Information in the literature and unpublished results of the authors on Dobinea were used to determine the kind [class(es)] of seed (true seed + endocarp) dormancy and of non-dormancy of genera in all five tribes of Anacardiaceae, and the results were examined in relation to the taxonomic position and endocarp anatomy within the family. Reports of both seed germination and endocarp anatomy were found for 15 genera in tribe Spondiadeae, 6 in tribe Anacardieae, 30 in tribe Rhoeae, 3 in tribe Semecarpeae and 1 in tribe Dobineeae. In Spondiadeae (Spondias-type endocarp), Anacardieae, Semecarpeae and Dobineeae (Anacardium-type endocarp), seeds are either non-dormant (ND) or have physiological dormancy (PD). In Rhoeae (Anacardium-type Rhoeae Groups A, B, C and D endocarps), on the other hand, seeds are ND or have physical dormancy (PY), PD or PY + PD. PY/PY + PD in this tribe seems to be restricted (or nearly so) to Rhus s.s. and closely related genera (e.g. Cotinus, Malosma and Toxicodendron) with an Anacardium-type Rhoeae Group A endocarp. However, seeds of other genera (e.g. Astronium and Schinus) with this type of endocarp and those with Rhoeae Group B (e.g. Pistacia), Group C (e.g. Pentaspadon) and Group D (e.g. Heeria) endocarps are either ND or have PD. The fossil fruit record strongly suggests that present-day relationships between diaspore dormancy (or non-dormancy), endocarp structure and taxonomic position within Anacardiaceae extend back to at least the Palaeogene.

The ability to initiate radicle elongation, or germination potential, occurs in developing embryos before the completion of seed maturation. Green embryos after the walking-stick stage in developing Arabidopsis thaliana seeds germinate when excised from seeds and incubated in Murashige–Skoog (MS) medium containing 1% sucrose. Germination potential is not observed during early embryogenesis at the globular, heart and torpedo stages. Here, we describe an Arabidopsis mutant with embryos arrested at early stages of development, but still exhibiting germination potential. The mutant, termed embryo ball (eb), produced shrunken seeds containing round or irregularly shaped embryos that did not germinate. The round embryos excised from developing eb seeds were capable of growing a primary root with root hairs when incubated in media. In contrast, cotyledons were absent at the apical region of the eb embryos, although the apical region produced leaf-like structures with trichomes, indicating vegetative leaf identity. These observations suggested that morphological maturation was not essential for the induction of germination potential. The eb embryos exhibited partial desiccation tolerance that is characteristic of mature embryos at later stages of development, suggesting that cell maturation was also independent of morphological maturation. The eb mutant provides novel information on cell and tissue identity in developmental biology, as well as a useful tool to dissect the mechanisms underlying the induction of germination potential in developing seeds.

Morphophysiological dormancy (MPD) is predominantly found in seeds of temperate regions and is uncommon in arid biomes. MPD has been reported in a number of Hibbertia (Dilleniaceae) species of temperate Australia, and in a single species of the arid zone, H. glaberrima. This study aimed to examine the dormancy and germination ecology of seeds of H. glaberrima. Seeds were subjected to temperature stratification treatments designed to mimic summer and autumn conditions in the Pilbara region of Western Australia. Seed germination and embryo growth were measured. We also tested the interaction between temperature stratification and cycles of drying and wetting designed to mimic sporadic rainfall events. All temperature and moisture treatments were tested in combination (+/–) with the smoke-derived chemical karrikinolide (KAR1). Exposing dormant seeds to temperatures suitable for warm stratification (35°C) for ≥ 8 weeks, followed by incubation at 25°C, resulted in significantly higher germination compared with non-stratified seeds. Exposing seeds to dry/wet cycling in conjunction with temperature stratification did not significantly increase germination. Exposure to KAR1 increased germination under most conditions. Once seeds are shed during October to December, they are exposed to hot and sporadically wet conditions over summer, allowing MPD to be overcome in a proportion of the seed population. Seeds may germinate in autumn (March to April), in conjunction with cooler temperatures. More deeply dormant individuals may require more than one summer to overcome dormancy. Similar to other species occurring in fire-prone ecosystems, fire also plays a crucial role in the germination ecology of H. glaberrima.

