The effect of zinc deficiency on the response of CBA mice to infection with the intestinal trematode Echinostoma caproni was examined. Young CBA mice were allocated to one of three dietary groups: a group fed a zinc deficient diet ad libitum, a control group pair fed a zinc sufficient diet and a control group fed a zinc sufficient diet ad libitum. The mice on the zinc deficient diet gained significantly less weight than the pair fed controls. In primary infections with six E. caproni metacercariae followed over a period of 128 days, zinc deficiency delayed worm expulsion. In addition, zinc deficiency resulted in a prolonged IgM response, a delayed IgG response and an increased IgA response towards the end of the experiment. Resistance to challenge infection day 21 following a primary infection with 25 E. caproni metacercariae was slightly, but not significantly, affected by zinc deficiency.

Two species of caryophyllids, L. longicollis sp. nov. and L. indicus (Moghe, 1926) were found in the fresh water cat fish, Clarias batrachus (L) (Clariidae) in India. The adult of L. longicollis is described in detail. It differs from all other species of the genus in having a receptaculum seminis.

The possibility of cryopreserving the eggs of Angiostrongylus cantonensis collected from the uterus of female worms was investigated. Eggs were cultured in NCTC 109 medium containing 50% rat serum, and various growth stages, from one-cell eggs to embryonated eggs, were used in this study.

As a cryoprotective agent, dimethylsulphoxide (Me2SO) was added to the medium at a final concentration of 1 M. Eggs suspended in 0·2 ml of the medium at 37°C were cooled to 0°C at a rate of l°C min−1, then an equal volume of 2M-Me2SO solution was added. After equilibration for 15min, the freezing procedures were started. In the freezing procedures, the effectiveness of (i) a seeding process, (ii) different cooling and warming rates and (iii) the relationship between the growth stages of the eggs and their tolerance to freezing at −20°C were investigated.

It was found the highest level of survival could be obtained with 32-cell eggs cooled at a rate of 0·3° C min−1 or more slowly with seeding at −4°C and warming at a rate of 5°C min−1. Survival was influenced more by cooling rate than by warming rate.

Using these optimum conditions, the survival of eggs was then investigated following cooling to various temperatures. While more than 50% of eggs were found to survive cooling to −30–C, extremely low survival was noted from lower temperatures.

Metacercariae of Steganoderma formosum Stafford, 1904 infected 17 of 358 Acadian hermit crabs (Pagurus acadianus) on the Scotian Shelf east of Nova Scotia, Canada, in the Northwest Atlantic Ocean in 1989, 1990 and 1995. Overall prevalence and abundance in P. acadianus on the Scotian Shelf were 5% and 0.4 metacercariae per crab, with a maximum intensity of 56. The highest prevalence (34%) and abundance (2.9) were recorded in the winter of 1989. All infected hermit crabs were greater than 9 mm in cephalothorax length. The parasite was not found in Acadian hermit crabs (n = 74) from Georges Bank, nor in any hairy hermit crabs (P. arcuatus and P. pubescens) on the Scotian Shelf (n = 808), on Georges Bank (n = 14), or in the northern Gulf of St. Lawrence (n = 87). A single sand shrimp (Crangon septemspinosa) of 1254 collected from the Scotian Shelf in 1989 and 1990 was infected with a metacercaria of S. formosum. None of 421 pink shrimps (Pandalus spp.) collected from the Scotian Shelf in 1989 and 1995 was infected. Male and female reproductive systems were well developed and spermatozoa were observed in the seminal receptacles of most parasites, suggesting that these metacercariae are progenetic. The distribution of the parasite among potential intermediate hosts suggests that S. formosum demonstrates greater specificity for its second intermediate host in the Northwest Atlantic than it does in the Pacific or in the Northeast Atlantic oceans.

A study of the life-cycle and epidemiology of Pseudanoplocephala crawfordi Baylis, 1927 is reported. Intermediate hosts are Tenebrio obscurus, T. molitor, Carpophilus dimidiatus and Tribolium castaneum. The first three species are new host records. Development in the intermediate host takes 1–1.5 months. The completion of the life-cycle takes 2–2.5 months. Surveys of intermediate hosts, pigs, humans and rats were carried out. In Rattus norvegicus, the prevalence was as high as 22%. Epidemiological factors and suggestions for control measures are discussed.

