Studies were conducted to determine if small embryos (i.e. low embryo length:seed length ratio) in mature dwarf seeds (0.2–2 mm) are underdeveloped. In this case, they would grow (inside the seed) prior to germination, and seeds would have morphological or morphophysiological dormancy. Prior to radicle emergence, embryo length in seeds of Drosera anglica (Droseraceae), Campanula americana, Lobelia appendiculata, L. spicata (Campanulaceae) and Sabatia angularis (Gentianaceae) increased 0, 103, 182, 83 and 57%, respectively. Since embryo growth did not occur in seeds of D. anglica prior to germination, embryos, although small, are fully developed; seeds have only physiological dormancy. The underdeveloped embryo in seeds of C. americana has little or no physiological dormancy; thus, seeds have morphological dormancy. On the other hand, underdeveloped embryos in seeds of L. appendiculata, L. spicata and S. angularis are physiologically dormant, and seeds have morphophysiological dormancy. Therefore, since small embryos in dwarf seeds may or may not be underdeveloped, assignment of seeds to a dormancy class requires that studies be done to determine if embryos grow inside the seed before germination can occur. Such information is important in understanding the evolutionary relationship of the different kinds of seed dormancy.

Storage of tomato seeds under deteriorating conditions reduced the rate of germination, the uniformity of germination, total germination and the proportion of normal seedlings. Pre-storage humidification and hydropriming of seeds resulted in a significant increase in resistance to deterioration. In contrast, osmoprimed seeds were more deterioration sensitive. Humidification or hydropriming for one day did not allow nuclei to enter the S phase of the cell cycle. Osmopriming strongly increased the percentage of nuclei with replicated DNA in the embryonic root tip, indicating initiation of the cell cycle and progression towards the G2 phase. The interaction between the prestorage treatments, cell cycle progression and deterioration resistance, is discussed. Based on the changes in moisture content equilibrium and cell cycle activity it is hypothesized that the beneficial effects of pre-storage humidification and hydropriming were related to metabolic activities induced by the partial hydration and that the adverse effects of osmopriming were caused by a decrease in DNA repair activity due to progression in the cell cycle.

Recruitment-related traits (development and dispersal of seeds, emergence, leaf dynamics, growth and survival of seedlings) were studied in Japanese elm (Ulmus davidiana var. japonica) in relation to disturbance at the soil level (bare soil, litter and control) or the canopy level (forest edge (FE), small gaps (SG) and forest understorey (FU)), or both, in a riverside forest from 1990 to 1993. Seed germination was also tested in the laboratory. Litter accumulation severely inhibited seedling emergence. Seedling emergence from a single seed rain occurred in summer immediately after seed dissemination and in the following spring. In summer, seedling emergence was higher in FE than in FU. In the following spring more seedlings emerged in FU than in FE and SG, resulting in non-significant differences in the percentage emergence among the three sites in bare soil. Darkness or far-red light induced seed dormancy, but after chilling they started to germinate again under white or red light conditions. These results explained emergence phenology under changing light conditions in the field. In FU, the spring emerging cohort grew and survived better than the summer emerging one. In FE the reverse was true. Japanese elm avoids unfavourable periods by escaping either in time (dormancy) or in space (dispersal). Such traits compensate for late and compressed seed dissemination, and improve seedling growth and survival not only when disturbance occurs at both canopy and soil level (bare soil in FE) but also only at soil level (bare soil in FU), which may occur more frequently in riverside forests.