Medicago truncatula, soybean (Glycine max), Arabidopsis thaliana and rice (Oryza sativa) all belong to the core angiosperm group of plants. Seed-specific genes are important for seed formation and development in these angiosperms. The identification of genes specifically expressed in angiosperm seeds and the comparison of the expression patterns of homologous genes among different angiosperm species can provide novel insights into the functions of genes that control seed development and the evolution of angiosperms. We downloaded the sequences and expression data from the relevant databases, and the seed-specific expression of genes was identified with cut-offs of a gene expression level ratio ≥ 5 and a Z-score ≥ 6. The genes were analysed using local BLAST software with an E-value ≤ 1.0E − 505. A total of 605, 581, 778 and 722 genes showed specific expression in the seeds of Medicago, soybean, Arabidopsis and rice, respectively. Additionally, we compared the expression patterns of seed-specific genes from each species with their homologues in the other three species, and found that the degree of variation in the expression patterns of homologous genes was low among closely related species but higher among more distantly related ones. The discrepancy between the homologous gene expression patterns may be caused by the different characteristics of the cis-elements in the promoter regions of the homologous genes.

The purpose of the present study was to investigate the dormancy break and germination requirements of seeds from the rhamnaceous vine Berchemia scandens. The fleshy fruit contains a two-locular stone with an endocarp described as ‘bony, thickish’. Scarified and non-scarified stones increased by about 30–50% in mass during imbibition over a 24-h period. The endocarp of the stone does not completely enclose the seeds and a soft tissue region is present. This region is the primary area of water entrance to the seed, as shown by dye-tracking and by sealing it. Freshly matured and overwintered seeds of B. scandens germinated to low percentages at all temperatures during 2 weeks of incubation in light, and they germinated from moderate to high percentages during 12–14 weeks of incubation in light. While cold stratification had a relatively modest effect on the promotion of total germination across most temperatures assessed (if seeds were left for long enough), it had a somewhat stronger effect on germination rate. Cold-stratified seeds germinated equally well in light and darkness. The class of dormancy found in seeds of B. scandens would be physiological. The anatomy of the stones readily allows water imbibition, showing that seeds of B. scandens lack physical dormancy, an uncommon trait in Rhamnaceae.

Ageing of wheat seeds (Triticum aestivum cv. Galahad, 1986 harvest) during storage is accompanied by an increase in the proportion of abnormal seedlings produced when aged seed lots germinate. The response of aleurone layers from normally and abnormally germinated seeds to heat shock has been investigated. [14C]-amino acid incorporation into aleurone layers from 3-d normally and abnormally germinated seeds diminished after 4 h at 42°C but the aleurone layers were able to recover significantly during a 5-h period at 20°C following heat shock. [35S]-methionine pulse-labelling of aleurone proteins showed that α-amylase isoenzyme synthesis was abolished upon heat shock in aleurone layers for both normally and abnormally germinated seeds and that only aleurone layers from normally germinated seeds were capable of recommencing substantial α-amylase synthesis during the recovery period. Heat shock caused aleurone layers from abnormally germinated seeds to synthesize polypeptides not synthesized by aleurone layers from normally germinated seeds under the same conditions. The synthesis of certain polypeptides by aleurone layers from abnormally germinated seeds was enhanced at 20°C, 42°C and during recovery compared with normally germinated seeds. These results are discussed in terms of the control of gene expression during heat shock and recovery during seed ageing.