Extracts of Dirofilaria immitis adults and microfilariae cause the dcgranulation of mast cells in rat mesentery when applied in vitro and also cause increased vascular permeability in guinea-pigs and rabbits. Adult worms DUt not microfilariac are able to activate complement in vitro and both stages contain a plasminogen activator. No protcolytic activity or hacmolysins could be detected in cither stage. The possibility that these factors may be important in the pathogencsis of dirofilariasis and in the adverse drug reactions that may follow treatment of microfilaraemic animals with dicthylcarbamazine is discussed.

The host-parasite relationships of a Schistosoma haematobium isolate, originating from Niger, and the white mouse are described. Swiss OF1 albino mice were exposed individually to 200 cercariae and worms were recovered 9, 12, 16 and 20 weeks post infection. The mean worm returns ranged between 10·54 and 13·5% and did not alter significantly between 9 and 20 weeks post infection. The sex ratio of worms was always in favour of males; from 7·9:1 at 9 weeks after infection it decreased regularly to 3·28:1 at 20 weeks. Male worms reached a mean length of 8·72 mm at 20 weeks. From the 12th week post infection, a high number of eggs was found in the liver and gut. At 20 weeks, eggs were also found in the bladder. Viable eggs and infective miracidia were obtained. The infection of Bulinus truncatus from Niger succeeded with a mean rate of 61% after the first passage through mice. The isolate of S. haematobium was maintained in the laboratory during 3 successive passages through mice. These entirely new results are very probably linked to genetic characteristics peculiar to the S. haematobium populations from Niger.

The trichostrongyloid nematode Ostertagia nianqingtanggulaensis K'ung & Li, 1965, previously recorded only from sheep in Tibet, is redescribed and is shown to be a common parasite of both sheep and goats at higher altitudes in the Himalayan foothills of western Nepal. Herbage examination indicated that infective larvae can overwinter on snow covered slopes. Use of tracer animals confirmed that migratory flocks are exposed to infection when grazing on alpine pastures during the summer months. In contrast, nearby non-migratory flocks kept at lower altitudes did not harbour O. nianqingtanggulaensis and tracer animals grazed with these flocks did not become infected with this species.

Total ecdysteroid-like immunoreactive material was assayed and quantified in adults of the parasitic nematode N. brasiliensis during the intestinal phase in the rat in order to detect possible physiological fluctuations in titre. Worms of the same sex isolated from one rat were pooled in order to quantify ecdysteroids using enzyme immunoassay (EIA). The concentration of ecdysteroids fluctuated during adult life according to the sex and the age of the parasite. Important differences of levels of ecdysteroid-like compounds between the two sexes of parasites were noted particularly at 128 and 168 h post-infection. The peak at 128 h, present in female, but not in male worms, corresponds to the time of egg-laying. Following HPLC-EIA analysis, the presence of ecdysone, 20-hydroxyecdysone and apolar ecdysteroid-like material was ascertained during this peak. The physiological role and the possible origin of ecdysteroids in this nematode are discussed.

This study was carried out to compare potential methods of transplanting adult Oesophagostomum dentatum from experimentally infected donor pigs to helminth naive recipient pigs. The following methods were each tested in five pigs: A. Transfer of worms by stomach tube to the gastric ventricle of pigs per os pretreated with 0.5 mg/kg cisapride to increase gastrointestinal peristalsis; B. Transfer by stomach tube to the gastric ventricle of pigs per os pre-treated with cisapride (0.5 mg/kg) and omeprazol 20 mg which blocks hydrochloric acid secretion; C. Surgical transfer of worms to caecum of pigs. Worms for transplantation to pigs were obtained after slaughter of experimentally infected donor pigs and following isolation from the contents of the large intestine, using an agar gel migration technique. A mean of 1054 nematodes were transferred into each recipient pig within 2 hours. Procedures A and B resulted in establishment rates corresponding to only 0.5% and 7.6% of the transferred worms. In contrast, surgical transfer allowed 74.2% of the transplanted worms to be established. In all groups the transplanted worms migrated to the normal predilection site, i.e. the middle part` of the large intestine. More female than male worms established in all groups. It was concluded from this study that surgical transfer was the most reliable of the methods tested for experimental establishment of adult O. dentatum in helminth naive pigs.

1. The anatomy of Apharyngostrigea (Apharyngostrigea) simplex (Johnston, 1904) new comb, from young cattle egrets is described in detail, involving the reconstruction of its internal organisation from serial sections.

