The Kinematics of Quasar Broad Emission Line Regions Using a Disk-Wind Model

The structure and kinematics of the broad line region in quasars are still unknown. One popular model is the disk-wind model that offers a geometric unification of a quasar based on the viewing angle. We construct a simple kinematical disk-wind model with a narrow outflowing wind angle. The model is combined with radiative transfer in the Sobolev, or high velocity, limit. We examine how angle of viewing affects the observed characteristics of the emission line. The line profiles were found to exhibit distinct properties depending on the orientation, wind opening angle, and region of the wind where the emission arises.
At low inclination angle (close to face-on), we find that the shape of the emission line is asymmetric, narrow, and significantly blueshifted. As the inclination angle increases (close to edge-on), the line profile becomes more symmetric, broader, and less blueshifted. Additionally, lines that arise close to the base of the disk wind, near the accretion disk, tend to be broad and symmetric. Single-peaked line profiles are recovered for the intermediate and equatorial wind. The model is also able to reproduce a faster response in either the red or blue sides of the line profile, consistent with reverberation mapping studies.
Abnormalities in serum biomarkers correlate with lower cardiac index in the Fontan population

Background
Fontan survivors have depressed cardiac index that worsens over time. Serum biomarker measurement is minimally invasive, rapid, widely available, and may be useful for serial monitoring. The purpose of this study was to identify biomarkers that correlate with lower cardiac index in Fontan patients.
Methods and resultsThis study was a multi-centre case series assessing the correlations between biomarkers and cardiac magnetic resonance-derived cardiac index in Fontan patients ⩾6 years of age with biochemical and haematopoietic biomarkers obtained ±12 months from cardiac magnetic resonance. Medical history and biomarker values were obtained by chart review. Spearman’s Rank correlation assessed associations between biomarker z-scores and cardiac index. Biomarkers with significant correlations had receiver operating characteristic curves and area under the curve estimated. In total, 97 cardiac magnetic resonances in 87 patients met inclusion criteria: median age at cardiac magnetic resonance was 15 (6–33) years. Significant correlations were found between cardiac index and total alkaline phosphatase (−0.26, p=0.04), estimated creatinine clearance (0.26, p=0.02), and mean corpuscular volume (−0.32, p<0.01). Area under the curve for the three individual biomarkers was 0.63–0.69. Area under the curve for the three-biomarker panel was 0.75. Comparison of cardiac index above and below the receiver operating characteristic curve-identified cut-off points revealed significant differences for each biomarker (p<0.01) and for the composite panel [median cardiac index for higher-risk group=2.17 L/minute/m2 versus lower-risk group=2.96 L/minute/m2, (p<0.01)].
ConclusionsHigher total alkaline phosphatase and mean corpuscular volume as well as lower estimated creatinine clearance identify Fontan patients with lower cardiac index. Using biomarkers to monitor haemodynamics and organ-specific effects warrants prospective investigation.

Parental Distress, Parenting Practices, and Child Adaptive Outcomes Following Traumatic Brain Injury

Moderate and severe pediatric traumatic brain injuries (TBI) are associated with significant familial distress and child adaptive sequelae. Our aim was to examine the relationship between parental psychological distress, parenting practices (authoritarian, permissive, authoritative), and child adaptive functioning 12–36 months following TBI or orthopedic injury (OI). Injury type was hypothesized to moderate the relationship between parental distress and child adaptive functioning, demonstrating a significantly stronger relationship in the TBI relative to OI group. Authoritarian parenting practices were hypothesized to mediate relationship between parental distress and child adaptive functioning across groups. Groups (TBI n = 21, OI n = 23) did not differ significantly on age at injury, time since injury, sex, race, or SES. Parents completed the Brief Symptom Inventory, Parenting Practices Questionnaire, and Vineland-II. Moderation and mediation hypotheses were tested using hierarchical multiple regression and a bootstrapping approach, respectively. Results supported moderation and revealed that higher parental psychological distress was associated with lower child adaptive functioning in the TBI group only. Mediation results indicated that higher parental distress was associated with authoritarian parenting practices and lower adaptive functioning across groups. Results suggest that parenting practices are an important area of focus for studies attempting to elucidate the relationship between parent and child functioning following TBI. Summary
The majority of therapeutic monoclonal antibodies (mAbs) on the market and in development focus primarily on a limited set of targets selected on the basis of a few well-studied pathways. Truly novel targets (and their corresponding therapeutic mAbs) are rare and carry increased risk and challenges to develop because they, or the pathways they are involved in, are often neither well characterized nor extensively validated. The Raven therapeutic mAb discovery platform is especially efficient in discovering novel targets. Because the platform utilizes intact, living cells as the immunogen – and thus targets antigens present on the membrane of living cells – it is not biased upfront toward a particular protein, protein family, or signaling pathway. In addition, the presentation of these membrane targets in their fully processed and modified configuration and orientation in the living cell enables the discovery of mAbs to conformational epitopes as well as post-translationally modified epitopes. These epitopes may have greater tumor specificity and antitumor activity than those raised from less biologically relevant input such as purified or recombinant proteins and peptides. These epitopes can include binding sites on carbohydrates or lipids as well as conformational epitopes. In fact, the ability to discover these specific and active epitopes, not obvious when looking at mRNA or protein sequences, may open an entirely new class of antibody targets for cancer and other diseases. RAV12 is one example of a mAb that targets a carbohydrate epitope.