In wetlands, dormancy may be a key functional trait enabling seeds to avoid underwater germination, which could be lethal for seedling establishment. Our objectives were to find out (i) if shallow dormant (i.e. conditionally dormant) Echinochloa crus-galli seeds from an anaerobic germination resistant accession can break dormancy under hypoxic submergence and (ii) if underwater germination can be restored in scarified, non-dormant seeds. Shallow dormant E. crus-galli seeds perceived diurnally alternating temperatures (AT) and red light (R) pulses (i.e. dormancy-breaking cues) under hypoxic submergence; however, an inhibitory far-red light pulse given at the end of the 4-d inundation period demonstrated that most of the seeds (85%) were unable to break dormancy. Scarified E. crus-galli seeds, which did not express dormancy under drained conditions, were unable to germinate under hypoxic submergence, despite being exposed to dormancy-breaking cues (AT + R). Lastly, the temporal window for germination sensitivity to the inhibitory action of hypoxia, once dormancy-breaking signals have been applied, is progressively lost and bounded to approximately 18 h for half of the seed lot. These results highlight the importance of dormancy as a trait enabling E. crus-galli seeds to avoid underwater germination, a risky scenario for seedling emergence and establishment in this facultative hydrophyte.

Zephyranthes tubispatha is an ornamental species distributed along several countries of South America. Although it can be multiplied through bulbs or scales, seed germination is a simpler and more cost-effective process. Temperature plays a major role in the control of germination; however, its effect has been scarcely investigated in this species. In the present work, we characterized the germination responses of Z. tubispatha seeds to different temperatures and analyzed the role of key components of the antioxidant metabolism and phytohormones in their control. Seeds showed an optimal temperature range for germination between 14 and 20°C, with higher temperatures (HTs) being progressively inhibitory. While germination was almost nil above 28°C, it could be recovered after transferring the seeds to 20°C, suggesting that thermoinhibition was the underlying phenomenon. The duration of the HT incubation period affected both the time to germination onset and the germination rate at 20°C. Similarly, the activity of antioxidant enzymes, the production of reactive oxygen species in the embryo and the sensitivity to some germination promoters varied depending on the duration of the HT treatment. The addition of 20 μM fluridone was sufficient to recover germination dynamics as in the control treatment when given after a long-term incubation period (25 d) at HT. Ethephon supply was more effective than gibberellins to suppress thermoinhibition, suggesting that changes in the balance and/or sensitivity to ethylene and abscisic acid over time play an important role in the regulation of germination responses to thermal cues in this species.

Seeds often survive in the soil in a dormant state, but their persistence is endangered by micro-organisms that could feed on them. Seed–microbe interactions in the soil are, however, poorly understood. We used dormant caryopses of red rice to study the defence response induced by pronase, a mixture of proteases secreted by Streptomyces griseus, a non-pathogenic bacterium. Pronase was shown to activate the plant immune reaction, indicating that its activity was recognized as a potential microbial attack. The defence reaction included extracellular alkalinization and superoxide production, and the former was necessary to activate the latter, since buffering at pH 6 inhibited the oxidative burst. Alkalinization was sufficient to trigger the oxidative burst, as superoxide production increased when caryopses were incubated in buffered solutions of increasing pH without pronase. Release of proanthocyanidins was observed, with or without pronase. These diverse mechanisms are hypothesized to cooperate in reinforcing seed protection. Finally, time profiles of superoxide production by dormant and non-dormant red rice caryopses during imbibition did not support a relationship between extracellular superoxide and dormancy breaking or germination. Thus, the role of this reactive oxygen species in red rice imbibed caryopses appears to be essentially aimed at defence against attacks by challenging micro-organisms.