2. The relationships of the species within the genus Apharyngostrigea Ciurea, 1927 is discussed using the method of Numerical Taxonomy by Sneath and Sokal (1902). It is found that the original genus can be split into two genera. The genus Apharyngostrigea comprises two subgenera; Apharyngostrigea (Apharyngostrigea) n. subgen. and A. (Brasiliana) n. subgen. made up of seven species groups. The generic diagnosis of Apharyngostrigea Ciurea, 1927 and Chaseostrigea n. gen. are given as well as a key for separating the species of the genus Apharyngostrigea.

It is well known that free-living nematodes are of frequent occurrence in various kinds of rotting organic materials both animal and vegetable under natural conditions. In a recent investigation of the nematodes occurring in a variety of substances such as pig-manure, sheep droppings taken from pastures, soil from a chicken run, decaying leaves from a drain, etc., the writer has made use of the Baermann method (Cort and others 1922) with a good deal of success. Amongst several interesting forms obtained from samples taken from a heap of pigmanure were six worms, four females and two males, belonging to the genus Aphelenchus, which at once attracted attention by reason of certain peculiarities in their appearance. They closely resemble Aphelenchus tenuicaudatus de Man (1895) in certain anatomical features, but differ on certain points, and the writer regards them as belonging to a hitherto undescribed species.

Examination of rajid skates off the Algerian coast in the Mediterranean Sea revealed that three of the 33 Dipturus batis Linnaeus, 1758 examined harboured a new tapeworm species: Echeneibothrium algeriensis n. sp. This new species, collected from the anterior half of the spiral valves, is described on the basis of morphological data from light and scanning electron microscopy. The new species differs from previously described Echeneibothrium species by details of the scolex and loculi, total length, the length of the myzorhynchus, the number of proglottides, and the number of testes. Comparison of the diets of the ten skate species common in the Mediterranean basin indicates some varying degree of overlap, suggesting that host specificity in this host–parasite system is determined by other host and/or ecological variables such as adaptations of the parasites to their respective hosts, either on the morpho-anatomical level, in physiological characteristics of the parasite's habitat, in the trophic requirements for the successful transmission of the parasite, or in adaptations to the behavioural characteristics of the host. Furthermore, restricted overlap of E. algeriensis n. sp. with congeners in parasite assemblages of D. batis indicates some structuring according to attachment-site preferences. However, attachment-site preferences are not explained solely by morphological compatibility between bothridia and villi. This study reiterates the need to examine multiple factors synergistically in studies on host specificity of parasites, and the need to examine the parasite fauna of hosts across their entire geographical range in order to truly appreciate the biodiversity they harbour.

Although the “potato-sickness” associated with the nematode Heterodera schachtii has been the subject of a good deal of investigation in Britain during the past twelve years, the only practical method of control so far devised is the adoption of a well-planned crop rotation in which potatoes are omitted for several years. The limitation of a general application of this method to the thousands of acres of land infected with this eelworm disease but otherwise admirably suited for the growing of potatoes has led to insistent demands for further study into chemical treatment of affected soil. In the present investigation, comparative control trials of soil treatment by certain chemical substances were carried out on land known to be thoroughly “potato-sick” and the last potato crop grown in it had been a failure. Most of the experimental treatments had already shown in other field experiments some measure of protection from “potato-sickness,” at least, judging by the healthier appearance of the foliage and increase in yield of crop. Although the results of previous experience indicated that there was a positive association between application of these various soil dressings and intensity of disease, it was felt that a more accurate computation of the latter, by measuring the eelworm cyst content of the soil, was desirable before any definite conclusion might be drawn.

Procyrnea choique n. sp. is described from the lesser rhea, Rhea pennata d´Orbigny (Aves: Rheidae), from the Protected Natural Area Península Valdés, Chubut province, Argentina. The new species resembles P. ficheuri, P. murrayi, P. excisiformis, P. dollfusi, P. haliasturi, P. anterovulvata, P. graculae, P. brevicaudata, P. uncinipenis, P. javaensis, P. ameerae, P. ornata, P. aegotheles, P. spiralis, P. ruschii and P. aptera, mainly in the absence of lateral alae; but differs from its congeners by having a left spicule without barbs, the position of the vulva which is post-equatorial, the absence of lateral ridges, absent median precloacal papilla, asymmetrical caudal alae, males more than 6 mm long, left spicule 1 mm long and spicule ratio 1:3. Our results extend the taxonomy of Procyrnea Chabaud, 1958 and comprise the first report of a habronematid from R. pennata. A key to species of Procyrnea is presented.

