other hand, antiethylenic substances did not decrease the germination of Al3+-stimulated seeds, but ACC oxidase activity and ethylene production by the seeds was decreased to a large extent. These data provide evidence that dormancy breakage by Al3+ differs from that caused by low pH and is not associated with ethylene production. Similarly to Al3+ action, methyl viologen (MV), a reactive oxygen species-generating compound, broke dormancy of Townsville stylo seeds. Sodium selenate and N-acetyl cysteine, antioxidant compounds, largely decreased germination of MV- and Al3+-stimulated dormant seeds. Altogether these data point to oxidative radicals constituting key molecules in the chain of events triggered by Al3+ leading to dormancy breakage.

Seeds of Celosia argentea L. displayed an absolute requirement for light for germination. Germination could be induced by far-red light as well as red light, and therefore, the effect of red light was not completely reversible by far-red light. A considerable proportion of the seed population was sensitive to red light within the range 0.01–0.1 μmol m−2, which suggested that phytochrome was operating in the very low fluence response mode. Seeds showed a gradual increase in germination in response to red light with an increasing duration of previous dark imbibition. Interruption of the dark imbibition period with very low fluence (10−4−10–1 μmol m−2) or low fluence (1–103 μmol m−2) red light prevented germination subsequent to a terminal, saturating red light irradiation. Increasing responsiveness to red light with increase in imbibition temperature and inhibition of development of photosensitivity by cycloheximide suggested that phytochrome synthesis occurred during the dark imbibition period. An absolute requirement for light for seed germination, inhibition of seed germination on interrupting the dark imbibition period with very low fluence or low fluence red light, and the recovery from photoinhibition of seeds in darkness are consistent with the hypothesis that PhyA is the principal phytochrome involved in the photoregulation of seed germination in C. argentea.

The effect of dry storage under ambient laboratory conditions on after-ripening and survivorship was tested on seeds of the geographically-widespread Solidago altissima and S. nemoralis and the narrow-endemic S. shortii. Freshly-matured seeds of S. altissima collected in 1991 and in 1992 germinated to low or moderate percentages in light at 15/6, 20/10 and 25/15°C and to high percentages at 30/15 and 35/20°C, whereas those of S. nemoralis and S. shortii germinated to low percentages over the range of temperature regimes. After 0.8–1.8 years of storage, 1991 seeds of S. altissima incubated in light germinated to high percentages at 25/15, 30/15 and 35/20°C, those of S. nemoralis did so at 30/15 and 35/20°C and those of S. shortii at 20/10, 25/15, 30/15 and 35/20°C; 1992 seeds of all three species germinated to high percentages at 20/10, 25/15, 30/15 and 35/20°C. Freshly-matured 1991 and 1992 seeds of the three species germinated to low percentages in darkness over the range of temperature regimes, and only seeds of S. shortii germinated to high percentages after 0.8–1.8 years of storage. Compared with cold stratification, dry storage was only moderately effective in breaking dormancy in these three species. The primary difference in after-ripening of seeds of the three species was that seeds of the narrow endemic germinated to higher percentages in darkness than those of its two geographically-widespread congeners. Survivorship curves for 1991 and 1992 seeds of S. altissima and S. nemoralis and for 1992 seeds of S. shortii were of Deevey Type I; the survivorship curve for 1991 seeds of S. shortii was closest to Type II. Longevity of 1991 seeds of S. altissima, 1992 seeds of S. nemoralis and 1991 and 1992 seeds of S. shortii was <4.0 years, whereas that of 1991 seeds of S. nemoralis was <2.3 years; 5% of 1992 seeds of S. altissima were viable after 4.0 years.