Red rice (Oryza sativa L.) dispersal units (florets) were dry-afterripened for 0–8 weeks and subsequently incubated at 30°C in polyethylene glycol (PEG) solutions with water potentials from 0 to − 1.6 MPa. Germination percentages and rates increased with dry-afterripening and water potential of the incubation medium. The seed population exhibited a normal distribution of base water potentials (Ψb, i.e. minimum water potential allowing germination) among individual seeds, characterized by three parameters: the hydrotime constant (θH), the mean base water potential (Ψb) and the standard deviation of the base water potential distribution (σΨb). Changes in germination during afterripening could be described by modifications of such parameters, particularly Ψb, which was employed to derive an index, DH(ARX = Ψb(ARX) − Ψb(ARN), where DH(ARX) represents a measure of dormancy of the seed population (in MPa) based on the hydrotime model, Ψb(ARX) is the mean base water potential of the seed population at any afterripening time X, and Ψb(ARN) is the mean water potential of the non-dormant (fully afterripened) population. The introduction of this index permitted interpretation of afterripening as a measurable reduction in the dormancy status of the seed, with progressive acquisition of both full germinative capacity and maximum germination rate, as anticipated by the hydrotime model. Moreover, secondary dormancy was induced proportionally to the reduction in water potential in the dark. Susceptibility to secondary dormancy induction was defined through DI(ARX), an index analogous to DH(ARX). These indices revealed that, in red rice, both breaking of primary dormancy and the inducibility of secondary dormancy followed decay kinetics with different sensitivities to the duration of dry storage.

A climate warming of 2–5°C by the end of the century will impact the likelihood of seed germination of sugar maple (Acer saccharum), a dominant tree species which possesses a restricted temperature range to ensure successful reproduction. We hypothesize that seed origin affects germination due to the species' local adaptation to temperature. We tested this by experimentally investigating the effect of incubation temperature and temperature shifting on sugar maple seed germination from seven different seed sources representing the current species range. Survival analysis showed that seeds from the northern range had the highest germination percentage, while the southern range had the lowest. The mean germination percentage under constant temperatures was best when temperatures were ≤5°C, whereas germination percentages plummeted at temperatures ≥11°C (5.8%). Cool shifting increased germination by 19.1% over constant temperature treatments and by 29.3% over warm shifting treatments. Both shifting treatments caused earlier germination relative to the constant temperature treatments. A climate warming of up to +5°C is shown to severely reduce germination of seeds from the southern range. However, under a more pronounced warming of 7°C, seed germination at the northern range become more affected and now comparable to those found from the southern range. This study states that the high seed germination percentage found in sugar maple at the northern range makes it fairly resilient to the warmest projected temperature increase for the next century. These findings provide forest managers with the necessary information to make accurate projections when considering strategies for future regeneration while also considering climate warming.

Embryos of Araucaria angustifolia seeds showed no dormancy; they germinated easily at temperatures ranging from 10° to 30°C, and the thermal optimum was about 25–30°C; they were recalcitrant. At harvest, their mean moisture content (dry weight basis) was about 120% and they completely lost viability when their moisture content fell to about 30%. The cotyledons were more sensitive to dehydration than the radicle. Dehydration induced deterioration of cell membranes as indicated by a high increase in leakage of solutes. It also resulted in damage in the nuclei, which was not repaired upon rehydration. During desiccation, respiratory activity decreased; however, O2 uptake was not an indication of germination ability, since it was significantly affected only when embryo moisture content reached the critical value of 30%. The decrease in the capacity to convert 1-aminocyclopropane 1-carboxylic acid to ethylene, which was observed at 30–60 min of dehydration, was a very early indicator of deterioration in embryos. Desiccation resulted also in a rapid decrease in the ability for protein synthesis as measured by [35S]methionine incorporation into total protein; 50% inhibition was observed after 30–60 min of desiccation for both axis and cotyledons.

As a consequence of previous lack of success in cryostoring axes excised from newly shed seeds of Trichilia dregeana, the effects of the mode of axis excision on seedling production were investigated. Although vigorous root production occurred, no shoots were produced when the cotyledons were severed as closely as possible to the axis surface (explant-type 0). In contrast, shoot production was increasingly facilitated when small to larger segments of cotyledonary tissue were left attached to the axes (explant-types 1, 2 and 4), which did not compromise axis drying rate. However, root growth of explant-types 1, 2 and 4 was negatively affected, probably by leakage into the medium of an inhibitory or toxic substance(s) from the cut surfaces of the cotyledonary tissue. Microscopical examination revealed that the cotyledons were sessile, and their insertions were contiguous with the shoot apex in axes from newly shed seeds, leading to the suggestion that failure of shoot production by type 0 explants in vitro was the direct consequence of the proximity of the wound sites to the apical meristem. When seeds were stored hydrated for 6 months, the shoot apex had elongated, positioning the apical meristem some distance from the top of the cotyledonary insertions. In contrast to axes excised as type 0 explants from newly shed seeds, the equivalent explants from the stored seeds rapidly formed shoots and leaves in vitro. This indicates that the developmental status of axes, when excised, dictates failure or success in their further development in vitro, and that this aspect needs to be resolved before any further manipulations for cryostorage are attempted.

