The structure and characteristics of a model suitable for estimating digestion within the rumen and rates and patterns of nutrient entry in lactating cows are presented. The model consists of 12 state variables comprising a large particle pool, small particle pools representing insoluble dietary nutrients, soluble pools representing soluble dietary nutrients, and fermentation intermediates and end products. The model was constructed assuming continuous feeding, using Michaelis–Menten or mass action kinetics. The computer program was written in ACSL to run on a VAX computer. A fourth-order Runge–Kutta procedure was used for numerical integration.

Sensitivity and behavioural analysis demonstrated that overall stability and sensitivity of the model to individual parameters was generally satisfactory, but the need to improve the description and parameterization of aspects such as particle size in relation to availability, rate and affinity constants for amino acid degradation and rate constants for particle outflow from the rumen was established. Adjustments of the model to examine discontinuous feeding regimes were undertaken and initial results with respect to changes in fermentation rates, rumen acetate levels and microbial metabolism were considered realistic.

Comparisons with experimental data were considered satisfactory on forage-based and medium concentrate-containing diets, but with diets comprising 90% cereal, some inconsistencies, especially with respect to predictions of volatile fatty acid production rates, were observed. Reasons for this are put forward and suggestions for improvements in the model are discussed.

Yogurt starter B3 was grown in cows' milk, goats' milk I (obtained 2–3 weeks after parturition) and goats' milk II (8 months after parturition), and a number of biochemical and bacteriological parameters were recorded at intervals during incubation for 8 h at 43°C. No appreciable differences between the milks were observed for total viable count, bacteriological ratio of rods to cocci, or decrease in pH. Production of acetaldehyde, however, was considerably lower in goats' milk I (6·2 ppm) and goats' milk II (8·9 ppm) than in cows' milk (18 ppm). Formation of diacetyl and acetoin showed similar patterns in all three milks. The highest production of α-acetolactic acid was observed in goats' milk II, apparently coincident with the highest production of CO2 in this milk.

A nutrient agar medium containing 0·1 % of a low melting point fraction of butterfat was shown to be suitable for detection, enumeration and isolation of lipolytic bacteria from milk. Bacterial growth was not inhibited by the butterfat and lipolytic reactions were clearly visible and easily interpreted. Lipolytic counts on the butterfat agar compared favourably with lipolytic counts obtained with other commonly used media.

TWO models of lactating dairy cows were constructed by combining mechanistic models of digestion and metabolism reported previously and by adding elements to allow simulation of changes in tissue metabolic capacities over time. One model (day version) had an integration interval of 0·005 d and was suitable for simulation of within-day dynamics of nutrient supply and partition. The other (lactation version) had an integration interval of 1 d and was suitable for simulation of full lactation cycles. A number of simulation analyses were conducted to characterize and evaluate the models, to examine quantitative and dynamic properties of mechanisms which influence partition of nutrients, to identify aspects requiring further study, and to illustrate the potential usefulness of mechanistic, as compared to empirical, models in analyses of energy balance in lactating dairy cows.

Creams containing 25–45% fat were made from whole milks which had been concentrated 2- to 5-fold by ultrafiltration. Each series of creams (from any one concentrate) was standardized with its own skimmed milk, giving creams with higher than normal protein contents. These were then pasteurized and cooled for at least 24 h before being whipped. Increasing the fat content of creams gave increased stiffness, an overrun (volume increase) that passed through a maximum, decreased serum leakage and shorter whipping time. Increasing the protein content gave increased stiffness, less serum leakage, reduced overrun and a whipping time that passed through a maximum. Low fat creams, and also creams containing high amounts of fat and protein, did not produce a satisfactory whipped product. However, 35% fat creams from 2-fold concentrated milks compared well with ordinary 38% whipping creams. By lowering the fat content of creams an increase in yield was obtained.

When a chromogenic Limulus Amoebocyte Lysate (LAL) assay and a tube gelation LAL assay were compared for the detection of Gram-negative bacteria, using a strain of Pseudomonas putida, the detection level (∼ 103 cfu/ml) and cost of the assays were approximately the same for both assays but the reading was more precise for the chromogenic substrate assay. A modified chromogenic assay was devised for detection of Ps. putida in milk.

Cultures of Pseudomonas P46 grown in whole milk to contain ∼ 2 × 107 or 1 × 108 viable cells ml−1 before ultra high temperature (UHT) treatment (140°C for 5 s) demonstrated near linear increases in the concentration of short-chain free fatty acids (FFA) during storage at 20°C. However with 5 × 106 cells ml−1 before UHT heat treatment there was no detectable increase in these FFA levels over a 6-month storage period. A novel heat treatment (140°C for 5 s followed by 60°C for 5 min) reduced the rate of production of volatile FFA to < 10% of the rates achieved after the normal UHT treatment.