Seed quality is a complex trait that is the result of a large variety of developmental processes. The molecular-genetic dissection of these seed processes and their relationship with seed and seedling phenotypes will allow the identification of the regulatory genes and signalling pathways involved and, thus, provide the means to predict and enhance seed quality. Natural variation for seed-quality aspects found in recombinant inbred line (RIL) populations is a great resource to help unravel the complex networks involved in the acquisition of seed quality. Besides extensive phenotyping, RILs can also be profiled by -omics technologies, such as transcriptomics, proteomics and metabolomics in a sophisticated so-called generalized genetical genomics approach. This combined use of physiology, genetics and several -omics technologies, followed by advanced data analysis, allows the construction of regulatory networks involved in the various attributes of seed and seedling quality. This type of analysis of the genetic variation in RIL populations in combination with genome-wide association (GWA) studies will allow a relatively rapid identification of genes that are responsible for quality-related traits of seeds and seedlings. New developments in several -omics technologies, especially the fast-evolving next-generation sequencing techniques, will make a similar system-wide approach more applicable to non-model species in the near future and this will be a huge boost for the potential to breed for seed quality.

Partial drying of non-dormant seeds had little effect on germination behaviour compared with undried controls. In contrast, partial drying resulted in a marked increase in the germination response of freshly harvested (dormant) seeds of Z. palustris. Partial drying also resulted in a 100-fold reduction in the concentration of gibberellins (GA4+7) required for maximum germination. Although the concentration range of applied abscisic acid (ABA) that permitted germination was unaltered, partial drying increased the final germination percentage at all concentrations tested. The concentration of ABA above which germination was reduced in freshly harvested (dormant) seeds of Z. palustris was 3.16 × 10−7m compared with 10−4m in fresh (non-dormant) seeds of P. coarctata. Changes in the germination response of Z. palustris seeds to applied growth regulators, following partial drying or cold stratification might be explained by similar changes in hormonal status.

The resilience of seed quality in rice (Oryza sativa L.) to flooding was investigated. Pot-grown plants of the japonica cv. Gleva, the indica cv. IR64, and the introgressed line IR64-Sub1 were submerged in water, to simulate flooding, for 3‒5 days at different stages of seed development and maturation. Mean seed weight, pre-harvest sprouting, ability to germinate, and subsequent longevity in air-dry storage were assessed. Whereas seed quality in both IR64 and IR64-Sub1 was resilient to submergence, in Gleva the longer the duration of submergence and the later in development when plants were submerged the greater the pre-harvest sprouting. Thousand seed dry weight was reduced more by submergence in Gleva than IR64 or IR64-Sub1. At harvest maturity, few pre-harvest sprouted seeds were able to germinate upon rehydration after desiccation to 11‒12% moisture content. Seed longevity of the non-sprouted seed fraction in air-dry hermetic storage (40°C, 15% moisture content) was not affected greatly by submergence, but longevity of the japonica rice was less than that of the indica rices due to the former's steeper seed survival curves. Longevity of the two indica rices was predicted well by the seed viability equation and previously published estimates of viability constants for rice. The greater dormancy of IR64 and IR64-Sub1, compared with Gleva, enhanced resilience to pre-harvest sprouting and reduced thousand seed dry weight from plant submergence. There was little or no effect of plant submergence on subsequent air-dry storage longevity of non-sprouted seeds in any genotype.

The impact of rehydration on a multicellular organism was studied in lettuce (Lactuca sativa L.) embryos, using cryo-scanning electron microscopy (cryo-SEM). Naked embryos were sensitive to imbibitional stress, whereas embryos with an intact, thick-walled endosperm were not. Imbibitional injury to naked embryos was mainly confined to the outer 2–3 cell layers of the axis. These cells failed to swell and appeared poorly hydrated. Deeper layers were not affected even after extended periods of cold rehydration. The proportion of damaged cells (6–7% of total) roughly corresponded with the additional K+ that gradually leached from the embryos. Damaged embryos were able to survive the loss of their surface layers and form adventitious roots. The swelling of inner tissues caused the dead surface layers to rupture into patches. Plasma membranes in dried embryos showed normal bilayer structure with a homogeneous distribution of intra-membrane particles (IMPs), also after non-injurious rehydration. Imbibitionally damaged plasma membranes showed many irregularities, such as globular insertions, that probably resulted from malfusions in the ruptured membrane, but the IMPs were still randomly distributed.