Seeds of Aconitum napellus subsp. castellanum were physiologically dormant at maturity in early autumn, with underdeveloped embryos. Thus they have morphophysiological dormancy (MPD). Embryos in fresh seeds were on average 1.01 mm long, and they had to grow to 3.60 mm before radicle emergence. Cold stratification at 5°C for 5 months with light enhanced the mean embryo length to 2.73 mm (SE = 0.13) and seed germination to 20%. However, with higher temperatures (15/4, 20/7, 25/10, 28/14 and 32/18°C) embryo growth was small, with no seeds germinating. Optimal germination was achieved after 4 months of cold stratification at 5°C followed by incubation at 20/7°C for 1 month with light, when germination ranged between 70 and 79%, depending on seed age, locality and year of collection. Cold stratification could be substituted by the application of GA3 solution, since mean embryo length in seeds incubated at 25/10°C for 1 month with light was 3.52 mm and the germination was 80%. Since cold stratification was the only requirement for the loss of MPD, the longest embryo growth occurred during this treatment, and GA3 promoted MPD loss, we concluded that A. napellus seeds have intermediate complex MPD. Germination was higher in 4-month stored than in freshly matured seeds. A pronounced variability in germinative patterns at inter-annual and inter-population level was recorded.

The role of smoke in fire-stimulated germination in the Mediterranean Basin has often been underestimated. A few records on smoke-enhanced germination are present in Mediterranean Lamiaceae species, but there is still a shortage of information to allow generalizations about this family to be made. To test the hypothesis that smoke enhances germination in Mediterranean Lamiaceae species, we performed a germination experiment, including aqueous smoke treatments in various concentrations (1:1, 1:10 and 1:100) on seven eastern Mediterranean Lamiaceae taxa. Six of the studied taxa (Lavandula stoechas, Origanum onites, Phlomis bourgaei, Stachys cretica ssp. smyrnaea, Satureja thymbra, Teucrium lamiifolium ssp. stachyophyllum) showed significant increments in germination percentage in at least one smoke treatment, as compared to the control. Moreover, L. stoechas, S. thymbra and T. lamiifolium ssp. stachyophyllum displayed faster germination in at least one smoke treatment than in the control. Of the species showing significant increments in germination percentages after aqueous smoke application, at least one single concentration of smoke solution did not stimulate germination, except in L. stoechas and S. thymbra which responded positively to all smoke treatments. Therefore, the concentration of aqueous smoke that improved germination was species-specific. Our results contribute to the current limited knowledge on smoke-enhanced germination in Mediterranean Lamiaceae, and support the idea that smoke is an important germination cue for this family.

For recalcitrant seeds, mortality curves of germination versus water content typically imply a wide range of desiccation sensitivities within a seed population. However, seed to seed differences in water content, during desiccation, may confound our interpretation of these mortality plots. Here, we illustrate this problem for two batches of Vitellaria paradoxa (Sapotaceae) seeds collected in 1996 and 2002. Whole seeds were desiccated to various target water contents (TWCs) using silica gel. During desiccation, smaller seeds in the population dried most rapidly. Consequently, there was a significant linear relationship between whole-seed water content and seed mass during the drying process. In addition, following desiccation to low TWCs, only the largest seeds in the population retained viability. Taken together, this suggests that the larger seeds survived, not as a consequence of great relative desiccation tolerance, but as a result of taking longer to desiccate. Subsequently, the critical water content (CWC) for viability loss was calculated, based on the assumptions that in the seed population whole-seed water content during desiccation was normally distributed and the smallest, and hence driest, seeds were killed first. Using this approach, the driest seeds in the population that were killed, at each TWC, were always below a single CWC (c. 20% and 26% in 1996 and 2002, respectively). In subsequent experiments the effect of seed size variation on the response to desiccation was confirmed by conducting desiccation screens on seeds sorted into two discrete size classes, i.e. the seed-lot heterogeneity in mass was reduced. Using this approach, the mortality curves had a steeper slope. Furthermore, data for 24 tropical tree species from the Database of Tropical Tree Seed Research (DABATTS) revealed that seed lots with less variability in mass had steeper mortality curves. Thus, taken together, the data suggest that, at least for whole seeds, the wide range of desiccation sensitivities typically inferred is an artefact of seed to seed variation in mass, and hence water contents, during drying.

