The causal chain of parasite–host–environment interactions, the so-called ‘dual parasite environment’, makes studying parasites more complicated than other wild organisms. A sample of 65 282 fleas taken from 336 different locations were analysed for changes in the distribution, diversity and compensation of flea communities found on small mammals along an elevational diversity gradient ranging from the Pannonian Plain to the base of the Carpathian summits. The fleas were divided into four groups, which were derived from changes in abundance and occurrence determined from cluster analysis. They are (1) flea species whose range seems unrelated to any change in elevation; (2) species that avoid high altitudes; (3) a group that can be subdivided into two types: (i) host-specific fleas and (ii) mountains species and (4) species opting for high altitudes on the gradient or preferring lower to middle elevations below 1000 m. Our study showed a unimodal pattern of flea diversity along the elevational gradient. It indicated that seasonality significantly conditions changes in biodiversity and patterns of spatial change along the elevational gradient, with specific flea species influenced by their host, while the impact of environmental conditions is more pronounced in opportunistic flea species.

The family Ctenophthalmidae (Order Siphonaptera) has been considered as a ‘catchall’ for a wide range of divergent taxa showing a paraphyletic origin. In turn, Ctenophthalmus sp. (Ctenophthalmidae) includes 300 valid described taxa. Within this genus, males are easily distinguishable basing on the size, shape, and chaetotaxy of their genitalia; however, females show slight morphological differences with each other. The main objective of this work was to carry out a comparative morphometric, phylogenetic, and molecular study of two different subspecies: Ctenophthalmus baeticus boisseauorum and Ctenophthalmus apertus allani in order to clarify and discuss its taxonomic status. From a morphological and biometrical point of view, we found clear differences between modified abdominal segments of males of both subspecies and slight differences in the margin of sternum VII of all female specimens which did not correspond with molecular and phylogenetic results based on four different molecular markers (Internal Transcribed Spacer 1 and 2 of ribosomal DNA, and the partial cytochrome c oxidase subunit 1 and cytochrome b of mitochondrial DNA). Thus, we observed a phenotypic plasticity between both subspecies, which did not correspond with a real genotypic variability nor different environmental or ecological conditions. Basing on these results, we could consider that there are no solid arguments to consider these two ‘morphosubspecies’ as two different taxa. We propose that C. b. boisseauorum should be considered as a junior synonym of C. a. allani.

Fleas have great medical relevance as vectors of the causative agents of several diseases in animals and humans and rodents are the principal reservoirs for these pathogens. Argentinian Patagonia has the highest diversity of rodent fleas in South America. However, parasitism rates of rodents by fleas, the factors that influence them and the ecological aspects that modulate geographical distributions of flea–host association remain unknown for this region. This is the first study to record the diversity, prevalence, abundance, geographical distributions and host ranges of fleas in Argentinian Patagonia. It also compares parasitism rates among Patagonian ecoregions and host species. We captured 438 rodents belonging to 13 species, which harboured 624 fleas from 11 species and subspecies (P = 46%; mean abundance = 1.44). The high parasitism rates obtained were consistent with previous records for other arid regions, suggesting that Patagonia favours the survival and development of Siphonaptera. Host geographic range and abundance were related to the parasitological indexes: host species with high-density populations had the highest mean flea abundance and prevalence, whereas widely distributed hosts had the highest richness and diversity of flea species. Our results contribute to the knowledge of the flea–host–environment complex. Our analysis of flea distributions and parasitism rate in Central Patagonia may be useful in epidemiological studies of flea-borne diseases and provide a basis for implementing surveillance systems for better risk assessment of emerging zoonoses in the region.

In the present work, we carried out a comparative molecular study of Stenoponia tripectinata tripectinata isolated from Mus musculus from the Canary Islands, Spain. The Internal Transcribed Spacers 1 and 2 (ITS1, ITS2) and 18S ribosomal RNA partial gene and cytochrome c-oxidase 1 (cox1) mitochondrial DNA partial gene sequences of this subspecies were determined to clarify the taxonomic status of this subspecies and to assess inter-population variation and inter-specific sequence differences. In addition, we have carried out a comparative phylogenetic study with other species of fleas using Bayesian, Maximum Parsimony, Maximum Likelihood and Neighbor-Joining analysis. A geographical signal was detected between the cox1 partial gene sequences of S. t. tripectinata isolated from M. musculus from different islands and those isolated from Apodemus sylvaticus from the Iberian Peninsula. Our results assess the monophyletic origin of Stenoponiinae and a different genetic lineage from Ctenophthalmidae. Thus, the elevation of subfamily Stenoponiinae to family level (Stenoponiidae) is suggested.

Oropsylla hirsuta is the primary flea of the black-tailed prairie dog and is a vector of the plague bacterium, Yersinia pestis. We examined the population genetic structure of O. hirsuta fleas collected from 11 prairie dog colonies, 7 of which had experienced a plague-associated die-off in 1994. In a sample of 332 O. hirsuta collected from 226 host individuals, we detected 24 unique haplotype sequences in a 480 nucleotide segment of the cytochrome oxidase II gene. We found significant overall population structure but we did not detect a signal of isolation by distance, suggesting that O. hirsuta may be able to disperse relatively quickly at the scale of this study. All 7 colonies that were recently decimated by plague showed signs of recent population expansion, whereas 3 of the 4 plague-negative colonies showed haplotype patterns consistent with stable populations. These results suggest that O. hirsuta populations are affected by plague-induced prairie dog die-offs and that flea dispersal among prairie dog colonies may not be dependent exclusively on dispersal of prairie dogs. Re-colonization following plague events from plague-free refugia may allow for rapid flea population expansion following plague epizootics.