A novel technique allows long-term monitoring of real-time oxygen consumption during seed germination in an open system. Most current techniques used to detect oxygen consumption by seeds measure the decrease in oxygen concentration in a closed chamber. This is not ideal for long-term experiments because the chamber must be replenished with air periodically, subjecting the seeds to abrupt changes in oxygen concentration. The current technique employs an open system, in which seeds are submerged in a continuously aerated aqueous environment. Oxygen electrodes are used to measure the steady-state concentration of oxygen in the solution, which is a function of both the rate of oxygen consumption by the seed and the rate of aeration from the atmosphere. The rate of aeration is directly dependent on the oxygen concentration of the bathing solution; therefore, previous calibration of the system allows the direct conversion of steady-state oxygen concentrations into oxygen consumption rates. Because oxygen is not limiting, the experimental design described here can monitor the same sample non-intrusively every minute for more than 24 h, allowing for greater precision than hourly readings often reported with current techniques. Multiple treatments and/or replicates can be run simultaneously, allowing sensitive comparison of various seed treatments or seed types. To illustrate its potential application, the technique was used to follow the rehydration and pre-emergence phases of germination of cucumber (Cucumis sativum), pea (Pisum sativum) and mustard (Brassica juncea) seeds, detect the inhibitory effects of surface sterilization techniques on seed respiration of cucumber, and follow the interaction of a bacterial biocontrol agent with germinating cucumber and pea seeds.

The endosperm and cotyledon tissues of Ricinus communis seeds and young seedlings were examined for changes in the mineral nutrient composition of globoids during early seedling growth. The effect on globoid composition of providing mineral nutrients to the developing seedling was also investigated. Globoids in endosperm and cotyledon tissues of castor bean seeds contained P, Mg and K, as well as trace amounts of Ca, Fe and Zn. Irrespective of the addition of mineral nutrients, K content in globoids of endosperm and cotyledon tissues declined significantly during initial seedling growth. During early seedling growth, amounts of Fe, Zn and Ca increased in cotyledon globoids. Ca contents of globoids of endosperm tissues also increased. The changes in Fe, Zn and Ca globoid contents were not influenced by providing mineral nutrients to growing castor bean seedlings.

Amaranthus albus L. seeds able to germinate in darkness can be prevented from germinating by prolonged irradiation with incandescent light (CIL). The germination in darkness is controlled by pre-existent Pfr. Allowing the seeds to remain in darkness for about 16 h allows an escape from the inhibition by CIL, about comparable with the time required to escape from pre-existent Pfr control. By examining escape from low fluence far-red (FR), it was found that no escape occurred in CIL. It was concluded that the mode of action of CIL in inhibiting A. albus seed germination involved prevention of the action of the pre-existent Pfr and a subsequent dormancy which does not allow a response to the Pfr generated by the incandescent light. Other actions of CIL were found but were considered less likely as causative for the inhibition of germination. One was a direct inhibition of radicle elongation and another involved prevention of an increased responsiveness to Pfr induced by treatment at 40°C.

Barriers to water permeability in prickly sida seeds were characterized using solvents and water-soluble salts. The seeds imbibed water when barriers in the chalazal area broke down. The major barrier was in the distal region of the palisade layer bordered internally by the light line. Soaking dormant seeds in certain organic solvents overcame the barrier and enabled the seeds to imbibe water. The most active were non-polar solvents with dielectric constants between 3.7 and 18.5, and with a high ability to hydrate and swell cellulosic materials. We concluded that the most active solvents (pyridine, diethylamine) overcame the major barrier, entered the proximal portion of the palisade layer and caused the swelling of cellulosic cell components. The cellular expansion created stresses that broke thin walls of the underlying subpalisade cells in the chalazal area. The breakage resulted in separation of the cell layers and rapid entry of water into the seed. Highly non-polar or highly polar solvents had little or no activity. Some solvents overcame the barrier but did not cause swelling and disruption of the cell layers. We propose that the natural breakdown of impermeability in prickly sida seeds was due to the gradual seepage of water past the barrier in the distal portion of the coat in the chalazal area and into the inner portions where swelling of hygroscopic components triggered the breakage of subpalisade cell walls resulting in cell layer separation.

