Blastocystis sp. is a prevalent protistan parasite found globally in the gastrointestinal tract of humans and various animals. This review aims to elucidate the advancements in research on axenic isolation techniques for Blastocystis sp. and their diverse applications. Axenic isolation, involving the culture and isolation of Blastocystis sp. free from any other organisms, necessitates the application of specific media and a series of axenic treatment methods. These methods encompass antibiotic treatment, monoclonal culture, differential centrifugation, density gradient separation, micromanipulation and the combined use of culture media. Critical factors influencing axenic isolation effectiveness include medium composition, culture temperature, medium characteristics, antibiotic type and dosage and the subtype (ST) of Blastocystis sp. Applications of axenic isolation encompass exploring pathogenicity, karyotype and ST analysis, immunoassay, characterization of surface chemical structure and lipid composition and understanding drug treatment effects. This review serves as a valuable reference for clinicians and scientists in selecting appropriate axenic isolation methods.

This paper examines various ways in which apocalyptic studies can benefit from the introduction of the term and concept of gilayon, a reconstructed Hebrew counterpart of the Judeo-Greek apocalypse. The term gilayon, which combines the meanings of “revealed book” and “book of revelation,” refers to a central image of early Jewish revealed literature and could serve to define an important corpus, the boundaries of which might well overlap with (but still differ from) what is understood by the “genre apocalypse” in modern research. Moreover, this reconstructed concept uncovers additional meanings and associations, which shed light on texts known as “apocalyptic,” and has explanatory power for many phenomena associated with them. The introduction of gilayon may modify the entire paradigm of our understanding of early Jewish mysticism and help to divert the discussion of textual genres associated with it from a phenomenological to a historical route.

In this article, I advance a recent epigraphic approach to historical study by foregrounding steles as a medium that functions both to communicate information and project authority publicly. Scholars taking this approach have explored distinctive genres of steles to transform our understanding of north China under Mongol rule. Through a case study, I show how a set of steles installed in the fifteenth-century rural world of north China transmitted authority and power not just through the content of their inscriptions but also through other written and unwritten information they stored. I give particular attention to the ways in which the inscriptions were materialized and visualized. In doing so, I argue that emphasizing the public communication function of steles challenges us to think beyond primary sources strictly in terms of their textual value to reflect more broadly on modes of transmission and the power dynamics contained within them.

To improve the efficiency of isolating and culturing lichen mycobionts, we performed a growth assay on an Australian strain of the soil-crust lichenized fungus Endocarpon pusillum Hedw. This assay determined the preferred nitrogen and carbon sources of the fungus by limiting the available nitrogen or carbon sources to single compounds found in soils, plants and lichen thalli. We found that the non-proteinaceous amino acid, GABA, produced the most growth of all nutrients when provided as the sole nitrogen source but was a poor carbon source. Fructose, glucose, cellobiose and sorbitol produced the most growth of all the carbon sources tested. Ammonium, nitrate and polyamines were poor nutrient sources. These findings correspond with reports of primary metabolite pools in other lichen species and may guide future studies involving growth of recalcitrant lichen mycobionts.

Fish consumption is associated with reduced risk of CVD, which may be partly mediated by alterations in plasma lipids, such as HDL-cholesterol. However, comprehensive analyses of associations between fatty fish consumption and lipoprotein subclass profile are limited and show inconsistent results. Therefore, the aim of the present exploratory study was to investigate the association between fatty fish consumption and lipoprotein subclass particle concentrations and composition, with an emphasis on HDL. We performed a comprehensive plasma metabolite profiling in 517 healthy adults, using a targeted high-throughput NMR spectroscopy platform. The participants were divided into tertiles based on consumption of fatty fish, reported through a validated FFQ. We compared the concentration of metabolites between the participants in the lowest and highest tertiles of fatty fish consumption. We show that high consumers of fatty fish (>223 g/week, median intake 294 g/week) had higher particle concentrations and content of total lipids, free cholesterol and phospholipids in large and extra-large HDL particles and higher content of total cholesterol, cholesteryl esters and TAG in large HDL particles than low consumers (<107 g/week, median intake 58 g/week). Using fatty fish consumption as a continuous variable, we found that fatty fish consumption was associated with lower levels of the inflammation marker glycoprotein acetyls. In conclusion, high consumers of fatty fish seem to have a more favourable HDL-cholesterol-related lipoprotein profile and anti-inflammatory phenotype than low consumers of fatty fish. Thus, these data support the current Norwegian dietary recommendations for fish consumption regarding CVD risk.