Seeds of Douglas fir (Pseudotsuga menziesii var. menziesii [Mirb.] Franco) were initially germinated at six constant temperatures (10–35°C), following 0–48 weeks incubation under moist conditions at 4°C, i.e. prechill or stratification. The best, single germination temperature was 15°C for determining the efficacy of subsequent dormancy breakage treatments. Seeds from the same seedlot were then adjusted to 10, 15, 20, 25, 30, 35 and 40% moisture contents (mcfw, fresh weight basis), prechilled for 0, 2, 4, 8, 16, 32, 64 and 128 weeks and transferred to 15°C. A smoothed bivariate spline was used to model the results and showed that virtually all combinations of moisture content and prechill duration significantly stimulated germination capacity, but that the optimal germination percentage (≥93%) was stimulated only by various combinations of between 30 and 35% mcfw and 25 and 48 weeks prechill. At optimal moisture contents (30 and 35%) , extending the prechill duration beyond 48 weeks led to a decrease in germination capacity. This was not due to dormancy reintroduction, but was caused largely by seed death. Regression models using a weighting function, to account for differences in standard deviations, demonstrated significant increases in the mean moisture content of individual seeds at higher moisture contents (≥25%) and longer prechill durations (≥64 weeks) that were concomitant with significant decreases in dry weight. The most likely explanation for this was seed respiration. The combined results suggest that dormancy breakage in Douglas fir seeds requires a hydration level sufficient for respiration to take place, and that, after maximal dormancy release, seeds at the highest mc (35–40%) exhaust their food reserves and begin to deteriorate.

Endosperms of fenugreek and carob which have been leached following their isolation exhibit increased activity of endo-β-mannanase, part of which is released into the surrounding incubation medium. This activity was suppressed by the addition to the endosperms of abscisic acid or endosperm/seed coat leachate. The leachate from carob inhibited the increase of endo-β-mannanase activity in fenugreek and that from fenugreek inhibited enzyme activity in carob but more weakly; neither of these leachates inhibited the production of α-amylase in wheat endosperm. Polyethylene glycol-induced stress on non-leached endosperms inhibited endo-β-mannanase production, but not when the stress was applied following endosperm leaching. However, the stress did reduce the amount of enzyme released into the surrounding incubation medium. Several isoenzymes of mannanase could be detected on isoelectric focusing activity gels in the endosperms of both fenugreek and carob, but their pis differed between the two species. When activity was suppressed by ABA or leachate, all pl forms declined equally.

Features of chemicals which break seed dormancy and the mechanisms of their action are discussed with reference to results obtained with red rice (Oryza sativa) and other relevant research. The significance of chemical structure, including the nature and location of functional groups, is considered, and the fact that similar structures can elicit different biochemical responses is illustrated with examples. As yet it is unclear whether changes in cell pH, highly correlated with the uptake of, and metabolism to, weak acids, and which some consider a possible trigger or marker in dormancy breaking, are causative, consequential or merely correlative. Uptake and metabolism of dormancy-breaking compounds is, or can be, very rapid and the importance of establishing the time course of dormancy-breaking, as opposed to germination events, is emphasized.