Calves and lambs heavily infected with Strongijloides papillosus develop cardiac arrest by ventricular fibrillation which is preceded by continuous sinus tachycardia and prolongation of the PQ interval during the patent infection. In the present study, cardiac rhythms following anthelmintic treatment were investigated in lambs infected with S. papillosus to ascertain whether cardiac disorders due to the infection are based on a reversible and curable change. Eight lambs were given a lethal dose of S. papillosus. Five of the animals were injected with ivermectin when they developed continuous sinus tachycardia and prolongation of the PQ interval. The other three animals served as untreated controls. In the treated animals, elevated heart rates and PQ intervals began to decrease between 10 and 21 h, then continuous sinus tachycardia and prolongation of PQ interval disappeared within 39 h of treatment. No arrhythmias were detected after the disappearance of sinus tachycardia. Faecal egg counts became negative within 61 h of treatment. Only a few worms were collected from the small intestine at necropsy. The control animals developed ventricular fibrillation by 349 h after infection, having high faecal egg counts and intestinal worm burdens. These results indicate that cardiac disorders generated by S. papillosus infection are reversible and curable following worm elimination.

An attempt was made to identify the filarial specific antigens that are capable of inducing immune response in human filariasis. Lymphocytes were taken from three clinically defined groups living in an endemic area in Madras, namely microfilaraemic (MF) subjects with microfilariae in their blood smear without any clinical symptoms, chronic pathology (CP) individuals with lymphangitis or lymphadenitis in combination with a history of recurrent filarial fevers or lymphoedema, and endemic normals (EN) subjects without microfilariae nor any clinical symptoms of pathology. Lymphocytes from the three groups responded with no significant difference (P = 0.21) in their proliferative index to PPD and PHA, although lymphocytes from MF individuals showed significantly (P < 0.001) less proliferative index to Brugia malayi antigen (BMA) than the CP and EN subjects. This antigen specific cellular unresponsiveness seen in MF patients was not reversed by the addition of recombinant IL-lα, IL-1β, and IFN-γ, but the addition of sera from EN individuals seemed to restore this unresponsiveness (P < 0.001). The peripheral blood mononuclear cells from MF patients secreted more IL-1 in response to BMA induction than the same from CP and EN individuals. A 58 kDa recombinant protein isolated from a Wuchereria bancrofti genomic library (58 kDa) had mounted a higher proliferative response to lymphocytes from all three groups compared to BMA (P < 0.001) indicating the possible use of recombinant filarial protein to mount immunological responses in filarial patients.

Fifty day-old chicks were each infected with 10 excysted metaccreariae of Philophthalimus nocturnus Looss. 1907 around each orbit and growth, development and allometry were studied. The growth rate showed two phases over a period of 35 days, a limited lag phase lasting two days post-infection in which flukes did not exceed 440 μm in length, and a rapid phase during which growth was rapid and flukes reached a size of 3·008–3·504 mm on day 35. Five developmental stages were noticed during the course of development of the metacercaria to the egg-producing adult stage. Eggs appeared in the uterus on day 14 and oculate miracidia on day 25. The hindhody, testes and ovary showed positive allometric growth, the pharnyx less so, whereas negative allometric growth was shown by the forebody. Body width, oral sucker and ventral sucker were close to isometry, growing at the same rate as the body length.

During the course of experimental work on the factors governing the escape of larvae from the cysts of Heterodera schachtii, a preliminary series of experiments was carried out to determine whether the admixture of artificial fertilisers with infected soil modified in any way the rate of hatching of the larvae. It was thought possible, firstly, that some form of fertiliser might have a lethal effect on the cyst contents or on the larvae during their passage through the soil before they succeeded in entering the roots of the host; secondly, that the fertiliser or its break-down products in soil might neutralise or retard the action of potato-root excretion, which normally stimulates the larvae to escape from the cysts and attracts them towards the roots of the plant; or thirdly, that the larvae might be stimulated to hatch to some extent even in the absence of the host-plant.

The cercarial emergence of Schistosoma japonicum from Oncomelania quadrasi was observed under outdoor conditions in Leyte, Philippines. Nocturnal periodicity of the emergence within 24 hours was recognized. Peak emergence usually occurs between 18:00 and 20:00 and occasionally between 8:00 and 10:00. The peaks were usually synchronized with a sudden lowering of water temperature and light intensity and occasionally with their increase. As the nocturnal periodicity of the emergence was observed in the dark and outdoors, it was suggested that emergence might be dependent on temperature, which is secondarily influenced by photoperiodicity and light intensity.