Men have earlier first-time event of CHD and higher postprandial TAG response compared with women. The aim of this exploratory sub-study was to investigate if intake of meals with the same amount of fat from different dairy products affects postprandial lipoprotein subclasses differently in healthy women and men. A total of thirty-three women and fourteen men were recruited to a randomised controlled cross-over study with four dairy meals consisting of butter, cheese, whipped cream or sour cream, corresponding to 45 g of fat (approximately 60 energy percent). Blood samples were taken at 0, 2, 4 and 6 h postprandially. Lipoprotein subclasses were measured using NMR and analysed using a linear mixed model. Sex had a significant impact on the response in M-VLDL (P=0·04), S-LDL (P=0·05), XL-HDL (P=0·009) and L-HDL (P=0·001) particle concentration (P), with women having an overall smaller increase in M-VLDL-P, a larger decrease in S-LDL-P and a larger increase in XL- and L-HDL-P compared with men, independent of meal. Men showed a decrease in XS-VLDL-P compared with women after intake of sour cream (P<0·01). In men only, XS-VLDL-P decreased after intake of sour cream compared with all other meals (v. butter: P=0·001; v. cheese: P=0·04; v. whipped cream: P=0·006). Meals with the same amount of fat from different dairy products induce different postprandial effects on lipoprotein subclass concentrations in men and women.

The present experiment examined how the interaction between senders’ communicative competence, veracity and the medium through which judgments were made affected observers’ accuracy. Stimuli were obtained from a previous study. Observers (N = 220) judged the truthfulness of statements provided by a good truth teller, a good liar, a bad truth teller, and a bad liar presented either via an audio-only, video-only, audio-video, or transcript format. Log-linear analyses showed that the data were best explained via the saturated model, therefore indicating that all the four variables interacted, G2(0) = 0, p = 1, Q2 = 1. Follow-up analyses showed that the good liar and bad liar were best evaluated via the transcript (z = 2.5) and the audio-only medium (z = 3.9), respectively. Both the good truth teller and the bad truth teller were best assessed through the audio-video medium (z = 2.1, good truth teller, z = 3.4, bad truth teller). Results indicated that all the factors interacted and played a joint role on observers’ accuracy. Difficulties and suggestions for choosing the right medium are presented.

In a criticism of their appeals to ancient media this essay works out the significance of cultural practices associated with orality, writing, and memory for further work on the synoptic problem, especially as regards the uneven patterns of variation and agreement in the tradition.

Dynamics of structural transformation of supported heteropoly compound was investigated by IR-spectroscopy, XRD, TPR, and TPO methods. The 15-30 wt.% H3PMo12O40 and H3PV2Mo10O40, supported on SiO2 catalysts were investigated in process under various conditions by interaction with O2 (air) in the presence of water vapor, H2, alkane in TPR regime, O2 in TPO regime (298-1273K) and H2O (298, 923K). The rank of reversible structural conversion of supported heteropoly compounds was established under influence of temperatureand medium. These new findings of reversible structural cycle explains the stability of the 12th series Mo heteropoly compounds supported on Si-containing carrier at high temperatures (873-1073K) which is close to the actual operation temperatures of the partial oxidation reactions and oxidative dehydrogenation of alkanes.

N6 medium, Murashige and Skoog (MS) medium and BNBN medium were adopted in tissue cultures of Nipponbare (japonica) scutellum for performance comparison. Results indicated that, among the three kinds of culture media, the NB medium worked best in callus induction and gene transformation and was most suitable for tissue culture of Nipponbare scutellum. Based on this, the elicitor-encoding gene pemG1 from Magnaporthe grisea was introduced into the genome of Nipponbare via the Agrobacterium-mediated method. Transgenic rice plants were obtained. The integration, transcription and expression of pemG1 in rice were confirmed by polymerase chain reaction (PCR), Northern blot and Western blot, respectively. Genetic analysis showed that the transgenes were segregated normally in the progenies.

Oviductal oocytes (n = 795) were obtained from ovulation-induced prepubertal gilts. In the first experiment, different parameters related to in vitro fertilisation (IVF) were compared in the presence and absence of cumulus matrix (which is shed with the oocytes at ovulation.) The results show that the presence of this matrix is beneficial because the rates of fertilisation (69%) (number of monospermic oocytes/total number of healthy mature oocytes; 42%), and the median number of spermatozoa per oocyte (1.52±0.06), were improved with respect to those obtained in its absence (54%, 22% and 2.33 ± 0.08, respectively; p<0.01). In the second experiment the effect of two different volumes of co-incubation medium (2 ml and 0.4 ml) on the same parameters of porcine IVF were compared. No significant differences between volumes were observed, except in the mean number of spermtozoa per oocyte and the percentage of dispermic oocytes.