Methanol accumulates in axis tissues of maturing soybean seeds, correlating with preharvest seed deterioration. Accumulation of methanol appears to be associated with the enzymic demethylation of pectin methyl esters by pectinesterase (PE; EC 3.1.1.11). To characterize PE in developing and maturing soybean (Glycine max (L.) Merrill) seeds, enzyme activity was assayed in axis and cotyledon tissues. Activity per g fresh weight was 20–25 times higher in axes than in cotyledons with highest activities between 45 and 60 days after flowering (DAF). Twenty to 33% of the total PE activity was in the ‘soluble’ form (extracted with water, 0.5 M sucrose, 1 M sucrose and water). Soluble and cell-wall-bound PE (subsequently extracted with 1 M NaCI) were purified and characterized from axes of seeds at 45–60 DAF. Purification of PE was achieved through concentration of extracts by ultra-filtration, precipitation with ammonium sulfate (30–80% saturation), dialysis, gel filtration on Sephadex G-75 columns, and ion exchange chromatography on CM Sepharose CL-6B. Further purification of both soluble and bound PE was by isoelectric focusing (IEF) on ultrathin layers of polyacrylamide gel with simultaneous detection of protein and PE activity. It was possible to follow seven bands exhibiting PE activity with pl values between 6.0 and 9.5 in 1 M NaCI-extracts of total homogenates. Differences in the IEF patterns of bound and soluble PE were observed. Whereas the bound enzyme exhibited more basic PE bands (pl 8–9.5), the soluble enzyme had more active bands at pl 6.5, 7.0 and 7.5. The Mr was close to 33 000 and the pH optimum was 7.8 for both soluble and bound PE.

For seed banks showing seasonal changes in their dormancy level, the possibility of predicting temporal patterns of emergence depends on establishing a robust relationship between temperature and the rate of dormancy loss and induction. However, although the effect of temperature on dormancy loss has been extensively studied, less work has been advocated to the quantification of temperature effects on dormancy induction. In the present work, we quantified temperature regulation of dormancy induction in Polygonum aviculare seeds using the hydrotime model. To study induction into secondary dormancy, seeds previously released from primary dormancy through stratification at 5°C were stored at dormancy-inductive temperatures of 10, 15, 20 and 25°C for different periods. During storage, seeds were germinated at different temperatures and water potentials, and hydrotime model parameters were derived. Changes in hydrotime model parameters (mean base water potential for germination and its standard deviation, and the hydrotime required for germination) during dormancy induction were described by adjusting exponential equations. Obtained results indicated a minimum temperature for dormancy induction of 8.7°C and the existence of a bi-linear relationship between rate of induction into secondary dormancy and storage temperature, in which storage temperatures around 25°C showed a higher dormancy induction rate than those below 20°C. Developed model equations were then used to predict changes in germination behaviour during dormancy induction at different temperatures, showing a good agreement between simulated and observed values.

The detection and quantification of the prevalence of genetically modified organism (GM) contamination in seed exports is a critical element of regulatory compliance. While the procedures to reliably detect high levels of GM contamination are well understood, no comparable statistical approaches are available for the quantification of levels of GM prevalence below the established detection rate of standard tests. We present a simple statistical approach based on simulation modelling for the quantification of low levels of GM contamination. The approach can be modified to match any sampling regime and can account for rates of false positive and negative assay results. The application of this method is demonstrated using the low level of contamination in Canadian flax breeder seed lots by the GM flax variety ‘Triffid’. We show that GM contamination is likely present in seed lots at rates between two GM seeds per million and six seeds per hundred thousand. We also show that this low level of presumed contamination is indistinguishable from the number of positive tests expected from a clean seed lot given the potential rates of false positive tests.

