The protective potential of killed Leishmania major (ALM) along with BCG was evaluated against L. donovani in Indian langur monkeys in single and triple dose schedules. A delayed protection was observed in monkeys after a single dose schedule of ALM (3 mg)+BCG (3 mg) given intradermally 2 months before intravenous challenge with L. donovani. Triple dose schedule each of 1 mg ALM+1 mg BCG was more effective. The status remained unchanged until the end of the experiment (approximately 8 months). The study indicates that a combination of ALM+BCG may be a good candidate vaccine for exploiting against human Kala-azar.

Three types of human leishmaniases are known to occur in Kenya: visceral leishmaniasis, caused by Leishmania donovani, and cutaneous leishmaniasis caused by L. aethiopica and L. major. Visceral leishmaniasis is the most important of the three in eastern Africa, not only because it is fatal but also because several epidemics have occurred periodically since the disease appeared in the country about 40 years ago resulting in many deaths. L. aethiopica and L. major are chiefly zoonotic and people contract the disease when they wander into areas containing animal reservoirs and infected vectors. Control measures were undertaken against vectors and reservoirs during an epidemic outbreak of kala-azar in Machakos District.

The vectors of Leishmania major were investigated in Marigat, Baringo District, Kenya, a focus of both visceral and dermal leishmaniases. These studies resulted in the discovery of the resting sites of Phlebotomus duboscqi, a known vector of L. major, and of alternate principal resting sites of Phlebotomus martini, a known vector of visceral leishmaniasis. Furthermore, Sergentomyia ingrami was incriminated as a possible vector of L. major among susceptible rodents inside burrows. Trapping methods which have resulted in the discovery of the resting sites of these phlebotomine sandflies are presented in this paper.

Various species of sandflies were collected from outdoor natural resting sites from three districts in Kenya which are endemic for kala-azar (Machakos, West Pokot and Baringo). Their blood-meals were analysed to find out host preferences. Female fed-flies were dissected for parasitological investigation. Lizards from these areas were also trapped and blood and internal organs were examined and cultured for isolation of leishmania parasites. Only the lizards obtained from West Pokot and Baringo revealed leishmania parasites in cultures. Bloodmeal analysis showed that Phlebotomus anten-natus, P. bedfordi, P. schwetzi, P. africanus, P. martini, P. affinis and P. rodhaini fed on reptiles. Additionally, promastigotes were seen and isolated from these sandfly species. The disease incidence appears to be less in areas where leishmania parasites were encountered both in lizards and the sandflies in the same locality.