Seed shelf-life or longevity is difficult to predict or to measure on a practical time scale. Predictive models suggest that water has the same effect on ageing rate for all seed lots within a species and that initial seed quality is the dominating factor explaining within-species variation. These assumptions are used in ‘accelerated ageing’ or ‘controlled deterioration’ (AA/CD) tests, which are commonly used to predict seed longevity in commercial and research applications. In this study, we describe within-species variation of longevity for seeds of Secale cereale and S. strictum (cultivated rye and its wild progenitor) under typical dry storage conditions of a genebank, and show that initial seed quality is an important, but not sole, factor explaining measured longevity. We also test the correlation of seed longevity, measured under humid and dry conditions, using 50 cultivars of rye, wheat (Triticum aestivum) and the intergeneric cross triticale, to assess how well AA/CD tests predict seed shelf-life during dry storage. Known differences in longevity between wheat and rye were confirmed at all water contents, and triticale seeds demonstrated intermediate behaviour. Longevity measured for humid and dry conditions were weakly correlated when comparisons included all grain types and were not correlated in within-grain-type comparisons. Response to moisture varied among cultivars. These findings do not support assumptions made in seed ageing models that use AA/CD tests. Our results suggest that more traits are involved in the expression of seed longevity than those typically measured in studies of initial seed vigour.

As part of a larger survey of biodiversity in private gardens in Sheffield, UK, we examined the composition and diversity of the soil seed banks in each of 56 gardens. Six soil cores from each garden revealed 2759 seedlings of 119 taxa, although the real species richness is likely to be much higher than this. By far the most abundant species were weedy natives, while the most common alien was Buddleja davidii. Seeds of perennial herbs were more abundant than hundreds of all other life forms combined. More frequent species were also more abundant, but the relationship was weak. Numbers of species in the seed bank and in the garden flora were positively but very weakly related. Seeds were quite evenly distributed between 0–5 cm and 5–10 cm soil layers, and most seeds were of species known to have persistent seed banks. Seeds of some species were largely confined to gardens in which the plant was growing, but others were not.

Freshly harvested tomato (Lycopersicon esculentum Mill, cv. Moneymaker) seeds were osmotically primed for 8 d in −1.0 MPa PEG-6000 solution and dried to about 6% water content for storage. Such so–called ‘fresh PEG priming’ enhanced seed germination and improved seedling performance as compared with the untreated control. Fresh PEG priming neither alleviated seed dormancy nor promoted DNA replication as was the case when seeds were dried upon harvest and subsequently primed in PEG (normal PEG priming). However, the addition of 10 μM GA4+7 to the osmotic priming solution triggered replicative DNA synthesis of fresh-priming seeds and further enhanced the germination process. After 5 months of storage in ambient temperature conditions, fresh PEG-primed seeds maintained more positive effects gained from priming, whereas, normal PEG-primed seeds had lost the promoting effects on germination. Normal PEG-primed seeds were much more susceptible to controlled deterioration than fresh PEG-primed seeds. It is suggested that the advancement of germination is negatively correlated with seed storability. The mechanisms of seed priming in relation to nuclear replication activities and physical changes are discussed.