The present study assessed the desiccation sensitivity of the excised embryonic axes of the following Amaryllidaceae species: Scadoxus puniceus, Amaryllis belladonna, Nerine huttoniae, N. humulus, N. bowdenii, N. filifolia, Haemanthus humulus humulus, H. coccineus, H. deformis, H. bakerae, Brunsvigia gregaria, Brunsvigia orientalis, Boophane disticha, Strumaria discifera, Crinum macowanii and C. bulbispermum. Excised embryonic axes were rapidly dehydrated (flash-dried). Curves of water content (g g− 1) and viability (%) versus drying time (min) were used to interpolate the water content (WC) and drying time (DT) values corresponding to 80% viability (WC/DT, V = 80) for individual species, and used to compare desiccation sensitivity between developmental stages and among years, provenances, species and genera. The seeds of all 16 species were shed with high axis water contents (2.25 ± 1.34 to 6.95 ± 0.89 g g− 1) and were sensitive to desiccation, with WC V = 80 values ranging from 0.13 to 3.15 g g− 1. Germination in amaryllids is unusual in that part of the cotyledon, to which the embryonic axis is attached, grows right out of the seed. It is at this stage that the axes were found to be more desiccation-sensitive in six out of nine species investigated. Seed fresh mass (SFM), axis shedding water content (SWC) and the degree of desiccation sensitivity were species characteristic. SWC, SFM and drying time were not significantly related to desiccation sensitivity. Parent plants were not confined to any particular biome type, occurring in areas characterized by seasonal dry spells. Parent plant habitat and seed characteristics were not useful in predicting the degree of desiccation sensitivity across species belonging to the same family.

Maternal effects on offspring seeds are mainly caused by seed position on, and the abiotic environment of, the mother plant. Seed heteromorphism, a special form of position effect, is the production by an individual plant of morphologically distinct seed types, usually with different ecological behaviours. Seed heteromorphism is assumed to be a form of bet hedging and provides an ideal biological model to test theoretical predictions. Most studies of maternal effects on seeds have focused on abiotic environmental factors and changes in mean seed traits of offspring. Suaeda salsa is an annual halophyte that produces dimorphic seeds within the same inflorescence. We tested the hypothesis that plants grown from brown seeds of S. salsa have a higher offspring brown seed:black seed morph ratio and variance in seed size than plants from black seeds. Results from a pot experiment showed that plants grown from brown seeds had a higher brown seed:black seed ratio than plants grown from black seeds. This is the first layer of dynamic bet hedging. Brown seeds had higher size variation than black seeds, and seeds produced by plants from brown seeds also had higher seed size variation than plants grown from black seeds. This is the second layer of dynamic bet hedging. Thus, the maternal effect of seed heteromorphism is dual dynamic bet hedging. Furthermore, for seed traits we verified for the first time the theoretical prediction that an increase in offspring size variability induces an increase in the mean size of offspring.

The Brazilian literature on seed germination testing of tropical forest species, although still insufficient, is extensive for species with economical potential. However, this information is still fragmented and often reported in scientific communications of modest circulation. In this summary, we present the results of a validation process for germination tests on Brazilian forest species, which had as a starting point the ample reviewing of current methods, besides exhaustive pre-testing in certified laboratories. Specific protocols were created for each species, including substrate, temperature, seed disinfection, breaking of dormancy, seedling classification and other information. These protocols were sent to research laboratories and accredited by the Ministry of Agriculture, together with seed lots of three distinct qualities. The presuppositions of normality of the residuals and homoscedasticity of the variations were previously tested and the completely randomized factorial design statistical model was applied to the normal seedlings of each species. After this analysis, germination test methods for seeds of Astronium fraxinifolium, Ceiba speciosa, Cybistax antisyphilitica, Enterolobium contortisiliquum, Guazuma ulmifolia, Lafoensia pacari, Mimosa caesalpiniaefolia, Peltophorum dubium, Pseudobombax tomentosum and Pterogyne nitens, all forest species of different Brazilian biomes, were considered to be validated.