A nuclear protein (QP47) is synthesized during the last stage of seed maturation when the embryo cells start to dehydrate and enter a condition of metabolic quiescence. This protein is localized in the nucleoplasm surrounding the chromosomes. The correlation existing between the synthesis of QP47 and arrest of cell proliferation, suggests that the presence of this protein in the nucleus could influence its metabolic activities. This hypothesis is supported by the fact that degradation of this protein precedes resumption of cell proliferation during the early stage of radicle elongation.

Cistus species have seeds with hard coats which impose physical seed dormancy that can be released after seed scarification. In fire-prone habitats, the break of physical seed dormancy is usually related to the heat produced during fires. It is commonly accepted that most hard-seeded species, including those of the genus Cistus, are able to germinate under a wide range of temperatures in light as well as in darkness, once the seed becomes permeable. However, although many studies have focused on the release of physical dormancy only, a few have done so on the effect of environmental factors once dormancy is released. In this research, through a factorial experiment, we analysed the effects of light (light and darkness) and a range of temperatures (10, 15, 20, 25 and 30°C) on the seed germination of eight Cistus species after a heat shock. On average, almost 60% of the seeds did not germinate despite being viable, and this lack of germination increased with higher temperatures during the treatment. Although an idiosyncratic germination response emerged, temperature had a significant effect in all the species, reaching the highest levels of germination between 10 and 20°C. Light interacted with temperature in four cases by increasing the germination, especially under the least favourable temperatures. Environmental factors, such as temperature and light, appear to modulate the germination of the studied Cistus species after the release of physical seed dormancy.

Functional traits related to regeneration responses to the environment are highly determinants of distribution patterns of plant communities. A large body of studies on seed traits suggests that regional climate may act as a strong filter of plant recruitment; however, few studies have evaluated the relative importance of seed traits and environmental filters for seed persistence at the population level. We tested the role of seed mass, water content and desiccation tolerance, as well as the germination time as proxies for seed tolerance to environmental filters (water deficit, heat shock and high temperatures) by comparing the response of tree species co-occurring in savannas located in different regions: Cerrado biome of Central Brazil and the Rio Branco savannas of northern Brazil. Seeds collected in savannas of Rio Branco showed a higher tolerance to environmental filters than those collected in savannas of the Cerrado. While the germination percentages largely varied in response to the treatments, the germination times were virtually unaffected by them, irrespective of seed origin, seed mass and water content. At the population level, the regional environment was a key determinant of seed tolerance to stress, irrespective of seed traits. Germination time was shown to represent a conservative seed trait and more linked to a species-specific germination strategy than to regional characteristics. Our results suggest that recruitment patterns of Cerrado savannas may be more impacted than Rio Branco savannas by the climate scenarios predicted for the future.

Protein disulfide isomerase (PDI E.C. 5.3.4.1) catalyses the formation of disulfide bonds in secretory or cell-surface proteins during their biosynthesis. PDI activity is associated with the microsomal fraction of the cell and has been found in several mammalian tissues and in developing wheat endosperm.

The embryo and aleurone layer of germinating seeds of the wheat cultivar Galahad showed PDI activity associated with a microsome-enriched fraction of cell homogenates PDI activity in microsome-enriched fractions from aleurone layers of normally germinating seeds was significantly higher than in their abnormally germinating counterparts and diminished as germination proceeded.

Dry embryos and germinating embryos showed less PDI activity than aleurone layers. There was no significant difference in PDI activity between microsome-enriched fractions from embryos of normally and abnormally germinating seeds at 3 or 6 days of germination.

Wheat aleurone PDI was partially purified using gel filtration and had a molecular mass of 110–130 kDa and a monomeric molecular mass of ca. 57 kDa on sodium dodecyl sulfate polyacrylamide gel electrophoresis.