The contents of the cytokinins zeatin (Z), zeatin riboside (ZR), and isopentenyladenine (IPA), the combined contents of gibberellins1 + 3 (GA), and the contents of indoleacetic acid (IAA) and abscisic acid (ABA) were measured during the development of the desiccation-sensitive seeds of Avicennia marina (Forssk.) Vierh. During the stage of histodifferentiation the amounts of these plant growth regulators (PGRs) were measured on whole fruits. During the phase of seed growth and reserve accumulation measurements were made on the embryonic axis, cotyledons and pericarp separately. Patterns in the amounts of PGRs present during histodifferentiation were similar to those reported for desiccation-tolerant seeds and it suggested that this process is under similar ‘hormonal’ control in A. marina as in orthodox seeds. Very high contents of cytokinins, particularly ZR, were present in both axes and cotyledons during reserve accumulation. This is thought to be related to the nature of the reserves accumulated (soluble sugars), rather than to the phenomenon of desiccation sensitivity. With the exception of ABA, embryonic contents of PGRs were relatively high at seed shedding, consistent with the rapid germination of this highly recalcitrant seed. ABA contents in the embryo were low during reserve accumulation, but concentrations in the pericarp increased throughout this development stage. ABA in the perticap could act to prevent precocious germination. The low concentrations of ABA in the embryo could be related to the desiccation-sensitivity of the seeds of A. marina.

Physiological dormancy has been described as a physiological inhibiting mechanism that prevents radicle emergence. It can be caused by the embryo (embryo dormancy) as well as by the structures that cover the embryo. One of its functions is to time plant growth and reproduction to the most optimal season and therefore, in nature, dormancy is an important adaptive trait that is under selective pressure. Dormancy is a complex trait that is affected by many loci, as well as by an intricate web of plant hormone interactions. Moreover, it is strongly affected by a multitude of environmental factors. Its induction, maintenance, cycling and loss come down to the central paradigm, which is the balance between two key hormonal regulators, i.e. the plant hormone abscisic acid (ABA), which is required for dormancy induction, and gibberellins (GA), which are required for germination. In this review we will summarize recent developments in dormancy research (mainly) in the model plant Arabidopsis thaliana, focusing on two key players for dormancy induction, i.e. the plant hormone ABA and the DELAY OF GERMINATION 1 (DOG1) gene. We will address the role of ABA and DOG1 in relation to various aspects of seed dormancy, i.e. induction during seed maturation, loss during dry seed afterripening, the rehydrated state (including dormancy cycling) and the switch to germination.

An osmotic priming treatment of 14 days in a −1.0 MPa polyethylene glycol solution improves the germination performance of a highvigour seed lot of leek (Allium porrum L., cv. Verina). Using in vivo pulse-labelling, two-dimensional polyacrylamide gel electrophoresis and fluorography, five polypeptides were found to be synthesized in embryonic tissue at 14 days of priming that were not present at 4 days of germination without prior priming. These polypeptides were still synthesized at 6 h of germination following priming. This time point lies in the 6–12 h lag phase in protein synthesis previously observed in leek embryo tissue during germination of primed seeds where there is little increase in the rate of synthesis over that seen at the end of priming. None of the polypeptides was synthesized at 2 days of germination after priming, a period of seedling growth. The five polypeptides appear to be specifically associated with the priming period. Two additional polypeptides were found in leek embryos that were synthesized at higher levels at the end of priming than during germination alone. These continued to be synthesized, at lower levels in leek embryo tissue upon germination after priming. Several polypeptides were identified in leek endosperm tissue which were synthesized at higher levels during priming than during germination and also two polypeptides whose synthesis appeared to be specific to germination.

Some of the seed-evolved volatiles, which were mainly composed of methanol, acetaldehyde, ethanol and acetone, caused the loss of seed germinability during storage. In general, the deleterious effects of the volatiles increased with increasing RH and temperature during storage. Acetaldehyde had the strongest deleterious effect regardless of RH and temperature, while ethanol caused seed deterioration only at high RHs. Acetone was slightly deleterious to some species, while methanol was almost inert in most seeds. Various aldehydes applied during storage showed some toxicity to seed germinability, which decreased, except for 3-methylbutanal, with increasing molecular size, suggesting that the endogenous volatile compounds with an aldehyde group cause seed deterioration. On the other hand, the contents of volatile compounds in seeds were higher when they were stored at 44% RH (water sorption zone 2), than when stored at 12% RH (water sorption zone 1). Acetaldehyde, the most deleterious volatile, was more abundantly accumulated within the seeds stored at −3.5°C than at 23°C. Based on these facts, it is suggested that endogenous volatiles, especially acetaldehyde, may be an important factor that accelerates seed deterioration which often occurs under lower RHs and/or temperatures throughout long-term storage.