To investigate the prevalence of a flea-borne protozoan (Trypanosoma (Herpetosoma) microti) in its field vole (Microtus agrestis) host, we monitored over a 2-year period a range of intrinsic and extrinsic parameters pertaining to host demographics, infection status and vector (flea) prevalence. Generalized Linear Mixed Modelling was used to analyse patterns of both flea and trypanosome occurrence. Overall, males of all sizes and ages were more likely to be infested with fleas than their female counterparts. Flea prevalence also showed direct density dependence during the winter, but patterns of density dependence varied amongst body mass (age) classes during the summer. Trypanosome prevalence did not vary between the sexes but was positively related to past flea prevalence with a lag of 3 months, with the highest levels occurring during the autumn season. A convex age-prevalence distribution was observed, suggesting that individuals develop a degree of immunity to trypanosome infection with age and exposure. An interaction between age and whether the individual was new or recaptured suggested that infected animals are less likely to become territory holders than their uninfected counterparts.

The potential of biological invasions to threaten native ecosystems is well recognized. Here we describe how an introduced species impacts on native host-parasite dynamics by acting as an alternative host. By sampling sites across an invasion front in Ireland, we quantified the influence of the introduced bank vole (Clethrionomys glareolus) on the epidemiology of infections caused by flea-transmitted haemoparasites of the genus Bartonella in native wood mice (Apodemus sylvaticus). Bartonella infections were detected on either side of the front but occurred exclusively in wood mice, despite being highly prevalent in both rodent species elsewhere in Europe. Bank vole introduction has, however, affected the wood mouse-Bartonella interaction, with the infection prevalence of both Bartonella birtlesii and Bartonella taylorii declining significantly with increasing bank vole density. Whilst flea prevalence in wood mice increases with wood mouse density in areas without bank voles, no such relationship is detected in invaded areas. The results are consistent with the dilution effect hypothesis. This predicts that for vector-transmitted parasites, the presence of less competent host species may reduce infection prevalence in the principal host. In addition we found a negative relationship between B. birtlesii and B. taylorii prevalences, indicating that these two microparasites may compete within hosts.

Jump performance was studied in seven flea species (Xenopsylla conformis mycerini, Xenopsylla ramesis, Xenopsylla dipodilli, Parapulex chephrenis, Synosternus cleopatrae pyramidis, Nosopsyllus iranus theodori and Stenoponia tripectinata medialis). Jump length was compared among individuals, between sexes and among species, and an examination made of whether: (1) there is a consistent difference in jump performance between male and female fleas; (2) sexual dimorphism in body size affects jump performance; (3) there is a size-specific difference in jump performance among species; (4) there are morphological correlates of jumping ability among individuals, between sexes and among species. Males were smaller than females in all species and size dimorphism did not increase with increasing body size. In all species, except S. t. medialis, both absolute and size-specific jumping abilities of the males were less than those of the females. Males and females of S. t. medialis demonstrated similar jumping ability despite their body size difference. The best jumpers were S. c. pyramidis, N. i. theodori and S. t. medialis, whereas X. dipodilli as well as female X. c. mycerini and X. ramesis demonstrated the poorest ability to jump. Female:male performance ratio differed among species, being the highest in X. c. mycerini and X. ramesis, and the lowest in S. t. medialis. The proportion of the overall dimorphism in jumping ability that was owing to size differences between males and females was low, ranging from 0.10 to 0.34. No correlation between jumping ability and pleural height or length of hind coxa, femur and tibia was found in any species, except in X. dipodilli. Interspecific comparisons using size-corrected and phylogeny-controlled values did not show any relationship between jumping ability and any morphological trait. Fleas occupying habitats with a soft sandy substrate were better jumpers than those living in habitats with a hard stony substrate.

The strongest evidence for host specificity of mammalian trypanosomes comes from parasites of the subgenus Trypanosoma (Herpetosoma). Laboratory studies have shown that T. (Herpetosoma) species will not infect an alternative host. However, this has not been demonstrated in wild populations. We screened 560 bank voles (Clethrionomys glareolus) and 148 wood mice (Apodemus sylvaticus) for trypanosomes by PCR amplification of the 18S rRNA gene. In total, 109 (19%) bank voles and 12 (8%) wood mice were infected. A HaeIII restriction site was discovered that could be used to discriminate between T. (H.) evotomys of the bank vole and T. (H.) grosi of the wood mouse. All the parasites in the bank voles were identified as T. (Herpetosoma) evotomys by RFLP-PCR. Out of the 12 wood mouse infections 10 were due to T. grosi. Two of the wood mice were infected with parasites with a novel genotype that was most similar to those of T. evotomys and T. microti of voles. Fifty-six fleas collected from the rodents were also screened for trypanosomes; 9 were infected with T. evotomys and 1 with T. grosi. One of the fleas infected with T. evotomys was collected from a wood mouse.