Oggtt is a dried fermented milk product made and marketed primitively under uncontrolled conditions in the Arabian peninsula. By applying controlled conditions for production, the keeping quality of oggtt was greatly improved. The two types (cooked and salted) were produced from fresh pasteurized goats' or cows' milk or from skim milk powder. Oggtts flavoured with chocolate, date, mint or different fruits were also produced. Chemical composition and organoleptic properties of the final products were evaluated. Taste panel results indicated that the plain products, including those made from reconstituted milk, were acceptable, but flavoured oggtts, with the exception of date flavour, were not popular.

When Havarti cheeses were made from milk concentrated 5-fold using ultrafiltration (UF) and with a direct vat set type starter containing Streptococcus lactis, Str. cremoris, Leuconostoc cremoris and Str. lactis subsp. diacetilactis (i.e. a BD-type starter) their colour, texture, body, and flavour were equal to those of traditionally produced Havarti. The somewhat slower flavour development was associated with a retarded breakdown of protein. Although αs1-casein was almost completely broken down in Havarti cheese made with the ultrafiltration method (UF Havarti) and traditional cheeses after 120 d of storage, degradation of β-casein was retarded in UF Havarti. Rennet contents and numbers of starter organisms were similar in the two types of cheese and thus could not explain the difference in ripening rate. Str. lactis subsp. diacetilactis was dominant in both UF and traditional Havarti cheeses when the BD-type starter was used. By varying the composition of the starter, large variations in the bacterial composition and citrate fermentation in UF Havarti cheeses were obtained. The BD-type starter gave the best organoleptic quality.

Pseudomonas fluorescens strains 240 and 32A expressed cell-associated peptidase activity which was shown by subcellular fractionation to be primarily intracellular. Two peptidases were partly purified from strain 32A. One specifically hydrolysed N-α-benzoyl-DL-arginine-4-nitroanilide and was termed endopeptidase and the other hydrolysed L-lysine- and L-leucine-4-nitroanilide and was termed aminopeptidase. The endopeptidase had very low activity on bovine serum albumin compared with that of trypsin and probably was not a proteinase. The endopeptidase had a mol. wt of 33000 and a pH optimum of 8·0. The enzyme was stimulated by Ca2+ and Mg2+ and inhibited by Co2+, Mn2+, Hg2+, Zn2+ and leupeptin. Soya bean trypsin inhibitor and phenylmethane sulphonyl fluoride (PMSF) had no effect on its activity. The aminopeptidase had a mol. wt of 44000 and a pH optimum of 8·0. It was inhibited by all the metal ions mentioned above and by PMSF. Little proteolysis was found when ultra high temperature (UHT) sterilized milk was treated with cell-free extract from strain 32A. It was concluded that the cell-associated peptidases from Pseudomonas strains normally present in raw milk may not contribute significantly to the deterioration of UHT sterilized milk.

The clarification of milk by addition of solutions of Triton X-100 and EDTA after digestion of added Hide Powder Azure (HPA) was found to provide a simple method of determining the extracellular proteinase activity of Gram-negative psychrotrophic bacteria in pasteurized whole milk. The light absorbance of the clarified HPA digestion product was measured directly, after a brief incubation period, and was stable to storage of samples in diffuse daylight for at least 2 d. Proteinase produced by growth in refrigerated whole milk of as few as 2·5 × 106 cfu ml−1 of Pseudomonas fluorescens AR11 was detected.

Milks whose compositions had been altered by deliberate manipulation or by contact with various environmental conditions relevant to cheesemaking were treated with rennet in such a way as normally to give a constant coagulation time. Rates of curd formation and whey loss, curd structure and fat retention were determined. Ca depletion in concentrated milks reduced firming and syneresis rates and gave curds with finer protein networks. Increased temperature of curd formation increased the curd-firming rate and curd coarseness, but decreased the syneresis rate at 30 °C. Prior treatment of concentrated milk with rennet in the cold gave a much finer protein network which retained fat better than curd formed normally. Despite increasing firming and syneresis rates, acidified milk gave a slightly finer curd with a better fat retention than normal. Addition of cationic materials stimulated aggregation and the curd retained fat better than normal, although the curd structure was unaffected. The results indicate that the processes of firming and syneresis have related mechanisms, and that the curd structure is not simply dependent on curd-forming conditions, but on the number of aggregating particles and the forces between them.