Wheat seed lots of different viability were obtained by an accelerated ageing technique in which seeds were hydrated up to 12.5–14.5% on a fresh weight basis and stored at 35°C. Ageing was accompanied by a progressive delay of the onset of germination and a decrease of [35S]-methionine uptake and incorporation into soluble proteins of the imbibing embryos. A detailed analysis of newly synthesized proteins by two-dimensional polyacrylamide gel electrophoresis showed that normal protein patterns during the germination process were impaired by ageing. Only two groups of polypeptides with molecular masses of 84.9–69.5 and 66.6–60.2 kDa, respectively, were actively produced while most proteins decreased or were definitely degraded. Our results suggest that the biochemical changes required for germination are gradually opposed by a deterioration process and that some newly-synthesized polypeptides can be taken as markers in assessing seed survival of ageing stress.

Legumes are crops that develop in cropping systems with relatively low inputs and are suitable to a more sustainable agriculture. Successful crop establishment, which is crucial for reliable plant production, depends on seed quality, environmental factors and genotypes. We studied pea and bean germination and seedling growth at various temperatures (5–40°C) and water potentials ( − 0.2 to − 1.5 MPa) using winter and spring pea and two common bean seeds produced in different conditions. The germination base temperature was − 1.1°C for pea seeds, and seeds of the winter genotype germinated more rapidly than those of the spring genotype. The base temperature for bean seed germination was 5.1–9.6°C, depending on the seed lot. The germination base water potential was about − 2 MPa for both species. The base temperatures for shoot elongation were higher (3–6°C) than those for germination. A review of the literature on other legumes confirmed that the differences in the responses of the legume seeds and seedlings to different temperatures were associated with their geographic origin. These results help understanding of pea and bean crop establishment, provide crop model parameter values and contribute to the search for genetic variability.

In fire-prone ecosystems, many species regenerate after fire from persistent soil seed banks. Species with physically dormant (PY) seeds have dormancy broken by fire-related heat. The magnitude of post-fire recruitment, to predict response to varying fire severity, is commonly estimated by testing dormancy-breaking temperature thresholds of fresh PY seeds. However, seeds spend years in the soil during the inter-fire period, and determining whether dormancy-breaking thresholds change over time is essential to accurately predict population persistence. Germination of four south-eastern Australian PY species from the Fabaceae family (Acacia linifolia, Aotus ericoides, Bossiaea heterophylla and Viminaria juncea) were studied. Dormancy-breaking temperature thresholds vary inter-specifically and the species represented either high or low dormancy-breaking threshold classes. Freshly collected seeds, and seeds that had been buried in the field or stored in dry laboratory conditions for 6 and 18 months were subjected to a fire-related range of heat treatments (40–100°C). Seed ageing increased germination response to heat treatments, effectively lowering the dormancy-breaking thresholds of three species. The fourth species, A. linifolia, initially had a relatively large non-dormant fraction which was lost as seeds aged, with older seeds then displaying PY broadly similar to the other study species. Patterns of threshold decay were species-specific, with the thresholds and viability of low-threshold species declining more rapidly than high-threshold species. The non-dormant fraction did not increase over time for any of our study species. Instead of increasing their non-dormant fraction, as is common in other vegetation types, these fire-prone PY species displayed a change of dormancy-breaking temperature thresholds. This is an important distinction, as maintaining dormancy during the inter-fire period is essential for population persistence. While changes in sensitivity to dormancy-breaking treatments have previously been reported as seeds age, our study provides the first test of changes to temperature thresholds, which increases the range of germination response from the seed bank under varying fire severity.