Eleven sunflower (Helianthus annuus L.) inbred lines were evaluated in field and laboratory studies under drought and irrigation. In the field, lines B59, R419 and B67 had reduced seed and oil yield under drought, while no reduction was observed for R432, HAR4 and B71. Lines HA89, R415, R049, RHA274 and R423 presented intermediate responses. In laboratory tests, seeds of line B59 had reduced germination percentages at 200 and 400 mM mannitol, while germination of seeds of lines R432, B71, HAR4, RHA274 and HA89 was reduced only at 400 mM mannitol. Drought-sensitive B59 and drought-tolerant B71 grown under irrigation and drought conditions in the field were selected for hormone assays. Abscisic acid (ABA) and its catabolites in pericarp, embryonic axis and cotyledons of dry and germinated seeds of B59 and B71 were determined. ABA was the major component of the pericarp of dry seeds from B71 and B59 plants grown under drought. The embryonic axis of B71 dry seeds from drought-grown plants also showed high ABA content. The major findings from this study are: (1) the drought-sensitive and -tolerant lines exhibited different ABA and catabolite profiles; (2) water environment during maternal plant growth affected ABA content and the composition of catabolites in mature and germinated seeds; (3) ABA content did not affect germination performance in our conditions; and (4) the dry and germinated seed parts showed different ABA and catabolite profiles.

Xenia is the direct effect of pollen genotype on the development and characteristics of the seed and fruit in the period that spans from fertilization to seed germination. Xenia effects cause phenotypic variations in the seed and fruit, which have importance for seed and fruit production but are not heritable to the progeny. Two hypotheses have been proposed as a mechanism for xenia: the hormonal hypothesis and the mobile mRNAs hypothesis. Although xenia effects have been studied in seeds and fruits in many crops, its effects and mechanism have been poorly studied at the molecular level. The aim of this work was to perform an initial screening of the xenia effect in the hybrid endosperm at the molecular level by differential gene expression among different pollen genotype sources from Paspalum notatum Flüggé. In order to characterize xenia effects and mechanisms, crosses were made between an emasculated mother plant with donors from two different pollen genotypes. RNA was isolated from ovaries 3 h after pollination. Some of the 24 differentially expressed transcript-derived fragments (DETDFs) provided relevant information. Four of those DETDFs were related to germination, pollen tube growth and pollen–pistil interaction. Seven DETDFs were associated with seed development and production. Finally, four DETDFs were predicted to encode for mobile mRNAs. These DETDFs might be involved in xenia effects and mechanisms in P. notatum.

Sucrose, raffinose and stachyose accumulate in soybean [Glycine max L. (Merrill)] embryos during seed maturation. To determine the relationship of plant maternal composition on seed composition, soluble carbohydrates in three 1-cm2 leaf punches at three plant growth stages (R2, R3, R6) and in seed coat cup exudates in planta were analysed at four 30-min intervals on soybean plants (R5) with low-raffinose, low-stachyose (LRS) seeds expressing the mutant stc1 phenotype; low-raffinose, low-stachyose and low-phytin (LRSP1, LRSP2) seeds expressing the mutant mips phenotype; or normal raffinose, stachyose and phytin (CHECK) seeds expressing the Stc1 and Mips phenotype. Leaf sucrose (23.6 μg cm− 2), myo-inositol (9.3 μg cm− 2), d-chiro-inositol (6.7 μg cm− 2), d-ononitol (0.76 μg cm− 2), d-pinitol (50.1 μg cm− 2) and total soluble carbohydrates (107.1 μg cm− 2) were not significantly different between phenotypes. d-chiro-Inositol, myo-inositol, d-pinitol and sucrose were unloaded from soybean seed coat cups in planta at decreasing rates over the four sequential periods of sampling. Unloading rates of sucrose and myo-inositol were highest for LRS, d-pinitol was highest for LRSP2, and d-chiro-inositol was not different between LRS, LRSP1, LRSP2 and CHECK. Free cyclitols were 60% of total soluble carbohydrates in leaves and 20% in seed coat cup exudates. Except for sucrose and d-pinitol, seed phenotype had little influence on the composition of compounds unloaded from seed coats to maturing embryos of low-raffinose, low-stachyose seeds. Maternally supplied cyclitols may contribute, in part, to changes in the composition of cyclitol galactosides stored in mature seeds.