Osmotic priming reduces the longevity of Vigna radiata (mung bean) seeds during subsequent dry storage. The present study has investigated the relationship between volumetric changes, modified water sorption properties and the decrease in seed longevity after priming. Volumetric analysis of control embryos revealed two major dehydration-related volumetric contractions in the biopolymer matrix, reflecting structural reorganization in biopolymer matrix during drying. These contractions occurred at hydration from 0.29 to 0.23 g g–1 dw and from 0.18 to 0.11 g g–1 dw, respectively. Volumetric contractions were reduced significantly after priming and also occurred at higher water contents (i.e. 0.48–0.32 g g–1 dw and 0.21–0.11 g g–1 dw). Consequently, at the same water content, primed seed embryos had higher specific biopolymer volume and lower specific density than control embryos. Water sorption study showed that priming did not change the monolayer hydration value, but altered the properties of multilayer water sorption in seed axes. The analysis of water clustering function suggested that priming enhanced the water–water association in seed axes, but not in cotyledon tissues. Solid-state 1H-NMR (nuclear magnetic resonance) investigation confirmed that priming did not cause a significant change of water dynamic properties in cotyledon tissues at water contents below 0.20 g g–1 dw. The relevance of volumetric modifications of the biopolymer matrix and changes in water clustering function to the reduced seed longevity after priming is discussed. It is proposed that priming-induced increases in surface reactivity may enhance deteriorative chemical reactions in re-dried seed tissues.

Information about seed dormancy cycling and germination in relation to temperature and moisture conditions in the natural environment is important for the conservation and restoration of rare species, including Begonia guishanensis and Paraisometrum mileense, two sympatric perennial limestone (karst) species. Dry afterripening (DAR) and wet and dry (WD) cycles at 15/5 and 25/15°C as well as moist chilling (MC) at 15/5°C were used to mimic the natural environment at different times of the year. A field experiment was conducted to monitor seasonal changes in germination responses of the seeds. About 40–65% of B. guishanensis and 5% of P. mileense seeds were dormant at maturity. DAR at 25/15 and 15/5°C as well as MC and WD cycles at 15/5°C alleviated dormancy for B. guishanensis but not P. mileense, and WD cycles at 25/15°C induced a deeper conditional dormancy for both species. Seeds of B. guishanensis exhibited dormancy cycling in the field, with increased dormancy under natural WD cycles at relatively high temperatures during the transition from the dry to the wet season in April to May and decreased dormancy during the wet season from June to October. KNO3 mitigated the dormancy-inducing effect of both artificial and natural WD cycles at relatively high temperatures for B. guishanensis. The field experiment indicated that seeds of B. guishanensis may be able to form a persistent soil seed bank, while almost all seeds of P. mileense germinate at the beginning of the wet season in the field.

We used two sunflower genotypes displaying pericarp-imposed dormancy at high incubation temperatures (i.e. 30°C) to investigate the role of the pericarp as a limitation to oxygen availability to the embryo (hypoxia), and its impact on embryo abscisic acid (ABA) content and sensitivity to ABA. Results showed that both genotypes displayed very different oxygen threshold values for inhibition of embryo germination when incubation was performed at 30°C. Expression of dormancy in one genotype was therefore related to exacerbated embryo sensitivity to hypoxia, whereas in the other genotype, the pericarp appeared to act as a more severe restraint to oxygen entry. Increased sensitivity to hypoxia was, in part, related to increased sensitivity to ABA, but not to alterations in ABA metabolism. The activity of pericarp-microbial communities (bacteria and fungi) at high temperatures was also assessed as a potential determinant of hypoxia to the embryo. Oxygen consumption in pericarps incubated at 30°C was attenuated with antibiotics, which concomitantly promoted achene germination. In agreement with the observed more severe oxygen deprivation to the embryo exerted by the pericarp, the bacterial load in the pericarp was significantly higher in the commercial hybrid than in the inbred line; however, the application of antibiotics strongly reduced the bacterial colony counts for each genotype. Different bacterial and fungal communities, assessed through their profiles of carbon-source utilization, were determined between genotypes and after treatment with antibiotics. This work highlights the relationship between enhancement of sensitivity to hypoxia with incubation temperature and seed dormancy expression, and suggests that microbial activity might be part of the mechanism through which hypoxia is imposed.
